Panax ginseng, a perennial medicinal plant, utilizes its dried roots and rhizomes for medicinal purposes. Currently, in China, ginseng cultivation employs two methods: under-forest and farmland planting. These methods create distinct habitats, significantly influencing the ginseng’s rhizome morphology and, consequently, its economic value. In this study, two-year-old ginsengs were transplanted into farmland (TCG), a larch forest (TLCG) and a Quercus mongolica forest (TQCG) to analyze the differences in rhizome phenotypes caused by habitat changes. The results showed that there were significant differences in light intensity and the soil’s available phosphorus and potassium contents between farmland and forest environments. The differences in habitats led to different adaptability of the ginseng’s rhizome morphology. Compared with TCG, the rhizomes of TLCG and TQCG were significantly elongated by 48.36% and 67.34%, respectively. After the rhizomes’ elongation in TLCG and TQCG, there was an increase in indole-3-acetic acid (IAA) contents and a decrease in lignin contents. By analyzing the expression of key genes, we found that, compared with TCG, the expression of key enzymes of lignin biosynthesis genes such as PgCOMT and PgCCR4 were down-regulated. The difference in ginseng’s rhizome length is related to the signal transduction pathway of auxin and gibberellin. In addition, we preliminarily screened out transcription factors PgWRKY75, PgDIV, and PgbHLH93.1, which can actively respond to habitat changes and play important roles in the elongation of ginseng rhizomes. In summary, this study elucidates the phenotypic plasticity strategy of ginseng rhizomes in response to habitat changes and delineates the regulatory mechanism behind phenotypic adaptation, offering novel insights into ginseng’s morphogenesis.

Dihydromyricetin (DMY), as the main active ingredient in Ampelopsis grossedentata, is a naturally occurring flavonoid that has attracted extensive attention for its multiple biological activities. For the quick and accurate measurement of DMY, a novel electrochemical sensor based on a glassy carbon electrode (GCE) modified with a cobalt metal-organic framework (Co-MOF) was proposed in this work. The Co-MOF was synthesized via a single-step hydrothermal process using Co(NO3)2·6H2O. Fourier infrared spectroscopy, X-ray photoelectron spectroscopy and scanning electron microscopy were used to study the morphology and structure of the synthesized Co-MOF. Utilizing differential pulse voltammetry and cyclic voltammetry methods, the effectiveness of DMY electro-oxidation on the Co-MOF/GCE was examined. The results showed that, in comparison to the bare GCE, the electro-oxidation peak current of DMY was considerably increased by the Co-MOF/GCE. The detection limit was 0.07 μM, and the peak current demonstrated two linear relationships in the ranges of 0.2−20 μM and 20−100 μM, with the linear equations of Ip (μA) = 0.4729c (μM) + 1.0822 (R2 = 0.9913) and Ip (μA) = 0.0939c (μM) + 8.4178 (R2 = 0.9971), respectively. The average DMY content in Ampelopsis grossedentata samples was measured to be 3.275 μM, with a good recovery of 108.27% and a relative standard deviation value of 3.46%. The proposed method is simple, rapid and sensitive and can be used for the determination of DMY in Ampelopsis grossedentata.

Reducing greenhouse gas (GHG) emissions and ammonia (NH3) volatilization by improving fertilization methods to increase crop yield is beneficial for the green and sustainable development of agriculture. This study evaluated the effects of farmer practice fertilization (FP), nutrient expert optimized fertilization (NE—optimized fertilizer usage and time), the application of stable compound fertilizer (SF), and the application of controlled-release coated urea (CRU) on greenhouse gases, NH3 volatilization, and corn yield through field experiments set up in the corn planting area in western Liaoning Province. The results showed that compared with FP treatment, NE could significantly reduce NH3 volatilization by 28% and increase N2O release by 41%. Compared with FP treatment, SF could significantly reduce NH3 volatilization by 48.54%, N2O release by 38.54%, CO2 release by 13.96%, global warming potential (GWP) by 16.60%, and greenhouse gas emission intensity (GHGI) by 27.23%, and could significantly increase corn yield by 15.86%. Compared with FP treatment, CRU could significantly reduce NH3 volatilization by 63.46%, CO2 release by 11.98%, GWP by 10.73%, and GHGI by 13.77%, while increasing N2O release by 6.71%. Overall, NE, SF, and CRU treatments all showed better effects than FP treatment in increasing corn yield or reducing NH3 volatilization and GHG emissions. Among them, SF treatment demonstrated superior performance over NE and CRU treatments in terms of NH3 volatilization, corn yield, and GHGI. Therefore, the application of stable compound fertilizer is the optimal choice for corn planting in western Liaoning, with broad application prospects.

Drought stress is one of the main factors affecting the growth and secondary metabolism of plants. Iris domestica, rich in isoflavones, is a common herbal medicine in China. In this study, the effects of drought stress and rehydration on resistance physiological characteristics and the secondary metabolism of two-year-old I. domestica during the vegetative and reproductive growth period were investigated. The results showed that the dry weight and fresh weight of rhizomes and roots under severe drought stress were significantly decreased, while those under moderate drought stress were not significantly changed. Meanwhile, the SOD activities, POD activities, MDA content and Pro content increased to resist drought at D1 and D2. In the vegetative growth period, the changes in isoflavone concentration and the expression levels of genes in isoflavone synthesis were more dramatic. Isoflavone accumulation was promoted, to some extent, in the reproductive growth period under the D1 drought treatment. In the actual production process, different measures, namely short-term stress regulation in the vegetative growth period and moderate drought stress (13.44% < soil water content ≤ 16.8%) in the reproductive growth period, need to be adopted to regulate isoflavone biosynthesis.

Platycodon grandiflorum, a dual-purpose herb for food and medicine, is widely distributed in Asia. Although P. grandiflorum has relatively low requirements for its growing environment, temperature remains an important ecological factor affecting its growth, development, and quality formation. In order to explore the effect of different temperatures on P. grandiflorum during their growth period, the diversity in growth physiology, platycodin contents, and gene expression of key enzymes were investigated under constant (8 °C, 18 °C, and 28 °C) and variable (8–18 °C, 8–28 °C, and 18–28 °C) temperature conditions at each of the three levels. The results suggested that both constant and variable temperatures at high levels significantly increased the aboveground fresh weight of P. grandiflorum. However, the low–variable temperature was beneficial for the accumulation of dry and fresh weight in the roots. Regardless of whether temperatures were constant or variable at low levels, this increased the content of soluble sugars, proline, and peroxidase in P. grandiflorum, while upregulating the expression levels of key enzyme genes involved in platycodin synthesis. Meanwhile, a low–constant temperature inhibited the photosynthetic rate of P. grandiflorum. Furthermore, medium–constant and large-scale variable temperatures were conducive to the accumulation of platycodins in the roots. This research provides a theoretical basis and data support for the influence of temperature variations on P. grandiflorum quality formation.

Monitoring air pollution is important for human health and the environment. Previous studies on the prediction of air pollutants from satellite images have employed machine learning, yet there are few enhancements to the constructure of model. Moreover, the existing models have been successful in predicting pollutants like PM2.5, PM10, and O3. They have not been as effective in predicting other primary air pollutants. To improve the overall prediction performance of the existing model, a novel residual learning backpropagation model, abs. as BresNet, has been proposed in this research. This model has revealed the availability to precisely predict the ground-surface concentration of the six primary air pollutants, PM2.5, PM10, O3, NO2, CO, and SO2, based on the satellite imagery of MODIS AOD. Two of the most commonly used machine learning models so far, viz. the multilayer backpropagation neural network (MLBPN) and random forest (RF), were employed as the control. In the conducted experiments, the proposed BresNet model demonstrated significant improvements of 18.75%/31.94%, 33.82%/85.71%, 15.00%/35.29%, 39.06%/134.21%, 23.23%/68.00%, and 137.14%/260.87% in terms of R2 for the six primary air pollutants, compared to the RF/MLBPN model. Moreover, the convergence speed and loss function of the BresNet model compared to that of the MLBPN decreased by 55.15%, revealing superior convergence speed with the lower loss function.

Panax ginseng taproot serves as a crucial storage organ and constitutes a significant component of Chinese herbal medicine. In China, ginseng is cultivated using two primary methods: under-forest planting and farmland planting. These methods create distinct habitats that notably influence the morphology of ginseng taproots. However, the precise regulatory mechanisms governing ginseng taproot expansion remain to be fully elucidated. This study aimed to delineate the patterns of ginseng taproot expansion by examining taproots transplanted into farmland (TCG) and forest environments (TLCG and TQCG). Our findings indicate that light intensity and soil available potassium levels in TCG plots significantly exceed those in TLCG and TQCG plots. Compared with TLCG and TQCG, the taproot diameter of TCG increased by 11.54% and 27.73%, respectively. At the same time, combined with the microstructure of the transverse section of the taproot, it showed that the expansion of the TCG taproot was closely related to cell expansion. During TCG taproot expansion, there was an increase in indole-3-acetic acid (IAA) content, significant starch accumulation, and a decrease in lignin content. By analyzing the expression of key genes, we found that compared with TLCG and TQCG, the expression of genes PgTPS1 and PgALDH1 was upregulated, and the expression of genes PgHCT1, PgPAL3, PgPER3, and PgPER51 were downregulated in TCG taproot. Additionally, the transcription factors PgARF18.1 and PgbHLH42 were identified as responsive to habitat changes, playing pivotal roles in taproot expansion. In conclusion, this study provides foundational insights into the regulatory mechanisms of ginseng taproot expansion, offering significant implications for enhancing the quality and value of ginseng.

Olanzapine is an atypical antipsychotic drug and a potent muscarinic M3 receptor (M3R) antagonist. Olanzapine has been reported to cause metabolic disorders, including dyslipidemia. Anaplastic lymphoma kinase (Alk), a tyrosine kinase receptor well known in the pathogenesis of cancer, has been recently identified as a key gene in the regulation of thinness via the regulation of adipose tissue lipolysis. This project aimed to investigate whether Olanzapine could modulate the hepatic Alk pathway and lipid metabolism via M3R. Female rats were treated with Olanzapine and/or Cevimeline (an M3R agonist) for 9 weeks. Lipid metabolism and hepatic Alk signaling were analyzed. Nine weeks’ treatment of Olanzapine caused metabolic disturbance including increased body mass index (BMI), fat mass accumulation, and abnormal lipid metabolism. Olanzapine treatment also led to an upregulation of Chrm3, Alk, and its regulator Ptprz1, and a downregulation of Lmo4, a transcriptional repressor of Alk in the liver. Moreover, there were positive correlations between Alk and Chrm3, Alk and Ptprz1, and a negative correlation between Alk and Lmo4. However, cotreatment with Cevimeline significantly reversed the lipid metabolic disturbance and adipose tissue accumulation, as well as the upregulation of the hepatic Alk signaling caused by Olanzapine. This study demonstrates evidence that Olanzapine may cause metabolic disturbance by modulating hepatic Alk signaling via M3R, which provides novel insight for modulating the hepatic Alk signaling and potential interventions for targeting metabolic disorders.

Camellia seed oil (CSO), as a nutrient-rich edible oil, is widely used in foods, cosmetics, and other fields. In this work, the extraction, deacidification, decolorization, and deodorization processes of CSO were respectively optimized for meeting injectable oil standards. The results showed that the CSO extraction rate reached the highest level of 94% at optimized conditions (ultrasonic time, 31.2 min; reaction pH, 9.2; and reaction time, 3.5 h). The physicochemical indexes of CSO and 10 other vegetable oils were evaluated by the principal component analysis method, and the overall scores of vegetable oils were ranked as camellia seed oil > olive oil > rice oil > peanut oil > sesame oil > corn oil > soybean oil > sunflower oil > rapeseed oil > walnut oil > flaxseed oil. The physicochemical indicators of CSO were the most ideal among the 11 vegetable oils, which means that CSO is suitable as an injectable oil. Through the optimized processes of the deacidification, decolorization, and deodorization, the CSO acid value was reduced to 0.0515 mg KOH/g, the decolorization rate reached a maximum of 93.86%, and the OD430 was 0.015, meeting the requirement (≤0.045 of OD430) of injectable oil. After the deodorization process, these parameters of the refractive index, acid value, saponification value, iodine value, absorbance, unsaponifiable, moisture and volatiles, fatty acid composition, and heavy metal limits all met the pharmacopoeia standards of injectable oil in many countries and regions. The possibility of CSO as an injectable oil was first verified through refining-process optimization and nutritional index analysis, providing an important technical reference for the high-value utilization of vegetable oil.

Extensive industrial activities and anthropogenic agricultural practices have led to substantial ammonia release to the environment. Although croplands can act as ammonia sinks, reduced crop production under high concentrations of ammonium has been documented. Alpha-ketoglutarate (AKG) is a critical carbon source, displaying pleiotropic physiological functions. The objective of the present study is to disclose the potential of AKG to enhance ammonium assimilation in poplars. It showed that AKG application substantially boosted the height, biomass, and photosynthesis activity of poplars exposed to excessive ammonium. AKG also enhanced the activities of key enzymes involved in nitrogen assimilation: glutamine synthetase (GS) and glutamate synthase (GOGAT), elevating the content of amino acids, sucrose, and the tricarboxylic acid cycle (TCA) metabolites. Furthermore, AKG positively modulated key genes tied to glucose metabolism and ATP synthesis, while suppressing ATP-depleting genes. Correspondingly, both H+-ATPase activity and ATP content increased. These findings demonstrate that exogenously applying AKG improves poplar growth under a high level of ammonium treatment. AKG might function through sufficient carbon investment, which enhances the carbon–nitrogen balance and energy stability in poplars, promoting ammonium assimilation at high doses of ammonium. Our study provides novel insight into AKG’s role in improving poplar growth in response to excess ammonia exposure.

Sheep testes undergo a dramatic rate of development with structural changes during pre-sexual maturity, including the proliferation and maturation of somatic niche cells and the initiation of spermatogenesis. To explore this complex process, 12,843 testicular cells from three males at pre-sexual maturity (three-month-old) were sequenced using the 10× Genomics ChromiumTM single-cell RNA-seq (scRNA-seq) technology. Nine testicular somatic cell types (Sertoli cells, myoid cells, monocytes, macrophages, Leydig cells, dendritic cells, endothelial cells, smooth muscle cells, and leukocytes) and an unknown cell cluster were observed. In particular, five male germ cell types (including two types of undifferentiated spermatogonia (Apale and Adark), primary spermatocytes, secondary spermatocytes, and sperm cells) were identified. Interestingly, Apale and Adark were found to be two distinct states of undifferentiated spermatogonia. Further analysis identified specific marker genes, including UCHL1, DDX4, SOHLH1, KITLG, and PCNA, in the germ cells at different states of differentiation. The study revealed significant changes in germline stem cells at pre-sexual maturation, paving the way to explore the candidate factors and pathways for the regulation of germ and somatic cells, and to provide us with opportunities for the establishment of livestock stem cell breeding programs.

One of the most significant transcription factors in plants, WRKYs, are crucial for plant growth and stress response. In this study, we analyzed the physicochemical properties, evolutionary relationships, conservation structure, and expression of the WRKY gene family in S. baicalensis. The WRKY family has highly conserved structural domains, which have been classified into three major categories, I, II, and III, based on the number of WRKY structural domains and zinc finger structural features. SbWRKYs of the same subgroup are functionally similar and essentially contain the same motif. Additionally, different drought stress situations resulted in varying levels of SbWRKYs expression, with the majority of these factors being up-regulated in moderate drought stress settings, and fewer of them were up-regulated under severe drought stress conditions. Under moderate drought stress, the expression of key enzymes increased, while under severe drought stress, the expression of key enzymes decreased. Mild drought stress resulted in a 26.42% increase in baicalin accumulation, while severe drought stress led to a 22.88% decrease. The protein interaction analysis of key enzyme genes and SbWRKYs revealed that the expression of key enzyme genes affected the expression of SbWRKYs. We screened nine SbWRKYs with a significant relationship with baicalin accumulation, and SbWRKY8 and SbWRKY16 showed the highest correlation with the baicalin content. These findings offer a theoretical framework for more research on the roles of SbWRKYs and show that SbWRKYs can respond to drought stress in S. baicalensis.

The quality and yield of ginseng are significantly affected by ecological factors, especially soil, which is a complex environment. This study compared the quality, yield, and soil differences of ginseng in a Mongolian oak forest (MOFG), birch forest (BFG), mixed wood forest (MWFG), poplar forest (PFG), farmland (FlG), and a felling forest (DfG). Based on these differences, the relationship between the three was explored, as was the most suitable forest type for developing understory ginseng cultivation. The soil bacteria diversity of DfG was similar to that of understory ginseng, and its ginsenoside content was at the same level as that of understory ginseng. The soil pH of FlG was significantly lower than that of the other five cultivation methods, and the soil bacteria were also different from them, especially those related to the nitrogen cycle. Ginsenoside Rg2 has a significant correlation with many soil factors and is sensitive to soil environmental differences. In conclusion, there were significant differences in the quality, yield, and soil environments between the different planting methods. MOFG was the most suitable for planting with excellent quality and high yield, while BFG was the worst; as such, planting in BFG should be avoided.

Tectona grandis is a traditional Dai medicine plant belonging to the Lamiaceae family, which can be used to treat malaria, inflammation, diabetes, liver disease, bronchitis, tumors, cholelithiasis, jaundice, skin disease and as an anti-helminthic. To find more novel therapeutic agents contained in this medicinal plant, the antioxidant, anti-inflammatory and anti-diabetic activities of T. grandis methanolic extract, fractions and compounds were evaluated. In this study, 26 compounds were isolated from the leaves and branches of T. grandis. Their structures were identified based on extensive spectral experiments, including NMR, ESI-MS and comparison with published spectral data. Among them, compounds 1–2, 4–6, 9–14 and 16–22 were reported for the first time for this plant. The antioxidant activity screening results showed that compounds 5, 15 and 23 had potent antioxidant capacities, with SC50 values from 0.32 to 9.92 µmol/L, 0.92 to 1.10 mmol Trolox/L and 1.02 to 1.22 mmol Trolox/L for DPPH, ABTS and FRAP, respectively. In addition, their anti-inflammatory effects were investigated by releasing TNF-α, IL-1β and IL-6 through the use of mouse monocytic macrophages (RAW 264.7). Compounds 1, 13, 18 and 23 had the effects of reducing the expression of inflammatory factors. Compounds 13 and 18 were reported for the first time for their anti-inflammatory activities. Furthermore, the methanolic extract (ME), petroleum ether extract (PEE) and EtOAc extract (EAE) of T. grandis showed significant glucose uptake activities; compounds 21 and 23 significantly promoted glucose uptake of 3T3-L1 adipocytes at 40 µM. Meanwhile, compounds 4, 5 and 7 showed significant inhibitory activities against α-glucosidase, with IC50 values of 14.16 ± 0.34 µmol/L, 19.29 ± 0.26 µmol/L and 3.04 ± 0.08 µmol/L, respectively. Compounds 4 and 5 were reported for the first time for their α-glucosidase inhibitory activities. Our investigation explored the possible therapeutic material basis of T. grandis to prevent oxidative stress and related diseases, especially inflammation and diabetes.

Light is essential for plants and plays a vital role in their growth and development. Light irradiation affects the physiological characteristics and synthesis of secondary metabolites in plants. As a semi-shade perennial plant, Panax ginseng C.A. Mey. is sensitive to changes in the light environment. Different light irradiations significantly affect the secondary metabolic processes of P. ginseng. However, few studies have investigated the changes in ginsenoside content in P. ginseng under different light irradiation conditions. In this study, 3-year-old P. ginseng was cultured under white (CK) light, blue (B) light, red (R) light, green (G) light, and natural light (NL) to explore the effects of light irradiation on the physiological characteristics and ginsenoside secondary metabolism of P. ginseng. The B and CK treatments significantly increased the photosynthetic level in P. ginseng leaves. The total saponin content under blue and red light treatments increased by 28.81% and 21.64%, respectively, compared with the CK treatment. Blue and red light improved the transcription levels of ginsenoside biosynthetic pathway genes. Blue light upregulated the expression of HMGR, SS, SE, DS, CYP716A52, and CYP716A47, and the expression of HMGR, SS, SE, DS, and CYP716A47 under red light treatment was significantly upregulated in P. ginseng roots. Principal component and correlation analyses revealed that the physiological and ecological processes of P. ginseng exhibited different responses to light irradiation. The total saponin content in the roots was positively correlated with the content of protopanaxatriol -type ginsenosides and water use efficiency in leaves. Our study indicates that light conditions can be improved by blue and red light or by blue and red film covering to facilitate the accumulation of saponin during the ecological cultivation of P. ginseng.

The red stretcher bacterium Sporidiobolus pararoseus is a high producer of carotenoids such as torularhodin, but its presence in vinegar has not been detected. Moreover, torularhodin has several biological activities, but its effect on the LPS-induced RAW 264.7 inflammatory cell model has also yet to be elucidated. In this study, S. pararoseus was identified in different vinegar samples from China by ITS sequencing. Meanwhile, one of the strains was deeply resolved by whole genome sequencing and functional annotation and named S. pararoseus ZQHL. Subsequently, the antioxidant effect of the fungal carotenoid torularhodin was investigated using in vitro DPPH, ABTS, and cellular models. Finally, LPS-induced RAW 264.7 cells were used as an inflammation model to assess torularhodin’s protective effect on inflammatory cells and to determine whether the TLR4 pathway is associated with this process. The results indicate that torularhodin has good free radical scavenging ability in vitro and can contribute to cell viability. More importantly, torularhodin alleviated LPS-induced cellular inflammatory damage and reduced the expression of inflammatory factors such as TLR4, MyD88, and TNF-a. The mechanism may attenuate the cellular inflammatory response by inhibiting the TLR4 inflammatory pathway. In conclusion, torularhodin produced by S. pararoseus fungi in vinegar samples significantly scavenged free radicals in vitro and alleviated RAW 264.7 cellular inflammation by modulating the TLR4 pathway.

Iris domestica is a popular gardening plant. Although the species is considered tolerant to drought, its growth and development are often affected by drought conditions. Therefore, revealing the regulatory mechanisms of drought tolerance in this species will aid in its cultivation and molecular breeding. In this study, morpho-physiological and transcriptome analyses of the roots of I. domestica plants were performed under persistent drought conditions. Peroxidase activity, proline content, and tectoridin content increased under sustained drought conditions. Transcriptome analysis showed that the roots of I. domestica seedlings respond to drought mainly by regulating the expression of drought-resistant genes and biosynthesis of secondary metabolites. This study provides basic data for identifying drought response mechanisms in Iris domestica.

The aim of this study was to establish a rapid detection method of rutin in food based on nitrogen-doped carbon quantum dots (N-CDs) as the fluorescent probe. N-CDs were prepared via a single-step hydrothermal process using citric acid as the carbon source and thiourea as the nitrogen source. The optical properties of N-CDs were characterized using an electron transmission microscope, X-ray diffractometer, Fourier-transform infrared spectrometer, and nanoparticle size potential analyzer. The UV/Vis absorption property and fluorescence intensity of N-CDs were also characterized using the respective spectroscopy techniques. On this basis, the optimal conditions for the detection of rutin by N-CDs fluorescent probes were also explored. The synthesized N-CDs were amorphous carbon structures with good water solubility and optical properties, and the quantum yield was 24.1%. In phosphate-buffered solution at pH = 7.0, Rutin had a strong fluorescence-quenching effect on N-CDs, and the method showed good linearity (R2 = 0.9996) when the concentration of Rutin was in the range of 0.1–400 μg/mL, with a detection limit of 0.033 μg/mL. The spiked recoveries in black buckwheat tea and wolfberry were in the range of 93.98–104.92%, the relative standard deviations (RSD) were in the range of 0.35–4.11%. The proposed method is simple, rapid, and sensitive, and it can be used for the rapid determination of rutin in food.

Saposhnikovia divaricata is a high-demand medicinal plant containing various bioactive metabolites (e.g., chromone). However, root rot disease leads to a dramatic reduction in the yield and quality of S. divaricata. The use of rhizospheric microorganisms is one of the best strategies for biological control. In this study, a total of 104 fungi isolated from the rhizospheric soil of S. divaricata plants were examined for their different antifungal properties. Subsequently, strain MR-57 was selected as a potential stock for biocontrol due to its broad-spectrum antagonistic activity against pathogens, including F. equiseti. Based on the analysis of morphological properties and rDNA internal transcribed spacers (ITSs), strain MR-57 was identified as Acrophialophora jodhpurensis (GenBank No. OK287150.1), a newly recorded species for China. In an in vitro antifungal assay, the culture filtrate of strain MR-57 significantly reduced the conidial germination rate and induced alterations in the mycelia morphology of F. equiseti, such as deformation and degradation. To assess the antifungal efficacy of MR-57 against root rot disease and the properties promoting the growth of S. divaricata, pot experiments were performed under natural outdoor conditions. The results indicated that co-inoculation with MR-57 delayed the occurrence of S. divaricata root rot and showed a control efficacy of 65.41% (p < 0.05) based on the measurement of suppressed disease lesions. Additionally, MR-57 successfully colonized and formed a stable population in the soil in which S. divaricata was grown, and it exhibited a consistently positive effect on the promotion of the growth of S. divaricata plants. In short, Acr. jodhpurensis MR-57 could be considered for the development of a potential biocontrol agent for the management of S. divaricata root rot caused by F. equiseti.

Hexokinase (HXK) family proteins exert critical roles in catalyzing hexose phosphorylation, sugar sensing, and modulation of plant growth and stress adaptation. Nevertheless, a large amount remains unknown about the molecular profile of HXK enzymes in Populus trichocarpa, a woody model tree species. A genome-wide survey of HXK-encoding genes, including phylogenies, genomic structures, exon/intron organization, chromosomal distribution, and conserved features, was conducted, identifying six putative HXK isogenes (PtHXK1-6) in the Populus genome. The evolutionary tree demonstrated that 135 homologous HXKs between 17 plant species were categorized into four major subfamilies (type A, B, C, and D), clustering one plastidic (PtHXK3) and five mitochondrial PtHXKs grouped into type A and B, respectively. The in silico deduction prompted the presence of the conserved sugar-binding core (motif 4), phosphorylation sites (motif 2 and 3), and adenosine-binding domains (motif 7). The transcriptomic sequencing (RNA-seq) and the quantitative real-time PCR (qRT-PCR) assays revealed that three isogenes (PtHXK2, 3, and 6) were abundantly expressed in leaves, stems, and roots, while others appeared to be dominantly expressed in the reproductive tissues. Under the stress exposure, PtHXK2 and 6 displayed a significant induction upon the pathogenic fungi (Fusarium solani) infection and marked promotions by glucose feeding in roots. In contrast, the PtHXK3 and 6 are ABA-responsive genes, following a dose-dependent manner. The comprehensive analyses of the genomic patterns and expression profiling provide theoretical clues and lay a foundation for unraveling the physiological and signaling roles underlying the fine-tuned PtHXKs responding to diverse stressors.

Objective: A sensitive and selective fluorescence-detection platform based on carbon quantum dots (CQDs) was designed and developed for the determination of methotrexate (MTX), for the purpose of minimizing the possible toxic threat of MTX in clinics. Methods: The approach was prepared for the first time by a simple, hydrothermal method, making the synthesis and modification processes realized in one step using polyethyleneimine (PEI), and the proposed PEI-CQDs were obtained with high fluorescence quantum yield (38%). Results: MTX was found highly responsive and effective in quenching the fluorescence of the PEI-CQDs, due to a suggested fluorescence resonance energy transfer mechanism or inner-filter effect. The linear range of MTX was between 1 and 600 μmol/L under optimum conditions, with a detection limit (LOD) as low as 0.33 μmol/L. Furthermore, the fluorescent method was established for the MTX assay, and satisfactory results were acquired in real-sample determination. The average labeled quantity was 98.2%, and the average added standard recovery was 100.9%. Conclusions: The proposed PEI-CQDs showed a remarkable potential for broad applications in biological molecule determination and environmental analysis.

Currently, there are only a limited number of Japanese-Chinese bilingual corpora of a sufficient amount that can be used as training data for neural machine translation (NMT). In particular, there are few corpora that include spoken language such as daily conversation. In this research, we attempt to construct a Japanese-Chinese bilingual corpus of a certain scale by crawling the subtitle data of movies and TV series from the websites. We calculated the BLEU scores of the constructed WCC-JC (Web Crawled Corpus—Japanese and Chinese) and the other compared corpora. We also manually evaluated the translation results using the translation model trained on the WCC-JC to confirm the quality and effectiveness.

Our research focused on the hypoglycemic capability and the possible mechanisms of extract and fractions from Polygoni Avicularis Herba (PAH) based on α-glucosidase, α-amylase inhibition assays, glucose uptake experiment, HPLC-MS analysis, and molecular docking experiment. In addition, DPPH, ABTS, and FRAP assays were used for determining the antioxidant capability. The results of total flavonoids and phenolics contents showed that ethyl acetate fraction (EAF) possessed the highest flavonoids and phenolics with values of 159.7 ± 2.5 mg rutin equivalents/g and 107.6 ± 2.0 mg galic acid equivalents/g, respectively. The results of in vitro hypoglycemic activity showed that all samples had effective α-glucosidase inhibition capacities, and EAF possessed the best inhibitory effect with IC50 value of 1.58 ± 0.24 μg/mL. In addition, n-butanol fraction (NBF) significantly promoted the glucose uptake rate of 3T3-L1 adipocytes. HPLC-MS analysis and molecular docking results proved the interactions between candidates and α-glucosidase. The results of antioxidation capacities showed that EAF possessed the best antioxidation abilities with DPPH, ABTS, and FRAP. In summary, the hypoglycemic activity of PAH might be related to the inhibition of α-glucosidase (EAF > PEF > NBF) and the promotion of glucose uptake in 3T3-L1 adipocytes (NBF). Simultaneously, the antioxidation capacity of PAH might be related to the abundant contents of flavonoids and other phenolics (EAF > PEF > NBF).

Neurodegenerative disorders have become a serious healthcare problem worldwide and there is no efficacious cure. However, regulating the fate of stem cells is an effective way to treat these neurological diseases. In previous work, stemazole was reported to maintain the survival of human neural stem cells in the absence of growth factors and to have therapeutic effects on neurodegenerative diseases. However, although it is a promising small molecule, the molecular mechanisms against apoptosis are ambiguous. In this study, tandem mass tag (TMT)-based proteomics were performed to obtain whole protein expression profiles of human neural stem cells in different groups under extreme conditions. Bioinformatics analysis based on protein–protein interaction (PPI) network construction, gene ontology (GO) and the Kyoto Encyclopaedia of Genes and Genomes (KEGG) pathway enrichment analysis were adopted to explore crucial proteins and possible pharmacological mechanisms. A total of 77 differentially expressed proteins were identified, comprising 38 upregulated proteins and 39 downregulated proteins. Combined with a diseases database of Alzheimer’s disease (AD), caspase-2 (CASP2), PKA C-alpha (PRKACA), fibronectin (FN1), large neutral amino acid transporter small subunit 1 (SLC7A5), which are involved in cell proliferation and apoptosis, this was further validated by enzyme activity assay and molecular docking, and regarded as putative targets regulated by stemazole. The present results give an insight into this small molecule and a better understanding for further elucidating the underlying mechanisms in the treatment of stem cells and neurodegenerative diseases.

The post-fire debris flow (PFDF) is a commonly destructive hazard that may persist for several years following the wildfires. Susceptibility mapping is an effective method for mitigating hazard risk. Yet, the majority of susceptibility prediction models only focus on spatial probability in the specific period while ignoring the change associated with time. This study improves the predictive model by introducing the temporal factor. The area burned by the 30 March 2020 fire in Xichang City, China is selected as an illustrative example, and the susceptibility of the PFDF was predicted for different periods of seven months after the wildfires. 2214 hydrological response events, including 181 debris flow events and 2033 flood events from the 82 watersheds are adopted to construct the sample dataset. Seven conditioning factors consist of temporal factors and spatial factors are extracted by the remote sensing interpretation, field investigations, and in situ tests, after correlation and importance analysis. The logistic regression (LR) is adopted to establish prediction models through 10 cross-validations. The results show that the susceptibility to PFDF has significantly reduced over time. After two months of wildfire, the proportions of very low, low, moderate, high, and very high susceptibility are 1.2%, 3.7%, 24.4%, 23.2%, and 47.6%, respectively. After seven months of wildfire, the proportions of high and very high susceptibility decreased to 0, while the proportions of very low to medium susceptibility increased to 35.4%, 35.6%, and 28.1%, respectively. The reason is that the drone seeding of grass seeds and artificial planting of trees accelerated the natural recovery of vegetation and soil after the fire. This study can give insight into the evolution mechanism of PFDF over time and reflect the important influence of human activity after the wildfire.

(1) Background: Carbon quantum dots (CQDs) are a new class of carbon nanomaterials with favorable features, such as tunable luminescence, unique optical properties, water solubility, and lack of cytotoxicity; they are readily applied in biomedicine. (2) Methods: S, N co-doped CQDs were prepared to develop a highly selective and sensitive fluorescence technique for the detection of methotrexate (MTX). For this purpose, citric acid and thiourea were used as C, N, and S sources in a single-step hydrothermal process to prepare the S, N co-doped CQDs, which displayed remarkable fluorescence properties. (3) Results: Two optimal emissions were observed at the excitation/emission wavelengths of 320/425 nm, respectively. The two emissions were significantly quenched in the presence of MTX. Under optimal conditions, MTX was detected in the linear concentration range of 1–300 μmol/L, with the detection limit of 0.33 μmol/L. The sensing mechanism was due to the fact that the effect of the inner filter on MTX and S, N-CQDs causes fluorescence quenching. The contents of MTX in real medicine samples were evaluated with acceptable recoveries of 98–101%. (4) Conclusions: This approach has great potential for detecting MTX in pharmaceutical analysis.

Zonula occludens-1 (ZO-1) is an intracellular scaffolding protein that orchestrates the anchoring of membrane proteins to the cytoskeleton in epithelial and specialized tissue including the heart. There is clear evidence to support the central role of intracellular auxiliary proteins in arrhythmogenesis and previous studies have found altered ZO-1 expression associated with atrioventricular conduction abnormalities. Here, using human cardiac tissues, we identified all three isoforms of ZO-1, canonical (Transcript Variant 1, TV1), CRA_e (Transcript Variant 4, TV4), and an additionally expressed (Transcript Variant 3, TV3) in non-failing myocardium. To investigate the role of ZO-1 on ventricular arrhythmogenesis, we generated a haploinsufficient ZO-1 mouse model (ZO-1+/−). ZO-1+/− mice exhibited dysregulated connexin-43 protein expression and localization at the intercalated disc. While ZO-1+/− mice did not display abnormal cardiac function at baseline, adrenergic challenge resulted in rhythm abnormalities, including premature ventricular contractions and bigeminy. At baseline, ventricular myocytes from the ZO-1+/− mice displayed prolonged action potential duration and spontaneous depolarizations, with ZO-1+/− cells displaying frequent unsolicited (non-paced) diastolic depolarizations leading to spontaneous activity with multiple early afterdepolarizations (EADs). Mechanistically, ZO-1 deficient myocytes displayed a reduction in sodium current density (INa) and an increased sensitivity to isoproterenol stimulation. Further, ZO-1 deficient myocytes displayed remodeling in ICa current, likely a compensatory change. Taken together, our data suggest that ZO-1 deficiency results in myocardial substrate susceptible to triggered arrhythmias.

Solanum aethiopicum is a very important vegetable for both rural and urban communities in Africa. The crop is rich in both macro- and micronutrients compared with other vegetables and is suitable for ensuring food and nutritional security. It also possesses several medicinal properties and is currently employed in the treatment of high blood pressure, diabetes, cholera, uterine complaints as well as skin infections in humans. The crop is predominantly cultivated by traditional farmers and plays an important role in the subsistence and economy of poor farmers and consumers throughout the developing world. It also holds potential for dietary diversification, greater genetic biodiversity and sustainable production in Africa. Despite the numerous benefits the crop presents, it remains neglected and underutilized due to the world’s over-dependence on a few plant species, as well as the little attention in research and development it has received over the years. This review highlights the importance of S. aethiopicum, its role in crop diversification, reducing hidden hunger, the potential for nutritive and medicinal benefits, agricultural sustainability and future thrusts for breeding and genetic improvement of the plant species.

This paper presents a recent rainfall-induced landslide in China that occurred on August 21, 2020 and resulted in nine deaths. The sliding material traveled a distance of 800 m, with an altitude difference of about 180 m. A field investigation, remote sensing based on an unmanned aerial vehicle (UAV), in situ monitoring, and a rainfall data analysis were carried out to reveal the deposit characteristics, causative factors, post-landslide behavior, and the mechanism of the landslide. A saltatory micro-relief of the original slope determined the multiple-stage failure type of the slide, and also promoted the entrainment effect during the landslide movement. After the first-initiation sliding stage, the motion of this landslide involved typical progressive movement, and over time, the style of the runout generally turned into a flow-like form. Furthermore, the antecedent cumulative rainfall of 149.5 mm directly contributed to the occurrence of the landslide. Using the GB-SAR early warning system, the post-landslide residual failure was successfully predicted 10 min in advance. The combination of the UAV and GB-SAR technique can surely be beneficial for other inaccessible landslide investigations as well and improves the emergency rescue security.

L-aspartate (Asp) serves as a central building block, in addition to being a constituent of proteins, for many metabolic processes in most organisms, such as biosynthesis of other amino acids, nucleotides, nicotinamide adenine dinucleotide (NAD), the tricarboxylic acid (TCA) cycle and glycolysis pathway intermediates, and hormones, which are vital for growth and defense. In animals and humans, lines of data have proved that Asp is indispensable for cell proliferation. However, in plants, despite the extensive study of the Asp family amino acid pathway, little attention has been paid to the function of Asp through the other numerous pathways. This review aims to elucidate the most important aspects of Asp in plants, from biosynthesis to catabolism and the role of Asp and its metabolic derivatives in response to changing environmental conditions. It considers the distribution of Asp in various cell compartments and the change of Asp level, and its significance in the whole plant under various stresses. Moreover, it provides evidence of the interconnection between Asp and phytohormones, which have prominent functions in plant growth, development, and defense. The updated information will help improve our understanding of the physiological role of Asp and Asp-borne metabolic fluxes, supporting the modular operation of these networks.

In order to select the optimal transportation route among alternative transportation routes more accurately and objectively, the choice of urban oversize cargo transportation routes was studied by taking the optimization weight–TOPSIS combination method for specific calculations. This model, based on an entropy weight method, cloud model, and TOPSIS method, combines the superiority of the cloud model for reflecting the randomness and discreteness of subjective evaluation with the advantages of the TOPSIS method in dealing with the problem of multi-objective programming. Through selecting and classifying several the main road influencing factors of urban oversize cargo transportation, based on the data of four urban roads, the entropy weight method is used to initially determine the weights of each influencing factor, the cloud model is used to optimize weights, the TOPSIS method is used to compare and evaluate the paths, and the optimal transportation route is selected on this basis. The results showed that the optimization weight–TOPSIS method is scientific and accurate for the multi-objective planning of oversize cargo transportation route selection, and solves the problem of the impact of subjective factors in existing methods and the difficulty of processing multiple influencing factors. The Pearson consistency test results show that the Pearson correlation coefficient between the proposed method and the actual oversize cargo transportation route selection is 0.995, which is higher than the calculation results without using the combination weight.

In order to solve the environmental protection problem of carbon emissions in the field of freight transportation, this article proposes to promote the transfer of road freight transportation to railway transportation within a reasonable range by levying carbon emission taxes. To propose an applicable solution, this paper establishes a comprehensive carbon emission system model in the field of road transportation and railway transportation to simulate a closed-loop system as comprehensively as a real transportation system, determines the system elements according to the actual situation, reasonably develops the model hypothesis scheme, and draws out the causal network. On this basis, the system flow diagram and corresponding structural equations are constructed, and the model parameters are estimated. Finally, the paper uses actual data to verify and simulate the system model. A reasonable carbon levy interval has been obtained, and the carbon levy within this interval can promote the transfer of road freight transportation to railway transportation, so as to achieve the purpose of decreasing total carbon emissions of road–rail transportation systems in an orderly way. The innovation of this paper is to construct the carbon emissions of the road–rail system systematically for the first time, and to conduct research and exploration of carbon levies on this basis.

Fusarium solani (Fs) is one of the notorious necrotrophic fungal pathogens that cause root rot and vascular wilt, accounting for the severe loss of Populus production worldwide. The plant–pathogen interactions have a strong molecular basis. As yet, the genomic information and transcriptomic profiling on the attempted infection of Fs remain unavailable in a woody model species, Populus trichocarpa. We used a full RNA-seq transcriptome to investigate the molecular interactions in the roots with a time-course infection at 0, 24, 48, and 72 h post-inoculation (hpi) of Fs. Concomitantly, the invertase and invertase inhibitor-like gene families were further analyzed, followed by the experimental evaluation of their expression patterns using quantitative PCR (qPCR) and enzyme assay. The magnitude profiles of the differentially expressed genes (DEGs) were observed at 72 hpi inoculation. Approximately 839 genes evidenced a reception and transduction of pathogen signals, a large transcriptional reprogramming, induction of hormone signaling, activation of pathogenesis-related genes, and secondary and carbohydrate metabolism changes. Among these, a total of 63 critical genes that consistently appear during the entire interactions of plant–pathogen had substantially altered transcript abundance and potentially constituted suitable candidates as resistant genes in genetic engineering. These data provide essential clues in the developing new strategies of broadening resistance to Fs through transcriptional or translational modifications of the critical responsive genes within various analyzed categories (e.g., carbohydrate metabolism) in Populus.

Plant host defense peptides (HDPs), also known as antimicrobial peptides (AMPs), are regarded as one of the most prevalent barriers elaborated by plants to combat various infective agents. Among the multiple classes of HDPs, the Snakin class attracts special concern, as they carry 12 cysteine residues, being the foremost cysteine-rich peptides of the plant HDPs. Also, their cysteines are present at very highly conserved positions and arranged in an extremely similar way among different members. Like other plant HDPs, Snakins have been shown to exhibit strong antifungal and antibacterial activity against a wide range of plant pathogens. Moreover, they display diversified biological activities in many aspects of plant growth and the development process. This review is devoted to present the general characters of the Snakin class of plant HDPs, as well as the individual features of different Snakin family members. Specifically, the sequence properties, spatial structures, distributions, expression patterns and biological activities of Snakins are described. In addition, further detailed classification of the Snakin family members, along with their possible mode of action and potential applications in the field of agronomy and pathology are discussed.

Arginine octamer (R8) and its derivatives were developed in this study for the enhanced mucosal permeation of insulin. R8 was substituted with different aminos, then modified with stearic acid (SA). We found that the SAR6EW-insulin complex had stronger intermolecular interactions and higher complex stability. The amphiphilic lipopeptide (SAR6EW) was significantly more efficient for the permeation of insulin than R8 and R6EW both in vitro and in vivo. Interestingly, different cellular internalization mechanisms were observed for the complexes. When the effectiveness of the complexes in delivering insulin in vivo was examined, it was found that the SAR6EW-insulin complex provided a significant and sustained (six hours) reduction in the blood glucose levels of diabetic rats. The improved absorption could be the comprehensive result of stronger intermolecular interactions, better enzymatic stability, altered internalization pathways, and increased transportation efficacy. In addition, no sign of toxicity was observed after consecutive administrations of SAR6EW. These results demonstrate that SAR6EW is a promising epithelium permeation enhancer for insulin and suggest that the chemical modification of cell-penetrating peptides is a feasible strategy to enhance their potential.

In the present study, tocotrienol rich fraction (TRF) nanoemulsions were produced as an alternative approach to improve solubility and absorption of tocotrienols. In the present study, droplet size obtained after 10 cycles of homogenization with increasing pressure was found to decrease from 120 to 65.1 nm. Nanoemulsions stabilized with Tween series alone or emulsifier blend Brij 35:Span 80 (0.6:0.4 w/w) homogenized at 25,000 psi and 10 cycles, produced droplet size less than 100 nm and a narrow size distribution with a polydispersity index (PDI) value lower than 0.2. However blend of Tween series with Span 80 produced nanoemulsions with droplet size larger than 200 nm. This work has also demonstrated the amount of tocols losses in TRF nanoemulsion stabilized Tweens alone or emulsifier blend Brij 35:Span 80 (0.6:0.4 w/w) ranged between 3%–25%. This can be attributed to the interfacial film formed surrounding the droplets exhibited different level of oxidative stability against heat and free radicals created during high pressure emulsification.

Panax ginseng C.A. Meyer is one of the most important medicinal plants in Northeast China, and ginsenosides are the main active ingredients found in medicinal ginseng. The biosynthesis of ginsenosides is regulated by environmental factors and the expression of key enzyme genes. Therefore, in this experiment, ginseng in the leaf opened stage, the green fruit stage, the red fruit stage, and the root growth stage was used as the test material, and nine individual ginsenosides and total saponins (the sum of the individual saponins) were detected by HPLC (High Performance Liquid Chromatography). There was a trend of synergistic increase and decrease, and saponin accumulation and transfer in different tissues. The expression of key enzyme genes in nine synthetic pathways was detected by real-time PCR, and the correlation between saponin content, gene expression, and ecological factors was analyzed. Correlation analysis showed that in root tissue, PAR (Photosynthetically Active Radiation) and soil water potential had a greater impact on ginsenoside accumulation, while in leaf tissue, temperature and relative humidity had a greater impact on ginsenoside accumulation. The results provide a theoretical basis for elucidating the relationship between ecological factors and genetic factors and their impact on the quality of medicinal materials. The results also have guiding significance for realizing the quality of medicinal materials.

Protopanaxatriol saponins obtained with AB-8 macroporous resin mainly consisted of ginsenosides Rg1 and Re. A novel mono-ester of ginsenoside-Rh1 (ginsenoside-ORh1) was synthesized through further enzymatic hydrolysis and octanoyl chloride modifications. A 53% yield was obtained by a facile synthetic method. The structures were identified on the basis of 1D-NMR and 2D-NMR, as well as ESI-TOF-MS mass spectroscopic analyses. The isolated and synthetic compounds were applied in an anti-tumor bioassay, in which ginsenoside ORh1 showed moderate effects on Murine H22 Hepatoma Cells.

There are six tocol analogs present in palm oil, namely α-tocopherol (α-T), α-tocomonoenol (α-T1), α-tocotrienol (α-T3), γ-tocotrienol (γ-T3), β-tocotrioenol (β-T3) and δ-tocotrienol (δ-T3). These analogs were difficult to separate chromatographically due to their similar structures, physical and chemical properties. This paper reports on the effect of pressure and injection solvent on the separation of the tocol analogs in palm oil. Supercritical CO2 modified with ethanol was used as the mobile phase. Both total elution time and resolution of the tocol analogs decreased with increased pressure. Ethanol as an injection solvent resulted in peak broadening of the analogs within the entire pressure range studied. Solvents with an eluent strength of 3.4 or less were more suitable for use as injecting solvents.

Metastatic cancers are the main cause of cancer-related death. In breast primary cancer, the five-year survival rate is close to 100%; however, for metastatic breast cancer, that rate drops to a mere 25%, due in part to the paucity of effective therapeutic options for treating metastases. Several in vitro and in vivo studies have indicated that consumption of natural polyphenols significantly reduces the risk of cancer metastasis. Therefore, this review summarizes the research findings involving the molecular mechanisms and metabolomics of natural polyphenols and how they may be blocking breast cancer metastasis. Most natural polyphenols are thought to impair breast cancer metastasis through downregulation of MMPs expression, interference with the VEGF signaling pathway, modulation of EMT regulator, inhibition of NF-κB and mTOR expression, and other related mechanisms. Intake of natural polyphenols has been shown to impact endogenous metabolites and complex biological metabolic pathways in vivo. Breast cancer metastasis is a complicated process in which each step is modulated by a complex network of signaling pathways. We hope that by detailing the reported interactions between breast cancer metastasis and natural polyphenols, more attention will be directed to these promising candidates as effective adjunct therapies against metastatic breast cancer in the clinic.
