In fair weather, the vertical atmospheric electric field is oriented downward (positive in the earth surface ordinate system) in the global atmospheric circuit. Some researchers have revealed the unique phenomenon whereby once an upward vertical atmospheric electric field is observed in fair weather, an earthquake (EQ) follows within 2–48 h regardless of the EQ magnitude. However, the mechanism has not been explained with a suitable physical model. In this paper, a physical model is presented considering four types of forces acting on charged particles in the air. It is demonstrated that the heavier positive ions and lighter negative ions are rapidly separated. Finally, a reversed fair weather electrostatic field is formed by the above charge separation process. MicroRNA-146b-5p (miR-146b-5p) is up-regulated during and to suppress the inflammation process, although mechanisms involved in the action of miR-146b-5p have not been fully elucidated. This study examined the anti-inflammation effects of miR-146b-5p in lipopolysaccharide (LPS)-stimulated human dental pulp cells (hDPCs). An increase in human miR-146b-5p (hsa-miR-146b-5p) expression following the mRNA expression of pro-inflammatory cytokines was observed in LPS-stimulated hDPCs. The expression of hsa-miR-146b-5p and pro-inflammatory cytokines was down-regulated by a nuclear factor-kappa B (NF-κB) inhibitor, and the expression of hsa-miR-146b-5p was also decreased by a JAK1/2 inhibitor. Enforced expression of hsa-miR-146b-5p abolished phosphorylation of NF-κB p65 and down-regulated the expression of pro-inflammatory cytokines and NF-κB signaling components, such as interleukin-1 receptor-associated kinase 1 (IRAK1), tumor necrosis factor receptor-associated factor 6 (TRAF6), and REL-associated protein involved in NF-κB (RELA). Expression of rat miR-146b-5p (rno-miR-146b-5p) and pro-inflammatory cytokine mRNA was also up-regulated in experimentally-induced rat pulpal inflammation in vivo, and rno-miR-146b-5p blocked the mRNA expression of pro-inflammatory mediators and NF-κB signaling components in LPS-stimulated ex vivo cultured rat incisor pulp tissues. These findings suggest that the synthesis of miR-146b-5p is controlled via an NF-κB/IL6/STAT3 signaling cascade, and in turn, miR-146b-5p down-regulates the expression of pro-inflammatory mediators by targeting TRAF6, IRAK1, and RELA in LPS-stimulated hDPCs. Adaptive array processing technology for a phased array radar is usually based on the assumption of a stationary environment; however, in real-world scenarios, nonstationary interference and noise deteriorate the performance of the traditional gradient descent algorithm, in which the learning rate of the tap weights is fixed, leading to errors in the beam pattern and a reduced output signal-to-noise ratio (SNR). In this paper, we use the incremental delta-bar-delta (IDBD) algorithm, which has been widely used for system identification problems in nonstationary environments, to control the time-varying learning rates of the tap weights. The designed iteration formula for the learning rate ensures that the tap weights adaptively track the Wiener solution. The results of numerical simulations show that in a nonstationary environment, the traditional gradient descent algorithm with a fixed learning rate has a distorted beam pattern and reduced output SNR; however, the IDBD-based beamforming algorithm, in which a secondary control mechanism is used to adaptively update the learning rates, showed a similar beam pattern and output SNR to a traditional beamformer in a Gaussian white noise background; that is, the main beam and null satisfied the pointing constraints, and the optimal output SNR was obtained. Although the proposed algorithm contains a matrix inversion operation, which has considerable computational complexity, this operation could be replaced by the Levinson–Durbin iteration due to the Toeplitz characteristic of the matrix; therefore, the computational complexity could be decreased to O(n), so additional computing resources are not required. The atmospheric electric field is an important physical parameter used to study the state of charge density distribution between the earth and the ionosphere, and its daily variation characteristics reflect the underlying atmospheric electrical properties in a region. In this paper, we used atmospheric electric field data from three global observation stations (the Xanthi station, the Zhuanghe station, and the University of Reading station) from January to December 2021 and then combined these data with meteorological data and geomagnetic activity data to filter the fair-day atmospheric electric field data and plot the average fair-weather atmospheric electric field curve. Then, we compared different sunrise and sunset times and PM 2.5 concentrations, analyzed the seasonal differences for each station, the differences among the three stations, and the difference between their curves and the Carnegie curve, and finally concluded that the difference among them showed positive correlations with PM 2.5 concentrations. The other possible reasons and the entire shape of the average fair-weather atmospheric electric field curve for a defined station are not affected by the season, including the location of peaks and valleys, and the overall variation have been also revealed. Due to at least 3 years of cultivation, Panax ginseng (ginseng) is susceptible to being attacked by pathogens which severely affect its quality and yield. Compared with other diseases of ginseng, Phytophthora blight caused by Phytophthora cactorum (P. cactorum) can spread rapidly and destroy almost the entire plant of ginseng, such as leaves, stems, and roots. However, little research was focused on this area, and how P. cactorum affected the metabolic profile of ginseng is still obscure. In the current study, we conducted a comprehensive analysis of metabolomics and transcriptomics to compare the differences in health and P. cactorum-affected ginseng leaves and stems. Metabolome analysis revealed that 110 and 113 significant differential metabolites were observably disturbed separately in ginseng leaves and stems. Transcriptome analysis demonstrated that 6424 and 9508 genes had remarkable variation in ginseng leaves and stems. Using conjoint analysis, we also revealed the changes in pathways "Alanine, aspartate and glutamate metabolism", "Glycine, serine and threonine metabolism", and "Biosynthesis of unsaturated fatty acids" and "Plant hormone signal transduction" in ginseng response to the P. cactorum. The current work provides an overview of the alteration of metabolic profile and gene expression profiles in ginseng leaves and stems in response to P. cactorum affection, which may help to further screen out the mechanism of plant-pathogen interaction at the molecular level. An hourly scale precursor of inland earthquakes (EQs) is revealed in this paper. Several EQ cases in China have been reported. As indicated by a table listing 23 inland EQs and their shock time, epicenter location, magnitude, near-epicenter weather conditions, precursor start time and precursor duration, when the weather conditions are fair near the epicenter, an anomalously negative atmospheric electrostatic signal is readily observable approximately 2–48 h before the EQ occurs. Moreover, a successful single-station alarm for nearby moderate-magnitude EQs is demonstrated, and a possible mechanism for the precursor signal is proposed. The change in the electrostatic field during an EQ process is explained as the release of radioactive gases from the subsurface into the atmosphere via large (regional-scale) preexisting microfractures in the rock at the source depth. These gases considerably ionize the atmosphere, and the separated positive and negative ions establish a special macroscopic electric field. We aimed to explore the medium- and long-term (≥12 weeks) effects of dapagliflozin on serum uric acid (SUA) level in patients with type 2 diabetes mellitus (T2DM) in the real world study and to explore the influencing factors of dapagliflozin on reducing SUA level. This observational, prospective cohort study was based on the real world. There were 77 patients included in this study. They were divided into two groups. Patients in treatment group (n = 38) were treated as dapagliflozin 10 mg/d combined with therapy of routine glucose-lowering drugs (GLDs), and patients in the control group (n = 39) were treated with their routine GLDs. All measurements of physical examinations, blood, and urine samples, including age, sex, weight, height, systolic blood pressure (SBP), diastolic blood pressure (DBP), fasting blood glucose (FBG), glycosylated hemoglobin (HbA1c), and SUA, were collected at baseline for all patients in these two groups and repeated after 12, 24, and 48 weeks of therapy. We compared the changes of metabolic indicators including SUA in these two groups to evaluate the effects of dapagliflozin and analyzed its influencing factors. In the dapagliflozin group, mean SUA levels significantly decreased from 334.2 ± 99.1 μmol/L at baseline to 301.9 ± 73.2 μmol/L after 12 weeks therapy (t = 2.378, p = 0.023). There was no significant statistical difference of SUA levels after 24 weeks treatment of dapagliflozin compared with 12-week and 48-week treatment with dapagliflozin (p > 0.05). We found that baseline SUA had a significant impact on the effect of dapagliflozin on reducing SUA (OR 1.014, 95%CI 1.003–1.025, p = 0.014) by logistic regression analysis. Receiver operating characteristic (ROC) curve showed that T2DM patients with SUA level ≥ 314.5 μmol/L had relative accuracy in recognizing the good effects of dapagliflozin on reducing SUA (sensitivity 76.9%, specificity 76.2%). Combination therapy of dapagliflozin with routine blood-glucose-lowering drugs in T2DM patients showed the significant and sustained stable effect of lowering SUA level in this real-world study. In this study, 1-methylhydantoin cinnamic imides were synthesized from 1-methylhydantoin and trans-cinnamic acid, and their anti-inflammatory activity was investigated. The anti-inflammatory activity in vitro was evaluated by measuring the contents of NO, TNF-α and IL-1β in the supernatant of RAW264.7 cells stimulated by LPS. The cytotoxicity of 1-methylhydantoin cinnamoyl imides on RAW264.7 cells was detected using the CCK-8 method. The results showed that compounds 2 and 4 can significantly inhibit the release of NO and reduce the secretion of TNF-α and IL-1β. Compound 3 inhibited the production of TNF-α. The inhibition rate of COX was evaluated in vitro. The in vivo anti-inflammatory activities of the five compounds were evaluated by establishing an animal model of xylene ear swelling. The results showed that 1-methylhydantoin cinnamic imides could alleviate xylene-induced ear edema in mice in a dose-dependent manner. Among them, the effect of compound 5 was the most significant. Featured Application: The proposed thermal comfort-based personalized models can be applied into personalized heating and cooling system in open-plan office buildings. Heating, ventilation and air-conditioning (HVAC) systems play a key role in shaping the built environment. However, centralized HVAC systems cannot guarantee the provision of a comfortable thermal environment for everyone. Therefore, a personalized HVAC system that aims to adapt thermal preferences has drawn much more attention. Meanwhile, occupant-related factors like skin temperature have not had standardized measurement methods. Therefore, this paper proposes to use infrared thermography to develop individual thermal models to predict thermal sensations using three different feature sets with the random forest (RF) and support vector machine (SVM). The results have shown the correlation coefficients between clothing surface temperature and thermal sensation are 11% and 3% higher than those between skin temperature and thermal sensation of two subjects, respectively. With cross-validation, SVM with a linear kernel and penalty number of 1, as well as RF with 50 trees and the maximum tree depth of 3 were selected as the model configurations. As a result, the model trained with the feature set, consisting of indoor air temperature, relative humidity, skin temperature and clothing surface temperature, and with linear kernel SVM has achieved 100% recall score on test data of female subjects and 95% recall score on that of male subjects. As nutrition and a health tonic for both medicine and food, the protein content of Oviductus Ranae is more than 40%, making it an ideal source to produce antioxidant peptides. This work evaluated the effects of six different proteases (pepsin, trypsin, papain, flavourzyme, neutral protease and alcalase) on the antioxidant activity of Oviductus Ranae protein, and analyzed the relationship between the hydrolysis time, the degree of hydrolysis (DH) and the antioxidant activity of the enzymatic hydrolysates. The results showed that the antioxidant activity of Oviductus Ranae protein was significantly improved and the optimal hydrolysis time was maintained between 3–4 h under the action of different proteases. Among them, the protein hydrolysate which was hydrolyzed by pepsin for 180 min had the strongest comprehensive antioxidant activity and was most suitable for the production of antioxidant peptides. At this time, the DH, the DPPH radical scavenging activity, the absorbance value of reducing power determination and the hydroxyl radical scavenging activity corresponding to the enzymatic hydrolysate were 13.32 ± 0.24%, 70.63 ± 1.53%, 0.376 ± 0.009 and 31.96 ± 0.78%, respectively. Correlation analysis showed that there was a significant positive correlation between the hydrolysis time, the DH and the antioxidant activity of the enzymatic hydrolysates, further indicating that the hydrolysates of Oviductus Ranae protein had great antioxidant potential. The traditional anti-aging efficacy of Oviductus Ranae is closely related to the scavenging of reactive oxygen species, and its hydrolysates have better antioxidant capacity, which also provides support for further development of its traditional anti-aging efficacy. As one of the featured products in northeast China, Oviductus Ranae has been widely used as a nutritious food, which contains a variety of bioactive unsaturated fatty acids (UFAs). It is necessary to establish a scientific and reliable determination method of UFA contents in Oviductus Ranae. In this work, six principal UFAs in Oviductus Ranae, namely eicosapentaenoic acid (EPA), linolenic acid (ALA), docosahexaenoic acid (DHA), arachidonic acid (ARA), linoleic acid (LA) and oleic acid (OA), were identified using UPLC-MS/MS. The UFAs identified in Oviductus Ranae were further separated based on the optimized RP-HPLC conditions. Quantitative analysis of multi-components by single-marker (QAMS) method was implemented in content determination of EPA, ALA, DHA, ARA and OA, where LA was used as the internal standard. The experiments based on Taguchi design verified the robustness of the QAMS method on different HPLC instruments and chromatographic columns. The QAMS and external standard method (ESM) were used to calculate the UFA content of 15 batches of Oviductus Ranae samples from different regions. The relative error (r < 0.73%) and cosine coefficient showed that the two methods obtained similar contents, and the method validations met the requirements. The results showed that QAMS can comprehensively and effectively control the quality of UFAs in Oviductus Ranae which provides new ideas and solutions for studying the active components in Oviductus Ranae. Rana chensinensis ovum oil (RCOO) is an emerging source of unsaturated fatty acids (UFAs), but it is lacking in green and efficient extraction methods. In this work, using the response surface strategy, we developed a green and efficient CO2 supercritical fluid extraction (CO2-SFE) technology for RCOO. The response surface methodology (RSM), based on the Box–Behnken Design (BBD), was used to investigate the influence of four independent factors (pressure, flow, temperature, and time) on the yield of RCOO in the CO2-SFE process, and UPLC-ESI-Q-TOP-MS and HPLC were used to identify and analyze the principal UFA components of RCOO. According to the BBD response surface model, the optimal CO2-SFE condition of RCOO was pressure 29 MPa, flow 82 L/h, temperature 50 °C, and time 132 min, and the corresponding predicted optimal yield was 13.61%. The actual optimal yield obtained from the model verification was 13.29 ± 0.37%, and the average error with the predicted value was 0.38 ± 0.27%. The six principal UFAs identified in RCOO included eicosapentaenoic acid (EPA), α-linolenic acid (ALA), docosahexaenoic acid (DHA), arachidonic acid (ARA), linoleic acid (LA), and oleic acid (OA), which were important biologically active ingredients in RCOO. Pearson correlation analysis showed that the yield of these UFAs was closely related to the yield of RCOO (the correlation coefficients were greater than 0.9). Therefore, under optimal conditions, the yield of RCOO and principal UFAs always reached the optimal value at the same time. Based on the above results, this work realized the optimization of CO2-SFE green extraction process and the confirmation of principal bioactive ingredients of the extract, which laid a foundation for the green production of RCOO. Oviductus Ranae is a nutritional product for both medicine and food. Its quality is uneven due to multiple factors. An efficient method was established to evaluate the quality of Oviductus Ranae using fingerprint techniques and chemometric methods based on the analysis of polyunsaturated fatty acids (PUFAs) in petroleum ether extract by high performance liquid chromatography (HPLC). During this process, 27 batches of Oviductus Ranae were analyzed by HPLC and two types of chromatographic fingerprints were established. The fingerprint that was obtained by matching six known peaks was used for the quantification of six PUFAs. Another fingerprint was obtained by matching sixteen peaks with a peak area ratio greater than 0.5% and it was used to classify the different qualities of Oviductus Ranae by further combining three different chemometric models. The 27 batches of Oviductus Ranae were divided into four categories, which was consistent with the analysis results of six PUFAs contents. The two high-quality samples with significantly higher contents were classified into one category, and samples with medium contents were divided into two categories, including eight and thirteen samples, respectively. The four inferior samples with lower contents were classified into one category. Natural products play an important role in drug discovery. This work employed a natural product 1-methylhydantoin as the lead compound to develop novel dual-active drugs. 1-Methylhydantoin was isolated from Oviductus Ranae, which is a traditional Chinese medicine that has been used for tussive and inflammation treatment for a long time. An in silico study screened the more active 1-methylhydantoin derivatives. Antitussive assessment indicated that the newly synthesized agent had similar bioactivity with the natural product. An anti-inflammatory model used xylene induced ear edema model. At the same dosage (100 mg/Kg), the newly prepared agent had an inhibition rate 53.18% which was much higher than that of the lead compound (22.69%). The results might be ascribed to the cyclooxygenases-1 (COX-1) and cyclooxygenases-2 (COX-2) selectivity, and the fitness of the compound, and the binding pocket. The anti-particulate matter (PM 2.5) acute pneumonia was evaluated through an in vivo model constructed by nasal instillation with PM 2.5 suspension. The results of the above models suggested that this novel agent had remarkable antitussive, anti-inflammatory, and anti-PM 2.5 acute pneumonia activities. [ABSTRACT FROM AUTHOR]

This work demonstrated a method combining reversed-phase high-performance liquid chromatography (RP-HPLC) with chemometrics analysis to identify the authenticity of Ranae Oviductus. The fingerprint chromatograms of the Ranae Oviductus protein were established through an Agilent Zorbax 300SB-C8 column and diode array detection at 215 nm, using 0.085% TFA (v/v) in acetonitrile (A) and 0.1% TFA in ultrapure water (B) as mobile phase. The similarity was in the range of 0.779–0.980. The fingerprint chromatogram of Ranae Oviductus showed a significant difference with counterfeit products. Hierarchical clustering analysis (HCA) and principal component analysis (PCA) successfully identified Ranae Oviductus from the samples.
