Many older adults are increasingly embracing digital technology in the Republic of Korea. This study investigated the relationship between the digital skills of Korean older adults and their perceived health status and digital technology application for health promotion. This mixed-method study comprised a community survey of 434 older adults aged ≥65 in two cities in South Korea, followed by focus group interviews. Five types of digital skills, ‘operational internet skills’, ‘information navigation skills’, ‘social skills’, ‘creative skills’, and ‘mobile skills’, were measured using the LSE digital skill measurement instrument. Multivariable analysis identified the influence of digital skills on health-related outcomes. Among them, ‘social skills’ associated positively with self-rated health (β 0.37, 95%CI 0.08, 0.65). ‘Information navigation skills’ contributed positively to the use of digital technology and the internet for a healthy lifestyle in terms of improving eating habits (β 0.43, 95%CI 0.09, 0.77), accessing healthcare (β 0.53, 95%CI 0.21, 0.85), and accessing long-term care services (β 0.45, 95%CI 0.11, 0.79). Thematic analysis revealed that the study participants use Korean language-based resources such as Naver and Kakao Talk for social connection to promote a healthy lifestyle. This study concludes that encouraging initial and sustained use of the internet and enhancing digital skills among Korean older adults can promote active and healthy aging.

Avian leukosis virus (ALV) is an avian oncogenic retrovirus that can impair immunological function, stunt growth and decrease egg production in avian flocks. The capsid protein (P27) is an attractive candidate for ALV diagnostics. In the present study, a new hybridoma cell (1F8) stably secreting an anti-P27 monoclonal antibody (mAb) was developed. The mAb exhibited a high affinity constant (Ka) of 8.65 × 106.0 L/mol, and it could be used for the detection of ALV-A/B/J/K strains. Moreover, a total of eight truncated recombinant proteins and five synthetic polypeptides were utilized for the identification of the B-cell epitopes present on P27. The results revealed that 218IIKYVLDRQK227 was the minimal epitope recognized by 1F8, which had never been reported before. Additionally, the epitopes could strongly react with different ALV subgroup’s specific positive serum and had a complete homology among all the ALV subgroups strains. Finally, a new sandwich ELISA method was created for the detection of ALV antigens, demonstrating increased sensitivity compared to a commercially available ELISA kit. These results offer essential knowledge for further characterizing the antigenic composition of ALV P27 and will facilitate the development of diagnostic reagents for ALV.

Reynoutria ciliinervis (Nakai) Moldenke (R. ciliinervis) root, a traditional Chinese medicine, was found to exhibit remarkable pharmacological properties through a series of comprehensive investigations. Our study commenced with a qualitative phytochemical analysis that identified 12 bioactive compounds within the plant. Subsequently, utilizing ultraviolet-visible spectrophotometry, the methanol extract emerged as the optimal solvent extract, which was abundant in diverse classes of compounds such as carbohydrates, phenolics, steroids, alkaloids, phenolic acids, and tannins. In vitro antioxidant assays underscored the exceptional free radical scavenging, metal ion chelation, hydrogen peroxide scavenging, singlet oxygen quenching, and β-carotene bleaching capabilities of the methanol extract, significantly outperforming other solvent extracts. Further ultra high-performance liquid chromatography–electrospray ionization–quadrupole time of flight–mass spectrometry analysis revealed the presence of 45 compounds, predominantly anthraquinones and phenolics, in the methanol extract. The extract demonstrated robust stability under various conditions, including high temperatures, varying pH levels, and simulated gastrointestinal digestion as well as efficacy in inhibiting the oxidation in edible oils. Acute toxicity tests in mice confirmed the safety of the methanol extract and provided a valuable dosage reference for future studies. Importantly, high-dose methanol extract exhibited a significant pre-protective effect against D-galactosamine-induced liver injury in rats, as evidenced by reduced alanine aminotransferase, aspartate aminotransferase, γ-glutamyl transpeptidase, malondialdehyde levels, and elevated catalase and albumin levels. These findings suggest a potential role for the methanol extract of R. ciliinervis root in treating oxidative stress-related disorders, highlighting the plant’s immense medicinal potential. Our research offers a thorough evaluation of the bioactive components, antioxidant properties, stability, and liver-protecting effects of the methanol extract, setting the stage for deeper investigation and potential clinical applications.

As one of the most important economic traits for domestic animal husbandry, skeletal muscle is regulated by an intricate molecular network. Adenosine deaminase acting on RNA (ADAR1) involves various physiological processes and diseases, such as innate immunity and the development of lung adenocarcinoma, breast cancer, gastric cancer, etc. However, its role in skeletal muscle growth requires further clarification. Here, we explored the functions of ADAR1 in the myogenic process of goat skeletal muscle satellite cells (MuSCs). The ADAR1 transcripts were noticeably enriched in goat visceral tissues compared to skeletal muscle. Additionally, its levels in slow oxidative muscles like the psoas major and minor muscles were higher than in the fast oxidative glycolytic and fast glycolytic muscles. Among the two common isoforms from ADAR1, p110 is more abundant than p150. Moreover, overexpressing ADAR1 enhanced the proliferation and myogenic differentiation of MuSCs. The mRNA-seq performed on MuSCs’ knockdown of ADAR1 obtained 146 differentially expressed genes (DEGs), 87 upregulated and 59 downregulated. These DEGs were concentrated in muscle development and process pathways, such as the MAPK and cAMP signaling pathways. Furthermore, many DEGs as the key nodes defined by protein–protein interaction networks (PPI), including STAT3, MYH3/8, TGFβ2, and ACTN4, were closely related to the myogenic process. Finally, RNA immunoprecipitation combined with qPCR (RIP-qPCR) showed that ADAR1 binds to PAX7 and MyoD mRNA. This study indicates that ADAR1 promotes the myogenic development of goat MuSCs, which provides a useful scientific reference for further exploring the ADAR1-related regulatory networks underlying mammal skeletal muscle growth.

Bananas are economically important fruits, but they are vulnerable to mechanical damage during harvesting and transport. This study examined the effects of methyl jasmonate (MeJA) on the cell membrane integrity and membrane lipid metabolism of wounded banana fruits after harvest. The results showed that 10 and 50 μM MeJA treatments on mechanically wounded bananas significantly delayed ripening and senescence in comparison with the control. At the end of storage, MeJA-treated groups showed a significant reduction in electrolyte leakage and malondialdehyde content, indicating that MeJA protected cell membrane integrity. MeJA also led to a significant decrease in the activity of antioxidant enzymes, including lipoxygenase, diacylglycerol kinase, and lipid phosphate phosphatase. Furthermore, MeJA reduced phospholipase (C and D), phosphatidic acid, and diacylglycerol levels, as well as slowed down the decrease in phosphatidylcholine and phosphatidylinositol contents. Compared to the control, MeJA significantly downregulated the expression of MaPLDγ, MaPLDα, and MaPLDζ. Therefore, MeJA treatment could be a reliable method to delay the senescence of harvested banana fruits subjected to mechanical wounding.

Since Teleostei fins have a strong regenerative capacity, further research was conducted on the regulation of gene expression during fin regeneration. This research focuses on miRNA, which is a key post-transcriptional regulatory molecule. In this study, a miRNA library for the fin regeneration of zebrafish was constructed to reveal the differential expression of miRNA during fin regeneration and to explore the regulatory pathway for fin regeneration. Following the injection of miRNA agomir into zebrafish, the proliferation of blastema cells and the overall fin regeneration area were significantly reduced. It was observed that the miRNAs impaired blastocyte formation by affecting fin regeneration through the inhibition of the expressions of genes and proteins associated with blastocyte formation (including yap1 and Smad1/5/9), which is an effect associated with the Hippo pathway. Furthermore, it has been demonstrated that miRNAs can impair the patterns and mineralization of newly formed fin rays. The miRNAs influenced fin regeneration by inhibiting the expression of a range of bone-related genes and proteins in osteoblast lineages, including sp7, runx2a, and runx2b. This study provides a valuable reference for the further exploration of morphological bone reconstruction in aquatic vertebrates.

Anemone baicalensis Turcz., a botanical species with a rich historical background in traditional medicine for detoxification and insecticidal applications, possesses a vast, yet largely unexplored, therapeutic potential. This study primarily focused on conducting a qualitative phytochemical analysis of the plant, determining the active ingredient content and antioxidant activity of various solvent extracts. The qualitative phytochemical analysis revealed the presence of 12 different types of phytochemicals within the plant. Utilizing ultraviolet-visible spectrophotometry, we identified 11 active ingredients in 4 solvent extracts. Notably, the methanol extract was found to contain high concentrations of total carbohydrate, total monoterpenoid, total phenolic, total tannin, and total triterpenoid. In the antioxidant experiment, the methanol extract demonstrated superior scavenging abilities against 1,1-diphenyl-2-picrylhydrazyl radical, 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonicacid) diammonium salt, superoxide anion radical, and hydrogen peroxide, outperforming other extracts in chelation experiments aimed at reducing iron and metal ions. Consequently, the methanol extract was selected for further investigation. Subsequent ultrahigh-performance liquid chromatography-electrospray ionization-quadrupole-time of flight-mass spectrometry analysis revealed that the methanol extract contained 39 compounds, primarily phenolic compounds and triterpenoid saponins. Three stability assessments confirmed the extract’s stability under high temperatures, varying pH levels, and simulated gastrointestinal processes. Additionally, oil stability testing demonstrated its antioxidant capacity in extra virgin olive oil and cold-pressed sunflower seed oil media. An oral acute toxicity experiment conducted on mice not only confirmed the absence of acute toxicity in the methanol extract but also provided a dose reference for subsequent gastric protection experiments. Notably, the methanol extract exhibited significant gastroprotective effects against ethanol-induced gastric lesions in rats, as evidenced by histopathological and biochemical analyses. Specifically, the extract reduced levels of malondialdehyde, alanine aminotransferase, and aspartate aminotransferase while increasing glutathione, nitric oxide, and catalase, indicating its gastroprotective mechanism. These findings suggest that the methanol extract from the aerial part of Anemone baicalensis could be a promising therapeutic agent for conditions associated with oxidative imbalances. They underscore the plant’s potential therapeutic benefits and offer valuable insights into its antioxidant properties, thereby broadening our understanding of its medicinal potential.

As an important driving device, the permanent magnet synchronous motor (PMSM) plays a critical role in modern industrial fields. Given the harsh working environment, research into accurate PMSM fault diagnosis methods is of practical significance. Time–frequency analysis captures the rich features of PMSM operating conditions, and convolutional neural networks (CNNs) offer excellent feature extraction capabilities. This study proposes an intelligent fault diagnosis method based on continuous wavelet transform (CWT) and CNNs. Initially, a mechanism analysis is conducted on the inter-turn short-circuit and demagnetization faults of PMSMs, identifying and displaying the key feature frequency range in a time–frequency format. Subsequently, a CNN model is developed to extract and classify these time–frequency images. The feature extraction and diagnosis results are visualized with t-distributed stochastic neighbor embedding (t-SNE). The results demonstrate that our method achieves an accuracy rate of over 98.6% for inter-turn short-circuit and demagnetization faults in PMSMs of various severities.

(1) Introduction: The global rise of gastrointestinal diseases, including colorectal cancer and inflammatory bowel diseases, highlights the need to understand their causes. Diet is a common risk factor and a crucial regulator of gene expression, with alterations observed in both conditions. This study aims to elucidate the specific biological mechanisms through which diet influences the risk of bowel diseases. (2) Methods: We analyzed data from 436 participants from the BarcUVa-Seq population-based cross-sectional study utilizing gene expression profiles (RNA-Seq) from frozen colonic mucosal biopsies and dietary information from a semi-quantitative food frequency questionnaire. Dietary variables were evaluated based on two dietary patterns and as individual variables. Differential expression gene (DEG) analysis was performed for each dietary factor using edgeR. Protein–protein interaction (PPI) analysis was conducted with STRINGdb v11 for food groups with more than 10 statistically significant DEGs, followed by Reactome-based enrichment analysis for the resulting networks. (3) Results: Our findings reveal that food intake, specifically the consumption of blue fish, alcohol, and potatoes, significantly influences gene expression in the colon of individuals without tumor pathology, particularly in pathways related to DNA repair, immune system function, and protein glycosylation. (4) Discussion: These results demonstrate how these dietary components may influence human metabolic processes and affect the risk of bowel diseases.

Presently, ethylene oxide (EtO) is posing a significant threat to both human health and the environment due to occasional or deliberate emissions. However, few works so far have focused on this issue. It is urgent to explore novel and effective technology to protect against the threat of EtO. Herein, a series of AC/ZSM-5 composites were prepared to improve the adsorption performance for EtO, evaluated by dynamic breakthrough experiments. Particularly, the AC/ZSM-20% composite demonstrated a more excellent adsorption capacity of 81.9 mg/g at 25 °C and 50% RH than that of pristine AC and ZSM-5 with 32.5 and 52.3 mg/g, respectively. Moreover, the adsorption capacity of the AC/ZSM-20% composite remained constant even after five adsorption-desorption cycles. The adsorption mechanism of EtO on the composite is further revealed by density functional theory (DFT) calculations.

The dimensions of tenons in solid wood furniture significantly influence the mechanical performance of mortise and tenon joints. While previous studies have primarily focused on tenon length, width, and thickness, they often overlooked the impact of clearance between the mortise and tenon. This study investigates the effects of tenon length, tenon width, and clearance on the mechanical performance of mortise and tenon joints, aiming to enhance their bending moment capacity (BMC) and stiffness. A three-factor, three-level orthogonal test was conducted, utilizing range analysis and variance analysis to assess the effects of each factor on BMC and stiffness. The LSD post hoc test was employed to identify significant differences between levels of the same factor, and nonlinear regression analysis was used to fit the experimental results. Based on orthogonal experiment outcomes, a grey relational theory-based evaluation system was developed to assess the comprehensive performance of joints, including both moment capacity and stiffness. Results indicate that tenon length has the most significant effect on BMC, followed by clearance and tenon width, while clearance has the greatest impact on stiffness, followed by tenon length and tenon width. These findings are consistent with those obtained from grey relational analysis. When considering both BMC and stiffness as a comprehensive evaluation, the optimal combination is a tenon length of 40 mm, a tenon width of 35 mm, and a clearance of −0.1 mm. This study offers valuable insights for the rational design of mortise and tenon joints, contributing to improved performance and reduced manufacturing costs.

Skeletal muscle development is spotlighted in mammals since it closely relates to animal health and economic benefits to the breeding industry. Researchers have successfully unveiled many regulatory factors and mechanisms involving myogenesis. However, the effect of N6-methyladenosine (m6A) modification, especially demethylase and its regulated genes, on muscle development remains to be further explored. Here, we found that the typical demethylase FTO (fat mass- and obesity-associated protein) was highly enriched in goats’ longissimus dorsi (LD) muscles. In addition, the level of m6A modification on transcripts was negatively regulated by FTO during the proliferation of goat skeletal muscle satellite cells (MuSCs). Moreover, a deficiency of FTO in MuSCs significantly retarded their proliferation and promoted the expression of dystrophin-associated protein 1 (DAG1). m6A modifications of DAG1 mRNA were efficiently altered by FTO. Intriguingly, the results of DAG1 levels and its m6A enrichment from FB23-2 (FTO demethylase inhibitor)-treated cells were consistent with those of the FTO knockdown, indicating that the regulation of FTO on DAG1 depended on m6A modification. Further experiments showed that interfering FTO improved m6A modification at site DAG1-122, recognized by Insulin-like growth factor 2 mRNA-binding protein 1 (IGF2BP1) and consequently stabilized DAG1 transcripts. Our study suggests that FTO promotes the proliferation of MuSCs by regulating the expression of DAG1 through m6A modification. This will extend our knowledge of the m6A-related mechanism of skeletal muscle development in animals.

Background: Identifying mediators between obesity-related traits and lower respiratory tract infections (LRTIs) would inform preventive and therapeutic strategies to reduce the burden of LRITs. We aimed to recognize whether lung function and inflammatory factors mediate their associations. Methods: We conducted a two-step, two-sample Mendelian randomization (MR) analysis. Two-sample MR was performed on (1) obesity-related traits (i.e., body mass index [BMI], waist circumference [WC], and waist-to-hip ratio [WHR]) and LRTIs (i.e., acute bronchitis, acute bronchiolitis, bronchiectasis, influenza, and pneumonia), (2) obesity-related traits and potential mediators, and (3) potential mediators and LRTIs. Next, two-step MR was applied to infer whether the mediation effects exist. Results: We found that C-reactive protein (CRP), interleukin-6 (IL-6), and forced expiratory volume in the first second (FEV1) mediated 32.59% (95% CI: 17.90%, 47.27%), 7.96% (95% CI: 1.79%, 14.14%), and 4.04% (95% CI: 0.34%, 7.74%) of the effect of BMI on pneumonia, and they mediated 26.90% (95% CI: 13.98%, 39.83%), 10.23% (95% CI: 2.72%, 17.73%), and 4.67% (95% CI: 0.25%, 9.09%) of the effect of WC on pneumonia, respectively. Additionally, CRP, forced vital capacity (FVC), and FEV1 mediated 18.66% (95% CI: 8.70%, 28.62%), 8.72% (95% CI: 1.86%, 15.58%), and 8.41% (95% CI: 2.77%, 14.06%) of the effect of BMI on acute bronchitis, and they mediated 19.96% (95% CI: 7.44%, 32.48%), 12.19% (95% CI: 2.00%, 22.39%), and 12.61% (95% CI: 2.94%, 22.29%) of the effect of WC on acute bronchitis, respectively. Conclusions: Health interventions linked to reducing inflammation and maintaining normal lung function could help mitigate the risk of obesity-related LRTIs.

7075 aluminum alloy has been widely used because of its excellent mechanical properties. The fatigue crack acceleration induced by compressive overloading is detrimental to 7075 aluminum alloy in service, and the corresponding mechanisms are complex. To further reveal these mechanisms, the effect of microstructure on fatigue damage response ahead of the crack tip is simulated based on the damage-coupled crystal plasticity model for 7075 aluminum alloy. Results show that the ratchetting strain ahead of the crack tip at the compressive overload ratio of −3 is greater than that at the compressive overload ratio of −2, which is attributed to the fact that higher compressive overloading induces greater tensile residual stress ahead of the crack tip. The proportion of material points with no less than four activated slip systems increases after compressive overloading, indicating the number of activated slip systems ahead of the crack tip increases because of the compressive overload effect. This causes the dislocation multiplication rate to be faster, thereby accelerating the fatigue damage accumulation after compressive overloading. Furthermore, the effect of compressive overload changes the slip system activity, and this change in slip system activity depends on grain orientation. It is also found that for a grain ahead of the crack tip, the acceleration of fatigue damage accumulation caused by the effect of compressive overload is influenced by the orientation of its nearest neighbor grains. The findings provide further insight into the acceleration mechanisms induced by the compressive overload effect from a microscopic perspective.

The process of skeletal muscle development is intricate and involves the regulation of a diverse array of genes. Accurate gene expression profiles are crucial for studying muscle development, making it essential to choose the right reference genes for real-time quantitative PCR (RT-qPCR). In the present study, eight candidate reference genes were identified from our previous transcriptome sequencing analysis of caprine skeletal muscle satellite cells (MuSCs), and two traditional reference genes (ACTB and GAPDH) were assessed. The quantitative levels of the candidate reference genes were determined through the RT-qPCR technique, while the stability of their expression was evaluated utilizing the GeNorm, NormFinder, BestKeeper, and RefFinder programs. Furthermore, the chosen reference genes were utilized for the normalization of the gene expression levels of PCNA and Myf5. It was determined that conventional reference genes, including ACTB and GAPDH, were not appropriate for normalizing target gene expression. Conversely, RPL14 and RPS15A, identified through RNA sequencing analysis, exhibited minimal variability and were identified as the optimal reference genes for normalizing gene expression during the proliferation and differentiation of goat MuSCs. Our research offers a validated panel of optimal reference genes for the detection of differentially expressed genes in goat muscle satellite cells using RT-qPCR.

Pre-clinical studies have discovered the neuroprotective function and the benefit for cognitive function of choline. However, it remains unclear whether these benefits observed in animal studies also work in humans. The aims of this study are to examine the effects of dietary choline intake on cognitive function and cognitive decline during ageing in middle-aged and elderly Chinese. We included 1887 subjects aged 55~79 years with 6696 observations from the China Health and Nutrition Survey cohort study. The subjects were followed up for 6 to 21 years, with an average of 12.2 years. A dietary survey was conducted over 3 consecutive days with a 24 h recall, using household weight-recording methods. Based on the China Food Composition, data from USDA, and published literature, the dietary choline intake was calculated as the sum of free choline, phosphocholine, phosphatidylcholine, sphingomyelin, and glycerophosphocholine. Cognitive function was assessed using a subset of the Telephone Interview for Cognitive Status-modified (TICS-m) items. In order to eliminate the different weight of scores in each domain, the scores were converted by dividing by the maximum score in each domain, which ranged from 0 to 3 points. Higher cognitive scores represented better cognition. We used two-level mixed effect models to estimate the effects of dietary choline intake on cognitive score and cognitive decline rate in males and females, respectively. The average dietary choline intake was 161.1 mg/d for the baseline. After adjusting for confounders, the dietary choline intake was significantly associated with higher cognitive score in both males and females. The cognitive score in the highest quartile group of dietary choline was 0.085 for males and 0.077 for females–higher than those in the lowest quartile group (p < 0.01 for males, p < 0.05 for females). For every 10-year increase in age, the cognitive score decreased by 0.266 for males and 0.283 for females. The cognitive score decline rate of the third quartile group of dietary choline was 0.125/10 years lower than that of the lowest quartile group in females (p < 0.05). Dietary choline intake not only improves cognitive function, but also postpones cognitive decline during the aging process. The findings of this study highlight the neuroprotective benefit of choline in the middle-aged and elderly Chinese population, especially among females.

Plant seedling morphogenesis is considerably related to photosynthesis, pigment synthesis, and circadian periodicity during seedling development. We identified and cloned a maize zebra or crossbanding leaves mutant wk3735, which produces pale white kernels and was identified and plays a role in the equilibrium of the Redox state the in/out of ETC by active oxygen scavenging. Interestingly, it produces the zebra leaves during the production of the first seven leaves, which is apparently different from the mutation of homologs AtPTOX in Arabidopsis. It is intriguing to investigate how and why yellow crossbands (zebra leaf phenotype) emerge on leaves. As expected, chlorophyll concentration and photosynthetic efficiency both significantly declined in the yellow sector of wk3735 leaves. Meanwhile, we observed the circadian expression pattern of ZmPTOX1, which was further validated by protein interaction assays of the circadian clock protein TIM1 and ZmPTOX1. The transcriptome data of yellow (muW) and green (muG) sectors of knock-out lines and normal leaves of overexpression lines (OE) at the 5th-leaf seedling stage were analyzed. Zebra leaf etiolated sections exhibit a marked defect in the expression of genes involved in the circadian rhythm and rhythmic stress (light and cold stress) responses than green sections. According to the analysis of co-DEGs of muW vs. OE and muG vs. OE, terms linked to cell repair function were upregulated while those linked to environmental adaptability and stress response were downregulated due to the mutation of ZmPTOX1. Further gene expression level analyses of reactive oxygen species (ROS) scavenging enzymes and detection of ROS deposition indicated that ZmPTOX1 played an essential role in plant stress resistance and ROS homeostasis. The pleiotropic roles of ZmPTOX1 in plant ROS homeostasis maintenance, stress response, and circadian rhythm character may collectively explain the phenotype of zebra leaves during wk3735 seedling development.

The land-based recirculating aquaculture system (RAS) has been widely applied to fish farming as a new eco-friendly culture model. This system consists of circular culture tanks on land integrated with water treatment and recycling systems. This study investigated the growth performance of juvenile gibel carp (Carassius gibelio) cultured at high stocking density (HSD, 0.3 kg/m3) and low stocking density (LSD, 0.15 kg/m3) conditions in RAS, and evaluated the comprehensive economic profit of RAS. The body weight, body length, weight gain rate, and condition factor of gibel carp in the LSD group were significantly higher than those in the HSD group (p < 0.05). The feed conversion ratio increased significantly in the HSD group (p < 0.05). A histological analysis revealed a significantly higher density of white muscle fibers in the LSD group (p < 0.05). Relative mRNA expression levels showed that ubiquitin–proteasome system (UPS)-related genes, ub, psma2, and mafbx, were significantly expressed in the HSD group, while the s6k1 expression was elevated in the LSD group (p < 0.05). The mRNA expression levels of keap1 and hsp70 in the dorsal muscle were significantly higher in the HSD group (p < 0.05). Throughout the rearing period, the water temperature remained consistent between the two density groups. The pH value gradually decreased and the dissolved oxygen levels in the HSD group were generally lower than in the LSD group. The nitrite nitrogen (NO2−-N) content was higher in the HSD group. Compared to the LSD group, the return on investment was significantly lower in the HSD group. In conclusion, the water quality and growth rates of juvenile gibel carp were better in the LSD group. An appropriate stocking density improved the growth performance and aquaculture economic efficiency.

As a novel succinate dehydrogenase inhibitor (SDHI), pydiflumetofen (PYD) exhibits broad-spectrum bactericidal activity in various crops; however, little is yet known about its absorption, translocation, and metabolic behavior within plants. Cucumber and tomato plants were cultured in hydroponic conditions spiked at 0.5 mg/L of PYD, and samples were collected at certain intervals to investigate the residual fate of PYD within the plants. The results demonstrated that PYD was readily absorbed by the roots of both plants, with mean root concentration factors (RCFs) of 5.6–12.3 for cucumber and 5.0–12.4 for tomato. PYD exhibited higher translocation ability from stems to leaves and limited from roots to stems in cucumber, while comparably weak root-to-stem and stem-to-leaf translocation were observed in tomato. By the end of the exposure period, a mass loss of 51.55% and 56.67% was observed, and six and three metabolites were found to be generated in the cucumber and tomato systems, respectively. This study provides a foundation for comprehending the uptake and translocation of PYD and offers novel insights into its potential risks to agricultural products and food safety.

Many cities in China are facing the dual challenge of PM2.5 and PM10 pollution. There is an urgent need to develop a cost-effective method that can apportion both with high-time resolution. A novel and practical apportionment method is presented in this study. It combines the measurement of particle mass size distribution (PMSD) with an optical particle counter (OPC) and the algorithm of normalized non-negative matrix factorization (N-NMF). Applied in the city center of Baoding, Hebei, this method separates four distinct pollution factors. Their sizes (ordered from the smallest to largest) range from 0.16 μm to 0.6 μm, 0.16 μm to 1.0 μm, 0.5 μm to 17.0 μm, and 2.0 μm to 20.0 μm, respectively. They correspondingly contribute to PM2.5 (PM10) with portions of 26% (17%), 37% (26%), 33% (41%), and 4% (16%), respectively, on average. The smaller three factors are identified as combustion, secondary, and industrial aerosols because of their high correlation with carbonaceous aerosols, nitrate aerosols, and trace elements of Fe/Mn/Ca in PM2.5, respectively. The largest-sized factor is linked to dust aerosols. The primary origin regions, oxidation degrees, and formation mechanisms of each source are further discussed. This provides a scientific basis for the comprehensive management of PM2.5 and PM10 pollution.

Dilated cardiomyopathy (DCM) is one of the major causes of heart failure. Although significant progress has been made in elucidating the underlying mechanisms, further investigation is required for clarifying molecular diagnostic and therapeutic targets. In this study, we found that the mRNA level of protein phosphatase 2 regulatory subunit B’ delta (Ppp2r5d) was altered in the peripheral blood plasma of DCM patients. Knockdown of Ppp2r5d in murine cardiomyocytes increased the intracellular levels of reactive oxygen species (ROS) and inhibited adenosine triphosphate (ATP) synthesis. In vivo knockdown of Ppp2r5d in an isoproterenol (ISO)-induced DCM mouse model aggravated the pathogenesis and ultimately led to heart failure. Mechanistically, Ppp2r5d-deficient cardiomyocytes showed an increase in phosphorylation of STAT3 at Y705 and a decrease in phosphorylation of STAT3 at S727. The elevated levels of phosphorylation at Y705 in STAT3 triggered the upregulation of interleukin 6 (IL6) expression. Moreover, the decreased phosphorylation at S727 in STAT3 disrupted mitochondrial electron transport chain function and dysregulated ATP synthesis and ROS levels. These results hereby reveal a novel role for Ppp2r5d in modulating STAT3 pathway in DCM, suggesting it as a potential target for the therapy of the disease.

Enteral nutrition (EN) and foods for special medical purposes (FSMPs) can be used to meet the specific nutritional needs of patients. There are multiple types of EN products and nutritionally complete FSMPs on the market. The peptides in these products are important nutritional components, while their presence in different products remains unclear. To provide better clinical guidelines, we analyzed and compared the molecular weight (MW) distribution, types, and quantities of peptides and bioactive peptides of two EN products (liquid products) and two FSMPs with nutritionally complete formulas (powder products). Our results showed that each product had a unique peptide profile. The two liquid products and one powder product (Samples 1–3) had a higher content of peptides. Sample 1 contained 75.60% peptides with an MW less than 375 Da and contained 95.21% peptides with an MW less than 1000 Da, being rich in short peptides. Sample 2 and 3 had high levels of peptides with MW values between 180 Da and 2000 Da. Additionally, Sample 4 contained high levels of proteins, containing 69.18% peptides with MW values larger than 10,000 Da. Further, Sample 1 had more bioactive dipeptides and Sample 2 had more long bioactive peptides. Our results suggest that peptides in different EN and FSMP products are very different and should be evaluated in more detail. This will provide valuable information for clinical medical professionals, help them to guide patients with different physiological conditions better, and ultimately benefit patients.

Flash drought (FD) has attracted much attention due to its severe stress on vegetation ecosystems. Yet to date, the impacts of FD on vegetation ecosystems in different regions have not been fully evaluated and explored, especially for ecologically fragile areas. In this study, we identified the FD events in the Loess Plateau from 2000 to 2023 based on the attenuation rate in soil moisture percentile over time. The evolution process of FD, the driving roles of meteorological conditions and the responses of different vegetation types to FD were explored by vegetation indicators such as solar-induced chlorophyll fluorescence (SIF), SIFyield, SIF-RCI, etc. The results showed that FD events were predominantly concentrated in wetter areas with dense vegetation, with the highest frequency being 29. Meteorological factors contributed differently to the occurrence and development of FD. The responses of vegetation to FD were not only related to vegetation types (cropland was more sensitive to FD than forest and grassland) but were also significantly influenced by background climate. The SIFyield anomaly of vegetation was more sensitive than SIF anomaly and SIF-RCI. The results advance our understanding of the formation mechanisms of FD and facilitate the exploration of vegetative photosynthetic responses to FD.

Faba bean is an important pulse. It provides proteins for the human diet and is used in industrial foodstuffs, such as flours. Drought stress severely reduces the yield of faba bean, and this can be efficiently overcome through the identification and application of key genes in response to drought. In this study, PacBio and Illumina RNA sequencing techniques were used to identify the key pathways and candidate genes involved in drought stress response. During seed germination, a total of 17,927 full-length transcripts and 12,760 protein-coding genes were obtained. There were 1676 and 811 differentially expressed genes (DEGs) between the varieties E1 and C105 at 16 h and 64 h under drought stress, respectively. Six and nine KEGG pathways were significantly enriched at 16 h and 64 h under drought stress, which produced 40 and 184 nodes through protein–protein interaction (PPI) analysis, respectively. The DEGs of the PPI nodes were involved in the ABA (abscisic acid) and MAPK (mitogen-activated protein kinase) pathways, N-glycosylation, sulfur metabolism, and sugar metabolism. Furthermore, the ectopic overexpression of a key gene, AAT, encoding aspartate aminotransferase (AAT), in tobacco, enhanced drought tolerance. The activities of AAT and peroxidase (POD), the contents of cysteine and isoleucine, were increased, and the contents of malonaldehyde (MDA) and water loss decreased in the overexpressed plants. This study provides a novel insight into genetic response to drought stress and some candidate genes for drought tolerance genetic improvements in this plant.

Alzheimer’s disease (AD) is characterized by progressive cognitive decline and late-stage neurobehavioral issues marked by amyloid-beta plaques and Tau protein tangles. This study aims to investigate Fibulin-1(FBLN1) gene expression in the hippocampal tissue of Presenilin-1/Presenilin-2 conditional double-knockout (DKO) and double-transgenic (DTG) mice, using single-cell sequencing and experimental methods to verify abnormal methylation status and correlation with AD. Genomic DNA from DKO and DTG mice was used for genotyping. Reduced Representation Bisulfite Sequencing (RRBS) identified 10 genes with abnormal methylation changes, with protein–protein interaction (PPI) analysis highlighting five core genes, including FBLN1. Single-cell sequencing, RT-PCR, and Western blotting (WB) were used to analyze FBLN1 mRNA and protein levels in the hippocampal tissues of early-stage and mid-stage AD DKO, DTG, and CBAC57 mice. RRBS identified 10 genes with abnormal methylation, with PPI highlighting five core genes. Single-cell sequencing showed significant FBLN1 expression in AD groups. RT-PCR and WB indicated elevated FBLN1 mRNA and protein levels in mid-stage AD DKO and DTG mice compared to CBAC57 mice, with no differences in early-stage AD DKO and CBAC57 mice. RRBS revealed hypomethylation of the FBLN1 gene in mid-stage AD DKO mice. Elevated FBLN1 expression in AD models suggests an age-dependent neurodegenerative mechanism independent of amyloid-beta deposition. This study enhances our understanding of AD’s epigenetic mechanisms, which will aid targeted diagnosis, treatment, and prognosis.

In the serviced components of a 7075 aluminum alloy, the propagation of fatigue crack can be retarded because of the overload effect; however, the corresponding retardation mechanisms are complex. To provide further insights into the retardation mechanisms of 7075 aluminum alloys, this study addresses the crack tip damage response of a cracked 7075 aluminum alloy under an overload effect. Based on the dual-scale modeling approach and the damage-coupled crystal plasticity model, the effect of the microstructure of a 7075 aluminum alloy on the damage behavior ahead of the crack tip under an overload was studied. The factors affecting fatigue damage accumulation ahead of the crack tip, such as dislocation density, the variation in the activities of slip systems, and the orientation effect of the nearest neighbor grains, are described. The results show that for the 7075 aluminum alloy, the compressive residual stress induced by the overload effect not only decreases the number of activated slip systems, but also lowers the rate of increase in dislocation density. This causes a decrease in fatigue damage accumulation during deformation. Moreover, the overload effect decreases the slip system activity as well as the resultant plastic slip; however, the decrease in plastic slip varies with the grain orientation, indicating that the overload effect depends on the grain orientation. It can also be found that both the damage strain energy release rate and lattice strain are influenced by the orientation of the nearest neighbor grains, which can eventually affect the overload effect. These findings contribute to understanding the retardation mechanisms from a microscopic perspective and provide guidance on improving the material design of a 7075 aluminum alloy to some extent.

Microorganisms harbor catabolic plasmids to tackle refractory organic pollutants, which is crucial for bioremediation and ecosystem health. Understanding the impacts of plasmids on hosts provides insights into the behavior and adaptation of degrading bacteria in the environment. Here, we examined alterations in the physiological properties and gene expression profiles of Rhodococcus sp. strain p52 after losing two conjugative dioxin-catabolic megaplasmids (pDF01 and pDF02). The growth of strain p52 accelerated after pDF01 loss, while it decelerated after pDF02 loss. During dibenzofuran degradation, the expression levels of dibenzofuran catabolic genes on pDF01 were higher compared to those on pDF02; accordingly, pDF01 loss markedly slowed dibenzofuran degradation. It was suggested that pDF01 is more beneficial to strain p52 under dibenzofuran exposure. Moreover, plasmid loss decreased biofilm formation, especially after pDF02 loss. Transcriptome profiling revealed different pathways enriched in upregulated and downregulated genes after pDF01 and pDF02 loss, indicating different adaptation mechanisms. Based on the transcriptional activity variation, pDF01 played roles in transcription and anabolic processes, while pDF02 profoundly influenced energy production and cellular defense. This study enhances our knowledge of the impacts of degradative plasmids on native hosts and the adaptation mechanisms of hosts, contributing to the application of plasmid-mediated bioremediation in contaminated environments.

Sulfonamides are not only widely applied in clinics but also highly valued in animal husbandry. Recently, it has become common for sulfonamide residues to exceed the standard limits in food, which can affect human health. Current regulations limit these residues. Therefore, we constructed a new limit test method to rapidly determine the levels of sulfonamide residues. Six sulfonamides were detected using the latest method called TLC-SERS, namely, sulfamethasone (A), sulfamethazine (B), sulfadoxine (C), sulfamethoxydiazine (D), sulfamethoxazole (E), and sulfathiazole (F). The optimal conditions for SERS detection were investigated for these six drugs, and the separation effects of different TLC spreaders on them were compared. Then, we successfully established a separation system using dichloromethane–methanol–ammonia in a ratio of 5:1:0.25 (v/v/v), which provided good separation effects on the six drugs. The residues were preliminarily separated via TLC. A silver sol solution was added to the spot on the silica gel G plate at the corresponding specific shift values, and SERS detection was performed. The sample solution was placed on the spot under a 532 nm laser, and the SERS spectrum was collected and analyzed for the six sulfonamides. The results showed obvious variations in the SERS spectrum among the six sulfonamides, with the LODs being 12.5, 6.4, 6.3, 7.1, 18.8, and 6.2 ng/mL from A to F, respectively, and an RSD of

(1) Background: Although daily time-use is associated with diet quality and cardiorespiratory fitness (CRF) in children, their interdependence remains unexplored. This study first examined the associations between reallocating daily movement time and diet quality and CRF, and second the mediating role of diet quality in the relationship between daily time-use and CRF. (2) Methods: This study included 1131 Chinese children (aged 8 to 10 years; median [interquartile range]: 8.5 [8.3, 8.8]) at baseline (September 2022) and 1268 children at the 9-month follow-up (June 2023) from the OptiChild study. Daily durations of moderate-to-vigorous physical activity (MVPA), sleep, and sedentary behavior (e.g., screen time) were self-reported or proxy-reported by parents. Diet quality was assessed via the Diet Quality Questionnaire (DQQ), which uses a 24 h dietary recall and is categorized according to the Global Dietary Recommendations (GDR) score and Food Group Diversity Score (FGDS). The CRF was measured using VO2max after the 20 m shuttle run test. Longitudinal associations between daily time-use, diet quality, and CRF were calculated using isotemporal substitution models. Mediation analyses were used to determine whether diet quality mediated the associations between daily time-use and CRF. (3) Results: Reallocation of 30 min from screen time to MVPA resulted in significant improvements in the GDR score (β baseline = 0.11, p = 0.024; β follow-up = 0.26, p < 0.001), FGDS (β baseline = 0.11, p = 0.006; β follow-up = 0.19, p < 0.001), and CRF (β baseline = 0.40, p < 0.001; β follow-up = 0.26, p = 0.001). Diet quality partially mediated the associations between MVPA, screen time, and CRF. Substituting 30 min of screen time for MVPA led to diet quality mediating a proportion of the association with CRF (GDR score: 11.4%, FGDS: 6.6%). (4) Conclusions: These findings underscore the importance of optimizing daily time-use of MVPA and screen time and improving diet quality to promote physical fitness in school-aged children.

There are many high dam hubs in the world, and the regular inspection of high dams is a critical task for ensuring their safe operation. Traditional manual inspection methods pose challenges related to the complexity of the on-site environment, the heavy inspection workload, and the difficulty in manually observing inspection points, which often result in low efficiency and errors related to the influence of subjective factors. Therefore, the introduction of intelligent inspection technology in this context is urgently necessary. With the development of UAVs, computer vision, artificial intelligence, and other technologies, the intelligent inspection of high dams based on visual perception has become possible, and related research has received extensive attention. This article summarizes the contents of high dam safety inspections and reviews recent studies on visual perception techniques in the context of intelligent inspections. First, this article categorizes image enhancement methods into those based on histogram equalization, Retinex, and deep learning. Representative methods and their characteristics are elaborated for each category, and the associated development trends are analyzed. Second, this article systematically enumerates the principal achievements of defect and obstacle perception methods, focusing on those based on traditional image processing and machine learning approaches, and outlines the main techniques and characteristics. Additionally, this article analyzes the principal methods for damage quantification based on visual perception. Finally, the major issues related to applying visual perception techniques for the intelligent safety inspection of high dams are summarized and future research directions are proposed.

(1) Background: Practice guidelines recommend neoadjuvant treatment for clinical T4 rectal cancer. The primary objective of this retrospective study was to assess whether compliance with guidelines correlates with patient outcomes. Secondarily, we evaluated predictors of adherence to guidelines and mortality. (2) Methods: A total of 397 qualified rectal cancer (RC) patients from 2017 to 2020 at West China Hospital of Sichuan University were included. Patients were divided into two groups depending on adherence to neoadjuvant treatment guidelines. The main endpoints were overall survival (OS) and disease special survival (DSS). We analyzed factors associated with guideline adherence and mortality. (3) Results: Compliance with guidelines was only 39.55%. Patients’ neoadjuvant therapy treated not according to the guidelines for clinical T4 RC was not associated with an overall survival (95.7% vs. 88.9%) and disease special survival (96.3% vs. 91.1%) benefit. Patients were more likely to get recommended therapy with positive patient compliance. Staging Ⅲ, medium/high differentiation and objective compliance were associated with increased risk of mortality. (4) Conclusions: Guideline adherence for clinical T4 RC in our system is low. Compliance with the relevant guidelines for neoadjuvant therapy seems not to lead to better overall survival for patients with clinical T4 RC.

The pricing of ride-hailing platforms (e.g., Didi Rider and Uber) is heavily and simultaneously influenced by the cross-group network effect and congestion effect. To analyze the bilateral pricing of ride-hailing platforms under the influence of these two effects, in this paper we construct a game-theoretic model under four different scenarios and analyze the equilibrium outcomes. The results show that: (1) when both passengers and drivers are sensitive to hassle costs, if the cross-group network effect on the passenger side is higher than that on the driver side, then the platform’s pricing on both sides increases with the increase in the congestion effect, otherwise the prices on both sides of the platform decrease with the increase in the congestion effect; (2) when passengers are sensitive to hassle costs and drivers are sensitive to price, if the ratio for passengers’ and drivers’ different perceptions of price and hassle cost is greater than a certain threshold, then the platform’s pricing on the passenger side increases with the increase in the congestion effect and the platform’s pricing on the driver side decreases with the increase in the congestion effect, otherwise the platform’s pricing on the passenger side decreases with the increase in the congestion effect and the platform’s pricing on the drivers’ side increases with the increase in the congestion effect; (3) when passengers are sensitive to price and drivers are sensitive to hassle costs, if the ratio for passengers’ and drivers’ different perceptions of price and hassle costs is greater than a certain threshold, then the platform’s pricing on the passenger side decreases with the increase in the congestion effect and the platform’s pricing on the drivers’ side increases with the increase in the congestion effect, otherwise the platform’s pricing on the passenger side increases with the increase of the congestion effect and the platform’s pricing on the driver side decreases with the increase in the congestion effect; (4) when both passengers and drivers are price-sensitive, if the cross-group network effect on the passengers’ side is larger than that on the drivers’ side, then the platform should decrease its pricing on both sides with the increase in the congestion effect, otherwise, if the cross-group network effect on the passengers’ side is less than that on the drivers’ side, the platform should increase its pricing on both sides with the increase in the congestion effect; (5) the platform is able to generate the highest profit in each scenario, and the results of the profit comparison between the four scenarios depends on the cross-group network effects and the congestion effects on both the passengers’ and the drivers’ sides.

Copper-based catalysts are widely used in methanol steam reforming to produce hydrogen. In this paper, the supportive effect of the crystal phase of ZrO2 on Cu-based catalysts in methanol steam reforming is discussed. Monoclinic(m-), Tetragonal(t-) and mixed ZrO2 phases were prepared, and structure–activity relationships were investigated with XRD, H2-TPR, BET, HR-TEM and XPS. It was found that the catalyst with a 81.4% monoclinic ZrO2 crystal phase exhibited the highest methanol conversion (88.5%) and the highest hydrogen production rate (104.6 μmol/gcat·s) at 275 °C as it displayed the best reducing properties and more oxygen vacancies on the catalyst surface. Oxygen vacancies can produce more Cu1+ + Cu0, which is the active species for methanol steam reforming on the catalyst surface, and therefore affect catalytic activity.

The discrete element method (DEM) is largely used to simulate the geotechnical behavior of granular materials. However, numerical modeling with this type of code is expensive and time consuming, especially when fine particles are involved. This leads researchers to make use of different approaches to shorten the time of calculation without verifying the stability and reliability of numerical results, even though a compromise between the time of calculation and accuracy is commonly claimed. The particle size distribution (PSD) curve of studied granular material is completely ignored or arbitrarily cut. It is unclear if the ensued numerical results are still representative of the studied granular materials. Additionally, one can see a large number of numerical models established on a basis of calibration by ignoring the physical meaning and even measured values of some model parameters. The representativeness and reliability of the obtained numerical results are questionable. All these partly contribute to reducing the public’s confidence in numerical modeling. In this study, a methodology is illustrated to obtain an optimal DEM model, which minimizes the time of calculation and ensures stable and reliable numerical results for the mechanical behavior of a waste rock. The results indicate that the PSD curve of the studied waste rock can indeed be cut by excluding a portion of fine particles, while the Young’s modulus of the waste rock particles can also be decreased to accelerate the numerical calculations. A physical explanation of why the time of calculation can be shortened by reducing the Young’s modulus of waste rock particles is provided for the first time. Overall, the PSD cut, reduction in Young’s modulus, and time step must be determined through sensitivity analyses to ensure stable and reliable results with the shortest time of calculation. In addition, it is important to minimize the number of model parameters determined through the process of calibration, especially for those having physical meanings. In this study, the only model parameter having a clear physical meaning but difficult to measure is the rolling resistance coefficient for repose angle tests on the studied waste rock. Its value has to be obtained through a process of calibration against some experimental results. The validity and predictability of the calibrated numerical model have been successfully verified against additional experimental results.

Domestic animals have multiple phenotypes of skin and coat color, which arise from different genes and their products, such as proteins and metabolites responsible with melanin deposition. However, the complex regulatory network of melanin synthesis remains to be fully unraveled. Here, the skin and tongue tissues of Liangshan black sheep (black group) and Liangshan semi-fine-wool sheep (pink group) were collected, stained with hematoxylin–eosin (HE) and Masson–Fontana, and the transcriptomic and metabolomic data were further analyzed. We found a large deposit of melanin granules in the epidermis of the black skin and tongue. Transcriptome and metabolome analysis identified 744 differentially expressed genes (DEGs) and 443 differentially expressed metabolites (DEMs) between the pink and black groups. Gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) enrichment analyses revealed the DEGs and DEMs were mainly enriched in the pathways of secondary metabolic processes, melanin biosynthesis processes, melanin metabolism processes, melanosome membranes, pigment granule membranes, melanosome, tyrosine metabolism, and melanogenesis. Notably, we revealed the gene ENSARG00020006042 may be a family member of YWHAs and involved in regulating melanin deposition. Furthermore, several essential genes (TYR, TYRP1, DCT, PMEL, MLANA, SLC45A2) were significantly associated with metabolite prostaglandins and compounds involved in sheep pigmentation. These findings provide new evidence of the strong correlation between prostaglandins and related compounds and key genes that regulate sheep melanin synthesis, furthering our understanding of the regulatory mechanisms and molecular breeding of pigmentation in sheep.

Locally Resonant Acoustic Metamaterials (LRAMs) have significant application potential because they can form subwavelength band gaps. However, most current research does not involve obtaining LRAMs with specified band gaps, even though such LRAMs are significant for practical applications. To address this, we propose a parameterized level-set-based topology optimization method that can use multiple materials to design LRAMs that meet specified frequency constraints. In this method, a simplified band-gap calculation approach based on the homogenization framework is introduced, establishing a restricted subsystem and an unrestricted subsystem to determine band gaps without relying on the Brillouin zone. These subsystems are specifically tailored to model the phenomena involved in band gaps in LRAMs, facilitating the opening of band gaps during optimization. In the multi-material representation model used in this method, each material, except for the matrix material, is depicted using a similar combinatorial formulation of level-set functions. This model reduces direct conversion between materials other than the matrix material, thereby enhancing the band-gap optimization of LRAMs. Two problems are investigated to test the method’s ability to use multiple materials to solve band-gap optimization problems with specified frequency constraints. The first involves maximizing the band-gap width while ensuring it encompasses a specified frequency range, and the second focuses on obtaining light LRAMs with a specified band gap. LRAMs with specified band gaps obtained in three-material or four-material numerical examples demonstrate the effectiveness of the proposed method. The method shows great promise for designing metamaterials to attenuate specified frequency spectra as required, such as mechanical vibrations or environmental noise.

A novel axially chiral all-hydrocarbon cyclo[7] (1,3-(4,6-dimethyl)benzene (CDMB-7) was designed and synthesized using atroposelective[2 + 5] cyclization through Suzuki–Miyaura coupling. CDMB-7 adopts an irregular bowl-like shape with C2 symmetry and exhibits two diastereoisomers in its crystallographic structure. The conformational isomers of CDMB-7 racemates remain stable at high temperatures (393 K). High-performance liquid chromatography (HPLC) confirmed that a single chiral isomer will spontaneously undergo racemization within 30 min at room temperature. This finding opens up possibilities for achieving adaptive chirality in all-hydrocarbon cyclo[7] m-benzene macrocycles.

Ti/IrO2-Ta2O5 electrodes are extensively utilized in the electrochemical industries such as copper foil production, cathodic protection, and wastewater treatment. However, their performance degrades rapidly under high current densities and severe oxygen evolution conditions. To address this issue, we have developed a composite anode of Ti/Ta-Ti/IrO2-Ta2O5 with a Ta-Ti alloy interlayer deposited on a Ti substrate by double-glow plasma surface alloying, and the IrO2-Ta2O5 surface coating prepared by the traditional thermal decomposition method. This investigation indicates that the electrode with Ta-Ti alloy interlayer reduces the agglomerates of precipitated IrO2 nanoparticles and refines the grain size of IrO2, thereby increasing the number of active sites and enhancing the electrocatalytic activity. Accelerated lifetime tests demonstrate that the Ti/Ta-Ti/IrO2-Ta2O5 electrode exhibits a much higher stability than the Ti/IrO2-Ta2O5 electrode. The significant improvement in electrochemical stability is attributed to the Ta-Ti interlayer, which offers high corrosion resistance and effective protection for the titanium substrate.

Neuroblastoma (NB) is a cancer of the peripheral nervous system found in children under 15 years of age. It is the most frequently diagnosed cancer during infancy, accounting for ~12% of all cancer-related deaths in children. Leucine-rich repeat-containing G-protein-coupled receptor 5 (LGR5) is a membrane receptor that is associated with the primary tumor formation and metastasis of cancers in the gastrointestinal system. Remarkably, high levels of LGR5 are found in NB tumor cells, and high LGR5 expression is strongly correlated with poor survival. Antibody–drug conjugates (ADCs) are monoclonal antibodies that are covalently linked to cell-killing cytotoxins to deliver the payloads into cancer cells. We generated an ADC with an anti-LGR5 antibody and pyrrolobenzodiazepine (PBD) dimer-based payload SG3199 using a chemoenzymatic conjugation method. The resulting anti-LGR5 ADC was able to inhibit the growth of NB cells expressing LGR5 with high potency and specificity. Importantly, the ADC was able to completely inhibit the growth of NB xenograft tumors in vivo at a clinically relevant dose for the PBD class of ADCs. The findings support the potential of targeting LGR5 using the PBD class of payload for the treatment of high-risk NBs.

The gastrointestinal (GI) tract is an organ actively involved in mechanical processes, where it detects forces via a mechanosensation mechanism. Mechanosensation relies on specialized cells termed mechanoreceptors, which convert mechanical forces into electrochemical signals via mechanosensors. The mechanosensitive Piezo1 and Piezo2 are widely expressed in various mechanosensitive cells that respond to GI mechanical forces by altering transmembrane ionic currents, such as epithelial cells, enterochromaffin cells, and intrinsic and extrinsic enteric neurons. This review highlights recent research advances on mechanosensitive Piezo channels in GI physiology and pathology. Specifically, the latest insights on the role of Piezo channels in the intestinal barrier, GI motility, and intestinal mechanosensation are summarized. Additionally, an overview of Piezo channels in the pathogenesis of GI disorders, including irritable bowel syndrome, inflammatory bowel disease, and GI cancers, is provided. Overall, the presence of mechanosensitive Piezo channels offers a promising new perspective for the treatment of various GI disorders.

To mitigate the control overhead of Software-Defined Mobile Ad Hoc Networks (SD-MANETs), this paper proposes a novel approach, termed Software-Defined Greedy Perimeter Stateless Routing (SD-GPSR), which integrates geographical location information. SD-GPSR optimizes routing functions by decentralizing them within the data plane of SD-MANET, utilizing the geographic location information of nodes to enhance routing efficiency. The controller is primarily responsible for providing location services and facilitating partial centralized decision-making. Within the data plane, nodes employ an enhanced distance and angle-based greedy forwarding algorithm, denoted as GPSR_DA, to efficiently forward data. Additionally, to address the issue of routing voids in the data plane, we employ the A* algorithm to compute an optimal routing path that circumvents such voids. Finally, we conducted a comparative analysis with several state-of-the-art approaches. The evaluation experiments demonstrate that SD-GPSR significantly reduces the control overhead of the network. Simultaneously, there is a notable improvement in both end-to-end latency and packet loss rate across the network.

The cross-dimensional matching of 2D images and 3D point clouds is an effective method by which to establish the spatial relationship between 2D and 3D space, which has potential applications in remote sensing and artificial intelligence (AI). In this paper, we propose a novel multi-task network, 2D3D-DescNet, to learn 2D and 3D local feature descriptors jointly and perform cross-dimensional matching of 2D image patches and 3D point cloud volumes. The 2D3D-DescNet contains two branches with which to learn 2D and 3D feature descriptors, respectively, and utilizes a shared decoder to generate the feature maps of 2D image patches and 3D point cloud volumes. Specifically, the generative adversarial network (GAN) strategy is embedded to distinguish the source of the generated feature maps, thereby facilitating the use of the learned 2D and 3D local feature descriptors for cross-dimensional retrieval. Meanwhile, a metric network is embedded to compute the similarity between the learned 2D and 3D local feature descriptors. Finally, we construct a 2D-3D consistent loss function to optimize the 2D3D-DescNet. In this paper, the cross-dimensional matching of 2D images and 3D point clouds is explored with the small object of the 3Dmatch dataset. Experimental results demonstrate that the 2D and 3D local feature descriptors jointly learned by 2D3D-DescNet are similar. In addition, in terms of 2D and 3D cross-dimensional retrieval and matching between 2D image patches and 3D point cloud volumes, the proposed 2D3D-DescNet significantly outperforms the current state-of-the-art approaches based on jointly learning 2D and 3D feature descriptors; the cross-dimensional retrieval at TOP1 on the 3DMatch dataset is improved by over 12%.

Accurate target identification of UAV (Unmanned Aerial Vehicle)-captured images is a prerequisite for maritime rescue and maritime surveillance. However, UAV-captured images pose several challenges, such as complex maritime backgrounds, tiny targets, and crowded scenes. To reduce the impact of these challenges on target recognition, we propose an efficient maritime rescue network (EMR-YOLO) for recognizing images captured by UAVs. In the proposed network, the DRC2f (Dilated Reparam-based Channel-to-Pixel) module is first designed by the Dilated Reparam Block to effectively increase the receptive field, reduce the number of parameters, and improve feature extraction capability. Then, the ADOWN downsampling module is used to mitigate fine-grained information loss, thereby improving the efficiency and performance of the model. Finally, CASPPF (Coordinate Attention-based Spatial Pyramid Pooling Fast) is designed by fusing CA (Coordinate Attention) and SPPF (Spatial Pyramid Pooling Fast), which effectively enhances the feature representation and spatial information integration ability, making the model more accurate and robust when dealing with complex scenes. Experimental results on the AFO dataset show that, compared with the YOLOv8s network, the EMR-YOLO network improves the mAP (mean average precision) and mAP50 by 4.7% and 9.2%, respectively, while reducing the number of parameters and computation by 22.5% and 18.7%, respectively. Overall, the use of UAVs to capture images and deep learning for maritime target recognition for maritime rescue and surveillance improves rescue efficiency and safety.

Smart highways represent a novel highway concept in the era of big data, emphasizing the synergy among people, vehicles, road facilities, and the environment. However, the operation and management of smart highways have become more intricate, surpassing the adaptability of traditional highway evaluation and management methods. This study integrates the distinctive characteristics of smart highway facilities and operational objectives to enhance and modernize the existing highway evaluation system. Drawing from research on smart highway construction projects, a smart highway evaluation system encompassing facility structure, electromechanical facilities, and operation services is formulated based on a hierarchical analysis method. The quantitative evaluation of each indicator is achieved by combining existing specifications and expert questionnaire solicitation. The group decision-making method is initially employed to optimize subjective weights, followed by the calculation of combined weights using both the entropy weight method and critic method in objective evaluation. Finally, a comprehensive evaluation model is established and validated through engineering projects. The results demonstrate that the evaluation system effectively highlights the advantages and disadvantages in the operation and management of smart highways, thereby fostering the advancement of smart highway iteration.

Hepatocellular carcinoma is a refractory tumor with poor prognosis and high mortality. Many oncolytic viruses are currently being investigated for the treatment of hepatocellular carcinoma. Based on previous studies, we constructed a recombinant GM-CSF-carrying Sindbis virus, named SINV-GM-CSF, which contains a mutation (G to S) at amino acid 285 in the nsp1 protein of the viral vector. The potential of this mutated vector for liver cancer therapy was verified at the cellular level and in vivo, respectively, and the changes in the tumor microenvironment after treatment were also described. The results showed that the Sindbis virus could effectively infect hepatocellular carcinoma cell lines and induce cell death. Furthermore, the addition of GM-CSF enhanced the tumor-killing effect of the Sindbis virus and increased the number of immune cells in the intra-tumor microenvironment during the treatment. In particular, SINV-GM-CSF was able to efficiently kill tumors in a mouse tumor model of hepatocellular carcinoma by regulating the elevation of M1-type macrophages (which have a tumor-resistant ability) and the decrease in M2-type macrophages (which have a tumor-promoting capacity). Overall, SINV-GM-CSF is an attractive vector platform with clinical potential for use as a safe and effective oncolytic virus.

To learn more about the nutritional composition and health benefits for human consumers of peas, we used a widely targeted metabolomics-based approach to reveal the metabolite components from three main varieties, and a total of 1095 metabolites were identified. A comparison of 487 differentially accumulated metabolites shared among three varieties of fresh and dried peas found most of the amino acids and derivatives were downregulated and most of the lipids and flavonoids were upregulated in dried peas. Furthermore, comparing the main nutrient profiles exclusively showed that there were few differences in free fatty acids, sugars, vitamins, and alkaloids between dried and fresh peas. Peas are especially enriched with B-group vitamins. Through detailed identification and classification, the flavonoid pathway of peas was revealed; a variety of glycosylated derivatives from kaempferol, quercetin, and luteolin were confirmed to be abundant in peas. It was also found that isoflavones are richer in peas than in many other plants, and putatively the isoflavone synthesis pathway originates from liquiritigenin and naringenin. Our study not only offers guidance for understanding the nutritional components of peas, but also provides the basis for healthy diet analysis of the edible value and health benefits of peas.

Quinoa is a nutritious crop that is tolerant to extreme environmental conditions; however, low-temperature stress can affect quinoa growth, development, and quality. Considering the lack of molecular research on quinoa seedlings under low-temperature stress, we utilized a Weighted Gene Co-Expression Network Analysis to construct weighted gene co-expression networks associated with physiological indices and metabolites related to low-temperature stress resistance based on transcriptomic data. We screened 11 co-expression modules closely related to low-temperature stress resistance and selected 12 core genes from the two modules that showed the highest associations with the target traits. Following the functional annotation of these genes to determine the key biological processes and metabolic pathways involved in low-temperature stress, we identified four important transcription factors involved in resistance to low-temperature stress: gene-LOC110731664, gene-LOC110736639, gene-LOC110684437, and gene-LOC110720903. These results provide insights into the molecular genetic mechanism of quinoa under low-temperature stress and can be used to breed lines with tolerance to low-temperature stress.

Loess is widely distributed in the northwest and other regions, and its unique structural forms such as large pores and strong water sensitivity lead to its collapsibility and collapse, which can easily induce slope instability. Guar gum and basalt fiber are natural green materials. For these reasons, this study investigated the solidification of loess by combining guar gum and basalt fiber and analyzed the impact of the guar gum content, fiber length, and fiber content on the soil shearing strength. Using scanning electron microscopy (SEM), the microstructure of loess was examined, revealing the synergistic solidification mechanism of guar gum and basalt fibers. On this basis, a shear strength model was established through regression analysis with fiber length, guar gum content, and fiber content. The results indicate that adding guar gum and basalt fiber increases soil cohesion, as do fiber length, guar gum content, and fiber content. When the fiber length was 12 mm, the fiber content was 1.00%, and the guar gum content was equal to 0.50%, 0.75%, or 1.00%, the peak strength of the solidified loess increased by 82.80%, 85.90%, and 90.40%, respectively. According to the shear strength model, the predicted and test data of the shear strength of solidified loess are evenly distributed on both sides of parallel lines, indicating a good fit. These findings are theoretically significant and provide practical guidance for loess solidification engineering.

Loess has the characteristics of loose, large pore ratio, and strong water sensitivity. Once it encounters water, its structure is damaged easily and its strength is degraded, causing a degree of subgrade settlement. The water sensitivity of loess can be evaluated by permeability and disintegration tests. This study analyzes the effects of guar gum content, basalt fiber content, and basalt fiber length on the permeability and disintegration characteristics of solidified loess. The microstructure of loess was studied through scanning electron microscopy (SEM) testing, revealing the synergistic solidification mechanism of guar gum and basalt fibers. A permeability model was established through regression analysis with guar gum content, confining pressure, basalt fiber content, and length. The research results indicate that the addition of guar gum reduces the permeability of solidified loess, the addition of fiber improves the overall strength, and the addition of guar gum and basalt fiber improves the disintegration resistance. When the guar gum content is 1.00%, the permeability coefficient and disintegration rate of solidified soil are reduced by 50.50% and 94.10%, respectively. When the guar gum content is 1.00%, the basalt fiber length is 12 mm, and the fiber content is 1.00%, the permeability of the solidified soil decreases by 31.9%, and the disintegration rate is 4.80%. The permeability model has a good fitting effect and is suitable for predicting the permeability of loess reinforced with guar gum and basalt fiber composite. This research is of vital theoretical worth and great scientific significance for guidelines on practicing loess solidification engineering.

Plant proline-rich proteins (PRPs) are cell wall proteins that are widely distributed in plants. Previous studies have shown that these proteins play a crucial role in adversity stress processes, but their function in the regulation of pollen fertility in rice remains unknown. In this study, we identified that OsPRP1 contains a Pollen_Ole-e_I allergenic structural domain, obtained the OsPRP1 quadruple mutant named osprp1.1/1.2/1.3/1.4, and observed significant reductions in pollen fertility, seed-setting rates, and the deformation and collapse of microspores during the late stages of pollen development. RNA-Seq analysis indicated the down-regulation of genes involved in anther development in osprp1.1/1.2/1.3/1.4, suggesting that OsPRP1 plays a role in regulating pollen fertility. In conclusion, a loss of function in OsPRP1.1/1.2/1.3/1.4 leads to decreased pollen fertility and seeding rates, which not only expands the functional spectrum of plant PRP genes but also provides new theoretical insight into the mechanism of fertility regulation in rice.
