The LABEXTRACT plant extract bank, featuring diverse members of the Myrtaceae family from Brazilian hot spot regions, provides a promising avenue for bioprospection. Given the pivotal roles of the Spike protein and 3CLpro and PLpro proteases in SARS-CoV-2 infection, this study delves into the correlations between the Myrtaceae species from the Atlantic Forest and these targets, as well as an antiviral activity through both in vitro and in silico analyses. The results uncovered notable inhibitory effects, with Eugenia prasina and E. mosenii standing out, while E. mosenii proved to be multitarget, presenting inhibition values above 72% in the three targets analyzed. All extracts inhibited viral replication in Calu-3 cells (EC50 was lower than 8.3 µg·mL−1). Chemometric analyses, through LC-MS/MS, encompassing prediction models and molecular networking, identified potential active compounds, such as myrtucommulones, described in the literature for their antiviral activity. Docking analyses showed that one undescribed myrtucommulone (m/z 841 [M − H]−) had a higher fitness score when interacting with the targets of this study, including ACE2, Spike, PLpro and 3CLpro of SARS-CoV-2. Also, the study concludes that Myrtaceae extracts, particularly from E. mosenii and E. prasina, exhibit promising inhibitory effects against crucial stages in SARS-CoV-2 infection. The implementation of a safety culture and awareness of emergency issues in buildings has been growing in more developed societies. It is essential that all occupants know how to act in an emergency situation, particularly during an emergency evacuation. In higher education institutions (HEIs), which annually host not only their many employees, but also national and international students, it is essential to know and understand the knowledge, attitudes, and behaviors that the academic community demonstrates in this matter. This study aimed to characterize the perception of occupants regarding safety, specifically in the emergency evacuation phase, within the academic community of an HEI. In this observational cross-sectional study, data on general knowledge and attitudes regarding safety and actions during emergency situations were collected through an anonymous questionnaire targeting students, faculty, and non-teaching staff, which was sent via institutional email. Valid responses were received from 392 participants and then scored and assessed on different domains. The results obtained showed that, despite a reasonable average regarding the general level of knowledge, attitudes, and behaviors of the occupants on the subject, the community falls somewhat short in terms of training. It is noteworthy that approximately 64% of the sample has never received awareness or training related to emergency evacuation, and around 68% are unaware of the location of their institution's meeting/gathering point. Finally, by identifying the most common gaps, namely the training dimension, some simple measures could be improved, such as the dissemination of safety instructions accessible through QR codes placed in strategic locations or even conducting small drills during class sessions, as well as learning with simulation concerning different scenarios of emergency. As part of a Ministry of Science, Technology, and Innovation initiative, the Brazilian Societies of Botany, Microbiology, Virology, and Zoology conducted a comprehensive evaluation of biological collections in Brazil. This assessment aimed to gather insights into the current state of these collections, with the goal of providing support for future public policies, including financial subsidies and prioritization policies. In this context, we present the findings related to microbiological collections, essential to ex situ biodiversity conservation and crucial in supporting research, development, and innovation. A survey was distributed to public and private institutions across Brazil, yielding responses from 168 microbiological collections representing 79 different entities. Notably, 73 of these collections are affiliated with public research institutions and universities, underscoring the State's pivotal role in preserving and safeguarding Brazilian microbial diversity. The primary taxonomic groups encompass bacteria (found in 70.24% of collections) and fungi (comprising 52.98% of collections), sourced from diverse Brazilian ecosystems and biomes, including those that contain several type strains. Furthermore, the collections preserve microorganisms harboring biotechnological potential applicable to environmental protection, public health, industry, and agribusiness. Despite these promising economic and biotechnological prospects, our meticulous data analysis has revealed significant limitations and vulnerabilities, especially regarding physical infrastructure and human resources, emphasizing the urgent need for interventions to guarantee their sustainability. The global food system is currently facing significant challenges that make it unsustainable and environmentally harmful. These challenges not only threaten food security but also have severe negative impacts on the environment. Efforts have been made to reform agrifood systems and align them with the built environment, but emerging obstacles have revealed the weaknesses in these systems, particularly in less self-sufficient countries. This review outlines the primary environmental problems associated with global agrifood systems and the challenges in promoting food security. It emphasizes that the increasing global population and urbanization need rational and equitable changes in food systems, including production, distribution, storage, and consumption. These changes should aim to minimize environmental impacts by protecting and efficiently utilizing natural resources such as air, water, soil, and biodiversity, reducing food loss and waste, and mitigating pollution that contributes to ecosystem degradation and climate change. In this context, hydroponics emerges as a sustainable, plant-based food production technique that can be employed as a solution in urban areas. It can be implemented in domestic microproduction systems, serving as a complementary alternative to conventional food production methods. This study also provides insights into the challenges that need to be addressed in order to enhance home hydroponic systems. The integration of hydroponics into urban food production offers the potential to tackle both food security and environmental sustainability issues, providing a path toward more resilient and efficient food systems. The main objective of this review was to determine the main risks that agricultural workers are exposed to during pesticide application, which may have a harmful effect on their health and on public health. This systematic review was based on the PRISMA guidelines. A search for articles was conducted in the Medline/PubMed, ScienceDirect, and Web of Science databases. Fifteen articles were selected considering their assessment of agricultural workers' knowledge, perceptions, attitudes, practices, and behaviours, identifying the main risks and risk factors for disease associated with the unsafe handling of pesticides. The main risk factors identified were age, education, pesticide safety training, farming experience, and contact with other farmers/intermediaries resulting in pesticide access. The most frequent risk behaviour was an application of pesticides without personal protective equipment (PPE), incorrect disposal of empty packaging and waste, and undervaluation of label information, as well as other unsafe practices. Multidisciplinary and more effective training must be delivered to enhance pesticide-safe usage. This will empower workers to adopt more conscious and safer behaviours while using pesticides. Variations in the rates of involuntary admission (IA) reflect the influence of unexplained contextual variables that are typically too heterogeneous to be included in systematic reviews. This paper attempts to gather and analyze factors unrelated to the patients that have been linked to IA. The articles included in this review were selected by iteratively searching four electronic databases (PubMed, PsychINFO, EMBASE, and Web of Science). A total of 54 studies from 19 different countries and regions, including 14 European countries, the United States, Canada, China, Vietnam, and Taiwan, were selected. The factors were categorized as service-related factors, impactful events, seasonal and temporal factors, mental health legislation, staff factors, and public attitudes. The factors rarely act in isolation but rather interact and reinforce each other, causing a greater influence on IA. This paper explains how these factors present opportunities for robust and sustainable interventions to reduce IAs. The paper also identifies future directions for research, such as examining the effects of economic recessions. Enhancing global reporting standards is essential to validate future research and support further in-depth studies. COVID-19 has infected humans worldwide, causing millions of deaths or prolonged symptoms in survivors. The transient or persistent symptoms after SARS-CoV-2 infection have been defined as post-COVID-19 conditions (PCC). We conducted a study of 151 Brazilian PCC patients to analyze symptoms and immunoglobulin profiles, taking into account sex, vaccination, hospitalization, and age. Fatigue and myalgia were the most common symptoms, and lack of vaccination, hospitalization, and neuropsychiatric and metabolic comorbidities were relevant to the development of PCC. Analysis of serological immunoglobulins showed that IgA was higher in PCC patients, especially in the adult and elderly groups. Also, non-hospitalized and hospitalized PCC patients produced high and similar levels of IgA. Our results indicated that the detection of IgA antibodies against SARS-CoV-2 during the course of the disease could be associated with the development of PCC and may be an immunological signature to predict prolonged symptoms in COVID-19 patients. 5-nitrouracil is a polarizable molecule with a permanent electric dipole moment. Its molecular properties caught the attention of physicists working on nonlinear optics or optoelectronics, but the translation of the molecular assets to the crystalline solid has not been straightforward. This review compares all the known crystal structures incorporating the neutral or ionic 5-nitrouracil, or the two species concomitantly, discussing the effect of the packing in the optimization of the crystalline optical properties. Two new centrosymmetric 5-nitrouracilate salts are also reported for the first time, showing extensive hydrogen bonding between anions and cations. This review also gathers data from nonlinear optical measurements of non-centrosymmetric crystals and thermal stabilities of known polymorphs, showing that a neutral 5-nitrouracil molecule in acentric crystalline environment allows efficient blue-light generation. Alkaline cements have been extensively tested for soil stabilisation in the last decade. However, only a few studies have focused on the assessment of such performance by establishing the constitutive behaviour of the cement. In this paper, we focus on the mechanical behaviour, using triaxial testing of a clay with high water content stabilised with an alkali-activated binder and the subsequent prediction of the experimental stress–strain response using a kinematic hardening constitutive model initially developed for natural clays. Monotonic consolidated undrained triaxial tests were conducted on reconstituted and stabilised clay specimens cured for 28 days to evaluate the effects of cementation on the overall shear behaviour. Alkali-activated binder was synthetised from blast furnace slag and sodium hydroxide. Scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS), and X-ray diffraction (XRD) were performed to study the microstructure, whereas leachate analyses were performed after 28 and 90 days of curing to investigate the contamination potential. The main product formed was calcium aluminosilicate hydrate (C-A-S-H) with a low CaO/SiO2 ratio, and no risk of soil contamination was found. The compressibility and undrained shear strength in the pre-yield state was found to be independent of the initial mean effective stress (p'0), unlike what was observed in the post-yield state, where the shear strength seemed to be affected by p'0. The model provided reliable predictions of the experimental results and captured the main features of the artificially cemented clay for the tested p'0 range. The SARS-CoV-2 virus infection led to millions of deaths during the COVID-19 pandemic. Hundreds of workers from several other Brazilian cities, as well as from other countries, arrive daily in Macaé to work in the oil supply chain, making this city a putative hotspot for the introduction of new viral lineages. In this study, we performed a genomic survey of SARS-CoV-2 samples from Macaé during the first outbreak of COVID-19, combined with clinical data and a molecular integrative analysis. First, phylogenomic analyses showed a high occurrence of viral introduction events and the establishment of local transmissions in Macaé, including the ingression and spread of the B.1.1.28 lineage in the municipality from June to August 2020. Second, SARS-CoV-2 mutations were identified in patients with distinct levels of COVID-19 severity. Third, molecular interactions of the mutated spike protein from three B.1.1.33 local samples and human ACE2 showed higher interactions than that of the wild-type spike protein from the ancestral virus. Altogether, these results elucidate the SARS-CoV-2 genomic profile in a strategic Brazilian city and further explore the functional aspects of SARS-CoV-2 with a characterization of emerging viral mutations associated with clinical data and the potential targets for drug development against SARS-CoV-2. Water scarcity is increasing in the Mediterranean and alternative sources of water are needed to meet food production needs, protect the environment and reduce the effects of climate change. Currently, many urban wastewater treatment plants (WWTP) produce high volumes of treated effluents which can be an alternative source of water for agriculture irrigation, since they fulfill the quality requirements for crops and the environment. This work analyzed the quantity and quality of a treated effluent produced by an urban WWTP in Algarve, and the environmental benefits of its use on the irrigation of a citrus orchard, as an alternative to groundwater. Carbon dioxide emissions related to orange production were quantified and the orchard's potential to sequester CO2 was estimated. The reuse of this urban wastewater is revealed to be technologically feasible and environmentally advantageous, avoiding the overexploitation of the local aquifer and preventing the eutrophication of aquatic ecosystems, contributing to the improvement of soil characteristics and decreasing the carbon emissions in orange production. Furthermore, it was found that during the five-month experimental period, the citrus orchard sequestered 87.5% of the CO2e emitted by WWTP in the effluent treatment, converting 72,623 kg of sequestered CO2 into orange biomass. Faba bean have been grown in Portugal for a long time and locally adapted populations are still maintained on farm. The genetic diversity of four Portuguese faba bean populations that are still cultivated in some regions of the country was evaluated using the Inter Retrotransposons Amplified Polymorphism (IRAP) technique. It was shown that molecular markers based on retrotransposons previously identified in other species can be efficiently used in the genetic variability assessment of Vicia faba. The IRAP experiment targeting Athila yielded the most informative banding patterns. Cluster analysis using the neighbor-joining algorithm generated a dendrogram that clearly shows the distribution pattern of V. faba samples. The four equina accessions are separated from each other and form two distinct clades while the two major faba bean accessions are not unequivocally separated by the IRAP. Fluorescent In Situ Hybridization (FISH) analysis of sequences amplified by IRAP Athila revealed a wide distribution throughout V. faba chromosomes, confirming the whole-genome coverage of this molecular marker. Morphological characteristics were also assessed through cluster analysis of seed characters using the unweighted pair group method arithmetic average (UPGMA) and principal component analysis (PCA), showing a clear discrimination between faba bean major and equina groups. It was also found that the seed character most relevant to distinguish accessions was 100 seed weight. Seed morphological traits and IRAP evaluation give similar results supporting the potential of IRAP analysis for genetic diversity studies. [ABSTRACT FROM AUTHOR]

Genomic diversity of Portuguese accessions of Avena species—diploid A. strigosa and hexaploids A. sativa and A. sterilis—was evaluated through molecular and cytological analysis of 45S rDNA, and other repetitive sequences previously studied in cereal species—rye subtelomeric sequence (pSc200) and cereal centromeric sequence (CCS1). Additionally, retrotransposons and microsatellites targeting methodologies—IRAP (inter-retrotransposon amplified polymorphism) and REMAP (retrotransposon-microsatellite amplified polymorphism)—were performed. A very high homology was detected for ribosomal internal transcribed sequences (ITS1 and ITS2) between the species analyzed, although nucleolar organizing regions (NOR) fluorescent in situ hybridization (FISH) analysis revealed distinct number of Nor loci between diploid and hexaploid species. Moreover, morphological diversity, evidenced by FISH signals with different sizes, was observed between distinct accessions within each species. pSc200 sequences were for the first time isolated from Avena species but proven to be highly similar in all genotypes analyzed. The use of primers designed for CCS1 unraveled a sequence homologous to the Ty3/gypsy retrotransposon Cereba, that was mapped to centromeric regions of diploid and hexaploid species, being however restricted to the more related A and D haplomes. Retrotransposon-based methodologies disclosed species- and accessions-specific bands essential for the accurate discrimination of all genotypes studied. Centromeric, IRAP and REMAP profiles therefore allowed accurate assessment of inter and intraspecific variability, demonstrating the potential of these molecular markers on future oat breeding programs. The union of different genomes in the same nucleus frequently results in hybrid genotypes with improved genome plasticity related to both genome remodeling events and changes in gene expression. Most modern cereal crops are polyploid species. Triticale, synthesized by the cross between wheat and rye, constitutes an excellent model to study polyploidization functional implications. We intend to attain a deeper knowledge of dispersed repetitive sequence involvement in parental genome reshuffle in triticale and in wheat-rye addition lines that have the entire wheat genome plus each rye chromosome pair. Through Random Amplified Polymorphic DNA (RAPD) analysis with OPH20 10-mer primer we unraveled clear alterations corresponding to the loss of specific bands from both parental genomes. Moreover, the sequential nature of those events was revealed by the increased absence of rye-origin bands in wheat-rye addition lines in comparison with triticale. Remodeled band sequencing revealed that both repetitive and coding genome domains are affected in wheat-rye hybrid genotypes. Additionally, the amplification and sequencing of pSc20H internal segments showed that the disappearance of parental bands may result from restricted sequence alterations and unraveled the involvement of wheat/rye related repetitive sequences in genome adjustment needed for hybrid plant stabilization. In this work, co-crystal screening was carried out for two important dihydrofolate reductase (DHFR) inhibitors, trimethoprim (TMP) and pyrimethamine (PMA), and for 2,4-diaminopyrimidine (DAP), which is the pharmacophore of these active pharmaceutical ingredients (API). The isomeric pyridinecarboxamides and two xanthines, theophylline (THEO) and caffeine (CAF), were used as co-formers in the same experimental conditions, in order to evaluate the potential for the pharmacophore to be used as a guide in the screening process. In silico co-crystal screening was carried out using BIOVIA COSMOquick and experimental screening was performed by mechanochemistry and supported by (solid + liquid) binary phase diagrams, infrared spectroscopy (FTIR) and X-ray powder diffraction (XRPD). The in silico prediction of low propensities for DAP, TMP and PMA to co-crystallize with pyridinecarboxamides was confirmed: a successful outcome was only observed for DAP + nicotinamide. Successful synthesis of multicomponent solid forms was achieved for all three target molecules with theophylline, with DAP co-crystals revealing a greater variety of stoichiometries. The crystalline structures of a (1:2) TMP:THEO co-crystal and of a (1:2:1) DAP:THEO:ethyl acetate solvate were solved. This work demonstrated the possible use of the pharmacophore of DHFR inhibitors as a guide for co-crystal screening, recognizing some similar trends in the outcome of association in the solid state and in the molecular aggregation in the co-crystals, characterized by the same supramolecular synthons. The influence of suction on the mechanical behaviour of unsaturated chemically stabilised soils is still mostly unknown and unquantified. This is also motivated by the difficulties associated with the experimental procedure required to fully characterise the unsaturated response of the soil, including its direct influence on traditional strength tests. The present paper presents the soil water retention curves obtained for a Portuguese soil before and after being stabilised with Portland cement (OPC) and an alkali-activated cement (AAC). Saturated undrained triaxial tests were also performed for the same curing conditions (0, 28, and 90 days). Previous attempts to characterise the retention curve of soils stabilised with AAC are unknown, and the results showed that the pore volume structure is already formed after 28 days, prior to the full development of the gel matrix responsible for the strength increase between 28 and 90 days. The curve changed after stabilisation, and with each binder, as the OPC presented a higher air-entry value and a narrower suction range compared to the AAC solution. The significant differences between the curves obtained from each binder suggest the future development of a methodology to assess the quality of the AAC stabilisation. [ABSTRACT FROM AUTHOR]

Wheat is one of the most important cereals for food and feed, and it is, therefore, necessary to determine the effects of short-term high temperature events (heatwaves) during grain filling. These heatwave events are increasingly common, especially in Portugal. In this work, seven commercial varieties recommended for production in Portugal were submitted to one-week high temperature (HT) treatment ten days after anthesis to evaluate heat effects on grain yield and quality. Grain yield parameters, such as grain number and weight, were evaluated as well as grain composition through attenuated total reflection Fourier transform infrared (ATR-FTIR) spectroscopy. Variation in HT response between varieties was detected. Grain number and weight tended to decrease in most varieties analyzed. However, two varieties proved to be more resilient since grain number and weight remain unaltered in the Bancal variety, which is the one with better yield results, and even increased in the Pata Negra variety. Regarding grain composition, the comparison between ATR-FTIR spectra of milled grains from control and HT plants revealed alterations in peaks assigned to polysaccharides and proteins. Additionally, a model was built based on nitrogen elemental analysis to predict protein content in flour samples through spectral data that corroborated the differences identified by spectra profile comparison. Moreover, both analyses showed that the intervarietal diversity observed in control conditions was significantly reduced in HT treated plants. Wheat is undoubtedly one of the most important crops worldwide and it is essential to study how the distinct varieties answer to heat waves associated with climatic changes, in order to design adequate wheat breeding strategies. To assess high temperature (HT) impact in wheat grain characteristics, seven commercial varieties, which have been recommended for production in Portugal, were submitted for one-week HT treatment ten days after anthesis. Firstly, predicted grain technological quality was determined by giving high scores for all varieties studied, based on the allelic compositions of genes encoding high molecular weight glutenins, granule-bound starch synthase and puroindolines. The effects of HT on transcription levels of those genes were, for the first time, evaluated in distinct wheat genotypes, in comparison with control plants. Finally, protein fraction content in mature grains were also estimated in untreated and treated plants. Immature grains from plants, maintained in control conditions, showed significant intervarietal differences in transcription levels of genes associated with grain quality traits, a variability that was significantly reduced in grains from HT treated plants. On the other hand, the influence of HT in mature grain protein-fractions and in gliadin/glutenin ratios revealed intervarietal diversity, even with opposite effects in some varieties. The present study, therefore, discloses marked variability in parameters associated with flour quality between the wheat varieties analyzed, which are differentially affected by HT treatments, similar to heat waves frequently observed in climate change scenarios.
