In order to enhance the comprehensive processing quality and production efficiency of seed melons, a seed melon crushing and seed-extraction separator has been developed and designed. Aiming at the issues of high impurity rate and scratch rate of melon seeds in the process of seed–flesh separation, the structure and parameters of the seed–flesh separation device were optimized in this study by simulation analysis and field testing. The simulation model of melon seed, melon flesh, and the seed–flesh separation device based on the discrete element method (DEM) was established, and the simulation parameters were calibrated. Subsequently, the melon seed impurity rate (G1) and the melon seed scratch rate (G2) were used as the evaluation indexes. The single-factor simulation test was carried out on the separation roller speed (A). The spacing between the scraper and the screen (B), the separation roller scraper inclination angle (C), and the influence rules of each factor on the separation effect of the seed–flesh were obtained. Finally, the three-factor and three-level orthogonal test was carried out. Using the method of ANOVA and multi-objective optimization, the optimal working parameters of the device were obtained as A-117.53 r/min, B-5 mm, and C-10°, at which time the optimal evaluation indexes were G1-5.59% and G2-2.85%. The prototype test was carried out with the optimization results. The values of G1 and G2 were measured at 5.71% and 2.91%, respectively, and the relative errors with the simulation values were 2.15% and 2.11%, respectively, which were basically the same between the simulation model and the prototype test. The results indicate that the designed separation roller speed, spacing between the scraper and screen, and separation roller scraper inclination angle can meet the requirements of seed–flesh separation in the seed melon crushing and seed-extraction separator. The traditional microstructure detecting methods such as metallography and electron backscatter diffraction are destructive to the sample and time-consuming and they cannot meet the needs of rapid online inspection. In this paper, a random forest regression microstructure characterization method based on a laser ultrasound technique is investigated for evaluating the microstructure of a titanium alloy (Ti-6Al-4V). Based on the high correlation between the longitudinal wave velocity of ultrasonic waves, the average grain size of the primary α phase, and the volume fraction of the transformed β matrix of the titanium alloy, and with the longitudinal wave velocity as the input feature and the average grain size of the primary α phase and the volume fraction of the transformed β matrix as the output features, prediction models for the average grain size of the primary α phase and the volume fraction of the transformed β matrix were developed based on a random forest regression. The results show that the mean values of the mean relative errors of the predicted mean grain size of the native α phase and the volume fraction of the transformed β matrix for the six samples in the two prediction models were 11.55% and 10.19%, respectively, and the RMSE and MAE obtained from both prediction models were relatively small, which indicates that the two established random forest regression models have a high prediction accuracy. Comprehensive vegetation surveys are crucial for species selection and layout during the restoration of degraded island ecosystems. However, due to the poor accessibility of uninhabited islands, traditional quadrat surveys are time-consuming and labor-intensive, and it is challenging to fully identify the specific species and their spatial distributions. With miniaturized sensors and strong accessibility, high spatial and temporal resolution, Unmanned Aerial Vehicles (UAVs) have been extensively implemented for vegetation surveys. By collecting UAVs multispectral images and conducting field quadrat surveys on Anyu Island, we employ four machine learning models, namely Gradient Boosting Decision Tree (GBDT), Support Vector Machine (SVM), Random Forest (RF) and Multiple Classifier Systems (MCS). We aim to identify the dominant species and analyze their spatial distributions according to spectral characteristics, vegetation index, topographic factors, texture features, and canopy heights. The results indicate that SVM model achieves the highest (88.55%) overall accuracy (OA) (kappa coefficient = 0.87), while MCS model does not significantly improve it as expected. Acacia confusa has the highest OA among 7 dominant species, reaching 97.67%. Besides the spectral characteristics, the inclusion of topographic factors and texture features in the SVM model can significantly improve the OA of dominant species. By contrast, the vegetation index, particularly the canopy height even reduces it. The dominant species exhibit significant zonal distributions with distance from the coastline on the Anyu Island (p < 0.001). Our study provides an effective and universal path to identify and map the dominant species and is helpful to manage and restore the degraded vegetation on uninhabited islands. To enhance the utilization of seed-used watermelon peel and mitigate environmental pollution, a hammer-blade seed-used watermelon peel crusher was designed and manufactured, and its structure and working parameters were optimized. Initially, the seed-used watermelon peel crusher and seed-used watermelon peel model were constructed, and the model's parameters were calibrated. Subsequently, the discrete element method (EDEM2022) was employed to investigate the effects of spindle speed (MSS), the number of hammers (NCB), and feeding volume (FQ) on the pulverizing process. Multivariate nonlinear regression prediction models were developed for the percentage of pulverized particle size less than 8 mm (Psv), pulverizing efficiency (Ge), and power density (Ppd), followed by the analysis of influencing factors and prediction models using ANOVA. The multiobjective optimization of the prediction model utilizing the improved hybrid metacellular genetic algorithm CellDE resulted in solutions of 90.02%, 89.57%, and 8.35 × 10−3 t/(h-kw) for Psv-opt, Ge-opt, and Ppd-opt, respectively. The corresponding optimal interaction values of MSS, NCB, and FQ were determined to be 1500 r/min, 108, and 150 kg/min. Finally, a prototype test was conducted by combining the optimal factor interaction values, yielding statistically calculated values of 96.63%, 92.37%, and 7.76 × 10−3 t/(h-kw) for Psv-pr, Ge-pr, and Ppd-pr, respectively. The results indicate that the optimized values of Psv-opt, Ge-opt, and Ppd-opt models have an error of less than 8% compared to the statistically calculated values of the prototype test and outperform the values of Psv-ori, Ge-ori, and Ppd-ori obtained under the original parameters. FHY3 and FAR1 serve as positive regulators of the phyA-mediated far-red light signal pathway, influencing plant growth and development by regulating the expression of downstream genes. However, little is known about the FAR1/FHY3 family in Brassica species. A total of 21 members of the BnFAR1/FHY3 gene family were identified in the Brassica napus genome, exhibiting an uneven distribution across ten B. napus chromosomes. A phylogenetic analysis showed that the BnFAR1/FHY3 family could be divided into four subfamilies. Putative cis-elements in the BnFAR1/FHY3 promoter regions were also identified, which were potentially involved in phytohormone, light and abiotic stress responses (shading, low-temperature, etc.). Additionally, qRT-PCR results indicated that the expression levels of BnFAR1-10, BnFAR1-11, BnFAR1-21 and BnFAR1-4 decreased under shading stress. The expression of BnFAR1-10, BnFAR1-11 and BnFAR1-21 increased under low-temperature stress, whereas the expression of BnFAR1-4 did not change. In addition, the results of the tissue expression analysis showed that most of the genes exhibited the lowest expression in pollen and the highest expression in the 54-day silique. This study screened a batch of BnFHY3/BnFAR1 gene resources, which will contribute to further research on the functional characteristics of BnFHY3/BnFAR1 family members in growth, development and the stress response. This study reveals the influence of word of mouth on destination reputation from the perspective of tourists' reputation judgment and explores the differentiating effect of judgment dimensions on the degree of consistency between word of mouth and reputation based on multinomial logistic regression analysis, aiming to advance the theoretical exploration of the complex relationship between word of mouth and reputation. This study finds that (1) tourists weigh word-of-mouth information that is identical or different from their views to make reputation judgments, and destination reputation is a systematic emergent outcome of word-of-mouth information that is recursively weighed and judged by tourists in the process of dissemination. (2) Tourists judge the reputation of a destination based on different dimensions, including scenic spots, local food, history and culture, natural environment, social atmosphere, activities and events, and facilities and services. (3) Tourists' reputation judgment dimensions have a differentiating effect on the influence of word of mouth on destination reputation; that is, there are interpersonal differences in the influence of word of mouth information on destination reputation by tourists based on different dimensions. This paper provides a new perspective and approach to the study of the relationship between word of mouth and destination reputation, which can contribute to destination reputation management and marketing efforts. Oligomeric Proanthocyanidins (OPCs), as a class of compounds widely found in plants, are particularly abundant in grapes and blueberries. It is a polymer comprising many different monomers, such as catechins and epicatechins. The monomers are usually linked to each other by two types of links, A-linkages (C-O-C) and B-linkages (C-C), to form the polymers. Numerous studies have shown that compared to high polymeric procyanidins, OPCs exhibit antioxidant properties due to the presence of multiple hydroxyl groups. This review describes the molecular structure and natural source of OPCs, their general synthesis pathway in plants, their antioxidant capacity, and potential applications, especially the anti-inflammatory, anti-aging, cardiovascular disease prevention, and antineoplastic functions. Currently, OPCs have attracted much attention, being non-toxic and natural antioxidants of plant origin that scavenge free radicals from the human body. In plants, the multi-gene family of dual-function hexokinases (HXKs) plays an important role in sugar metabolism and sensing, that affects growth and stress adaptation. Sugarcane is an important sucrose crop and biofuel crop. However, little is known about the HXK gene family in sugarcane. A comprehensive survey of sugarcane HXKs, including physicochemical properties, chromosomal distribution, conserved motifs, and gene structure was conducted, identifying 20 members of the SsHXK gene family that were located on seven of the 32 Saccharum spontaneum L. chromosomes. Phylogenetic analysis showed that the SsHXK family could be divided into three subfamilies (group I, II and III). Motifs and gene structure were related to the classification of SsHXKs. Most SsHXKs contained 8–11 introns which was consistent with other monocots. Duplication event analysis indicated that HXKs in S. spontaneum L. primarily originated from segmental duplication. We also identified putative cis-elements in the SsHXK promoter regions which were involved in phytohormone, light and abiotic stress responses (drought, cold et al.). During normal growth and development, 17 SsHXKs were constitutively expressed in all ten tissues. Among them, SsHXK2, SsHXK12 and SsHXK14 had similar expression patterns and were more highly expressed than other genes at all times. The RNA-seq analysis showed that 14/20 SsHXKs had the highest expression level after cold stress for 6 h, especially SsHXK15, SsHXK16 and SsHXK18. As for drought treatment, 7/20 SsHXKs had the highest expression level after drought stress for 10 days, 3/20 (SsHKX1, SsHKX10 and SsHKX11) had the highest expression level after 10 days of recovery. High levels of cadmium (Cd) in soil can cause crop yield reduction or death. Cadmium accumulation in crops affects human and animal health as it passes through the food chain. Therefore, a strategy is needed to enhance the tolerance of crops to this heavy metal or reduce its accumulation in crops. Abscisic acid (ABA) plays an active role in plants' response to abiotic stress. The application of exogenous ABA can reduce Cd accumulation in shoots of some plants and enhance the tolerance of plants to Cd; therefore, ABA may have good application prospects. In this paper, we reviewed the synthesis and decomposition of ABA, ABA-mediated signal transduction, and ABA-mediated regulation of Cd-responsive genes in plants. We also introduced physiological mechanism underlying Cd tolerance because of ABA. Specifically, ABA affects metal ion uptake and transport by influencing transpiration and antioxidant systems, as well as by affecting the expression of metal transporter and metal chelator protein genes. Brassica oleracea displays remarkable morphological variations. It intrigued researchers to study the underlying cause of the enormous diversification of this organism. However, genomic variations in complex heading traits are less known in B. oleracea. Herein, we performed a comparative population genomics analysis to explore structural variations (SVs) responsible for heading trait formation in B. oleracea. Synteny analysis showed that chromosomes C1 and C2 of B. oleracea (CC) shared strong collinearity with A01 and A02 of B. rapa (AA), respectively. Two historical events, whole genome triplication (WGT) of Brassica species and differentiation time between AA and CC genomes, were observed clearly by phylogenetic and Ks analysis. By comparing heading and non-heading populations of B. oleracea genomes, we found extensive SVs during the diversification of the B. oleracea genome. We identified 1205 SVs that have an impact on 545 genes and might be associated with the heading trait of cabbage. Overlapping the genes affected by SVs and the differentially expressed genes identified by RNA-seq analysis, we identified six vital candidate genes that may be related to heading trait formation in cabbage. Further, qRT-PCR experiments also verified that six genes were differentially expressed between heading leaves and non-heading leaves, respectively. Collectively, we used available genomes to conduct a comparison population genome analysis and identify candidate genes for the heading trait of cabbage, which provides insight into the underlying reason for heading trait formation in B. oleracea. The VQ motif-containing proteins play a vital role in various processes such as growth, resistance to biotic and abiotic stresses and development. However, there is currently no report on the VQ genes in sugarcane (Saccharum spp.). Herein, 78 VQ genes in Saccharum spontaneum were identified and classified into nine subgroups (I-IX) by comparative genomic analyses. Each subgroup had a similar structural and conservative motif. These VQ genes expanded mainly through whole-genome segmental duplication. The cis-regulatory elements (CREs) of the VQ genes were widely involved in stress responses, phytohormone responses and physiological regulation. The RNA-seq data showed that SsVQ gene expression patterns in 10 different samples, including different developmental stages, revealed distinct temporal and spatial patterns. A total of 23 SsVQ genes were expressed in all tissues, whereas 13 SsVQ genes were not expressed in any tissues. Sequence Read Archive (SRA) data showed that the majority of SsVQs responded to cold and drought stress. In addition, quantitative real-time PCR analysis showed that the SsVQs were variously expressed under salicylic acid (SA), jasmonic acid (JA), abscisic acid (ABA) and cold treatment. This study conducted a full-scale analysis of the VQ gene family in sugarcane, which could be beneficial for the functional characterization of sugarcane VQ genes and provide candidate genes for molecular resistance breeding in cultivated sugarcane in the future. Tourism safety perception is one of the factors influencing destination image, but there is a lack of systematic research on the multidimensional influence of tourism safety perception on destination image. In this paper, based on the survey data collected from 623 tourists traveling in Xinjiang, China, we classified the respondents into three types of high, medium, and low levels of tourism safety perception by cluster analysis, and studied the influence of tourism safety perception on destination image in multiple dimensions by means of content analysis, diversity index, one-way ANOVA, and factor analysis. We found that tourism safety perception significantly affects the cognitive image, affective image, and conative image of the destination. Tourists with a high safety perception evaluate and affectively experience destination attributes more positively with higher satisfaction and stronger willingness to revisit and recommend. Tourism safety perception affects the stereotype image of the destination to a certain extent. Tourists in general produce a broadly homogeneous stereotype image, but there are differences in diversity and emotions. Tourists with a high safety perception have a richer and more positive stereotype image. This study enriches and deepens the theory regarding the influence of tourism safety perception on destination image, and also provides a richer theoretical basis for destination image construction and precision marketing. As the core components of abscisic acid (ABA) signal pathway, Clade A PP2C (PP2C-A) phosphatases in ABA-dependent stress responses have been well studied in Arabidopsis. However, the roles and natural variations of maize PP2C-A in stress responses remain largely unknown. In this study, we investigated the expression patterns of ZmPP2C-As treated with multiple stresses and generated transgenic Arabidopsis plants overexpressing most of the ZmPP2C-A genes. The results showed that the expression of most ZmPP2C-As were dramatically induced by multiple stresses (drought, salt, and ABA), indicating that these genes may have important roles in response to these stresses. Compared with wild-type plants, ZmPP2C-A1, ZmPP2C-A2, and ZmPP2C-A6 overexpression plants had higher germination rates after ABA and NaCl treatments. ZmPP2C-A2 and ZmPP2C-A6 negatively regulated drought responses as the plants overexpressing these genes had lower survival rates, higher leaf water loss rates, and lower proline accumulation compared to wild type plants. The natural variations of ZmPP2C-As associated with drought tolerance were also analyzed and favorable alleles were detected. We widely studied the roles of ZmPP2C-A genes in stress responses and the natural variations detected in these genes have the potential to be used as molecular markers in genetic improvement of maize drought tolerance.
