Phthalic acid esters (PAEs), which are widespread environmental contaminants, can be efficiently biodegraded, mediated by enzymes such as hydrolases. Despite great advances in the characterization of PAE hydrolases, which are the most important enzymes in the process of PAE degradation, their molecular catalytic mechanism has rarely been systematically investigated. Acinetobacter sp. LUNF3, which was isolated from contaminated soil in this study, demonstrated excellent PAE degradation at 30 °C and pH 5.0–11.0. After sequencing and annotating the complete genome, the gene dphAN1, encoding a novel putative PAE hydrolase, was identified with the conserved motifs catalytic triad (Ser201-Asp295-His325) and oxyanion hole (H127GGG130). DphAN1 can hydrolyze DEP (diethyl phthalate), DBP (dibutyl phthalate) and BBP (benzyl butyl phthalate). The high activity of DphAN1 was observed under a wide range of temperature (10–40 °C) and pH (6.0–9.0). Moreover, the metal ions (Fe2+, Mn2+, Cr2+ and Fe3+) and surfactant TritonX-100 significantly activated DphAN1, indicating a high adaptability and tolerance of DphAN1 to these chemicals. Molecular docking revealed the catalytic triad, oxyanion hole and other residues involved in binding DBP. The mutation of these residues reduced the activity of DphAN1, confirming their interaction with DBP. These results shed light on the catalytic mechanism of DphAN1 and may contribute to protein structural modification to improve catalytic efficiency in environment remediation. [ABSTRACT FROM AUTHOR]

Yersinia enterocolitica (Y. enterocolitica) is an important food-borne and zoonotic pathogen. It can form biofilm on the surface of food, increasing the risk to food safety. Generally, outer membrane vesicles (OMVs) are spherical nanostructures secreted by Gram-negative bacteria during growth. They play a role in biological processes because they contain biologically active molecules. Several studies have reported that OMVs secreted by various bacteria are associated with the formation of biofilms. However, the interactions between Y. enterocolitica OMVs and biofilm are unknown. This study aims to investigate the effect of Y. enterocolitica OMVs on biofilm formation. Firstly, OMVs were extracted from Y. enterocolitica Y1083, which has a strong biofilm-forming ability, at 15 °C, 28 °C and 37 °C and then characterized. The characterization results showed differences in the yield and protein content of three types of OMVs. Next, by co-culturing the OMVs with Y. enterocolitica, it was observed that the OMVs inhibited the initial stage of Y. enterocolitica biofilm formation but did not affect the growth of Y. enterocolitica. Furthermore, biofilm formation by Salmonella enteritidis and Staphylococcus aureus were also inhibited by OMVs. Subsequently, it was proved that lipopolysaccharides (LPS) in OMVs inhibited biofilm formation., The proteins, DNA or RNA in OMVs could not inhibit biofilm formation. Bacterial motility and the expression of the biofilm-related genes pgaABC, motB and flhBD were inhibited by LPS. LPS demonstrated good anti-biofilm activity against various bacteria. The Qinghai–Tibet Plateau includes the Himalayas and Hengduan Mountains and is well known for its rich biodiversity. Evolutionary radiation is one of the main ways by which plants diversify in mountains, particularly the Qinghai–Tibet Plateau. It presents a large challenge to the classification of taxa that radiate quickly. One way to overcome these challenges is to continue conducting detailed field studies while integrating morphological and molecular evidence to classify these taxa. The aim of this research was to provide a case for the systematic study of the complex taxa Rhodiola, which rapidly radiate. During the field study, we found two unique variants of Rhodiola in an alpine dry meadow and beds of pebbles on beaches, respectively. We utilized a morphological principal component analysis, scanning electron microscopy and molecular phylogenetic analysis to propose two new species: Rhodiola wangii S.Y. Meng and Rhodiola namlingensis S.Y. Meng. R. wangii is similar to R. stapfii (Hamet) S.H. Fu, but it differs in having an intensely broad rhombus and alternate leaves, a distinct petiole, stamens gathered together and reflexed purple scales. R. namlingensis is similar to R. prainii (Hamet) H. Ohba, but it differs in its exerted alternate leaves, the presence of more than four leaves on the stem, thick leaf blades, an obovate to inverted triangle, and short petioles. In order to provide a simplified and low-cost solution of the terminal for a distributed actuation system, this paper proposes an electro-hydrostatic actuator (EHA) based on the linear drive principle. The proposed actuator is directly driven by a linear pump with a collaborative rectification mechanism, whose performance relies on the collaboration of the internal two units. A pair of linear oscillating motors are employed to drive the two pump units respectively. The control of the actuator is based on the modulation of the oscillating amplitude, frequency, and phase difference of the two motors. The advantage of this actuator is that no more valve control is needed to rectify the linear pump besides the high efficiency of the direct pump drive. In this paper, both schematic and detailed structure of the actuator is presented. The kinematic and dynamic characteristics are analyzed and modeled, based on which the control method is proposed. PEMFC are considered to be the most promising for automotive energy because of their good working effect, low temperature, high efficiency, and zero pollution. Stainless steel as a PEMFC bipolar plate has unparalleled advantages in strength, cost, and processability, but it is easy to corrode in a PEMFC working environment. In order to improve the corrosion resistance, the surface modification of 316L stainless steel is a feasible solution for PEMFC bipolar plates. In the present study, the plasma-nitrided coating and CrNx coating were prepared by the plasma-enhanced balanced magnetron sputtering technology on the 316L stainless steel surface. The microstructures, phase compositions, and corrosion resistance behavior of the coatings were investigated. The corrosion behavior of the prepared plasma-nitrided coating and CrNx coating was investigated by potentiodynamic polarization, potentiostatic polarization, and electrochemical impedance spectroscopy (EIS) in both cathodic and anodic environments. The experimental results show that corrosion resistance of the CrNx coating was better than the plasma-nitrided coating. Mechanical properties of Q370qE carbon steel can be improved by cladding it with 316L stainless steel. After rolling these materials together, microstructure, hardness, shear strength, and corrosion properties of the cladded metals were evaluated. Hardness and shear tests were performed according to appropriate standards to evaluate the bonds. The results show that the remarkable diffusion of Ni and Cr formed a hard transition zone. The width of this diffusion zone increases with increasing temperature. The shear strength of the clad interface reached a minimum of 385 MPa. In addition, the surfaces of samples were examined by neutral salt spray (NSS) tests and potentiodynamic polarization tests to examine corrosion behavior. The 316L side primarily exhibited pitting corrosion, while the Q370qE side was dominated by uniform corrosion. [ABSTRACT FROM AUTHOR]

As major component in cereals grains, starch has been one of the most important carbohydrate consumed by a majority of world's population. However, the molecular mechanism for regulation of biosynthesis of starch remains elusive. In the present study, ZmES22, encoding a MADS-type transcription factor, was modestly characterized from maize inbred line B73. ZmES22 exhibited high expression level in endosperm at 10 days after pollination (DAP) and peaked in endosperm at 20 DAP, indicating that ZmES22 was preferentially expressed in maize endosperm during active starch synthesis. Transient expression of ZmES22 in tobacco leaf revealed that ZmES22 protein located in nucleus. No transactivation activity could be detected for ZmES22 protein via yeast one-hybrid assay. Transformation of overexpressing plasmid 35S::ZmES22 into rice remarkedly reduced 1000-grain weight as well as the total starch content, while the soluble sugar was significantly higher in transgenic rice lines. Moreover, overexpressing ZmES22 reduced fractions of long branched starch. Scanning electron microscopy images of transverse sections of rice grains revealed that altered expression of ZmES22 also changed the morphology of starch granule from densely packed, polyhedral starch granules into loosely packed, spherical granules with larger spaces. Furthermore, RNA-seq results indicated that overexpressing ZmES22 could significantly influence mRNA expression levels of numerous key regulatory genes in starch synthesis pathway. Y1H assay illustrated that ZmES22 protein could bind to the promoter region of OsGIF1 and downregulate its mRNA expression during rice grain filling stages.
