Macrobrachium rosenbergii is a warm water species, and low temperature is a limiting factor for its growth and survival. In order to explore the role of the acetyl-CoA-carboxylase (ACC) gene in response to the cold stress of M. rosenbergii, we investigated the effects of RNA interference (RNAi) with the ACC gene on the expression of fatty acid metabolism-related genes and the mortality of M. rosenbergii under cold stress. The results showed that different siRNA sequences and different injection concentrations had different inhibiting effects on ACC gene expression, and siRNA-III with an injection concentration of 2.0 μg/g (siRNA/prawn body weight) had the best interference effect. With the optimal siRNA and the optimal concentration under cold stress, the expressions of three fatty acid metabolism-related genes, FabD, echA, and ACOT, were generally significantly down-regulated. Compared to negative (scrambled-siRNA) and blank (PBS) control groups, the expression of FabD in the interference group was extremely significantly down-regulated at 12 h in the hepatopancreas and at 18 h in the muscles and gills; EchA was highly significantly down-regulated at 6 and 12 h in the muscles and gills; and ACOT was extremely significantly down-regulated and kept declining in the gills. Within 6–18 h after injection under cold stress, the mortality rate of the siRNA interference group (75%) was much lower than that of the negative (95%) or blank control group (97.5%), and all prawns died after 24 h. In conclusion, RNA interference with the ACC gene inhibited the expression of some fatty acid metabolism-related genes, and could partly improve the tolerance of M. rosenbergii to cold stress, indicating that the ACC gene might play an important role in the response of M. rosenbergii to cold stress. Surface features are crucial for assessing welding quality because they serve as an intuitive depiction of the quality of the joint and have a major influence on welding strength. According to the characteristics of the refill friction stir spot welding (RFSSW) process and an analysis of the surface-state and internal morphology of RFSSW joints, a method of predicting the mechanical properties of RFSSW joints based on surface-state characteristics was proposed. In this paper, a laser-ranging sensor was used to characterize the surface state of RFSSW joints, and parametric characterization methods of the surface-state features of RFSSW joints were proposed. On this basis, a support vector machine was used to predict and analyze the fracture mode of RFSSW joints. The accuracy of the analysis of the test samples reached 95.8%. For covert communication in lossy channels, it is necessary to consider that the carrier of the hidden watermark will undergo multiple image-processing attacks. In order to ensure that secret information can be extracted without distortion from the watermarked images that have undergone attacks, in this paper, we design a novel fragmented secure communication system. The sender will fragment the secret data to be transmitted and redundantly hide it in a large number of multimodal carriers of messenger accounts on multiple social platforms. The receiver receives enough covert carriers, extracts each fragment, and concatenates the transmitted secret data. This article uses the image carrier as an example to fragment the text file intended for transmission and embeds it into a large number of images, with each fragment being redundant and embedded into multiple images. In this way, at the receiving end, only enough stego images need to be received to extract the information in each image, and then concatenate the final secret file. In order to resist various possible attacks during image transmission, we propose a strong robust image watermarking method. This method adopts a watermark layer based on DFT, which has high embedding and detection efficiency and good invisibility. Secondly, a watermark layer based on DCT is adopted, which can resist translation attacks, JPEG attacks, and other common attacks. Experiments have shown that our watermarking method is very fast; both the embedding time and the extraction time are less than 0.15 s for images not larger than 2000 × 2000 . Our watermarking method has very good invisibility with 41 dB PSNR on average. And our watermarking method is more robust than existing schemes and robust to nearly all kinds of attacks. Based on this strong robust image watermarking method, the scheme of fragmenting and hiding redundant transmission content into a large number of images is effective and practical. Our scheme can 100% restore the secret file completely under different RST or hybrid attacks, such as rotation by 1 degree and 5 degrees, scaling by 1.25 and 0.8, and cropping by 10% and 25%. Our scheme can successfully restore the secret file completely even if 30% of received images are lost. When 80% of received images are lost, our scheme can still restore 61.1% of the secret file. Market uncertainty has a significant impact on market performance. Previous studies have dedicated much effort towards investigations into market uncertainty related to information asymmetry and risk. However, they have neglected the uncertainty inherent in market transactions, which is also an important aspect of market performance, besides the quantity of transactions and market efficiency. In this paper, we put forward a concept of transaction entropy to measure market uncertainty and see how it changes with price. Transaction entropy is defined as the ratio of the total information entropy of all traders to the quantity of transactions, reflecting the level of uncertainty in making successful transactions. Based on the computational and simulated results, our main finding is that transaction entropy is the lowest at equilibrium, it will decrease in a shortage market, and increase in a surplus market. Additionally, we make a comparison of the total entropy of the centralized market with that of the decentralized market, revealing that the price-filtering mechanism could effectively reduce market uncertainty. Clinically, early brain injury (EBI), which refers to the acute injuries to the whole brain in the phase of the first 72 h following subarachnoid hemorrhage (SAH), is intensely investigated to improve neurological and psychological function. Additionally, it will be meaningful to explore new therapeutic approaches for EBI treatment to improve the prognosis of patients with SAH. To investigate the underlying neuroprotection mechanism in vitro, the Protein tyrosine phosphatase 1B inhibitor (PTP1B-IN-1) was put in primary neurons induced by OxyHb to observe neuroapoptosis, neuroinflammation, and ER stress. Then, one hundred forty male mice were subjected to Experiment two and Experiment three. The mice in the SAH24h + PTP1B-IN-1 group were given an intraperitoneal injection of 5 mg/kg PTP1B-IN-1 30 min before anesthesia. SAH grade, neurological score, brain water content, Western blot, PCR, and Transmission Electron Microscopy (TEM) were performed to observe the underlying neuroprotection mechanism in vivo. Overall, this study suggests that PTP1B-IN-1 could ameliorate neuroapoptosis, neuroinflammation, and ER stress in vitro and in vivo by regulating the IRS-2/AKT signaling pathway, suggesting that PTP1B-IN-1 may be a candidate drug for the treatment of early brain injury after SAH. Protected cropping produces more food per land area than field-grown crops. Protected cropping includes low-tech polytunnels utilizing protective coverings, medium-tech facilities with some environmental control, and high-tech facilities such as fully automated glasshouses and indoor vertical farms. High crop productivity and quality are maintained by using environmental control systems and advanced precision phenotyping sensor technologies that were first developed for broadacre agricultural and can now be utilized for protected-cropping applications. This paper reviews the state of the global protected-cropping industry and current precision phenotyping methodology and technology that is used or can be used to advance crop productivity and quality in a protected growth environment. This review assesses various sensor technologies that can monitor and maintain microclimate parameters, as well as be used to assess plant productivity and produce quality. Protected cropping offers a way to bolster food production in the face of climate change and deliver healthy food sustainably with fewer resources. However, to make this way of farming economically viable, we need to consider the status of protected cropping in the context of available technologies and corresponding target horticultural crops. This review outlines existing opportunities and challenges that must be addressed by ongoing research and innovation in this exciting but complex field in Australia. Indoor farm facilities are broadly categorised into the following three levels of technological advancement: low-, medium- and high-tech with corresponding challenges that require innovative solutions. Furthermore, limitations on indoor plant growth and protected cropping systems (e.g., high energy costs) have restricted the use of indoor agriculture to relatively few, high value crops. Hence, we need to develop new crop cultivars suitable for indoor agriculture that may differ from those required for open field production. In addition, protected cropping requires high start-up costs, expensive skilled labour, high energy consumption, and significant pest and disease management and quality control. Overall, protected cropping offers promising solutions for food security, while reducing the carbon footprint of food production. However, for indoor cropping production to have a substantial positive impact on global food security and nutritional security, the economical production of diverse crops will be essential. Simple Summary: This review focuses on plant growth and development harmed by abiotic stress, primarily salt stress. Salt stress raises the intracellular osmotic pressure, leading to hazardous sodium buildup. Plants react to salt stress signals by regulating ion homeostasis, activating the osmotic stress pathway, modulating plant hormone signaling, and altering cytoskeleton dynamics and cell wall composition. Understanding the processes underlying these physiological and biochemical responses to salt stress could lead to more effective agricultural crop yield measures. In this review, researchers outline recent advances in plant salt stress control. The study of plant salt tolerance processes is essential, both theoretically and practically, to improve agricultural output, produce novel salt-tolerant cultivars, and make full use of saline soil. Based on past research, this paper discusses the adverse effects of salt stress on plants, including photosynthesis suppression, ion homeostasis disturbance, and membrane peroxidation. The authors have also covered the physiological mechanisms of salt tolerance, such as the scavenging of reactive oxygen species and osmotic adjustment. This study further identifies specific salt stress-responsive mechanisms linked to physiological systems. Based on previous studies, this article reviews the current methodologies and techniques for improving plant salt tolerance. Overall, it is hoped that the above-mentioned points will impart helpful background information for future agricultural and crop plant production. Salinity is significant abiotic stress that affects the majority of agricultural, irrigated, and cultivated land. It is an issue of global importance, causing many socio-economic problems. Salt stress mainly occurs due to two factors: (1) soil type and (2) irrigation water. It is a major environmental constraint, limiting crop growth, plant productivity, and agricultural yield. Soil salinity is a major problem that considerably distorts ecological habitats in arid and semi-arid regions. Excess salts in the soil affect plant nutrient uptake and osmotic balance, leading to osmotic and ionic stress. Plant adaptation or tolerance to salinity stress involves complex physiological traits, metabolic pathways, the production of enzymes, compatible solutes, metabolites, and molecular or genetic networks. Different plant species have different salt overly sensitive pathways and high-affinity K+ channel transporters that maintain ion homeostasis. However, little progress has been made in developing salt-tolerant crop varieties using different breeding approaches. This review highlights the interlinking of plant morpho-physiological, molecular, biochemical, and genetic approaches to produce salt-tolerant plant species. Most of the research emphasizes the significance of plant growth-promoting rhizobacteria in protecting plants from biotic and abiotic stressors. Plant growth, survival, and yield can be stabilized by utilizing this knowledge using different breeding and agronomical techniques. This information marks existing research areas and future gaps that require more attention to reveal new salt tolerance determinants in plants—in the future, creating genetically modified plants could help increase crop growth and the toleration of saline environments. Alginate is derived from brown algae, which can be cultivated in large quantities. It can be broken down by alginate lyase into alginate oligosaccharides (AOSs), which exhibit a higher added value and better bioactivity than alginate. In this study, metagenomic technology was used to screen for genes that code for high-efficiency alginate lyases. The candidate alginate lyase gene alg169 was detected from Psychromonas sp. SP041, the most abundant species among alginate lyase bacteria on selected rotten kelps. The alginate lyase Alg169 was heterologously expressed in Escherichia coli BL21 (DE3), Ni-IDA-purified, and characterized. The optimum temperature and pH of Alg169 were 25 °C and 7.0, respectively. Metal ions including Mn2+, Co2+, Ca2+, Mg2+, Ni2+, and Ba2+ led to significantly increased enzyme activity. Alg169 exhibited a pronounced dependence on Na+, and upon treatment with Mn2+, its activity surged by 687.57%, resulting in the highest observed enzyme activity of 117,081 U/mg. Bioinformatic analysis predicted that Alg169 would be a double-domain lyase with a molecular weight of 65.58 kDa. It is a bifunctional enzyme with substrate specificity to polyguluronic acid (polyG) and polymannuronic acid (polyM). The kidney plays a crucial role in glucose homeostasis by regulating glucose transport. We aimed to investigate the impact of alterations in glucose transport on glucose metabolism during ageing. Adult male Sprague Dawley rats were divided into five groups: 3-month, 6-month, and 12-month control groups, and 6- and 12-month groups receiving the hydrogen sulfide donor molecule GYY4137. The study found that, as age increased, daily urinary uric acid and protein levels increased in the 12-month group. Blood sugar level and HOMA-IR index increased in the 12-month group, and were partially improved by GYY4137. The kidney tissue showed mild glomerulosclerosis in the 12-month group, which was diminished by GYY4137. Gene expression analysis showed decreased sirtuin and increased p21 expression in the aging groups. Increased SGLT1 and SGLT2 expression was observed in the 12-month group, which was reversed by GYY4137. Both GLUT1 and GLUT2 expression was increased in the 6- and 12-month groups, and reversed by GYY4137 in the 12-month group. The study concluded that aging was associated with increased blood sugar levels and the HOMA-IR index, and the abundance of renal glucose transporters increased as aging progressed. Nowadays robots have been commonly adopted in various manufacturing industries to improve product quality and productivity. The selection of the best robot to suit a specific production setting is a difficult decision making task for manufacturers because of the increase in complexity and number of robot systems. In this paper, we explore two key issues of robot evaluation and selection: the representation of decision makers' diversified assessments and the determination of the ranking of available robots. Specifically, a decision support model which utilizes cloud model and TODIM (an acronym in Portuguese of interactive and multiple criteria decision making) method is developed for the purpose of handling robot selection problems with hesitant linguistic information. Besides, we use an entropy-based combination weighting technique to estimate the weights of evaluation criteria. Finally, we illustrate the proposed cloud TODIM approach with a robot selection example for an automobile manufacturer, and further validate its effectiveness and benefits via a comparative analysis. The results show that the proposed robot selection model has some unique advantages, which is more realistic and flexible for robot selection under a complex and uncertain environment. Several environmental stresses, including biotic and abiotic factors, adversely affect the growth and development of crops, thereby lowering their yield. However, abiotic factors, e.g., drought, salinity, cold, heat, ultraviolet radiations (UVr), reactive oxygen species (ROS), trace metals (TM), and soil pH, are extremely destructive and decrease crop yield worldwide. It is expected that more than 50% of crop production losses are due to abiotic stresses. Moreover, these factors are responsible for physiological and biochemical changes in plants. The response of different plant species to such stresses is a complex phenomenon with individual features for several species. In addition, it has been shown that abiotic factors stimulate multi-gene responses by making modifications in the accumulation of the primary and secondary metabolites. Metabolomics is a promising way to interpret biotic and abiotic stress tolerance in plants. The study of metabolic profiling revealed different types of metabolites, e.g., amino acids, carbohydrates, phenols, polyamines, terpenes, etc, which are accumulated in plants. Among all, primary metabolites, such as amino acids, carbohydrates, lipids polyamines, and glycine betaine, are considered the major contributing factors that work as osmolytes and osmoprotectants for plants from various environmental stress factors. In contrast, plant-derived secondary metabolites, e.g., phenolics, terpenoids, and nitrogen-containing compounds (alkaloids), have no direct role in the growth and development of plants. Nevertheless, such metabolites could play a significant role as a defense by protecting plants from biotic factors such as herbivores, insects, and pathogens. In addition, they can enhance the resistance against abiotic factors. Therefore, metabolomics practices are becoming essential and influential in plants by identifying different phytochemicals that are part of the acclimation responses to various stimuli. Hence, an accurate metabolome analysis is important to understand the basics of stress physiology and biochemistry. This review provides insight into the current information related to the impact of biotic and abiotic factors on variations of various sets of metabolite levels and explores how primary and secondary metabolites help plants in response to these stresses. The effect of Ti and B microalloying on the hardenability, prior austenite grain size (PAGS), mechanical properties, and sulfide stress cracking (SSC) of C110 grade steel was studied by Jominy testing, static tensile testing, an optical microscope (OM), scanning electron microscopy (SEM) and double cantilever beam (DCB) testing. The results show that the addition of 0.015% Ti and 0.002% B into a medium-carbon Fe-Cr-Mo-Nb-V steel increased the hardenability and refined the PAGS and quenched martensite packets, and the size of carbides was reduced. The present study emphasizes the efficacy of a biosurfactant-producing bacterial strain Klebsiella sp. KOD36 in biodegradation of azo dyes and hexavalent chromium individually and in a simultaneous system. The bacterial strain has exhibited a considerable potential for biodegradation of chromium and azo dyes in single and combination systems (maximum 97%, 94% in an individual and combined system, respectively). Simultaneous aerobic biodegradation of azo dyes and hexavalent chromium (SBAHC) was modeled using machine learning programming, which includes gene expression programming, random forest, support vector regression, and support vector regression-fruit fly optimization algorithm. The correlation coefficient includes the dispersion index, and the Willmott agreement index was employed as statistical metrics to assess the performance of each model separately. In addition, the Taylor diagram was used to further investigate the methods used. The findings of the present study were that the support vector regression-fruitfly optimization algorithm (SVR-FOA) with correlation coefficient (CC) of 0.644, (scattered index) SI of 0.374, and (Willmott's index of agreement) WI of 0.607 performed better than the autonomous support vector regression (SVR), gene expression programming (GEP), and random forest (RF) methods. In addition, the standalone SVR model with CC of 0.146, SI of 0.473, and WI of 0.408 ranked the second best. In summary, the SBAHC can be accurately estimated using the hybrid SVR-FOA method. The domain of unknown function (DUF) superfamily encodes proteins of unknown functions in plants. Among them, DUF668 family members in plants possess a 29 amino-acid conserved domain, and this family has not been described previously. Here, we report this plant-specific novel DUF668 gene family containing 12 OsDUF668 genes in rice (Oryza sativa) and 91 DUF668s for the other seven plant species. In our study, DUF668 genes were present in both dicot and monocot plants, indicating that DUF668 is a conserved gene family that originated by predating the dicot–monocot divergence. Based on the gene structure and motif composition, the DUF668 family consists of two distinct clades, I and II in the phylogenetic tree. Remarkably, OsDUF668 genes clustered on the chromosomes merely show close phylogenetic relationships, suggesting that gene duplications or collinearity seldom happened. Cis-elements prediction display that over 80% of DUF668s contain phytohormone and light responsiveness factors. Further comprehensive experimental analyses of the OsDUF668 family are implemented in 22 different tissues, five hormone treatments, seven environmental factor stresses, and two pathogen-defense related stresses. The OsDUF668 genes express ubiquitously in analyzed rice tissues, and seven genes show tissue-specific high expression profiles. All OsDUF668s respond to drought, and some of Avr9/Cf-9 rapidly elicited genes resist to salt, wound, and rice blast with rapidly altered expression patterns. These findings imply that OsDUF668 is essential for drought-enduring and plant defense. The projected increase of the world's population, coupled with the shrinking area of arable land required to meet future food demands, is building pressure on Earth's finite agricultural resources. As an alternative to conventional farming methods, crops can be grown in protected environments, such as traditional greenhouses or the more modern plant factories. These are usually more productive and use resources more efficiently than conventional farming and are now receiving much attention—especially in urban and peri-urban areas. Traditionally, protected cropping has been predominantly practised in temperate climates, but interest is rapidly rising in hot, arid areas and humid, tropical regions. However, maintaining suitable climatic conditions inside protected cropping structures in warm climates—where warm is defined as equivalent to climatic conditions that require cooling—is challenging and requires different approaches from those used in temperate conditions. In this paper, we review the benefits of protected cropping in warm climates, as well as the technologies available for maintaining a controlled growing environment in these regions. In addition to providing a summary of active cooling methods, this study summarises photovoltaic (PV)-based shading methods used for passive cooling of greenhouses. Additionally, we also summarise the current humidity-control techniques used in the protected cropping industry and identify future research opportunities in this area. Furfuryl alcohol (FA) is an important building block in polymer, food, and pharmaceutical industries. In this work, we reported the biocatalytic reduction of furfural, one of the top value-added bio-based platform chemicals, to FA by immobilized Meyerozyma guilliermondii SC1103 cells. The biocatalytic process was optimized, and the tolerance of this yeast strain toward toxic furfural was evaluated. It was found that furfural of 200 mM could be reduced smoothly to the desired product FA with the conversion of 98% and the selectivity of >98%, while the FA yield was only approximately 81%. The gap between the substrate conversion and the product yield might partially be attributed to the substantial adsorption of the immobilization material (calcium alginate) toward the desired product, but microbial metabolism of furans (as carbon sources) made a negligible contribution to it. In addition, FA of approximately 156 mM was produced within 7 h in a scale-up reaction, along with the formation of trace 2-furoic acid (1 mM) as the byproduct. The FA productivity was up to 2.9 g/L/h, the highest value ever reported in the biocatalytic synthesis of FA. The crude FA was simply separated from the reaction mixture by organic solvent extraction, with the recovery of 90% and the purity of 88%. Recently, extensive research has been done on evolute curves in Minkowski space-time. However, the special characteristics of curves demand advanced level observations that are lacking in existing well-known literature. In this study, a special kind of generalized evolute and involute curve is considered in four-dimensional Minkowski space. We consider (1,3)-evolute curves with respect to the casual characteristics of the (1,3)-normal plane that are spanned by the principal normal and the second binormal of the vector fields and the (0,2)-evolute curve that is spanned by the tangent and first binormal of the given curve. We restrict our investigation of (1,3)-evolute curves to the (1,3)-normal plane in four-dimensional Minkowski space. This research contribution obtains a necessary and sufficient condition for the curve possessing the generalized evolute as well as the involute curve. Selective photodynamic therapy (PDT) for cancer cells is more efficient and much safer. Most selective PDTs are realized by antigene-biomarker or peptide-biomarker interactions. Here, we modified dextran with hydrophobic cholesterol as a photosensitizer carrier to selectively target cancer cells, including colon cancer cells, and fulfilled selective PDT. The photosensitizer was designed with regular Aggregation-Induced Emission (AIE) units, including triphenylamine and 2-(3-cyano-4,5,5-trimethylfuran-2-ylidene)propanedinitrile. The AIE units can help to decrease the quenching effect in the aggregate state. The efficiency of the photosensitizer is further improved via the heavy atom effect after bromination modification. We found that the obtained photosensitizer nanoparticles could selectively target and ablate cancer cells after encapsulation into the dextran-cholesterol carrier. Simple Summary: Endothelial cell senescence has been considered as an initiation in the progress of vascular aging leading to the advancement of cardiovascular diseases, while the mechanism of endothelial cell senescence remains elusive. This study aimed to investigate the critical role of cathepsinL/V in endothelial cell senescence. We found that cathepsinL/V was decreased in senescent endothelial cells, which enhanced aldehyde dehydrogenase 1 family member A2 (ALDH1A2) expression and activated AKT/ERK1/2-P21 pathway, and therefore promoted cellular senescence, which may play an important role in vascular aging. These findings suggest that cathepsinL/V may be a potential therapeutic target in endothelial cell senescence. The senescence of vascular endothelial cells (ECs) is characterized as a hallmark of vascular aging, which leads to the initiation, progress, and advancement of cardiovascular diseases. However, the mechanism of the ECs senescence remains elusive. In this study, thoracic aortas were separated from young (8-week-old) and aged (18-month-old) mice. Decreased Ctsl expression and increased vascular remodeling were observed in senescent aorta. H2O2 was used to induce human umbilical vein endothelial cells (HUVECs) senescence, as shown by increased SA-β-gal positive cells and upregulated p21 level. CTSV significantly decreased after H2O2 treatment, while over-expression of CTSV by adenovirus reduced cellular senescence. RNA sequencing analysis was conducted subsequently, and ALDH1A2 was observed to significantly increased in H2O2 group and decreased after over-expression of CTSV. This result was further confirmed by RT-PCR and WB. Moreover, over-expression of CTSV reduced the increase of ERK1/2 and AKT phosphorylation induced by H2O2. Additionally, retinoic acid (RA), the major production of ALDH1A2, was added to CTSV over-expressed senescent HUVECs. Administration of RA activated AKT and ERK1/2, induced the expression of p21, and enhanced SA-β-gal positive cells, while not affecting the expression of CTSV and ALDH1A2. These results were further confirmed in doxorubicin (DOX)-induced senescent ECs. In conclude, we have identified that Ctsl/CTSV plays a key role in ECs senescence by regulating ALDH1A2 to activate AKT/ ERK1/2-P21 pathway. Simple Summary: Schwann cells (SCs) have been reported to support tumor spreading and metastasis formation in both the in vitro and in vivo models. However, the role of SCs in the progression of small-cell lung cancer (SCLC) has not been investigated. This study demonstrated the crosstalk between SCLC and tumor-associated SCs (TA-SCs) and constructed the mRNA-miRNA-lncRNA network that may regulate tumor cell-SC interaction and cancer progression. Small-cell lung cancer (SCLC), representing 15–20% of all lung cancers, is an aggressive malignancy with a distinct natural history, poor prognosis, and limited treatment options. We have previously identified Schwann cells (SCs), the main glial cells of the peripheral nervous system, in tumor tissues and demonstrated that they may support tumor spreading and metastasis formation in the in vitro and in vivo models. However, the role of SCs in the progression of SCLC has not been investigated. To clarify this issue, the cell proliferation assay, the annexin V apoptosis assay, and the transwell migration and invasion assay were conducted to elucidate the roles in SCLC of tumor-associated SCs (TA-SCs) in the proliferation, apoptosis, migration, and invasion of SCLC cells in vitro, compared to control group. In addition, the animal models to assess SC action's effects on SCLC in vivo were also developed. The result confirmed that TA-SCs have a well-established and significant role in facilitating SCLC cell cancer migration and invasion of SCLC in vitro, and we also observed that SC promotes tumor growth of SCLC in vivo and that TA-SCs exhibited an advantage and show a repair-like phenotype, which allowed defining them as tumor-associated repair SCs (TAR-SCs). Potential molecular mechanisms of pro-tumorigenic activity of TAR-SCs were investigated by the screening of differentially expressed genes and constructing networks of messenger-, micro-, and long- non-coding RNA (mRNA-miRNA-lncRNA) using DMS114 cells, a human SCLC, stimulated with media from DMS114-activated SCs, non-stimulated SCs, and appropriate controls. This study improves our understanding of how SCs, especially tumor-activated SCs, may promote SCLC progression. Simple Summary: Immune checkpoint inhibitors (ICIs) have made an indelible mark on metastatic colorectal cancer patients with microsatellite instability-high (MSI-H) instead of microsatellite stability (MSS) tumors. Clinical studies have explored the introduction of ICIs into treatment for locally advanced rectal cancer patients with MSS; however, the outcomes are still limited. Different studies have provided valuable information for understanding the expression and clinical significance of PD-1 and PD-L1 during neoadjuvant chemoradiotherapy (nCRT). However, the value of CTLA-4 and CD86 after nCRT has not been fully studied. We found that CD86 expression was significantly lower in the nCRT-treated group rather than CTLA-4. Improved overall survival in patients with lower CD86 expression could only be observed in patients with low rather than high t/sCD8+ cell densities. Our findings reveal the relationship between the immunosuppressive microenvironment and the CTLA-4/CD86 pathway after nCRT. The synergistic effect of combining immune checkpoint inhibitors (ICIs) with neoadjuvant chemo(radio)therapy (nCRT) in colorectal cancer is still limited. We aimed to understand the impact of nCRT on the tumor microenvironment and to explore favorable immune markers of this combination. Herein, we investigated the expression of cytotoxic T lymphocyte-associated antigen 4 (CTLA-4), CD86, CD4, and CD8 after nCRT and its association with clinicopathological characteristics. Immunostaining of immune-related molecules was performed in 255 surgically resected specimens from rectal cancer patients treated with nCRT. CD4 and CD8 expression on the tumor (tCD4/CD8), stroma (sCD4/CD8), and invasive front (iCD4/CD8) was evaluated. The expression levels of immune-related molecules were significantly lower in the nCRT-treated group, except for CTLA-4 and sCD8. However, patients with higher sCD8+ cell density and CTLA-4 expression had better progression-free survival (PFS) and distant metastasis-free survival (DMFS). In addition, higher CD86 expression was associated with poorer overall survival (OS). Higher CTLA-4 expression was associated with higher tCD8+ cell density, whereas CD86 expression was correlated with the cell density of t/sCD8. Prognostic analysis confirmed that the relationships between CTLA-4 and DMFS as well as CD86 and OS were significantly correlated in low rather than high CD8+ cell density. Further the combination of CD8+ cell density and CD86 expression was shown to be an independent prognostic factor of OS, whereas the combination of CTLA-4 was not for DMFS. Together, these results demonstrate significant correlations between CD86 expression and t/sCD8+ cell density in rectal cancer after nCRT and could potentially have clinical implications for combining ICIs and nCRT. The proliferation of massive polarimetric Synthetic Aperture Radar (SAR) data helps promote the development of SAR image interpretation. Due to the advantages of powerful feature extraction capability and strong adaptability for different tasks, deep learning has been adopted in the work of SAR image interpretation and has achieved good results. However, most deep learning methods only employ single-polarization SAR images and ignore the water features embedded in multi-polarization SAR images. To fully exploit the dual-polarization SAR data and multi-scale features of SAR images, an effective flood detection method for SAR images is proposed in this paper. In the proposed flood detection method, a powerful Multi-Scale Deeplab (MS-Deeplab) model is constructed based on the dual-channel MobileNetV2 backbone and the classic DeeplabV3+ architecture to improve the ability of water feature extraction in SAR images. Firstly, the dual-channel feature extraction backbone based on the lightweight MobileNetV2 separately trains the dual-polarization SAR images, and the obtained training parameters are merged with the linear weighting to fuse dual-polarization water features. Given the multi-scale space information in SAR images, then, a multi-scale feature fusion module is introduced to effectively utilize multi-layer features and contextual information, which enhances the representation of water features. Finally, a joint loss function is constructed based on cross-entropy and a dice coefficient to deal with the imbalanced categorical distribution in the training dataset. The experimental results on the time series of Sentinel-1A SAR images show that the proposed method for flood detection has a strong ability to locate water boundaries and tiny water bodies in complex scenes. In terms of quantitative assessment, MS-Deeplab can achieve a better performance compared with other mainstream semantic segmentation models, including PSPNet, Unet and the original DeeplabV3+ model, with a 3.27% intersection over union (IoU) and 1.69% pixel accuracy (PA) improvement than the original DeeplabV3+ model. The purpose of this meta-analysis was to evaluate the efficacy and safety of celecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, in addition to standard anticancer therapy. Randomized controlled trials (RCTs) that evaluated the efficacy and safety of celecoxib-combined cancer therapy were systematically searched in PubMed and Embase databases. The endpoints were overall survival (OS), progression-free survival (PFS), disease-free survival (DFS), objective response rate (ORR), disease control rate (DCR), pathological complete response (pCR), and adverse events (AEs). The results of 30 RCTs containing 9655 patients showed limited benefits in celecoxib-combined cancer therapy. However, celecoxib-combined palliative therapy prolonged PFS in epidermal growth factor receptor (EGFR) wild-type patients (HR = 0.57, 95%CI = 0.35–0.94). Moreover, despite a slight increase in thrombocytopenia (RR = 1.35, 95%CI = 1.08–1.69), there was no increase in other toxicities. Celecoxib combined with adjuvant therapy indicated a better OS (HR = 0.850, 95%CI = 0.725–0.996). Furthermore, celecoxib plus neoadjuvant therapy improved the ORR in standard cancer therapy, especially neoadjuvant therapy (overall: RR = 1.13, 95%CI = 1.03–1.23; neoadjuvant therapy: RR = 1.25, 95%CI = 1.09–1.44), but not pCR. Our study indicated that adding celecoxib to palliative therapy prolongs the PFS of EGFR wild-type patients, with good safety profiles. Celecoxib combined with adjuvant therapy prolongs OS, and celecoxib plus neoadjuvant therapy improves the ORR. Thus, celecoxib-combined cancer therapy may be a promising therapy strategy. Soil salinity is a major constraint that affects plant growth and development. Rice is a staple food for more than half of the human population but is extremely sensitive to salinity. Among the several known mechanisms, the ability of the plant to exclude cytosolic Na+ is strongly correlated with salinity stress tolerance in different plant species. This exclusion is mediated by the plasma membrane (PM) Na+/H+ antiporter encoded by Salt Overly Sensitive (SOS1) gene and driven by a PM H+-ATPase generated proton gradient. However, it is not clear to what extent this mechanism is operational in wild and cultivated rice species, given the unique rice root anatomy and the existence of the bypass flow for Na+. As wild rice species provide a rich source of genetic diversity for possible introgression of abiotic stress tolerance, we investigated physiological and molecular basis of salinity stress tolerance in Oryza species by using two contrasting pairs of cultivated (Oryza sativa) and wild rice species (Oryza alta and Oryza punctata). Accordingly, dose- and age-dependent Na+ and H+ fluxes were measured using a non-invasive ion selective vibrating microelectrode (the MIFE technique) to measure potential activity of SOS1-encoded Na+/H+ antiporter genes. Consistent with GUS staining data reported in the literature, rice accessions had (~4–6-fold) greater net Na+ efflux in the root elongation zone (EZ) compared to the mature root zone (MZ). Pharmacological experiments showed that Na+ efflux in root EZ is suppressed by more than 90% by amiloride, indicating the possible involvement of Na+/H+ exchanger activity in root EZ. Within each group (cultivated vs. wild) the magnitude of amiloride-sensitive Na+ efflux was higher in tolerant genotypes; however, the activity of Na+/H+ exchanger was 2–3-fold higher in the cultivated rice compared with their wild counterparts. Gene expression levels of SOS1, SOS2 and SOS3 were upregulated under 24 h salinity treatment in all the tested genotypes, with the highest level of SOS1 transcript detected in salt-tolerant wild rice genotype O. alta (~5–6-fold increased transcript level) followed by another wild rice, O. punctata. There was no significant difference in SOS1 expression observed for cultivated rice (IR1-tolerant and IR29-sensitive) under both 0 and 24 h salinity exposure. Our findings suggest that salt-tolerant cultivated rice relies on the cytosolic Na+ exclusion mechanism to deal with salt stress to a greater extent than wild rice, but its operation seems to be regulated at a post-translational rather than transcriptional level. Xinyang Maojian (XYMJ) green tea is one of the top ten teas in China, and the consumers prefer spring tea due to its umami taste and pleasurable aroma. However, the knowledge about temporal variation of the volatile compounds in XYMJ green teas harvested during different seasons is very limited. In the present work, the main non-volatile compounds that endowed the taste and volatile compounds responsible for the aroma in XYMJ green teas harvested during the spring and autumn seasons were determined. The average contents of free amino acids (FAA) were significantly higher and gradually declined in the spring teas, whereas the caffeine was significantly lower and gradually increased in the spring teas. A total of 39 volatile compounds of six chemical classes were detected in XYMJ green teas, and they displayed various change trends during the spring and autumn seasons, among which 15 volatile compounds were identified as the key odorants based on odor activity value (OAV). The highest OAV of 2195.05 was calculated for the violet-like smelling trans-β-ionone followed by decanal, nonanal, dimethyl sulfide, linalool, geraniol and naphthalene. The OAVs of geraniol, (Z)-3-hexenyl hexanoate, heptanal, benzaldehyde and hexanal in XYMJ spring teas were higher than XYMJ autumn teas. The hierarchical clustering analysis indicated that XYMJ green teas were divided into three clusters and the quality of XYMJ green teas changed greatly within spring season. The influence of tempering temperature on the microstructure of 0.5Cr0.4W steels was investigated by scanning electron microscope, and the roles of grain boundary character, dislocation, and Taylor factor in sulfide stress cracking (SSC) resistance were interpreted using the election backscattered diffraction technique. The 0.5Cr0.4W steels tempered at 690 °C, 700 °C, and 715 °C all showed tempered martensites. The specimen tempered at 715 °C exhibited a higher critical stress intensity factor (KISSC) of 34.58 MPa·m0.5, but the yield strength of 800 MPa did not meet the criterion of 125 ksi (862 MPa) grade. When the specimen was tempered at 690 °C, the yield strength reached 960 MPa and the KISSC was only 21.36 MPa·m0.5, displaying poorer SSC resistance. The 0.5Cr0.4W steel tempered at 700 °C showed a good combination of yield strength (887 MPa) and SSC resistance (KISSC: 31.16 MPa·m0.5). When increasing the tempering temperature, the local average misorientation and Taylor factor of the 0.5Cr0.4W steels were decreased. The reduced dislocation density, and greater number of grains amenable to slippage, produced less hydrogen transport and a lower crack sensitivity. The SSC resistance was, thus, increased, owing to the minor damage to hydrogen aggregation. Therefore, 700 °C is a suitable tempering temperature for 0.5Cr0.4W casing steel. [ABSTRACT FROM AUTHOR]

Marine biofouling is a worldwide problem in coastal areas and affects the maritime industry primarily by attachment of fouling organisms to solid immersed surfaces. Biofilm formation by microbes is the main cause of biofouling. Currently, application of antibacterial materials is an important strategy for preventing bacterial colonization and biofilm formation. A natural three-dimensional carbon skeleton material, TRP (treated rape pollen), attracted our attention owing to its visible-light-driven photocatalytic disinfection property. Based on this, we hypothesized that TRP, which is eco-friendly, would show antifouling performance and could be used for marine antifouling. We then assessed its physiochemical characteristics, oxidant potential, and antifouling ability. The results showed that TRP had excellent photosensitivity and oxidant ability, as well as strong anti-bacterial colonization capability under light-driven conditions. Confocal laser scanning microscopy showed that TRP could disperse pre-established biofilms on stainless steel surfaces in natural seawater. The biodiversity and taxonomic composition of biofilms were significantly altered by TRP (p < 0.05). Moreover, metagenomics analysis showed that functional classes involved in the antioxidant system, environmental stress, glucose–lipid metabolism, and membrane-associated functions were changed after TRP exposure. Co-occurrence model analysis further revealed that TRP markedly increased the complexity of the biofilm microbial network under light irradiation. Taken together, these results demonstrate that TRP with light irradiation can inhibit bacterial colonization and prevent initial biofilm formation. Graphene has been widely used as a lubricating additive to reduce the energy consumption of engines and improve fuel economy because of its unique crystal structure. Herein, graphene (GR) and fluorinated graphene (F-GR) nanoparticles were prepared by ball milling and liquid-phase exfoliation. The SEM/EDS, HRTEM, XPS, Raman spectrometer, X-ray spectrometer, FTIR were used to investigate the morphologies, surface groups, and crystal structure of two kinds of graphene materials. The influence of loads on the tribological properties of two kinds of particles was investigated in Poly Alpha Olefin (PAO6) using a UMT-2 reciprocating tribometer. Results showed that the crystal structure of GR is better than F-GR. F-GR can improve the lubrication performance of PAO6. For PAO6 containing 1 wt% F-GR at 10 N, the average friction coefficient and average wear rate decreased by 12.3% and 87% relative to pure PAO6, respectively. However, the high load resulted in an inconspicuous anti-wear and anti-friction effect. The influence of F-GR on the tribological behavior of PAO6 was more substantial than that of GR. The friction and wear mechanisms attributed to F-GR quickly entered the interface between the friction pairs. Friction-induced F-GR nanosheets mainly took the tribo-chemical reactions to participate in the lubrication film formation and helped achieve a low friction coefficient and wear rate. Cooperative supramolecular polymerization of π-conjugated compounds into one-dimensional nanostructures has received tremendous attentions in recent years. It is commonly achieved by incorporating amide linkages into the monomeric structures, which provide hydrogen bonds for intermolecular non-covalent complexation. Herein, the effect of amide linkages is elaborately studied, by comparing supramolecular polymerization behaviors of two structurally similar monomers with the same platinum(II) acetylide cores. As compared to the N-phenyl benzamide linkages, N-[(1S)-1-phenylethyl] benzamide linkages give rise to effective chirality transfer behaviors due to the closer distances between the chiral units and the platinum(II) acetylide core. They also provide stronger intermolecular hydrogen bonding strength, which consequently brings higher thermo-stability and enhanced gelation capability for the resulting supramolecular polymers. Supramolecular polymerization is further strengthened by varying the monomers from monotopic to ditopic structures. Sustainable food production in protected cropping is increasing rapidly in response to global climate change and population growth. However, there are significant knowledge gaps regarding energy consumption while achieving optimum environmental conditions for greenhouse crop production. A capsicum crop cultivated in a high-tech greenhouse facility in Australia was analysed in terms of relationships between key environmental variables and the comparative analysis of energy consumption during different seasons. We showed that daily energy consumption varied due to the seasonal nature of the external environment and maintenance of optimal growing temperatures. Total power consumption reported throughout the entire crop cycle for heating (gas hot water system) and cooling (pad and fan) was 12,503 and 5183 kWh, respectively; hence, heating consumed ca. 70% of the total energy requirement over the 8-month growing period (early spring to late autumn) in the greenhouse facility. Regressions of daily energy consumption within each season, designated either predominantly for heating or cooling, indicated that energy consumption was 14.62 kWh per 1 °C heating and 2.23 kWh per 1 °C cooling. Therefore, changing the planting date to late spring is likely to significantly reduce heating energy costs for greenhouse capsicum growers in Australia. The findings will provide useful guidelines to maximise the greenhouse production of capsicum with better economic return by taking into consideration the potential optimal energy saving strategy during different external environment conditions and seasons. Plant cell walls play a fundamental role in several ways, providing structural support for cells, resistance against pathogens and facilitating the communication between cells. The glycosyltransferase family 8 (GT8) is involved in the formation of the plant cell wall. However, the evolutionary relationship and the functional differentiation of this important gene family remain obscure in Gramineae crop genomes. In the present investigation, we identified 269 GT8 genes in the seven Gramineae representative crop genomes, namely, 33 in Hordeum vulgare, 37 in Brachypodium distachyon, 40 in Oryza sativa ssp. japonica, 41 in Oryza rufipogon, 36 in Setaria italica, 37 in Sorghum bicolor, and 45 in Zea mays. Phylogenetic analysis suggested that all identified GT8 proteins belonged to seven subfamilies: galacturonosyltransferase (GAUT), galacturonosyltransferase-like (GATL), GATL-related (GATR), galactinol synthase (GolS), and plant glycogenin-like starch initiation proteins A (PGSIP-A), PGSIP-B, and PGSIP-C. We estimated that the GAUT subfamily might be further divided into four subgroups (I–IV) due to differentiation of gene structures and expression patterns. Our orthogroup analysis identified 22 orthogroups with different sizes. Of these orthogroups, several orthogroups were lost in some species, such as S. italica and Z. mays. Moreover, lots of duplicate pairs and collinear pairs were discovered among these species. These results indicated that multiple duplication modes led to the expansion of this important gene family and unequal loss of orthogroups and subfamilies might have happened during the evolutionary process. RNA-seq, microarray analysis, and qRT-PCR analyses indicated that GT8 genes are critical for plant growth and development, and for stresses responses. We found that OsGolS1 was significantly up-regulated under salt stress, while OsGAUT21, OsGATL2, and OsGATL5 had remarkable up-regulation under cold stress. The current study highlighted the expansion and evolutionary patterns of the GT8 gene family in these seven Gramineae crop genomes and provided potential candidate genes for future salt- and cold- resistant molecular breeding studies in O. sativa. Sugars are critical for plant growth and development as suppliers of carbon and energy, as signal molecules, or as solute molecules for osmotic homeostasis. Monosaccharide transporter (MST) genes are involved in various processes of plant growth and development as well as in response to abiotic stresses. However, the evolution and their roles of MST genes in growth and development and in coping with abiotic stresses in rice are poorly known. Here, we identified 64 MST genes in rice genome, which are classified into seven subfamilies: STP, PLT, AZT, ERD, pGlcT, INT, and XTPH. MST genes are not evenly distributed between chromosomes (Chrs) with a bias to Chr 3, 4, 7, and 11, which could be a result of duplication of fragments harboring MST genes. In total, 12 duplication events were found in the rice MST family, among which, two pairs were derived from fragmental duplications and ten pairs were from tandem duplications. The synonymous and nonsynonymous substitution rates of duplicate gene pairs demonstrated that the MST family was under a strong negative selection during the evolution process. Furthermore, a comprehensive expression analysis conducted in 11 different tissues, three abiotic stresses, five hormone treatments, and three sugar treatments revealed different expression patterns of MST genes and indicated diversified functions of them. Our results suggest that MST genes play important roles not only in various abiotic stresses but also in hormone and sugar responses. Acanthopanax senticosus (Rupr. Maxim.) Harms (ASH) and Acanthopanax sessiliflorus (Rupr. Maxim.) Seem (ASS), are members of the Araliaceae family, and both are used in Asian countries. These herbals have drawn much attention in recent years due to their strong biological activity, with innocuity and little side effects. However, the common and distinct mode of compound profiles between ASH and ASS is still unclear. In this study, a high performance liquid chromatograph-mass spectrometry (HPLC-MS) method was developed to simultaneously quantify the seven major active compounds, including protocatechuate, eleutheroside B, eleutheroside E, isofraxidin, hyperoside, kaempferol and oleanolic acid. Then the targeted metabolomics were conducted to identify 19 phenolic compounds, with tight relation to the above mentioned active compounds, including nine C6C3C6-type, six C6C3-type and four C6C1-type in the two Acanthopanax species studied here. The results showed that the seven active compounds presented a similar trend of changes in different tissues, with more abundant accumulation in roots and stems for both plants. From the view of plant species, the ASH plants possess higher abundance of compounds, especially in the tissues of roots and stems. For phenolics, the 19 phenols detected here could be clearly grouped into five main clusters based on their tissue-specific accumulation patterns. Roots are the tissue for the most abundance of their accumulations. C6C3C6-type compounds are the most widely existing type in both plants. Worldwide, drought affects crop yields; therefore, understanding plants' strategies to adapt to drought is critical. Chloroplasts are key regulators of plant responses, and signals from chloroplasts also regulate nuclear gene expression during drought. However, the interactions between chloroplast-initiated retrograde signals and ion channels under stress are still not clear. In this review, we summarise the retrograde signals that participate in regulating plant stress tolerance. We compare chloroplastic transporters that modulate retrograde signalling through retrograde biosynthesis or as critical components in retrograde signalling. We also discuss the roles of important plasma membrane and tonoplast ion transporters that are involved in regulating stomatal movement. Ginsenosides, the major compounds present in ginseng, are known to have numerous physiological and pharmacological effects. The physiological processes, enzymes and genes involved in ginsenoside synthesis in P. ginseng have been well characterized. However, relatively little information is known about the dynamic metabolic changes that occur during ginsenoside accumulation in ginseng. To explore this topic, we isolated metabolites from different tissues at different growth stages, and identified and characterized them by using gas chromatography coupled with mass spectrometry (GC-MS). The results showed that a total of 30, 16, 20, 36 and 31 metabolites were identified and involved in different developmental stages in leaf, stem, petiole, lateral root and main root, respectively. To investigate the contribution of tissue to the biosynthesis of ginsenosides, we examined the metabolic changes of leaf, stem, petiole, lateral root and main root during five development stages: 1-, 2-, 3-, 4- and 5-years. The score plots of partial least squares-discriminate analysis (PLS-DA) showed clear discrimination between growth stages and tissue samples. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis in the same tissue at different growth stages indicated profound biochemical changes in several pathways, including carbohydrate metabolism and pentose phosphate metabolism, in addition, the tissues displayed significant variations in amino acid metabolism, sugar metabolism and energy metabolism. These results should facilitate further dissection of the metabolic flux regulation of ginsenoside accumulation in different developmental stages or different tissues of ginseng. The effect of a naphthalimide pharmacophore coupled with diverse substituents on the interaction between naphthalimide-polyamine conjugates 1-4 and bovine serum albumin (BSA) was studied by UV absorption, fluorescence and circular dichroism (CD) spectroscopy under physiological conditions (pH = 7.4). The observed spectral quenching of BSA by the compounds indicated that they could bind to BSA. Furthermore, caloric fluorescent tests revealed that the quenching mechanisms of compounds 1-3 were basically static type, but that of compound 4 was closer to a classical type. The Ksv values at room temperature for compound-BSA complexes-1-BSA, 2-BSA, 3-BSA and 4-BSA were 1.438 × 104, 3.190 × 104, 5.700 × 104 and 4.745 × 105, respectively, compared with the value of MINS, 2.863 × 104 at Ex = 280 nm. The obtained quenching constant, binding constant and thermodynamic parameter suggested that the binding between compounds 1-4 with BSA protein, significantly affected by the substituted groups on the naphthalene backbone, was formed by hydrogen bonds, and other principle forces mainly consisting of charged and hydrophobic interactions. Based on results from the analysis of synchronous three-dimensional fluorescence and CD spectra, we can conclude that the interaction between compounds 1-4 and BSA protein has little impact on the BSA conformation. Calculated results obtained from in silico molecular simulation showed that compound 1 did not prefer either enzymatic drug sites I or II over the other. However, DSII in BSA was more beneficial than DSI for the binding between compounds 2-4 and BSA protein. Rice (Oryza sativa), a staple crop for a substantial part of the world's population, is highly sensitive to soil salinity; however, some wild Oryza relatives can survive in highly saline environments. Sodium/hydrogen antiporter (NHX) family members contribute to Na+ homeostasis in plants and play a major role in conferring salinity tolerance. In this study, we analyzed the evolution of NHX family members using phylogeny, conserved domains, tertiary structures, expression patterns, and physiology of cultivated and wild Oryza species to decipher the role of NHXs in salt tolerance in Oryza. Phylogenetic analysis showed that the NHX family can be classified into three subfamilies directly related to their subcellular localization: endomembrane, plasma membrane, and tonoplast (vacuolar subfamily, vNHX1). Phylogenetic and structural analysis showed that vNHX1s have evolved from streptophyte algae (e.g., Klebsormidium nitens) and are abundant and highly conserved in all major land plant lineages, including Oryza. Moreover, we showed that tissue tolerance is a crucial trait conferring tolerance to salinity in wild rice species. Higher Na+ accumulation and reduced Na+ effluxes in leaf mesophyll were observed in the salt-tolerant wild rice species O. alta, O. latifolia, and O. coarctata. Among the key genes affecting tissue tolerance, expression of NHX1 and SOS1/NHX7 exhibited significant correlation with salt tolerance among the rice species and cultivars. This study provides insights into the evolutionary origin of plant NHXs and their role in tissue tolerance of Oryza species and facilitates the inclusion of this trait during the development of salinity-tolerant rice cultivars. Eleven novel naphthalimide-diamine conjugates were synthesized and their structures were confirmed by elemental analysis, 1H-NMR, 13C-NMR and MS. Their in vitro antitumor activities were assessed using MTT assays on two cancerous cell lines K562, HCT116, and one normal hepatoma cell line QSG 7701. Compound 7f exhibited potent antitumor activity on HCT116 cells and favorable cell selectivity toward QSG 7701 compared with the positive control, amonafide. Moreover, 7f could block HeG2 cells in the G2/M phase and induce HeG2 cells apoptosis. The interaction of compound 7f with herring sperm DNA was studied by UV/vis absorption and fluorescence spectroscopy under physiological conditions (pH = 7.4). The observed spectral quenching of compound 7f by DNA and the displacement of EB from DNA-EB complex by compound 7f indicated that compound 7f could intercalate into DNA base pairs, which was also corroborated by the effect of KI on compound-DNA interaction. Further caloric fluorescent tests revealed that the quenching mechanism was a static type. Meanwhile, the binding constants, thermodynamic parameters and the effect of NaCl on compound-DNA interaction showed that the type of interaction force was mainly hydrogen bonds and the binding process was driven by hydrogen and van der Waals bonding. Adaptation to unfavorable abiotic stresses is one of the key processes in the evolution of plants. Calcium (Ca2+) signaling is characterized by the spatiotemporal pattern of Ca2+ distribution and the activities of multi-domain proteins in integrating environmental stimuli and cellular responses, which are crucial early events in abiotic stress responses in plants. However, a comprehensive summary and explanation for evolutionary and functional synergies in Ca2+ signaling remains elusive in green plants. We review mechanisms of Ca2+ membrane transporters and intracellular Ca2+ sensors with evolutionary imprinting and structural clues. These may provide molecular and bioinformatics insights for the functional analysis of some non-model species in the evolutionarily important green plant lineages. We summarize the chronological order, spatial location, and characteristics of Ca2+ functional proteins. Furthermore, we highlight the integral functions of calcium-signaling components in various nodes of the Ca2+ signaling pathway through conserved or variant evolutionary processes. These ultimately bridge the Ca2+ cascade reactions into regulatory networks, particularly in the hormonal signaling pathways. In summary, this review provides new perspectives towards a better understanding of the evolution, interaction and integration of Ca2+ signaling components in green plants, which is likely to benefit future research in agriculture, evolutionary biology, ecology and the environment. Halophytes species can be used as a highly convenient model system to reveal key ionic and molecular mechanisms that confer salinity tolerance in plants. Earlier, we reported that quinoa (Chenopodium quinoa Willd.), a facultative C3 halophyte species, can efficiently control the activity of slow (SV) and fast (FV) tonoplast channels to match specific growth conditions by ensuring that most of accumulated Na+ is safely locked in the vacuole (Bonales-Alatorre et al. (2013) Plant Physiology). This work extends these finding by comparing the properties of tonoplast FV and SV channels in two quinoa genotypes contrasting in their salinity tolerance. The work is complemented by studies of the kinetics of net ion fluxes across the plasma membrane of quinoa leaf mesophyll tissue. Our results suggest that multiple mechanisms contribute towards genotypic differences in salinity tolerance in quinoa. These include: (i) a higher rate of Na+ exclusion from leaf mesophyll; (ii) maintenance of low cytosolic Na+ levels; (iii) better K+ retention in the leaf mesophyll; (iv) a high rate of H+ pumping, which increases the ability of mesophyll cells to restore their membrane potential; and (v) the ability to reduce the activity of SV and FV channels under saline conditions. Energy management in protected cropping is critical due to the high cost of energy use in high-tech greenhouse facilities. The main purpose of this research was to investigate the optimal strategy to reduce cooling energy consumption, by regulating the settings (opening/closing) of either vents or curtains during the day, at the protected cropping facility at Western Sydney University. We measured daily changes in air temperature and energy consumption under four treatments (open/closed combinations of vents and shade screens) and developed an optimal cooling strategy for energy management using multi-temperature acquisition points at different heights within a greenhouse compartment. The optimal treatment (vents open/curtains closed) reduced energy load at the rooftop, thereby maintaining a desirable plant canopy temperature profile, and reducing cooling energy. Daily energy consumption was lowest for vents open/curtains closed (70.5 kWh) and highest for vents closed/curtains open (121 kWh). It was also found that delaying the operation of opening and closing of vents and curtains until the plant canopy temperature reached 25 °C reduced cooling energy consumption and decreased heating energy consumption in the morning (e.g., 08:00 to 10:00). The estimated savings of 1.83 kWh per 1 °C cooling between the optimal (vents open/curtains closed) and least optimal (vents closed/curtains open) conditions had the potential for significant energy savings at 494 kWh per °C over a crop cycle of nine months in warm weather conditions. However, selection of the optimal cooling strategy utilising control of vents and curtains must also account for the impact from other greenhouse environmental factors, including light, humidity, and CO2 concentration, which may be crop specific. Rice (Oryza sativa L.) is a widely cultivated food crop around the world, especially in Asia. However, rice seedlings often suffer from cold stress, which affects their growth and yield. Here, RNA-seq analysis and Meta-QTLs mapping were performed to understand the molecular mechanisms underlying cold tolerance in the roots of 14-day-old seedlings of rice (RPY geng, cold-tolerant genotype). A total of 4779 of the differentially expressed genes (DEGs) were identified, including 2457 up-regulated and 2322 down-regulated DEGs. The GO, COG, KEEG, and Mapman enrichment results of DEGs revealed that DEGs are mainly involved in carbohydrate transport and metabolism, signal transduction mechanisms (plant hormone signal transduction), biosynthesis, transport and catabolism of secondary metabolites (phenylpropanoid biosynthesis), defense mechanisms, and large enzyme families mechanisms. Notably, the AP2/ERF-ERF, NAC, WRKY, MYB, C2H2, and bHLH transcription factors participated in rice's cold–stress response and tolerance. On the other hand, we mapped the identified DEGs to 44 published cold–stress-related genes and 41 cold-tolerant Meta-QTLs regions. Of them, 12 DEGs were the published cold–stress-related genes and 418 DEGs fell into the cold-tolerant Meta-QTLs regions. In this study, the identified DEGs and the putative molecular regulatory network can provide insights for understanding the mechanism of cold stress tolerance in rice. In addition, DEGs in KEGG term-enriched terms or cold-tolerant Meta-QTLs will help to secure key candidate genes for further functional studies on the molecular mechanism of cold stress response in rice. Drought and salinity are two of the most frequently co-occurring abiotic stresses. Despite recent advances in the elucidation of the effects of these stresses individually during the vegetative stage of plants, significant gaps exist in our understanding of the combined effects of these two frequently co-occurring stresses. Here, Tibetan wild barley XZ5 (drought tolerant), XZ16 (salt tolerant), and cultivated barley cv. CM72 (salt tolerant) were subjected to drought (D), salinity (S), or a combination of both treatments (D+S). Protein synthesis is one of the primary activities of the green part of the plant. Therefore, leaf tissue is an important parameter to evaluate drought and salinity stress conditions. Sixty differentially expressed proteins were identified by mass spectrometry (MALDI-TOF/TOF) and classified into 9 biological processes based on Gene Ontology annotation. Among them, 21 proteins were found to be expressed under drought or salinity alone; however, under D+S, 7 proteins, including S-adenosylmethionine synthetase 3 (SAMS3), were exclusively upregulated in drought-tolerant XZ5 but not in CM72. HvSAMS3 carries both N-terminal and central domains compared with Arabidopsis and activates the expression of several ethylene (ET)-responsive transcription factors. HvSAMS3 is mainly expressed in the roots and stems, and HvSAMS3 is a secretory protein located in the cell membrane and cytoplasm. Barley stripe mosaic virus-based virus-induced gene silencing (BSMV-VIGS) of HvSAMS3 in XZ5 severely compromised its tolerance to D+S and significantly reduced plant growth and K+ uptake. The reduced tolerance to the combined stress was associated with the inhibition of polyamines such as spermidine and spermine, polyamine oxidase, ethylene, biotin, and antioxidant enzyme activities. Furthermore, the exogenous application of ethylene and biotin improved the tolerance to D+S in BSMV-VIGS:HvSAMS3-inoculated plants. Ultraviolet-B (UV-B) radiation (280–320 nm) may induce photobiological stress in plants, activate the plant defense system, and induce changes of metabolites. In our previous work, we found that between the two Astragalus varieties prescribed by the Chinese Pharmacopoeia, Astragalus mongholicus has better tolerance to UV-B. Thus, it is necessary to study the metabolic strategy of Astragalus under UV-B radiation further. In the present study, we used untargeted gas chromatography-mass spectrometry (GC-MS) and targeted liquid chromatography-mass spectrometry (LC-MS techniques) to investigate the profiles of primary and secondary metabolic. The profiles revealed the metabolic response of Astragalus to UV-B radiation. We then used real-time polymerase chain reaction (RT-PCR) to obtain the transcription level of relevant genes under UV-B radiation (UV-B supplemented in the field, λmax = 313 nm, 30 W, lamp-leaf distance = 60 cm, 40 min·day−1), which annotated the responsive mechanism of phenolic metabolism in roots. Our results indicated that supplemental UV-B radiation induced a stronger shift from carbon assimilation to carbon accumulation. The flux through the phenylpropanoids pathway increased due to the mobilization of carbon reserves. The response of metabolism was observed to be significantly tissue-specific upon the UV-B radiation treatment. Among phenolic compounds, C6C1 carbon compounds (phenolic acids in leaves) and C6C3C6 carbon compounds (flavones in leaves and isoflavones in roots) increased at the expense of C6C3 carbon compounds. Verification experiments show that the response of phenolics in roots to UV-B is activated by upregulation of relevant genes rather than phenylalanine. Cystosepiment-like cobalt nanoparticles@N-doped carbon composite named Co-NPs@NC with highly efficient electrocatalytic performance for oxygen evolution reaction was prepared from carbonization of N-doped Co-MOFs. The optimized Co-NPs@NC-600 shows overpotentials of 315 mV to afford a current density of 10 mA·cm−2. Meanwhile, the electrocatalys presents excellent long-term durability. The outstanding electrocatalytic performance can be attributed to the unique cystosepiment-like architecture with high specific surface area (214 m2/g), high conductivity of N-doped carbon and well-distributed active sites. Tibetan wild barley has been identified to show large genetic variation and stress tolerance. A genome-wide association (GWA) analysis was performed to detect quantitative trait loci (QTLs) for drought tolerance using 777 Diversity Array Technology (DArT) markers and morphological and physiological traits of 166 Tibetan wild barley accessions in both hydroponic and pot experiments. Large genotypic variation for these traits was found; and population structure and kinship analysis identified three subpopulations among these barley genotypes. The average LD (linkage disequilibrium) decay distance was 5.16 cM, with the minimum on 6H (0.03 cM) and the maximum on 4H (23.48 cM). A total of 91 DArT markers were identified to be associated with drought tolerance-related traits, with 33, 26, 16, 1, 3, and 12 associations for morphological traits, H+K+-ATPase activity, antioxidant enzyme activities, malondialdehyde (MDA) content, soluble protein content, and potassium concentration, respectively. Furthermore, 7 and 24 putative candidate genes were identified based on the reference Meta-QTL map and by searching the Barleymap. The present study implicated that Tibetan annual wild barley from Qinghai–Tibet Plateau is rich in genetic variation for drought stress. The QTLs detected by genome-wide association analysis could be used in marker-assisting breeding for drought-tolerant barley genotypes and provide useful information for discovery and functional analysis of key genes in the future.
