Simple Summary: Cancer patients frequently develop a syndrome named cachexia that causes severe muscle loss and frailty, eventually representing the cause of death. Muscle atrophy and muscle weakness are characterized by massive degradation of endogenous proteins, potentially consequent to excessive disposal of mitochondria through the selective autophagic process of mitophagy. This study explored whether selectively silencing BNIP3, a mitophagy-related protein upregulated in the muscle of both mouse and human cancer hosts, could help in preventing muscle loss. Two distinct methodological silencing approaches were tested, either by electroporation of a plasmid or via adenoviral particle injection. Although the first method was ineffective in tumor-bearing mice, the adenovirus-based approach significantly reduced BNIP3 levels and moderately increased muscle fiber size, suggesting partial prevention of muscle atrophy. BNIP3 silencing also maintained mitochondrial mass without disrupting oxidative balance, highlighting BNIP3's central role in cancer cachexia and suggesting that targeting BNIP3 may help in supporting muscle health in cancer patients. Background and Aims: Cancer cachexia is a complex syndrome affecting most cancer patients and is directly responsible for about 20% of cancer-related deaths. Previous studies showed muscle proteolysis hyper-activation and mitophagy induction in tumor-bearing animals. While basal mitophagy is required for maintaining muscle mass and quality, excessive mitophagy promotes uncontrolled protein degradation, muscle loss and impaired function. BNIP3, a key mitophagy-related protein, is significantly increased in the muscles of both mice and human cancer hosts. This study aimed to define the potential of mitigating mitophagy via BNIP3 downregulation in preserving mitochondrial integrity, counteracting skeletal muscle loss in experimental cancer cachexia. Methods: Two in vivo gene delivery methods were performed to knock down muscle BNIP3: electroporation of a BNIP3-specific shRNA expression vector or adenovirus injection. Results: The electroporation effectively reduced muscle BNIP3 in healthy mice but was ineffective in C26 tumor-bearing mice. In contrast, adenovirus-mediated BNIP3 knockdown successfully decreased BNIP3 levels also in tumor hosts. Although BNIP3 knockdown did not impact overall on body or muscle mass, it improved muscle fiber size in C26-bearing miceh2, suggesting partial prevention of muscle atrophy. Mitochondrial respiratory chain complexes (OxPhos) and TOM20 protein levels were consistently rescued, indicating improvements in mitochondrial mass, while H2O2 levels were unchanged among the groups, suggesting that BNIP3 downregulation does not impair the endogenous control of oxidative balance. Conclusions: These findings suggest that a fine balance between mitochondrial disposal and biogenesis is fundamental for preserving muscle homeostasis and highlight a potential role for BNIP3 modulation against cancer-induced muscle wasting. Neuregulin 4 (Nrg4) is an adipokine that belongs to the epidermal growth factor family and binds to ErbB4 tyrosine kinase receptors. In 3T3-L1 adipocytes, the downregulation of Nrg4 expression enhances inflammation and autophagy, resulting in insulin resistance. Here, we searched for the causes of this phenotype. Nrg4 knockdown (Nrg4 KD) adipocytes showed a significant reduction in mitochondrial content and elongation, along with a lower content of the mitochondria fusion protein mitofusin 2 (MFN2), and increased H2O2 production compared to the control scrambled cells (Scr). The antioxidant N-acetylcysteine reversed the oxidative stress and reduced the gene expression of the pro-inflammatory cytokine tumor necrosis factor α (TNFα). Nrg4 KD adipocytes showed enhanced lipolysis and reduced lipogenesis, in addition to a significant reduction in several intermediates of the Krebs cycle. In summary, Nrg4 downregulation in adipocytes affects mitochondrial content and functioning, causing impaired cellular metabolism, which in turn results in oxidative stress, inflammation, and insulin resistance. Ozone plays a key role in both atmospheric chemistry and UV absorption in planetary atmospheres. On Mars, upper-tropospheric ozone has been widely characterized by space-based instruments. However, surface ozone remains poorly characterized, hindered by the limited sensitivity of orbiters to the lowest scale height of the atmosphere and challenges in delivering payloads to the surface of Mars, which have prevented, to date, the measurement of ozone from the surface of Mars. Systematic measurements from the Martian surface could advance our knowledge of the atmospheric chemistry and habitability potential of this planet. NASA's Mars 2020 mission includes the first ozone detector deployed on the Martian surface, which is based on discrete photometric observations in the ultraviolet band, a simple technology that could obtain the first insights into total ozone abundance in preparation for more sophisticated measurement techniques. This paper describes the Mars 2020 ozone detector and its retrieval algorithm, including its performance under different sources of uncertainty and the potential application of the retrieval algorithm on other missions, such as NASA's Mars Science Laboratory. Pre-landing simulations using the UVISMART radiative transfer model suggest that the retrieval is robust and that it can deal with common issues affecting surface operations in Martian missions, although the expected low ozone abundance and instrument uncertainties could challenge its characterization in tropical latitudes of the planet. Many natural and artificial liquid environments, such as rivers, oceans, lakes, water storage tanks, aquariums, and urban water distribution systems, are difficult to access. As a result, technology is needed to enable autonomous liquid sampling to monitor water quality and ecosystems. Existing in situ sample acquisition and handling systems for liquid environments are currently limited to a single use and are semi-autonomous, relying on an operator. Liquid sampling systems should be robust and light and withstand long-term operation in remote locations. The system components involved in liquid sampling should be sterilisable to ensure reusability. Here, we introduce a prototype of a liquid sampler that can be used in various liquid environments and may be valuable for the scientific characterisation of different natural, remote, and planetary settings. The Autonomous Planetary Liquid Sampler (APLS) is equipped with pre-programmed, fully autonomous extraction, cleaning, and sterilisation functionalities. It can operate in temperatures between −10 °C and 60 °C and pressure of up to 0.24 MPa (~24 m depth below mean sea level on Earth). As part of the control experiment, we demonstrate its safe and robust autonomous operation in a laboratory environment using a liquid media with Bacillus subtilis. A typical sampling procedure required 28 s to extract 250 mL of liquid, 5 s to fill the MilliQ water, 25 s for circulation within the system for cleaning and disposal, and 200 s to raise the system temperature from ~30 °C ambient laboratory temperature to 150 °C. The temperature is then maintained for another 3.2 h to sterilise the critical parts, allowing a setup reset for a new experiment. In the future, the liquid sampler will be combined with various existing analytical instruments to characterise the liquid solution and enable the autonomous, systematic monitoring of liquid environments on Earth. Background and Objectives: Modern classification and categorization of individuals' health requires personalized variables such as nutrition, physical activity, lifestyle, and medical data through advanced analysis and clustering methods involving machine learning tools. The objective of this project was to categorize Mediterranean dwellers' health factors and design metabotypes to provide personalized well-being in order to develop professional implementation tools in addition to characterizing nutritional and lifestyle features in such populations. Materials and Methods: A two-phase observational study was conducted by the Pharmacists Council to identify Spanish nutritional and lifestyle characteristics. Adults over 18 years of age completed questionnaires on general lifestyle habits, dietary patterns (FFQ, MEDAS-17 p), physical activity (IPAQ), quality of life (SF-12), and validated well-being indices (LS7, MEDLIFE, HHS, MHL). Subsequently, exploratory factor, clustering, and random forest analysis methods were conducted to objectively define the metabotypes considering population determinants. Results: A total of 46.4% of the sample (n = 5496) had moderate-to-high adherence to the Mediterranean diet (>8 points), while 71% of the participants declared that they had moderate physical activity. Almost half of the volunteers had a good self-perception of health (49.9%). Regarding lifestyle index, population LS7 showed a fair cardiovascular health status (7.9 ± 1.7), as well as moderate quality of life by MEDLIFE (9.3 ± 2.6) and MHL scores (2.4 ± 0.8). In addition, five metabotype models were developed based on 26 variables: Westernized Millennial (28.6%), healthy (25.1%), active Mediterranean (16.5%), dysmetabolic/pre-morbid (11.5%), and metabolically vulnerable/pro-morbid (18.3%). Conclusions: The support of tools related to precision nutrition and lifestyle integrates well-being characteristics and contributes to reducing the impact of unhealthy lifestyle habits with practical implications for primary care. Mitochondrial network architecture plays a critical role in cellular physiology. Indeed, alterations in the shape of mitochondria upon exposure to cellular stress can cause the dysfunction of these organelles. In this scenario, mitochondrial dynamics proteins and the phospholipid composition of the mitochondrial membrane are key for fine-tuning the modulation of mitochondrial architecture. In addition, several factors including post-translational modifications such as the phosphorylation, acetylation, SUMOylation, and o-GlcNAcylation of mitochondrial dynamics proteins contribute to shaping the plasticity of this architecture. In this regard, several studies have evidenced that, upon metabolic stress, mitochondrial dynamics proteins are post-translationally modified, leading to the alteration of mitochondrial architecture. Interestingly, several proteins that sustain the mitochondrial lipid composition also modulate mitochondrial morphology and organelle communication. In this context, pharmacological studies have revealed that the modulation of mitochondrial shape and function emerges as a potential therapeutic strategy for metabolic diseases. Adipocyte dysfunction is the driver of obesity and correlates with insulin resistance and the onset of type 2 diabetes. Protein kinase N1 (PKN1) is a serine/threonine kinase that has been shown to contribute to Glut4 translocation to the membrane and glucose transport. Here, we evaluated the role of PKN1 in glucose metabolism under insulin-resistant conditions in primary visceral adipose tissue (VAT) from 31 patients with obesity and in murine 3T3-L1 adipocytes. In addition, in vitro studies in human VAT samples and mouse adipocytes were conducted to investigate the role of PKN1 in the adipogenic maturation process and glucose homeostasis control. We show that insulin-resistant adipocytes present a decrease in PKN1 activation levels compared to nondiabetic control counterparts. We further show that PKN1 controls the adipogenesis process and glucose metabolism. PKN1-silenced adipocytes present a decrease in both differentiation process and glucose uptake, with a concomitant decrease in the expression levels of adipogenic markers, such as PPARγ, FABP4, adiponectin and CEBPα. Altogether, these results point to PKN1 as a regulator of key signaling pathways involved in adipocyte differentiation and as an emerging player of adipocyte insulin responsiveness. FTY720 is an FDA-approved sphingosine derivative drug for the treatment of multiple sclerosis. This compound blocks lymphocyte egress from lymphoid organs and autoimmunity through sphingosine 1-phosphate (S1P) receptor blockage. Drug repurposing of FTY720 has revealed improvements in glucose metabolism and metabolic diseases. Studies also demonstrate that preconditioning with this compound preserves the ATP levels during cardiac ischemia in rats. The molecular mechanisms by which FTY720 promotes metabolism are not well understood. Here, we demonstrate that nanomolar concentrations of the phosphorylated form of FTY720 (FTY720-P), the active ligand of S1P receptor (S1PR), activates mitochondrial respiration and the mitochondrial ATP production rate in AC16 human cardiomyocyte cells. Additionally, FTY720-P increases the number of mitochondrial nucleoids, promotes mitochondrial morphology alterations, and induces activation of STAT3, a transcription factor that promotes mitochondrial function. Notably, the effect of FTY720-P on mitochondrial function was suppressed in the presence of a STAT3 inhibitor. The Echus-Kasei region on Mars has been exposed to different episodic volcanic, fluvial, and glacial events in Amazonian time. The goal of the present work is to demonstrate the usefulness of radar instruments to find preserved late Amazonian subsurface structures that may have been encapsulated underneath recent lava flows on Mars. We have analysed 27 radar observations of the SHAllow RADar (SHARAD) instrument on board the Mars Reconnaissance Orbiter (MRO), over the region of Echus Chasma. We discovered the presence of subsurface reflectors in five consecutive SHARAD radargrams at a depth from 35 to 79 m beneath the structure of a lava fan that formed about 59 ± 4 Ma ago. Some vents are preserved above the surface of this lava flow, which stands at a height of 80 m above the surrounding surface. A few kilometres to the north, we find other subsurface reflectors at a depth of about 30 m and a long pit chain formed by the collapse of a lava tube. These kinds of subsurface late Amazonian structures are of interest for astrobiology because they date from the last period when the planet still experienced intense volcanic activity over regions that were previously extensively covered by water. The Earth's atmosphere plays a critical role in transporting and dispersing biological aerosols. Nevertheless, the amount of microbial biomass in suspension in the air is so low that it is extremely difficult to monitor the changes over time in these communities. Real-time genomic studies can provide a sensitive and rapid method for monitoring changes in the composition of bioaerosols. However, the low abundance of deoxyribose nucleic acid (DNA) and proteins in the atmosphere, which is of the order of the contamination produced by operators and instruments, poses a challenge for the sampling process and the analyte extraction. In this study, we designed an optimized, portable, closed bioaerosol sampler based on membrane filters using commercial off-the-shelf components, demonstrating its end-to-end operation. This sampler can operate autonomously outdoors for a prolonged time, capturing ambient bioaerosols and avoiding user contamination. We first performed a comparative analysis in a controlled environment to select the optimal active membrane filter based on its ability to capture and extract DNA. We have designed a bioaerosol chamber for this purpose and tested three commercial DNA extraction kits. The bioaerosol sampler was tested outdoors in a representative environment and run for 24 h at 150 L/min. Our methodology suggests that a 0.22-µm polyether sulfone (PES) membrane filter can recover up to 4 ng of DNA in this period, sufficient for genomic applications. This system, along with the robust extraction protocol, can be automated for continuous environmental monitoring to gain insights into the time evolution of microbial communities within the air. CBL is rapidly phosphorylated upon insulin receptor activation. Mice whole body CBL depletion improved insulin sensitivity and glucose clearance; however, the precise mechanisms remain unknown. We depleted either CBL or its associated protein SORBS1/CAP independently in myocytes and assessed mitochondrial function and metabolism compared to control cells. CBL- and CAP-depleted cells showed increased mitochondrial mass with greater proton leak. Mitochondrial respiratory complex I activity and assembly into respirasomes were reduced. Proteome profiling revealed alterations in proteins involved in glycolysis and fatty acid degradation. Simple Summary: Many countries have implemented, or are implementing, general age-based screening programs to reduce mortality due to colorectal cancer. Fecal hemoglobin immunodetection (FIT) in combination with endoscopy has already been relatively successful in achieving this goal. However, there are issues that can be improved in relation to participation rates and reduction in false positives. We studied whether the blood biomarker soluble-CD26 (sCD26), a glycoprotein with dipeptidyl peptidase enzyme activity (DPP4), could help in the early diagnosis of colorectal cancer and advanced adenomas in combination with FIT, also reducing false positives. We propose a sequential testing strategy for FIT positive individuals, offering an alternative blood test with our biomarker for a confirmation prior to colonoscopy in the short term. Fecal hemoglobin immunodetection (FIT) in combination with endoscopy has been implemented to reduce mortality from colorectal cancer (CRC), although there are issues that can be improved in relation to participation rates. We studied whether the blood biomarker soluble-CD26 (sCD26), related at least in part to the immune system and inflammation, and/or its dipeptidyl peptidase enzyme activity (DPP4), could help reduce false positives. In a cohort of 1703 individuals who underwent colonoscopy and had a serum sample, sCD26 and DPP4 activity showed statistically significant differences regarding sex and age. According to the colonoscopy findings, sCD26 and DPP4 activity progressively decreased in advanced adenomas and CRC, with statistically significant differences, even between both groups; 918 of them had a FIT result (n = 596 positive cases) with approximately 70% of these (n = 412) false positives. With cut-offs of 440 ng/mL for sCD26, 42 mU/mL for DPP4, and 11 ng/mU for their ratio, the combined information of the three biomarkers (at least positive for one biomarker) identified almost all advanced adenomas and CRC cases in the FIT cohort with approximately half of the false positives compared to FIT. Flow (shear stress)-mediated dilation (FMD) of resistance arteries is a rapid endothelial response involved in tissue perfusion. FMD is reduced early in cardiovascular diseases, generating a major risk factor for atherosclerosis. As alteration of mitochondrial fusion reduces endothelial cells' (ECs) sprouting and angiogenesis, we investigated its role in ECs responses to flow. Opa1 silencing reduced ECs (HUVECs) migration and flow-mediated elongation. In isolated perfused resistance arteries, FMD was reduced in Opa1+/− mice, a model of the human disease due to Opa1 haplo-insufficiency, and in mice with an EC specific Opa1 knock-out (EC-Opa1). Reducing mitochondrial oxidative stress restored FMD in EC-Opa1 mice. In isolated perfused kidneys from EC-Opa1 mice, flow induced a greater pressure, less ATP, and more H2O2 production, compared to control mice. Opa1 expression and mitochondrial length were reduced in ECs submitted in vitro to disturbed flow and in vivo in the atheroprone zone of the mouse aortic cross. Aortic lipid deposition was greater in Ldlr−/--Opa1+/- and in Ldlr−/--EC-Opa1 mice than in control mice fed with a high-fat diet. In conclusion, we found that reduction in mitochondrial fusion in mouse ECs altered the dilator response to shear stress due to excessive superoxide production and induced greater atherosclerosis development. The construction of new high voltage overhead power lines (HVOPLs) has become a controversial issue for electricity companies due to social opposition. Citizens are concerned about how these power lines may have an impact on their lives, basically caused by their effects on health and safety. Visual impact is one of the most easily perceived. Although there are several published works that deal with the assessment of the visual impact produced by HVOPLs, no methodology has been proposed to assess this impact from an objective perspective. This work presents an original methodology which helps to identify the optimal routes for a new HVOPL under an objective observability criterion, enabling the selection of those with the lowest visibility in a zone. The application of the proposed methodology achieves a set of routes that links new HVOPL origin and destination points creating a corridor which includes all possible routes with an observability of its towers under a threshold limit. This methodology is illustrated by a real-life use corresponding to the selection of the route with least observability for a new power line in La Rioja (Spain). The results obtained may help to achieve a consensus between key stakeholders since it is focused on the specific issues of the planned HVOPL and its observability from an objective perspective. Browning of white adipocytes has been proposed as a powerful strategy to overcome metabolic complications, since brown adipocytes are more catabolic, expending energy as a heat form. However, the biological pathways involved in the browning process are still unclear. Aquaglyceroporins are a sub-class of aquaporin water channels that also permeate glycerol and are involved in body energy homeostasis. In the adipose tissue, aquaporin-7 (AQP7) is the most representative isoform, being crucial for white adipocyte fully differentiation and glycerol metabolism. The altered expression of AQP7 is involved in the onset of obesity and metabolic disorders. Herein, we investigated if aquaglyceroporins are implicated in beige adipocyte differentiation, similar to white cells. Thus, we optimized a protocol of murine 3T3-L1 preadipocytes browning that displayed increased beige and decreased white adipose tissue features at both gene and protein levels and evaluated aquaporin expression patterns along the differentiation process together with cellular lipid content. Our results revealed that AQP7 and aquaporin-9 (AQP9) expression was downregulated throughout beige adipocyte differentiation compared to white differentiation, which may be related to the beige physiological role of heat production from oxidative metabolism, contrasting with the anabolic/catabolic lipid metabolism requiring glycerol gateways occurring in white adipose cells. The impacts of climate change are affecting ski tourism in Europe’s southernmost ski resorts, such as those in France, and Spain. This is leading to changes in the scheduling of activities which, in turn, imply changes in how ski resorts are managed. The main aim of this work is to analyze whether ski resort websites are developing an effective marketing strategy and if they are adapted to e-commerce and the needs of contemporary society, including the adaptations needed to better face climate change and the stagnation the sector is suffering. In order to achieve this, we have developed a model to analyze ski resort websites; it is based on web content analysis, taking four factors into account: information, communication, e-commerce, and additional functions. We will, secondly, apply the eMICA (Extended Model of Internet Commerce Adoption) methodology to analyze the maturity of such e-commerce activities in ski tourism. Fifty-two ski resorts in Spain and 173 in France were analyzed. Results show that, while alpine ski resorts in general, and those in France in particular, are better prepared, resorts still have to facilitate more effective communication and interaction with their target public. We study the bias induced by a weak (200 mT) external magnetic field on the preferred handedness of sodium chlorate crystals obtained by slow evaporation at ambient conditions of its saturated saline solution with 20 ppm of added racemic (DL) hydrophobic amino acids. By applying the Fisher test to pairs of experiments with opposing magnetic field orientation we conclude, with a confidence level of 99.7%, that at the water-air interface of this saline solution there is an enantioselective magnetic interaction that acts upon racemic mixtures of hydrophobic chiral amino acids. This interaction has been observed with the three tested racemic hydrophobic amino acids: DL-Phe, DL-Try and DL-Trp, at ambient conditions and in spite of the ubiquitous chiral organic contamination. This enantioselective magnetic dependence is not observed when there is only one handedness of added chiral amino-acid, if the added amino acid is not chiral or if there is no additive. This effect has been confirmed with a double blind test. This novel experimental observation may have implications for our view of plausible initial prebiotic scenarios and of the roles of the geomagnetic field in homochirality in the biosphere. Abstract: Hydrogen cyanide (HCN) is a ubiquitous molecule in the Universe. It is a compound that is easily produced in significant yields in prebiotic simulation experiments using a reducing atmosphere. HCN can spontaneously polymerise under a wide set of experimental conditions. It has even been proposed that HCN polymers could be present in objects such as asteroids, moons, planets and, in particular, comets. Moreover, it has been suggested that these polymers could play an important role in the origin of life. In this review, the simple organics and biomonomers that have been detected in HCN polymers, the analytical techniques and procedures that have been used to detect and characterise these molecules and an exhaustive classification of the experimental/environmental conditions that favour the formation of HCN polymers are summarised. Nucleobases, amino acids, carboxylic acids, cofactor derivatives and other compounds have been identified in HCN polymers. In previous studies we described a decreased alpha-L-fucosidase activity in colorectal tumors, appearing as a prognostic factor of tumoral recurrence. The aim of this work was to extend the knowledge about tissue alpha-L-fucosidase in colorectal cancer by quantifying the expression of its encoding gene FUCA1 in tumors and healthy mucosa. FUCA1 mRNA levels were measured by RT-qPCR in paired tumor and normal mucosa tissues from 31 patients. For the accuracy of the RT-qPCR results, five candidate reference genes were validated in those samples. In addition, activity and expression of alpha-L-fucosidase in selected matched tumor and healthy mucosa samples were analyzed. According to geNorm and NormFinder algorithms, RPLP0 and HPRT1 were the best reference genes in colorectal tissues. These genes were used for normalization of FUCA1 expression levels. A significant decrease of more than 60% in normalized FUCA1 expression was detected in tumors compared to normal mucosa (p = 0.002). Moreover, a gradual decrease in FUCA1 expression was observed with progression of disease from earlier to advanced stages. These findings were confirmed by Western blot analysis of alpha-L-fucosidase expression. Our results demonstrated diminished FUCA1 mRNA levels in tumors, suggesting that expression of tissue alpha-L-fucosidase could be regulated at transcriptional level in colorectal cancer. Aiming to find new diagnostic proteins, we applied the proteomic DIGE technology to analyze changes in the secretome before/after differentiation of the colon adenocarcinoma Caco-2 cell line, an accepted in vitro model to study colorectal tumorigenesis. When the secretomes from undifferentiated (tumor-like) and differentiated cells (resembling healthy enterocytes) were compared, we found 96 spots differentially expressed. After MS/MS analysis, 22 spots corresponding to 15 different proteins were identified. Principal component analysis demonstrated these 22 spots could serve as a discriminatory panel between the tumor-like and normal-like cells. Among the identified proteins, the translationally-controlled tumor protein (TCTP), the transforming growth factor-beta-induced protein ig-h3 (TGFßIp), and the Niemann-Pick disease type C2 protein (NPC2) are interesting candidates for future studies focused on their utility as serum biomarkers of colorectal cancer. We describe the parameters that drive the design and modeling of the Rover Environmental Monitoring Station (REMS) Ground Temperature Sensor (GTS), an instrument aboard NASA's Mars Science Laboratory, and report preliminary test results. REMS GTS is a lightweight, low-power, and low cost pyrometer for measuring the Martian surface kinematic temperature. The sensor's main feature is its innovative design, based on a simple mechanical structure with no moving parts. It includes an in-flight calibration system that permits sensor recalibration when sensor sensitivity has been degraded by deposition of dust over the optics. Since the earliest development of the eye (and vision) around 530 million years ago (Mya), it has evolved, adapting to different habitats, species, and changing environmental conditions on Earth. We argue that a radiation environment determined by the atmosphere played a determining role in the evolution of vision, specifically on the human eye, which has three vision regimes (photopic-, scotopic-, and mesopic vision) for different illumination conditions. An analysis of the irradiance spectra, reaching the shallow ocean depths, revealed that the available radiation could have determined the bandwidth of the precursor to vision systems, including human vision. We used the radiative transfer model to test the existing hypotheses on human vision. We argue that, once on the surface, the human photopic (daytime) and scotopic (night-time) vision followed different evolutionary directions, maximum total energy, and optimum information, respectively. Our analysis also suggests that solar radiation reflected from the moon had little or no influence on the evolution of scotopic vision. Our results indicate that, apart from human vision, the vision of only a few birds, rodents, and deep-sea fish are strongly correlated to the available radiation within their respective habitats. The adipokine Neuregulin 4 (Nrg4) protects against obesity-induced insulin resistance. Here, we analyze how the downregulation of Nrg4 influences insulin action and the underlying mechanisms in adipocytes. Validated shRNA lentiviral vectors were used to generate scramble (Scr) and Nrg4 knockdown (KD) 3T3-L1 adipocytes. Adipogenesis was unaffected in Nrg4 KD adipocytes, but there was a complete impairment of the insulin-induced 2-deoxyglucose uptake, which was likely the result of reduced insulin receptor and Glut4 protein. Downregulation of Nrg4 enhanced the expression of proinflammatory cytokines. Anti-inflammatory agents recovered the insulin receptor, but not Glut4, content. Proteins enriched in Glut4 storage vesicles such as the insulin-responsive aminopeptidase (IRAP) and Syntaxin-6 as well as TBC1D4, a protein involved in the intracellular retention of Glut4 vesicles, also decreased by Nrg4 KD. Insulin failed to reduce autophagy in Nrg4 KD adipocytes, observed by a minor effect on mTOR phosphorylation, at the time that proteins involved in autophagy such as LC3-II, Rab11, and Clathrin were markedly upregulated. The lysosomal activity inhibitor bafilomycin A1 restored Glut4, IRAP, Syntaxin-6, and TBC1D4 content to those found in control adipocytes. Our study reveals that Nrg4 preserves the insulin responsiveness by preventing inflammation and, in turn, benefits the insulin regulation of autophagy. [ABSTRACT FROM AUTHOR]

The water content of the upper layers of the surface of Mars is not yet quantified. Laboratory simulations are the only feasible way to investigate this in a controlled way on Earth, and then compare it with remote and in situ observations of spacecrafts on Mars. Describing the processes that may induce changes in the water content of the surface is critical to determine the present-day habitability of the Martian surface, to understand the atmospheric water cycle, and to estimate the efficiency of future water extraction procedures from the regolith for In Situ Resource Utilization (ISRU). This paper illustrates the application of the SpaceQ facility to simulate the near-surface water cycle under Martian conditions. Rover Environmental Monitoring Station (REMS) observations at Gale crater show a non-equilibrium situation in the atmospheric H2O volume mixing ratio (VMR) at night-time, and there is a decrease in the atmospheric water content by up to 15 g/m2 within a few hours. This reduction suggests that the ground may act at night as a cold sink scavenging atmospheric water. Here, we use an experimental approach to investigate the thermodynamic and kinetics of water exchange between the atmosphere, a non-porous surface (LN2-chilled metal), various salts, Martian regolith simulant, and mixtures of salts and simulant within an environment which is close to saturation. We have conducted three experiments: the stability of pure liquid water around the vicinity of the triple point is studied in experiment 1, as well as observing the interchange of water between the atmosphere and the salts when the surface is saturated; in experiment 2, the salts were mixed with Mojave Martian Simulant (MMS) to observe changes in the texture of the regolith caused by the interaction with hydrates and liquid brines, and to quantify the potential of the Martian regolith to absorb and retain water; and experiment 3 investigates the evaporation of pure liquid water away from the triple point temperature when both the air and ground are at the same temperature and the relative humidity is near saturation. We show experimentally that frost can form spontaneously on a surface when saturation is reached and that, when the temperature is above 273.15 K (0 °C), this frost can transform into liquid water, which can persist for up to 3.5 to 4.5 h at Martian surface conditions. For comparison, we study the behavior of certain deliquescent salts that exist on the Martian surface, which can increase their mass between 32% and 85% by absorption of atmospheric water within a few hours. A mixture of these salts in a 10% concentration with simulant produces an aggregated granular structure with a water gain of approximately 18- to 50-wt%. Simple Summary: Hepatocellular carcinoma is a leading cause of cancer-related death worldwide. Major risk factors in liver cancer development include chronic hepatitis B or C virus, autoimmune hepatitis, diabetes mellitus, alcohol abuse, and several metabolic diseases, among others. Standard therapy shows low efficacy, and there is an urgent need for novel therapies. Recent data permit to propose that proteins that control mitochondrial morphology through changes in mitochondrial fusion or mitochondrial fission, confer susceptibility or resistance to the development of liver cancer in mouse models. Here, we review the data that suggest mitochondrial dynamics to be involved in the development of liver tumors. Hepatocellular carcinoma (HCC) is the most prevalent primary liver cancer. Due to its rising incidence and limited therapeutic options, HCC has become a leading cause of cancer-related death worldwide, accounting for 85% of all deaths due to primary liver cancers. Standard therapy for advanced-stage HCC is based on anti-angiogenic drugs such as sorafenib and, more recently, lenvatinib and regorafenib as a second line of treatment. The identification of novel therapeutic strategies is urgently required. Mitochondrial dynamics describes a group of processes that includes the movement of mitochondria along the cytoskeleton, the regulation of mitochondrial morphology and distribution, and connectivity mediated by tethering and fusion/fission events. In recent years, mitochondrial dynamic processes have emerged as key processes in the maintenance of liver mitochondrial homeostasis. In addition, some data are accumulating on the role played by mitochondrial dynamics during cancer development, and specifically on how such dynamics act directly on tumor cells or indirectly on cells responsible for tumor aggression and defense. (1) Background: The transforming growth factor (TGF)-β plays a dual role in liver carcinogenesis. At early stages, it inhibits cell growth and induces apoptosis. However, TGF-β expression is high in advanced stages of hepatocellular carcinoma (HCC) and cells become resistant to TGF-β induced suppressor effects, responding to this cytokine undergoing epithelial–mesenchymal transition (EMT), which contributes to cell migration and invasion. Metabolic reprogramming has been established as a key hallmark of cancer. However, to consider metabolism as a therapeutic target in HCC, it is necessary to obtain a better understanding of how reprogramming occurs, which are the factors that regulate it, and how to identify the situation in a patient. Accordingly, in this work we aimed to analyze whether a process of full EMT induced by TGF-β in HCC cells induces metabolic reprogramming. (2) Methods: In vitro analysis in HCC cell lines, metabolomics and transcriptomics. (3) Results: Our findings indicate a differential metabolic switch in response to TGF-β when the HCC cells undergo a full EMT, which would favor lipolysis, increased transport and utilization of free fatty acids (FFA), decreased aerobic glycolysis and an increase in mitochondrial oxidative metabolism. (4) Conclusions: EMT induced by TGF-β in HCC cells reprograms lipid metabolism to facilitate the utilization of FFA and the entry of acetyl-CoA into the TCA cycle, to sustain the elevated requirements of energy linked to this process. The Sample Analysis at Mars (SAM) instrument on the Curiosity rover has detected evidence of oxychlorine compounds (i.e., perchlorates and chlorates) in Gale crater, which has implications for past habitability, diagenesis, aqueous processes, interpretation of in situ organic analyses, understanding the martian chlorine cycle, and hazards and resources for future human exploration. Pure oxychlorines and mixtures of oxychlorines with Mars-analog phases have been analyzed for their oxygen (O2) and hydrogen chloride (HCl) releases on SAM laboratory analog instruments in order to constrain which phases are present in Gale crater. These studies demonstrated that oxychlorines evolve O2 releases with peaks between ~200 and 600 °C, although the thermal decomposition temperatures and the amount of evolved O2 decrease when iron phases are present in the sample. Mg and Fe oxychlorines decompose into oxides and release HCl between ~200 and 542 °C. Ca, Na, and K oxychlorines thermally decompose into chlorides and do not evolve HCl by themselves. However, the chlorides (original or from oxychlorine decomposition) can react with water-evolving phases (e.g., phyllosilicates) in the sample and evolve HCl within the temperature range of SAM (<~870 °C). These laboratory analog studies support that the SAM detection of oxychlorine phases is consistent with the presence of Mg, Ca, Na, and K perchlorate and/or chlorate along with possible contributions from adsorbed oxychlorines in Gale crater samples. There is, recently, a global concern about the gender gap in Science, Technology, Engineering, and Mathematics (STEM) areas, starting from education role models, student applications, through the academic, industrial and management career progression. Given the high visibility and popularity of all subjects related to space exploration, female participation in this field may be used to change the existing stereotypes and provide role models to the younger generations, thus having a positive influence on education while also demonstrating to other organizations how to promote diversity in the working environment. Previous studies on spacecraft science teams, considering only principal and co-Investigators, PIs and co-investigators (CoIs), respectively, show that the percentage of women in the role of investigators has remained flat at 15.8% since 2000. The NASA Mars Science Laboratory (MSL) mission is taken here as an example to perform a statistical analysis of the gender profile for the period 2004–2018. The results are compared with: (1) data from the US National Science Foundation (NSF) about gender distribution in STEM postdoctoral profiles and faculty members; (2) the trend of planetary exploration team profiles; (3) research and innovation statistics in Europe; (4) proposals of the EU FP6 funding program; and (5) the percentage of female researchers from the Elsevier status report. This analysis shows that the process of continually holding open calls for Participating Scientists based on individual merit and the application of a flat working structure have allowed gender balance within the MSL team to improve naturally while maximizing individual and team performance. Women represent approximately 30.6% of the team, in agreement with the current percentage of female planetary exploration researchers and senior faculty members in academia. Interestingly, the percentage of female-led articles has been above the MSL women percentage trend. While the percentage of women in planetary science appears to be increasing, their role on the proposing teams is still low. As in other STEM fields, attention should be paid to secure the adequate promotion of younger generations to achieve the United Nation's Sustainable Development Goal 5 of achieving gender equality and empowering all women and girls by 2030. Platelet activation plays a key role in cardiovascular diseases. The generation of mitochondrial reactive oxygen species (ROS) has been described as a critical step required for platelet activation. For this reason, it is necessary to find new molecules with antiplatelet activity and identify their mechanisms of action. Mitoquinone (MitoQ) is a mitochondria-targeted antioxidant that reduces mitochondrial overproduction of ROS. In this work, the antiplatelet effect of MitoQ through platelet adhesion and spreading, secretion, and aggregation was evaluated. Thus MitoQ, in a non-toxic effect, decreased platelet adhesion and spreading on collagen surface, and expression of P-selectin and CD63, and inhibited platelet aggregation induced by collagen, convulxin, thrombin receptor activator peptide-6 (TRAP-6), and phorbol 12-myristate 13-acetate (PMA). As an antiplatelet mechanism, we showed that MitoQ produced mitochondrial depolarization and decreased ATP secretion. Additionally, in platelets stimulated with antimycin A and collagen MitoQ significantly decreased ROS production. Our findings showed, for the first time, an antiplatelet effect of MitoQ that is probably associated with its mitochondrial antioxidant effect. [ABSTRACT FROM AUTHOR]

Metabolt is a portable soil incubator to characterize the metabolic activity of microbial ecosystems in soils. It measures the electrical conductivity, the redox potential, and the concentration of certain metabolism-related gases in the headspace just above a given sample of regolith. In its current design, the overall weight of Metabolt, including the soils (250 g), is 1.9 kg with a maximum power consumption of 1.5 W. Metabolt has been designed to monitor the activity of the soil microbiome for Earth and space applications. In particular, it can be used to monitor the health of soils, the atmospheric-regolith fixation, and release of gaseous species such as N2, H2O, CO2, O2, N2O, NH3, etc., that affect the Earth climate and atmospheric chemistry. It may be used to detect and monitor life signatures in soils, treated or untreated, as well as in controlled environments like greenhouse facilities in space, laboratory research environments like anaerobic chambers, or simulating facilities with different atmospheres and pressures. To illustrate its operation, we tested the instrument with sub-arctic soil samples at Earth environmental conditions under three different conditions: (i) no treatment (unperturbed); (ii) sterilized soil: after heating at 125 °C for 35.4 h (thermal stress); (iii) stressed soil: after adding 25% CaCl2 brine (osmotic stress); with and without addition of 0.5% glucose solution (for control). All the samples showed some distinguishable metabolic response, however there was a time delay on its appearance which depends on the treatment applied to the samples: 80 h for thermal stress without glucose, 59 h with glucose; 36 h for osmotic stress with glucose and no significant reactivation in the pure water case. This instrument shows that, over time, there is a clear observable footprint of the electrochemical signatures in the redox profile which is complementary to the gaseous footprint of the metabolic activity through respiration. We describe a versatile simulation chamber that operates under representative space conditions (pressures from < 10−5 mbar to ambient and temperatures from 163 to 423 K), the SpaceQ chamber. This chamber allows to test instrumentation, procedures, and materials and evaluate their performance when exposed to outgassing, thermal vacuum, low temperatures, baking, dry heat microbial reduction (DHMR) sterilization protocols, and water. The SpaceQ is a cubical stainless-steel chamber of 27,000 cm3 with a door of aluminum. The chamber has a table which can be cooled using liquid nitrogen. The chamber walls can be heated (for outgassing, thermal vacuum, or dry heat applications) using an outer jacket. The chamber walls include two viewports and 12 utility ports (KF, CF, and Swagelok connectors). It has sensors for temperature, relative humidity, and pressure, a UV–VIS–NIR spectrometer, a UV irradiation lamp that operates within the chamber as well as a stainless-steel syringe for water vapor injection, and USB, DB-25 ports to read the data from the instruments while being tested inside. This facility has been specifically designed for investigating the effect of water on the Martian surface. The core novelties of this chamber are: (1) its ability to simulate the Martian near-surface water cycle by injecting water multiple times into the chamber through a syringe which allows to control and monitor precisely the initial relative humidity inside with a sensor that can operate from vacuum to Martian pressures and (2) the availability of a high-intensity UV lamp, operating from vacuum to Martian pressures, within the chamber, which can be used to test material curation, the role of the production of atmospheric radicals, and the degradation of certain products like polymers and organics. For illustration, here we present some applications of the SpaceQ chamber at simulated Martian conditions with and without atmospheric water to (i) calibrate the ground temperature sensor of the Engineering Qualification Model of HABIT (HabitAbility: Brines, Irradiation and Temperature) instrument, which is a part of ExoMars 2022 mission. These tests demonstrate that the overall accuracy of the temperature retrieval at a temperature between −50 and 10 °C is within 1.3 °C and (ii) investigate the curation of composite materials of Martian soil simulant and binders, with added water, under Martian surface conditions under dry and humid conditions. Our studies have demonstrated that the regolith, when mixed with super absorbent polymer (SAP), water, and binders exposed to Martian conditions, can form a solid block and retain more than 80% of the added water, which may be of interest to screen radiation while maintaining a low weight. Volcanic-aeolian interactions and processes have played a vital role in landscape evolution on Mars. Martian lava fields and associated caves have extensive geomorphological, astrobiological, and in-situ resource utilization (ISRU) implications for future Mars missions which might be focused on subsurface exploration. Although several possible cave "skylights" of tens to >100 m diameter have been spotted in lava fields of Mars, there is a possibility of prevalence of meter-scale features which are an order of magnitude smaller and difficult to identify but could have vital significance from the scientific and future exploration perspectives. The Icelandic volcanic-aeolian environment and fissure volcanoes can serve as analogs to study lava flow-related small caves such as surface tubes, inflationary caves, liftup caves, and conduits. In the present work, we have tried to explore the usability of unmanned aerial vehicle (UAV)-derived images for characterizing a solidified lava flow and designing a sequential methodology to identify small caves in the lava flow. In the mapped area of ~0.33 km2, we were able to identify 81 small cave openings, five lava flow morphologies, and five small cave types using 2 cm/pixel high-resolution images. The results display the usefulness of UAV imaging for such analogous research, and also highlight the possibility of the widespread presence of similar small cave openings in Martian lava fields. Such small openings can facilitate optimal air circulation within the caves while sheltering the insides from physical weathering and harmful radiations. Using the available best resolution remote sensing images, we extend the analogy through the contextual and geomorphological analysis of several possible pit craters in the Tharsis region of Mars, in a region of extremely vesicular and fragile lava crust with pahoehoe-type morphology. We report two possible pit craters in this region, with diameters as small as ~20 m. The possibility that such small cave openings can lead to vast subterranean hollow spaces on Mars cannot be ruled out considering its low gravity. [ABSTRACT FROM AUTHOR]

The surface and near-surface air temperature observations are primary data for glacio-hydro-climatological studies. The in situ air temperature (Ta) observations require intense logistic and financial investments, making it sparse and fragmented particularly in remote and extreme environments. The temperatures in Himalaya are controlled by a complex system driven by topography, seasons, and cryosphere which further makes it difficult to record or predict its spatial heterogeneity. In this regard, finding a way to fill the observational spatiotemporal gaps in data becomes more crucial. Here, we show the comparison of Ta recorded at 11 high altitude stations in Western Himalaya with their respective land surface temperatures (Ts) recorded by Moderate Resolution Imagining Spectroradiometer (MODIS) Aqua and Terra satellites in cloud-free conditions. We found remarkable seasonal and spatial trends in the Ta vs. Ts relationship: (i) Ts are strongly correlated with Ta (R2 = 0.77, root mean square difference (RMSD) = 5.9 °C, n = 11,101 at daily scale and R2 = 0.80, RMSD = 5.7 °C, n = 3552 at 8-day scale); (ii) in general, the RMSD is lower for the winter months in comparison to summer months for all the stations, (iii) the RMSD is directly proportional to the elevations; (iv) the RMSD is inversely proportional to the annual precipitation. Our results demonstrate the statistically strong and previously unreported Ta vs. Ts relationship and spatial and seasonal variations in its intensity at daily resolution for the Western Himalaya. We anticipate that our results will provide the scientists in Himalaya or similar data-deficient extreme environments with an option to use freely available remotely observed Ts products in their models to fill-up the spatiotemporal data gaps related to in situ monitoring at daily resolution. Substituting Ta by Ts as input in various geophysical models can even improve the model accuracy as using spatially continuous satellite derived Ts in place of discrete in situ Ta extrapolated to different elevations using a constant lapse rate can provide more realistic estimates. Lysinuric protein intolerance (LPI) is a rare autosomal disease caused by defective cationic amino acid (CAA) transport due to mutations in SLC7A7, which encodes for the y+LAT1 transporter. LPI patients suffer from a wide variety of symptoms, which range from failure to thrive, hyperammonemia, and nephropathy to pulmonar alveolar proteinosis (PAP), a potentially life-threatening complication. Hyperammonemia is currently prevented by citrulline supplementation. However, the full impact of this treatment is not completely understood. In contrast, there is no defined therapy for the multiple reported complications of LPI, including PAP, for which bronchoalveolar lavages do not prevent progression of the disease. The lack of a viable LPI model prompted us to generate a tamoxifen-inducible Slc7a7 knockout mouse (Slc7a7−/−). The Slc7a7−/− model resembles the human LPI phenotype, including malabsorption and impaired reabsorption of CAA, hypoargininemia and hyperammonemia. Interestingly, the Slc7a7−/− mice also develops PAP and neurological impairment. We observed that citrulline treatment improves the metabolic derangement and survival. On the basis of our findings, the Slc7a7−/− model emerges as a promising tool to further study the complexity of LPI, including its immune-like complications, and to design evidence-based therapies to halt its progression. Understanding extraterrestrial environments and landforms through remote sensing and terrestrial analogy has gained momentum in recent years due to advances in remote sensing platforms, sensors, and computing efficiency. The seasonal brines of the largest salt plateau on Earth in Salar de Uyuni (Bolivian Altiplano) have been inadequately studied for their localized hydrodynamics and the regolith volume transport across the freshwater-brine mixing zones. These brines have recently been projected as a new analogue site for the proposed Martian brines, such as recurring slope lineae (RSL) and slope streaks. The Martian brines have been postulated to be the result of ongoing deliquescence-based salt-hydrology processes on contemporary Mars, similar to the studied Salar de Uyuni brines. As part of a field-site campaign during the cold and dry season in the latter half of August 2017, we deployed an unmanned aerial vehicle (UAV) at two sites of the Salar de Uyuni to perform detailed terrain mapping and geomorphometry. We generated high-resolution (2 cm/pixel) photogrammetric digital elevation models (DEMs) for observing and quantifying short-term terrain changes within the brines and their surroundings. The achieved co-registration for the temporal DEMs was considerably high, from which precise inferences regarding the terrain dynamics were derived. The observed average rate of bottom surface elevation change for brines was ~1.02 mm/day, with localized signs of erosion and deposition. Additionally, we observed short-term changes in the adjacent geomorphology and salt cracks. We conclude that the transferred regolith volume via such brines can be extremely low, well within the resolution limits of the remote sensors that are currently orbiting Mars, thereby making it difficult to resolve the topographic relief and terrain perturbations that are produced by such flows on Mars. Thus, the absence of observable erosion and deposition features within or around most of the proposed Martian RSL and slope streaks cannot be used to dismiss the possibility of fluidized flow within these features. Metabolic adaptation may happen in response to the pressure exerted by the microenvironment and is a key step in survival of metastatic cells. Brain metastasis occurs as a consequence of the systemic dissemination of tumor cells, a fact that correlates with poor prognosis and high morbidity due to the difficulty in identifying biomarkers that allow a more targeted therapy. Previously, we performed transcriptomic analysis of human breast cancer patient samples and evaluated the differential expression of genes in brain metastasis (BrM) compared to lung, bone and liver metastasis. Our network approach identified upregulation of glucose-regulated protein 94 (GRP94) as well as proteins related to synthesis of fatty acids (FA) in BrM. Here we report that BrM cells show an increase in FA content and decreased saturation with regard to parental cells measured by Raman spectroscopy that differentiate BrM from other metastases. Moreover, BrM cells exerted a high ability to oxidize FA and compensate hypoglycemic stress due to an overexpression of proteins involved in FA synthesis and degradation (SREBP-1, LXRα, ACOT7). GRP94 ablation restored glucose dependence, down-regulated ACOT7 and SREBP-1 and decreased tumorigenicity in vivo. Aeolian processes are believed to play a major role in the landscape evolution of Mars. Investigations on Martian aeolian landforms such as ripples, transverse aeolian ridges (TARs), and dunes, and aeolian sediment flux measurements are important to enhance our understanding of past and present wind regimes, the ongoing dust cycle, landscape evolution, and geochemistry. These aeolian bedforms are often comprised of loose sand and sharply undulating topography and thus pose a threat to mobility and maneuvers of Mars rovers. Here we present a first-hand account of the distribution, morphologies, and morphometrics of TARs in Oxia Planum, the recently selected ExoMars 2020 Rover landing site. The gridded mapping was performed for contiguous stretches of TARs within all the landing ellipses using 57 sub-meter high resolution imaging science experiment (HiRISE) scenes. We also provide the morphological descriptions for all types of TARs present within the landing ellipses. We use HiRISE digital terrain models (DTMs) along with the images to derive morphometric information for TARs in Oxia Planum. In general, the average areal TAR coverage was found to be 5.4% (±4.9% standard deviation), increasing from west to east within the landing ellipses. We report the average TAR morphometrics in the form of crest–ridge width (131.1 ± 106.2 m), down-wind TAR length (17.6 ± 10.1 m), wavelength (37.3 ± 11.6 m), plan view aspect ratio (7.1 ± 2.3), inter-bedform spacing (2.1 ± 1.1), slope (10.6° ± 6.1°), predominant orientations (NE-SW and E-W), and height (1.2 ± 0.8 m). While simple TARs are predominant, we report other TAR morphologies such as forked TAR, wavy TAR with associated smaller secondary ripples, barchan-like TAR, networked TAR, and mini-TARs from the region. Our results can help in planning the rover traverses in terms of both safe passage and scientific returns favoring aeolian research, particularly improving our understanding of TARs.
