Guided wave array-based structural health monitoring (SHM) is a promising solution for diagnosing damage in metal-connected structures. In this field, the reconstruction algorithm for probabilistic inspection (RAPID) is one of the most widely used algorithms for performing damage localization. In this paper, a density clustering RAPID based on an array-compensated damage index is proposed. A new probability distribution function was constructed based on a new damage index, which is adaptive to different elements in the sensor array to compensate for performance variation. Then, the imaging matrix of the RAPID algorithm was density-clustered to obtain the location and degree of damage. Finally, the method was verified by experiments on a stiffened aluminum plate. The experimental results demonstrate that the method achieves damage localization and enables quantitative damage diagnosis.

Aiming at the existing high-temperature liquid metal flow online accurate measurement by the metal melt characteristics, installation space, and high-temperature environment adaptability limitations, this paper innovatively puts forward a soft measurement method based on the impact force generated in the fluid flow process as an observational variable series. Fluid mechanics theory and simulation software are used to analyze and verify the feasibility of the impact force as an observable variable to measure the flow rate, followed by the construction of the CNN-LSTM-CNN-Double (CLCD) flow measurement model of impact force and flow rate based on the parameters of the learning rate and the number of training times, and finally the construction of a test platform for the flow measurement, and the validity of the method is verified through actual operation.

Cultivated peanut ( Arachis hypogaea L.) is a key oil- and protein-providing legume crop of the world. It is full of nutrients, and its nutrient profile is comparable to that of other nuts. Peanut is a unique plant as it showcases a pegging phenomenon, producing flowers above ground, and after fertilization, the developing peg enters the soil and produces seeds underground. This geocarpic nature of peanut exposes its seeds to soil pathogens. Peanut seeds are protected by an inedible pericarp and testa. The pericarp- and testa-specific promoters can be effectively used to improve the seed defense. We identified a pericarp- and testa-abundant expression gene ( AhN8DT-2 ) from available transcriptome expression data, whose tissue-specific expression was further confirmed by the qRT-PCR. The 1827bp promoter sequence was used to construct the expression vector using the pMDC164 vector for further analysis. Quantitative expression of the GUS gene in transgenic Arabidopsis plants showed its high expression in the pericarp. GUS staining showed a deep blue color in the pericarp and testa. Cryostat sectioning of stained Arabidopsis seeds showed that expression is only limited to seed coat (testa), and staining was not present in cotyledons and embryos. GUS staining was not detected in any other tissues, including seedlings, leaves, stems, and roots, except for some staining in flowers. Under different phytohormones, this promoter did not show an increase in expression level. These results indicated that the AhN8DT-2 promoter drives GUS gene expression in a pericarp- and testa-specific manner. The identified promoter can be utilized to drive disease resistance genes, specifically in the pericarp and testa, enhancing peanut seed defense against soil-borne pathogens. This approach has broader implications for improving the resilience of peanut crops and other legumes, contributing to sustainable agricultural practices and food security.

Considering that heavy-metal contamination of seawater is getting worse, building a quick, accurate and portable device for detecting trace zinc in seawater in real time would be very beneficial. In this work, a microfluidic system was developed that includes a planar disc electrode, a micro-cavity for detection, an electrochemical workstation, a computer, a container for waste liquid reprocessing, an external pipeline and other components as well as a graphene/cerium oxide/nano-cerium oxide/Nafion composite membrane was used to modify the planar disc electrode (GR/CeO 2 /Nafion/Au) to investigate the electrochemical behaviour of Zn(II) using cyclic voltammetry, square-wave voltammetry and orthogonal test methods. Under optimal experimental conditions, the peak reaction current of Zn(II) showed a good linear relationship with the concentration of Zn(II) in the range of 1-900 μg/L with a correlation coefficient of 0.998, and the detection limit of the method was 0.87 μg/L. In addition, the microfluidic system had good stability, reproducibility and anti-interference. The system was used for determining zinc ions in real seawater samples, and the results were very similar to those of inductively coupled plasma-emission spectrometry, demonstrating the practicality of the system for the detection of trace zinc.

In this paper, a sensor using a complementary split ring resonator (CSRR) is proposed for non-destructive testing of blood glucose. By depicting the complementary split ring structure on the ground, the electromagnetic field strength between the split rings can be enhanced effectively. The structure size of the sensor by CSRR is determined by simulation, so that the insertion loss curve of the device has a resonance point at the frequency of 3.419 GHz. With a special holder created by three-dimensional (3D) printing technology, the test platform was established when the concentration of the solution varied from 0 mg/mL to 20 mg/mL. The experimental results indicate that there is an obvious linear relationship between the insertion loss S 21 and the glucose concentration at the resonant frequency. Similarly, the measured real part and imaginary part of the S 21 both vary with glucose concentration linearly. Based on the above experimental results, the feasibility of the sensor using a CSRR proposed in this paper for non-destructive detection of blood glucose is preliminarily verified.

Binge drinking in obese patients positively correlates with accelerated liver damage and liver-related death. However, the underlying mechanism and the effect of alcohol use on the progression of metabolic-dysfunction-associated steatotic liver disease (MASLD) remain unexplored. Here, we show that short-term feeding of a metabolic-dysfunction-associated steatohepatitis (MASH) diet plus daily acute alcohol binges for three days induce liver injury and activation of the NLRP3 inflammasome. We identify that a MASH diet plus acute alcohol binges promote liver inflammation via increased infiltration of monocyte-derived macrophages, neutrophil recruitment, and NET release in the liver. Our results suggest that both monocyte-derived macrophages and neutrophils are activated via NLRP3, while the administration of MCC950, an NLRP3 inhibitor, dampens these effects.In this study, we reveal important intercellular communication between hepatocytes and neutrophils. We discover that the MASH diet plus alcohol induces IL-1β via NLRP3 activation and that IL-1β acts on hepatocytes and promotes the production of CXCL1 and LCN2. In turn, the increase in these neutrophils recruits chemokines and causes further infiltration and activation of neutrophils in the liver. In vivo administration of the NLRP3 inhibitor, MCC950, improves the early phase of MetALD by preventing liver damage, steatosis, inflammation, and immune cells recruitment.

Bryophyllum pinnatum (BP) is a medicinal plant used to treat many conditions when taken as a leaf juice, leaves in capsules, as an ethanolic extract, and as herbal tea. These preparations have been chemically analyzed except for decoctions derived from boiled green leaves. In preparation for a clinical trial to validate BP tea as a treatment for kidney stones, we used NMR and MS analyses to characterize the saturation kinetics of the release of metabolites. During boiling of the leaves, (a) the pH decreased to 4.8 within 14 min and then stabilized; (b) regarding organic acids, citric and malic acid were released with maximum release time (t max ) = 35 min; (c) for glycoflavonoids, quercetin 3-O-α-L-arabinopyranosyl-(1 → 2)-α-L-rhamnopyranoside (Q-3O-ArRh), myricetin 3-O-α-L-arabinopyranosyl-(1 → 2)-α-L-rhamnopyranoside (M-3O-ArRh), kappinatoside, myricitrin, and quercitrin were released with t max = 5-10 min; and (d) the total phenolic content (TPC) and the total antioxidant capacity (TAC) reached a t max at 55 min and 61 min, respectively. In summary, 24 g of leaves boiled in 250 mL of water for 61 min ensures a maximal release of key water-soluble metabolites, including organic acids and flavonoids. These metabolites are beneficial for treating kidney stones because they target oxidative stress and inflammation and inhibit stone formation.

Elevated glucose uptake and production of lactate are common features of cancer cells. Among many tumor-promoting effects, lactate inhibits immune responses and is positively correlated with radioresistance. Dichloroacetate (DCA) is an inhibitor of pyruvate dehydrogenase kinase that decreases lactate production. Quercetin is a flavonoid compound found in fruits and vegetables that inhibits glucose uptake and lactate export. We investigated the potential role and mechanisms of DCA, quercetin, and their combination, in the treatment of HPV-positive head and neck squamous cell carcinoma, an antigenic cancer subtype in need of efficacious adjuvant therapies. C57Bl/6-derived mouse oropharyngeal epithelial cells, a previously developed mouse model that was retrovirally transduced with HPV type-16 E6/E7 and activated Ras, were used to assess these compounds. Both DCA and quercetin inhibited colony formation and reduced cell viability, which were associated with mTOR inhibition and increased apoptosis through enhanced ROS production. DCA and quercetin reduced tumor growth and enhanced survival in immune-competent mice, correlating with decreased proliferation as well as decreased acidification of the tumor microenvironment and reduction of Foxp (+) Treg lymphocytes. Collectively, these data support the possible clinical application of DCA and quercetin as adjuvant therapies for head and neck cancer patients.

Anthocyanins are ubiquitous pigments derived from the phenylpropanoid compound conferring red, purple and blue pigmentations to various organs of horticultural crops. The metabolism of flavonoids in the cytoplasm leads to the biosynthesis of anthocyanin, which is then conveyed to the vacuoles for storage by plant glutathione S-transferases ( GST ). Although GST is important for transporting anthocyanin in plants, its identification and characterization in eggplant ( Solanum melongena L.) remains obscure. In this study, a total of 40 GST genes were obtained in the eggplant genome and classified into seven distinct chief groups based on the evolutionary relationship with Arabidopsis thaliana GST genes. The seven subgroups of eggplant GST genes ( SmGST ) comprise: dehydroascorbate reductase (DHAR), elongation factor 1Bγ (EF1Bγ), Zeta (Z), Theta(T), Phi(F), Tau(U) and tetra-chlorohydroquinone dehalogenase TCHQD. The 40 GST genes were unevenly distributed throughout the 10 eggplant chromosomes and were predominantly located in the cytoplasm. Structural gene analysis showed similarity in exons and introns within a GST subgroup. Six pairs of both tandem and segmental duplications have been identified, making them the primary factors contributing to the evolution of the SmGST . Light-related cis-regulatory elements were dominant, followed by stress-related and hormone-responsive elements. The syntenic analysis of orthologous genes indicated that eggplant, Arabidopsis and tomato ( Solanum lycopersicum L.) counterpart genes seemed to be derived from a common ancestry. RNA-seq data analyses showed high expression of 13 SmGST genes with SmGSTF1 being glaringly upregulated on the peel of purple eggplant but showed no or low expression on eggplant varieties with green or white peel. Subsequently, SmGSTF1 had a strong positive correlation with anthocyanin content and with anthocyanin structural genes like SmUFGT ( r = 0.9), SmANS ( r = 0.85), SmF3H ( r = 0.82) and SmCHI2 ( r = 0.7). The suppression of SmGSTF1 through virus-induced gene silencing (VIGs) resulted in a decrease in anthocyanin on the infiltrated fruit surface. In a nutshell, results from this study established that SmGSTF1 has the potential of anthocyanin accumulation in eggplant peel and offers viable candidate genes for the improvement of purple eggplant. The comprehensive studies of the SmGST family genes provide the foundation for deciphering molecular investigations into the functional analysis of SmGST genes in eggplant.

In this work, we propose a SiC-NSFET structure that uses a PTS scheme only under the gate, with SiC layers under the source and drain, to improve the leakage current and thermal reliability. Punch-through stopper (PTS) doping is widely used to suppress the leakage current, but aggressively high PTS doping will cause additional band-to-band (BTBT) current. Therefore, the bottom oxide isolation nanosheet field-effect transistor (BOX-NSFET) can further reduce the leakage current and become an alternative to conventional structures with PTS. However, thermal reliability issues, like bias temperature instability (BTI), hot carrier injection (HCI), and time-dependent dielectric breakdown (TDDB), induced by the self-heating effect (SHE) of BOX-NSFET, become more profound due to the lower thermal conductivity of SiO2 than silicon. Moreover, the bottom oxide will reduce the stress along the channel due to the challenges associated with growing high-quality SiGe material on SiO2. Therefore, this method faces difficulties in enhancing the mobility of p-type devices. The comprehensive TCAD simulation results show that SiC-NSFET significantly suppresses the substrate leakage current compared to the conventional structure with PTS. In addition, compared to the BOX-NSFET, the stress reduction caused by the bottom oxide is avoided, and the SHE is mitigated. This work provides significant design guidelines for leakage and thermal reliability optimization of next-generation advanced nodes.

In this paper, we investigate the effects of negative bias instability (NBTI) and self-heating effect (SHE) on threshold voltage in NSFETs. To explore accurately the interaction between SHE and NBTI, we established an NBTI simulation framework based on trap microdynamics and considered the influence of the self-heating effect. The results show that NBTI weakens the SHE effect, while SHE exacerbates the NBTI effect. Since the width of the nanosheet in NSFET has a significant control effect on the electric field distribution, we also studied the effect of the width of the nanosheet on the NBTI and self-heating effect. The results show that increasing the width of the nanosheet will reduce the NBTI effect but will enhance the SHE effect. In addition, we extended our research to the SRAM cell circuit, and the results show that the NBTI effect will reduce the static noise margin (SNM) of the SRAM cell, and the NBTI effect affected by self-heating will make the SNM decrease more significantly. In addition, our research results also indicate that increasing the nanosheet width can help slow down the NBTI effect and the negative impact of NBTI on SRAM performance affected by the self-heating effect.

Chlorogenic acids (CGAs) are bioactive compounds widely used in the food, pharmaceutical, and cosmetic industries. Carthamus tinctorius is an important economic crop, and its suspension cells are rich in CGAs. However, little is known about the biosynthesis and regulation of CGAs in Carthamus tinctorius cells. This study first elucidated the regulatory mechanism of CGA biosynthesis in methyl jasmonate (MeJA)-treated Carthamus tinctorius cells and the role of the MeJA-responsive hydroxycinnamoyl transferase ( HCT ) gene in enhancing their CGA accumulation. Firstly, temporal changes in intracellular metabolites showed that MeJA increased the intracellular CGA content up to 1.61-fold to 100.23 mg·g -1 . Meanwhile, 31 primary metabolites showed significant differences, with 6 precursors related to increasing CGA biosynthesis. Secondly, the transcriptome data revealed 3637 new genes previously unannotated in the Carthamus tinctorius genome and 3653 differentially expressed genes. The genes involved in the plant signaling pathway and the biosynthesis of CGAs and their precursors showed a general up-regulation, especially the HCT gene family, which ultimately promoted CGA biosynthesis. Thirdly, the expression of a newly annotated and MeJA-responsive HCT gene ( CtHCT , CtNewGene_3476) was demonstrated to be positively correlated with CGA accumulation in the cells, and transient overexpression of CtHCT enhanced CGA accumulation in tobacco. Finally, in vitro catalysis kinetics and molecular docking simulations revealed the ability and mechanism of the Ct HCT protein to bind to various substrates and catalyze the formation of four hydroxycinnamic esters, including CGAs. These findings strengthened our understanding of the regulatory mechanism of CGA biosynthesis, thereby providing theoretical support for the efficient production of CGAs.

The hippocampal formation, particularly the CA2 subregion, is critical for social memory formation and memory processing, relying on synaptic plasticity-a fundamental mechanism by which synapses strengthen. Given the role of the ubiquitin-proteasome system (UPS) in various nervous system processes, including learning and memory, we were particularly interested in exploring the involvement of RING-type ubiquitin E3 ligases, such as UHRF2 (NIRF), in social behavior and synaptic plasticity. Our results revealed altered social behavior in mice with systemic Uhrf2 knockout, including changes in nest building, tube dominance, and the three-chamber social novelty test. In Uhrf2 knockout mice, the entorhinal cortex-CA2 circuit showed significant reductions in synaptic plasticity during paired-pulse facilitation and long-term potentiation, while the inability to evoke synaptic plasticity in the Schaffer-collateral CA2 synapses remained unaffected. These changes in synaptic plasticity correlated with significant changes in gene expression including genes related to vesicle trafficking and transcriptional regulation. The effects of Uhrf2 knockout on synaptic plasticity and the observed gene expression changes highlight UHRF2 as a regulator of learning and memory processes at both the cellular and systemic levels. Targeting E3 ubiquitin ligases, such as UHRF2, may hold therapeutic potential for memory-related disorders, warranting further investigation.

The reliability of the resistor-capacitor (RC) time constant of a continuous-time (CT) filter has long been an obstacle with integrated circuits. Due to process and temperature variations in complementary metal-oxide semiconductor (CMOS) technology, the absolute value of the RC time constant may vary over ±50%, which is a big issue for many integrated continuous-time analog circuits. This study proposes an on-chip RC time constant auto-tuning scheme. The proposed scheme is based on the discrete master-slave auto-tuning concept. Considering the limitations in conventional works, a higher tuning accuracy is achieved by adopting two techniques: firstly, parasitic capacitance cancelation is proposed to eliminate the effects caused by parasitic capacitance; secondly, symmetric comparison is introduced to minimize the influence of the DC offset of the comparator. A successive approximation procedure is applied to improve the tuning speed. The proposed auto-tuning scheme has been validated in 55 nm CMOS technology with a fourth-order active-RC low-pass filter under PVT variations and 60 mV input offset voltage. The average tuning error is 2.21%, and the maximum error is 3.67%. The tuning error of the proposed scheme is considerably lower than the conventional scheme.

Berberine (BBR) is a natural alkaloid with multiple biotical effects that has potential as a treatment for fatty liver hemorrhagic syndrome (FLHS). However, the mechanism underlying the protective effect of BBR against FLHS remains unclear. The present study aimed to investigate the effect of BBR on FLHS induced by a high-energy, low-protein (HELP) diet and explore the involvement of the gut microbiota and bile acid metabolism in the protective effects. A total of 90 healthy 140-day-old Hy-line laying hens were randomly divided into three groups, including a control group (fed a basic diet), a HELP group (fed a HELP diet), and a HELP+BBR group (high-energy, high-protein diet supplemented with BBR instead of maize). Our results show that BBR supplementation alleviated liver injury and hepatic steatosis in laying hens. Moreover, BBR supplementation could significantly regulate the gut's microbial composition, increasing the abundance of Actinobacteria and Romboutsia. In addition, the BBR supplement altered the profile of bile acid. Furthermore, the gut microbiota participates in bile acid metabolism, especially taurochenodeoxycholic acid and α-muricholic acid. BBR supplementation could regulate the expression of genes and proteins related to glucose metabolism, lipid synthesis (FAS, SREBP-1c), and bile acid synthesis (FXR, CYP27a1). Collectively, our findings demonstrate that BBR might be a potential feed additive for preventing FLHS by regulating the gut microbiota and bile acid metabolism.

Hexavalent chromium (Cr(VI)) is a hazardous substance that poses significant risks to environmental ecosystems and animal organisms. However, the specific consequences of Cr(VI) exposure in terms of liver damage remain incompletely understood. This study aims to elucidate the mechanism by which Cr(VI) disrupts mitochondrial dynamics, leading to hepatic injury in ducks. Forty-eight healthy 8-day-old ducks were divided into four groups and subjected to diets containing varying doses of Cr(VI) (0, 9.28, 46.4, and 232 mg/kg) for 49 days. Our results demonstrated that Cr(VI) exposure resulted in disarranged liver lobular vacuolation, along with increasing the serum levels of ALT, AST, and AKP in a dose-dependent manner, which indicated liver damage. Furthermore, Cr(VI) exposure induced oxidative stress by reducing the activities of T-SOD, SOD, GSH-Px, GSH, and CAT, while increasing the contents of MDA and H 2 O 2 . Moreover, Cr(VI) exposure downregulated the activities of CS and MDH, resulting in energy disturbance, as evidenced by the reduced AMPK/p-AMPK ratio and PGC-1α protein expression. Additionally, Cr(VI) exposure disrupted mitochondrial dynamics through decreased expression of OPA1, Mfn1, and Mfn2 and increased expression of Drp-1, Fis1, and MFF proteins. This disruption ultimately triggered mitochondria-mediated apoptosis, as evidenced by elevated levels of caspase-3, Cyt C, and Bax, along with decreased expression of Bcl-2 and the Bcl-2/Bax ratio, at both the protein and mRNA levels. In summary, this study highlights that Cr(VI) exposure induces oxidative stress, inhibits the AMPK-PGC-1α pathway, disrupts mitochondrial dynamics, and triggers liver cell apoptosis in ducks.

Adding discrete fibres to sand has been seen as a feasible technique to improve sand's strength as well as liquefaction resistance. Considering the anisotropic distribution of fibre orientations, the anisotropy in the liquefaction resistance of the reinforced sand is also introduced using fibres. Here, the triaxial compression and extension test results of unreinforced and fibre-reinforced sand in different density states are provided, from which the anisotropy in the liquefaction resistance of fibre-reinforced sand is demonstrated. Fibre reinforcement improves the liquefaction resistance of sand by introducing both the densifying effect and the confining effect. The inclusion of fibres increases both the slope and the intercept of the strength envelope in comparison with the unreinforced sand under triaxial compression, while the strength envelope is not affected by fibres under triaxial extension. Stress contribution of fibres makes the ESP of the composite under undrained loading reverse its direction to develop even though the phase transformation is absent. The stress ratio initiating the ESP reversal is irrespective of the fibre content but dependent on the density state under triaxial compression. Under triaxial extension, the stress ratio initiating the ESP reversal remains the same in the samples with varied density states and fibre contents. The mechanism correlating to the strength envelope and ESP reversal of the fibre-reinforced sand was demonstrated following a rule of mixture based constitutive modelling framework. By introducing an alternatively defined pore pressure ratio that incorporates the stress contribution of fibres, the liquefaction state of the fibre reinforced sand is reasonably assessed. Liquefaction remains absent in the sand once the fibres are mixed. The anisotropy in the liquefaction resistance of fibre-reinforced sand arises, as the predominant role played by the fibres to suppress the liquefaction is different when varied loading paths are involved, which is sourced from the anisotropic distribution of fibre orientations.

Canthaxanthin(CX) is a ketocarotenoid, which is widely used in poultry production as a lipophilic antioxidant. Huaixiang chickens are a local breed in China famous for their excellent meat quality; improving their laying rate via nutritional regulation has attracted extensive attention. The aim of this study was to evaluate the effects of dietary CX on the laying rate and follicular development in Huaixiang hens. A total of 180 Huaixiang hens were randomly divided into five groups with six replicates, and six chickens per replication. The control group (CON) were fed a basal diet, and the treatment group (NT) were fed a basal diet supplemented with 4, 6, 8 and 10 mg/kg CX. All chickens were 26 weeks old, living at an average environmental temperature of 25 ± 2 °C with a relative humidity of 65-75%. The results showed that supplementing the CX improved the laying rate and large white follicles (LWF) number ( p < 0.05) and increased the concentration of reproductive hormones (LH, FSH, E 2 and Prog) ( p < 0.05), and the basal diet supplemented with 6 mg/kg CX worked best. Moreover, CX could increase the activities of antioxidant enzymes SOD and GSH-Px ( p < 0.05) and reduce the content of the lipid peroxidation product MDA in Huaixiang chickens ( p < 0.05); again, 6 mg/kg CX was best. In conclusion, dietary CX had positive effects on the laying rate, ovarian structure, reproductive hormone secretion, follicle development, and the antioxidant capacity of Huaixiang hens, and 6 mg/kg CX was recommended to be added to the diet of Huaixiang chickens.

Deep learning (DL) models in breast ultrasound (BUS) image analysis face challenges with data imbalance and limited atypical tumor samples. Generative Adversarial Networks (GAN) address these challenges by providing efficient data augmentation for small datasets. However, current GAN approaches fail to capture the structural features of BUS and generated images lack structural legitimacy and are unrealistic. Furthermore, generated images require manual annotation for different downstream tasks before they can be used. Therefore, we propose a two-stage GAN framework, 2s-BUSGAN, for generating annotated BUS images. It consists of the Mask Generation Stage (MGS) and the Image Generation Stage (IGS), generating benign and malignant BUS images using corresponding tumor contours. Moreover, we employ a Feature-Matching Loss (FML) to enhance the quality of generated images and utilize a Differential Augmentation Module (DAM) to improve GAN performance on small datasets. We conduct experiments on two datasets, BUSI and Collected. Moreover, results indicate that the quality of generated images is improved compared with traditional GAN methods. Additionally, our generated images underwent evaluation by ultrasound experts, demonstrating the possibility of deceiving doctors. A comparative evaluation showed that our method also outperforms traditional GAN methods when applied to training segmentation and classification models. Our method achieved a classification accuracy of 69% and 85.7% on two datasets, respectively, which is about 3% and 2% higher than that of the traditional augmentation model. The segmentation model trained using the 2s-BUSGAN augmented datasets achieved DICE scores of 75% and 73% on the two datasets, respectively, which were higher than the traditional augmentation methods. Our research tackles imbalanced and limited BUS image data challenges. Our 2s-BUSGAN augmentation method holds potential for enhancing deep learning model performance in the field.

This research systematically analyzes the behaviors of correlations among stock prices and the eigenvalues for correlation matrices by utilizing random matrix theory (RMT) for Chinese and US stock markets. Results suggest that most eigenvalues of both markets fall within the predicted distribution intervals by RMT, whereas some larger eigenvalues fall beyond the noises and carry market information. The largest eigenvalue represents the market and is a good indicator for averaged correlations. Further, the average largest eigenvalue shows similar movement with the index for both markets. The analysis demonstrates the fraction of eigenvalues falling beyond the predicted interval, pinpointing major market switching points. It has identified that the average of eigenvector components corresponds to the largest eigenvalue switch with the market itself. The investigation on the second largest eigenvalue and its eigenvector suggests that the Chinese market is dominated by four industries whereas the US market contains three leading industries. The study later investigates how it changes before and after a market crash, revealing that the two markets behave differently, and a major market structure change is observed in the Chinese market but not in the US market. The results shed new light on mining hidden information from stock market data.

Coating the perovskite layer via a two-step method is an adaptable solution for industries compared to the anti-solvent process. But what about the impact of unreacted PbI 2 ? Usually, it is generated during perovskite conversion in a two-step method and considered beneficial within the grain boundaries, while also being accused of enhancing the interface defects and nonradiative recombination. Several additives are mixed in PbI 2 precursors for the purpose of improving the perovskite crystallinity and hindering the Pb 2+ defects. Herein, in lieu of adding additives to the PbI 2 , the effects of the PbI 2 residue via the electron transport layer/perovskite interface modification are explored. Consequently, by introducing artemisinin decorated with hydrophobic alkyl units and a ketone group, it reduces the residual PbI 2 and improves the perovskites' crystallinity by coordinating with Pb 2+ . In addition, artemisinin-deposited perovskite enhances both the stability and efficiency of perovskite solar cells by suppressing nonradiative recombination.

Amino acid permeases (AAPs) are proteins of the integral membrane that play important roles in plant growth, development, and responses to various stresses. The molecular functions of several AAPs were characterized in Arabidopsis and rice, but there is still limited information on wheat. Here, we identified 51 AAP genes ( TaAAPs ) in the wheat genome, classified into six groups based on phylogenetic and protein structures. The chromosome location and gene duplication analysis showed that gene duplication events played a crucial role in the expansion of the TaAAPs gene family. Collinearity relationship analysis revealed several orthologous AAPs between wheat and other species. Moreover, cis-element analysis of promoter regions and transcriptome data suggested that the TaAAPs can respond to salt stress. A TaAAP1 gene was selected and transformed in wheat. Overexpressing TaAAP1 enhanced salt tolerance by increasing the expression of ethylene synthesis genes ( TaACS6 / TaACS7 / TaACS8 ) and accumulating more ethylene. The present study provides an overview of the AAP family in the wheat genome as well as information on systematics, phylogenetics, and gene duplication, and shows that overexpressing TaAAP1 enhances salt tolerance by regulating ethylene production. These results serve as a theoretical foundation for further functional studies on TaAAPs in the future.

With the emergence of high-throughput sequencing technology, a number of non-avian reptile species have been sequenced at the genome scale, shedding light on various scientific inquiries related to reptile ecology and evolution. However, the routine requirement of tissue or blood samples for genome sequencing often poses challenges in many elusive reptiles, hence limiting the application of high-throughput sequencing technologies to reptile studies. An alternative reptilian DNA resource suitable for genome sequencing is in urgent need. Here, we used the corn snake ( Pantherophis guttatus ) as a reptile model species to demonstrate that the shed skin is a high-quality DNA source for genome sequencing. Skin sheds provide a noninvasive type of sample that can be easily collected without restraining or harming the animal. Our findings suggest that shed skin from corn snakes yields DNA of sufficient quantity and quality that are comparable to tissue DNA extracts. Genome sequencing data analysis revealed that shed skin DNA is subject to bacteria contamination at variable levels, which is a major issue related to shed skin DNA and may be addressed by a modified DNA extraction method through introduction of a 30 min pre-digestion step. This study provides an enhanced method for the use of reptile shed skins as a high-quality DNA source for whole genome sequencing. Utilizing shed skin DNA enables researchers to overcome the limitations generally associated with obtaining traditional tissue or blood samples and promises to facilitate the application of genome sequencing in reptilian research.

To overcome the shortcomings of traditional wet styrene-butadiene-styrene (SBS) modification technology, such as its high energy consumption and thermal decomposition, a warm mix and fast-melting SBS modifier was developed. Based on the theory of rheology, a dynamic shear rheometer (DSR) was applied to investigate the viscoelastic properties of the warm mix and fast-melting SBS-modified asphalt using a frequency scanning test. Atomic force microscopy (AFM) was used to reveal the modification mechanism of the SBS-modified asphalt. An investigation of the thermal stability of the asphalt binder was conducted using a thermogravimetric test (TG). The results exhibited that the SBS-modified asphalt had good viscoelastic properties, as well as thermal stability. The "bee structure" of the SBS-modified asphalt was finer and more stable. In addition, the adhesion and the Derjaguin-Muller-Toporov (DMT) modulus of the SBS-modified asphalt at a warm mixing speed was higher than that of regular SBS-modified asphalt.

Phenanthrene (PHE), as a structurally simple, tricyclic, polycyclic aromatic hydrocarbon (PAHs), is widely present in marine environments and organisms, with serious ecological and health impacts. It is crucial to study fast and simple high-sensitivity detection methods for phenanthrene in seawater for the environment and the human body. In this paper, a immunosensor was prepared by using a multi-wall carbon nanotube (MWCNTs)-chitosan oligosaccharide (COS) nanocomposite membrane loaded with phenanthrene antibody. The principle was based on the antibody-antigen reaction in the immune reaction, using the strong electron transfer ability of multi-walled carbon nanotubes, coupled with chitosan oligosaccharides with an excellent film formation and biocompatibility, to amplify the detection signal. The content of the phenanthrene in seawater was studied via differential pulse voltammetry (DPV) using a potassium ferricyanide system as a redox probe. The antibody concentration, pH value, and probe concentration were optimized. Under the optimal experimental conditions, the response peak current of the phenanthrene was inversely proportional to the concentration of phenanthrene, in the range from 0.5 ng·mL -1 to 80 ng·mL -1 , and the detection limit was 0.30 ng·mL -1 . The immune sensor was successfully applied to the detection of phenanthrene in marine water, with a recovery rate of 96.1~101.5%, and provided a stable, sensitive, and accurate method for the real-time monitoring of marine environments.

Due to the heterogeneity of ultrasound (US) images and the indeterminate US texture of liver fibrosis (LF), automatic evaluation of LF based on US images is still challenging. Thus, this study aimed to propose a hierarchical Siamese network that combines the information from liver and spleen US images to improve the accuracy of LF grading. There were two stages in the proposed method. In stage one, a dual-channel Siamese network was trained to extract features from paired liver and spleen patches that were cropped from US images to avoid vascular interferences. Subsequently, the L1 distance was used to quantify the liver-spleen differences (LSDs). In stage two, the pretrained weights from stage one were transferred into the Siamese feature extractor of the LF staging model, and a classifier was trained using the fusion of the liver and LSD features for LF staging. This study was retrospectively conducted on US images of 286 patients with histologically proven liver fibrosis stages. Our method achieved a precision and sensitivity of 93.92% and 91.65%, respectively, for cirrhosis (S4) diagnosis, which is about 8% higher than that of the baseline model. The accuracy of the advanced fibrosis (≥S3) diagnosis and the multi-staging of fibrosis (≤S2 vs. S3 vs. S4) both improved about 5% to reach 90.40% and 83.93%, respectively. This study proposed a novel method that combined hepatic and splenic US images and improved the accuracy of LF staging, which indicates the great potential of liver-spleen texture comparison in noninvasive assessment of LF based on US images.

Oxidative stress can adversely affect the health status of the body, more specifically by causing intestinal damage by disrupting the permeability of the intestinal barrier. This is closely related to intestinal epithelial cell apoptosis caused by the mass production of reactive oxygen species (ROS). Baicalin (Bai) is a major active ingredient in Chinese traditional herbal medicine that has antioxidant, anti-inflammatory, and anti-cancer properties. The purpose of this study was to explore the underlying mechanisms by which Bai protects against hydrogen peroxide (H 2 O 2 )-induced intestinal injury in vitro. Our results indicated that H 2 O 2 treatment caused injury to IPEC-J2 cells, resulting in their apoptosis. However, Bai treatment attenuated H 2 O 2 -induced IPEC-J2 cell damage by up-regulating the mRNA and protein expression of ZO-1, Occludin, and Claudin1. Besides, Bai treatment prevented H 2 O 2 -induced ROS and MDA production and increased the activities of antioxidant enzymes (SOD, CAT, and GSH-PX). Moreover, Bai treatment also attenuated H 2 O 2 -induced apoptosis in IPEC-J2 cells by down-regulating the mRNA expression of Caspase-3 and Caspase-9 and up-regulating the mRNA expression of FAS and Bax, which are involved in the inhibition of mitochondrial pathways. The expression of Nrf2 increased after treatment with H 2 O 2 , and Bai can alleviate this phenomenon. Meanwhile, Bai down-regulated the ratio of phosphorylated AMPK to unphosphorylated AMPK, which is indicative of the mRNA abundance of antioxidant-related genes. In addition, knockdown of AMPK by short-hairpin RNA (shRNA) significantly reduced the protein levels of AMPK and Nrf2, increased the percentage of apoptotic cells, and abrogated Bai-mediated protection against oxidative stress. Collectively, our results indicated that Bai attenuated H 2 O 2 -induced cell injury and apoptosis in IPEC-J2 cells through improving the antioxidant capacity through the inhibition of the oxidative stress-mediated AMPK/Nrf2 signaling pathway.

Yangmai-13 (YM13) is a wheat cultivar with weak gluten fractions. In contrast, Zhenmai-168 (ZM168) is an elite wheat cultivar known for its strong gluten fractions and has been widely used in a number of breeding programs. However, the genetic mechanisms underlying the gluten signatures of ZM168 remain largely unclear. To address this, we combined RNA-seq and PacBio full-length sequencing technology to unveil the potential mechanisms of ZM168 grain quality. A total of 44,709 transcripts were identified in Y13N (YM13 treated with nitrogen) and 51,942 transcripts in Z168N (ZM168 treated with nitrogen), including 28,016 and 28,626 novel isoforms in Y13N and Z168N, respectively. Five hundred and eighty-four differential alternative splicing (AS) events and 491 long noncoding RNAs (lncRNAs) were discovered. Incorporating the sodium-dodecyl-sulfate (SDS) sedimentation volume (SSV) trait, both weighted gene coexpression network analysis (WGCNA) and multiscale embedded gene coexpression network analysis (MEGENA) were employed for network construction and prediction of key drivers. Fifteen new candidates have emerged in association with SSV, including 4 transcription factors (TFs) and 11 transcripts that partake in the post-translational modification pathway. The transcriptome atlas provides new perspectives on wheat grain quality and would be beneficial for developing promising strategies for breeding programs.

Sensor ontology provides a standardized semantic representation for information sharing between sensor devices. However, due to the varied descriptions of sensor devices at the semantic level by designers in different fields, data exchange between sensor devices is hindered. Sensor ontology matching achieves data integration and sharing between sensors by establishing semantic relationships between sensor devices. Therefore, a niching multi-objective particle swarm optimization algorithm (NMOPSO) is proposed to effectively solve the sensor ontology matching problem. As the sensor ontology meta-matching problem is essentially a multi-modal optimization problem (MMOP), a niching strategy is introduced into MOPSO to enable the algorithm to find more global optimal solutions that meet the needs of different decision makers. In addition, a diversity-enhancing strategy and an opposition-based learning (OBL) strategy are introduced into the evolution process of NMOPSO to improve the quality of sensor ontology matching and ensure the solutions converge to the real Pareto fronts (PFs). The experimental results demonstrate the effectiveness of NMOPSO in comparison to MOPSO-based matching techniques and participants of the Ontology Alignment Evaluation Initiative (OAEI).

It is well established that PRRSV elimination is an effective strategy for PRRS control, but published reports concerning successful PRRSV elimination cases in farrow-to-finishing herds are rare. Here, we have reported a successful PRRSV elimination case in a farrow-to-finish herd by employing a "herd closure and rollover" approach with some modifications. Briefly, the introduction of pigs to the herd was stopped and normal production processes were maintained until the herd reached a PRRSV provisional negative status. During the herd closure, strict biosecurity protocols were implemented to prevent transmission between nursery pigs and sows. In the current case, introducing gilts before herd closure and live PRRSV exposure were skipped. In the 23rd week post-outbreak, the pre-weaning piglets started to show 100% PRRSV negativity in qPCR tests. In the 27th week, nursery and fattening barns fully launched depopulation. In the 28th week, nursery and fattening houses reopened and sentinel gilts were introduced into gestation barns. Sixty days post-sentinel gilt introduction, the sentinel pigs maintained being PRRSV antibody negative, manifesting that the herd matched the standard of the provisional negative status. The production performance of the herd took 5 months to bounce back to normal. Overall, the current study provided additional information for PRRSV elimination in farrow-to-finish pig herds.

Two polysaccharides, named APS2-I and APS3-I, were purified from the water extract of Radix Astragali. The average molecular weight of APS2-I was 1.96 × 10 6 Da and composed of Man, Rha, GlcA, GalA, Glc, Gal, Xyl, and Ara in a molar ratio of 2.3:4.8:1.7:14.0:5.8:11.7:2.8:12.6, while the average molecular weight of APS3-I was 3.91 × 10 6 Da and composed of Rha, GalA, Glc, Gal, and Ara in a molar ratio of 0.8:2.3:0.8:2.3:4.1. Biological evaluation showed APS2-I and APS3-I had significant antioxidant activity and myocardial protection activity. Furthermore, total polysaccharide treatment could significantly enhance hemodynamic parameters and improve cardiac function in rat ischemia and reperfusion isolated heart models. These results provided important information for the clinical application of APS in the field of cardiovascular disease and implied that Astragalus polysaccharides (APS) could be considered as a reference for the quality control of Radix Astragali.

Unlike conventional lead ion (Pb2+) detecting methods, electrochemical methods have the attractive advantages of rapid response, good portability and high sensitivity. In this paper, a planar disk electrode modified by multiwalled carbon nanotube (MWCNTs)/chitosan (CS)/lead (Pb2+) ionophore IV nanomaterial and its matched system are proposed. This system presented a good linear relationship between the concentration of Pb2+ ions and the peak current in differential pulse stripping voltammetry (DPSV), under optimized conditions of -0.8 V deposition potential, 5.5 pH value, 240 s deposition time, performed sensitive detection of Pb2+ within sensitivity of 1.811 μA · μg -1 and detection limit of 0.08 μg · L -1 . Meanwhile, the results of the system in detecting lead ions in real seawater samples are highly similar to that of inductively coupled plasma emission spectrometer (ICP-MS), which proved a practicability for the system in detection of trace-level Pb2+.

Usher syndrome (USH) is characterised by degenerative vision loss known as retinitis pigmentosa (RP), sensorineural hearing loss, and vestibular dysfunction. RP can cause degeneration and the loss of rod and cone photoreceptors, leading to structural and functional changes in the retina. Cep250 is a candidate gene for atypical Usher syndrome, and this study describes the development of a Cep250 KO mouse model to investigate its pathogenesis. OCT and ERG were applied in Cep250 and WT mice at P90 and P180 to access the general structure and function of the retina. After recording the ERG responses and OCT images at P90 and P180, the cone and rod photoreceptors were visualised using an immunofluorescent stain. TUNEL assays were applied to observe the apoptosis in Cep250 and WT mice retinas. The total RNA was extracted from the retinas and executed for RNA sequencing at P90. Compared with WT mice, the thickness of the ONL, IS/OS, and whole retina of Cep250 mice was significantly reduced. The a-wave and b-wave amplitude of Cep250 mice in scotopic and photopic ERG were lower, especially the a-wave. According to the immunostaining and TUNEL stain results, the photoreceptors in the Cep250 retinas were also reduced. An RNA-seq analysis showed that 149 genes were upregulated and another 149 genes were downregulated in Cep250 KO retinas compared with WT mice retinas. A KEGG enrichment analysis indicated that cGMP-PKG signalling pathways, MAPK signalling pathways, edn2-fgf2 axis pathways, and thyroid hormone synthesis were upregulated, whereas protein processing in the endoplasmic reticulum was downregulated in Cep250 KO eyes. Cep250 KO mice experience a late-stage retinal degeneration that manifests as the atypical USH phenotype. The dysregulation of the cGMP-PKG-MAPK pathways may contribute to the pathogenesis of cilia-related retinal degeneration.

It is well known that solid diet supplementation in early life can significantly promote rumen development and metabolic function in young ruminants. However, the changes in the expressed proteome and related metabolism in rumen epithelium in response to a supplemented solid diet remain unclear. In this study, rumen epithelial tissue from goats in three diet regimes including milk replacer only (MRO), milk replacer supplemented concentrate (MRC), and milk replacer supplemented concentrate plus alfalfa pellets (MCA) were collected for measurement of the expression of epithelial proteins using proteomic technology (six per group). The results showed that solid diet significantly improved the growth performance of goats, enhanced the ability of rumen fermentation, and promoted the development of epithelial papilla ( p < 0.05). Proteome analysis revealed the distinct difference in the expressed protein in the MRC and MCA group compared with the MRO group (42 upregulated proteins and 79 downregulated proteins in MRC; 38 upregulated proteins and 73 downregulated proteins in MCA). Functional analysis showed that solid diet supplementation activated a variety of molecular functions in the epithelium, including protein binding, ATP binding, structural constituent of muscle, etc., in the MRC and MCA groups. Meanwhile, the expression of proteins related to fatty acid metabolism, the PPAR signaling pathway, valine, leucine, and isoleucine degradation, and butanoate metabolism were upregulated, being stimulated by solid feed. In contrast, the proteins associated with carbohydrate digestion and absorption and glycosaminoglycan degradation were downregulated. In addition, the protein expression of enzymes involved in ketone body synthesis in the rumen was generally activated, which was caused by solid feed. In summary, solid feed promoted the development of rumen epithelium by changing the expression of proteins related to fatty acid metabolism, energy synthesis, and signal transduction. The ketone body synthesis pathway might be the most important activated pathway, and provides energy for rumen development.

Peanut ( Arachis hypogaea L.) is an important food and feed crop worldwide and is affected by various biotic and abiotic stresses. The cellular ATP levels decrease significantly during stress as ATP molecules move to extracellular spaces, resulting in increased ROS production and cell apoptosis. Apyrases ( APYs ) are the nucleoside phosphatase (NPTs) superfamily members and play an important role in regulating cellular ATP levels under stress. We identified 17 APY homologs in A. hypogaea ( AhAPYs ), and their phylogenetic relationships, conserved motifs, putative miRNAs targeting different AhAPYs , cis -regulatory elements, etc., were studied in detail. The transcriptome expression data were used to observe the expression patterns in different tissues and under stress conditions. We found that the AhAPY2-1 gene showed abundant expression in the pericarp. As the pericarp is a key defense organ against environmental stress and promoters are the key elements regulating gene expression, we functionally characterized the AhAPY2-1 promoter for its possible use in future breeding programs. The functional characterization of AhAPY2-1P in transgenic Arabidopsis plants showed that it effectively regulated GUS gene expression in the pericarp. GUS expression was also detected in flowers of transgenic Arabidopsis plants. Overall, these results strongly suggest that APYs are an important future research subject for peanut and other crops, and AhPAY2-1P can be used to drive the resistance-related genes in a pericarp-specific manner to enhance the defensive abilities of the pericarp.

Rapid urbanization has led to a series of environmental problems that are harmful to the physical and mental health of residents. Not only can increasing urban tree canopy (UTC) improve sustainable city development, but it can also effectively promote the quality of life for residents; however, the uneven spatial distribution of UTC can also bring about social justice problems. Currently, there are few studies related to the justice of UTC distribution in China. Based on this, the paper adopts object-oriented image classification technology to interpret and extract UTC data from satellite images, and it uses house price to explore the justice of the spatial distribution of UTC in the main urban area of Guangzhou from the perspective of environmental justice by ANOVA, Pearson correlation coefficient, and bivariate local spatial autocorrelation. The results show that: (1) There is a significant positive correlation between UTC and house price in the main urban area of Guangzhou, and there are regional differences in the distribution of UTC, with the UTC in the "very high" house price group being significantly higher than that in other groups. (2) The spatial clustering of UTC and house price in the main urban area of Guangzhou is found to be "low-low" and "high-high"; thus, it can be concluded that the spatial distribution of UTC in the main urban area of Guangzhou is uneven. This is an environmental injustice, as the areas with low UTC are spatially clustered in old residential areas, while the areas with high UTC are spatially clustered in commercial housing estates with high house prices. The study shows that urban tree planting should not only focus on quantitative improvement but also on equitable spatial layouts to promote social equity and justice thereby improving the urban ecological environment and promoting healthy urban development.

Novel monomethoxy poly(ethylene glycol) ( m PEG) modified hydroxylated tung oil (HTO), denoted as m PEG-HTO- m PEG, was designed and synthesized for drug delivery. m PEG-HTO- m PEG consists of a hydroxylated tung oil center joined by two m PEG blocks via a urethane linkage. The properties of m PEG-HTO- m PEG were affected by the length of the mPEG chain. Three m PEG with different molecular weights were used to prepare m PEG-HTO- m PEG. The obtained three m PEG-HTO- m PEG polymers were characterized by nuclear magnetic resonance (NMR), Fourier transformation infrared spectroscopy (FT-IR), differential scanning calorimetry (DSC) and gel permeation chromatography (GPC), respectively. Furthermore, the particle sizes of m PEG-HTO- m PEG micelles were evaluated by dynamic light scattering (DLS) and transmission electron microscope (TEM). A critical aggregation concentration (CAC) ranged from 7.28 to 11.73 mg/L depending on the chain length of mPEG. The drug loading and release behaviors of m PEG-HTO- m PEG were investigated using prednisone acetate as a model drug, and results indicated that hydrophobic prednisone acetate could be effectively loaded into m PEG-HTO- m PEG micelles and exhibited a long-term sustained release. Moreover, compared with HTO, m PEG-HTO- m PEG had no obvious cytotoxicity to HeLa and L929 cells. Therefore, monomethoxy poly(ethylene glycol) modified hydroxylated tung oil m PEG-HTO- m PEG may be a promising drug carrier.

Early metastasis and resistance to traditional therapy are responsible for the poor prognosis of pancreatic adenocarcinoma patients. Metal-dependent protein phosphatases (PPMs) have been proven to play a crucial role in the initiation and progression of various tumors. Nevertheless, the expression and function of distinct PPMs in pancreatic adenocarcinoma have not been fully elucidated. In this study, we investigated the mRNA expression level, prognostic value, and the relationship between the expression of PPMs and the tumor microenvironment in pancreatic adenocarcinoma using Oncomine, TCGA and GTEx, GEO, Kaplan-Meier plotter, STRING, GeneMANIA, and HPA databases and R packages. GO and KEGG analysis revealed that PPMs and their differential co-expression genes are attributed to cell-cell adhesion and immune cell infiltration. Among these, PPM1K was downregulated in the tissue and peripheral blood of PAAD patients, whose expression level was negatively related to poor prognosis. Further to this, PPM1K was found to play a role in the epithelial-mesenchymal transition and immune infiltration. ROC curves showed that PPM1K had a good predictive value for pancreatic adenocarcinoma. The knockdown of PPM1K markedly promoted the proliferation and migration of pancreatic cancer cells, confirming its role in tumor suppressor activity in PAAD. This study demonstrates the potential clinical utility of PPM1K in tumor immunotherapy and brings about novel insights into the prognostic value of PPM1K in pancreatic adenocarcinoma.

Objective: In this paper, we propose a Sine chaos mapping-based improved sparrow search algorithm (SSA) to optimize the BP neural network for trajectory prediction of inland river vessels because of the problems of poor accuracy and easy trapping in local optimum in BP neural networks., Method: First, a standard BP model is constructed based on the AIS data of ships in the Yangtze River section. A Sine-BP model is built using Sine chaos mapping to assign neural network weights and thresholds. Finally, a Sine-SSA-BP model is built using the sparrow search algorithm (SSA) to solve the optimal solutions of the neural network weights and thresholds., Result: The Sine-SSA-BP model effectively improves the initialized population of uniform distribution, and reduces the problem that population intelligence algorithms tend to be premature., Conclusions: The test results show that the Sine-SSA-BP neural network has higher prediction accuracy and better stability than conventional LSTM and SVM, especially in the prediction of corners, which is in good agreement with the real ship navigation trajectory.

Urban vitality is a major indicator used for evaluating the sustainability and attractiveness of an urban environment. Global experience indicates that urban vitality can be stimulated through a reasonable urban design. However, it remains incompletely understood in the literature which building-related indicators can substantially affect urban vitality in Asian countries. To give an insight into this question, our study took a step forward by focusing specifically on the influence of the three-dimensional built environment on urban vitality, based on which decision makers could enhance urban vitality from the perspective of vertical building design. A machine-learning-based framework was developed in this study. First, we utilized several building-related indicators to thoroughly measure the spatial characteristics of buildings at the township level. Second, the relationship between a three-dimensional built environment and urban vitality was revealed based on a combined use of the correlation method, scatter charts, and a random forest. In the random forest, both a benchmark and a new model were constructed to evaluate the importance of those building-related indicators. The results suggested that urban vitality was closely related to the three-dimensional built environment, which played an even more important role than common benchmark factors in stimulating urban vitality. The building coverage ratio, density of tall buildings, and floor area ratio were essential spatial drivers behind urban vitality. Therefore, urban designers and decision makers should not only take traditional factors into account but also carefully consider the potential influence of high-rise buildings and the outdoor thermal environment so that urban vitality can be enhanced. Our study's results can offer practical recommendations for improving urban vitality from the perspective of vertical building design. The proposed framework can also be used for measuring the potential influence of the three-dimensional built environment in other areas.

Pathogenic bacteria can trigger active defense responses in higher plants, leading to hypersensitive programmed cell death (PCD) to against those bacteria. However, related research on seaweeds is very limited. Pseudoalteromonas piscicida X-8 (PpX-8) has been identified as the pathogen that causes bleaching disease in commercially farmed Saccharina japonica. In this study, using an inoculation assay and microscopic observations, we found that the proportion of bleaching tissue pieces inoculated with PpX-8 extracellular compounds was significantly higher (p < 0.05) than that inoculated with heated extracellular compounds, indicating that the virulence factors of PpX-8 exist in extracellular compounds and they are heat-sensitive. Using TEM, we observed typical morphological characteristics of PCD after inoculation with extracellular compounds, including chloroplast shrinkage, cytoplasmic vacuolation, and intact mitochondrial structures. Moreover, we detected biochemical characteristics of PCD, such as 3′-OH ends resulting from DNA cleavage and caspase-3-like enzymatic activity, using a TUNEL assay and fluorescence staining. Therefore, PpX-8 extracellular compounds can induce PCD, thus triggering active defense responses in S. japonica. These results indicate that seaweeds and higher plants are conservative in their active defense responses against pathogenic bacteria. The results of this study lay the foundation for further investigation of the virulence mechanisms of PpX-8.

Robust and accurate visual feature tracking is essential for good pose estimation in visual odometry. However, in fast-moving scenes, feature point extraction and matching are unstable because of blurred images and large image disparity. In this paper, we propose an unsupervised monocular visual odometry framework based on a fusion of features extracted from two sources, that is, the optical flow network and the traditional point feature extractor. In the training process, point features are generated for scene images and the outliers of matched point pairs are filtered by FlannMatch. Meanwhile, the optical flow network constrained by the principle of forward-backward flow consistency is used to select another group of corresponding point pairs. The Euclidean distance between the matching points found by FlannMatch and the corresponding point pairs by the flow network is added to the loss function of the flow network. Compared with SURF, the trained flow network shows more robust performance in complicated fast-motion scenarios. Furthermore, we propose the AvgFlow estimation module, which selects one group of the matched point pairs generated by the two methods according to the scene motion. The camera pose is then recovered by Perspective-n-Point (PnP) or the epipolar geometry. Experiments conducted on the KITTI Odometry dataset verify the effectiveness of the trajectory estimation of our approach, especially in fast-moving scenarios.

Neoagaro-oligosaccharides (NAOs) belong to the algae oligosaccharides. NAOs have been found to have diverse biological activities. However, the effects of NAOs on depression and their underlying mechanism have not been thoroughly studied. A chronic restraint stress (CRS)-induced C57BL/6J mouse model was used to assess the antidepressant effects of NAOs. Anxiety and depression behaviors were assessed by open field tests (OFT) and forced swimming tests (FST), while interleukin 18 (IL-18), 5-hydroxytryptamine (5-HT) and brain-derived neurotrophic factor (BDNF) were the molecular biomarkers of depression. Fecal microbiota transplantation (FMT) was performed. The results showed that NAO treatment significantly improved the body weight of depressed mice and reduced the central area time in the OFT and immobility time in the FST. NAO treatment decreased the levels of IL-18 in the serum and increased the levels of 5-HT in the serum and whole brain and of BDNF in the whole brain. NAO treatment mitigated the gut microbiota dysbiosis in the depressed mice and reversed the decreased levels of short-chain fatty acids (SCFAs) in the cecum of the depressed mice. FMT indicated that the gut microbiota is, indeed, linked to depression, which was reflected in the changes in weight gain and behaviors. In a word, NAOs effectively reversed the CRS-induced mice model of depression, which depended on the changes in the gut microbiota and SCFAs, as well as its modulation of 5-HT and BDNF.

The color of fruit peel is an economically important character of eggplant, and black-purple eggplant has received much attention for being rich in anthocyanin. However, the reason why different fruit peel colors form in eggplant is not well understood. In the present study, an integrative analysis of the metabolome and transcriptome profiles was performed in five eggplant varieties with different fruit colors. A total of 260 flavonoids were identified, and most of them showed significantly higher abundance in black-purple varieties than in other varieties. The transcriptome analysis indicated the activation of early phenylpropanoid biosynthesis genes ( SmPAL , SmC4H, and Sm4CL ) was more responsible for anthocyanin accumulation, while SmF3'5'H was the key factor for the formation of a purple color. Furthermore, two transcription factors, SmGL2 and SmGATA26, were identified as new hub genes associated with anthocyanin accumulation. The silencing of SmGL2 and SmGATA26 reduced anthocyanin accumulation in eggplant fruit peels, suggesting the possible involvement of SmGL2 and SmGATA26 in regulating anthocyanin biosynthesis. In addition, the pathway of plant hormone signal transduction was significantly enriched, indicating that phytohormones may cooperatively interact to modulate flavonoid biosynthesis. This study provides comprehensive information of flavonoid metabolites and new insights into the regulatory network of fruit coloration, which might be useful for the molecular breeding of eggplant.

Exposure to ionizing radiation (IR) is a lingering threat from accidental or terroristic nuclear events, but is also widely used in cancer therapy. In both cases, host inflammatory responses to IR damage normal tissue causing morbidity and possibly mortality to the victim/patient. Opaganib, a first-in-class inhibitor of sphingolipid metabolism, has broad anti-inflammatory and anticancer activity. Opaganib elevates ceramide and reduces sphingosine 1-phosphate (S1P) in cells, conditions that increase the antitumor efficacy of radiation while concomitantly suppressing inflammatory damage to normal tissue. Therefore, opaganib may suppress toxicity from unintended IR exposure and improve patient response to chemoradiation. To test these hypotheses, we first examined the effects of opaganib on the toxicity and antitumor activity of radiation in mice exposed to total body irradiation (TBI) or IR with partial bone marrow shielding. Oral treatment with opaganib 2 h before TBI shifted the LD 75 from 9.5 Gy to 11.5 Gy, and provided substantial protection against gastrointestinal damage associated with suppression of radiation-induced elevations of S1P and TNFα in the small intestines. In the partially shielded model, opaganib provided dose-dependent survival advantages when administered 4 h before or 24 h after radiation exposure, and was particularly effective when given both prior to and following radiation. Relevant to cancer radiotherapy, opaganib decreased the sensitivity of IEC6 (non-transformed mouse intestinal epithelial) cells to radiation, while sensitizing PAN02 cells to in vitro radiation. Next, the in vivo effects of opaganib in combination with radiation were examined in a syngeneic tumor model consisting of C57BL/6 mice bearing xenografts of PAN02 pancreatic cancer cells and a cross-species xenograft model consisting of nude mice bearing xenografts of human FaDu cells. Mice were treated with opaganib and/or IR (plus cisplatin in the case of FaDu tumors). In both tumor models, the optimal suppression of tumor growth was attained by the combination of opaganib with IR (± cisplatin). Overall, opaganib substantially protects normal tissue from radiation damage that may occur through unintended exposure or cancer radiotherapy.

This bedside-to-bench study aimed to systematically investigate the value of applying BMP2-loaded calcium phosphate cement (BMP2-CPC) in the restoration of large-scale alveolar bone defects. Compared to deproteinized bovine bone (DBB), BMP2-CPC was shown to be capable of inducing a favorable pattern of bone regeneration and bone remodeling accompanied by active osteoclastogenesis and optimized biomaterial resorption when applied in reconstructive periodontally accelerated osteogenic orthodontics (PAOO) surgery. To verify the regulatory role of osteoclasts in the BMP2-CPC-induced pattern of bone regeneration, in vitro and in vivo studies were designed to elucidate the underlying mechanism. Our results revealed that osteoclasts played a multifaceted role (facilitating osteogenesis, bone remodeling and biomaterial resorption) in the BMP2-CPC-induced bone regeneration. Osteoclasts contributed to the osteogenic differentiation of mesenchymal stem cells (MSCs) by secreting calcium ions, CTHRC1 and PDGF-B. Moreover, the increased osteoclasts promoted the remodeling of new bone and BMP2-CPC resorption, leading to a harmonized replacement of biomaterials with mature bone. In conclusion, the in vitro and in vivo experimental results corresponded with the clinical results and showed the optimized properties of BMP2-CPC in activating osteoclast-driven bone regeneration and remodeling, thus indicating the highly promising prospects of BMP2-CPC as an ideal therapeutic for alveolar bone defects.

Fine particulate matter (PM 2.5 ) exposure has been linked to numerous adverse health effects, with some disadvantaged subgroups bearing a disproportionate exposure burden. Few studies have been conducted to estimate the exposure and inequality of different subgroups due to a lack of adequate characterization of disparities in exposure to air pollutants in urban areas, and a mechanistic understanding of the causes of these exposure inequalities. Based on a long-term series of PM 2.5 concentrations, this study analyzed the spatial and temporal characteristics of PM 2.5 in 31 provincial capital cities of China from 2000 to 2016 using the coefficient of variation and trend analyses. A health risk assessment of human exposure to PM 2.5 from 2000 to 2016 was then undertaken. A cumulative population-weighted average concentration method was applied to investigate exposures and inequality for education level, job category, age, gender and income population subgroups. The relationships between socioeconomic factors and PM 2.5 exposure concentrations were quantified using the geographically and temporally weighted regression model (GTWR). Results indicate that the PM 2.5 concentrations in most of the capital cities in the study experienced an increasing trend at a rate of 0.98 μg m -3 per year from 2000 to 2016. The proportion of the population exposed to high PM 2.5 (above 35 μg m -3 ) increased annually, mainly due to the increase of population migrating into north, east, south and central China. The higher educated, older, higher income and urban secondary industry share (SIS) subgroups suffered from the most significant environmental inequality, respectively. The per capita GDP, population size, and the share of the secondary industry played an essential role in unequal exposure to PM 2.5 .

Background and objectives: In clinical practice, we observed that the prognoses of patients with heart failure and atrial fibrillation were worse than those of patients with only heart failure or atrial fibrillation. The study aims to get a better understanding of the common pathogenesis of the two diseases and find new therapeutic targets. Materials and Methods: We downloaded heart failure datasets and atrial fibrillation datasets from the gene expression omnibus database. The common DEGs (differentially expressed genes) in heart failure and atrial fibrillation were identified by a series of bioinformatics methods. To better understand the functions and possible pathways of DEGs, we performed Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses. Results: We identified 22 up-regulated genes and 14 down-regulated genes in two datasets of heart failure and 475 up-regulated and 110 down-regulated genes in atrial fibrillation datasets. In addition, two co-upregulated (FRZB, SFRP4) and three co-downregulated genes (ENTPPL, AQP4, C1orf105) were identified. GO enrichment results showed that these common differentially expressed genes were mainly concentrated in the signal regulation of the Wnt pathway. Conclusions: We found five crucial genes in heart failure and atrial fibrillation, which may be potential therapeutic targets for patients with heart failure and atrial fibrillation.

Global navigation satellite system (GNSS) plays a crucial role in many fields, such as aerospace and transportation. Integrity is the measure of trust used in GNSS positioning especially in safety-critical applications. Advanced receiver autonomous integrity monitoring (ARAIM), taking full advantage of multi-constellation GNSS, shows huge potential to provide vertical navigation in civil aviation en route navigation and terminal approaches. However, the multi-constellation ARAIM also greatly exposes computational complexity and potential performance hazards in fault modes determination and fault-tolerant positioning. From the perspective of integrity risk control, rather than the pursuit of better positioning accuracy blindly for safety-critical applications, the concept of constellation dynamic selection is proposed and implemented in ARAIM and the performance analysis is discussed in this paper. Only the best two constellations which have the best vertical geometry performance are involved in ARAIM calculation anytime anywhere. The proposed method shows superiority in both integrity availability and computational complexity in both simulations and actual GNSS signal experiments. While the computational complexity is less than 10% of that using four constellations, 100% availability under LPV-200 criteria can be achieved in worldwide coverage experiment. The proposed method also overcomes the shortcomings of ARAIM with two fixed constellations and shows good robustness under depleted scenarios. Furthermore, the statistics results from observation stations proved the applicability and generality of the proposed method under current developing GNSS constellations.

Metabolism-associated fatty liver disease (MAFLD) is one of the most common causes of liver disease; however, the underlying processes remain unknown. This study aimed to investigate the changes of free fatty acids (FFA) on the expression of genes related to the AMP-activated protein kinase (AMPK) signaling pathway in the primary hepatocytes of laying hens. The primary hepatocytes of laying hens were treated with FFA (containing a 2:1 ratio of oleic and palmitic acids) for 24 h. FFA significantly increased lipid droplet accumulation, decreased glycogen synthesis, increased the levels of triglycerides (TG), total cholesterol (TC), reactive oxygen species (ROS), malondialdehyde (MDA), and glucose content in the supernatant (GLU) in the primary hepatocytes of laying hens, and decreased the levels of total antioxidant capacity (T-AOC) and superoxide dismutase (SOD), as well as mitochondrial membrane potential (MMP). The results of the PCR array combined with Western blotting experiments showed that the activity of AMPK was inhibited. Inhibition of AMPK signaling pathway decreases the expression of genes involved in fatty acid oxidation, increases the expression of genes involved in lipid synthesis, decreases the expression of genes involved in glycogen synthesis, increases the expression of genes involved in glycolysis, increases the expression of genes involved in oxidative stress, and increases the expression of genes involved in cell proliferation and apoptosis. Taken together, our results suggest that FFA can affect the homeostasis of the AMPK signaling pathway by altering energy metabolic homeostasis, inducing oxidative stress, and adjusting the onset of cell proliferation and apoptosis.
