Sapoviruses, like noroviruses, are single-stranded positive-sense RNA viruses classified in the family Caliciviridae and are recognized as a causative pathogen of diarrhea in infants and the elderly. Like human norovirus, human sapovirus (HuSaV) has long been difficult to replicate in vitro. Recently, it has been reported that HuSaV can be replicated in vitro by using intestinal epithelial cells (IECs) derived from human tissues and cell lines derived from testicular and duodenal cancers. In this study, we report that multiple genotypes of HuSaV can sufficiently infect and replicate in human-induced pluripotent stem cell-derived IECs. We also show that this HuSaV replication system can be used to investigate the conditions for inactivation of HuSaV by heat and alcohol, and the effects of virus neutralization of antisera obtained by immunization with vaccine antigens, under conditions closer to the living environment. The results of this study confirm that HuSaV can also infect and replicate in human normal IECs regardless of their origin and are expected to contribute to future virological studies.

Pedestrian deaths account for the highest percentage of fatality caused by traffic accidents in Japan. Increasing pedestrian safety is a key objective for reducing such deaths. For pedestrian fatality caused by vehicles at low speed, turning the vehicle toward the right was the most common behavior under left-hand traffic. Autonomous emergency braking (AEB) systems for pedestrian safety have great potential to mitigate pedestrian injuries and fatalities in traffic accidents. However, pedestrian-AEB systems, especially for vehicles turning right, are still under development. This study identified the characteristics of dangerous traffic scenarios between vehicles turning right and pedestrians, focusing on two directions of pedestrian crossing: from the left to the right side (Left-Pedestrian) and from the right to the left side (Right-Pedestrian). The ego vehicle recorded near-miss incidents using a drive recorder. The results revealed that the Left-Pedestrian and Right-Pedestrian scenarios had different features for both the width of roads going to and through the intersection and the average of the travel speeds of the ego vehicles. They had similar characteristics in terms of the presence of other vehicle categories, but differences in the relationship of numbers and/or travel directions of other vehicles. The findings of this study will contribute to the development and evaluation of safety systems for preventing collisions between right-turning vehicles and pedestrians at intersections.

Basic helix-loop-helix (BHLH) transcription factors differentiated embryonic chondrocyte gene 1 (DEC1) and gene 2 (DEC2) regulate circadian rhythms, apoptosis, epithelial mesenchymal transition (EMT), invasions and metastases in various kinds of cancer. The stem cell markers SOX2 and c-MYC are involved in the regulation of apoptosis and poor prognosis. In cervical cancer, however, their roles are not well elucidated yet. To determine the function of these genes in human cervical cancer, we examined the expression of DEC1, DEC2, SOX2 and c-MYC in human cervical cancer tissues. In immunohistochemistry, they were strongly expressed in cancer cells compared with in non-cancerous cells. Notably, the strong rate of DEC1 and SOX2 expressions were over 80% among 20 cases. We further examined the roles of DEC1 and DEC2 in apoptosis. Human cervical cancer HeLa and SiHa cells were treated with cisplatin—HeLa cells were sensitive to apoptosis, but SiHa cells were resistant. DEC1 expression decreased in the cisplatin-treated HeLa cells, but had little effect on SiHa cells. Combination treatment of DEC1 overexpression and cisplatin inhibited apoptosis and affected SOX2 and c-MYC expressions in HeLa cells. Meanwhile, DEC2 overexpression had little effect on apoptosis and on SOX2 and c-MYC expressions. We conclude that DEC1 has anti-apoptotic effects and regulates SOX2 and c-MYC expressions on apoptosis.

We investigated the effect of driver posture on driving control following a takeover request (TOR) from autonomous to manual driving in level 3 autonomous vehicles. When providing a TOR, driving behaviors need to be investigated to develop driver monitoring systems, and it is important to clarify the effect of driver postures. Experiments were conducted using driver postures that are likely to be adopted in autonomous driving. Driver postures were set based on combinations of two types of upper-body posture and three types of foot posture. The driver’s upper body and head were set to either a forward or sideways orientation. For each of these there were three types of foot posture: both feet on the floor, crossed legs, and cross-legged sitting. Following a TOR, we compared the braking and steering maneuvers of subjects driving normally and examined the effects of posture on driver reaction time. The results show that both the upper-body and foot postures of the driver affect the steering and braking reaction time. The driver monitoring system should be able to detect posture and activate a TOR warning, and detection times up to 2 and 1.3 times faster than those for normal postures should be considered for different upper-body and foot postures, respectively.

This study aimed to devise a novel physique index and investigate its accuracy in identifying newborns with skeletal dysplasia in comparison with head circumference (HC)/height (HT) ratio. The birth weight (W), HT, and HC at birth of 1500 newborns were retrospectively collected. The linear regression equations and coefficients of determination (R2) were determined. The formulated equation was corrected by the mean weight for gestational age at birth (Wcorr) as a novel physique index for screening skeletal dysplasia. The index accuracy was assessed using receiver operating characteristic (ROC) curves in 11 newborns by fetal ultrasound and compared with that of the HC/HT ratio. The R2 values between W and HT, (HT)2, and (HT) 3 were 0.978, 0.990, and 0.993, respectively. Those between W and HC, (HC)2, and (HC)3 were 0.974, 0.984, and 0.988, respectively. W/Wcorr × (HC/HT)3 was used as a novel physique index. Seven newborns had skeletal dysplasia. Our novel physique index had a higher area under the curve (AUC), sensitivity, and specificity than the HC/HT ratio (AUC: 1.00 vs. 0.86, sensitivity: 1.00 vs. 0.86, and specificity: 1.00 vs. 0.75, respectively). Our novel physique index was more accurate than HC/HT ratio and has the potential to accurately identify newborns with skeletal dysplasia.

Virus–host cell interactions in rubella virus (RuV) are of great interest in current research in the field, as their mechanism is not yet well understood. By hypothesizing that the epithelial-to-mesenchymal transition (EMT) may play a role in RuV infection, this study aimed to investigate the influence of TGF-β1-induced EMT of human lung epithelial A549 cells on the infectivity of RuV. A549 cells were cultured and treated with TGF-β1 for 1 to 2 days prior to virus infection (with a clinical strain). Viral infectivity was determined by flow cytometry analysis of cells harvested at 24 and 48 h post-infection (hpi) and by titration of supernatants collected at 48 hpi. The results showed that the percentages of the TGF-β1-treated A549 cells that were positive for RuV were at least twofold higher than those of the control, and the viral progeny titers in the supernatants collected at 48 hpi were significantly higher in the treatment group than in the control group. In addition, the virus binding assay showed a strong increase (more than threefold) in the percentages of RuV-positive cells, as determined by flow cytometry analysis and further confirmed by real-time PCR. Such an enhancement effect on RuV infectivity was abolished using LY364947 or SB431542, inhibitors of the TGF-β/Smad signaling pathway. The findings suggest that the TGF-β1-induced EMT-like process enhances RuV binding and infection in A549 cells via the Smad pathway. Further studies are necessary to identify possible proteins that facilitate viral binding and entry into treated cells.

We investigated a driver’s gazing time during backing vehicle maneuvers in a 90-degree reverse parking system for the phase from start to the 5-m point (start phase) and the phase from backing from the 5-m point to stop (stop phase). The objective of this study was to elucidate the driver’s gaze while approaching the parking area and in the parking area during backing. We used a sport utility vehicle with the driver’s seat at the right-hand side. We modified the vehicle with four technological conditions: no-tech vehicle, sonar vehicle, backup-camera vehicle, and backup camera with sonar vehicle. The monitor, rearview mirror, left-side mirror, and right-side mirror were designated as the area of interest (AOI) in the gaze. For the start phase, there were no significant differences in the total gazing time of all the AOIs in the four technologies. For the stop phase, however, the total gazing time of all the AOIs for the vehicle with a backup camera was significantly longer than that of the vehicle without a backup camera owing to the increase in the gazing time of the monitor. The results obtained from these experiments will form a basis for the future development of vehicle camera systems.

The purpose of this study was to determine the effect of vaccination and beta-carotene supplementation on blood oxidative stress and antibody response in calves. Thirty Japanese Black calves were randomly assigned to two groups. Fifteen calves received 20 mg of beta-carotene supplemented into their daily provided rations from 2 to 8 weeks of age (BC group), and the other 15 calves did not receive the daily beta-carotene supplement (control group). All calves received a commercially available modified live bovine respiratory syncytial (RS) virus vaccine at 4 and 8 weeks of age. Blood samples were taken at 2, 4, 8, and 12 weeks of age. At 4 weeks of age, the concentration of reactive oxygen metabolites within serum were significantly lower in the BC group than the concentrations measured in the control group. Also at 4 weeks of age, the concentration of biological antioxidant capacity within serum was significantly higher in the BC group than the concentrations measured in the control group. Both groups showed a gradual decrease of antibody titers to live bovine RS virus in the samples taken from 2 to 12 weeks of age. These results confirmed that beta-carotene supplementation decreased oxidative stress. However, beta-carotene supplementation did not affect the antibody response to live bovine RS virus vaccination, perhaps due to the presence of the maternal antibody.

Vehicle occupants were killed in 33% of all traffic accidents in Japan in 2017. Of the vehicles in vehicle-to-vehicle accidents, 54% were impacted from the front. In frontal impact accidents, when the lap belt moves away from the iliac crests of the pelvis of a vehicle occupant, the belt moves directly into the abdomen. Here, we investigated causes of abdominal injuries to vehicle occupants, because the abdomen is associated with the highest rates of severe injury and fatality. The purpose of this study was to clarify the correlation between downward movement of the seat and of the lap belt away from the iliac crests of a human occupant of a car, in the event of a frontal impact. We investigated this phenomenon by conducting simulations using an anthropomorphic 50th percentile male (AM50) human model wearing a three-point seatbelt. We set two deformable seat conditions: Vertical movement and lean forward movement. Our results revealed that the lap belt came off from both of the iliac crests during lean forward movement but only from one of the iliac crests during vertical movement. We concluded that abdominal injuries can be caused by downward movement together with forward rotation in the seat during vehicle-to-vehicle frontal impacts.

A novel nor-phragmalin-type limonoid, named carapanosin D (1), and two novel mexicanolide-type limonoids, carapanosins E (2) and F (3), were isolated from the seed oil of andiroba (Carapa guianensis Aublet), a traditional medicine in Brazil and Latin American countries. Their structures were unambiguously determined on the basis of spectroscopic analyses using one-dimensional (1D) and two-dimensional (2D) NMR techniques and High resolution Fast Atom Bombardment Mass Spectrometry (HRFABMS). Compounds 1–3 were evaluated for their effects on the production of nitric oxide (NO) in Lipopolysaccharide (LPS)-activated mouse peritoneal macrophages. The NO inhibitory assay suggested that compounds 2 and 3 have high potency as inhibitors of macrophage activation.

The paper presents an emerging issue of fine-grained pedestrian action recognition that induces an advanced pre-crush safety to estimate a pedestrian intention in advance. The fine-grained pedestrian actions include visually slight differences (e.g., walking straight and crossing), which are difficult to distinguish from each other. It is believed that the fine-grained action recognition induces a pedestrian intention estimation for a helpful advanced driver-assistance systems (ADAS). The following difficulties have been studied to achieve a fine-grained and accurate pedestrian action recognition: (i) In order to analyze the fine-grained motion of a pedestrian appearance in the vehicle-mounted drive recorder, a method to describe subtle change of motion characteristics occurring in a short time is necessary; (ii) even when the background moves greatly due to the driving of the vehicle, it is necessary to detect changes in subtle motion of the pedestrian; (iii) the collection of large-scale fine-grained actions is very difficult, and therefore a relatively small database should be focused. We find out how to learn an effective recognition model with only a small-scale database. Here, we have thoroughly evaluated several types of configurations to explore an effective approach in fine-grained pedestrian action recognition without a large-scale database. Moreover, two different datasets have been collected in order to raise the issue. Finally, our proposal attained 91.01% on National Traffic Science and Environment Laboratory database (NTSEL) and 53.23% on the near-miss driving recorder database (NDRDB). The paper has improved +8.28% and +6.53% from baseline two-stream fusion convnets.
