This contribution deals with the focusing and time–depth conversion of GPR data in a two-layer scenario. In a layered scenario, the different propagation velocities of the electromagnetic waves in different layers can induce distortion in the reconstruction. In particular, it is not possible, with common data processing, to achieve the focusing of both the targets embedded in the upper and deeper layers at the same time. We will present a strategy for mitigating this problem. The strength points of this strategy are its simplicity and the fact that it only requires a commercial code for the GPR data processing plus some steps easily implementable through a homemade code. This work extends a contribution of the same authors at the 20th International GPR conference held in Chanchung, China, in June 2024, and the conclusions are updated with three-dimensional numerical results, including depth slices determined directly in the spatial domain. Both numerical and experimental data will be shown.

Meagre (Argyrosomus regius) is one of the fast-growing species considered for sustainable aquaculture development along the Mediterranean and Eastern Atlantic coasts. The emergence of Systemic Granulomatosis (SG), a disease marked by multiple granulomas in various tissues, poses a significant challenge in meagre aquaculture. In the current study, we investigate the association of Mycobacterium spp. and SG in offshore aquaculture facilities in Sardinia, Italy. A total of 34 adult seemingly healthy meagre were arbitrarily collected and analyzed, combining histological, microbiological, molecular, metagenomics, and in situ techniques to investigate the presence of pathogens. Ziehl–Neelsen (ZN), periodic acid–schiff (PAS), and Giemsa stains were performed for the detection of acid-fast bacteria, common parasites, and fungi within granulomas, respectively. Granulomas were detected in 91% (31/34) of fish. The affected organs were kidney (88%), liver (47%), heart (41%), intestine (17.6%), and brain (5%). Acid-fast staining, along with Mycobacterium spp. specific quantitative PCR (qPCR), in situ hybridization (ISH) assay, and microbiological analyses showed negative results for the detection of Mycobacterium spp. and other bacteria implicated in granuloma formation. However, PCR amplification and sequencing of the 65-kDa heat shock protein gene revealed the presence of M. chelonae in 13% of both formalin-fixed and frozen liver tissues. Bacterial isolation failed to detect nontuberculous mycobacteria (NTM) and other bacteria typically associated with granulomas. Consistently, the use of an M. chelonae-specific probe in ISH failed to identify this bacterial species in granulomas. Collectively, results do not support the role of M. chelonae in the development of granulomas and suggest rejecting the hypothesis of a potential link between NTM and SG.

Lactococcus garvieae has been considered for a long time the only causal agent of lactococcosis. In recent years, different papers reported the involvement of other two bacterial species: Lactococcus petauri and Lactococcus formosensis. A different host tropism has been described for these species where L. garvieae and L. petauri are predominant species in fish and humans’ infections, while L. formosensis in bovine. L. garvieae has been reported as rare infectious agent in dog. This paper represents the first isolation of L. petauri in two domesticated dog cases from urine and skin samples, respectively. The recovered L. petauri has been identified using PCR and sequencing based on Internal Transcribe Spacer (ITS) and phylogenetic analysis showed that it belongs to the L. petauri cluster with a 100% of identity with sequences previously reported from fish isolates while there were differences with L. petauri isolated from urinary tract infection from humans. L. petauri in human infection has been considered not necessarily deriving from the ingestion of contaminated food but rather as an opportunistic pathogen colonization intestinal tract. Differences among virulotypes have been reported for humans and dogs, and a comparison was also made between the virulotyping of L. petauri and L. garvieae in dogs. The antimicrobial pattern showed susceptibility for the election treatment molecules. These data contribute to our understanding of the host trophism of this species which was misclassified for long time and provide new data on its virulence factors and antimicrobial resistance.

Sediments of alpine lakes serve as crucial records that reveal the history of lacustrine basins, offering valuable insights into the effects of global changes. One significant effect is the variation in rainfall regimes, which can substantially influence nutrient loads and sedimentation rates in lacustrine ecosystems, thereby playing a pivotal role in shaping biotic communities. In this study, we analyze subfossil chironomid assemblages within a sediment core from an alpine lake (western Italian Alps) to investigate the effects of rainfall and flood regime variations over the past 1200 years. Sediment characterization results highlight changes in sediment textures and C/N ratio values, indicating phases of major material influx from the surrounding landscape into the lake basin. These influxes are likely associated with intense flooding events linked to heavy rainfall periods over time. Flooding events are reflected in changes in chironomid assemblages, which in our samples are primarily related to variations in sediment texture and nutrient loads from the surrounding landscape. Increased abundances of certain taxa (i.e., Brillia, Chaetocladius, Cricotopus, Psectrocladius, Cricotopus/Orthocladius Parorthocladius) may be linked to higher organic matter and vegetation inputs from the surrounding landscape. Biodiversity decreased during certain periods along the core profile due to intense flood regimes and extreme events. These results contribute to our understanding of alpine lake system dynamics, particularly those associated with intense flooding events, which are still understudied.

The major histocompatibility complex (MHC) consists of genes involved in immune response and molecular discrimination between self and non-self. MHC genes are the most polymorphic in vertebrates. The origin and maintenance of polymorphism in MHC genes in populations is still unresolved. Mechanisms such as sexual selection and heterozygote advantage have been suggested as explanations for this high variability. In this study, a farmed population of rainbow trout (Oncorhynchus mykiss) characterized by the presence of specific MHC class IIB gene haplotypes at a frequency higher (30%) than that expected from random matings was investigated. Therefore, it was hypothesized that disassortative matings occur with an adaptive advantage for females, resulting in improved reproductive performance when mated with individuals with similar MHC haplotypes. Genetic analyses of the breeders were performed to define the MHC haplotypes and to perform specific matings. The effect of mating was evaluated by analyzing the survival rate of the offspring at various stages of incubation until swim-up. The reproductive performance of the offspring derived from specimens with similar haplotypes showed a better survival trend during the first life stages and reduced malformations. The results obtained are in contrast with the heterozygous advantage theory, therefore it was hypothesized, as for other salmonid species, the presence of a positive selection towards locally adapted MHC genes that promotes reproduction between genetically similar individuals.

Trace element pollution in the Persian Gulf originates from industrial activities, urbanization, shipping, and oil extraction, leading to accumulation in sediments, water, and marine life such as jellyfish. This study investigated trace element bioaccumulation in two jellyfish species, Crambionella orsini and Cassiopea andromeda, across different locations. Jellyfish samples were collected from the Mahshahr and Dilam ports, and their trace element concentrations were analyzed using inductively coupled plasma mass spectrometry (ICP-MS). The study revealed no significant differences in trace element concentrations between C. orsini and C. andromeda. However, levels of copper, iron, manganese, and nickel were significantly higher in specimens from Mahshahr compared to those from Dilam. No significant differences were observed for cadmium, mercury, vanadium, tin, magnesium, and selenium across sites. Lower concentrations of lead, arsenic, zinc, and cobalt were found in C. orsini from Dilam compared to Mahshahr. Additionally, the study found no significant correlation between trace element concentrations in the water and their accumulation in jellyfish bodies. These findings offer valuable insights into the distribution and bioaccumulation of trace elements in jellyfish populations across different marine environments in the Persian Gulf.

Background: The literature lacks a combined analysis of neurosurgical microvascular anastomosis training models. We performed a systematic literature search to provide an overview of the existing models and proposed a classification system based on the level of simulation and reproducibility of the microvascular anastomosis. Methods: The systematic literature search followed the PRISMA guidelines. We consulted MEDLINE, Web of Knowledge, and EMBASE independently for papers about bypass training models. Every training model was analyzed according to six tasks supposed to esteem their fidelity to the real operative setting by using a scoring system from zero to two. Finally, authors classified the models into five classes, from A to E, by summing the individual scores. Results: This study included 109 papers for analysis. Training models were grouped into synthetic tubes, ex vivo models (animal vessels, fresh human cadavers, human placentas) and in vivo simulators (live animals—rats, rabbits, pigs). By applying the proposed classification system, live animals and placentas obtained the highest scores, falling into class A (excellent simulators). Human cadavers and animal vessels (ex vivo) were categorized in class B (good simulators), followed by synthetic tubes (class C, reasonable simulators). Conclusions: The proposed classification system helps the neurosurgeon to analyze the available training models for microvascular anastomosis critically, and to choose the most appropriate one according to the skills they need to improve

Oysters are sessile, filter-feeding bivalve molluscs widely distributed in estuarine and coastal habitats worldwide. They constitute a valuable resource for fisheries and extensive aquaculture and provide essential ecological services. Yet, their genetic diversity and distribution remain understudied. The variability in shell morphology complicates species classification, which is influenced by environmental and genetic factors. Although molecular phylogenetics research has refined oyster taxonomy and identified approximately 100 extant species, numerous taxonomic uncertainties persist. In the present study, we aimed to document the occurrence of small flat oysters of the genus Ostrea along the Mediterranean coastal areas of Liguria and Sardinia (Italy). Specifically, 16S rRNA sequence data were used to identify Ostrea species. Our findings offer novel insights into the O. stentina species complex and O. neostentina, a new species in the Mediterranean coastal areas of Italy. The study data further our understanding of Ostrea species diversity, distribution, and evolutionary patterns.

In the context of sea state monitoring, reconstructing the wave field and estimating the sea state parameters from radar data is a challenging problem. To reach this goal, this paper proposes a fully data-driven, deep learning approach based on a convolutional neural network. The network takes as input the radar image spectrum and outputs the sea wave directional spectrum. After a 2D fast Fourier transform, the wave elevation field is reconstructed, and accordingly, the sea state parameters are estimated. The reconstruction strategy, herein presented, is tested using numerical data generated from a synthetic sea wave simulator, considering the spectral proprieties of the Joint North Sea Wave Observation Project model. A performance analysis of the proposed deep-learning estimation strategy is carried out, along with a comparison to the classical modulation transfer function approach. The results demonstrate that the proposed approach is effective in reconstructing the directional wave spectrum across different sea states.

We propose an artificial intelligence approach based on deep neural networks to tackle a canonical 2D scalar inverse source problem. The learned singular value decomposition (L-SVD) based on hybrid autoencoding is considered. We compare the reconstruction performance of L-SVD to the Truncated SVD (TSVD) regularized inversion, which is a canonical regularization scheme, to solve an ill-posed linear inverse problem. Numerical tests referring to far-field acquisitions show that L-SVD provides, with proper training on a well-organized dataset, superior performance in terms of reconstruction errors as compared to TSVD, allowing for the retrieval of faster spatial variations of the source. Indeed, L-SVD accommodates a priori information on the set of relevant unknown current distributions. Different from TSVD, which performs linear processing on a linear problem, L-SVD operates non-linearly on the data. A numerical analysis also underlines how the performance of the L-SVD degrades when the unknown source does not match the training dataset.

Revascularization surgery for the symptomatic hemisphere with hemodynamic impairment is effective for Moyamoya vasculopathy patients. However, careful patient selection is crucial and ideally supported by advanced quantitative hemodynamic imaging. Recently, blood oxygenation level-dependent cerebrovascular reactivity (BOLD-CVR) and quantitative magnetic resonance angiography with non-invasive optimal vessel analysis (qMRA-NOVA) have gained prominence in assessing these patients. This study aims to present the results of BOLD-CVR and qMRA-NOVA imaging along with the changes in cerebral hemodynamics and flow status following flow augmentation with superficial temporal artery–middle cerebral artery (STA-MCA) bypass in our Moyamoya vasculopathy patient cohort. Symptomatic patients with Moyamoya vasculopathy treated at the Clinical Neuroscience Center of the University Hospital Zurich who underwent hemodynamic and flow imaging (BOLD-CVR and qMRA-NOVA) before and after bypass were included in the analysis. Reduced hemispheric volume flow rates, as well as impaired BOLD-CVR, were measured in all 12 patients with Moyamoya vasculopathy before STA-MCA bypass surgery. Following the surgical procedure, post-operative BOLD-CVR demonstrated a non-significant increase in BOLD-CVR values within the revascularized, symptomatic middle cerebral artery territory and cerebral hemisphere. The results of the statistical tests should be viewed as indicative due to the small sample size. Additionally, post-operative qMRA-NOVA revealed a significant improvement in the hemispheric volume flow rate of the affected hemisphere due to the additional bypass flow rate. Our findings affirm the presence of hemodynamic and flow impairments in the symptomatic hemisphere of the Moyamoya vasculopathy patients. Bypass surgery proves effective in improving both BOLD-CVR impairment and the hemispheric volume flow rate in our patient cohort.

High-mountain lakes are freshwater ecosystems situated above the tree line which are known for their remote locations and limited accessibility. These ecosystems host simplified biotic communities primarily concentrated in the littoral zone and dominated by benthic macroinvertebrates that serve as bioindicators of environmental pressures. A two-year monitoring investigation was performed in July 2022 and July 2023 at Nero Lake (Cesana Torinese, Northwest Italy). Five sites along the lakeshore were selected for sampling physicochemical water parameters and macrobenthos. All collected data were analysed to compare trends across years and within specific sites. The results revealed that Nero Lake exhibited consistent macrobenthic communities across the two years studied, but significant differences were observed in its microhabitats. This suggests that substrate type and physicochemical water parameters strongly influence community composition. Chironomidae larvae and Mollusca were the dominant species, showing distinct associations with different substrates and environmental factors from one year to another. These findings contribute to our understanding of the intricate relationships between benthic macroinvertebrates and their environments, highlighting the necessity of detailed, small-scale assessments to comprehend ecosystem dynamics and develop effective conservation strategies.

The paper deals with a combined time–depth conversion strategy able to improve the reconstruction of voids embedded in an opaque medium, such as cavities, caves, empty hypogeal rooms, and similar targets. The combined time–depth conversion accounts for the propagation velocity of the electromagnetic waves both in free space and in the embedding medium, and it allows better imaging and interpretation of the underground scenario. To assess the strategy’s effectiveness, ground penetrating radar (GPR) data referred to as an experimental test in controlled conditions are accounted for and processed by two different approaches to achieve focused images of the scenario under test. The first approach is based on a classical migration algorithm, while the second one faces the imaging as a linear inverse scattering approach. The results corroborate that the combined time–depth conversion improves the imaging in both cases.

Red mullet (Mullus barbatus) is a commercially relevant fish species, yet epidemiological data on anisakid nematode infestation in M. barbatus are scarce. To fill this gap, we report the occurrence of Anisakis larvae in red mullet in the Ligurian Sea (western Mediterranean). This survey was performed between 2018 and 2020 on fresh specimens of M. barbatus (n = 838) from two commercial fishing areas (Imperia, n = 190; Savona, n = 648) in the Ligurian Sea. Larvae morphologically identified as Anisakis spp. (n = 544) were characterized using PCR-RFLP as Anisakis pegreffii. The overall prevalence of A. pegreffii was 24.46%; the prevalence at each sampling site was 6.32% for Imperia and 29.78% for Savona. Furthermore, 3300 larvae of Hysterothylacium spp. were detected in the visceral organs of fish coinfected with A. pegreffii, showing that coinfection with two parasitic species is not rare. This study provides a timely update on the prevalence of ascaridoid nematodes in red mullet of the Ligurian Sea, an important commercial fishing area in the Mediterranean.

The agro-livestock sector produces about one third of global greenhouse gas (GHG) emissions. Since more energy is needed to meet the growing demand for food and the industrial revolution in agriculture, renewable energy sources could improve access to energy resources and energy security, reduce dependence on fossil fuels, and reduce GHG emissions. Hydrogen production is a promising energy technology, but its deployment in the global energy system is lagging. Here, we analyzed the theoretical and practical application of green hydrogen generated by electrolysis of water, powered by renewable energy sources, in the agro-livestock sector. Green hydrogen is at an early stage of development in most applications, and barriers to its large-scale deployment remain. Appropriate policies and financial incentives could make it a profitable technology for the future.

Soft tissue regeneration holds significant promise for addressing various clinical challenges, ranging from craniofacial and oral tissue defects to blood vessels, muscle, and fibrous tissue regeneration. Mesenchymal stem cells (MSCs) have emerged as a promising tool in regenerative medicine due to their unique characteristics and potential to differentiate into multiple cell lineages. This comprehensive review explores the role of MSCs in different aspects of soft tissue regeneration, including their application in craniofacial and oral soft tissue regeneration, nerve regeneration, blood vessel regeneration, muscle regeneration, and fibrous tissue regeneration. By examining the latest research findings and clinical advancements, this article aims to provide insights into the current state of MSC-based therapies in soft tissue regenerative medicine.

The gut microbiota has become a topic of increasing importance in various fields, including aquaculture. Several fish species have been the subject of investigations concerning the intestinal microbiota, which have compared different variables, including the intestinal portions, the environment, and diet. In this study, the microbiota of farmed and wild brook trout (Salvelinus fontinalis) were analyzed, in which the wall and content of the medial portion of the intestine were considered separately. A total of 66 fish (age class 2+) were sampled, of which 46 were wild and 20 were farmed brook trout, in two different years. Microbiota data were obtained using a 16S metabarcoding approach by analyzing the V3–V4 hypervariable regions of the corresponding 16S rRNA. The data showed that the core microbiota of these species consist of Proteobacteria (Alpha- and Gammaproteobacteria), Actinobacteria, Firmicutes (Bacilli and Clostridia), and, only for farmed animals, Fusobacteria. The latter taxon’s presence is likely related to the fishmeal-based diet administered to farmed brook trout. Indeed, alpha and beta diversity analysis showed differences between wild and farmed fish. Finally, statistically significant differences in the microbiota composition were observed between the intestinal walls and contents of wild fish, while no differences were detected in reared animals. Our work represents the first study on the intestinal microbiota of brook trout with respect to both farmed and wild specimens. Future studies might focus on the comparison of our data with those pertaining to other fish species and on the study of other portions of the brook trout intestine.

As of October 2022, the COVID-19 pandemic continues to pose a major public health conundrum, with increased rates of symptomatic infections in vaccinated individuals. An ideal vaccine candidate for the prevention of outbreaks should be rapidly scalable, easy to administer, and able to elicit a potent mucosal immunity. Towards this aim, we proposed an engineered Escherichia coli (E. coli) Nissle 1917 (EcN) strain with SARS-CoV-2 spike protein (SP)-coding plasmid, which was able to expose SP on its cellular surface by a hybridization with the adhesin involved in diffuse adherence 1 (AIDA1). In this study, we presented the effectiveness of a 16-week intragastrically administered, engineered EcN in producing specific systemic and mucosal immunoglobulins against SARS-CoV-2 SP in mice. We observed a time-dependent increase in anti-SARS-CoV-2 SP IgG antibodies in the sera at week 4, with a titre that more than doubled by week 12 and a stable circulating titre by week 16 (+309% and +325% vs. control; both p < 0.001). A parallel rise in mucosal IgA antibody titre in stools, measured via intestinal and bronchoalveolar lavage fluids of the treated mice, reached a plateau by week 12 and until the end of the immunization protocol (+300, +47, and +150%, at week 16; all p < 0.001 vs. controls). If confirmed in animal models of infection, our data indicated that the engineered EcN may be a potential candidate as an oral vaccine against COVID-19. It is safe, inexpensive, and, most importantly, able to stimulate the production of both systemic and mucosal anti-SARS-CoV-2 spike-protein antibodies.

The directions of primary strain lines of local deformation in Epicardial and Endocardial layers have been the subject of debate in recent years. Different methods led to different conclusions and a complete assessment of strain direction patterns in large and variable (in terms of pathology) cohorts of healthy and diseased patients is still lacking. Here, we use local deformation tensors in order to evaluate the angle of strain lines with respect to the horizontal circumferential direction in both Epi- and Endo-layers. We evaluated this on a large group of 193 subjects including 82 healthy control and 111 patients belonging to a great variety of pathological conditions. We found that Epicardial strain lines obliquely directed while those of Endocardium are almost circumferential. This result occurs irrespective of pathological condition. We propose that the geometric vinculum characterizing Endocardium and Epicardium in terms of different lever arm length and orientation of muscular fibers during contraction inescapably requires Endocardial strain lines to be circumferentially oriented and this is corroborated by experimental results. Further investigations on transmural structure of myocytes could couple results presented here in order to furnish additional experimental explanations.

Copper (Cu) is a ubiquitous trace element in the aquatic environment, and is usually found at low levels. Copper environmental concentrations can be altered as a result anthropogenic activities. Shellfish are useful bioindicators to ensure adequate environmental monitoring. Thus, the aim of the present study was as follows: (a) determine the LC50 of copper(I) chloride in grooved carpet shell (Ruditapes decussatus) collected in the Santa Gilla lagoon (Sardinia, Italy), and (b) analyze the antioxidant biomarkers in digestive gland and gills of same specimens exposed to different concentrations of the above-mentioned metal (0.045, 0.45, and 0.90 mg/L) for 96 h. A withdrawal period of 96 h was considered for the treated clam, carrying out the same biochemical analyses, superoxide dismutase (SOD), catalase (CAT), selenium-dependent glutathione peroxidase (Se-GPx), glutathione S-transferases (GSTs), and total glutathione (GSH+2GSSG) in the two tissues. Different time and dose responses of the antioxidant biomarkers were recorded in the digestive glands and gills. Oxidative stress biomarkers highlighted the ability of Cu to induce oxidative stress in R. decussatus. Clam, following the withdrawal period of 96 h, has not been able to achieve the control levels of all biochemical markers in the digestive gland and gills. R. decussatus can be a suitable model to assess the ecotoxicity of copper in aquatic ecosystems. These findings may advance knowledge on the role and the effects of copper on oxidative stress biomarkers in grooved carpet shell. The metal ecotoxicity response can be useful to perform accurate biomarker-based monitoring programs using this bivalve species.

Striped venus (Chamelea gallina) is one of the most important fish resources on the west coast of the Adriatic Sea. Recently, there has been a widespread die-off of C. gallina populations in Friuli-Venezia Giulia (northern Adriatic Sea, Italy), probably due to unfavorable climatic events. Overall, wild populations have become increasingly rare due to many factors affecting the ecological balance of the species. In this study, the available literature was reviewed to determine the current state of knowledge on the biology, ecology, fisheries, and status of C. gallina populations with reference to populations in Friuli-Venezia Giulia. However, few data are available in terms of peer-reviewed articles; much of it can be found in the gray literature (e.g., project reports, ministerial reports, institutional websites, etc.). However, a critical review of the sources reveals that the species is as endangered as the habitats it inhabits. As a result, conservation and restoration efforts have been undertaken to date as part of some larger project to protect the species. Therefore, considering the ecological and economic importance of this species, the results of the new studies will be useful for the scientific community and will be a key element in the conservation of this species.

Infectious diseases place an economic burden on aquaculture and a limitation to its growth. An innovative approach to mitigate their impact on production is breeding for disease resistance: selection for domestication, family-based selection, marker-assisted selection, and more recently, genomic selection. Advances in genetics and genomics approaches to the control of infectious diseases are key to increasing aquaculture efficiency, profitability, and sustainability and to reducing its environmental footprint. Interaction and co-evolution between a host and pathogen can, however, turn breeding to boost infectious disease resistance into a potential driver of pathogenic change. Parallel molecular characterization of the pathogen and its virulence and antimicrobial resistance genes is therefore essential to understand pathogen evolution over time in response to host immunity, and to apply appropriate mitigation strategies.

Marketed globally, freshwater turtles are popular pets. Two species of the Chelydridae family are increasingly reported in Italy: the snapping turtle (Chelydra serpentina) and the alligator snapping turtle (Macrochelys temminckii). Both pose potential threats to public safety and habitat biodiversity. This update reports on their distribution and impact on biodiversity and human health. The recent increase in the number of C. serpentina in urban and rural areas suggests illegal importation into the country. Findings are reported for the north (35% and 100% for C. serpentina and M. temminckii, respectively) and the central-northern regions (60% for C. serpentina), predominantly Umbria and Latium, and the Tiber River catchment area in particular. Because omnivorous, Chelydridae species can affect native biodiversity; because they are carriers of pathogens, they endanger public health. Monitoring plans need to take account of this neglected threat.

Similar to canine inflammatory enteropathy, inflammatory bowel disease (IBD) is a chronic idiopathic condition characterized by remission periods and recurrent flares in which diarrhea, visceral pain, rectal bleeding/bloody stools, and weight loss are the main clinical symptoms. Intestinal barrier function alterations often persist in the remission phase of the disease without ongoing inflammatory processes. However, current therapies include mainly anti-inflammatory compounds that fail to promote functional symptoms-free disease remission, urging new drug discoveries to handle patients during this step of the disease. ALIAmides (ALIA, autacoid local injury antagonism) are bioactive fatty acid amides that recently gained attention because of their involvement in the control of inflammatory response, prompting the use of these molecules as plausible therapeutic strategies in the treatment of several chronic inflammatory conditions. N-palmitoyl-D-glucosamine (PGA), an under-researched ALIAmide, resulted in being safe and effective in preclinical models of inflammation and pain, suggesting its potential engagement in the treatment of IBD. In our study, we demonstrated that micronized PGA significantly and dose-dependently reduces colitis severity, improves intestinal mucosa integrity by increasing the tight junction proteins expression, and downregulates the TLR-4/NLRP3/iNOS pathway via PPAR-α receptors signaling in DNBS-treated mice. The possibility of clinically exploiting micronized PGA as support for the treatment and prevention of inflammation-related changes in IBD patients would represent an innovative, effective, and safe strategy.

Radar imaging from unmanned aerial vehicles (UAVs) is a dynamic research topic attracting huge interest due to its practical fallouts. In this context, this article provides a comprehensive review of the current state of the art and challenges related to UAV-based ground-penetrating radar (GPR) imaging systems. First, a description of the available prototypes is provided in terms of radar technology, UAV platforms, and navigation control devices. Afterward, the paper addresses the main issues affecting the performance of UAV-based GPR imaging systems. such as the control of the UAV platform during the flight to collect high-quality data, the necessity to provide accurate platform position information in terms of probing wavelength, and the mitigation of clutter and other electromagnetic disturbances. A description of the major applicative areas for UAV GPR systems is reported with the aim to show their potential. Furthermore, the main signal-processing approaches currently adopted are detailed and two experimental tests are also reported to prove the actual imaging capabilities. Finally, open challenges and future perspectives regarding this promising technology are discussed.

Adelmidrol is a promising palmitoylethanolamide (PEA) analog which displayed up-and-coming anti-inflammatory properties in several inflammatory conditions. Recent studies demonstrated that Adelmidrol is an in vitro enhancer of PEA endogenous production, through the so called “entourage” effect. The present study investigated the ability of Adelmidrol (1 and 10 mg/Kg per os) to increase the endogenous level of PEA in the duodenum and colon of mice after 21-day oral administration in the presence and absence of PPAR-γ inhibitor (1 mg/kg). The level of PEA was analyzed by HPLC-MS. The expression of PEA-related enzymatic machinery was evaluated by western blot and RT-PCR analysis. Our findings demonstrated that Adelmidrol significantly increased PEA levels in the duodenum and colon in a dose/time-dependent manner. We also revealed that Adelmidrol up regulated the enzymatic machinery responsible for PEA metabolism and catabolism. Interestingly, the use of the selective irreversible PPAR-γ antagonist did not affect either PEA intestinal levels or expression/transcription of PEA metabolic enzymes following Adelmidrol administration. The “entourage effect” with Adelmidrol as an enhancer of PEA was thus PPAR-γ-independent. The findings suggest that Adelmidrol can maximize a PEA therapeutic-based approach in several intestinal morbidities.

The first case of infection of Streptococcus iniae in Adriatic sturgeon (Acipenser naccarii) was recently reported in a raceway system located in Northern Italy. A second episode of infection in sturgeons with absence of mortality and evident clinical signs, was registered in November 2020 in the same farm and is reported in this study. Histopathological changes observed in infected organs are described. The strains isolated in the two episodes were compared using molecular analysis based on PCR, phylogeny and virulence factors analysis. Not all the major virulence factors were detected for the two strains; in particular the strains 78697, isolated in November, lacks cpsD, compared to the strains 64844, isolated in September. Moreover, genetic variations were reported for lctO and pmg genes. These findings let us hypothesize a different virulence of the strains in accordance with clinical findings related to the sturgeons.

Crohn’s disease (CD) is a chronic inflammatory gastrointestinal disorder requiring lifelong medications. The currently approved drugs for CD are associated with relevant side effects and several studies suggest an increased use of nutraceuticals among CD patients, seeking for what is perceived as a more “natural” approach in controlling this highly morbid condition. Nutraceuticals are foods or foods’ components with beneficial health properties that could aid in CD treatment for their anti-inflammatory, analgesic and immunoregulatory activities that come along with safety, high tolerability, easy availability and affordability. Depending on their biological effect, nutraceuticals’ support could be employed in different subsets of CD patients, both those with active disease, as adjunctive immunomodulatory therapies, and/or in quiescent disease to provide symptomatic relief in patients with residual functional symptoms. Despite the increasing interest of the general public, both limited research and lack of education from healthcare professionals regarding their real clinical effectiveness account for the increasing number of patients turning to unconventional sources. Professionals should recognize their widespread use and the evidence base for or against their efficacy to properly counsel IBD patients. Overall, nutraceuticals appear to be safe complements to conventional therapies; nonetheless, little quality evidence supports a positive impact on underlying inflammatory activity.

The genus Vibrio currently contains 147 recognized species widely distributed, including pathogens for aquatic organisms. Vibrio infections in elasmobranchs are poorly reported, often with identifications as Vibrio sp. and without detailed diagnostic insights. The purpose of this paper is the description of the isolation and identification process of Vibrio spp. following a mortality event of Scyliorhinus canicula juvenile reared in an Italian public aquarium. Following investigations aimed at excluding the presence of different pathogens of marine fish species (parasites, bacteria, Betanodavirus), several colonies were isolated and subjected to species identification using the available diagnostic techniques (a biochemical test, MALDI-TOF MS, and biomolecular analysis). Discrepancies were observed among the methods; the limits of biochemistry as a unique tool for Vibrio species determination were detected through statistical analysis. The use of the rpoB gene, as a diagnostic tool, allowed the identification of the isolates as V. crassostreae and V. cyclotrophicus. Although the pathogenic role of these microorganisms in lesser-spotted dogfish juveniles has not been demonstrated, and the presence of further pathogens cannot be excluded, this study allowed the isolation of two Vibrio species in less-studied aquatic organisms, highlighting the weaknesses and strengths of the different diagnostic methods applied.

Acute inflammation is particularly relevant in the pathogenesis of visceral hypersensitivity associated with inflammatory bowel diseases. Glia within the enteric nervous system, as well as within the central nervous system, contributes to neuroplasticity during inflammation, but whether enteric glia has the potential to modify visceral sensitivity following colitis is still unknown. This work aimed to investigate the occurrence of changes in the neuron–glial networks controlling visceral perception along the gut–brain axis during colitis, and to assess the effects of peripheral glial manipulation. Enteric glia activity was altered by the poison fluorocitrate (FC; 10 µmol kg−1 i.p.) before inducing colitis in animals (2,4-dinitrobenzenesulfonic acid, DNBS; 30 mg in 0.25 mL EtOH 50%), and visceral sensitivity, colon damage, and glia activation along the pain pathway were studied. FC injection significantly reduced the visceral hyperalgesia, the histological damage, and the immune activation caused by DNBS. Intestinal inflammation is associated with a parallel overexpression of TRPV1 and S100β along the gut–brain axis (colonic myenteric plexuses, dorsal root ganglion, and periaqueductal grey area). This effect was prevented by FC. Peripheral glia activity modulation emerges as a promising strategy for counteracting visceral pain induced by colitis.

Despite its possible therapeutic potential against COVID-19, the exact mechanism(s) by which palmitoylethanolamide (PEA) exerts its beneficial activity is still unclear. PEA has demonstrated analgesic, anti-allergic, and anti-inflammatory activities. Most of the anti-inflammatory properties of PEA arise from its ability to antagonize nuclear factor-κB (NF-κB) signalling pathway via the selective activation of the PPARα receptors. Acting at this site, PEA can downstream several genes involved in the inflammatory response, including cytokines (TNF-α, Il-1β) and other signal mediators, such as inducible nitric oxide synthase (iNOS) and COX2. To shed light on this, we tested the anti-inflammatory and immunomodulatory activity of ultramicronized(um)-PEA, both alone and in the presence of specific peroxisome proliferator-activated receptor alpha (PPAR-α) antagonist MK886, in primary cultures of murine alveolar macrophages exposed to SARS-CoV-2 spike glycoprotein (SP). SP challenge caused a significant concentration-dependent increase in proinflammatory markers (TLR4, p-p38 MAPK, NF-κB) paralleled to a marked upregulation of inflammasome-dependent inflammatory pathways (NLRP3, Caspase-1) with IL-6, IL-1β, TNF-α over-release, compared to vehicle group. We also observed a significant concentration-dependent increase in angiotensin-converting enzyme-2 (ACE-2) following SP challenge. um-PEA concentration-dependently reduced all the analyzed proinflammatory markers fostering a parallel downregulation of ACE-2. Our data show for the first time that um-PEA, via PPAR-α, markedly inhibits the SP induced NLRP3 signalling pathway outlining a novel mechanism of action of this lipid against COVID-19.

Annual and interannual dynamics of shellfish toxins and associated harmful algal species (HAS) were analyzed from 2015 to 2020 in Tortolì Lagoon (Sardinia, west Mediterranean Sea). Analysis of seasonal occurrence of different harmful algae, such as Dinophysis spp., Prorocentrum spp., Pseudo-nitzschia spp. and Alexandrium minutum, was performed. The species Dinophysis acuminata and Dinophysis sacculus were responsible for the accumulation of lipophilic toxins belonging to the okadaic acid group (OAs) and pectenotoxins2 (PTX2) in bivalve mollusks. The highest HAS detection was recorded in the winter months; in particular, Dinophysis spp. was mostly present in January–February. Out of 1090 analyzed mollusk samples, 39 were non-compliant, exceeding the legal limits (160 μg OA eq/kg e.p.) reported in Regulation 853/2004 of the European Commission. A statistical analysis related to the presence of OA and PTX2 in mollusks with various environmental parameters (pH, water temperature, dissolved oxygen, algal density) was implemented, proving a clear winter seasonality. The present study highlights the necessity to better understand the different factors able to influence the production and accumulation of toxins in bivalve mollusks bred in an important Sardinian production area. The contribution of this research is important not only from an environmental and productive point of view but also from the view of implementing management in order to mitigate any harm to human health.

The parrotfish Sparisoma cretense, a marine species native to the eastern and southern coastal areas of the Mediterranean, has extended its distribution northward. Here, we provide an update on its distribution based on currently published data and two new records from the coastline of Sardinia, Italy (central-western Mediterranean). The survey methods were scuba diving and spearfishing: one specimen of S. cretense was caught along the Argentiera coastline (northwest Mediterranean) and the others were photographed in the Gulf of Orosei, Osalla Bay (central-eastern Mediterranean). A literature update, together with new records, documents the distribution of this species in the northernmost areas of the Mediterranean. Probably a result of global warming, the ongoing northward expansion of S. cretense highlights the need for sampling campaigns to obtain timely updates on population and distribution of this thermophilic species.

The Mycobacterium fortuitum group (MFG) consists of about 15 species of fast-growing nontuberculous mycobacteria (NTM). These globally distributed microorganisms can cause diseases in humans and animals, especially fish. The increase in the number of species belonging to MFG and the diagnostic techniques panel do not allow to clarify their real clinical significance. In this study, biomolecular techniques were adopted for species determination of 130 isolates derived from fish initially identified through biochemical tests as NTM belonging to MFG. Specifically, gene sequencing and phylogenetic analysis were used based on a fragment of the gene encoding the 65 KDa heat shock protein (hsp65). The analyzes made it possible to confirm that all the isolates belong to MFG, allowing to identify the strains at species level. Phylogenetic analysis substantially confirmed what was obtained by gene sequencing, except for six strains; this is probably due to the sequences present in NCBI database. Although the methodology used cannot represent a univocal identification system, this study has allowed us to evaluate its effectiveness as regards the species of MFG. Future studies will be necessary to apply these methods with other gene fragments and to clarify the real pathogenic significance of the individual species of this group of microorganisms.

The fan mussel, Pinna nobilis, represents the largest bivalve endemic to the Mediterranean Sea. Since 2016, dramatic mass mortality of this species has been observed in several areas. The first surveys suggested that Haplosporidium pinnae (currently considered species-specific) was the main etiological agent, but recent studies have indicated that a multifactorial disease may be responsible for this phenomenon. In this study, we performed molecular diagnostic analyses on P. nobilis, P. rudis, and bivalve heterologous host species from the island of Sardinia to shed further light on the pathogens involved in the mass mortality. The results support the occurrence of a multifactorial disease and that Mycobacterium spp. and H. pinnae are not necessarily associated with the illness. Indeed, our analyses revealed that H. pinnae is not species-specific for P. nobilis, as it was present in other bivalves at least three years before the mass mortality began, and species of Mycobacterium were also found in healthy individuals of P. nobilis and P. rudis. We also detected the species Rhodococcus erythropolis, representing the first report in fan mussels of a bacterium other than Mycobacterium spp. and Vibrio spp. These results depict a complicated scenario, further demonstrating how the P. nobilis mass mortality event is far from being fully understood.

The present manuscript faces the problem of performing high-resolution Unmanned Aerial Vehicle (UAV) radar imaging in sounder modality, i.e., into the vertical plane defined by the along-tack and the nadir directions. Data are collected by means of a light and compact UAV radar prototype; flight trajectory information is provided by two positioning estimation techniques: standalone Global Positioning System (GPS) and Carrier based Differential Global Positioning System (CDGPS). The radar imaging is formulated as a linear inverse scattering problem and a motion compensation (MoCo) procedure, accounting for GPS or CDGPS positioning, is adopted. The implementation of the imaging scheme, which is based on the Truncated Singular Value Decomposition, is made efficient by the Shift and Zoom approach. Two independent flight tests involving different kind of targets are considered to test the imaging strategy. The results show that the CDGPS supports suitable imaging performance in all the considered test cases. On the other hand, satisfactory performance is also possible by using standalone GPS when the meter-level positioning error exhibits small variations during the radar integration time.

This paper deals with a feasibility study assessing the reconstruction capabilities of a small Multicopter-Unmanned Aerial Vehicle (M-UAV) based radar system, whose flight positions are determined by using the Carrier-Phase Differential GPS (CDGPS) technique. The paper describes the overall radar imaging system in terms of both hardware devices and data processing strategy for the case of a single flight track. The data processing is cast as the solution of an inverse scattering problem and is able to provide focused images of on surface targets. In particular, the reconstruction is approached through the adjoint of the functional operator linking the unknown contrast function to the scattered field data, which is computed by taking into account the actual flight positions provided by the CDGPS technique. For this inverse problem, we provide an analysis of the reconstruction capabilities by showing the effect of the radar parameters, the flight altitude and the spatial offset between target and flight path on the resolution limits. A measurement campaign is carried out to demonstrate the imaging capabilities in controlled conditions. Experimental results referred to two surveys performed on the same scene but at two different UAV altitudes verify the consistency of these results with the theoretical resolution analysis.

Abandoned quarries are frequently used as sites of illegal dumping of solid urban waste. These sites often occur nearby or within urban areas so that their detection may turn out to be quite difficult from the surface. This study focuses on the detection and geometrical characterization of a hidden landfill site located along the coastline of the Campi Flegrei, near Naples, Italy. Our approach is based on the analysis of historical topographic maps and aerial photographs, coupled with quantitative comparison of multitemporal digital elevation models obtained by digital photogrammetry and lidar techniques. The comparative analysis of topographic maps and aerial photos clearly shows modifications of the landscape associated with the urban development and quarrying activity, as well as the later filling of the quarry. The change detection analysis reveals that remarkable elevation changes occurred in the study area between 1956 and 2008. The average thickness of the landfill deposits is ca. 8 m, whereas the average volume is ca. 100,000 m3. The results of this work confirm the suitability of the used methodological approach that combines both qualitative and quantitative techniques for the detection of buried landfill sites. The geometric characterization of a landfill represents a fitting starting point for the further planning of geophysical site surveys and direct investigations aimed at the assessment of environmental hazards.

The aim of the present study was to evaluate the marketing policies of lobsters as influenced by different practices related to product quality in seven supermarkets located in Italy. Retailers were divided in two categories: large scale and medium scale. The two groups were compared to screen for differences and to assess differences in score distribution attributed to different practices related to product quality. Our results showed no statistical differences (p > 0.05) between the two categories. Lobsters were often marketed alive on ice and/or stocked for long periods in supermarket aquariums, highlighting the need to improve the specific European regulations on health, welfare, and quality at the market stage. Retail shop managers should be encouraged to develop better practices and policies in terms of marketing of lobsters. This will help in keeping the animals in good health and improve product quality at the marketing stages.
