Coronavirus disease 2019 (COVID-19) is an infection characterized by the dysregulation of systemic cytokine levels. The measurement of serum levels of inflammatory cyto-/chemokines has been suggested as a tool in the management of COVID-19. The aim of this study is to highlight the significance of measured levels of interleukin (IL)-1α, IL-1β, IL-6, IL-8, IL-10, IL-12(p70), IL-27, interferon (IFN)γ, interferon gamma-induced protein (IP)-10, monocyte chemoattractant protein (MCP)-1, and tumor necrosis factor (TNF)-α in serum samples from infected and recovered subjects, possibly predictive of severity and/or duration of the disease. Serum samples from healthy (HD), positive at hospital admittance (T0), and recovered subjects (T1, 31–60, or 70–200 days post-negativization) were collected and tested through a bead-based cytometric assay and confirmed through ELISA. IL-10 levels were increased in the T0 group compared to both HD and T1. IL-27 significantly decreased in the 31–60 group. IL-1β significantly increased in the 70–200 day group. TNF-α significantly decreased in T0 compared to HD and in the 31–60 group versus HD. IP-10 significantly increased in T0 compared to HD. These results suggest that IP-10 could represent an early marker of clinical worsening, whereas IL-10 might be indicative of the possible onset of post-COVID-19 long syndrome.

The synthesis of a new tetracationic porphyrin derivative is described. Contrary to the best known derivatives in the literature, which are derived from 5,10,15,20-tetrakis-4-pyridylporphyrin (TPyP), in this procedure we start from 5,10,15,20-tetrakis-(4-carboxymethoxyphenyl)porphyrin (TPPCOOMe), obtained by the condensation reaction between pyrrole and 4-formylbenzoate. The reaction is carried out in refluxed xylene, avoiding the use of halogenated solvents. The final product, 5,10,15,20-tetrakis-(4-(3-carbamoyl-pyridyl)-methylphenyl)porphyrin bromide (P15p), exhibits four cationic portions that make it soluble in water and suitable for G4 stabilization. The choice to synthesize a derivative of TPPCOOMe is based on the idea of having a possible stabilizer that, unlike those obtained from TPyP, shows the cationic moieties farther from the porphyrin core and thus closer to the phosphate groups present on the G4 loops.

The impact of disability on people’s lives is often underestimated because the extra costs of living with a disability are not accounted for. This paper analyzes several different methodologies for estimating those costs and explores their usefulness in designing inclusive social policies. For example, one approach is to measure what is currently being spent, while another is to estimate what would need to be spent for equal participation. These can be measured using statistical techniques or through a more qualitative methodology. Each of these methods has its advantages and disadvantages. The paper concludes with recommendations for which methodology fits which purpose, and how they can be used together to obtain a full accounting of the extra costs incurred by people with disabilities.

The production of nanofibers has become a significant area of research due to their unique properties and diverse applications in various fields, such as biomedicine, textiles, energy, and environmental science. Electrospinning, a versatile and scalable technique, has gained considerable attention for its ability to fabricate nanofibers with tailored properties. Among the wide array of conductive polymers, poly(3,4-ethylenedioxythiophene) (PEDOT) has emerged as a promising material due to its exceptional conductivity, environmental stability, and ease of synthesis. The electrospinning of PEDOT-based nanofibers offers tunable electrical and optical properties, making them suitable for applications in organic electronics, energy storage, biomedicine, and wearable technology. This review, with its comprehensive exploration of the fabrication, properties, and applications of PEDOT nanofibers produced via electrospinning, provides a wealth of knowledge and insights into leveraging the full potential of PEDOT nanofibers in next-generation electronic and functional devices by examining recent advancements in the synthesis, functionalization, and post-treatment methods of PEDOT nanofibers. Furthermore, the review identifies current challenges, future directions, and potential strategies to address scalability, reproducibility, stability, and integration into practical devices, offering a comprehensive resource on conductive nanofibers.

Monitoring surface displacements of landslides is essential for evaluating their evolution and the effectiveness of mitigation works. Traditional methods like robotic total stations (RTSs) and GNSS provide high-accuracy measurements but are limited to discrete points, potentially missing the broader landslide’s behavior. On the contrary, laser scanner surveys offer accurate 3D representations of slopes and the possibility of inferring their movements, but they are often limited to infrequent, high-cost surveys. Monitoring techniques based on ground-based digital photogrammetry may represent a new, robust, and cost-effective alternative. This study demonstrates the use of multi-temporal images from fixed and calibrated cameras to achieve the 3D reconstruction of landslide displacements. The method presented offers the important benefit of obtaining spatially dense displacement data across the entire camera view and quasi-continuous temporal measurement. This paper outlines the framework for this prototyping technique, along with a description of the necessary hardware and procedural steps. Furthermore, strengths and weaknesses are discussed based on the activities carried out in a landslide case study in northeastern Italy. The results from the photo-monitoring are reported, discussed, and compared with traditional topographical data, validating the reliability of this new approach in monitoring the time evolution of surface displacements across the entire landslide area.

Electromagnetic forces can lead to the structural failure of power transformers due to extreme loading conditions, vibration, and/or fatigue. Therefore, studying the nature and the magnitude of these forces is a key task in the design and failure analysis of such important equipment. Keeping this issue in mind, this work aims at conducting a numerical analysis in order to evaluate the mechanical stresses and displacements of windings in power transformers due to the action of electromagnetic forces. With this purpose, a finite element model is developed considering electromagnetic and mechanical effects assuming short-circuit conditions. The study compares the cases employing copper and aluminum windings with header tap, in different temperatures. The model developed in this work is verified against analytical solutions. The numerical calculations allow for performing a detailed analysis in terms of the distribution of both displacements and stresses along the windings, which is of great relevance for identifying critical structural points and avoiding structural failure. Overall, the obtained results demonstrate that the finite element model is a useful tool for the structural design of power transformers that allow for investigating and optimizing key aspects before manufacturing.

Background: Myotonic dystrophy type 1 (DM1) is a rare multisystemic genetic disorder with motor hallmarks of myotonia, muscle weakness and wasting. DM1 patients have an increased risk of falling of multifactorial origin, and proprioceptive and vestibular deficits can contribute to this risk. Abnormalities of muscle spindles in DM1 have been known for years. This observational cross-sectional study was based on the hypothesis of impaired cervical proprioception caused by alterations in the neck spindles. Methods: Head position sense was measured in 16 DM1 patients and 16 age- and gender-matched controls. A head-to-target repositioning test was requested from blindfolded participants. Their head was passively rotated approximately 30° leftward or rightward and flexed or extended approximately 25°. Participants had to replicate the imposed positions. An optoelectronic system was adopted to measure the angular differences between the reproduced and the imposed positions (joint position error, JPE, °) concerning the intended (sagittal, horizontal) and unintended (including the frontal) planar projections. In DM1 patients, JPEs were correlated with clinical and balance measures. Static balance in DM1 patients was assessed through dynamic posturography. Results: The accuracy and precision of head repositioning in the intended sagittal and horizontal error components did not differ between DM1 and controls. On the contrary, DM1 patients showed unintended side-bending to the left and the right: the mean [95%CI] of frontal JPE was −1.29° [−1.99°, −0.60°] for left rotation and 0.98° [0.28°, 1.67°] for right rotation. The frontal JPE of controls did not differ significantly from 0° (left rotation: 0.17° [−0.53°, 0.87°]; right rotation: −0.22° [−0.91°, 0.48°]). Frontal JPE differed between left and right rotation trials (p < 0.001) only in DM1 patients. No correlation was found between JPEs and measures from dynamic posturography and clinical scales. Conclusions: Lateral head bending associated with head rotation may reflect a latent impairment of neck proprioception in DM1 patients.

Background: Methylmalonic Aciduria (MA) without homocystinuria (or isolated MA) is a group of rare inherited metabolic disorders which leads to the accumulation of methylmalonic acid (MMA), a toxic molecule that accumulates in blood, urine, and cerebrospinal fluid, causing acute and chronic complications including metabolic crises, acute kidney injury (AKI), and chronic kidney disease (CKD). Detailed Case Description: Herein, we report a case of a 39-year-old male with MA and stage IV CKD who experienced acute metabolic decompensation secondary to gastrointestinal infection. The patient underwent a single hemodialysis (HD) session to correct severe metabolic acidosis unresponsive to medical therapy and to rapidly remove MMA. The HD session resulted in prompt clinical improvement and shortening of hospitalization. Discussion: MMA accumulation in MA patients causes acute and life-threatening complications, such as metabolic decompensations, and long-term complications such as CKD, eventually leading to renal replacement therapy (RRT). Data reported in the literature show that, overall, all dialytic treatments (intermittent HD, continuous HD, peritoneal dialysis) are effective in MMA removal. HD, in particular, can be useful in the emergency setting to control metabolic crises, even with GFR > 15 mL/min. Kidney and/or liver transplantations are often needed in MA patients. While a solitary transplanted kidney can be rapidly affected by MMA exposure, with a decline in renal function even in the first year of follow-up, the combined liver–kidney transplantation showed better long-term results due to a combination of reduced MMA production along with increased urinary excretion. Conclusions: Early diagnosis, multidisciplinary management and preventive measures are pivotal in MA patients to avoid recurrent AKI episodes and, consequently, to slow down CKD progression.

Reduced-order electrothermal models play a key role in the design and control of lithium-ion cell stacks, calling for accurate model parameter calibration. This paper presents a complete electrical and thermal experimental characterization procedure for the coupled modeling of cylindrical lithium-ion cells in order to implement them in a prototype Formula SAE hybrid racing car. The main goal of the tests is to determine how the cell capacity varies with the temperature and the discharge current to predict the open-circuit voltage of the cell and its entropic component. A simple approach for the characterization of the battery equivalent electrical circuit and a two-step thermal characterization method are also shown. The investigations are carried out on four commercial 18650 NMC lithium cells. The model was shown to predict the battery voltage with an RMS error lower than 20 mV and the temperature with an RMS error equal to 0.5 °C. The authors hope that this manuscript can contribute to the development of standardized characterization techniques for such cells while offering experimental data and validated models that can be used by researchers and BMS designers in different applications.

Conductive polymers, such as polyaniline (PANI), have interesting applications, ranging from flexible electronics, energy storage devices, sensors, antistatic or anticorrosion coatings, etc. However, the full exploitation of conductive polymers still poses a challenge due to their low processability. The use of compatible stabilizers to obtain dispersible and stable colloids is among the possible solutions to overcome such drawbacks. In this work, potato starch was used as a steric stabilizer for the preparation of colloidal polyaniline (emeraldine salt, ES)/starch composites by exploiting the oxidative polymerization of aniline in aqueous solutions with various starch-to-aniline ratios. The polyaniline/starch bio-composites were subjected to structural, spectroscopic, thermal, morphological, and electrochemical analyses. The samples were then tested for their dispersibility/solubility in a range of organic solvents. The results demonstrated the formation of PANI/starch biocomposites with a smaller average size than starch particles, showing improved aqueous dispersion and enhanced solubility in organic solvents. With respect to previously reported PANI-EB (emeraldine base)/starch composites, the novel colloids displayed a lower overall crystallinity, but the conductive nature of PANI-ES enhanced its electrochemical properties, resulting in richer redox chemistry, particularly evident in its oxidation behavior, as observed through cyclic voltammetry. Finally, as proof of the improved processability, the colloids were successfully integrated into a thin polyether sulfone (PES) membrane.

Background: Institutionalized individuals with dysphagia are particularly at risk for malnutrition. This study investigated two texture-modified models for patients with dysphagia, as follows: (i) traditional homemade pureed food (PF) and (ii) homogenized meals obtained from dehydrated and rehydrated instantaneous preparations (IPs). Methods: A retrospective pilot study was performed. It included patients affected by medium-severity dysphagia admitted to the nursing home “Sacra Famiglia” Institute of Cocquio Trevisago, Varese. The patients were aged 41–81 years old and all had complex disabilities. They underwent anthropometric and biochemical parameter assessments at baseline, as well as at two months and four months follow-up. Results: The study involved 30 patients, 15 received the IP meal. The comparison between the baseline and the follow-up did not show significant anthropometric and biochemical parameter differences. Conversely, the IP group reported significantly higher levels of consumption and satisfaction, evaluated using a modified Chernoff scale based on three levels of smiles, than the PF group. Conclusions: The present findings provide promising indications to improve the diet of patients affected by dysphagia, since meal satisfaction is a relevant factor that has been shown to be associated with better patient mood, motivation to eat, and adherence to prescribed diet.

Adherent cells perceive mechanical feedback from the underlying matrix and convert it into biochemical signals through a process known as mechanotransduction. The response to changes in the microenvironment relies on the cell’s mechanical properties, including elasticity, which was recently identified as a biomarker for various diseases. Here, we propose the design, development, and characterization of a new system for the measurement of adherent cells’ strain drop, a parameter correlated with cells’ elasticity. To consider the interplay between adherent cells and the host extracellular matrix, cell stretching was combined with adhesion on substrates with different stiffnesses. The technique is based on the linear stretching of silicone chambers, high-speed image acquisition, and feedback for image centering. The system was characterized in terms of the strain homogeneity, impact of collagen coating, centering capability, and sensitivity. Subsequently, it was employed to measure the strain drop of two osteosarcoma cell lines, low-aggressive osteoblast-like SaOS-2 and high-aggressive 143B, cultured on two different substrates to recall the stiffness of the bone and lung extracellular matrices. Results demonstrated good substrate homogeneity, a negligible effect of the collagen coating, and an accurate image centering. Finally, the experimental results showed an average strain drop that was lower in the 143B cells in comparison with the SaOS-2 cells in all the tested conditions.

Infant attention is a cognitive function that underlines sensory–motor integration processes at the interface between the baby and the surrounding physical and socio-relational environment, mainly with the caregivers. The investigation of the role of non-visual inputs (i.e., vocal and tactile) provided by the caregivers in shaping infants’ attention in the context of visual impairment is relevant from both a theoretical and clinical point of view. This study investigated the social attention (i.e., gaze orientation) skills in a group of visually impaired (VI) and age-matched sighted controls (SCs) between 9 and 12 months of age. Moreover, the role of VI severity and maternal vocalizations and touch in shaping the social attention were investigated. Overall, 45 infants and their mothers participated in a video-recorded 4 min interaction procedure, including a play and a still-face episode. The infants’ gaze orientation (i.e., mother-directed, object-directed, or unfocused) and the types of maternal vocalizations and touch (i.e., socio-cognitive, affective) were micro-analytically coded. Maternal vocalizations and touch were found to influence gaze orientation differently in VI infants compared SCs. Moreover, the group comparisons during the play episode showed that controls were predominantly oriented to the mothers, while VI infants were less socially oriented. Visual impairment severity did not emerge as linked with social attention. These findings contribute to our understanding of socio-cognitive developmental trajectories in VI infants and highlight the need for tailored interventions to promote optimal outcomes for VI populations.

The application of sanitary sewage using subsurface drip irrigation can mitigate current challenges related to water availability and food production. However, before implementing these techniques, it is crucial to conduct studies to assess their impacts on soil and plants. The objective of this study was to evaluate chemical changes in the soil and the development of cowpeas subjected to sanitary sewage applied by drippers with different flow rates and installation depths. Drippers were positioned at various depths (0 to 30 cm) and operated with flow rates of 1.6 and 3.8 L h−1. Cowpeas were cultivated in pots with clayey soil, using synthetic sanitary sewage based on the maximum limit of nitrogen fertilization. Irrigation management was controlled in terms of soil moisture, which was monitored using TDR probes. The results indicated that reducing the depth of the drippers positively affected grain production and the development of cowpeas. Fertigation with sanitary sewage at greater depths increased soil phosphorus concentrations and base saturation. Dripper depth also influenced soil concentrations of phosphorus, potassium, calcium, and magnesium, while sodium concentrations decreased with greater depth. It is concluded that dripper flow rates did not impact soil chemical parameters or the agronomic characteristics of cowpeas. However, despite nutrient supply at greater depths, the subsurface drip irrigation system proved unsuitable for cowpea production in clayey soil.

Aryl and heteroaryl iodides have been efficiently converted into the corresponding thioacetates in cyclopentyl methyl ether (CPME), a green solvent, under Cu catalysis. The chemoselectivity of the reaction is mainly controlled by electronic factors, enabling the conversion of both electron-rich and electron-deficient substrates into the corresponding thioacetates in good to excellent yields. The products can be easily deprotected to the corresponding thiolates to carry out additional synthetic transformations in situ. Surprisingly, despite CPME’s relatively low dielectric constant, the reaction rate significantly increased when conducted under microwave irradiation conditions. This synthetic methodology exhibits a remarkable tolerance to functional groups, mild reaction conditions, and a wide substrate scope, utilizing a safe and inexpensive CuI pre-catalyst in the green solvent CPME. A non-aqueous workup allowing for the complete recovery of both catalyst and solvent makes this approach an environmentally sustainable protocol for C(sp2) sulfur functionalization. Additionally, the reaction shows selective cross-coupling with iodides in competition with chlorides and bromides, allowing its use in multistep syntheses. To demonstrate the potential of this methodology, it was applied to the high-yield synthesis of a photochromic dithienylethene, where a selective synthesis had not been reported before.

Traders and investors are interested in accurately predicting cryptocurrency prices to increase returns and minimize risk. However, due to their uncertainty, volatility, and dynamism, forecasting crypto prices is a challenging time series analysis task. Researchers have proposed predictors based on statistical, machine learning (ML), and deep learning (DL) approaches, but the literature is limited. Indeed, it is narrow because it focuses on predicting only the prices of the few most famous cryptos. In addition, it is scattered because it compares different models on different cryptos inconsistently, and it lacks generality because solutions are overly complex and hard to reproduce in practice. The main goal of this paper is to provide a comparison framework that overcomes these limitations. We use this framework to run extensive experiments where we compare the performances of widely used statistical, ML, and DL approaches in the literature for predicting the price of five popular cryptocurrencies, i.e., XRP, Bitcoin (BTC), Litecoin (LTC), Ethereum (ETH), and Monero (XMR). To the best of our knowledge, we are also the first to propose using the temporal fusion transformer (TFT) on this task. Moreover, we extend our investigation to hybrid models and ensembles to assess whether combining single models boosts prediction accuracy. Our evaluation shows that DL approaches are the best predictors, particularly the LSTM, and this is consistently true across all the cryptos examined. LSTM reaches an average RMSE of 0.0222 and MAE of 0.0173, respectively, 2.7% and 1.7% better than the second-best model. To ensure reproducibility and stimulate future research contribution, we share the dataset and the code of the experiments.

Rare sarcomas present significant treatment challenges compared to more prevalent soft tissue sarcomas due to limited treatment options and a poor understanding of their biology. This study investigates a unique case of penile sarcoma, providing a comprehensive morphological and molecular analysis. Through the creation of experimental patient-derived models—including patient-derived xenograft (PDX), 3D, and monolayer primary cultures—we successfully replicated crucial molecular traits observed in the patient’s tumor, such as smooth muscle actin and CD99 expression, along with specific mutations in genes like TSC2 and FGFR4. These models are helpful in assessing the potential for an in-depth exploration of this tumor’s biology. This comprehensive approach holds promise in identifying potential therapeutic avenues for managing this exceedingly rare soft tissue sarcoma.

The search for cleaner and more sustainable energy sources is increasingly growing. Aligning this demand with another environmental problem, such as sewage treatment/disposal, is a strategic priority. In light of this, the aim of this study was to estimate the energy potential of sewage generated in the Brazilian state of São Paulo (SP) by using it to produce biomethane. The study also evaluated the viability of using this byproduct of sewage treatment (biomethane) as a substitute for natural gas (NG), as both of them have similar lower heat values. To do this, information was gathered regarding the population, gross domestic product per capita, sewage collected, and natural gas consumption for each of the state’s 645 cities, and, based on this, the sewage energy potential, the amount of NG to be substituted by biomethane, and the reduction in CO2 emissions were calculated. Moreover, in order to address a possible allocation of biomethane that could potentially be produced in each SP city and sent to currently NG-consuming cities, an optimization algorithm was proposed. The results indicated a sewage energy potential of 4.68 × 109 kWh/yr for the entire SP state, which would be enough to supply around 10% of the energy value of all the NG currently consumed. It was also observed that from 130 cities with NG consumption, 10 could produce enough biomethane to fully satisfy the natural gas demand. In the elected scenario of optimization, 291 cities were found to be capable of supplying the demand of 26 cities that currently use NG. The potential to reduce CO2 emissions is between 1.81 × 106 and 2.42 × 106 ton/yr, and this range could increase if sewage treatment coverage grows. Despite the challenges inherent in extrapolating a potential study to scenarios that require significative investment, the results obtained are useful for formulating public policies for decarbonization in the near future.

The scarcity of natural resources makes it essential to develop products that meet environmental requirements. This is also true for the water and wastewater treatment business, where even consolidated processes, such as coagulation and flocculation, must be improved, opening opportunities for searching for alternative options to conventional processes. Among the existing options, tannin-based agents (TBAs) have been highlighted in recent years due to their biodegradability and proven efficiency. However, little is known about the impacts of the production process of these agents on an operational/industrial scale. In this study, an examination of the environmental impacts of the full-scale production (more than 500 tons yearly) of a TBA from Acacia spp. (known as black acacia or mimosa) was carried out. To accomplish this, a life cycle assessment (LCA) was developed using openLCA version 2.0.0 to assess a cradle-to-gate system of 1 kg of packed TBA produced. Additionally, a comparison was made between the impacts of the production of TBA and a conventional water treatment agent, aluminum sulfate, to verify the benefits of producing the former. The most relevant impacts resulting from the production of 1 kg of TBA are observed in the following categories: global warming (1.52 kgCO2-eq); terrestrial (7.67 kg1.4-DCB-eq), freshwater (0.06 kg1.4-DCB-eq), and marine (0.08 kg1.4-DCB-eq) ecotoxicities; carcinogenic (0.10 kg1.4-DCB-eq) and non-carcinogenic (1.36 kg1.4-DCB-eq) human toxicities; and water use (0.02 m3). The main contributors to the impacts were the chemicals ammonium chloride and formaldehyde used, the transport of inputs, and the energy used. The aluminum sulfate showed better performance than the TBA for a greater number of categories; however, the normalization of the impacts showed the TBA as a very interesting option. The results obtained here can be used by TBA producers to act on the most impactful categories so that the production process becomes increasingly sustainable.

Melanoma is the principal cause of death in skin cancer due to its ability to invade and cause metastasis. Hypoxia, which characterises the tumour microenvironment (TME), plays an important role in melanoma development, as cancer cells can adapt and acquire a more aggressive phenotype. Carbonic anhydrases (CA) activity, involved in pH regulation, is related to melanoma cell migration and invasion. Furthermore, the Hedgehog (Hh) pathway, already known for its role in physiological processes, is a pivotal character in cancer cell growth and can represent a promising pharmacological target. In this study, we targeted Hh pathway components with cyclopamine, glabrescione B and C22 in order to observe their effect on carbonic anhydrase XII (CAXII) expression especially under hypoxia. We then performed a migration and invasion assay on two melanoma cell lines (SK-MEL-28 and A375) where Smoothened, the upstream protein involved in Hh regulation, and GLI1, the main transcription factor that determines Hh pathway activation, were chemically inhibited. Data suggest the existence of a relationship between CAXII, hypoxia and the Hedgehog pathway demonstrating that the chemical inhibition of the Hh pathway and CAXII reduction resulted in melanoma migration and invasion impairment especially under hypoxia. As in recent years drug resistance to small molecules has arisen, the development of new chemical compounds is crucial. The multitarget Hh inhibitor C22 proved to be effective without signs of cytotoxicity and, for this reason, it can represent a promising compound for future studies, with the aim to reach a better melanoma disease management.

The early identification of a subclinical rejection (SCR) can improve the long-term outcome of the transplanted kidney through intensified immunosuppression. However, the only approved diagnostic method is the protocol biopsy, which remains an invasive method and not without minor and/or major complications. The protocol biopsy is defined as the sampling of allograft tissue at pre-established times even in the absence of an impaired renal function; however, it does not avoid histological damage. Therefore, the discovery of new possible biomarkers useful in the prevention of SCR has gained great interest. Among all the possible candidates, there are microRNAs (miRNAs), which are short, noncoding RNA sequences, that are involved in mediating numerous post-transcriptional pathways. They can be found not only in tissues, but also in different biological fluids, both as free particles and contained in extracellular vesicles (EVs) released by different cell types. In this study, we firstly performed a retrospective miRNA screening analysis on biopsies and serum EV samples of 20 pediatric transplanted patients, followed by a second screening on another 10 pediatric transplanted patients’ urine samples at one year post-transplant. In both cohorts, we divided the patients into two groups: patients with histological SCR and patients without histological SCR at one year post-transplantation. The isolated miRNAs were analyzed in an NGS platform to identify different expressions in the two allograft states. Although no statistical data were found in sera, in the tissue and urinary EVs, we highlighted signatures of miRNAs associated with the histological SCR state.

Ester is one of the most significant functional groups in organic chemistry and is enclosed in several valued molecules. Usually, esters are prepared through the acid-catalyzed esterification reaction of carboxylic acids with alcohols, transesterification of esters with alcohols, or via activation of carboxylic acids followed by the addition of alcohols. However, these procedures typically imply the excess use of reactants and harsh reaction conditions. Visible light-mediated photoreactions have been disclosed to display a safe, sustainable, and accessible alternative to traditional methods, and to lead new reactivity modes in organic procedures. In this context, we propose a transition metal-based and organic-based photocatalyst-free synthesis of esters from alcohols induced by visible light. The methodology can be carried out using sunlight or artificial visible light as a solar simulator or a blue LED source.

This study investigated the synergistic effects of combining Saccharomyces cerevisiae and Oenococcus oeni during the alcoholic fermentation of a low-acidity cider. The initial population of indigenous wild lactic acid bacteria (LAB) in the apple must was 104 CFU/mL. Alcoholic fermentations were carried out without (Cider I) and with (Cider II) the O. oeni inoculation at 105 CFU/mL. As S. cerevisiae grows, a declining trend was observed in indigenous and inoculated LAB populations. While the wild LAB exhibited higher sensitivity than O. oeni, they were not eliminated during alcoholic fermentation. The addition of O. oeni impacted the growth and metabolic activity of S. cerevisiae. The bioconversion of malic acid into lactic acid predominantly occurred during the growth phase (43–66%) and stationary phase (4–27%). The resurgence of O. oeni following alcoholic fermentation significantly impacted the production of volatile compounds. After 20 days of fermentation, Cider II displayed a twofold increase in these compounds, resulting in a more favorable sensory profile according to evaluators. Consequently, malolactic fermentation (MLF) coincided with alcoholic fermentation, leading to a reduction in malic acid content. Furthermore, post alcoholic fermentation, MLF positively enhanced the aromatic quality of low-acid cider made from apples with low acidity.

Margarine exhibits significant variations in composition, allowing it to cater to diverse consumer segments. This study aimed to characterize the physical and sensory attributes of margarine samples available in the Brazilian market. Twelve commercial samples from six different brands, encompassing 30% to 80% of lipid contents, were subjected to instrumental texture analysis and affective assessment. A total of 112 consumers participated in acceptance tests and Check-All-That-Apply (CATA) evaluations, while another group of 62 subjects performed Projective Mapping. Samples with lipid percentages exceeding 70% achieved the highest average acceptance scores for taste and overall impression. The brand with the lowest lipid content (30%) exhibited a stronger association with negative attributes, including rancid flavor and aroma, bitterness, and metallic flavor, resulting in lower average scores for aroma, flavor, texture, and overall impression. However, these scores were not statistically different from samples with 50% and 60% lipid content. Reducing lipid levels in fat-based products such as margarine poses a challenge to food manufacturers, as consumers generally perceive higher lipid percentages as indicative of superior flavor quality.

Fabry disease (FD) is an X-linked lysosome storage disease that results in the accumulation of globotriaosylceramide (Gb3) throughout the body leading to irreversible target organ damage. As the role of secondary mediators (inflammatory molecules) and their mechanisms has not been fully elucidated, we focused on the interleukin (IL)-6 system in adult FD patients and in matched healthy subjects. To obtain insights into the complex regulation of IL-6 actions, we used a novel approach that integrates information from plasma and exosomes of FD patients (n = 20) and of healthy controls (n = 15). Soluble IL-6 receptor (sIL-6R) levels were measured in plasma with the ELISA method, and membrane-bound IL-6R was quantified in plasma and urinary exosomes using flow cytometry. In FD patients, the levels of soluble IL-6R in plasma were higher than in control subjects (28.0 ± 5.4 ng/mL vs. 18.9 ± 5.4 ng/mL, p < 0.0001); they were also higher in FD subjects with the classical form as compared to those with the late-onset form of the disease (36.0 ± 11.4 ng/mL vs. 26.1 ± 4.5 ng/mL, p < 0.0001). The percentage of urinary exosomes positive for IL-6R was slightly lower in FD (97 ± 1 vs. 100 ± 0% of events positive for IL-6R, p < 0.05); plasma IL-6 levels were not increased. These results suggest a potential role of IL-6 in triggering the inflammatory response in FD. As in FD patients only the levels of sIL-6Rs are consistently higher than in healthy controls, the IL-6 pathogenic signal seems to prevail over the homeostatic one, suggesting a potential mechanism causing multi-systemic damage in FD.

Chrono-nutrition studies dietary habits and their role in the onset of metabolic diseases. The aim of this study is to describe chrono-nutritional patterns based on the analysis of the eating habits of patients with severe obesity during the 24-h cycle and investigate a possible relationship between these profiles, the comorbidities, and the psychological status. From the overall evaluation of the chrono-nutritional profiles of 173 patients with severe obesity, four predominant eating patterns were obtained with a refined statistical model. A regression analysis was performed to determine the relationship between chrono-nutritional patterns, medical comorbidities, and psychological status. Profile 1 was the most frequent (46.2%) and characterised by the regular presence of the three main meals. The distribution of the chrono-nutritional profiles did not vary with BMI. Chrono-nutritional profiles affected predominantly psychological variables, with lower performances among chrono-nutritional profiles 3 (to eat during all the 24-h, with nibbling and snacking also during the night) and 4 (like the fourth but without night-eating). This finding could be useful in the assessment and treatment of patients with obesity, allowing the identification of patients with a higher probability of suffering from a psychopathological condition simply by knowing the patients’ dietary profiles.

Nitric oxide (NO) is produced within the airways and released with exhalation. Nasal NO (nNO) can be measured in a non-invasive way, with different devices and techniques according to the age and cooperation of the patients. Here, we conducted a narrative review of the literature to examine the relationship between nNO and some respiratory diseases with a particular focus on primary ciliary dyskinesia (PCD). A total of 115 papers were assessed, and 50 were eventually included in the review. nNO in PCD is low (below 77 nL/min), and its measurement has a clear diagnostic value when evaluated in a clinically suggestive phenotype. Many studies have evaluated the role of NO as a molecular mediator as well as the association between nNO values and genotype or ciliary function. As far as other respiratory diseases are concerned, nNO is low in chronic rhinosinusitis and cystic fibrosis, while increased values have been found in allergic rhinitis. Nonetheless, the role in the diagnosis and prognosis of these conditions has not been fully clarified.

Polymeric membranes are useful tools for water filtration processes, with their performance strongly dependent on the presence of hydrophilic dopants. In this study, polyaniline (PANI)-capped aluminosilicate (halloysite) nanotubes (HNTs) are dispersed into polyether sulfone (PES), with concentrations ranging from 0.5 to 1.5 wt%, to modify the properties of the PES membrane. Both undoped and HNT-doped PES membranes are investigated in terms of wettability (static and time-dependent contact angle), permeance, mechanical resistance, and morphology (using scanning electron microscopy (SEM)). The higher water permeance observed for the PES membranes incorporating PANI-capped HNTs is, finally, assessed and discussed vis-à-vis the real distribution of HNTs. Indeed, the imaging and characterization in terms of composition, spatial arrangement, and counting of HNTs embedded within the polymeric matrix are demonstrated using non-destructive Micro Particle Induced X-ray Emission (µ-PIXE) and Scanning Transmission Ion Microscopy (STIM) techniques. This approach not only exhibits the unique ability to detect/highlight the distribution of HNTs incorporated throughout the whole thickness of polymer membranes and provide volumetric morphological information consistent with SEM imaging, but also overcomes the limits of the most common analytical techniques exploiting electron probes. These aspects are comprehensively discussed in terms of practical analysis advantages.

Increased genetic risk for melanoma can occur in the context of germline pathogenic variants in high-penetrance genes, such as CDKN2A and CDK4, risk variants in low- to moderate-penetrance genes (MC1R and MITF), and possibly due to variants in emerging genes, such as ACD, TERF2IP, and TERT. We aimed to identify germline variants in high- and low- to moderate-penetrance melanoma risk genes in Brazilian patients with clinical criteria for familial melanoma syndrome. We selected patients with three or more melanomas or melanoma patients from families with three tumors (melanoma and pancreatic cancer) in first- or second-degree relatives. Genetic testing was performed with a nine-gene panel (ACD, BAP1, CDK4, CDKN2A, POT1, TERT, TERF2IP, MC1R, and MITF). In 36 patients, we identified 2 (5.6%) with germline pathogenic variants in CDKN2A and BAP1 and 4 (11.1%) with variants of uncertain significance in the high-penetrance genes. MC1R variants were found in 86.5%, and both red hair color variants and unknown risk variants were enriched in patients compared to a control group. The low frequency of germline pathogenic variants in the high-penetrance genes and the high prevalence of MC1R variants found in our cohort show the importance of the MC1R genotype in determining the risk of melanoma in the Brazilian melanoma-prone families.

Background: Multiple primary melanoma (MPM) is a diagnostic challenge even with ancillary imaging technologies available to dermatologists. In selected patients’ phenotypes, the use of imaging approaches can help better understand lesion characteristics, and aid in early diagnosis and management. Methods: Under a 5-year prospective single-center follow-up, 58 s primary melanomas (SPMs) were diagnosed in two first-degree relatives, with fair skin color, red hair, green eyes, and personal history of one previous melanoma each. Patients’ behavior and descriptive demographic data were collected from medical records. The information on the first two primary melanomas (PMs) were retrieved from pathology reports. The characteristics of 60 melanomas were collected from medical records, video dermoscopy software, and pathology reports. Reflectance confocal microscopy (RCM) was performed prior to excision of 22 randomly selected melanomas. Results: From February 2018 to May 2023, two patients underwent a pooled total of 214 excisional biopsies of suspect lesions, resulting in a combined benign versus malignant treatment ratio (NNT) of 2.0:1.0. The number of moles excised for each melanoma diagnosed (NNE) was 1.7:1.0 and 6.9:1.0 for the female and male patient respectively. The in-situ melanoma/invasive melanoma ratio (IIR) demonstrated a higher proportion of in-situ melanomas for both patients. From June 2018 to May 2023, a total of 58 SPMs were detected by the combination of total body skin exam (TBSE), total body skin photography (TBSP), digital dermoscopy (DD), and sequential digital dermoscopy imaging (SDDI) via comparative approach. The younger patient had her PM one month prior to the second and third cutaneous melanomas (CMs), characterizing a case of synchronous primary CM. The male older relative had a total of 7 nonsynchronous melanomas. Conclusions: This CM cohort is composed of 83.3% in-situ melanoma and 16.7% invasive melanoma. Both patients had a higher percentage of SPM with clinical nevus-like morphology (84.5%), global dermoscopic pattern of asymmetric multiple component (60.3%) and located on the lower limbs (46.6%). When RCM was performed prior to excision, 81% of SPM had features suggestive of malignancy. As well, invasive melanomas were more frequent in the lower limbs (40%). In the multivariate model, for the two high-risk patients studied, the chance of a not associated with nevus (“de novo”) invasive SPM diagnosis is 25 times greater than the chance of a diagnosis of a nevus-associated invasive SPM.

Small cell carcinoma of the ovary, hypercalcemic type (SCCOHT) is a rare and aggressive condition that is associated with the SMARCA4 mutation and has a dismal prognosis. It is generally diagnosed in young women. Here, we report a case of a young woman with SCCOHT harboring a rare molecular finding with a highly aggressive biological behavior. The patient had a somatic SMARCB1 mutation instead of an expected SMARCA4 alteration. Even though the patient was treated with high-dose chemotherapy followed by stem cell transplantation, she evolved with disease progression and died 11 months after her first symptoms appeared. We present a literature review of this rare disease and discuss the findings in the present patient in comparison to expected molecular alterations and options for SCCOHT treatment.

Detailed analysis of kinematics of the Milky Way disk in the solar neighborhood based on the GAIA DR3 catalog reveals the existence of peculiarities in the stellar velocity distribution perpendicular to the galactic plane. We study the influence of resonances—the outer Lindblad resonance and the outer vertical Lindblad resonance—of a rotating bar with stellar oscillations perpendicular to the plane of the disk, and their role in shaping the spatial and the velocity distributions of stars. We find that the Z and VZ distributions of stars with respect to LZ are affected by the outer Lindblad resonance. The existence of bar resonance with stellar oscillations perpendicular to the plane of the disk is demonstrated for a long (large semi-axis 5 kpc) and fast rotating bar with Ωb=60.0kms−1kpc−1. We show also that, in the model with the long and fast rotating bar, some stars in the 2:1 OLR region deviate far from their original places, entering the bar region. A combination of resonance excitation of stellar motions at the 2:1 OLR region together with strong interaction of the stars with the bar potential leads to the formation of the group of ‘escapees’, i.e., stars that deviate in R and Z—directions at large distances from the resonance region. Simulations, however, do not demonstrate any noticeable effect on VZ-distribution of stars in the solar neighborhood.

Dry yeast extracts (DYE) are applied to vineyards to improve aromatic and secondary metabolic compound content and wine quality; however, systematic information on the underpinning molecular mechanisms is lacking. This work aimed to unravel, through a systematic approach, the metabolic and molecular responses of Sauvignon Blanc berries to DYE treatments. To accomplish this, DYE spraying was performed in a commercial vineyard for two consecutive years. Berries were sampled at several time points after the treatment, and grapes were analyzed for sugars, acidity, free and bound aroma precursors, amino acids, and targeted and untargeted RNA-Seq transcriptional profiles. The results obtained indicated that the DYE treatment did not interfere with the technological ripening parameters of sugars and acidity. Some aroma precursors, including cys-3MH and GSH-3MH, responsible for the typical aromatic nuances of Sauvignon Blanc, were stimulated by the treatment during both vintages. The levels of amino acids and the global RNA-seq transcriptional profiles indicated that DYE spraying upregulated ROS homeostatic and thermotolerance genes, as well as ethylene and jasmonic acid biosynthetic genes, and activated abiotic and biotic stress responses. Overall, the data suggested that the DYE reduced berry oxidative stress through the regulation of specific subsets of metabolic and hormonal pathways.

Electrical power systems are exposed to transient disturbances that change the voltage and current signals of the network, which can interrupt power and damage equipment. In high-frequency phenomena, it is essential to study the transient recovery voltage (TRV) to ensure the electrical insulation limits of circuit breakers are not violated, thus leading to a safe and reliable operation. Adequate models are crucial to achieving satisfactory results in the studies, according to the range of frequency of the transient being evaluated. This paper presents a comprehensive literature review of methods and models for studying electromagnetic transients, focusing on TRV requests imposed on circuit breakers, in addition to fault-clearing simulations on real system modeling. The analyses are fundamental both for the evaluation of the amplitude of the voltage signal and for its rate of rise. We also compare the reviewed models and techniques to provide a handy resource for researchers.

Background: Asthma is the most common chronic condition in children. It is a complex non-communicable disease resulting from the interaction of genetic and environmental factors and characterized by heterogeneous underlying molecular mechanisms. Metabolomics, as with the other omic sciences, thanks to the joint use of high-throughput technologies and sophisticated multivariate statistical methods, provides an unbiased approach to study the biochemical–metabolic processes underlying asthma. The aim of this narrative review is the analysis of the metabolomic studies in pediatric asthma published in the past 10 years, focusing on the prediction of asthma development, endotype characterization and pharmaco-metabolomics. Methods: A total of 43 relevant published studies were identified searching the MEDLINE/Pubmed database, using the following terms: “asthma” AND “metabolomics”. The following filters were applied: language (English), age of study subjects (0–18 years), and publication date (last 10 years). Results and Conclusions: Several studies were identified within the three areas of interest described in the aim, and some of them likely have the potential to influence our clinical approach in the future. Nonetheless, further studies are needed to validate the findings and to assess the role of the proposed biomarkers as possible diagnostic or prognostic tools to be used in clinical practice.

People living with HIV (PLWH) remain at high risk of mortality and morbidity from vaccine-preventable diseases, even though antiretroviral therapy (ART) has restored life expectancy and general well-being. When, which, and how many doses of vaccine should be administered over the lifetime of PLWH are questions that have become clinically relevant. Immune responses to most vaccines are known to be impaired in PLWH. Effective control of viremia with ART and restored CD4+ T-cell count are correlated with an improvement in responsiveness to routine vaccines. However, the presence of immune alterations, comorbidities and co-infections may alter it. In this article, we provide a comprehensive review of the literature on immune responses to different vaccines in the setting of HIV infection, emphasizing the potential effect of HIV-related factors and presence of comorbidities in modulating such responses. A better understanding of these issues will help guide vaccination and prevention strategies for PLWH.

The microbiological quality assessment of drinking water (DW) and drinking water sources (DWSs) is based on the detection of indicator microorganisms (IMs). However, the relationship between IMs and pathogens has been questioned, as pathogens have been detected even in the absence of IMs, and vice versa. Therefore, the aim of this review was to evaluate the reliability of IMs by analysing the correlation between the presence of IMs and pathogens in water. This review focused on studies that reported statistical analyses of the relationship between traditional and alternative IMs and enteric pathogens in DWSs (groundwater, surface water, and rainwater) and in DW. Additionally, the main DW guidelines and regulations, along with a focus on the application of Quantitative Microbial Risk Assessment (QMRA), were also reported. The overall analysis of publications revealed a controversial correlation, characterised by high spatiotemporal variability, indicating the impossibility of identifying a reliable IM for any specific pathogen or water type. The association was also influenced by numerous factors, such as intrinsic characteristics of microorganisms, seasonal variations, sample number, water sample volume, and the detection method used. In conclusion, the detection of IMs should be considered complementary to, rather than a substitute for, the detection of pathogens.

Background: Monoclonal antibodies are designed to target specific proteins of COVID-19 and can be used as a treatment for people with mild to moderate infection and at a high risk of severe disease. Casirivimab/imdevimab, sotrovimab, and Bamlanivimab/etesevimab have been authorized for emergency use in the treatment of COVID-19. However, during pregnancy, these drugs have not been extensively studied. Methods: A total of 22 pregnant women with mild to moderate infection were treated with three different monoclonal antibodies, and efficacy and safety were evaluated in the first period and until six months of follow-up. Results: No infusion/allergic reactions occurred. No fatal or adverse events were observed in the pregnant women or fetus. The time of negativization with sotrovimab was shorter in comparison to Imdevimav/casirivimab (p = 0.0187) and Bamlanivimab/etesevimab (p < 0.00001). The time of negativization with sotrovimab was earlier in comparison to Imdevimav/casirivimab (t-value: 2.92; p = 0.0052) in vaccinated patients and similar in comparison to Imdevimav/casirivimab (t-value: 1.48; p = 0.08). In unvaccinated patients, sotrovimab was faster to achieve negativization in comparison to Bamlanivimab/etesevimab (t-value: 10.75; p < 0.0005). Conclusions: Pregnant COVID-19 patients receiving sotrovimab obtained better clinical outcomes. Pregnancy or neonatal complications were not observed after monoclonal treatment, confirming the safety and tolerability of these drugs in pregnant women.

Planning the best path for the energy system decarbonization is currently one of the issues of high global interest. At the European level, the recent policies dealing with the transportation sector have decided to ban the registration of light-duty vehicles powered by internal combustion engines fed by fossil fuels, from 2035. Regardless of the official positions, the major players (industries, politicians, economic and statistical institutions, etc.) manifest several opinions on this decision. In this paper, a mathematical model of a nation’s energy system is used to evaluate the economic impact of this decision. The model considers a superstructure that incorporates all energy conversion and storage units, including the entire transportation sector. A series of succeeding simulations was run and each of them was constrained to the achievement of the decarbonization level fixed, year by year, by the European community road-map. For each simulation, an optimization algorithm searches for a less costly global energy system, by including/excluding from the energy system the energy conversion units, storage devices, using a Mixed Integer Linear approach. Three optimization scenarios were considered: (1) a “free” scenario in which the only constraint applied to the model is the achievement of the scheduled decarbonization targets; (2) an “e-fuels” scenario, in which all new non-battery-electric light-duty vehicles allowed after 2035 must be fed with e-fuels; (3) a “pure electric” scenario, in which all new light-duty vehicles allowed after 2035 are battery-electric vehicles. The comparison of the optimum solutions for the three scenarios demonstrates that the less costly transition to a fully renewable energy system decarbonizes the transportation sector only when the share of renewable energy sources exceeds 90%. E-fueled light-duty vehicles always turn out to be a less expensive alternative than the electric vehicles, mainly because of the very high cost of the energy supply infrastructure needed to charge the batteries. Most of all, the costs imposed to society by the “e-fuels” and “pure electric” light-duty-vehicle decarbonizing scenarios exceed by 20% and 60%, respectively, the “free” transition scenario.

Circulating tumor cells (CTCs) and/or circulating tumor microemboli (CTM) from non-small cell lung cancer (NSCLC) patients may be a non-invasive tool for prognosis, acting as liquid biopsy. CTCs interact with platelets through the transforming growth factor-β/transforming growth factor-β receptor type 1 (TGF-β/TGFβRI) forming clusters. CTCs also may express the Cluster of Differentiation 47 (CD47) protein, responsible for the inhibition of phagocytosis, the “don’t eat me” signal to macrophages. Objectives: To isolate, quantify and analyze CTCs/CTMs from metastatic NSCLC patients, identify TGFβRI/CD47 expression in CTCs/CTMs, and correlate with progression-free survival (PFS). Methods: Blood (10 mL) was collected at two time-points: T1 (before the beginning of any line of treatment; T2 (60 days after initial collection). CTCs were isolated using ISET®. Immunocytochemistry was conducted to evaluate TGFβRI/CD47 expression. Results: 45 patients were evaluated. CTCs were observed in 82.2% of patients at T1 (median: 1 CTC/mL; range: 0.33–11.33 CTCs/mL) and 94.5% at T2 (median: 1.33 CTC/mL; 0.33–9.67). CTMs were observed in 24.5% of patients and significantly associated with poor PFS (10 months vs. 17 months for those without clusters; p = 0.05) and disease progression (p = 0.017). CTMs CD47+ resulted in poor PFS (p = 0.041). TGFβRI expression in CTCs/CTMs was not associated with PFS. Conclusion: In this study, we observed that CTC/CTM from NSCLC patients express the immune evasion markers TGFβRI/CD47. The presence of CTMs CD47+ is associated with poor PFS. This was the first study to investigate CD47 expression in CTCs/CTM of patients with NSCLC and its association with poor PFS.

Background: Different methods are used for the analysis of marginal and internal fit of partial crowns, but not all of them are applicable for in vivo studies. The aim of this review is to search the available methods, described in the current literature, to assess marginal and internal fit in partial crowns. Methods: an electronic search was performed on Pubmed and Web of Science databases to find studies published from 1 January 2017 up to 2 March 2023, following PRISMA guidelines and Cochrane handbook for systematic reviews. The search strategy applied was: “(marginal) AND (fit OR gap OR adaptation OR discrepancy) AND (inlay OR onlay OR partial crown)”. In vitro studies which evaluated marginal and internal fit on CAD CAM or 3D printed partial crowns were included in this review. Quality of the studies was assessed by using Quality Assessment Tool For In Vitro Studies (QUIN tool). Results: 22 studies were included. Among conventional methods, direct view with microscope, indirect view on resin replicas, and silicone replica technique (SRT) were used. Considering new digital methods, micro-CT, SRT 3D and triple scan technique (TST) were applied. Conclusions: Among 2D methods, direct view technique is the most used marginal fit analysis. For a more comprehensive evaluation, a 3D digital analysis is suggested. SRT and indirect view are the only 2D methods available for in vivo analysis. A protocol for the application of TST for assessment in vivo is now available, but no studies are reported in literature yet.

Buildings are responsible for a large part of energy demand worldwide. To collaborate to reduce this demand, this paper aims to present a computational model to analyze the performance of an earth–air heat exchanger (EAHE) based on computational fluid dynamics using the ANSYS/Fluent® software in the simulations. This passive air conditioning system uses the soil as a heat exchanger, taking advantage of the fact that the temperature of the soil at a certain depth remains relatively constant, regardless of the weather conditions above the surface, promoting heating, cooling, or ventilation for buildings. The air temperature values obtained were compared with experimental data from sensors installed in an EAHE at the Federal University of Technology—Parana, Ponta Grossa/Brazil (25.1° South, 50.16° West) to validate the computational model. A high computational effort would be demanded to perform these simulations involving the whole soil domain and the climatic boundary conditions. In order to optimize the numerical analysis of EAHE, two reduced models for the soil and heat exchanger domains were verified. First, a constant temperature of 23.7 °C was imposed on the surface of the exchanger tube, corresponding to the average soil temperature at a depth of 1.5 m. Afterward, a reduced soil domain extending 0.5 m in all directions from the heat exchanger serpentine was considered. Likewise, constant temperatures were imposed on the upper and lower surfaces of the soil domain, also obtained experimentally. In both cases, the temperature values obtained through the fast simulations showed good agreement compared to the experimental values. Barely explored in the literature, the thermal behavior of the two identical indoor environments at the university was also compared, in which the climatized environment, with the EAHE working in a closed loop, obtained milder and smaller amplitude air temperatures.

Anaplastic large cell lymphoma (ALCL) is a histological subtype of non-Hodgkin lymphoma, largely characterized by anaplastic lymphoma kinase (ALK) positivity, resulting from the chromosomal translocation t(2;5). We report a pediatric case of ALK-positive ALCL with primary concomitant involvement of bone and central nervous system (CNS); thereafter, a literature review about pediatric primary bone and primary CNS ALCL was conducted. According to the analyzed data, our case is unique because it is characterized by the contemporary involvement of the spine and CNS. During and after chemotherapy, our patient was monitored by detecting minimal residual disease (MRD) through the analysis of fusion transcript nucleophosmin-ALK. MRD assessment, not only in bone marrow but also in peripheral blood, seems to be a very powerful tool for predicting the prognosis of pediatric ALCL patients, as already described in the literature. Moreover, as shown in our case, it could be used during the follow-up for early recognition of relapse.

Using a complete sample of about 330,000 dwarf stars, well measured by Gaia DR3, limited to the galactic north and south solid angles |b|<75° and up to a vertical distance of 2 kpc, we analyze the vertical structure of the Milky Way stellar disks, based on projected tangential velocities. From selected subsamples dominated by their corresponding population, we obtain the thin and thick disk scale heights as hZ=279.76±12.49 pc and HZ=797.23±12.34 pc, respectively. Then from the simultaneous fitting of the sum of two populations over the whole sample, assuming these scale heights, we estimate the thick-to-thin disk number density ratio at the galactic plane to be ρT/ρt=0.750±0.049, which is consistent with a previous result by the authors: in the galactic plane there is a significant number of thick disk stars, possibly as many as thin disk ones, which also points to the existence of more thick disk stars than generally thought. The overall fit does not closely follow the data for |Z|>700 pc and points to the presence of more stars beyond the thin disk that cannot be accounted for by the two-disk model.

Online approaches for monitoring water quality can be an alternative aid to rapid decision-making in watershed management, especially reservoirs, given their vulnerability to the process of eutrophication. In this study, a modified water quality index (WQI) was developed using parameters that are easily measured with sensors, which would allow for the online monitoring of reservoirs. The modified WQI was based on WQICETESB and we used regression models to obtain values for the parameters: total phosphorus (TP), total nitrogen (TN), biochemical oxygen demand (BOD) and total solids (TS). Water quality data from reservoirs from 2003 to 2020 were used, which were provided by the Environmental Company of the State of São Paulo (CETESB), Brazil. The adjusted modified WQI employing weight redistribution (WQIRWAdj or WQISOL) presented the most promising results, with a Pearson correlation of 0.92 and a success rate of 72.6% and 97.0% for the CETESB and simplified classifications, respectively. WQISOL, which was proposed in the present study, exhibited a satisfactory performance, allowing the water quality of reservoirs to be monitored remotely and in real-time.

Viral infections can lead to transplant dysfunction, and their possible role in rejection is described. In total, 218 protocol biopsies performed in 106 children at 6, 12 and 24 months after transplantation were analyzed according to Banff ’15. RT-PCR for cytomegalovirus, Epstein-Barr virus, BK virus and Parvovirus B19 was performed on blood and bioptic samples at the time of transplant and each protocol biopsy. The prevalence of intrarenal viral infection increases between 6 and 12 months after transplantation (24% vs. 44%, p = 0.007). Intrarenal Parvovirus B19 infection is also associated with antibody-mediated rejection (ABMR) (50% ABMR vs. 19% T-cell-mediated rejection, p = 0.04). Moreover, Parvovirus infection is higher at 12 months of follow-up and it decreases at 48 months (40.4% vs. 14%, p = 0.02), while in 24% of grafts, Parvovirus is already detectable at the moment of transplantation. Intrarenal Parvovirus B19 infection seems to be related to ABMR in pediatric kidney recipients. The graft itself may be the way of transmission for Parvovirus, so performance of a PCR test for Parvovirus B19 should be considered to identify high-risk patients. Intrarenal Parvovirus infection presents mainly during the first-year post-transplantation; thus, we recommend an active surveillance of donor-specific antibodies (DSA) in patients with intrarenal Parvovirus B19 infection during this period. Indeed, it should be considered a treatment with intravenous immunoglobulins in patients with intrarenal Parvovirus B19 infection and DSA positivity, even in the absence of ABMR criteria for kidney biopsy.

Fabry disease (FD) is a rare genetic disorder caused by a deficiency in the α-galactosidase A enzyme, which results in the globotriaosylceramide accumulation in many organs, including the kidneys. Nephropathy is a major FD complication that can progress to end-stage renal disease if not treated early. Although enzyme replacement therapy and chaperone therapy are effective, other treatments such as ACE inhibitors and angiotensin receptor blockers can also provide nephroprotective effects when renal damage is also established. Recently, SGLT2 inhibitors have been approved as innovative drugs for treating chronic kidney disease. Thus, we plan a multicenter observational prospective cohort study to assess the effect of Dapagliflozin, a SGLT2 inhibitor, in FD patients with chronic kidney disease (CKD) stages 1–3. The objectives are to evaluate the effect of Dapagliflozin primarily on albuminuria and secondarily on kidney disease progression and clinical FD stability. Thirdly, any association between SGT2i and cardiac pathology, exercise capacity, kidney and inflammatory biomarkers, quality of life, and psychosocial factors will also be evaluated. The inclusion criteria are age ≥ 18; CKD stages 1–3; and albuminuria despite stable treatment with ERT/Migalastat and ACEi/ARB. The exclusion criteria are immunosuppressive therapy, type 1 diabetes, eGFR < 30 mL/min/1.73 m2, and recurrent UTIs. Baseline, 12-month, and 24-month visits will be scheduled to collect demographic, clinical, biochemical, and urinary data. Additionally, an exercise capacity and psychosocial assessment will be performed. The study could provide new insights into using SGLT2 inhibitors for treating kidney manifestations in Fabry disease.

The demonstration that F1FO (F)-ATP synthase and adenine nucleotide translocase (ANT) can form Ca2+-activated, high-conductance channels in the inner membrane of mitochondria from a variety of eukaryotes led to renewed interest in the permeability transition (PT), a permeability increase mediated by the PT pore (PTP). The PT is a Ca2+-dependent permeability increase in the inner mitochondrial membrane whose function and underlying molecular mechanisms have challenged scientists for the last 70 years. Although most of our knowledge about the PTP comes from studies in mammals, recent data obtained in other species highlighted substantial differences that could be perhaps attributed to specific features of F-ATP synthase and/or ANT. Strikingly, the anoxia and salt-tolerant brine shrimp Artemia franciscana does not undergo a PT in spite of its ability to take up and store Ca2+ in mitochondria, and the anoxia-resistant Drosophila melanogaster displays a low-conductance, selective Ca2+-induced Ca2+ release channel rather than a PTP. In mammals, the PT provides a mechanism for the release of cytochrome c and other proapoptotic proteins and mediates various forms of cell death. In this review, we cover the features of the PT (or lack thereof) in mammals, yeast, Drosophila melanogaster, Artemia franciscana and Caenorhabditis elegans, and we discuss the presence of the intrinsic pathway of apoptosis and of other forms of cell death. We hope that this exercise may help elucidate the function(s) of the PT and its possible role in evolution and inspire further tests to define its molecular nature.

Antimicrobial growth promoters (AGPs) in animal production have been related to the increase in multidrug-resistant bacteria. The AGP ban in many countries has highlighted the growing need for alternatives for feed additives. Considering the non-antibiotic anti-inflammatory theory of AGPs, chicks received three different doses of sodium salicylate (SS) in feed (10, 30, 90 mg/kg), basal diet (BD) was used as a negative control, and zinc bacitracin (ZB) was used as a positive control. Chicks were individually housed to increase the accuracy of the dose of SS ingested. Performance parameters and footpad dermatitis were evaluated weekly, while haematology, serum biochemistry, histopathology, and tibial dyschondroplasia were determined on Days 21 and 42. A linear dose-dependent decrease in haemoglobin concentration was observed, but the values were within the normal reference range. Among all the other evaluated parameters, no relevant differences between treatments were observed; however, not even the AGP group performed better than the control group. It is possible that the conditions in which the birds were raised were not stressful enough to allow for anti-inflammatories to demonstrate their beneficial effects on performance. Studies should be conducted where the animals are exposed to commercial conditions, as the presence of natural stressors could allow a better evaluation of the efficacy of the anti-inflammatory agent as a growth promoter.

High rates of fluorosis were reported worldwide as a result of human consumption of water with fluoride contents. Adjusting fluoride concentration in water as recommended by the World Health Organization (−1) is a concern and it needs to be conducted through inexpensive, but efficient techniques, such as phytoremediation. The application of phytohormones was investigated as a strategy to improve this process. Thus, the main goal of this research was to evaluate the effect of exogenous auxin and gibberellin on the tropical duckweed Eichhornia crassipes performance for fluoride phytoremediation. Definitive screening and central composite rotatable designs were used for experiments where fluoride concentration (5~15 mg L−1), phosphorus concentration (1~10 mg L−1), and pH (5~9) were assessed as well throughout 10 days. Fluoride contents were determined in solution and plant tissues by potentiometry. Higher concentrations of fluoride reflected on greater absorptions by plants, though in relative terms removal efficiencies were quite similar for all treatments (~60%). Auxin and acidic conditions favored fluoride removals per mass of plant. Fluoride accumulated mostly in leaves and auxin probably alleviated toxic effects on E. crassipes while gibberellin showed no effect. Therefore, E. crassipes could be employed as a fluoride accumulator plant for water treatment and exogenous auxin may be used to improve the process.
