Cystic fibrosis (CF) is a rare autosomal recessive disease most commonly affecting the Caucasian population. CF diagnosis can be a challenge due to the large spectrum of pathogenic variants in the CFTR gene and the effects of complex alleles. Next-generation sequencing has improved our understanding of the contribution of these complex alleles to the wide spectrum of CF clinical symptoms and to the response to medications. Herein, we studied nine CF patients from six unrelated families carrying the complex allele p.[Ile148Thr;Ile1023_Val1024del] with a frequency of 0.18%. All patients were from Central Mexico. This complex allele was found in trans with Class I and II pathogenic variants such as p.(Phe508del), and p.(Phe1078Profs*77)]. A targeted search of a dataset of 2217 exomes from healthy individuals revealed that eight individuals (0.18%) carried the p.(Ile148Thr) variant, but only one (0.022%), who was also born in Central Mexico, was a carrier of the complex allele. These findings show an enrichment of this p.[Ile148Thr;Ile1023_Val1024del] complex allele in Mexican CF patients in this region of Mexico. Finally, protein modeling revealed that this complex allele disrupts the secondary structure of the CFTR protein and might alter the ion flow.

Robotic Astronomical Observatories (RAOs) have provided very good results in different research projects in astrophysics/astronomy. Their applications in the detection, tracking, and identification of near-Earth objects have contributed to the identification of potentially dangerous objects for our security, such as near-Earth Objects (NEOs), near-Earth Asteroids (NEAs), meteors, and comets, whose trajectory changes can cause an impact on our planet. If advances in astrometry techniques (measuring the position and trajectory of Earth-orbiting objects) and photometry (variation in light curves) are considered together with the new sensors that work in the optical and near-infrared spectral ranges, a new observatory system that allows for the detection of nearby satellite objects in different spectral ranges and with better-defined optics can be developed. The present paper describes the design of a new observatory applied to the surveillance and tracking of satellites and other debris objects, the Satellite Robotic Observatory (SRO). Starting from general constraints from astronomy observatories, the design process has been determined, considering the main objectives, the necessary sensors, and several technical improvements that have contributed to a final configuration proposed for the SRO. The result is the design of a portable observatory model that can host at least two sensors to track and monitor satellite objects simultaneously.

Background: Type 2 diabetes (T2D) is one of the leading causes of mortality and is a public health challenge worldwide. Metformin is the first-choice treatment for T2D; its pharmacokinetics (PK) is facilitated by members of the solute carrier (SLC) superfamily of transporters, it is not metabolized, and it is excreted by the kidney. Although interindividual variability in metformin pharmacokinetics is documented in the Mexican population, its pharmacogenomics is still underexplored. We aimed to identify variants in metformin SLC transporter genes associated with metformin PK and response in Mexican patients. Methods: Using exome data from 2217 Mexican adults, we identified 86 biallelic SNVs in the eight known genes encoding SLC transporters, with a minor allele frequency ≥ 1%, which were analyzed in an inadequate glycemic control (IGC) association study in T2D metformin treated patients. Metformin PK was evaluated in a pediatric cohort and the effect of associated SNVs was correlated. Results: Functional annotation classified two SNVs as pathogenic. The association study revealed two blocks associated with IGC. These haplotypes comprise rs622591, rs4646272, rs4646273, and rs4646276 in SLC22A1; and rs1810126 and rs668871 in SLC22A3. PK profiles revealed that homozygotes of the SLC22A1 haplotype reached lower plasma metformin concentrations 2 h post administration than the other groups. Conclusions: Our findings highlight the potential of pharmacogenomics studies to enhance precision medicine, which may involve dosage adjustments or the exploration of alternative therapeutic options. These hold significant implications for public health, particularly in populations with a high susceptibility to develop metabolic diseases, such as Latin Americans.

The total antioxidant capacity (TAC) has been related to the development of and complications associated with chronic diseases, but its importance during obesity is not entirely clear. We conducted a systematic review and meta-analysis to clarify whether there are differences or similarities in the TAC between subjects with obesity (SO) and subjects with normal weight (NW). Following the recommendations of PRISMA and Cochrane, we performed a systematic search in the PubMed, Scopus, Web of Science, Cochrane, and PROSPERO databases, identifying 1607 studies. Among these, 22 studies were included in the final analysis, comprising 3937 subjects (1665 SO and 2272 NW) in whom serum TAC was measured, and from these 19,201 subjects, the correlation of serum TAC with anthropo-metabolic parameters was also estimated. The Newcastle–Ottawa method was used for the evaluation of the risk of bias. Using a random-effect model (REM), TAC was reduced in SO independently of age (SMD, −0.86; 95% CI −1.38 to −0.34; p = 0.0012), whereas malondialdehyde (SMD, 1.50; 95% CI 0.60 to 2.41), oxidative stress index (SMD, 1.0; 95% CI 0.16 to 1.84), and total oxidant status (SMD, 0.80; 0.22 to 1.37) were increased. There were seven significant pooled correlations of TAC with anthropometric and metabolic parameters: weight (r = −0.17), hip circumference (r= −0.11), visceral adipose index (r = 0.29), triglycerides (r = 0.25), aspartate aminotransferase (r = 0.41), alanine aminotransferase (r = 0.38), and uric acid (r = 0.53). Our results confirm a decrease in TAC and an increase in markers of oxidative stress in SO and underpin the importance of these serum biomarkers in obesity.

Climate change may shift species outside of their current climatic tolerances, which can be problematic for oceanic islands with limited options for species migration. Their alpine habitats, which represent islands within islands, are a special concern due to high endemism rates. The Canary Islands have two examples for such fragile ecosystems above 2000 m, on La Palma and Tenerife. This study contributes an assessment of the Canary Islands’ climatic habitat for 47 alpine plant species under observed climate change (1959–1989 versus 1990–2019 periods) as well as for future projections (2041–2060 and 2061–2080 periods). We analyzed the contraction of climatic habitats and migration requirements (biotic velocity) for each species to compensate for observed and predicted climate change. Our results suggest that temporary climate refugia exist on Tenerife but are insufficient on La Palma, where habitat loss due to historical climate change has already created an area of significant concern: the Cumbre Vieja Natural Park. On Tenerife, current alpine species can find suitable climate habitats in the caldera area and on the Teide-Pico Viejo volcano. That said, migration paths toward these refugia are long and complex, and human-assisted migration may be required. Species-specific statistics to support such management interventions are provided in this study.

Intracellular hydrogen peroxide (H2O2) levels can oscillate from low, physiological concentrations, to intermediate, signaling ones, and can participate in toxic reactions when overcoming certain thresholds. Fluorescent protein-based reporters to measure intracellular H2O2 have been developed in recent decades. In particular, the redox-sensitive green fluorescent protein (roGFP)-based proteins fused to peroxiredoxins are among the most sensitive H2O2 biosensors. Using fission yeast as a model system, we recently demonstrated that the gradient of extracellular-to-intracellular peroxides through the plasma membrane is around 300:1, and that the concentration of physiological H2O2 is in the low nanomolar range. Here, we have expressed the very sensitive probe roGFP2-Tpx1.C169S in two other model systems, budding yeast and human Jurkat cells. As in fission yeast, the biosensor is ~40–50% oxidized in these cell types, suggesting similar peroxide steady-state levels. Furthermore, probe oxidation upon the addition of extracellular peroxides is also quantitatively similar, suggesting comparable plasma membrane H2O2 gradients. Finally, as a proof of concept, we have applied different concentrations of zinc to all three model systems and have detected probe oxidation, demonstrating that an excess of this metal can cause fluctuations of peroxides, which are moderate in yeasts and severe in mammalian cells. We conclude that the principles governing H2O2 fluxes are very similar in different model organisms.

This article presents the design and implementation of a linear actuator based on NiTi Shape Memory Alloys with temperature and position measurements based on a magnetic sensor array and a set of thermistors. The position instrumentation is contact free to avoid friction perturbations; the position signal conditioning is carried out through the calculation of the response of each magnetic sensor, selecting the closest sensor to ensure accurate results on the full range of movement. Experimental results validate the accuracy of the position sensing with a competitive behaviour.

This paper presents the development of the UPMSat-2 sun sensor, from the design to on-orbit operation. It also includes the testing of the instrument, one of the most important tasks that needs to be performed to operate a sensor with precision. The UPMSat-2 solar sensor has been designed, tested, and manufactured at the Universidad Politécnica de Madrid (UPM) using 3D printing and COTS (photodiodes). The work described in this paper was carried out by students and teachers of the Master in Space Systems (Máster Universitario en Sistemas Espaciales—MUSE). The solar sensor is composed of six photodiodes that are divided into two sets; each set is held and oriented on the satellite by its corresponding support printed in Delrin. The paper describes the choice of components, the electrical diagram, and the manufacture of the supports. The methodology followed to obtain the response curve of each photodiode is simple and inexpensive, as it requires a limited number of instruments and tools. The selected irradiance source was a set of red LEDs and halogen instead of an AM0 spectrum irradiance simulator. Some early results from the UPMSat-2 mission have been analyzed in the present paper. Data from magnetometers and the attitude control system have been used to validate the data obtained from the sun sensor. The results indicate a good performance of the sensors during flight, in accordance with the data from the ground tests.

Fluorescent protein-based reporters used to measure intracellular H2O2 were developed to overcome the limitations of small permeable dyes. The two major families of genetically encoded redox reporters are the reduction-oxidation sensitive green fluorescent protein (roGFP)-based proteins fused to peroxiredoxins and HyPer and derivatives. We have used the most sensitive probes of each family, roGFP2-Tpx1.C169S and HyPer7, to monitor steady-state and fluctuating levels of peroxides in fission yeast. While both are able to monitor the nanomolar fluctuations of intracellular H2O2, the former is two-five times more sensitive than HyPer7, and roGFP2-Tpx1.C169S is partially oxidized in the cytosol of wild-type cells while HyPer7 is fully reduced. We have successfully expressed HyPer7 in the mitochondrial matrix, and it is ~40% oxidized, suggesting higher steady-state levels of peroxides, in the low micromolar range, than in the cytosol. Cytosolic HyPer7 can detect negligible H2O2 in the cytosol from mitochondrial origin unless the main H2O2 scavenger, the cytosolic peroxiredoxin Tpx1, is absent, while mitochondrial HyPer7 is oxidized to the same extent in wild-type and ∆tpx1 cells. We conclude that there is a bidirectional flux of H2O2 across the matrix and the cytosol, but Tpx1 rapidly and efficiently scavenges mitochondrial-generated peroxides and stops their steady-state cytosolic levels rising.

In this paper, we propose very simple analytical methodologies for modeling the behavior of photovoltaic (solar cells/panels) using a one-diode/two-resistor (1-D/2-R) equivalent circuit. A value of a = 1 for the ideality factor is shown to be very reasonable for the different photovoltaic technologies studied here. The solutions to the analytical equations of this model are simplified using easy mathematical expressions defined for the Lambert W-function. The definition of these mathematical expressions was based on a large dataset related to solar cells and panels obtained from the available academic literature. These simplified approaches were successfully used to extract the parameters from explicit methods for analyzing the behavior of solar cells/panels, where the exact solutions depend on the Lambert W-function. Finally, a case study was carried out that consisted of fitting the aforementioned models to the behavior (that is, the I-V curve) of two solar panels from the UPMSat-1 satellite. The results show a fairly high level of accuracy for the proposed methodologies.

In this work, the use of a calibration satellite (L2-CalSat) flying in formation with a Cosmic Microwave Background (CMB) polarization mission in an orbit located at the second Lagrange point, is proposed. The new generation of CMB telescopes are expected to reach unprecedented levels of sensitivity to allow a very precise measurement of the B-mode of polarization, the curl-like polarization component expected from gravitational waves coming from Starobinski inflationary models. Due to the CMB polarized signal weakness, the instruments must be subjected to very precise calibration processes before and after launching. Celestial sources are often used as external references for calibration after launch, but these sources are not perfectly characterized. As a baseline option, L2-CalSat is based on the CubeSat standard and serves as a perfectly known source of a reference signal to reduce polarization measurements uncertainty. A preliminary design of L2-CalSat is described and, according to the scanning strategy followed by the telescope, the influence of the relative position between the spacecrafts in the calibration process is studied. This new calibration element will have a huge impact on the performance of CMB space missions, providing a significant improvement in the measurements accuracy without requiring new and costly technological developments.

Swarm intelligence has appeared as an active field for solving numerous machine-learning tasks. In this paper, we address the problem of clustering data with missing values, where the patterns are described by mixed (or hybrid) features. We introduce a generic modification to three swarm intelligence algorithms (Artificial Bee Colony, Firefly Algorithm, and Novel Bat Algorithm). We experimentally obtain the adequate values of the parameters for these three modified algorithms, with the purpose of applying them in the clustering task. We also provide an unbiased comparison among several metaheuristics based clustering algorithms, concluding that the clusters obtained by our proposals are highly representative of the “natural structure” of data.

In the present work, an extensive analysis of the putative glutathione peroxidases (GPx) of the eukaryotic microorganism model Tetrahymena thermophila is carried out. A comparative analysis with GPx present in other Tetrahymena species and other very taxonomically diverse ciliates is also performed. A majority of ciliate GPx have replaced the selenocysteine (Sec) by Cys in its catalytic center, so they can be considered as phospholipid hydroperoxide glutathione peroxidases (PHGPx). Selenocysteine insertion sequence (SECIS) elements have been detected in several ciliate GPx that do not incorporate Sec in their amino acid sequences, and conversely, in other ciliate GPx with Sec, no SECIS elements are detected. These anomalies are analyzed and discussed. From the phylogenetic analysis using the ciliate GPx amino acid sequences, the existence of extensive intra- and interspecific gene duplications that produced multiple GPx isoforms in each species is inferred. The ancestral character of the selenoproteins is also corroborated. The analysis by qRT-PCR of six selected T. thermophila GPx genes has shown a quantitative differential expression between them, depending on the stressor (oxidizing agents, apoptotic inducer or metals) and the time of exposure.

The results of several research campaigns investigating cup anemometer performance carried out since 2008 at the IDR/UPM Institute are included in the present paper. Several analysis of large series of calibrations were done by studying the effect of the rotor’s geometry, climatic conditions during calibration, and anemometers’ ageing. More specific testing campaigns were done regarding the cup anemometer rotor aerodynamics, and the anemometer signals. The effect of the rotor’s geometry on the cup anemometer transfer function has been investigated experimentally and analytically. The analysis of the anemometer’s output signal as a way of monitoring the anemometer status is revealed as a promising procedure for detecting anomalies.

Due to the high dependence of photovoltaic energy efficiency on environmental conditions (temperature, irradiation...), it is quite important to perform some analysis focusing on the characteristics of photovoltaic devices in order to optimize energy production, even for small-scale users. The use of equivalent circuits is the preferred option to analyze solar cells/panels performance. However, the aforementioned small-scale users rarely have the equipment or expertise to perform large testing/calculation campaigns, the only information available for them being the manufacturer datasheet. The solution to this problem is the development of new and simple methods to define equivalent circuits able to reproduce the behavior of the panel for any working condition, from a very small amount of information. In the present work a direct and completely explicit method to extract solar cell parameters from the manufacturer datasheet is presented and tested. This method is based on analytical formulation which includes the use of the Lambert W-function to turn the series resistor equation explicit. The presented method is used to analyze commercial solar panel performance (i.e., the current-voltage–I-V–curve) at different levels of irradiation and temperature. The analysis performed is based only on the information included in the manufacturer’s datasheet.

Accurate and simple mathematical models are usually required to assess the performances of photovoltaic devices. In particular, it is common practice to use explicit models to evaluate the current–voltage (I–V) performance curves, mainly based on simple analytical expressions that enable the parameters determination with a little computational effort. Six different explicit photovoltaic models (i.e., explicit I–V equations) by different authors (Akbaba & Alattawi; El-Tayyan; Karmalkar & Haneefa; Das/Saetre et al.; Das; and Pindado & Cubas) are analyzed and compared. This comparison is carried out by fitting these models to eight I–V curves for different technologies, including Si, Si polycrystalline, Ga–As, and plastic solar cells. The accuracy of each model depends on the photovoltaic technology to which it is applied. The best fit to each I–V curve studied is normally obtained with a different model, with an average deviation under 2% in terms of short-circuit current (normalized RMSE). In general, the model proposed by Karmalkar & Haneefa shows the highest level of accuracy, and is a good fit for all I–V curves studied.

In the present work, the research derived from a wide experience on cup anemometer calibration works at IDR/UPM Institute (Instituto Universitario de Microgravedad “Ignacio Da Riva”) is summarized. This research started in 2008, analyzing large series of calibrations, and is focused on two main aspects: (1) developing a procedure to predict the degradation level of these wind sensors when working on the field and (2) modeling cup anemometer performances. The wear and tear level of this sensor is evaluated studying the output signal and its main frequencies through Fourier analysis. The modeling of the cup anemometer performances is carried out analyzing first the cup aerodynamics. As a result of this process, carried out through several testing and analytical studies since 2010, a new analytical method has been developed. This methodology might represent an alternative to the classic approach used in the present standards of practice such as IEC 64000-12.

This work proposes a new analytical model to extract the 1-Diode/2-Resistor solar cell/panel equivalent circuit parameters. The methodology is based on a reduced amount of experimentally measured information: short-circuit current, the slope of the I-V curve at that point, the open-circuit voltage, and the current and voltage levels, together with the slope of the I-V curve at the instantaneous operation point. This procedure is specially designed to analyze the performance of autonomous photovoltaic systems, which are most of the primary sources for spacecraft power. Results show good agreement with experimental data. Furthermore, this methodology allows for fast and accurate I-V curve maximum power point (MPP) identification.

The official data in relation to salaries paid in Spain from 1999 to 2014 has been analyzed. The inadequate data format does not reflect the whole salary distribution. Fréchet distributions have been fitted to the data. This simple distribution has similar accuracy in relation to the data when compared to other distributions (Log-Normal, Gamma, Dagum, GB2). Analysis of the data through the fitted Fréchet distributions reveals a tendency towards more balanced (i.e., less skewed) salary distributions from 2002 to 2014 in Spain.
