Simple Summary: The molecular basis of prostate cancer progression to advanced disease is not fully understood. Small non-coding RNAs, called microRNAs regulate gene expression post-translationally and regulate crucial cellular processes. It has been reported that microRNAs are dysregulated during prostate cancer progression. The objective of this study is to determine the role of miR-410-3p, a little-studied microRNA in prostate cancer progression. Our results demonstrate for the first time that this miRNA plays a context-dependent role in prostate cancer. We found that miR-410 expression is regulated in a biphasic manner in prostate cancer, with its expression decreased in primary prostate cancer and increased in the advanced disease. We further performed functional studies that show that miR-410 regulates critical cellular processes that are involved in cancer progression and metastasis in prostate cancer. Our study suggests that miR-410-mediated regulation of target genes is context-dependent. Our findings would lead to an increased understanding of the mechanistic basis of prostate cancer progression and metastasis that would be helpful in guiding novel therapies. The molecular basis of prostate cancer (PCa) progression from the primary disease to metastatic castration-resistant prostate cancer (CRPC) followed by therapy-induced neuroendocrine prostate cancer is not fully understood. In this study, we elucidate the role of miR-410, a little-studied microRNA located on chromosome 14q32.31 within the DLK1-DIO3 cluster, in PCa. miR-410 expression analyses in primary and metastatic PCa tissues and cell lines show that its levels are decreased in initial stages and increased in advanced PCa. Functional studies were performed in a series of PCa cell lines. In LNCaP cells, miR-410 overexpression led to decreases in cellular viability, proliferation, invasiveness, and migration. On the other hand, miR-410 overexpression in PC3 and C42B cells led to increased viability, proliferation, and invasiveness. Our data suggest that miR-410 represses epithelial-to-mesenchymal transition (EMT) in LNCaP cells by directly repressing SNAIL. However, it promotes EMT and upregulates PI3K/Akt signaling in PC3 and C42B cells. In vivo studies with PC3 xenografts support an oncogenic role of miR-410. These data suggest that miR-410 acts as a tumor suppressor in the initial stages of PCa and play an oncogenic role in advanced PCa. The expected increase in urbanization and population in coming years is going to increase the impervious land area, leading to substantial increases in stormwater runoff and hydrological challenges, and presents significant challenges for urban potable water supply. These are worldwide challenges that can potentially be ameliorated by harvesting stormwater for potable use or for other uses that can reduce the pressure on potable water supply. This study sought to assist the local water authority in planning for future potable water supply through a review of the scientific literature to determine the likely chemical and microbial characteristics of stormwater, the treatment train (TT) requirements, and the likely costs of treatment to achieve potable standards for the high-growth metropolitan region of Melbourne, Australia. Literature stormwater quality statistical data and treatment process performance data were used to model the expected product water microbial and chemical quality after treatment using a number of advanced TT options. The results of the modelling were compared with literature microbial log reduction targets (LRTs) for the potable use of stormwater and with the Australian Drinking Water Guidelines (ADWG). It was found that a reverse osmosis (RO)-based TT with microfiltration pre-treatment and post-RO advanced oxidation and chlorination in storage reservoirs is a conservative stormwater potable use treatment option. A less conservative and less expensive ozone-and-biologically active filtration (O3/BAF)-based TT option is also proposed if RO concentrate disposal is deemed to be too challenging. These results could be useful in climate change adaptation involving the evaluation of options for the mitigation of future population-growth- and climate-change-driven water supply challenges, as well as urbanization-driven stormwater hydrology and receiving water pollution challenges. This study discovers the complement protein profile in the aqueous humor (AH) of human subjects and investigates its association with primary open-angle glaucoma (POAG) pathogenesis. Among the 32 complement proteins identified, 22 were highly abundant and detected in more than 50% of AH samples. The most predominant active complement proteins in the AH are C3, C4B, C4A, CFB, CFD, and C9. Additionally, the most prevalent complement regulators and receptors include CLU, SERPING1, F2, CFH, CFI, and VTN. Significant alterations in complement proteins were observed in individuals with POAG compared to those with cataracts. Specifically, complement protein F2 was upregulated, while C8G, C6, and CFH were downregulated in POAG samples. Stratification of the samples by race and sex revealed distinct alterations of complement proteins in patients with POAG. In the African American cohort, five complement proteins (C4A, C4B, F2, C7, and C3) were upregulated in POAG compared to cataract patients. In the Caucasian cohort, eight complement proteins (C3, SERPING1, CFI, CLU, CFHR1, C8G, C6, and CFH) were downregulated in the POAG samples compared to the cataract samples. Within the male cohort, three complement proteins (CLU, C6, and CFH) were downregulated in POAG patients compared to those with cataracts. Whereas, within the female cohort, two complement proteins (C4B and F2) were upregulated and one (C8G) downregulated in the POAG samples when compared to cataracts. Discerning these changes in the AH complement protein profile will assist in the development of tailored therapies to modulate the complement system for managing ocular disorders. The Urban Heat Island (UHI) effect is a significant concern in today's rapidly urbanising cities, with exacerbating heatwaves' impact, urban livelihood, and environmental well-being. This study aims to assess the cooling effect of blue-green spaces in Bhubaneswar, India, and explore their implications for mitigating UHI effects. Satellite images were processed with Google Earth Engine (GEE) to produce information on the blue-green spaces' land surface temperatures (LST). The Normalised Difference Vegetation Index (NDVI) and Modified Normalised Difference Water Index (MNDWI) were employed to quantify the presence and characteristics of these blue-green spaces. The findings revealed significant spatial variations in the LST, with higher temperatures observed in bare land and built-up areas and lower temperatures in proximity to the blue-green spaces. In addition, a correlation analysis indicated the strong influence of the built-up index (NDBI) on the LST, emphasising the impact of urbanisation on local climate dynamics. The analysis demonstrated the potential of blue-green spaces in reducing surface temperatures and mitigating UHI effects. Based on these results, strategic interventions were proposed, such as increasing the coverage of green spaces, optimising access to water bodies, and integrating water-sensitive design principles into urban planning to enhance the cooling effects and foster a more sustainable and resilient urban environment. This study highlighted the importance of leveraging remote sensing and GEE for urban UHI analyses. It provides valuable insights for policymakers and urban planners to prioritise nature-based solutions for heat mitigation in Bhubaneswar and other similar cities. Future research could delve deeper into a quantitative assessment of the cooling benefits of specific blue-green infrastructure interventions and explore their socio-economic impacts on urban communities. Simple Summary: Zinc is an essential mineral supplemented to human and pig diets as inorganic (inorganic salts) or organic (bound to an organic compound) sources. Some studies reported that pigs fed diets with organic zinc sources respond better to stress conditions than those fed diets with inorganic sources. In this study, we identified genes in the small intestine that may explain those differences. We fed finisher pigs with diets containing organic or inorganic zinc sources for 32 days and conducted gene expression analysis of intestinal ileum samples. The organic Zn source induced lesser level of pro-inflammatory cytokine interleukin 18 (IL18), but greater Toll-like receptor 2 (TLR2) gene expression than inorganic Zn, suggesting that an organic Zn source may improve the condition and immune response of animals under stress by modulating gene expression in the intestine. We also explored if intestinal epithelial cells, which are in direct contact with diet components, are the main cells showing these changes. We exposed intestinal organoids or epithelial "mini-guts" to zinc sources in vitro. Organoids showed changes related to Zn supplementations but did not reproduce the changes of immune-related genes observed in the animal samples, indicating cells other than epithelial are affected by Zn sources. Animals receiving Zinc (Zn) dietary supplementation with organic sources respond better to stress than inorganic Zn sources supplementation. The study aimed to identify the effect of different Zn sources on intestinal epithelial gene expression. In total, 45 pigs (9 per treatment) (77.5 ± 2.5 kg weight) were fed for 32 days, a corn-soybean meal diet without supplemented Zn (ZnR) or supplemented with 50 and 100 ppm of inorganic ZnCl2 (Zn50 and Zn100), and amino acid-bound organic Zn sources (LQ50 and LQ100). Gene expression changes form RNA-seq in ileum tissues of ZnR revealed changes associated with Zn insufficiency. Comparing organic with inorganic Zn sources by one-way ANOVA, pro-inflammatory cytokine interleukin 18 (IL18) was downregulated (p = 0.03) and Toll-like receptor 2 (TLR2) upregulated (p = 0.02). To determine the role of epithelial cells in response to dietary Zn, swine intestinal organoids (enteroids) were exposed to Zn restriction, ZnCl2 or LQ-Zn. In enteroids, ZIP4 expression decreased with added Zn compared with Zn-restriction (p = 0.006) but Zn sources did not affect (p > 0.05) IL18 or TLR2 expression. These results suggest that organic Zn may stimulate TLR2 signaling possibly affecting immune response, while decreasing the proinflammatory cytokine IL18 expression in non-epithelial cells of intestinal mucosa. Access to inexpensive, safe, consistent, and clean energy is a critical necessity for all to achieve the SDGs. India's renewable energy (RE) currently accounts for more than a third of the 482 GW of installed capacity and more than 40 percent of power production (including large-scale hydropower). Reforms such as the establishment of a single national power grid have improved access to electricity for people, and the ambitious development of renewable energy, which is the world's third-largest energy generator and third-largest electricity user, has helped in achieving these aims. As a result, the expansion of national targets signifies and reflects the country's optimism and goal for the forthcoming generation. Standardization of the guidelines and development of the stable grid and transmission networks will only enable the country to achieve the ambitious target of 500 GW of green and clean energy by 2030. This paper highlights the important development in the power sector regarding the energy security of India. As well as specifically examining the initiative of NSMs for achieving the 2030 targets, the key challenges, and the way forward to increase the cumulative installed capacity, comprehensive studies of various policies and government initiatives are also discussed. Furthermore, the key challenges usually faced by the developers in the industry, along with the steep decline and rise in the tariffs of solar projects and the previous trends in capacity installation, are also pointed out. This research work also highlights the potential key challenges to achieving the targets, and will thus provide a focus for power developers, policy makers, researchers, and industry practitioners and help with their planning. Globally, researchers have devoted consistent efforts to producing excellent coating properties since coating plays an essential role in enhancing electrochemical performance and surface quality. In this study, TiO2 nanoparticles in varying concentrations of 0.5, 1, 2, and 3 wt.% were added into the acrylic-epoxy polymeric matrix with 90:10 wt.% (90A:10E) ratio incorporated with 1 wt.% graphene, to fabricate graphene/ TiO 2 -based nanocomposite coating systems. Furthermore, the properties of the graphene/ TiO 2 composites were investigated by Fourier-transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), ultraviolet-visible (UV-Vis) spectroscopy, water contact angle (WCA) measurements, and cross-hatch test (CHT), respectively. Moreover, the field emission scanning electron microscope (FESEM) and the electrochemical impedance spectroscopy (EIS) tests were conducted to investigate the dispersibility and anticorrosion mechanism of the coatings. The EIS was observed by determining the breakpoint frequencies over a period of 90 days. The results revealed that the TiO 2 nanoparticles were successfully decorated on the graphene surface by chemical bonds, which resulted in the graphene/ TiO 2 nanocomposite coatings exhibiting better dispersibility within the polymeric matrix. The WCA of the graphene/ TiO 2 coating increased along with the ratio of TiO 2 to graphene, achieving the highest CA of 120.85° for 3 wt.% of TiO2. Excellent dispersion and uniform distribution of the TiO 2 nanoparticles within the polymer matrix were shown up to 2 wt.% of TiO2 inclusion. Among the coating systems, throughout the immersion time, the graphene/ TiO 2 (1:1) coating system exhibited the best dispersibility and high impedance modulus values ( Z 0.01   Hz ), exceeding 10 10 Ω   cm 2 . Integrated urban water management approaches (IUWM) are implemented to address challenges from increases in water demand as a result of population growth and the impact of climate change. IUWM aims to utilize all water resources (stormwater, wastewater, and rainwater) based on fit-for-purpose concepts. Here, a local water utility in Melbourne's Melton growth area explored the availability of stormwater as an alternative water resource for water service planning for a proposed residential development in an existing greenfield area of 13,890 hectares for 160,000 new houses by 2040. A methodology was developed for assessing the stormwater quantity and quality under land use change and different climatic conditions considering the availability of stormwater from the proposed urban development. The modelling results indicated that the amount of annual stormwater generated in the region increased by nearly four times to 32 GL/year under the 2040 full urban land use with high climate change. The provision of constructed wetlands in proposed development blocks was found to be efficient at removing TSS, TP, and TN, and able to retain over 90% of TSS, 77% of TP, and 52% of TN in all scenarios. Osteoarthritis (OA) is a highly prevalent degenerative joint condition that disproportionately affects females. The pathophysiology of the disease is not well understood, which makes diagnosis and treatment difficult. Given the physical connection of synovial fluid (SF) with articular tissues, the SF's composition can reflect relevant biological modifications, and has therefore been a focus of research. Previously, we demonstrated that extracellular vesicles isolated from the synovial fluid of OA patients carry different cargo (protein and miRNA) in a sex-specific manner. Given the increased prevalence and severity of OA in females, this study aims to identify differential protein content within the synovial fluid of female OA and non-osteoarthritic (non-OA) patients. We found that several proteins were differentially expressed in osteoarthritic females compared with age-matched controls. Presenilin, Coagulation Factor X, Lysine-Specific Demethylase 2B, Tenascin C, Leucine-Rich Repeat-Containing Protein 17 fragments, and T-Complex Protein 1 were negatively regulated in the OA group, with PGD Synthase, Tubulointerstitial Nephritis Antigen, and Nuclear Receptor Binding SET Domain Protein 1 positively regulated in the OA group. Database for Annotation, Visualization, and Integrated Discovery (DAVID) and QuickGO analyses established these proteins as significantly involved in many biological, cellular, and molecular processes. MicroRNAs are non-coding RNAs that serve as regulatory molecules in a variety of pathways such as inflammation, metabolism, homeostasis, cell machinery, and development. With the progression of sequencing methods and modern bioinformatics tools, novel roles of microRNAs in regulatory mechanisms and pathophysiological states continue to expand. Advances in detection methods have further enabled larger adoption of studies utilizing minimal sample volumes, allowing the analysis of microRNAs in low-volume biofluids, such as the aqueous humor and tear fluid. The reported abundance of extracellular microRNAs in these biofluids has prompted studies to explore their biomarker potential. This review compiles the current literature reporting microRNAs in human tear fluid and their association with ocular diseases including dry eye disease, Sjögren's syndrome, keratitis, vernal keratoconjunctivitis, glaucoma, diabetic macular edema, and diabetic retinopathy, as well as non-ocular diseases, including Alzheimer's and breast cancer. Ischemia-reperfusion (I/R) injury is a complication impacting multiple organs and tissues in clinical conditions ranging from peripheral arterial disease to musculoskeletal trauma and myocardial infarction. Stem cell-derived extracellular vesicles (EVs) may represent one therapeutic resource for preventing the tissue damage associated with I/R injury. Here we tested the hypothesis that lyophilized extracellular vesicles derived from adipose stem cells could serve as an "off-the-shelf" treatment modality for I/R injury in a mouse hindlimb ischemia model. Ischemia was induced for 90 min using a rubber band tourniquet and extracellular vesicles (0, 50, or 100 µg) administered via tail vein injection immediately prior to reperfusion. Perfusion was measured prior to, during, and after ischemia using laser Doppler imaging. Serum and tissue were collected 24 h after reperfusion. Mass spectrometry (MS)-based proteomics was used to characterize the EV cargo and proteins from the ischemic and non-ischemic hindlimb. Inflammatory cytokines were measured in muscle and serum using a multiplex array. Results indicate that EVs significantly increase reperfusion and significantly increase expression of the anti-inflammatory factor annexin a1 in skeletal muscle; however, the increased reperfusion was also associated with a marked decrease in muscle structural proteins such as dystrophin, plectin, and obscurin. Circulating inflammatory cytokines TNF-alpha and IL-6 were increased with EV treatment, and serum TNF-alpha showed a significant, positive correlation with reperfusion level. These findings suggest that, while EVs may enhance reperfusion, the increased reperfusion can negatively impact muscle tissue and possibly remote organs. Extreme rainfall events cause an increase in the flow into aging sewer networks, which can lead to Sanitary Sewer Overflows (SSOs). This literature review presents a complete assessment of the application of Water Sensitive Urban Design (WSUD) approaches as mitigation strategies for reducing rainfall-induced SSOs. The review highlights the various WSUD techniques identified in past studies for reducing sewer overflows. In these studies, it was identified that permeable pavements, green roofs, raingardens/bio-retention cells and rainwater tanks were the most popular WSUD strategies that have been extensively used in the past for the mitigation of sewer overflows. WSUD or "green" approaches also have enormous environmental, social and economic benefits when compared to the conventional "gray" approaches for sewer overflow mitigation. However, there have been limited studies conducted in the past that highlight and quantify the benefits of WSUD approaches for sewer overflow mitigation, particularly when such strategies are applied at a large scale (e.g., city scale). This review has identified the modelling software, SWMM, to be the most widely applied tool that has been used in the literature for WSUD modelling. It was also identified that with climate change-induced extreme rainfall events on the increase, WSUD-based "green" strategies alone may not be enough for the mitigation of sewer overflows. A suitable sewer overflow mitigation strategy could be green or a hybrid green-gray strategy, which would need to be identified based on a detailed context specific analysis. [ABSTRACT FROM AUTHOR]

The Nepalese Sunsari Morang Irrigation district is the lifeblood of millions of people in the Koshi River basin. Despite its fundamental importance to food security, little is known about the impacts of climate change on future irrigation demand and grain yields in this region. Here, we examined the impacts of climate change on the irrigation demand and grain yield of wheat crop. Climate change was simulated using Representative Concentration Pathways (RCPs) of 4.5 and 8.5 for three time horizons (2016–2045, 2036–2065, and 2071–2100) in the Agricultural Production Systems Simulator (APSIM). For the field data's measured period (2018–2020), we showed that farmers applied only 25% of the irrigation water required to achieve the maximum potential grain yield. Actual yields were less than 50% of the potential yields. Projected irrigation water demand is likely to increase for RCP4.5 (3%) but likely to decrease under RCP8.5 (8%) due to the truncated crop duration and lower maturity biomass by the end of the 21st century. However, simulated yields declined by 20%, suggesting that even irrigation will not be enough to mitigate the severe and detrimental effects of climate change on crop production. Hydrological modelling at a catchment scale was conducted to investigate the impact of climate change and land-use change individually and in combination with the available streamflow in the Painkalac catchment using an eWater Source hydrological model. This study compares the performance of three inbuilt conceptual models within eWater Source, such as the Australian water balance model (AWBM), Sacramento and GR4J for streamflow simulation. The three-model performance was predicted by bivariate statistics (Nash–Sutcliff efficiency) and univariate (mean, standard deviation) to evaluate the efficiency of model runoff predictions. Potential evapotranspiration (PET) data, daily rainfall data and observed streamflow measured from this catchment are the major inputs to these models. These models were calibrated and validated using eight objective functions while further comparisons of these models were made using objective functions of a Nash–Sutcliffe efficiency (NSE) log daily and an NSE log daily bias penalty. The observed streamflow data were split into three sections. Two-thirds of the data were used for calibration while the remaining one-third of the data was used for validation of the model. Based on the results, it was observed that the performance of the GR4J model is more suitable for the Painkalac catchment in respect of prediction and computational efficiency compared to the Sacramento and AWBM models. Further, the impact of climate change, land-use change and combined scenarios (land-use and climate change) were evaluated using the GR4J model. The results of this study suggest that the higher climate change for the year 2065 will result in approximately 45.67% less streamflow in the reservoir. In addition, the land-use change resulted in approximately 42.26% less flow while combined land-use and higher climate change will produce 48.06% less streamflow compared to the observed flow under the existing conditions. Water and wastewater services have been provided through centralised systems for more than a century. The operational and management approaches of the water systems face challenges induced by population growth, urbanisation, and ageing infrastructure. Recent advancements in water system engineering include the development of intelligent water networks. These intelligent networks address management and operational challenges associated with pressure and flow variations in the water network and it reduces the time for identification of pipe bursts and leakages. Research is required into the development of intelligent water networks to ensure consistent data collection and analysis that can filter and aggregate into actionable events to reduce water leakage, leakage cost, customer disruptions, and damages. Implementation of an intelligent algorithm with an integrated Supervisory Control and Data Acquisition (SCADA) system, high-efficiency smart sensors, and flow meters, including a tracking mechanism, will significantly reduce system management and operational issues and ensure improved service delivery for the community. This paper discusses the history of water systems, traditional water supply systems, need for intelligent water network, and design/development of the intelligent water networks. This paper presents a monitoring-based investigation of rainwater collection systems using economic performance indicators in a group of households with nonconventional end-uses for rainwater that are not traditionally associated with rainwater supply. The monitored data for five household rainwater tank systems were analysed in two stages. For the first stage, the data was empirically analysed to develop a method to predict effective roof catchment areas. For the second stage, the effective roof catchment areas, together with roof area connection percentages, were analysed against different types of water demands in individual households. The individual systems were investigated for yield capacities, costs and water security using a modified Roof Runoff Harvesting Systems average annual yield model based on daily water balance procedures. The Life Cycle Costing analysis of the systems using the model was based on the Capital Recovery Method by taking into consideration the capital costs as well as ongoing costs for maintenance, replacement and operation of the systems. Anaplastic thyroid carcinoma (ATC) is almost universally fatal. Elevated keratin-8 (KRT8) protein expression is an established diagnostic cancer biomarker in several epithelial cancers (but not ATC). Several keratins, including KRT8, have been suggested to have a role in cell biology beyond that of structural cytoskeletal proteins. Here, we provide evidence that KRT8 plays a direct role in the growth of ATCs. Genomic and transcriptomic analysis of >5000 patients demonstrates that KRT8 mutation and copy number amplification are frequently evident in epithelial-derived cancers. Carcinomas arising from diverse tissues exhibit KRT8 mRNA and protein overexpression when compared to normal tissue levels. Similarly, in a panel of patient-derived ATC cell lines and patient tumors, KRT8 expression shows a similar pattern. sh-RNA-mediated KRT8 knockdown in these cell lines increases apoptosis, whereas forced overexpression of KRT8 confers resistance to apoptosis under peroxide-induced cell stress conditions. Short duration intense rainfall causes an increase in rainfall derived infiltration and inflow (RDII) into aging sewer networks, which leads to Sanitary Sewer Overflows (SSOs). This study presents a generalised framework for assessing and mitigating the impacts of intense rainfall on sanitary sewer networks. The first part of the proposed framework involves a detailed hydraulic modelling to evaluate the performance of the sewer network. The second part deals with the development of SSO mitigation strategies based on Water Sensitive Urban Design (WSUD) approaches. This paper also demonstrates the application of the first part of the proposed framework for a case study catchment in Melbourne, Australia. The hydraulic performance of the case study sewer network during a wet and a dry year is presented. The analysis found that for the wet year, 11 manholes had sewer overflows, whereas 53 of 57 manholes in the network of 3.2 km had surcharges. Large scale centralised water, wastewater and stormwater systems have been implemented for over 100 years. These systems have provided a safe drinking water supply, efficient collection and disposal of wastewater to protect human health, and the mitigation of urban flood risk. The sustainability of current urban water systems is under pressure from a range of challenges which include: rapid population growth and resulting urbanisation, climate change impacts, and infrastructure that is ageing and reaching capacity constraints. To address these issues, urban water services are now being implemented with Integrated Urban Water Management (IUWM) and Water Sensitive Urban Design (WSUD) approaches. WSUD systems can deliver multiple benefits including water conservation, stormwater quality improvement, flood control, landscape amenity and a healthy living environment. These systems can be provided as stand-alone systems or in combination with centralised systems. These systems are still novel and thus face knowledge gaps that are impeding their mainstream uptake. Knowledge gaps cover technical, economic, social, and institutional aspects of their implementation. This paper is based on the outcomes of a comprehensive study conducted in South Australia which investigated impediments for mainstream uptake of WSUD, community perceptions of WSUD and potential of WSUD to achieve water conservation through the application of alternative resources, and in flood management. The outcomes are discussed in this paper for the benefit of water professionals engaged with WSUD planning, implementation, community consultation and regulation. Although the paper is based on a study conducted in South Australia, the comprehensive framework developed to conduct this detailed study and investigation can be adopted in any part of the world. Urban water managers around the world are adopting decentralized water supply systems, often in combination with centralized systems. While increasing demand for water arising from population growth is one of the primary reasons for this increased adoption of alternative technologies, factors such as climate change, increased frequency of extreme weather events and rapid urbanization also contribute to an increased rate of adoption of these technologies. This combination of centralized-decentralized water systems approach is referred to as "hybrid water supply systems" and is based on the premise that the provision of alternative water sources at local scales can both extend the capacity of existing centralized water supply infrastructures, and improve resilience to variable climatic conditions. It is important to understand, however, that decentralized water production and reuse may change the flow and composition of wastewater and stormwater, thereby potentially also having negative impacts on its effectiveness and performance. This paper describes a framework to assess the interactions between decentralized water supply systems and existing centralized water servicing approaches using several analytical tools, including water balance modelling, contaminant balance modelling and multi-criteria decision analysis. The framework enables the evaluation of impacts due to change in quantity and quality of wastewater and stormwater on the existing centralized system arising from the implementation of hybrid water supply systems. The framework consists of two parts: (1) Physical system analysis for various potential scenarios and (2) Ranking of Scenarios. This paper presents a critical review of the physical impacts of decentralized water supply systems on existing centralized water infrastructures. This paper highlights the combination of centralized and decentralized systems, which is referred to as hybrid water supply systems. The system is hypothesized to generate more sustainable and resilient urban water systems. The basic concept is to use decentralized water supply options such as rainwater tanks, storm water harvesting and localized wastewater treatment and reuse in combination with centralized systems. Currently the impact of hybrid water supply technologies on the operational performance of the downstream infrastructure and existing treatment processes is yet to be known. The paper identifies a number of significant research gaps related to interactions between centralized and decentralized urban water services. It indicates that an improved understanding of the interaction between these systems is expected to provide a better integration of hybrid systems by improved sewerage and drainage design, as well as facilitate operation and maintenance planning. The tear film is a multi-layer fluid that covers the corneal and conjunctival epithelia of the eye and provides lubrication, nutrients, and protection from the outside environment. Tear fluid contains a high concentration of proteins and has thus been recognized as a potential source of biomarkers for ocular disorders due to its proximity to disease sites on the ocular surface and the non-invasive nature of its collection. This is particularly true in the case of dry eye disease, which directly impacts the tear film and its components. Proteomic analysis of tear fluid is challenging mainly due to the wide dynamic range of proteins and the small sample volumes. However, recent advancements in mass spectrometry have revolutionized the field of proteomics enabling unprecedented depth, speed, and accuracy, even with small sample volumes. In this study using the Orbitrap Fusion Tribrid mass spectrometer, we compared four different mass spectrometry workflows for the proteomic analysis of tear fluid collected via Schirmer strips. We were able to establish a method of in-strip protein digestion that identified >3000 proteins in human tear samples from 11 healthy subjects. Ionizing radiation results in extensive damage to biological systems. The massive amount of ionizing radiation from nuclear accidents, radiation therapy (RT), space exploration, and the nuclear battlefield leads to damage to biological systems. Radiation injuries, such as inflammation, fibrosis, and atrophy, are characterized by genomic instability, apoptosis, necrosis, and oncogenic transformation, mediated by the activation or inhibition of specific signaling pathways. Exposure of tumors or normal cells to different doses of ionizing radiation could lead to the generation of free radical species, which can release signal mediators and lead to harmful effects. Although previous FDA-approved agents effectively mitigate radiation-associated toxicities, their use is limited due to their high cellular toxicities. Preclinical and clinical findings reveal that phytochemicals derived from plants that exhibit potent antioxidant activities efficiently target several signaling pathways. Emerging infectious diseases (EID) are serious problems caused by fungi in humans and plant species. They are a severe threat to food security worldwide. In our current work, we have developed a support vector machine (SVM)-based model that attempts to design and predict therapeutic plant-derived antifungal peptides (PhytoAFP). The residue composition analysis shows the preference of C, G, K, R, and S amino acids. Position preference analysis shows that residues G, K, R, and A dominate the N-terminal. Similarly, residues N, S, C, and G prefer the C-terminal. Motif analysis reveals the presence of motifs like NYVF, NYVFP, YVFP, NYVFPA, and VFPA. We have developed two models using various input functions such as mono-, di-, and tripeptide composition, as well as binary, hybrid, and physiochemical properties, based on methods that are applied to the main data set. The TPC-based monopeptide composition model achieved more accuracy, 94.4%, with a Matthews correlation coefficient (MCC) of 0.89. Correspondingly, the second-best model based on dipeptides achieved an accuracy of 94.28% under the MCC 0.89 of the training dataset. [ABSTRACT FROM AUTHOR]

Purpose: The purpose of this study was to discover the aqueous humor proteomic changes associated with visual field indices in glaucoma patients. Methods: Aqueous humor samples were analyzed using Liquid Chromatography Tandem Mass Spectrometry (LC-MS/MS). The visual fields were analyzed with the Humphrey Visual Field analyzer. Statistical analyses were performed to discover the relationship between the aqueous humor proteins and visual field parameters including Pattern Standard Deviation (PSD), Visual Field Index (VFI), Mean Deviation (MD) and Glaucoma Hemifield Test (GHT). Results: In total, 222 proteins were identified in 49 aqueous humor samples. A total of 11, 9, 7, and 6 proteins were significantly correlated with PSD, VFI, MD, and GHT respectively. These proteins include apolipoprotein D, members of complement pathway (C1S, C4A, C4B, C8B, and CD14), and immunoglobulin family (IKHV3-9, IGKV2-28). Conclusion: Several proteins involved in immune responses (immunoglobulins and complement factors) and neurodegeneration (apolipoprotein D) were identified to be associated with abnormal visual field parameters. The present study examines the optimal water supply portfolio under the impact of climate change constructed by the authors previously. It incorporates feasibility planning for water supply projects, assesses a feasible water supply portfolio for central Taiwan, and uses the shadow price method to assess the rationality of the compensation policy for transferring agricultural water to ensure water supply security for the industrial sector. The study finds that Changhua and Yunlin have the highest per-unit costs of raw water, and the Nantou region has the highest carbon emission coefficient (carbon footprint) per unit of water produced. The cumulative value (2021–2031) of the water resources policy to reallocate agricultural water to achieve water supply security is about TWD 15.904–31.13 billion. The shadow price of industrial water is about TWD 40.18/cubic meter. Therefore, a compensation price for agricultural water transfer of less than TWD 40.18/cubic meter represents a rational policy. High water vapor flux at low brine temperatures without surface fouling is needed in membrane distillation-based desalination. Brine crossflow over surface-modified hydrophobic hollow fiber membranes (HFMs) yielded fouling-free operation with supersaturated solutions of scaling salts and their precipitates. Surface modification involved an ultrathin porous polyfluorosiloxane or polysiloxane coating deposited on the outside of porous polypropylene (PP) HFMs by plasma polymerization. The outside of hydrophilic MicroPES HFMs of polyethersulfone was also coated by an ultrathin coating of porous plasma-polymerized polyfluorosiloxane or polysiloxane rendering the surface hydrophobic. Direct contact membrane distillation-based desalination performances of these HFMs were determined and compared with porous PP-based HFMs. Salt concentrations of 1, 10, and 20 wt% were used. Leak rates were determined at low pressures. Surface and cross-sections of two kinds of coated HFMs were investigated by scanning electron microscopy. The HFMs based on water-wetted MicroPES substrate offered a very thin gas gap in the hydrophobic surface coating yielding a high flux of 26.4–27.6 kg/m2-h with 1 wt% feed brine at 70 °C. The fluxes of HFMs on porous PP substrates having a long vapor diffusion path were significantly lower. Coated HFM performances have been compared with flat hydrophilic membranes of polyvinylidene fluoride having a similar plasma-polymerized hydrophobic polyfluorosiloxane coating. Women are at a significantly higher risk of developing osteoarthritis (OA) compared to males. The pathogenesis of osteoarthritis (OA) in women is poorly understood. Extracellular vesicles (EVs) have been shown to play an essential role in numerous signaling processes during the pathogenesis of age-related diseases via paracrine signaling. Molecular profiling of the synovial fluid-derived EVs cargo in women may help in the discovery of novel biomarkers and therapeutics for the treatment of OA in women. Previously, we reported that synovial fluid-derived EV miRNA cargo differs in a sex-specific manner. This study aims to characterize synovial fluid-derived EV protein cargo in OA patients. Our data showed sex-specific EVs protein content in OA. We found haptoglobin, orosomucoid, and ceruloplasmin significantly up-regulated, whereas apolipoprotein down-regulated in female OA EVs. In males, we discovered β-2-glycoprotein, and complement component 5 proteins significantly up-regulated and Spt-Ada-Gcn5 acetyltransferase (SAGA)-associated factor 29 down-regulated in male OA EVs. Database for Annotation, Visualization, and Integrated Discovery (DAVID) and QuickGO analysis revealed OA-specific protein involvement in several biological, molecular, and cellular pathways, specifically in inflammatory processes. Rainwater tanks are increasingly being implemented as part of the integrated urban water management paradigm where all sources of water, including potable, stormwater and recycled, are considered eligible to contribute to the urban water supply. Over the last decade or so, there has been a rapid uptake of rainwater tank systems in urban areas, especially in Australian cities, encouraged through financial incentives, but more importantly, from change in residential building codes effectively mandating the installation of rainwater tanks. Homes with rainwater tanks in Australian cities have increased from 15% to 28% over six years to 2013. These building codes specify certain rainwater tank specifications to achieve a stated rainwater use, and hence potable water savings. These specifications include minimum rainwater tank size, minimum connected roof area, plumbing for internal supply for toilets and washing machines, and external supply for garden watering. These expected potable water savings from households are often factored into regional strategic water planning objectives. Hence if rainwater tanks do not deliver the expected saving due to sub-standard installation and/or poor maintenance, it will have an adverse impact on the regional water plan in the longer term. In this paper, a methodology to assess the effectiveness of a government rainwater tank policy in achieving predicted potable water savings is described and illustrated with a case study from South East Queensland, Australia. It is anticipated that water professionals across the globe should be able to use the same methodology to assess the effectiveness of similar rainwater policies, or indeed any other distributed water saving policy, in their local planning communities. Diabetic retinopathy (DR), a sight-threatening neurovasculopathy, is the leading cause of irreversible blindness in the developed world. DR arises as the result of prolonged hyperglycemia and is characterized by leaky retinal vasculature, retinal ischemia, retinal inflammation, angiogenesis, and neovascularization. The number of DR patients is growing with an increase in the elderly population, and therapeutic approaches are limited, therefore, new therapies to prevent retinal injury and enhance repair are a critical unmet need. Besides vascular endothelial growth factor (VEGF)-induced vascular proliferation, several other mechanisms are important in the pathogenesis of diabetic retinopathy, including vascular inflammation. Thus, combining anti-VEGF therapy with other new therapies targeting these pathophysiological pathways of DR may further optimize treatment outcomes. Technological advancements have allowed for high-throughput proteomic studies examining biofluids such as aqueous humor, vitreous humor, tear, and serum. Many DR biomarkers have been identified, especially proteins involved in retinal inflammatory processes. There are pressures on existing centralized water infrastructures in urban centers which justify the search for alternatives. An increasingly important alternative is to shift from centralized to hybrid systems, often in response to climate variability and demographic changes. In a hybrid system, water is supplied and discharged through a mix of centralized and decentralized systems. There is usually no single objective that justifies the choice of hybrid water systems, but they typically are justified based on the consideration of a number of different criteria in order to evaluate the overall quality of service provision. The most important criteria include meeting water demand, as well as reducing demand for fresh water and instead using local alternative water supplies. Integration of multiple objectives to evaluate the hybrid water supply systems can be accomplished by multi-criteria decision aid techniques. This paper evaluates a number of hybrid water supply scenarios using a case study based on the Northern Growth Area of Melbourne, Australia. It uses the Preference Ranking Organization METHod for Enrichment Evaluations (PROMETHEE) and Geometrical Analysis for Interactive Decision Aid (GAIA), one of the multi-criteria decision-making methods through D-Sight software, to rank the hybrid water supply scenarios, and this ranking is validated by means of sensitivity analysis. The centralized system combined with stormwater harvesting and the centralized system combined with treated wastewater and rainwater tanks yielded the first and second most preferred scenarios, while the centralized water supply system combined with treated wastewater yielded the worst hybrid water supply option. The molecular basis of prostate cancer (PCa) progression from the primary disease to metastatic castration-resistant prostate cancer (CRPC) followed by therapy-induced neuroendocrine prostate cancer is not fully understood. In this study, we elucidate the role of miR-410, a little-studied microRNA located on chromosome 14q32.31 within the DLK1-DIO3 cluster, in PCa. miR-410 expression analyses in primary and metastatic PCa tissues and cell lines show that its levels are decreased in initial stages and increased in advanced PCa. Functional studies were performed in a series of PCa cell lines. In LNCaP cells, miR-410 overexpression led to decreases in cellular viability, proliferation, invasiveness, and migration. On the other hand, miR-410 overexpression in PC3 and C42B cells led to increased viability, proliferation, and invasiveness. Our data suggest that miR-410 represses epithelial-to-mesenchymal transition (EMT) in LNCaP cells by directly repressing SNAIL. However, it promotes EMT and upregulates PI3K/Akt signaling in PC3 and C42B cells. In vivo studies with PC3 xenografts support an oncogenic role of miR-410. These data suggest that miR-410 acts as a tumor suppressor in the initial stages of PCa and play an oncogenic role in advanced PCa. Our findings have important implications in understanding the molecular basis of PCa progression with potential translational implications.
