Carotenoids are high added-value products primarily known for their intense coloration and high antioxidant activity. They can be extracted from a variety of natural sources, such as plants, animals, microalgae, yeasts, and bacteria. Gordonia alkanivorans strain 1B is a bacterium recognized as a hyper-pigment producer. However, due to its adaptations to its natural habitat, hydrocarbon-contaminated soils, strain 1B is resistant to different organic solvents, making carotenoid extraction through conventional methods more laborious and inefficient. Ionic liquids (ILs) have been abundantly shown to increase carotenoid extraction in plants, microalgae, and yeast; however, there is limited information regarding bacterial carotenoid extraction, especially for the Gordonia genus. Therefore, the main goal of this study was to evaluate the potential of ILs to mediate bacterial carotenoid extraction and develop a method to achieve higher yields with fewer pre-processing steps. In this context, an initial screening was performed with biomass of strain 1B and nineteen different ILs in various conditions, revealing that tributyl(ethyl)phosphonium diethyl phosphate (IL#18), combined with ethyl acetate (EAc) as a co-solvent, presented the highest level of carotenoid extraction. Afterward, to better understand the process and optimize the extraction results, two experimental designs were performed, varying the amounts of IL#18 and EAc used. These allowed the establishment of 50 µL of IL#18 with 1125 µL of EAc, for 400 µL of biomass (cell suspension with about 36 g/L), as the ideal conditions to achieve maximal carotenoid extraction. Compared to the conventional extraction method using DMSO, this novel procedure eliminates the need for biomass drying, reduces extraction temperatures from 50 °C to 22 ± 2 °C, and increases carotenoid extraction by 264%, allowing a near-complete recovery of carotenoids contained in the biomass. These results highlight the great potential of ILs for bacterial carotenoid extraction, increasing the process efficiency, while potentially reducing energy consumption, related costs, and emissions. (1) Background: We examined the effect of the acute administration of olive oil (EVOO), linseed oil (GLO), soybean oil (SO), and palm oil (PO) on gastric motility and appetite in rats. (2) Methods: We assessed food intake, gastric retention (GR), and gene expression in all groups. (3) Results: Both EVOO and GLO were found to enhance the rate of stomach retention, leading to a decrease in hunger. On the other hand, the reduction in food intake caused by SO was accompanied by delayed effects on stomach retention. PO caused an alteration in the mRNA expression of NPY, POMC, and CART. Although PO increased stomach retention after 180 min, it did not affect food intake. It was subsequently verified that the absence of an autonomic reaction did not nullify the influence of EVOO in reducing food consumption. Moreover, in the absence of parasympathetic responses, animals that received PO exhibited a significant decrease in food consumption, probably mediated by lower NPY expression. (4) Conclusions: This study discovered that different oils induce various effects on parameters related to food consumption. Specifically, EVOO reduces food consumption primarily through its impact on the gastrointestinal tract, making it a recommended adjunct for weight loss. Conversely, the intake of PO limits food consumption in the absence of an autonomic reaction, but it is not advised due to its contribution to the development of cardiometabolic disorders. Due to the high abrasiveness and anisotropic nature of composites, along with the need to machine different materials at the same time, drilling multi-materials is a difficult task, and usually results in material damage, such as uncut fibres and delamination, hindering hole functionality and reliability. Image processing and analysis algorithms can be developed to effectively assess such damage, allowing for the calculation of delamination factors essential to the quality control of hole inspection in composite materials. In this study, a digital image processing and analysis algorithm was developed in Python to perform the delamination evaluation of drilled holes on a carbon fibre reinforced polymer (CFRP) and aluminium (Al) multi-material. This algorithm was designed to overcome several limitations often found in other algorithms developed with similar purposes, which frequently lead to user mistakes and incorrect results. The new algorithm is easy to use and, without requiring manual pre-editing of the input images, is fully automatic, provides more complete and reliable results (such as the delamination factor), and is a free-of-charge software. For example, the delamination factors of two drilled holes were calculated using the new algorithm and one previously developed in Matlab. Using the previous Matlab algorithm, the delamination factors of the two holes were 1.380 and 2.563, respectively, and using the new Python algorithm, the results were equal to 3.957 and 3.383, respectively. In several industrial fields, environmental and operational data are acquired with numerous purposes, potentially generating a huge quantity of data containing valuable information for management actions. This work proposes a methodology for clustering time series based on the K-medoids algorithm using a convex combination of different time series correlation metrics, the COMB distance. The multidimensional scaling procedure is used to enhance the visualization of the clustering results, and a matrix plot display is proposed as an efficient visualization tool to interpret the COMB distance components. This is a general-purpose methodology that is intended to ease time series interpretation; however, due to the relevance of the field, this study explores the clustering of time series judiciously collected from data of a wind farm located on a complex terrain. Using the COMB distance for wind speed time bands, clustering exposes operational similarities and dissimilarities among neighboring turbines which are influenced by the turbines' relative positions and terrain features and regarding the direction of oncoming wind. In a significant number of cases, clustering does not coincide with the natural geographic grouping of the turbines. A significant and increasing number of studies have been dedicated to complex concentrated alloys (CCAs) due to the improved properties that these metallic materials can exhibit. However, while most of these studies employ melting techniques, only a few explore powder metallurgy and pressureless sintering as production methods. In this work, a microstructural characterization of AlCrCuFeMnNi CCA samples obtained by powder metallurgy and pressureless sintering using mixtures of powders with different compositions was carried out. One batch of samples (B1) was prepared using commercial powders of Al, Cr, Cu, Fe, Mn, and Ni. Another batch (B2) used mixtures of CrFeMn, AlNi, and Cu powders. A third set of samples (B3) was obtained by adding 1% at. of Mg to the B2 powder. The samples were characterized by X-ray diffraction, scanning and transmission electron microscopy, energy dispersive spectroscopy, density measurements, and hardness tests. Thermodynamic calculations were also used to complement the microstructural characterization. All the obtained samples exhibited high relative density and hardness values. However, B3 samples showed a higher hardness, attributed to the finer distribution of oxide particles, which was promoted by the presence of Mg during sintering. These last samples presented a hardness/density ratio of 62 HV/(g cm−3), surpassing that of some martensitic stainless steels and nickel–titanium alloys. [ABSTRACT FROM AUTHOR]

Hospital accreditation has become ubiquitous in developing countries. While research acknowledges that accreditation can enhance healthcare quality, efficiency, and safety, concerns persist regarding hospitals' management of conflicts stemming from the diverse institutional logic inherent in this process. Therefore, this study aimed to investigate how professional and market logic, alongside conflicts arising from institutional demands, affect compliance with hospital accreditation. To this end, we conducted a multiple-case study in four Brazilian hospitals employing in-depth interviews and on-site observations. The triangulation of narrative analysis and the outcomes of multiple correspondence analysis revealed that when professional logic predominates, there is a greater propensity to tailor accredited activities by segmenting the tasks between physicians and nurses with the intention of mitigating existing conflicts. Conversely, when conflicts occur over established goals between professionals and orientated marked logic executives, the accreditation process is impeded, resulting in non-compliance. Ultimately, the findings underscore the alignment between the pursuit of legitimacy and efficiency within the accreditation process. Simple Summary: Because the spineless cactus has a low dry matter, neutral detergent fiber and crude protein content, its use alone is not recommended, and it should be combined with other bulky food sources, such as gliricidia, pornunça and buffel grass, as they are found in abundance in semi-arid regions and nutritionally complement the cactus. Thus, a homogeneous mixture of spineless cactus-based silage in combination with these tropical forages can reduce the selection of components by animals, benefiting their performance and reducing costs compared to their conventional diet, given the more efficient intake. This study investigated the chemical composition, fractionation of carbohydrates and proteins, ruminal degradation kinetics and in vitro gas production of silages composed of cactus and tropical forage plants and their potential use as exclusive feed for sheep. Mixed spineless cactus silages with tropical forages have positive effects on ruminal degradation, digestibility and gas production. Mixed spineless cactus silage with tropical forages can be used as exclusive feed for sheep and can replace corn silage in diet composition. The aim was to evaluate the chemical composition, carbohydrates, protein fractionation and in vitro gas production of silages composed of spineless cactus and tropical forages and their effect on sheep performance. Treatments consisted of silages: corn silage (CS), spineless cactus silage (SCS), spineless cactus + gliricidia (SCG), spineless cactus + buffel grass silage (SCBG) and spineless cactus + pornunça (SCP). Silos were opened 60 days after ensiling, and analyses were carried out. The digestibility test lasted for 36 days, with eight animals per treatment. A completely randomized design was adopted. Considering carbohydrate fractionation, CS, SCS and SCBG silages had higher total carbohydrate content (p = 0.001). The SCS silage presented a higher A + B1 fraction (p = 0.001). The SCBG and SCG silages showed a higher B2 fraction (p < 0.0001) compared to the CS and SCS silages. The SCBG and SCP silages presented a higher C fraction (p = 0.001). For protein fractionation, the SCP and SCG silages showed higher crude protein contents (p = 0.001). The CS and SCS silages showed a higher A fraction (p = 0.001). The SCBG silage presented a higher B1 + B2 fraction (p = 0.001). The SCG silage showed a higher B3 fraction (p = 0.006) compared to SCBG silage. The SCS and SCP silages showed a higher C fraction (p = 0.001). Exclusive SCS silage showed higher in vitro dry matter digestibility (p = 0.001), dry matter degradability (p = 0.001) and total gas production (p = 0.001). The use of the SCBG, SCP and SCG silages to feed sheep increased the dry matter intake (p < 0.001). Sheep fed the SCG silage showed greater dry matter and crude protein digestibility compared to the sheep fed the CS, SCS and SCP silages (p = 0.002). There was a higher water intake (p < 0.001) with the use of the SCS and SCG silages to feed the sheep. The SCP and SCG silages provided a greater intake (p < 0.001) and excretion (p < 0.001) of nitrogen by the animals. Although there were no differences between the treatments for daily gains, lambs that received the spineless cactus-based silage associated with tropical forages showed higher gains (160–190 g/day) than lambs that received CS silage (130 g/day). Thus, the use of spineless cactus associated with buffelgrass, pornunça and gliricidia to prepare mixed silages (60:40) to feed sheep has potential use to feed sheep, with positive effects on nutrient degradation and increases in dry matter intake. Under experimental conditions, we recommend the exclusive use of spineless cactus silage associated with buffel grass, pornunça and gliricidia in feeding sheep in semi-arid regions, as it provides nutrients, water and greater daily gains compared to corn silage. Chondrosia reniformis is a collagen-rich marine sponge that is considered a sustainable and viable option for producing an alternative to mammalian-origin collagens. However, there is a lack of knowledge regarding the properties of collagen isolated from different sponge parts, namely the outer region, or cortex, (ectosome) and the inner region (choanosome), and how it affects the development of biomaterials. In this study, a brief histological analysis focusing on C. reniformis collagen spatial distribution and a comprehensive comparative analysis between collagen isolated from ectosome and choanosome are presented. The isolated collagen characterization was based on isolation yield, Fourier-transformed infrared spectroscopy (FTIR), circular dichroism (CD), SDS-PAGE, dot blot, and amino acid composition, as well as their cytocompatibility envisaging the development of future biomedical applications. An isolation yield of approximately 20% was similar for both sponge parts, as well as the FTIR, CD, and SDS-PAGE profiles, which demonstrated that both isolated collagens presented a high purity degree and preserved their triple helix and fibrillar conformation. Ectosome collagen had a higher OHpro content and possessed collagen type I and IV, while the choanosome was predominately constituted by collagen type IV. In vitro cytotoxicity assays using the L929 fibroblast cell line displayed a significant cytotoxic effect of choanosome collagen at 2 mg/mL, while ectosome collagen enhanced cell metabolism and proliferation, thus indicating the latter as being more suitable for the development of biomaterials. This research represents a unique comparative study of C. reniformis body parts, serving as a support for further establishing this marine sponge as a promising alternative collagen source for the future development of biomedical applications. A comprehensive investigation regarding the hygrothermal behavior of a constructive solution containing phase change materials (PCMs) was performed on a full-scale test cell, divided into two similar compartments. This involved hygrothermal monitoring (indoor air temperature) of the two compartments, in which one had PCM incorporated into the floor mortar. The main goal of this research was to investigate the potential of this kind of solution for overheating mitigation. The numerical study was conducted using EnergyPlus® software (version 9.0), exploring different natural ventilation flow rates to gauge the novel solution's potential to reduce overheating rates. The results from the monitoring studies revealed prolonged periods of thermal discomfort in both test cells, particularly overheating. However, it was proven that the PCM application in one of the test cells led to a reduction of almost 10 °C in the maximum peak of air temperature. In the simulation analysis, the increase in the ventilation rate led to a linear decrease in the overheating hours of up to one renovation per hour, and then the reductions were attenuated. The effectiveness of feed additives delivered through free-choice mineral mixtures (MMs) to grazing cattle remains unclear. Two farm-scale and one in vitro experiment (Exp.) were conducted to investigate the effects of salinomycin and virginiamycin, delivered through an MM, on growing bulls grazing tropical pastures. In Exp. 1, 316 zebu (Bos indicus) Nellore bulls (225 ± 26.7 kg liveweight (LW)) were randomly allocated to four treatments: (1) MM no additives (CON), (2) MM with salinomycin at 1950 mg/kg (SLI), (3) MM with salinomycin at 780 mg/kg (SHI), and (4) MM with virginiamycin at 1950 mg/kg (VGN). Over 123 days, these bulls grazed tropical grasses on pastures of guinea grass, palisade grass, or Bermuda grass. No significant treatment effects were observed for oocyst eggs or ruminal parameters. Bulls fed VGN had higher average daily gain (ADG) compared to CON (p = 0.02) and SLI (p = 0.03) but similar compared to SHI (p = 0.07). In Exp. 2, 308 zebu cross bulls (237 ± 23.0 kg LW) grazed Bermuda grass paddocks and were allocated into two treatments: (1) MM with no additives (CON) and (2) MM containing virginiamycin at 2522 mg/kg (VGN). Cattle fed VGN had a significantly higher ADG (p = 0.007). Exp. 3 tested salinomycin's effectiveness in vitro at different exposure times to MM, revealing no impact of exposure time on short-chain fatty acid production. In conclusion, virginiamycin delivered through free-choice MM can increase grazing beef bulls' ADG by 12% compared with CON, with no clear link to rumen fermentation or coccidiostat effects. A no-tillage system (NT) is an alternative to replace soil management with intense soil tillage and degradation. Our objective was to evaluate the physical–hydric properties of soil under NT for four years after undergoing 13 years of minimum (4NTM) and conventional tillage (4NTC) with reference to continuous NT for 17 years (17NT). The soil bulk density, porosity, storage capacities of water and air, visual evaluation of soil structure (VESS), and saturated hydraulic conductivity were determined. The root dry biomass of soybean, maize, and palisade grass was also measured. NT during four years established after receiving 13 years of tillage did not significantly affect soil properties compared to 17NT, which means that four years of NT reached structural stability similar to continuous 17NT. The VESS scores were less sensitive to identifying soil compaction. Crops have no significative influence on soil properties, but the roots of maize seem to be less sensitive to soil compaction, which is important considering it is in a crop rotation system. The treatments (soil tillage and crop rotation) presented soil compaction below a 10 cm depth, and to our loamy textural class soil, a Bd > 1.60 Mg m−3 restricted soil aeration due to macroporosity < 0.10 m3 m−3. SiOx films, frequently derived from amino silane precursors, have found several applications with high added value. Although frequently used, the deposition of coatings from Tris(dimethyl amino) silane (TDMAS) has been reported to demand considerable amounts of energy, mainly due to the difficulty of oxidizing such compounds. As is well known, Plasma-enhanced atomic layer deposition (PEALD) is able to improve the oxidation efficiency, even under low processing temperatures. Owing to this, PEALD can be considered a very promising technique for the deposition of SiOx coatings. In this work, the deposition of silicon oxide films using TDMAS at 150 °C has been investigated. The effect of the plasma oxidation time (6 to 18 s) and atmosphere composition (pure O2 or O2 + Ar) on the chemical structure, elemental composition, and chemical bonding state of the films has also been evaluated. Increasing the plasma oxidation time in pure O2 resulted in a larger proportion of retained C (Si-CH3), whereas N was preserved in the structure (Si-N). Background: Exercise testing is key in the risk stratification of patients with heart failure (HF). There are scarce data on its prognostic power in women. Our aim was to assess the predictive value of the heart transplantation (HTx) thresholds in HF in women and in men. Methods: Prospective evaluation of HF patients who underwent cardiopulmonary exercise testing (CPET) from 2009 to 2018 for the composite endpoint of cardiovascular mortality and urgent HTx. Results: A total of 458 patients underwent CPET, with a composite endpoint frequency of 10.5% in females vs. 16.0% in males in 36-month follow-up. Peak VO2 (pVO2), VE/VCO2 slope and percent of predicted pVO2 were independent discriminators of the composite endpoint, particularly in women. The International Society for Heart Lung Transplantation recommended values of pVO2 ≤ 12 mL/kg/min or ≤14 if the patient is intolerant to β-blockers, VE/VCO2 slope > 35, and percent of predicted pVO2 ≤ 50% showed a higher diagnostic effectiveness in women. Specific pVO2, VE/VCO2 slope and percent of predicted pVO2 cut-offs in each sex group presented a higher prognostic power than the recommended thresholds. Conclusion: Individualized sex-specific thresholds may improve patient selection for HTx. More evidence is needed to address sex differences in HF risk stratification. [ABSTRACT FROM AUTHOR]

Background and Objectives: Cardiopulmonary exercise testing (CPET) is a cornerstone of risk stratification in heart failure with reduced ejection fraction (HFrEF). However, there is a paucity of evidence on its predictive power in older patients. The aim of this study was to evaluate the prognostic power of current heart transplantation (HTx) listing criteria in HFrEF stratified according to age groups. Materials and Methods: Consecutive patients with HFrEF undergoing CPET between 2009 and 2018 were followed-up for cardiac death and urgent HTx. Results: CPET was performed in 458 patients with HFrEF. The composite endpoint occurred in 16.8% of patients ≤50 years vs. 14.1% of patients ≥50 years in a 36-month follow-up. Peak VO2 (pVO2), VE/VCO2 slope and percentage of predicted pVO2 were strong independent predictors of outcomes. The International Society for Heart and Lung Transplantation thresholds of pVO2 ≤ 12 mL/kg/min (≤14 if intolerant to β-blockers), VE/VCO2 slope > 35 and percentage of predicted pVO2 ≤ 50% presented a higher overall diagnostic effectiveness in younger patients (≤50 years). Specific thresholds for each age subgroup outperformed the traditional cut-offs. Conclusions: Personalized age-specific thresholds may contribute to an accurate risk stratification in HFrEF. Background: New therapies with prognostic benefits have been recently introduced in heart failure with reduced ejection fraction (HFrEF) management. The aim of this study was to evaluate the prognostic power of current listing criteria for heart transplantation (HT) in an HFrEF cohort submitted to cardiopulmonary exercise testing (CPET) between 2009 and 2014 (group A) and between 2015 and 2018 (group B). Methods: Consecutive patients with HFrEF who underwent CPET were followed-up for cardiac death and urgent HT. Results: CPET was performed in 487 patients. The composite endpoint occurred in 19.4% of group A vs. 7.4% of group B in a 36-month follow-up. Peak VO2 (pVO2) and VE/VCO2 slope were the strongest independent predictors of mortality. International Society for Heart and Lung Transplantation (ISHLT) thresholds of pVO2 ≤ 12 mL/kg/min (≤14 if intolerant to β-blockers) and VE/VCO2 slope > 35 presented a similar and lower Youden index, respectively, in group B compared to group A, and a lower positive predictive value. pVO2 ≤ 10 mL/kg/min and VE/VCO2 slope > 40 outperformed the traditional cut-offs. An ischemic etiology subanalysis showed similar results. Conclusion: ISHLT thresholds showed a lower overall prognostic effectiveness in a contemporary HFrEF population. Assessments of the water quality in coastal zones often rely on indirect indicators from contributing river inputs and the neighbouring ocean. Using a novel combination of distance accumulation cost methods and an ocean-colour product derived from SENTINEL-3 data, we developed a probabilistic method for the assessment of dissolved inorganic nitrogen (DIN) in Liverpool Bay (UK) for the period from 2017 to 2020. Using our approach, we showed the annual and monthly likelihood of DIN exposure from its 12 major contributory rivers. Furthermore, we generated monthly risk maps showing the probability of DIN exposure from all rivers, which revealed a seasonal variation of extent and location around the bay. The highest likelihood of high DIN exposure throughout the year was in the estuarine regions of the Dee, Mersey, and Ribble, along with near-shore areas along the north Wales coast and around the mouth of the rivers Mersey and Ribble. There were seasonal changes in the risk of DIN exposure, and this risk remained high all year for the Mersey and Dee estuary regions. In contrast, for the mouth and near the coastal areas of the Ribble, the DIN exposure decreased in spring, remained low during the summer and early autumn, before displaying an increase during winter. Our approach offers the ability to assess the water quality within coastal zones without the need of complex hydrodynamic models, whilst still having the potential to apportion nutrient exposure to specific riverine inputs. This information can help to prioritise how direct mitigation strategies can be applied to specific river catchments, focusing the limited resources for coastal zone and river basin management. Given the current EU decarbonization targets, the railway transport is a key player to boost mobility toward more sustainable transportation, as it is currently the cleanest high-volume mode of locomotion available. However, a study analyzing the life cycle environmental impact of the existing conventional Portuguese railway has never been performed. Aiming to address this research gap, this paper presents an attributional life cycle assessment (LCA) to quantify the environmental impacts of the Portuguese railway infrastructure and rolling stock, using the Douro line case study. Through the LCA methodology, the current setting (using electric and diesel rolling stock) and three scenarios of full-line electrification (considering 2019, 2030, and 2050 electricity mixes) were analyzed for hotspot identification and an outlook on EU-aligned long-term sustainability prospects. In the current scenario, railway operation accounts for 74% of the total carbon footprint, mostly due to the fuel use of diesel trains and the expended electricity of electric train and infrastructure operation. The total electrification of the line and rolling stock can reduce carbon emissions by 38%, 56%, and 63%, if the 2019, 2030, and 2050 electricity mixes are considered, respectively. The wide occurrence of resistance to Cry1A and Cry2A insecticidal toxins from Bacillus thuringiensis (Bt) in the corn earworm/bollworm Helicoverpa zea (Boddie) leaves the Vip3A toxin produced during the vegetative stage of Bt as the only fully active toxin expressed in transgenic crops to control H. zea in the U.S.A. During 2021, the first unexpected survival of H. zea and injury (UXI) on a maize hybrid expressing Cry1A.105, Cry2Ab2, and Vip3Aa in Louisiana, U.S.A. were observed in two sentinel plots used for resistance monitoring. A follow-up intensive investigation was conducted with two H. zea populations established from larvae collected from the two UXI plots. The main goal of this study was to reveal if the unexpected damage was due to resistance development in the insect to the Bt toxins expressed in the maize hybrid. Diet-overlay bioassays showed that the two populations were highly resistant to Cry1A.105, moderately resistant to Cry2Ab2, but still highly susceptible to Vip3Aa when compared to a reference susceptible strain. In 10 d assays with detached ears, the larvae of the two UXI populations exhibited survival on ears expressing only Cry toxins but presented near 100% mortality on maize hybrids containing both cry and vip3A transgenes. Multiple field trials over three years demonstrated that natural H. zea populations in Louisiana were highly resistant to maize expressing only Cry toxins but remained susceptible to all tested hybrids containing cry and vip3A genes. Altogether, the results of this study suggest that the observed UXIs in Louisiana were associated with a resistance to Cry toxins but were not due to a resistance to Vip3A. The possible causes of the UXIs are discussed. Bio-based polyols were obtained from the thermochemical liquefaction of two biomass feedstocks, pinewood and Stipa tenacissima, with conversion rates varying between 71.9 and 79.3 wt.%, and comprehensively characterized. They exhibit phenolic and aliphatic moieties displaying hydroxyl (OH) functional groups, as confirmed by attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR) and nuclear magnetic resonance spectroscopy (NMR) analysis. The biopolyols obtained were successfully employed as a green raw material to produce bio-based polyurethane (BioPU) coatings on carbon steel substrates, using, as an isocyanate source, a commercial bio-based polyisocyanate—Desmodur® Eco N7300. The BioPU coatings were analyzed in terms of chemical structure, the extent of the reaction of the isocyanate species, thermal stability, hydrophobicity, and adhesion strength. They show moderate thermal stability at temperatures up to 100 °C, and a mild hydrophobicity, displaying contact angles between 68° and 86°. The adhesion tests reveal similar pull-off strength values (ca. 2.2 MPa) for the BioPU either prepared with pinewood and Stipa-derived biopolyols (BPUI and BPUII). Electrochemical impedance spectroscopy (EIS) measurements were carried out on the coated substrates for 60 days in 0.05 M NaCl solution. Good corrosion protection properties were achieved for the coatings, with particular emphasis on the coating prepared with the pinewood-derived polyol, which exhibited a low-frequency impedance modulus normalized for the coating thickness of 6.1 × 1010 Ω cm at the end of the 60 days test, three times higher than for coatings prepared with Stipa-derived biopolyols. The aim of this study was to analyze the trend of severe obesity in the capitals of the Brazilian states and the Federal District, from 2006 to 2021. For this purpose, a time-series, population-based, observational study was designed using data from the VIGITEL Survey. The dependent variable of this study was the prevalence of severe obesity, which was defined as a body mass index ≥ 40 kg/m2. Time series analysis was conducted using Joinpoint Regression Analysis Software v.4.9.1.0. In this study, a normal distribution was assumed, and the average annual percentage change (AAPC) and 95% confidence intervals (95% CIs) were presented. In total, 778,445 individuals participated in the study (38.2% were male and 61.8% were female). The prevalence of severe obesity has increased from 1.1% in 2006 to 1.9% in 2021. The average annual percentage change indicates an upward trend for the period (AAPC: 4.7; 95% CI: 3.8; 5.7). When stratifying the trend of severe obesity by sex, a significant upward trend was observed for females (AAPC: 4.3; 95% CI: 1.9; 6.8). The objective of this study was to evaluate the characteristics and recovery of minerals in silages of sorghum cv. IPA 2502 irrigated with different leaching fractions of brackish water. Sorghum cultivation was carried out in the field in a randomised block design, with four replications and four irrigation leaching fractions (0%, 5%, 10%, and 15%). From the harvested plant material, five silos per treatment, with five repetitions, were arranged in a completely randomised design for ensiling. Leaching fractions did not alter the levels of nitrogen, phosphorus, potassium, calcium, magnesium, and sulfur in sorghum. The sodium content showed a quadratic response to the leaching fractions, which was also observed for iron, manganese, and chloride. In silage, an increasing linear effect was found for nitrogen, whereas for phosphorus, potassium, and sulfur, the contents were reduced with the addition of the leaching fraction. The contents of copper, iron, zinc, and chloride in silages responded in a quadratic way. The use of brackish water, especially the 15% leaching fraction, in the cultivation of sorghum enhances the mineral contents and the recovery of some macro- and micro-nutrients after the ensiling process. Current management for diabetes has stimulated the development of versatile 3D-based hydrogels as in vitro platforms for insulin release and as support for the encapsulation of pancreatic cells and islets of Langerhans. This work aimed to create agarose/fucoidan hydrogels to encapsulate pancreatic cells as a potential biomaterial for diabetes therapeutics. The hydrogels were produced by combining fucoidan (Fu) and agarose (Aga), marine polysaccharides derived from the cell wall of brown and red seaweeds, respectively, and a thermal gelation process. The agarose/fucoidan (AgaFu) blended hydrogels were obtained by dissolving Aga in 3 or 5 wt % Fu aqueous solutions to obtain different proportions (4:10; 5:10, and 7:10 wt). The rheological tests on hydrogels revealed a non-Newtonian and viscoelastic behavior, while the characterization confirmed the presence of the two polymers in the structure of the hydrogels. In addition, the mechanical behavior showed that increasing Aga concentrations resulted in hydrogels with higher Young's modulus. Further, the ability of the developed materials to sustain the viability of human pancreatic cells was assessed by encapsulation of the 1.1B4HP cell line for up to 7 days. Inconel 718 is a highly valued material in the aerospace and nuclear industries due to the fact of its exceptional properties. However, the processing of this material is quite difficult, especially through machining processes. Machining this material results in rapid tool wear, even when low material removal rates are considered. In this study, instrumented turning experiments were employed to evaluate the machinability of Inconel 718 alloy using PCBN tools while assessing the usage of two distinct binder phases, TiN and TiC, for those cutting tools. It was found that the tool life was highly sensitive to the cutting speeds but also affected by the workpiece mechanical properties. At lower cutting speeds, notch wear significantly impacted the tool integrity, whereas at higher cutting speeds, flank wear was the primary failure mode of the tool. The flank wear of the tools with TiN-based binder outperformed TiC by almost 30%, presenting a more consistent behavior when machining. [ABSTRACT FROM AUTHOR]

Fucoidan has been reported to present diverse bioactivities, but each extract has specific features from which a particular biological activity, such as immunomodulation, must be confirmed. In this study a commercially available pharmaceutical-grade fucoidan extracted from Fucus vesiculosus, FE, was characterized and its anti-inflammatory potential was investigated. Fucose was the main monosaccharide (90 mol%) present in the studied FE, followed by uronic acids, galactose, and xylose that were present at similar values (3.8–2.4 mol%). FE showed a molecular weight of 70 kDa and a sulfate content of around 10%. The expression of cytokines by mouse bone-marrow-derived macrophages (BMDMs) revealed that the addition of FE upregulated the expression of CD206 and IL-10 by about 28 and 22 fold, respectively, in respect to control. This was corroborated in a stimulated pro-inflammatory situation, with the higher expression (60 fold) of iNOS being almost completely reversed by the addition of FE. FE was also capable of reverse LPS-caused inflammation in an in vivo mouse model, including by reducing macrophage activation by LPS from 41% of positive CD11C to 9% upon fucoidan injection. Collagen is the major structural protein in extracellular matrix present in connective tissues, including skin, being considered a promising material for skin regeneration. Marine organisms have been attracting interest amongst the industry as an alternative collagen source. In the present work, Atlantic codfish skin collagen was analyzed, to evaluate its potential for skincare. The collagen was extracted from two different skin batches (food industry by-product) using acetic acid (ASColl), confirming the method reproducibility since no significant yield differences were observed. The extracts characterization confirmed a profile compatible with type I collagen, without significant differences between batches or with bovine skin collagen (a reference material in biomedicine). Thermal analyses suggested ASColl's native structure loss at 25 °C, and an inferior thermal stability to bovine skin collagen. No cytotoxicity was found for ASColl up to 10 mg/mL in keratinocytes (HaCaT cells). ASColl was used to develop membranes, which revealed smooth surfaces without significative morphological or biodegradability differences between batches. Their water absorption capacity and water contact angle indicated a hydrophilic feature. The metabolic activity and proliferation of HaCaT were improved by the membranes. Electrochemical sensors consisting of screen-printed electrodes (SPEs) are recurrent devices in the recent literature for applications in different fields of interest and contribute to the expanding electroanalytical chemistry field. This is due to inherent characteristics that can be better (or only) achieved with the use of SPEs, including miniaturization, cost reduction, lower sample consumption, compatibility with portable equipment, and disposability. SPEs are also quite versatile; they can be manufactured using different formulations of conductive inks and substrates, and are of varied designs. Naturally, the analytical performance of SPEs is directly affected by the quality of the material used for printing and modifying the electrodes. In this sense, the most varied carbon nanomaterials have been explored for the preparation and modification of SPEs, providing devices with an enhanced electrochemical response and greater sensitivity, in addition to functionalized surfaces that can immobilize biological agents for the manufacture of biosensors. Considering the relevance and timeliness of the topic, this review aimed to provide an overview of the current scenario of the use of carbonaceous nanomaterials in the context of making electrochemical SPE sensors, from which different approaches will be presented, exploring materials traditionally investigated in electrochemistry, such as graphene, carbon nanotubes, carbon black, and those more recently investigated for this (carbon quantum dots, graphitic carbon nitride, and biochar). Marine-origin gelatin has been increasingly used as a safe alternative to bovine and porcine ones due to their structural similarity, avoiding the health-related problems and sociocultural concerns associated with using mammalian-origin materials. Another benefit of marine-origin gelatin is that it can be produced from fish processing-products enabling high production at low cost. Recent studies have demonstrated the excellent capacity of gelatin-methacryloyl (GelMA)-based hydrogels in a wide range of biomedical applications due to their suitable biological properties and tunable physical characteristics, such as tissue engineering applications, including the engineering of cartilage. In this study, fish gelatin was obtained from Greenland halibut skins by an acidic extraction method and further functionalized by methacrylation using methacrylic anhydride, developing a photosensitive gelatin-methacryloyl (GelMA) with a degree of functionalization of 58%. The produced marine GelMA allowed the fabrication of photo-crosslinked hydrogels by incorporating a photoinitiator and UV light exposure. To improve the biological performance, GelMA was combined with two glycosaminoglycans (GAGs): hyaluronic acid (HA) and chondroitin sulfate (CS). GAGs methacrylation reaction was necessary, rendering methacrylated HA (HAMA) and methacrylated CS (CSMA). Three different concentrations of GelMA were combined with CSMA and HAMA at different ratios to produce biomechanically stable hydrogels with tunable physicochemical features. The 20% (w/v) GelMA-based hydrogels produced in this work were tested as a matrix for chondrocyte culture for cartilage tissue engineering with formulations containing both HAMA and CSMA showing improved cell viability. Background: Data on the impact of sacubitril/valsartan (SV) therapy on phasic left atrial (LA) and left ventricular (LV) strain in heart failure with reduced ejection fraction (HFrEF) are limited. The aim of this study was to evaluate changes in two-dimensional speckle tracking (2D-STE) parameters with SV therapy in HFrEF patients. Methods: Prospective evaluation of HFrEF patients receiving optimized medical therapy. Two-dimensional speckle tracking (2D-STE) parameters were assessed at baseline and after 6 months of SV therapy. LA strain and strain rate (SR) in reservoir, conduit, and contraction phases were compared with LV longitudinal, radial, and circumferential strain and SR and stratified according to heart rhythm and HFrEF etiology. Results: A total of 35 patients completed the 6-month follow-up, with a mean age of 59 ± 11 years, 40% in atrial fibrillation, 43% with ischemic etiology, and LVEF of 29 ± 6%. There were significant improvements in LA reservoir, conduit, and contractile strain and SR following SV therapy, particularly among patients in sinus rhythm. There were significant improvements in longitudinal, radial, and circumferential LV function indices. Conclusion: SV therapy in HFrEF was associated with improved longitudinal, radial, and circumferential function, particularly among patients in sinus rhythm. The self-repair capacity of human tissue is limited, motivating the arising of tissue engineering (TE) in building temporary scaffolds that envisage the regeneration of human tissues, including articular cartilage. However, despite the large number of preclinical data available, current therapies are not yet capable of fully restoring the entire healthy structure and function on this tissue when significantly damaged. For this reason, new biomaterial approaches are needed, and the present work proposes the development and characterization of innovative polymeric membranes formed by blending marine origin polymers, in a chemical free cross-linking approach, as biomaterials for tissue regeneration. The results confirmed the production of polyelectrolyte complexes molded as membranes, with structural stability resulting from natural intermolecular interactions between the marine biopolymers collagen, chitosan and fucoidan. Furthermore, the polymeric membranes presented adequate swelling ability without compromising cohesiveness (between 300 and 600%), appropriate surface properties, revealing mechanical properties similar to native articular cartilage. From the different formulations studied, the ones performing better were the ones produced with 3 % shark collagen, 3% chitosan and 10% fucoidan, as well as with 5% jellyfish collagen, 3% shark collagen, 3% chitosan and 10% fucoidan. Overall, the novel marine polymeric membranes demonstrated to have promising chemical, and physical properties for tissue engineering approaches, namely as thin biomaterial that can be applied over the damaged articular cartilage aiming its regeneration. Dilated cardiomyopathy (DCM) represents one of the most common causes of non-ischemic heart failure, characterised by ventricular dilation alongside systolic dysfunction. Despite advances in therapy, DCM mortality rates remain high, and it is one of the leading causes of heart transplantation. It was recently recognised that many patients present minor structural cardiac abnormalities and express different arrhythmogenic phenotypes before overt heart-failure symptoms. This has raised several diagnostic and management challenges, including the differential diagnosis with other phenotypically similar conditions, the identification of patients at increased risk of malignant arrhythmias, and of those who will have a worse response to medical therapy. Recent developments in complementary diagnostic procedures, namely cardiac magnetic resonance and genetic testing, have shed new light on DCM understanding and management. The present review proposes a comprehensive and systematic approach to evaluating DCM, focusing on an improved diagnostic pathway and a structured stratification of arrhythmic risk that incorporates novel imaging modalities and genetic test results, which are critical for guiding clinical decision-making and improving outcomes. Arthropods, the largest animal phylum, including insects, spiders and crustaceans, are characterized by their bodies being covered primarily in chitin. Besides being a source of this biopolymer, crustaceans have also attracted attention from biotechnology given their cuticles' remarkable and diverse mechanical properties. The goose barnacle, Pollicipes pollicipes, is a sessile crustacean characterized by their body parts covered with calcified plates and a peduncle attached to a substrate covered with a cuticle. In this work, the composition and structure of these plates and cuticle were characterized. The morphology of the tergum plate revealed a compact homogeneous structure of calcium carbonate, a typical composition among marine invertebrate hard structures. The cuticle consisted of an outer zone covered with scales and an inner homogenous zone, predominantly organic, composed of successive layers parallel to the surface. The scales are similar to the tergum plate and are arranged in parallel and oriented semi-vertically. Structural and biochemical characterization confirmed a bulk composition of ɑ-chitin and suggested the presence of elastin-based proteins and collagen. The mechanical properties of the cuticle showed that the stiffness values are within the range of values described in elastomers and soft crustacean cuticles resulting from molting. The removal of calcified components exposed round holes, detailed the structure of the lamina, and changed the protein properties, increasing the rigidity of the material. This flexible cuticle, predominantly inorganic, can provide bioinspiration for developing biocompatible and mechanically suitable biomaterials for diverse applications, including in tissue engineering approaches. The industrial processing of fish for food purposes also generates a considerable number of by-products such as viscera, bones, scales, and skin. From a value-added perspective, fish by-products can act also as raw materials, especially because of their collagen content (particularly in fish skin). Interestingly, the potential of marine collagen for cosmetic applications is enormous and, remarkably, the extraction of this protein from fish skins has been established for different species. Using this approach, we investigated the integration of marine collagen (COLRp_I) extracted from the skin of the Greenland halibut as an active ingredient in a cosmetic hydrogel formulation. In this study, extracts of marine collagen at concentrations up to 10 mg/mL showed a non-cytotoxic effect when cultured with fibroblast cells for 3 days. In addition, marine collagen extract, when incorporated into a cosmetic hydrogel formulation, met criterion A of ISO 11930:2019 regarding the efficacy of the preservative system (challenge test). In addition, the cosmetic formulations based on marine collagen at dosages of 0.1, 0.25 and 0.5% were tested in a clinical study on the skin of the forearms of 23 healthy volunteers, showing a sightly hydration effect, suggesting its potential for beauty applications. Moreover, this work illustrates that the circular economy concept applied to the fish processing industry can represent important benefits, at innovation, environmental and economic levels. The demand for bio-based and safer composite materials is increasing due to the growth of the industry, human population, and environmental concerns. In this framework, sustainable and safer cork-polymer composites (CPC), based on green low-density polyethylene (LDPE) were developed using melt-based technologies. Chitosan and polyethylene-graft-maleic anhydride (PE-g-MA) were employed to enhance the CPC's properties. The morphology, wettability, mechanical, thermal, and antibacterial properties of the CPC against Pseudomonas putida (P. putida) and Staphylococcus aureus (S. aureus) were examined. The CPC showed improved stiffness when compared with that of the LDPE matrix, preferably when combined with chitosan and PE-g-MA (5 wt. %), reinforcing the stiffness (58.8%) and the strength (66.7%). Chitosan also increased the composite stiffness and strength, as well as reduced the surface hydrophilicity. The CPCs' antibacterial activity revealed that cork significantly reduces the biofilm on the polymer matrix. The highest biofilm reduction was found with CPC containing cork and 5 wt. % chitosan for both P. putida (54% reduction) and S. aureus (36% reduction), confirming their potential to extend the lifespan of products for packaging and healthcare, among other applications. Chlorella vulgaris is a green microalga with a high chlorophyll content, representing a valuable source of green pigments for food applications. As the application of whole biomass can promote an unpleasant fish-like flavor, the use of chlorophyll extract can overcome this drawback. However, chlorophylls tend to easily degrade when out of the chloroplasts, decreasing their potential as a food ingredient. Thus, to study the suitable conditions for isolated chlorophylls preservation, in this work, the influence of temperature (4 to 60 °C), light (dark or 24 h photoperiod), alkaline conditions (with or without aqueous NaOH addition), and modified atmosphere (air or argon atmosphere) on the stability of the color in ethanolic solutions obtained from C. vulgaris were studied. The loss of green color with temperature followed the first-order kinetics, with an activation energy of 74 kJ/mol. Below 28 °C and dark conditions were suitable to preserve isolated chlorophylls. The addition of NaOH and an inert argon-rich atmosphere did not exhibit a statistically positive effect on color preservation. In the case study, cooked cold rice was colored to be used in sushi. The color remained stable for up to 3 days at 4 °C. Therefore, this work showed that C. vulgaris chlorophylls could be preserved in ethanolic solutions at room or lower temperatures when protected from light, allowing them to obtain a suitable natural food ingredient to color foodstuffs. Corneal pathologies from infectious or noninfectious origin have a significant impact on the daily lives of millions of people worldwide. Despite the risk of organ rejection or infection, corneal transplantation is currently the only effective treatment. Finding safe and innovative strategies is the main goal of tissue-engineering-based approaches. In this study, the potential of gelatin methacryloyl (GelMA) hydrogels produced from marine-derived gelatin and loaded with ascorbic acid (as an enhancer of the biological activity of cells) was evaluated for corneal stromal applications. Marine GelMA was synthesized with a methacrylation degree of 75%, enabling effective photocrosslinking, and hydrogels with or without ascorbic acid were produced, encompassing human keratocytes. All the produced formulations exhibited excellent optical and swelling properties with easy handling as well as structural stability and adequate degradation rates that may allow proper extracellular matrix remodeling by corneal stromal cells. Formulations loaded with 0.5 mg/mL of ascorbic acid enhanced the biological performance of keratocytes and induced collagen production. These results suggest that, in addition to marine-derived gelatin being suitable for the synthesis of GelMA, the hydrogels produced are promising biomaterials for corneal regeneration applications. Bio-oils produced from three different biomass sources, namely cork, pinewood, and olive stones, are evaluated concerning their suitability and prospects of including their electrochemical transformations in a biorefinery scenario for the production of added-value compounds. Different types and concentrations of electrolytes (e.g., H2SO4, KOH) are added to the bio-oils to increase the samples' initially low ionic conductivity. The samples prepared by mixing bio-oil with 2 M KOH aqueous solution (50 vol.%) lead to a stable and homogeneous bio-oil alkaline emulsion suitable for electrochemical studies. The bio-oil samples are characterized by physicochemical methods (e.g., density, viscosity, conductivity), followed by analyzing their electrochemical behavior by voltammetric and chronoamperometric studies. The organics electrooxidation and the hydrogen evolution reaction in the bio-oils are assessed using Pt electrodes. Single- and two-compartment cell laboratory bio-oil electrolyzers are assembled using nickel plate electrodes. Electrolysis is carried out at 2.5 V for 24 h. Attenuated Total Reflection-Fourier-Transform Infrared Spectroscopy and Mass Spectrometry are applied to identify possible changes in the bio-oil samples' chemical structure during the electrolysis experiments. Comparing the analyses of the bio-oil samples subjected to electrolysis with the blank samples demonstrates that bulk electrolysis significantly changes the bio-oil composition. The bio-oil obtained from cork biomass shows the most promising results, but further studies are required to understand the nature of the actual changes. [ABSTRACT FROM AUTHOR]

(1) Background: Peripheral nerve injuries represent a major clinical challenge. If nerve ends retract, there is no spontaneous regeneration and grafts are required to proximate the nerve ends and give continuity to the nerve. (2) Methods: GDNF-loaded NPs were characterized physicochemically. For that, NPs stability at different pH's was assessed, and GDNF release was studied through ELISA. In vitro studies are performed with Schwann cells, and the NPs are labeled with fluorescein-5(6)-isothiocyanate for uptake experiments with SH-SY5Y neural cells. (3) Results: GDNF-loaded NPs are stable in physiological conditions, releasing GDNF in a two-step profile, which is beneficial for nerve repair. Cell viability is improved after 1 day of culture, and the uptake is near 99.97% after 3 days of incubation. Despite its low prevalence, pancreatic cancer (PC) is one of the deadliest, typically characterised as silent in early stages and with a dramatically poor prognosis when in its advanced stages, commonly associated with a high degree of metastasis. Many efforts have been made in pursuing innovative therapeutical approaches, from the search for new cytotoxic drugs and other bioactive compounds, to the development of more targeted approaches, including improved drug delivery devices. Marine biotechnology has been contributing to this quest by providing new chemical leads and materials originating from different organisms. In this review, marine biodiscovery for PC is addressed, particularly regarding marine invertebrates (namely sponges, molluscs, and bryozoans), seaweeds, fungi, and bacteria. In addition, the development of biomaterials based on marine-originating compounds, particularly chitosan, fucoidan, and alginate, for the production of advanced cancer therapies, is also discussed. Collagen is a ubiquitous protein present in the extracellular matrix of all major metazoan animals, with approximately 28 different human collagen types described in the literature, each with unique physicochemical properties. Collagens found broad application in the cosmeceutical, pharmaceutical, and biomedical fields and can be isolated from environmentally sustainable sources such as marine byproducts, which are abundant in the fish processing industry and are highly appealing low-cost sources. In this study, marine collagen was isolated from the skins of Greenland halibut (Reinhardtius hippoglossoides), an unexplored byproduct from fish processing plants, using three different collagen extraction methods, due to the use of distinct salting-out methods using a solution of 2.6 M NaCl + 0.05 M Tris-HCl pH = 7.5, (method I); a combination of 0.7 M NaCl followed by a solution of 2.3 M NaCl + 0.05 M Tris-HCl pH = 7.5 (method II); and one method using only 0.9 M NaCl (method III), yielding COLRp_I, COLRp_II, and COLRp_III collagens. These extracted type I collagens were produced with a yield of around 2 and 4% and characterized regarding the physicochemical properties, considering possible biotechnological applications. This work evidenced that the typical triple helix structure conformation was preserved in all extraction methods, but influenced the thermal behavior, intrinsic morphology, and moisture capacity of the collagens, with interest for biotechnological application, as the incorporation as an ingredient in cosmetic formulation. Furthermore, the use of collagen isolated from skin byproducts represents a high economic value with decreasing collagen cost for industrial purposes and is also an environmentally sustainable source for industrial uses. The successful integration of transplanted three-dimensional tissue engineering (TE) constructs depends greatly on their rapid vascularization. Therefore, it is essential to address this vascularization issue in the initial design of constructs for perfused tissues. Two of the most important variables in this regard are scaffold composition and cell sourcing. Collagens with marine origins overcome some issues associated with mammal-derived collagen while maintaining their advantages in terms of biocompatibility. Concurrently, the freshly isolated stromal vascular fraction (SVF) of adipose tissue has been proposed as an advantageous cell fraction for vascularization purposes due to its highly angiogenic properties, allowing extrinsic angiogenic growth factor-free vascularization strategies for TE applications. In this study, we aimed at understanding whether marine collagen 3D matrices could support cryopreserved human SVF in maintaining intrinsic angiogenic properties observed for fresh SVF. For this, cryopreserved human SVF was seeded on blue shark collagen sponges and cultured up to 7 days in a basal medium. The secretome profile of several angiogenesis-related factors was studied throughout culture times and correlated with the expression pattern of CD31 and CD146, which showed the formation of a prevascular network. Upon in ovo implantation, increased vessel recruitment was observed in prevascularized sponges when compared with sponges without SVF cells. Immunohistochemistry for CD31 demonstrated the improved integration of prevascularized sponges within chick chorioalantoic membrane (CAM) tissues, while in situ hybridization showed human cells lining blood vessels. Fibre metal laminates (FML) are significantly adopted in the aviation industry due to their convenient combination of specific strength, impact resistance and ductility. Drilling of such materials is a regular pre-requisite which enables assembly operations, typically through rivet joining. However, the hole-making operation is of increased complexity due to the dissimilarity of the involved materials, often resulting in defects (i.e., material interface delamination), which can significantly compromise the otherwise excellent fatigue strength. This work explores the potential of three different drill geometries, operating under variable cutting speeds and feeds on CFRP-AA laminates. In addition, the usage of sacrificial back support is investigated and cutting load, surface roughness and delamination extension are examined. In order to predict delamination occurrence, ADCB tests are performed, enabling the calculation of fracture energy threshold. Drill geometry presents a very significant influence on delamination occurrence. The usage of specific step-tools with secondary cutting edge showed superior performance. Despite its simplicity, the applied critical force threshold model was able to successfully predict interface delamination with good accuracy. [ABSTRACT FROM AUTHOR]

Background: Dapagliflozin has been shown to reduce morbidity and mortality in Heart Failure with reduced Ejection Fraction (HFrEF), but its impact on exercise capacity of non-diabetic HF outpatients is unknown. Methods: Adult non-diabetic HF patients with a left ventricular ejection fraction (LVEF) <50% were randomized 1:1 to receive dapagliflozin 10 mg or to continue with HF medication. Patients underwent an initial evaluation which was repeated after 6 months. The variation of several clinical parameters was compared, with the primary endpoint being the 6 month peak oxygen uptake (pVO2) variation. Results: A total of 40 patients were included (mean age 61 ± 13 years, 82.5% male, mean LVEF 34 ± 5%), half being randomized to dapagliflozin, with no significant baseline differences between groups. The reported drug compliance was 100%, with no major safety events. No statistically significant difference in HF events was found (p = 0.609). There was a 24% reduction in the number of patients in New York Heart Association (NYHA) class III in the treatment group as opposed to a 15.8% increase in the control group (p = 0.004). Patients under dapagliflozin had a greater improvement in pVO2 (3.1 vs. 0.1 mL/kg/min, p = 0.030) and a greater reduction in NT-proBNP levels (−217.6 vs. 650.3 pg/mL, p = 0.007). Conclusion: Dapagliflozin was associated with a significant improvement in cardiopulmonary fitness at 6 months follow-up in non-diabetic HFrEF patients. [ABSTRACT FROM AUTHOR]

Marine origin polymers represent a sustainable and natural alternative to mammal counterparts regarding the biomedical application due to their similarities with proteins and polysaccharides present in extracellular matrix (ECM) in humans and can reduce the risks associated with zoonosis and overcoming social- and religious-related constraints. In particular, collagen-based biomaterials have been widely explored in tissue engineering scaffolding applications, where cryogels are of particular interest as low temperature avoids protein denaturation. However, little is known about the influence of the parameters regarding their behavior, i.e., how they can influence each other toward improving their physical and chemical properties. Factorial design of experiments (DoE) and response surface methodology (RSM) emerge as tools to overcome these difficulties, which are statistical tools to find the most influential parameter and optimize processes. In this work, we hypothesized that a design of experiments (DoE) model would be able to support the optimization of the collagen-chitosan-fucoidan cryogel manufacturing. Therefore, the parameters temperature (A), collagen concentration (B), and fucoidan concentration (C) were carefully considered to be applied to the Box–Behnken design (three factors and three levels). Data obtained on rheological oscillatory measurements, as well as on the evaluation of antioxidant concentration and adenosine triphosphate (ATP) concentration, showed that fucoidan concentration could significantly influence collagen-chitosan-fucoidan cryogel formation, creating a stable internal polymeric network promoted by ionic crosslinking bonds. Additionally, the effect of temperature significantly contributed to rheological oscillatory properties. Overall, the condition that allowed us to have better results, from an optimization point of view according to the DoE, were the gels produced at −80 °C and composed of 5% of collagen, 3% of chitosan, and 10% fucoidan. Biosurfactants and bioemulsifiers (BS/BE) are naturally synthesized molecules, which can be used as alternatives to traditional detergents. These molecules are commonly produced by microorganisms isolated from hydrocarbon-rich environments. Gordonia alkanivorans strain 1B was originally found in such an environment, however little was known about its abilities as a BS/BE producer. The goal of this work was to access the potential of strain 1B as a BS/BE producer and perform the initial characterization of the produced compounds. It was demonstrated that strain 1B was able to synthesize lipoglycoprotein compounds with BS/BE properties, both extracellularly and adhered to the cells, without the need for a hydrophobic inducer, producing emulsion in several different hydrophobic phases. Using a crude BS/BE powder, the critical micelle concentration was determined (CMC = 16.94 mg/L), and its capacity to reduce the surface tension to a minimum of 35.63 mN/m was demonstrated, surpassing many commercial surfactants. Moreover, after dialysis, emulsification assays revealed an activity similar to that of Triton X-100 in almond and sunflower oils. In benzene, the E24 value attained was 83.45%, which is 30% greater than that of the commercial alternative. This work was developed within the scope of the research project "Easyfloor—Development of composite sandwich panels for building floor rehabilitation", which aims at developing an innovative hybrid sandwich panel as an alternative construction system to conventional floor solutions, mainly for building rehabilitation. The developed hybrid sandwich panel is composed of a top face layer of steel-fibre-reinforced self-compacting concrete (FRC), a core of polyurethane (PUR) closed-cell foam, a bottom face sheet, and lateral webs of glass-fibre-reinforced polymer (GFRP). Full-scale experimental tests on the developed sandwich panels were carried out to characterize their short- and long-term (creep) flexural behaviour. The present work includes a detailed description of the developed panels and the experimental programme carried out and presents and discusses the relevant results. The experimental results showed an almost linear behaviour up to failure. The creep tests were carried out for a period of 180 days, using a creep load equal to 20% of its ultimate loading capacity. An average creep coefficient of 0.27 was obtained for this period. The composed creep model used to simulate the sandwich panel's creep deflections by considering the individual viscoelastic contributions was able to predict the observed structural response with good accuracy. Due to the lack of a general drought definition, water users and managers have developed and used different indices. Many studies using drought indices have been made so as to detect drought events or just to compare their results and assess their advantages and disadvantages. In Portugal, these studies have been done for common drought indices; however, an integrated evaluation and comparison using recent data is needed. Therefore, this study is intended to give an updated overview of the behaviour of the proposed indices. This study proposes the usage of PDSI, scPDSI, SPI and SPEI. With the exception of the PDSI, all indices have been calculated through R packages. The results for the studied regions in mainland Portugal suggest that the drought situations are, in general, most significant and frequent than the wet periods. From our results, we can conclude that the SPI model is more sensitive to extreme drought events and can detect them earlier. The PDSI, scPDSI and SPEI are more reliable for drought monitorization at medium and long spells, which might represent the environmental interactions more closely to the reality. Also, the scPDSI tends to reduce the importance of short period recovering. It is then advisable that impact and scientifical studies consider all of these indices or at least some of them to have a broader and complete understanding of the drought situations to be studied.
