Transplanting has been widely used in American ginseng (Panax quinquefolium L.) cultivation in Northwest China to mitigate the negative effects of continuous cropping obstacles. Because of the accumulation of pathogenic microorganisms and the change in soil properties, transplanting American ginseng to newly cultivated fields after two years of growth has become a major planting pattern. Despite transplanting improving the quality of American ginseng, the effects of soil properties and microbiota on growth during the transplanting process are poorly understood. In the present study, microbial communities, soil physico-chemical properties and morpho-physiological parameters were analyzed to investigate the effects of microbiota and soil characteristics on American ginseng growth in both soil and ginseng root microhabitats. Results indicated that the structure and species of bacterial and fungal communities changed significantly in different microhabitats before and after transplantation. Moreover, the assemblage process of the bacterial community was dominated by deterministic processes. The stochastic process ratio increased and niche breadth decreased significantly after transplanting. While the assembly of the fungal community was dominated by stochastic process, and there was no significant difference in NST, βNTI or niche breadth before and after transplanting. Bacterial co-occurrence networks demonstrated a higher connectivity but a lower aggregation in soil microhabitat, while the fungal community networks remained stable before and after transplantation. Gammaproteobacteria was the biomarker in the soil microhabitat, while Alphaproteobacteria, Betaproteobacteria and Gemmatimonadetes were biomarkers in the ginseng root microhabitat. Sordariomycetes was a biomarker with high relative abundance in the fungal community before and after transplanting. The bacterial functional and important ASVs were significantly correlated with pH, organic matter, total nitrogen, available phosphorus, total potassium root fresh weight, taproot diameter and stem height of American ginseng. Partial least squares path modeling showed that soil properties significantly affected the formation of different microbial specific ASVs. The important functional ASVs in ginseng root microhabitat had a positive effect on American ginseng growth, while the rare taxa had a negative effect. Our results provide a good starting point for future studies of microbial community succession in different microhabitats influenced by the transplantation pattern of American ginseng.

The influence of Glucagon-like peptide-1 (GLP-1) and Exendin-4 on development of intrahepatic cholangiocarcinoma (ICC) is evaluated in the study. In vitro tests, including acute toxicity test, cell colony formation assays, cells proliferation and apoptosis, transwell assay, were performed. An ICC in situ tumor animal model was established. Then, animals were randomly divided into four groups (n = 6): control, Exendin-4 treatment, oxaliplatin treatment and Exendin-4-oxaliplatin treatment. Animals in the Exendin-4 treatment and Exendin-4-oxaliplatin treatment groups received a subcutaneous injection of Exendin-4 (100 μg/kg/day) for 1 week, and then received oxaliplatin (10 mg/kg/week) by tail vein injection. Animals in the control group received PBS. Immunohistochemistry tests were used for PCNA, Ki67, Caspase 3 expression in tumor tissue. Results show that that, after incubation of human cholangiocarcinoma cell lines, HuCCTI and GLP-1, or HuCCTI and Exendin-4, colony formation number was sharply decreased. However, GLP-1, HuCCTI or Exendin-4 did not affect the colony of normal cells. Combination treatment with oxaliplatin and Exendin-4 can significantly inhibit tumor cells’ proliferation and promote apoptosis. The combined effect is stronger than that of oxaliplatin or Exendin-4. Combination treatment with oxaliplatin and Exendin4 can significantly decrease Ki67 and PCNA proteins’ expression in subcutaneous tumors of nude mice. The inhibitory effect of Combination treatment with oxaliplatin and Exendin4 is clearly stronger than that of oxaliplatin. In addition, Combination treatment with oxaliplatin and Exendin4 can significantly increase Caspase3 protein positive expression. In short, these results show that combination treatment with oxaliplatin and Exendin4 can inhibit tumor cells’ proliferation, and promote apoptosis.

In host-parasitoid interactions, antagonistic relationship drives parasitoids to vary in virulence in facing different hosts, which makes these systems excellent models for stress-induced evolutionary studies. Venom compositions varied between two strains of Tetrastichus brontispae, Tb-Bl and Tb-On. Tb-Bl targets Brontispa longissima pupae as hosts, and Tb-On is a sub-population of Tb-Bl, which has been experimentally adapted to a new host, Octodonta nipae. Aiming to examine variation in parasitoid virulence of the two strains toward two hosts, we used reciprocal injection experiments to compare effect of venom/ovarian fluids from the two strains on cytotoxicity, inhibition of immunity and fat body lysis of the two hosts. We found that Tb-Onvenom was more virulent towards plasmatocyte spreading, granulocyte function and phenoloxidase activity than Tb-Blvenom. Tb-Blovary was able to suppress encapsulation and phagocytosis in both hosts; however, Tb-Onovary inhibition targeted only B. longissima. Our data suggest that the venom undergoes rapid evolution when facing different hosts, and that the wasp has good evolutionary plasticity.

The most significant threat to pepper production worldwide is the Phytophthora blight, which is caused by the oomycete pathogen, Phytophthora capsici Leonian. In an effort to help control this disease, we isolated and characterized a P. capsici resistance gene, CaRGA2, from a high resistant pepper (C. annuum CM334) and analyzed its function by the method of real-time PCR and virus-induced gene silencing (VIGS). The CaRGA2 has a full-length cDNA of 3,018 bp with 2,874 bp open reading frame (ORF) and encodes a 957-aa protein. The protein has a predicted molecular weight of 108.6 kDa, and the isoelectric point is 8.106. Quantitative real-time PCR indicated that CaRGA2 expression was rapidly induced by P. capsici. The gene expression pattern was different between the resistant and susceptible cultivars. CaRGA2 was quickly expressed in the resistant cultivar, CM334, and reached to a peak at 24 h after inoculation with P. capsici, five-fold higher than that of susceptible cultivar. Our results suggest that CaRGA2 has a distinct pattern of expression and plays a critical role in P. capsici stress tolerance. When the CaRGA2 gene was silenced via VIGS, the resistance level was clearly suppressed, an observation that was supported by semi-quantitative RT-PCR and detached leave inoculation. VIGS analysis revealed their importance in the surveillance to P. capsici in pepper. Our results support the idea that the CaRGA2 gene may show their response in resistance against P. capsici. These analyses will aid in an effort towards breeding for broad and durable resistance in economically important pepper cultivars.
