This study examines the issues of privacy protection, data security, and query efficiency in blockchain-based electronic medical record (EMR) sharing. It proposes a secure storage and sharing scheme for EMR based on Hyperledger Fabric and the InterPlanetary File System (IPFS). To mitigate the privacy risks of data mining that could reveal patient identities, we establish an attribution channel in Hyperledger Fabric to store EMR ownership information and a data channel to store the storage location, digest, and usage records of medical data. Encrypted medical data are stored in the IPFS. To improve query efficiency in the blockchain, we integrate queryable medical data attributes into a composite key for conditional queries, avoiding complex data filtering processes. Additionally, we use a zero-knowledge proof combined with smart contracts for decentralized identity verification, eliminating reliance on third-party centralized verification services and enhancing system security. We also integrate AES and proxy re-encryption techniques to ensure data security during sharing. This scheme provides a more secure, efficient, and privacy-preserving approach for EMR systems, with significant practical implications and broad application potential.

Lactiplantibacillus plantarum, a typical ecological species against pathogens, used due to its bacteriocin yield in fermented foods, was proven to have the capacity to lower cholesterol. In this study, using L. plantarum ATCC8014 as the control, L. plantarum WLPL21 and ZDY04 were probed with whole-genome sequencing to ascertain their potential ability to lower cholesterol and yield bacteriocins, as well as to further evaluate their survival capacity in vitro. Our results showed 386 transport-system genes in both L. plantarum WLPL21 and ZDY04. Correspondingly, the in vitro results showed that L. plantarum WLPL21 and ZDY04 could remove cholesterol at 49.23% and 41.97%, respectively, which is 1.89 and 1.61 times that of L. plantarum ATCC8014. The survival rates of L. plantarum WLPL21 and ZDY04 in 1% H2O2, pH 3.0, and 0.3% bile salt were higher than those of L. plantarum ATCC8014. Our results exhibited a complete gene cluster for bacteriocin production encoded by L. plantarum WLPL21 and ZDY04, including plnJKR, plnPQAB, plnEFI, plnSUVWY, and plnJK; and plnMN, plnPQA and plnEFI, respectively, compared with only plnEF in L. plantarum ATCC8014. The present study suggests that the combination of genomic analysis with in vitro evaluations might be useful for exploring the potential functions of probiotics.

In order to meet the increasing pollutants discharge standard, the selective catalytic reduction (SCR) module in the diesel engine after-treatment system is an important means to reduce nitrogen oxide (NOx) emissions. SCR systems are prone to urea crystallization at lower temperatures, especially during the cold-start conditions of diesel engines. In this study, we use the diesel engine after-treatment system test bench to obtain the boundary parameter of the simulation modules, and the urea crystallization risk assessment model of the diesel SCR system is established. Comparing the computational fluid dynamics (CFD) results with the test bench results, it is shown that the predicted urea film distribution of the assessment model is in good agreement with the experimental results. In order to clarify the various factors that affect the urea crystallization risk, this paper conducts a simulation analysis on a nozzle and mixer structure and operating parameters. The CFD results indicate that the increase in urea spray time will increase the maximum urea film thickness on the SCR system mixer surface. Exhaust temperature is the most important influencing factor. When the diesel engine exhaust temperature increases from 190 °C to 300 °C, the maximum urea film thickness decreases by 32 and the urea film mass accumulation decreases by 5%. Exhaust flow has a small impact on urea crystallization risk. When the exhaust flow increases from 300 kg/h to 600 kg/h, the maximum urea film thickness decreases by 39% and the urea film mass accumulation decreases by about 1%. In addition, urea spray rate, nozzle numbers, spray angle, and spray cone angle are also factors that affect urea crystallization risk.

The stability of the filling roof—as an important bearing unit in the stope of the access stope in the filling mining method—is of great significance to guarantee the safe and efficient production of the mine. Arrangement of the mining approach in downward cemented filling stope is the key factor affecting the stability of the filling body roof. Based on a combination of laboratory tests, theoretical analysis, and numerical simulation, the influence of different mining approach arrangements on the stability of the filling body roof is analyzed. The weak filling surface is formed between adjacent mining paths. The mechanical strength of the weak filling surface is significantly reduced by laboratory experiments. The relationship between the distribution of the weak filling surface, azimuth angle, and the stability of the filling roof is further studied by numerical simulation. The results show that, when the upper and lower layered mining approaches are arranged vertically or nearly vertically, the areas of stress concentration and the plastic zone in the numerical simulation results are the smallest, and the stability of the filler roof is the best.

Probiotics are prospective for the prevention and treatment of cardiovascular diseases. Until now, systematic studies on the amelioration of hypercholesterolemia have been rare in terms of (cholesterol metabolism and transportation, reshaping of gut microbiota, as well as yielding SCFAs) intervention with lactic acid bacteria (LAB). In this study, strains of Lactiplantibacillus plantarum, WLPL21, WLPL72, and ZDY04, from fermented food and two combinations (Enterococcus faecium WEFA23 with L. plantarum WLPL21 and WLPL72) were compared for their effect on hypercholesterolemia. Comprehensively, with regard to the above aspects, L. plantarum WLPL21 showed the best mitigatory effect among all groups, which was revealed by decreasing total cholesterol (TC) and low-density lipoprotein cholesterol (LDL-C) levels, upregulated cholesterol metabolism (Cyp27a1, Cyp7b1, Cyp7a1, and Cyp8b1) levels in the liver, cholesterol transportation (Abca1, Abcg5, and Abcg8) in the ileum or liver, and downregulated Npc1l1. Moreover, it reshaped the constitution of gut microbiota; specifically, the ratio of Firmicutes to Bacteroidetes (F/B) was downregulated; the relative abundance of Allobaculum, Blautia, and Lactobacillus was upregulated by 7.48–14.82-fold; and that of Lachnoclostridium and Desulfovibrio was then downregulated by 69.95% and 60.66%, respectively. In conclusion, L. plantarum WLPL21 improved cholesterol metabolism and transportation, as well as the abundance of gut microbiota, for alleviating high-cholesterol-diet-induced hypercholesterolemia.

Orf is an important zoonotic disease caused by the Orf virus (ORFV) which can cause contagious pustular dermatitis in goats and sheep. Orf is widespread in most sheep-raising countries in the world, causing huge economic losses. Although diagnostic methods for ORFV infection already exist, it is still necessary to develop a time-saving, labor-saving, specific, low-cost and visual diagnostic method for rapid detection of ORFV in the field and application in grassroots laboratories. This study establishes a DNA extraction–free, real-time, visual recombinase–aided amplification (RAA) method for the rapid detection of ORFV. This method is specific to ORFV and does not cross-react with other common DNA viruses. The detection limits of the real-time RAA and visual judgment of the RAA assay at 95% probability were 13 and 21 copies per reaction for ORFV, respectively. Compared with qPCR, the sensitivity and specificity of the real-time RAA assay were 100%, and those of the visual RAA assay were 92.31% and 100.0%, respectively. The DNA extraction–free visual detection method of RAA established in this study can meet the needs of rapid onsite detection and grassroots laboratories and has important reference value and significance for the early diagnosis of diseased animals.

COVID-19 was officially declared a global pandemic disease on 11 March 2020, with severe implications for healthcare systems, economic activity, and human life worldwide. Fast and sensitive amplification of the severe acute respiratory syndrome virus 2 (SARS-CoV-2) nucleic acids is critical for controlling the spread of this disease. Here, a real-time reverse transcription recombinase-aided amplification (RT-RAA) assay, targeting conserved positions in the nucleocapsid protein gene (N gene) of SARS-CoV-2, was successfully established for SARS-CoV-2. The assay was specific to SARS-CoV-2, and there was no cross-reaction with other important viruses. The sensitivity of the real-time RT-RAA assay was 142 copies per reaction at 95% probability. Furthermore, 100% concordance between the real-time RT-RAA and RT-qPCR assays was achieved after testing 72 clinical specimens. Further linear regression analysis indicated a significant correlation between the real-time RT-RAA and RT-qPCR assays with an R2 value of 0.8149 (p < 0.0001). In addition, the amplicons of the real-time RT-RAA assay could be directly visualized by a portable blue light instrument, making it suitable for the rapid amplification of SARS-CoV-2 in resource-limited settings. Therefore, the real-time RT-RAA method allows the specific, sensitive, simple, rapid, and reliable detection of SARS-CoV-2.

Avian influenza viruses (AIVs) have the potential for cross-species transmission and pandemics. In recent years, clade 2.3.4.4 H5N6 AIVs are prevalent in domestic poultry, posing a threat to the domestic poultry industry and public health. In this study, two strains of H5N6 AIVs were isolated from chickens in Hebei, China, in 2019: A/chicken/Hebei/HB1907/2019(H5N6) and A/chicken/Hebei/HB1905/2019(H5N6). Phylogenetic analysis showed that both viral HA genes clustered in the 2.3.4.4h clade. Receptor binding analysis showed that the HB1905 strain preferentially binds to α-2,3-linked sialic acid (SA) receptors, while the HB1907 strain preferentially binds to α-2,3- and α-2,6-linked sialic acid (SA) receptors. During early infection, the HB1907 strain is highly replicable in MDCK cells, more so than the HB1905 strain. Pathogenicity assays in mice showed that both viruses could replicate in the lungs without prior adaptation, with HB1907 being more highly pathogenic in mice than the HB1905 strain. Significantly, both the HB1905 and HB1907 strains can be transmitted through direct contact among guinea pigs, but the transmission efficiency of the HB1907 strain through contact between guinea pigs is much greater than that of the HB1905 strain. These results strengthen the need for ongoing surveillance and early warning of H5N6 AIVs in poultry.

To investigate the effect of dimensional changes on the mechanical properties of cemented tailings backfill (CTB), uniaxial compression tests are performed on square CTBs of four different sizes. Combining digital image correlation (DIC) and acoustic emission (AE) methods, the fracture process and AE behavior characteristics of backfills with different sizes are analyzed. The results show that as the backfill size increases, its uniaxial compressive strength decreases, and its strength stabilizes gradually when it measures 100 mm. Under uniaxial compression loading, surface cracks on the smaller specimens evolve rapidly and aggressively, with no significant shedding area, whereas the larger specimens show plastic failure. The cracks expand and penetrate gradually, forming a large shedding area. As the specimen size increases, the backwards trend of the peak value of the ringing count relative to the peak value of the stress becomes increasingly evident. Combining the change law between the r value and the average frequency centroid, the sudden drop point of r value and the lowest value of average frequency centroid can be regarded as the precursor of macroscopic damage.

The laboratory discrimination tests of the rockburst tendency and AE tests under the uniaxial compression were conducted on the limestone. Three criterions were used to judge the rockburst tendency of the limestone with a buried depth of 600–1000 m under the same borehole. The rock damages were quantified by the AE events, ring count and energy. The fractal characteristics of these parameters were compared and analyzed. The characterization accuracy of these parameters on the damage process of the limestone with different rockburst tendencies was discussed. The AE parameters of the limestone with different rockburst tendencies increase sharply when the stress is near its peak. With the increase in the rockburst tendency, the variation of the AE event rate and ring count of the limestone is more and more consistent with the AE energy. Based on the characterization of the AE events and ring count, the damage process is characterized by a gradual evolution, while based on the energy characterization, it is characterized by a stepped evolution. The damage represented by the AE energy reveals the intensity and instantaneity of the rockburst failure. According to the verification of the CV value, the fractal dimension D of the AE energy fluctuates more gently with the increase in the rockburst tendency. It reflects the energy storage characteristics of the rock with a rockburst tendency in the loading process, which is consistent with the failure mechanism of the rockburst. Therefore, AE energy is more accurate to characterize and predict the failure of the limestone with a rockburst tendency.

Contagious ecthyma (Orf) is a highly contagious disease caused by Orf virus (ORFV) infection. Orf is prevalent all over the world and, not only affects the healthy development of sheep husbandry, but also threatens human health. However, there are no safe and effective vaccines or drugs for the prevention and treatment of Orf at present. In this study, we constructed a DNA plasmid expressing ORFV B2L and F1L genes as a DNA vaccine candidate, with purified B2L full-length protein and F1L truncated protein as subunit vaccine candidates. BALB/c mice were immunized with the DNA vaccine, subunit vaccine, as well as DNA prime-protein boost strategies. The results showed that compared with the DNA vaccine and subunit vaccine alone, the DNA prime-protein boost immunization group had a higher level of specific antibodies, stronger lymphocyte proliferation, and higher expression of cytokines such as IL-2, IL-4, IL-6, IFN-γ, and TNF-α, which are considered to cause a Th1/Th2 mixed cytokine response. Our results demonstrated that the DNA prime-protein boost immunization strategy induced stronger humoral and cellular immune responses, which have potential advantages in preventing ORFV infection.

To study the characteristics of the quiet period of acoustic emission (AE) during the rock failure process, rock models with an abnormal elastic modulus were established based on PFC2D. The calibration of the mesoscopic parameters was performed by the macroscopic mechanical parameters of granite samples obtained in uniaxial compression tests. An abnormal area of 2 × 100 mm2 was set at the center of the model and had two to six times the elastic modulus of the normal area. The evolution law of cracks and the characteristics of the AE quiet period were analyzed in detail. The main conclusions are as follows: (1) The axial stress of the abnormal area rises to the maximum value before the occurrence of the main fracture; in the AE quiet period, the axial stress in the abnormal area of elastic modulus increased by 2%–5%, then decreases rapidly after the AE quiet period. (2) As the elastic modulus increases, the interval time of the AE quiet period is prolonged by one to five times. Furthermore, many cracks occurred mainly in the abnormal area, and then a few cracks were generated before the AE quiet period. Cracks mainly focus on the elastic modulus abnormal area and propagate beyond the normal area after the AE quiet period. (3) The Z value is used as the indicator of the significance of the AE quiet period, and it changes with the elastic modulus of the abnormal area. The Z value without an abnormal area is the minimum of 1.1, and the elastic modulus of the abnormal area was set to three times the normal area, the Z value reaches the maximum of 6.08, and the Z value changes with the elastic modulus. The distribution of different elastic modulus is an important factor concerned with the characteristics of the AE quiet period.

With the development of modern pig raising technology, the increasing density of animals in pig houses leads to the accumulation of microbial aerosols in pig houses. It is an important prerequisite to grasp the characteristics of bacteria in aerosols in different pig houses to solve the problems of air pollution and disease prevention and control in different pig houses. This work investigated the effects of growth stages on bacterial aerosol concentrations and bacterial communities in pig houses. Three traditional types of closed pig houses were studied: farrowing (FAR) houses, weaning (WEA) houses, and fattening (FAT) houses. The Andersen six-stage sampler and high-volume air sampler were used to assess the concentrations and size distribution of airborne bacteria, and 16S rRNA gene sequencing was used to identify the bacterial communities. We found that the airborne bacterial concentration, community richness, and diversity index increased with pig age. We found that Acinetobacter, Erysipelothrix, Streptococcus, Moraxella, and Aerococcus in the microbial aerosols of pig houses have the potential risk of causing disease. These differences lead us to believe that disinfection strategies for pig houses should involve a situational focus on environmental aerosol composition on a case-by-case basis.

The strength of cemented paste backfill (CPB) directly affects mining safety and progress. At present, in-situ backfill strength is obtained by conducting uniaxial compression tests on backfill core samples. At the same time, it is time-consuming, and the integrity of samples cannot be guaranteed. Therefore guided wave technique as a nondestructive inspection method is proposed for the strength development monitoring of cemented paste backfill. In this paper, the acoustic parameters of guided wave propagation in the different cement-tailings ratios (1:4, 1:8) and different curing times (within 42 d) of CPBs were measured. Combined with the uniaxial compression strength of CPB, relationships between CPB strength and the guided wave acoustic parameters were established. Results indicate that with the increase of backfill curing time, the guided wave velocity decreases sharply at first; on the contrary, attenuation of guided waves increases dramatically. Finally, both velocity and attenuation tend to be stable. When the CPB strength increases with curing time, guided wave velocity shows an exponentially decreasing trend, while the guided wave attenuation shows an exponentially increasing trend with the increase of the CPB strength. Based on the relationship curves between CPB strength and guided wave velocity and attenuation, the guided wave technique in monitoring the strength development of CPB proves feasible.

A high-salt diet (HSD) is one of the key risk factors for hypertension and kidney injury. In this study, a HSD C57BL/6J mice model was established with 4% NaCl, and then different concentrations of Lactobacillus plantarum ZDY2013 were intragastrically administered for 2 weeks to alleviate HSD-induced renal injury. For the study, 16S rRNA gene sequencing, non-targeted metabonomics, real-time fluorescent quantitative PCR, and Masson’s staining were used to investigate the mechanism of L. plantarum ZDY2013 in alleviating renal damage. Results showed that HSD caused intestinal inflammation and changed the intestinal permeability of mice, disrupted the balance of intestinal flora, and increased toxic metabolites (tetrahydrocorticosteron (THB), 3-methyhistidine (3-MH), creatinine, urea, and L-kynurenine), resulting in serious kidney damage. Interestingly, L. plantarum ZDY2013 contributed to reconstructing the intestinal flora of mice by increasing the level of Lactobacillus and Bifidobacterium and decreasing that of Prevotella and Bacteroides. Moreover, the reconstructed intestinal microbiota significantly changed the concentration of the metabolites of hosts through metabolic pathways, including TCA cycle, ABC transport, purine metabolism, and histidine metabolism. The content of uremic toxins such as L-kynurenine, creatinine, and urea in the serum of mice was found to be decreased by L. plantarum ZDY2013, which resulted in renal injury alleviation. Our data suggest that L. plantarum ZDY2013 can indeed improve chronic kidney injury by regulating intestinal flora, strengthening the intestinal barrier, limiting inflammatory response, and reducing uremic toxins.

The Przewalski’s Gazelles (P. przewalskii) are affected by disorders that are characterized by deprived appetites, pica, emaciation and dyskinesia. The purpose of this study was to investigate the possibility of high molybdenum (Mo) in forage leading to copper (Cu) deprivation. The mineral contents in forage, soil, and samples of hair and blood from affected ranges were compared to healthy pasture. Blood parameters were also determined. Our results showed that the mean content of Mo in forage and soil in studied pasture was 5.17 and 4.17 μg/g, respectively. The Cu to Mo ratio in forage from affected and healthy areas was 1.26 and 5.89 μg/g, respectively. The Cu concentrations in hair and blood from gazelles in affected pasture were extremely lower (p < 0.01) than those in unaffected animals. The Mo contents in hair were higher (p < 0.01) than those in unaffected gazelles. The levels of Hb, RBC, PCV, MCV, and MCH in unaffected gazelles were significantly lower (p < 0.01) than those in unaffected gazelles. The levels of TP, ALB and GLB in blood were significantly lower (p < 0.01) than those in unaffected gazelles, while the levels of AST, LDH, CPK and ALP in serum were significantly higher (p < 0.01) than those in unaffected gazelles. The activities in serum T-AOC, SOD, GSH-Px, and CAT in affected gazelles were extremely lower (p < 0.01) than those in unaffected gazelles, while MDA was significantly higher (p < 0.01) than that in unaffected gazelles. Supplementation in copper sulphate (CuSO4) has prevented and cured this disorder. In summary, high molybdenum in feed and soil may lead to secondary Cu deficiency in gazelles, which can be alleviated by supplement of copper sulfate.

In order to investigate the influence of the ion exchange process on the permeability of rare earth ore bodies in the leaching process, a laboratory-scale comparative experiment with ammonium sulfate solution and deionized (DI) water as leaching solutions is conducted. Compared with the DI water leaching test, the permeability coefficient of the rare earth ore sample leached by the ammonium sulfate solution gradually decreases at the beginning and then increases with the completion of leaching. The physical and morphological evolutions of rare earth ore samples in this comparative experiment are also monitored by nuclear magnetic resonance and scanning electron microscopy. It is concluded that the change in the permeability coefficient arises from the adsorption–desorption of a large number of clay microparticles, resulting in a dynamic evolution of pore structures. Further mechanism analysis suggests that the change in internal ionic strength caused by ion exchange and leaching solution seepage promotes the adsorption–desorption behavior of clay microparticles.

Interior permanent magnet (IPM) motors are widely used in electric vehicles (EVs), benefiting from the excellent advantages of a more rational use of energy. For further improvement of energy utilization, this paper presents an analytical method of d- and q-axis inductance calculation for IPM motors with V-shaped rotor in no-load condition. A lumped parameter magnetic circuit model (LPMCM) is adopted to investigate the saturation and nonlinearity of the bridge. Taking into account the influence of magnetic field distribution on inductance, the winding function theory (WFT) is employed to accurately calculate the armature reaction airgap magnetic field and d- and q-axis inductances. The validity of the analytical technique is verified by the finite element method (FEM).

Chemokines are cytokines with chemoattractant capacities that exert their physiological functions through the binding of chemokine receptors. Thus, chemokine and receptor complexes exert important roles in regulating development and homeostasis during routine immune surveillance and inflammation. Compared to mammals, the physiology and structure of chemokine receptors in fish have not been systematically studied. Furthermore, the salmonid-specific whole genome duplication has significantly increased the number of functional paralogs of chemokine receptors. In this context, in the current study, trout exhibited 17 cxcr genes, including 12 newly identified and 5 previously identified receptors. Interestingly, gene expression of brain cxcr1 and cxcr4, kidney cxcr3 and cxcr4, and spleen cxcr3, cxcr4, and cxcr5 subtypes were altered by bacterial infection, whereas brain cxcr1, kidney cxcr1 and cxcr7, and liver cxcr2, cxcr3, and cxcr4 subtypes were changed in response to environmental changes. Based on protein structures predicted by ColabFold, the conserved amino acids in binding pockets between trout CXCR4.1 subtypes and human CXCR4 were also analyzed. Our study is valuable from a comparative point of view, providing new insights into the identification and physiology of salmonid chemokine receptors.
