Background: In current clinical practice, intensity-modulated proton therapy (IMPT) head and neck cancer (HNC) plans are generated using a constant relative biological effectiveness (cRBE) of 1.1. The primary goal of this study was to explore the dosimetric impact of proton range uncertainties on RBE-weighted dose (RWD) distributions using a variable RBE (vRBE) model in the context of bilateral HNC IMPT plans. Methods: The current study included the computed tomography (CT) datasets of ten bilateral HNC patients who had undergone photon therapy. Each patient’s plan was generated using three IMPT beams to deliver doses to the CTV_High and CTV_Low for doses of 70 Gy(RBE) and 54 Gy(RBE), respectively, in 35 fractions through a simultaneous integrated boost (SIB) technique. Each nominal plan calculated with a cRBE of 1.1 was subjected to the range uncertainties of ±3%. The McNamara vRBE model was used for RWD calculations. For each patient, the differences in dosimetric metrices between the RWD and nominal dose distributions were compared. Results: The constrictor muscles, oral cavity, parotids, larynx, thyroid, and esophagus showed average differences in mean dose (Dmean) values up to 6.91 Gy(RBE), indicating the impact of proton range uncertainties on RWD distributions. Similarly, the brachial plexus, brain, brainstem, spinal cord, and mandible showed varying degrees of the average differences in maximum dose (Dmax) values (2.78–10.75 Gy(RBE)). The Dmean and Dmax to the CTV from RWD distributions were within ±2% of the dosimetric results in nominal plans. Conclusion: The consistent trend of higher mean and maximum doses to the OARs with the McNamara vRBE model compared to cRBE model highlighted the need for consideration of proton range uncertainties while evaluating OAR doses in bilateral HNC IMPT plans.

Efficient triaging and referral assessments are critical in ensuring prompt medical intervention in the community healthcare (CHC) system. However, the existing triaging systems in many community health services are an intensive, time-consuming process and often lack accuracy, particularly for various symptoms which might represent heart failure or other health-threatening conditions. There is a noticeable limit of research papers describing AI technologies for triaging patients. This paper proposes a novel quantitative data-driven approach using machine learning (ML) modelling to improve the community clinical triaging process. Furthermore, this study aims to employ the feature selection process and machine learning power to reduce the triaging process’s waiting time and increase accuracy in clinical decision making. The model was trained on medical records from a dataset of patients with “Heart Failure”, which included demographics, past medical history, vital signs, medications, and clinical symptoms. A comparative study was conducted using a variety of machine learning algorithms, where XGBoost demonstrated the best performance among the other ML models. The triage levels of 2,35,982 patients achieved an accuracy of 99.94%, a precision of 0.9986, a recall of 0.9958, and an F1-score of 0.9972. The proposed diagnostic model can be implemented for the CHC decision system and be developed further for other medical conditions.

Background: Our aim was to examine the frequency and prescription pattern of breakthrough (BTO) and scheduled (SCH) opioids and their ratio (BTO/SCH ratio) of use, prior to and after referral to an inpatient supportive care consult (SCC) for cancer pain management (CPM). Methods and Materials: Patients admitted at the MD Anderson Cancer Center and referred to a SCC were retrospectively reviewed. Cancer patients receiving SCH and BTO opioids for ≥24 h were eligible for inclusion. Patient demographics and clinical characteristics, including the type and route of SCH and BTO opioids, daily opioid doses (MEDDs) of SCH and BTO, and BTO/SCH ratios were reviewed in patients seen prior to a SCC (pre-SCC) and during a SCC. A normal BTO ratio was defined as 0.5–0.2. Results: A total of 665/728 (91%) patients were evaluable. Median pain scores (p < 0.001), BTO MEDDs (p < 0.001), scheduled opioid MEDDs (p < 0.0001), and total MEDDs (p < 0.0001) were higher, but the median number of BTO doses was fewer (2 vs. 4, p < 0.001), among patients seen at SCC compared to pre-SCC. A BTO/SCH ratio over the recommended ratio (>0.2) was seen in 37.5% of patients. The BTO/SCH ratios in the pre-SCC and SCC groups were 0.10 (0.04, 0.21) and 0.17 (0.10, 0.30), respectively, p < 0.001. Hydromorphone and Morphine were the most common BTO and SCH opioids prescribed, respectively. Patients in the early supportive care group had higher pain scores and MEDDs. Conclusions: BTO/SCH ratios are frequently prescribed higher than the recommended dose. Daily pain scores, BTO MEDDs, scheduled opioid MEDDs, and total MEDDs were higher among the SCC group than the pre-SCC group, but the number of BTO doses/day was lower.

The phase behavior of lipids extracted from Astrocaryum vulgare (AV) and Astrocaryum aculeatum (AA) pulp and kernels and their microstructural, thermal and flow properties were studied. The lipid profiles, crystal structures, microstructures, thermal stabilities and flow behaviors of these lipids provided important structure–function information that are useful to assess potential applications in the food, cosmetic and pharmaceutical industries. AV and AA fruits were sourced from the lowlands and rainforests, respectively, of Guyana. AV and AA pulp oils (AVP and AAP) were distinguished from each other in composition and unsaturation, with AVP oils being predominated by a di-unsaturated TAG (2-(palmitoyloxy)propane-1,3-diyl dioleate (POO)) and AAP oils predominated by propane-1,2,3-triyl trioleate (OOO); there were unsaturation levels of 65% and 80%, respectively. The main fatty acids in AVP oils were oleic, palmitic and stearic; for AAP, these were oleic, linoleic, palmitic and stearic. The kernel fats of AV and AA were similar in composition and had saturation levels of 80%, being mainly comprised of tri-saturated TAGs propane-1,2,3-triyl tridodecanoate (LLL) and 3-(tetradecanoyloxy)propane-1,2-diyl didodecanoate (LML). The onset of mass loss (T5%on) of AV and AA pulp oils were similar at 328 ± 6 °C, which were 31 °C ± 9 higher compared to that of the kernel fats, which demonstrated similar T5%on = 293 ± 7 °C. AA and AV pulp oils were liquid at room temperature, with melting points of −5 ± 1 °C and 3 ± 1 °C, respectively; both kernel fats were solid at room temperature, packing in β′ (90% of crystals) and β (10% of crystals) polymorphic forms and melting almost identically at 30 ± 1 °C. Pulp oils demonstrated sporadic nucleation at the onset of crystallization with slow growth into rod-shaped crystallites, leading to an approximately 50% degree of crystallization at undercooling of approximately 40K. Nucleation for kernel fats was instantaneous at undercooling of approximately 23K, demonstrating a spherulitic growth pattern incorporating crystalline lamella and a 90% degree of crystallization. Kernel fats and pulp oils demonstrated Newtonian flow behavior and similar dynamic viscosity in the melt, approximately 28.5 mPa·s at 40 °C. The lipid profiles of AVP and AAP oils were dominated by unsaturated TAGs, suggesting potential nutrition and health benefits, particularly compared to other tropical oils with higher saturation levels, such as palm oil. AAP oil in particular is as unsaturated as olive oil, contains high levels of beta carotene and provides a unique flavor profile. The AAK and AVK lipid profiles and phase transformation indicate potential for applications where a high solid fat content and medium-chain fatty acids are required. Their high lauric and myristic acid content makes them similar to industrially important tropical oils (coconut and palm kernel), suggesting their use in similar formulations. The melting point and plasticity of the kernel fats are similar to that of cocoa and shea butters, suggesting use as replacements in cosmetics, foods and confections. There is, however, the need to better understand their nutritional status and effects on health.

Echinacea, a herbaceous, perennial flowering plant from the Compositae (Asteraceae) family, exhibits stimulating effects on myeloid progenitors (CFU-GMs) in rat bone marrow, as demonstrated in our previous study using a 75% (v v−1) ethanol extract of aerial parts. Expanding on this work, we have investigated eleven different Echinacea samples that belong to three species for their myeloid progenitor-stimulating activity. Simultaneously, we employed 1H-NMR spectroscopy (400 MHz, 0.02–10.02 ppm) and chemometric analysis to predict constituents responsible for activity. Female Sprague–Dawley rats received oral doses of ethanol extracts (0–200 mg extract dry weight kg−1 body weight) of Echinacea for seven days. Bone marrow cells were then cultured with CFU-GM growth factors. Extracts showing a statistically significant (p < 0.05) increase in CFU-GM, compared to the control, were considered active. Significant CFU-GM increases were observed in rats treated with seven Echinacea samples, ranging from 39% to 91% higher than the control, while four samples were inactive. All five Echinacea purpurea samples showed myeloid progenitor-stimulating activity, while one sample each of Echinacea angustifolia and Echinacea pallida also exhibited the same activity. By applying orthogonal partial least squares discriminant analysis (OPLS-DA) to the 1H-NMR spectra, we identified specific spectral bins (0.70–1.98 ppm aliphatic and 6.38–7.76 ppm aromatic) correlating with myelopoiesis stimulation. These findings highlight the potential of chemometric analysis using 1H-NMR spectroscopy to infer the chemical classes that could be responsible for the bioactive properties of complex herbal mixtures, like Echinacea.

We report two cases of pancytopenia in patients after recovering from a mild COVID-19, now presenting as paroxysmal nocturnal hemoglobinuria (PNH) and aplastic anemia. These cases illustrate a common pathway whereby a viral trigger causes the clonal expansion of a hematological disorder. Although the association of both cases with COVID-19 is temporal and COVID-19 may be an incidental diagnosis, the growing evidence related to the hematological effects of SARS-CoV-2 infection highlights the need for further investigation into the hematological consequences of COVID-19, particularly in the post-pandemic era.

An imperative requisite of tissue-engineered scaffolds is to promote host cell regeneration and concomitantly thwart microbial growth. Antibacterial agents are often added to prevent implant-related infections, which, however, aggravates the risk of bacterial resistance. For the first time, we report a fiber-based platform that selectively promotes the growth of mammalian cells and alleviates bacteria by varying fiber size, orientation, and material of polymeric yarns. The interactions of Gram-positive and -negative bacterial species with mammalian mesenchymal stem cells (MSC) were investigated on poly-€-caprolactone (PCL) yarns, polyethylene terephthalate (PET), poly-L-lactic acid (PLLA), and cotton. Various yarn configurations were studied by altering the fiber diameter (from nano- to microscale) and fiber orientations (aligned, twisted, and random) of PCL yarns. PCL nanofibrous yarn decreased the adhesion of S. aureus and E. coli, with a 2.7-fold and 1.5-fold reduction, respectively, compared to PCL microfibrous yarn. Among different fiber orientations, nanoaligned fibers resulted in an 8-fold and 30-fold reduction of S. aureus and E. coli adhesion compared to random fibers. Moreover, aligned orientation was superior in retarding the S. aureus adhesion by 14-fold compared to nanotwisted fibers. Our data demonstrate that polymeric yarns comprising fibers with nanoscale features and aligned orientation promote mammalian cell adhesion and spreading and concomitantly mitigate bacterial interaction. Moreover, we unveil the wicking of cells through polymeric yarns, facilitating early cell adhesion in fibrous scaffolds. Overall, this study provides insight to engineer scaffolds that couple superior interaction of mammalian cells with high-strength fibrous yarns for regenerative applications devoid of antibacterial agents or other surface modification strategies.

The COVID-19 pandemic due to the SARS-CoV-2 coronavirus showed acute and prolonged effects on human health. In addition, over the past four years, there has been a tremendous surge in COVID-19-related scientific publications, as shown by bibliometric and scientometric studies. However, such analysis of the scientific literature is lacking in the area of male reproduction. The current scientometric study analyzes publication characteristics of articles related to male reproduction and COVID-19 infection. We used the Scopus database to analyze scientometric data (the number of publications, journals, countries, type of documents, and subject area) related to COVID-19 and male reproductive research. Our literature search identified 345 articles related to COVID-19 and male reproductive research. Most of the articles were published in the USA (n = 72), Italy (n = 55), and China (n = 51). Such research was mainly focused around medicine (57.1%), followed by biochemistry, genetics, and molecular biology (25.7%). Also, in the area of male reproduction, only 37.1% (n = 128) of the articles contributed towards original research, whereas 52.8% (n = 182) were review articles and editorials focusing more on sexual dysfunction than infertility. Such a small number of studies published on COVID-19-related effects on male reproduction warrants a significant increase in research, which is required to decipher the mechanism(s) underlying SARS-CoV-2 infection-associated impairment of male reproductive function.

Herein, we evaluate the conversion efficiency of dye-sensitized solar cells (DSSCs) photosensitized using two different natural dyes extracted from Alpinia purpurata and Alstroemeria flower petals. The appreciable absorption capacity of the extracts in the visible light region was examined through absorption spectroscopy. The functional groups of the corresponding pigments were identified through Fourier transform spectroscopy (FTIR) technique thus indicating the presence of cyanidin 3-glycosides and piperine in the flowers of Alstroemeria and Alpinia purpurata. The extracted dyes were immobilized on TiO2 on transparent conducting FTO glass, which were used as photoanode. The dye-coated TiO2 photoanode, pt photocathode and iodide/triiodide redox electrolyte assembled into a cell module was illuminated by a light source intensity 100 mW/cm2 to measure the photovoltaic conversion efficiency of DSSCs. The TiO2 anode and Pt counter electrode surface roughness and morphological studies were evaluated using atomic force microscope (AFM) and field emission scanning electron microscopy (FESEM), respectively. Through the photoelectric characterizations, it was promising to verify that the solar conversion efficiency was calculated with the photovoltaic cell sensitized by Alstroemeria and Alpinia purpurata. This was achieved with a yield (η) of 1.74% and 0.65%, with an open-circuit voltage (Voc) of 0.39 and 0.53 V, short-circuit current density (Jsc) of 2.04 and 0.49 mA/cm2, fill factor (FF) of 0.35 and 0.40, and Pmax of 0.280 and 0.100 mW/cm2, respectively. The results are promising and demonstrate the importance of the search for new natural dyes to be used in organic solar cells for the development of devices that generate electricity in a sustainable way.

Mechanical structural systems are subject to multiple dynamic disturbances during service. While several possible scenarios can be examined to determine their design loading conditions, only a relatively small set of such scenarios is considered critical. Therefore, only such particular deterministic set of critical load cases is commonly employed for the structural design and optimization. Nevertheless, during the design and optimization stages, the mass and stiffness distributions of such assemblies vary, and, in consequence, their dynamic response also varies. Thus, it is important to consider the variations in the dynamic loading conditions during the design-and-optimization cycles. This paper studies the modal participation factors at length and proposes an alternative to the current point-wise treatment of the dynamic equations of motion of flexible bodies during design optimization. First, the most relevant-to-structural-dynamics definitions available in the literature are reviewed in depth. Second, the analysis of those definitions that have the potential to be adopted as point-wise constraint equations during structural optimization is extended. Finally, a proof of concept is presented to demonstrate the usability of each definition, followed by a case study in which the potential advantages of the proposed extended analysis are shown.

The manufacturing of copper oxide (CuO) nanoparticles has been accomplished utilizing a green technique that relies on biologically reliable mechanisms. Aqueous solutions of copper nitrate and Ixora Coccinea leaf extract are used in an environmentally safe process for creating CuO nanoparticles. The characterization of the synthesized CuO nanoparticles involves the utilization of techniques such as X-ray diffractometry (XRD), scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FTIR), and thermogravimetricanalysis (TGA). CuO nanoparticles are confirmed by XRD and FTIR peak results. When the particles are measured, they range between 93.75 nm and 98.16 nm, respectively. The produced CuO nanoparticles are used to prepare the nanofluid. While conventional water exhibits a 3 °C temperature difference, nanofluid achieves a considerable temperature differenceof 7 °C. As a result, it is clear that the nanofluid performs better at dispersing heat into the environment. The experiment’s overall findings support the possibility of ecologically friendly, green-synthesized CuO nanoparticle-induced nanofluid as an effective heattransfer fluid that can be applied to heattransfer systems.

The objectives of this study were to (1) investigate the association between human blood type and COVID-19 in both inpatient and longitudinal populations and (2) identify the association between blood type and severity of COVID-19 via presence of cellular biomarkers of severe infection in hospitalized individuals at our institution in South Florida. This study consisted of (1) a single-center retrospective analysis of 669 out of 2741 COVID-19-positive, screened patients seen from 1 January 2020 until 31 March 2021 at the University of Miami Emergency Department (ED) who tested positive for COVID-19 and had a documented ABO blood type and (2) a longitudinal SARS-CoV-2 immunity study (“CITY”) at the University of Miami Miller School of Medicine, consisting of 185 survey participants. In an inpatient setting, blood type appeared to be associated with COVID-19 severity and mortality. Blood type O sustained less risk of COVID-19 mortality, and blood type O- demonstrated less risk of developing COVID-19 pneumonia. Inpatients with O- blood type exhibited less biomarkers of severe infection than did other blood types. In a longitudinal setting, there was no association found between blood type and SARS-CoV-2 infection.

Motivated by the saturation of drag reduction effectiveness at high non-dimensional riblet spacing in turbulent boundary layer flows, this study seeks to investigate the influence of a secondary blade riblet structure on flow statistics and friction drag reduction effectiveness in comparison to the widely explored single-scale blade riblet surface. The turbulent flow dynamics and drag reduction performance over single- and multi-scale blade riblet surfaces were experimentally examined in a flow visualization channel across various non-dimensional riblet spacings. The shear velocity was quantified by the streamwise velocity distributions from the logarithmic layer via planar Particle Image Velocimetry (PIV) measurements, whereas the near-wall flow dynamics were characterized by a Micro Particle Image Velocimetry (micro-PIV) system. The results highlighted that although both riblet surfaces exhibited similar drag reduction performances at low non-dimensional riblet spacings, the presence of a secondary riblet blade structure can effectively extend the drag reduction region with the non-dimensional riblet spacing up to 32 and achieve approximately 10% lower friction drag in comparison to the single-scale riblet surface when the non-dimensional riblet spacing increases to 44.2. The average number of uniform momentum zones (UMZs) on the multi-scaled blade riblet has also reduced by 9% compared to the single-scaled riblet which indicates the reduction of strong shear layers within a turbulent boundary layer. The inspection of near-wall flow statistics demonstrated that at high non-dimensional riblet spacings, the multi-scale riblet surface produces reduced wall-normal velocity fluctuations and Reynolds shear stresses. Quadrant analysis revealed that this design allows for the suppression of both the sweep and ejection events. This experimental result demonstrated that surfaces with spanwise variations of riblet heights have the potential to maintain drag reduction effectiveness across a wider range of flow speeds.

Node localization is critical for accessing diverse nodes that provide services in remote places. Single-anchor localization techniques suffer co-linearity, performing poorly. The reliable multiple anchor node selection method is computationally intensive and requires a lot of processing power and time to identify suitable anchor nodes. Node localization in wireless sensor networks (WSNs) is challenging due to the number and placement of anchors, as well as their communication capabilities. These senor nodes possess limited energy resources, which is a big concern in localization. In addition to convention optimization in WSNs, researchers have employed nature-inspired algorithms to localize unknown nodes in WSN. However, these methods take longer, require lots of processing power, and have higher localization error, with a greater number of beacon nodes and sensitivity to parameter selection affecting localization. This research employed a nature-inspired crow search algorithm (an improvement over other nature-inspired algorithms) for selecting the suitable number of anchor nodes from the population, reducing errors in localizing unknown nodes. Additionally, the weighted centroid method was proposed for identifying the exact location of an unknown node. This made the crow search weighted centroid localization (CS-WCL) algorithm a more trustworthy and efficient method for node localization in WSNs, with reduced average localization error (ALE) and energy consumption. CS-WCL outperformed WCL and distance vector (DV)-Hop, with a reduced ALE of 15% (from 32%) and varying communication radii from 20 m to 45 m. Also, the ALE against scalability was validated for CS-WCL against WCL and DV-Hop for a varying number of beacon nodes (from 3 to 2), reducing ALE to 2.59% (from 28.75%). Lastly, CS-WCL resulted in reduced energy consumption (from 120 mJ to 45 mJ) for varying network nodes from 30 to 300 against WCL and DV-Hop. Thus, CS-WCL outperformed other nature-inspired algorithms in node localization. These have been validated using MATLAB 2022b.

Extensive hydrologic and water quality modeling within a watershed benefits from long-term flow and nutrient data sets for appropriate model calibration and validation. However, due to a lack of local water quality data, simpler water quality modeling techniques are generally adopted. In this study, the monitoring sites were established at two different locations to collect hydraulic data for the hydraulic calibration and validation of the model. In addition, water quality samples were collected at eight monitoring sites and analyzed in the lab for various parameters for calibration. This includes total suspended solids (TSS), soluble phosphorus, five-day biochemical oxygen demand (BOD5), and dissolved oxygen (DO). The Personal Computer Storm Water Management Model (PCSWMM) 7.6 software was used to simulate all the pollutant loads using event mean concentrations (EMCs). The performance of the model for streamflow calibration at the two USGS gauging stations was satisfactory, with Nash–Sutcliffe Efficiency (NSE) values ranging from 0.51 to 0.54 and coefficients of determination (R2) ranging from 0.71 to 0.72. The model was also validated with the help of historical flow data with NSE values ranging from 0.5 to 0.79, and R2 values ranging from 0.6 to 0.95. The hydraulic calibration also showed acceptable results with reasonable NSE and R2 values. The water quality data recorded at the monitoring stations were then compared with the simulated water quality modeling results. The model reasonably simulated the water quality, which was evaluated through visual inspection using a scatter plot. Our analysis showed that the upstream tributaries, particularly from agricultural areas, were contributing more pollutants than the downstream tributaries. Overall, this study demonstrates that the PCSWMM, which was typically used for modeling urban watersheds, could also be used for modeling larger mixed land use watersheds with reasonable accuracy.

Laser machining by ultra-short (sub-ps) pulses at high intensity offers high precision, high throughput in terms of area or volume per unit time, and flexibility to adapt processing protocols to different materials on the same workpiece. Here, we consider the challenge of optimization for high throughput: how to use the maximum available laser power and larger focal spots for larger ablation volumes by implementing a fast scan. This implies the use of high-intensity pulses approaching ∼PW/cm2 at the threshold where tunneling ionization starts to contribute to overall ionization. A custom laser micromachining setup was developed and built to enable high speed, large-area processing, and easy system reconfiguration for different tasks. The main components include the laser, stages, scanners, control system, and software. Machining of metals such as Cu, Al, or stainless steel and fused silica surfaces at high fluence and high exposure doses at high scan speeds up to 3 m/s were tested for the fluence scaling of ablation volume, which was found to be linear. The largest material removal rate was 10 mm3/min for Cu and 20 mm3/min for Al at the maximum power 80 W (25 J/cm2 per pulse). Modified surfaces are color-classified for their appearance, which is dependent on surface roughness and chemical modification. Such color-coding can be used as a feedback parameter for industrial process control.

This study presents a GIS-based Multi-Criteria Decision Analysis (MCDA) spatial model to assess land suitability for oil palm (OP) cultivation in rainfed conditions. Initially, twelve parameters, viz., rainfall, number of rainy days, mean temperature, RH, ground water level, soil pH, salinity, soil depth, surface texture, stoniness, slope, and drainage, were selected for assessing OP suitability in one of the states (Kerala). However, subsequent ground verification revealed significant discrepancies, which prompted refining the model by focusing on key parameters with greater accuracy and relevance. Accordingly, only five the most critical parameters affecting OP cultivation under rainfed conditions were selected through the rank sum method, and weights were assigned ac-cording to their significance. This study was aimed at creating a comprehensive tool for informed decision making in agricultural planning. District-level spatial data from reliable sources were utilized for Multi-Criteria Decision Analysis. Thematic rasters, representing key factors influencing land suitability, were created in a GIS. Utilizing MCDA techniques, a digital suitability map was generated in ArcGIS 10.3, delineating three distinct classes over an extensive area of 10.5 million hectares. Further, with an aim to focus on actual locations that can be readily planted with oil palm, the suitable locations identified were restricted to eight selected land use/land cover (LULC) classes. This strategic limitation aimed to facilitate the expansion of OP cultivation exclusively to areas deemed most suitable based on the identified criteria. The validation of this developed model involved comparing the suitability map generated with the performance of existing oil palm plantations across diverse locations. The reasonable similarity between the model’s predictions and real-world plantation outcomes validated the effectiveness of this MCDA spatial model. This model not only helps identify suitable locations for rainfed oil palm cultivation but also serves as a valuable tool for strategic decision making in agricultural land use planning.

Cutting tool coolant channels play a pivotal role in machining processes, facilitating the efficient supply of cooling agents to high-stress areas and effective heat dissipation. Achieving optimal cooling at the tool’s cutting-edge is essential for enhancing production processes. Experimental investigations into tribological stress analysis can be limited in accessing complex tool–workpiece contact zones, prompting the use of numerical modelling to explore fluid dynamics and tribology. In this study, the coolant flow dynamics and heat dissipation in drilling operations were comprehensively investigated through computational fluid dynamics (CFD) modelling. Four twist drill models with varying coolant channel arrangements were studied: standard model drill, standard model drill with notch, profile model drill, and profile model drill with notch. Two distinct approaches are applied to the coolant inlet to assess the impact of operating conditions on fluid flow and heat dissipation at the cutting-edge. The findings emphasize that cutting-edge zones have insufficient coolant supply, particularly in modified drill models such as the standard model drill with notch and profile model drills with and without notch. Moreover, enhanced coolant supply at the cutting-edge is achieved under high-pressure inlet conditions. The standard model drill with a notch exhibited exceptional performance in reducing thermal load, facilitating efficient coolant escape to the flute for improved heat dissipation at the cutting-edge. Despite challenges like dead zones in profile models, the standard-with-notch model yielded the most promising results. Further analyses under constant pressure conditions at 40 and 60 bar exhibited enhanced fluid flow rates, particularly at the cutting-edge, leading to improved heat dissipation. The temperature distribution along the cutting-edge and outer corner demonstrated a decrease as the pressure increased. This study underscores the critical role of both coolant channel design and inlet pressure in optimizing coolant flow dynamics and heat transfer during drilling operations. The findings provide valuable insights for designing and enhancing coolant systems in machining processes, emphasizing the significance of not only coolant channel geometry but also inlet pressure for effective heat dissipation and enhanced tool performance.

Human milk reduces risk for necrotizing enterocolitis in preterm infants. Necrotizing enterocolitis occurs in the ileocecal region where thousands of milk protein-derived peptides have been released from digestion. Digestion-released peptides may exert bioactivity, such as antimicrobial and immunomodulatory activities, in the gut. In this study, we applied mass spectrometry-based peptidomics to characterize peptides present in colostrum before and after in vitro digestion. Sequence-based computational modeling was applied to predict peptides with antimicrobial activity. We identified more peptides in undigested samples, yet the abundances were much higher in the digested samples. Heatmapping demonstrated highly different peptide profiles between undigested and digested samples. Four peptides (αS1-casein [157–163], αS1-casein [157–165], β-casein [153–159] and plasminogen [591–597]) were selected, synthesized and tested against common pathogenic bacteria associated with necrotizing enterocolitis. All four exhibited bacteriostatic, though not bactericidal, activities against Klebsiella aerogenes, Citrobacter freundii and Serratia marcescens, but not Escherichia coli.

The study highlights the importance of integrating organic resources, such as vermicompost and biofertilizers with inorganic fertilizers to sustain coconut yields and manage costs. The experimental trial was conducted from 2016 to 2020 in a 47-year-old East Coast Tall coconut garden at Coconut Research Station in South India. The research evaluated four nutrient management treatments viz., T1 (75% Recommended Dose of Fertilizer (RDF) + 25% N organic), T2 (50% RDF + 50% N organic), T3 (100% N organic) and a control (100% RDF). Intercrops included black pepper (Piper nigrum), banana (Musa acuminata) and cocoa (Theobroma cacao). Organic manure significantly improved soil physical properties, water retention and overall soil health. The T2 treatment achieved the highest yields for coconut, cocoa, banana and black pepper. The added soil health parameters supported to these findings, with T2 showing the highest fungal (15.27 × 103 cfu/g of soil) and bacterial populations (17.25 × 105 cfu/g of soil), along with a significant earthworm population (26/m2), indicating enhanced soil ecosystem health. Additionally, soil moisture content was highest under T3 (100% organic) across various depths, followed by T2, highlighting the critical role of organic matter in improving soil moisture conservation. The economic feasibility analysis, including a net present value (NPV), benefit/cost ratio (B/C ratio) and an internal rate of return (IRR), revealed T2 to be the most economically viable nutrient management strategy. This study highlights the economic benefits of intercropping coconuts using an integrated nutrient management (INM) approach, demonstrating its superiority over traditional monocropping practices.

Synchronous fluorescence spectroscopy (SFS) is a technique that involves the simultaneous detection of fluorescence excitation and emission at a constant wavelength difference. The spectrum yields bands that are narrower and less complex than the original excitation and emission bands. The SFS bands correspond uniquely to the fluorescing molecule. Our investigation focuses on evaluating the sensitivity of the SFS technique for the detection and quantitation of PAHs relevant to astrochemistry. Results are presented for naphthalene, anthracene, and pyrene in three different solvents: n-hexane, water, and ethanol. SF bands are obtained with a constant wavelength difference between the peak excitation and emission wavelength (Δλ = λex − λem) at a concentration ranging from 10−4 to 10−10 M. Limit of detection (LOD) and limit of quantitation (LOQ) calculations are based on integrated SF band areas at different concentrations. Spectra of 23 pg/g of anthracene, 16 pg/g, and 2.6 pg/g of pyrene are recorded using ethanol as the solvent. The PAHs exhibit detection limits in the fractions of parts-per-billion (ng/g) range. Through comparison with similar prior studies employing fluorescence emission, our findings reveal a better detectability limit, demonstrating the effectiveness and applicability of the SFS technique.

This study focuses on the development and characterization of an intranasal vaccine platform using adjuvanted nanoparticulate delivery of swine influenza A virus (SwIAV). The vaccine employed whole inactivated H1N2 SwIAV as an antigen and STING-agonist ADU-S100 as an adjuvant, with both surface adsorbed or encapsulated in mannose–chitosan nanoparticles (mChit-NPs). Optimization of mChit-NPs included evaluating size, zeta potential, and cytotoxicity, with a 1:9 mass ratio of antigen to NP demonstrating high loading efficacy and non-cytotoxic properties suitable for intranasal vaccination. In a heterologous H1N1 pig challenge trial, the mChit-NP intranasal vaccine induced cross-reactive sIgA antibodies in the respiratory tract, surpassing those of a commercial SwIAV vaccine. The encapsulated mChit-NP vaccine induced high virus-specific neutralizing antibody and robust cellular immune responses, while the adsorbed vaccine elicited specific high IgG and hemagglutinin inhibition antibodies. Importantly, both the mChit-NP vaccines reduced challenge heterologous viral replication in the nasal cavity higher than commercial swine influenza vaccine. In summary, a novel intranasal mChit-NP vaccine platform activated both the arms of the immune system and is a significant advancement in swine influenza vaccine design, demonstrating its potential effectiveness for pig immunization.

The transition from internal combustion engine vehicles to electric vehicles (EVs) is gaining momentum due to their significant environmental and economic benefits. This study addresses the challenges of integrating renewable energy sources, particularly solar power, into EV charging infrastructures by using deep learning models to predict photovoltaic (PV) power generation and EV charging demand. The study determines the optimal battery energy storage capacity and charging schedule based on the prediction result and actual data. A dataset of a 15 kWp rooftop PV system and simulated EV charging data are used. The results show that simple RNNs are most effective at predicting PV power due to their adept handling of simple patterns, while bidirectional LSTMs excel at predicting EV charging demand by capturing complex dynamics. The study also identifies an optimal battery storage capacity that will balance the use of the grid and surplus solar power through strategic charging scheduling, thereby improving the sustainability and efficiency of solar energy in EV charging infrastructures. This research highlights the potential for integrating renewable energy sources with advanced energy storage solutions to support the growing electric vehicle infrastructure.

Beira Lake, located in Colombo, Sri Lanka, has suffered severe anthropogenic impacts, with previous restoration attempts failing due to a limited understanding of pollutant dynamics. Aiming to fill this gap, a comprehensive study was conducted during dry and wet seasons to assess the spatiotemporal water pollution of Beira Lake, employing key physicochemical parameters, numerical indices, and remote sensing analysis. The water pollution index (WPI) results categorize Beira Lake as highly polluted, with WPI values ranging from 2.38 ± 0.92 in the wet season to 2.53 ± 1.32 in the dry season. Comparatively higher COD levels recorded in the Beira Lake network, especially for Gangarama Lake show significant pollution levels during both the dry and wet seasons, e.g., the highest COD levels, at 306.40 mg/L, were observed during the wet season. The Trophic State Index (TSI) results indicate eutrophic and hypereutrophic conditions in Beira Lake, which are particularly pronounced during the wet season. The heavy metal pollution index (HPI) results suggest elevated heavy metal concentrations in Beira Lake, especially in the wet season. Combined with field investigation results, a remote sensing data analysis between 2016 and 2023 reveals significant improvements in water transparency, suggesting positive effects of recent management interventions. Parameters demanding attention include COD, nitrate, and total phosphate levels due to their consistent exceedance of permissible limits. The PCA results of indices correlations between wet and dry seasons offer valuable insights into the complex dynamics of Beira Lake’s water quality. The study makes recommendations for restoring Beira Lake, including stringent pollution controls, regular dredging, green infrastructure implementation, implementing new rules and regulations, and community engagement.

False positive reactive plasmin reagin (RPR) reactivity following a COVID-19 vaccine has been reported, and it is therefore conceivable that individuals who receive frequent coronavirus disease 2019 (COVID-19) vaccinations may exhibit durable RPR responses. Here, we sought to investigate the extent to which repeated mRNA COVID-19 vaccines can elicit chronic false RPR reactivity in a longitudinal cohort. Participants (n = 119) in an IRB-approved (#20201026), longitudinal SARS-CoV-2 cohort study were screened for RPR reactivity via manual RPR card assays. Samples with reactive results underwent additional testing, including follow-on RPR screening at additional timepoints, confirmatory fluorescent treponemal antibody (FTA-ABS) testing and anti-nuclear antibody (ANA) testing. Medical histories were collected. We observed (n = 2) screen-positive RPR results (1.7% [2/119]) following booster vaccination, for which two individuals exhibited chronic, vaccine-induced RPR reactivity for up to 9 months following booster vaccination. Both participants were ANA-negative. It is imperative for clinicians to be mindful of the potential immunologic interference of COVID-19 vaccines with standard infectious disease assays, including RPR testing. Detailed medical histories and clinical contexts, including recent vaccination, should be reviewed prior to proceeding with distressing and invasive workups.

In this paper, the electrochemical non-enzymatic detection of Riboflavin (RF) was proposed based on its catalytic reduction in a Thionine-coated Cadmium Selenide Quantum dots (TH@CdSe QDs)-modified paraffin wax-impregnated graphite electrode (PIGE) that was prepared using a novel approach. The synthesized TH@CdSe QDs were confirmed by UV-Vis spectroscopy, Confocal Raman Microscopy and High Resolution Transmission Electron Microscopy (HRTEM) studies. The electrochemical response of the TH@CdSe QDs-modified PIGE was studied by cyclic voltammetry. The voltammetric response of RF at the TH@CdSe QDs-modified PIGE showed higher current than the bare PIGE. Under optimum conditions, the electrocatalytic reduction currents of RF was found to be linearly related to its concentration over the range of 1.6 × 10−7 M to 1.4 × 10−4 M with a detection limit of 53 × 10−9 M (S/N = 3). The TH@CdSe QDs-modified PIGE was utilized as an amperometric sensor for the detection of RF in flow systems was performed by carrying out hydrodynamic and chronoamperometric experiments. The TH@CdSe QDs-modified PIGE showed very good stability and a longer shelf life. The applicability of the fabricated electrode was justified by the quantification of RF in commercial tablets.

Zinc oxide nanomaterials have been the cynosure of this decade because of their immense potential in different biomedical applications. It includes their usage in the prognosis and treatment of different infectious and cellular diseases, owing to their peculiar physiochemical properties such as variable shape, size, and surface charge etc. Increasing demand and usage of the ZnO nanomaterials raise concerns about their cellular and molecular toxicity and their biocompatibility with human cells. This review comprehensively details their physiochemical properties for usage in biomedical applications. Furthermore, the toxicological concerns of ZnO nanomaterials with different types of cellular systems have been reviewed. Moreover, the biomedical and biocompatible efficacy of ZnO nanomaterials for cancer specific pathways has been discussed. This review offers insights into the current scenario of ZnO nanomaterials usage and signifies their potential future extension usage on different types of biomedical and environmental applications.

The aging population and increasing incidence of trauma among younger age groups have heightened the increasing demand for reliable implant materials. Effective implant materials must demonstrate rapid osseointegration and strong antibacterial properties to ensure optimal patient outcomes and decrease the chance of implant rejection. This study aims to enhance the bone–implant interface by utilizing 45S5 bioglass modified with various concentrations of Fe3O4 as a coating material. The effect of the insertion of Fe3O4 into the bioglass structure was studied using Raman spectroscopy which shows that with the increase in Fe3O4 concentration, new vibration bands associated with Fe-related structural units appeared within the sample. The bioactivity of the prepared glasses was evaluated using immersion tests in simulated body fluid, revealing the formation of a calcium phosphate-rich layer within 24 h on the samples, indicating their potential for enhanced tissue integration. However, the sample modified with 8 mol% of Fe3O4 showed low reactivity, developing a calcium phosphate-rich layer within 96 h. All the bioglasses showed antibacterial activity against the Gram-positive and Gram-negative bacteria. The modified bioglass did not present significant antibacterial properties compared to the bioglass base.

This paper presents the results of adsorption and permeation experiments of hydrogen and methane at elevated temperatures on a carbon-based nanoporous membrane material provided by Fraunhofer IKTS. The adsorption of pure components was measured between 90 °C and 120°C and pressures up to 45 bar. The Langmuir adsorption isotherm shows the best fit for all data points. Compared to available adsorption isotherms of H2 and CH4 on carbon, the adsorption on the investigated nanoporous carbon structures is significantly lower. Single-component permeation experiments were conducted on membranes at temperatures up to 220 °C. After combining the experimental results with a Maxwell–Stefan surface diffusion model, Maxwell–Stefan surface diffusion coefficients Dis were calculated. The calculated values are in line with an empirical model and thus can be used in future multi-component modeling approaches in order to better analyze and design a membrane system. The published adsorption data fill a gap in the available adsorption data for CH4 and H2.

The management of early-stage hepatocellular carcinoma (HCC) is complex, with multiple treatment strategies available. There is a paucity of literature regarding variations in the patterns of care and outcomes between transplant and non-transplant centres. We conducted this real-world multi-centre cohort study in two liver cancer referral centres with an integrated liver transplant program and an additional eight non-transplant HCC referral centres across Australia to identify variation in patterns of care and key survival outcomes. Patients with stage Barcelona Clinic Liver Cancer (BCLC) 0/A HCC, first diagnosed between 1 January 2016 and 31 December 2020, who were managed at a participating site, were included in the study. Patients were excluded if they had a history of prior HCC or if they received upfront liver transplantation. A total of 887 patients were included in the study, with 433 patients managed at a liver cancer centre with a transplant program (LTC) and 454 patients managed at a non-transplant centre (NTC). Management at an LTC did not significantly predict allocation to resection (adjusted OR 0.75, 95% CI 0.50 to 1.11, p = 0.148). However, in those not receiving resection, LTC and NTC patients were systematically managed differently, with LTC patients five times less likely to receive upfront ablation than NTC patients (adjusted OR 0.19, 95% CI 0.13 to 0.28, p < 0.001), even after adjusting for tumour burden, as well as for age, gender, liver disease aetiology, liver disease severity, and medical comorbidities. LTCs exhibited significantly higher proportions of patients undergoing TACE for every tumour burden category, including those with a single tumour measuring 2 cm or less (p < 0.001). Using multivariable Cox proportional hazards analysis, management at a transplant centre was associated with reduced all-cause mortality (adjusted HR 0.71, 95% CI 0.51 to 0.98, p = 0.036), and competing-risk regression analysis, considering liver transplant as a competing event, demonstrated a similar reduction in risk (adjusted HR 0.70, 95% CI 0.50 to 0.99, p = 0.041), suggesting that the reduced risk of death is not fully explained by higher rates of transplantation. Our study highlights systematic differences in HCC care between large volume liver transplant centres and other sites, which has not previously been well-described. Further work is needed to better define the reasons for differences in treatment allocation and to aim to minimise unwarranted treatment variation to maximise patient outcomes across Australia.

With additive manufacturing (AM) processes such as Wire Arc Additive Manufacturing (WAAM), components with complex shapes or with functional properties can be produced, with advantages in the areas of resource conservation, lightweight construction, and load-optimized production. However, proving component quality is a challenge because it is not possible to produce 100% defect-free components. In addition to this, statistically determined fluctuations in the wire quality, gas flow, and their interaction with process parameters result in a quality of the components that is not 100% reproducible. Complex testing procedures are therefore required to demonstrate the quality of the components, which are not cost-effective and lead to less efficiency. As part of the project “3DPrintFEM”, a sound emission analysis is used to evaluate the quality of AM components. Within the scope of the project, an approach was being developed to determine the quality of an AM part dependent not necessarily on its geometry. Samples were produced from WAAM, which were later cut and milled to precision. To determine the frequencies, the samples were put through a resonant frequency test (RFM). The unwanted modes were then removed from the spectrum produced by the experiments by comparing it with FEM simulations. Later, defects were introduced in experimental samples in compliance with the ISO 5817 guidelines. In order to create a database of frequencies related to the degree of the sample defect, they were subjected to RFM. The database was further augmented through frequencies from simulations performed on samples with similar geometries, and, hence, a training set was generated for an algorithm. A machine-learning algorithm based on regression modelling was trained based on the database to sort samples according to the degree of flaws in them. The algorithm’s detectability was evaluated using samples that had a known level of flaws which forms the test dataset. Based on the outcome, the algorithm will be integrated into an equipment developed in-house to monitor the quality of samples produced, thereby having an in-house quality assessment routine. The equipment shall be less expensive than conventional acoustic equipment, thus helping the industry cut costs when validating the quality of their components.

The alkaline oxygen evolution reaction (OER) remains a bottleneck in green hydrogen production owing to its slow reaction kinetics and low catalytic efficiencies of earth abundant electrocatalysts in the alkaline OER reaction. This study investigates the OER performance of hierarchically porous cobalt electrocatalysts synthesized using the dynamic hydrogen bubble templating (DHBT) method. Characterization studies revealed that electrocatalysts synthesized under optimized conditions using the DHBT method consisted of cobalt nanosheets, and hierarchical porosity with macropores distributed in a honeycomb network and mesopores distributed between cobalt nanosheets. Moreover, X-ray photoelectron spectroscopy studies revealed the presence of Co(OH)2 as the predominant surface cobalt species while Raman studies revealed the presence of the cubic Co3O4 phase in the synthesized electrocatalysts. The best performing electrocatalyst required only 360 mV of overpotential to initiate a current density of 10 mA cm−2, exhibited a Tafel slope of 37 mV dec−1, and stable OER activity over 24 h. The DHBT method offers a facile, low cost and rapid synthesis approach for preparation for highly efficient cobalt electrocatalysts.

This study investigates the impact of the Energy-related Uncertainty Index (EUI) on corporate investment among Chinese non-financial listed companies, focusing on two aspects: the effect of EUI fluctuations on investment behavior, and its differential impact on energy versus non-energy sectors. Utilizing a dataset of 2487 firms from 2007 to 2022, encompassing 22,346 firm-year observations, our analysis reveals that a 1% increase in the EUI leads to a 0.045% decrease in overall corporate investment. Notably, this effect is more pronounced in energy-related firms, where a 1% increase in EUI leads to a 0.057% reduction in investment. In comparison, non-energy-related firms exhibit a milder response, with a 1% increase in EUI resulting in a 0.026% decrease in investment. Given the average annual change in EUI over the sample period [2007–2022] of 27.710%, a 0.045% decrease in investment implies a substantial 1.246% per annum change in investment. This highlights the economically significant impact of EUI fluctuations on corporate investment decisions, particularly during periods of heightened uncertainty. These findings, validated through alternative EUI measures and investment metrics, provide crucial insights for understanding investment behavior under energy uncertainty. Conclusively, our study contributes to the literature by highlighting how energy uncertainty uniquely impacts corporate investment, taking into account the specific financial and operational conditions within different sectors. The findings highlight the importance of incorporating energy policy considerations into corporate strategic planning, particularly for energy-intensive industries within transitional economies like China.

MTX-211 is a first-in-class dual inhibitor of epidermal growth factor receptor (EGFR) and phosphoinositide-3 kinase (PI3K) signaling pathways with a compelling pharmaceutical profile and could enhance the effectiveness of mitogen-activated protein kinase kinase (MEK) inhibitor therapy in colorectal tumors with KRAS mutations. However, the specific mechanisms contributing to the acquired resistance to MTX-211 in human cancers remain elusive. Here, we discovered that the overexpression of the ATP-binding cassette (ABC) drug transporter ABCG2, a prevalent mechanism associated with multidrug resistance (MDR), could diminish the effectiveness of MTX-211 in human cancer cells. We showed that the drug efflux activity of ABCG2 substantially decreased the intracellular accumulation of MTX-211 in cancer cells. As a result, the cytotoxicity and effectiveness of MTX-211 in suppressing the activation of the EGFR and PI3K pathways were significantly attenuated in cancer cells overexpressing ABCG2. Moreover, the enhancement of the MTX-211-stimulated ATPase activity of ABCG2 and the computational molecular docking analysis illustrating the binding of MTX-211 to the substrate-binding sites of ABCG2 offered a further indication for the interaction between MTX-211 and ABCG2. In summary, our findings indicate that MTX-211 acts as a substrate for ABCG2, underscoring the involvement of ABCG2 in the emergence of resistance to MTX-211. This finding carries clinical implications and merits further exploration.

People with HIV (PWH) frequently suffer from Opioid (OP) Use Disorder (OUD). In an investigation of the impact of OUD on underlying immune dysfunction in PWH, we previously reported that OP use exacerbates inflammation in virally controlled PWH followed in the Infectious Diseases Elimination Act (IDEA) Syringe Services Program (SSP). Unexpectedly, Flu vaccination-induced antibody responses in groups with OUD were superior to PWH without OUD. Here, we investigated the profile of 48 plasma biomarkers comprised of TNF and Ig superfamily (SF) molecules known to impact interactions between T and B cells in 209 participants divided into four groups: (1) HIV+OP+, (2) HIV−OP+, (3) HIV+OP−, and (4) HIV−OP−. The differential expression of the top eight molecules ranked by median values in individual Groups 1–3 in comparison to Group 4 was highly significant. Both OP+ groups 1 and 2 had higher co-stimulatory TNF SF molecules, including 4-1BB, OX-40, CD40, CD30, and 4-1BBL, which were found to positively correlate with Flu Ab titers. In contrast, HIV+OP− exhibited a profile dominant in Ig SF molecules, including PDL-2, CTLA-4, and Perforin, with PDL-2 showing a negative correlation with Flu vaccine titers. These findings are relevant to vaccine development in the fields of HIV and OUD.

The L 1 region of bovine adenovirus (BAdV)-3 encodes a multifunctional protein named protein VII. Anti-protein VII sera detected a protein of 26 kDa in transfected or BAdV-3-infected cells, which localizes to nucleus and nucleolus of infected/transfected cells. Analysis of mutant protein VII identified four redundant overlapping nuclear/nucleolar localization signals as deletion of all four potential nuclear/nucleolar localization signals localizes protein VII predominantly to the cytoplasm. The nuclear import of protein VII appears to use importin α (α-1), importin-β (β-1) and transportin-3 nuclear transport receptors. In addition, different nuclear transport receptors also require part of protein VII outside nuclear localization sequences for efficient interaction. Proteomic analysis of protein complexes purified from recombinant BAdV-3 expressing protein VII containing Strep Tag II identified potential viral and cellular proteins interacting with protein VII. Here, we confirm that protein VII interacts with IVa2 and protein VIII in BAdV-3-infected cells. Moreover, amino acids 91–101 and 126–137, parts of non-conserved region of protein VII, are required for interaction with IVa2 and protein VIII, respectively.

Excessive fibrosis and resultant poor control of intraocular pressure (IOP) reduce the efficacy of glaucoma surgeries. Historically, corticosteroids and anti-fibrotic agents, such as mitomycin C (MMC) and 5-fluorouracil (5-FU), have been used to mitigate post-surgical fibrosis, but these have unpredictable outcomes. Therefore, there is a need to develop novel treatments which provide increased effectiveness and specificity. This review aims to provide insight into the pathophysiology behind wound healing in glaucoma surgery, as well as the current and promising future wound healing agents that are less toxic and may provide better IOP control.

Background: In planning radiotherapy treatments, computed tomography (CT) has become a crucial tool. CT scans involve exposure to ionizing radiation, which can increase the risk of cancer and other adverse health effects in patients. Ionizing radiation doses for medical exposure must be kept “As Low As Reasonably Achievable”. Very few articles on guidelines for radiotherapy-computed tomography scans are available. This paper reviews the current literature on radiation dose optimization based on the effective dose and diagnostic reference level (DRL) for head, neck, and pelvic CT procedures used in radiation therapy planning. This paper explores the strategies used to optimize radiation doses, and high-quality images for diagnosis and treatment planning. Methods: A cross-sectional study was conducted on 300 patients with head, neck, and pelvic region cancer in our institution. The DRL, effective dose, volumetric CT dose index (CTDIvol), and dose-length product (DLP) for the present and optimized protocol were calculated. DRLs were proposed for the DLP using the 75th percentile of the distribution. The DLP is a measure of the radiation dose received by a patient during a CT scan and is calculated by multiplying the CT dose index (CTDI) by the scan length. To calculate a DRL from a DLP, a large dataset of DLP values obtained from a specific imaging procedure must be collected and can be used to determine the median or 75th-percentile DLP value for each imaging procedure. Results: Significant variations were found in the DLP, CTDIvol, and effective dose when we compared both the standard protocol and the optimized protocol. Also, the optimized protocol was compared with other diagnostic and radiotherapy CT scan studies conducted by other centers. As a result, we found that our institution’s DRL was significantly low. The optimized dose protocol showed a reduction in the CTDIvol (70% and 63%), DLP (60% and 61%), and effective dose (67% and 62%) for both head, neck, and pelvic scans. Conclusions: Optimized protocol DRLs were proposed for comparison purposes.

In cholinergic urticaria (CholU), small, itchy wheals are induced by exercise or passive warming and reduced sweating has been reported. Despite the described reduced muscarinic receptor expression, sweat duct obstruction, or sweat allergy, the underlying pathomechanisms are not well understood. To gain further insights, we collected skin biopsies before and after pulse-controlled ergometry and sweat after sauna provocation from CholU patients as well as healthy controls. CholU patients displayed partially severely reduced local sweating, yet total sweat volume was unaltered. However, sweat electrolyte composition was altered, with increased K+ concentration in CholU patients. Formalin-fixed, paraffin-embedded biopsies were stained to explore sweat leakage and tight junction protein expression. Dermcidin staining was not found outside the sweat glands. In the secretory coils of sweat glands, the distribution of claudin-3 and -10b as well as occludin was altered, but the zonula occludens-1 location was unchanged. In all, dermcidin and tight junction protein staining suggests an intact barrier with reduced sweat production capability in CholU patients. For future studies, an ex vivo skin model for quantification of sweat secretion was established, in which sweat secretion could be pharmacologically stimulated or blocked. This ex vivo model will be used to further investigate sweat gland function in CholU patients and decipher the underlying pathomechanism(s).

Groundwater is an important drinking water resource in the coastal regions of island countries and has suffered from heavy seawater intrusion. However, the areas specifically affected by seawater intrusion and their groundwater hydrogeochemical compositions and evolution processes remain unclear. This study analyzed the hydrogeochemical compositions, water quality, and evolution processes of groundwater in the Mannar district, Sri Lanka, during the dry season. A total of 56 samples were collected from shallow wells and tube wells across the region, and about 64.28% of groundwater samples had good quality (WQI < 100). Geochemical compositions and water quality parameters had a high level in the north and south mainland regions, where they severely suffered from seawater intrusion with a high content of Cl− and Na+. The geochemical compositions of groundwater in the Mannar district were predominantly affected by rock weathering and/or evaporation processes. Cl-Na and HCO3-Ca facies were the main hydrochemical types, and the corresponding ions were mainly from silicate and halite dissolution. The reverse cation exchange process mainly occurred in seawater intrusion areas. The study highlights the impacts of seawater intrusion on the hydrogeochemical compositions and evolution processes in Mannar region groundwater, which will enhance the understanding of the local water quality and seawater intrusion situation and aid in protecting drinking water safety by routinely monitoring the groundwater quality and implementing targeted desalination techniques in the key areas.

Zika virus (ZIKV) remains a public health concern, with epidemics in endemic regions and sporadic outbreaks in new areas posing significant threats. Several mosquito-borne flaviviruses that can cause human illness, including West Nile, Usutu, and St. Louis encephalitis, have associations with birds. However, the susceptibility of chickens to ZIKV and their role in viral epidemiology is not currently known. We investigated the susceptibility of chickens to experimental ZIKV infection using chickens ranging from 1-day-old chicks to 6-week-old birds. ZIKV caused no clinical signs in chickens of all age groups tested. Viral RNA was detected in the blood and tissues during the first 5 days post-inoculation in 1-day and 4-day-old chicks inoculated with a high viral dose, but ZIKV was undetectable in 6-week-old birds at all timepoints. Minimal antibody responses were observed in 6-week-old birds, and while present in younger chicks, they waned by 28 days post-infection. Innate immune responses varied significantly between age groups. Robust type I interferon and inflammasome responses were measured in older chickens, while limited innate immune activation was observed in younger chicks. Signal transducer and activator of transcription 2 (STAT2) is a major driver of host restriction to ZIKV, and chicken STAT2 is distinct from human STAT2, potentially contributing to the observed resistance to ZIKV infection. The rapid clearance of the virus in older chickens coincided with an effective innate immune response, highlighting age-dependent susceptibility. Our study indicates that chickens are not susceptible to productive ZIKV infection and are unlikely to play a role in the ZIKV epidemiology.

This research aims to comprehensively examine the clearness index (KT), total ozone column (TOC), and ultraviolet A (UVA) and ultraviolet B radiation (UVB) over Lumbini, Nepal (27°28’ N, 83°16’ E, and 150 m above sea level) throughout the 11 years of solar cycle 24 (2008 to 2018). The Lumbini, a highly polluted region, is important in advancing the identification and analysis of TOC variations across regions with similar geographical and climatic attributes. Data from the Ozone Monitoring Instrument (OMI) of the EOS-AURA satellite of NASA were used to analyze the daily, monthly, seasonal, and annual trends in the clearness index (KT), ultraviolet A (UVA), ultraviolet B (UVB), and TOC from the Comprehensive Environmental Data Archive (CEDA). The study found that the yearly averages for KT, TOC, UVA, and UVB were 0.55 ± 0.13, 272 ± 14 DU, 12.61 ± 3.50 W/m2, and 0.32 ± 0.11 W/m2, respectively. These values provide insights into the long-term variations in atmospheric parameters at Lumbini. The study also applied the continuous wavelet transform (CWT) to analyze KT, TOC, UVA, and UVB temporal variations. The power density peak of 35,000 DU2 in the TOC was observed from the end of 2010 to the end of 2011, within 8.5 years, underscoring the significance of analyzing TOC dynamics over extended durations to understand atmospheric behavior comprehensively.

In recent decades, the requirements for implantable medical devices have increased, but the risks of implant rejection still exist. These issues are primarily associated with poor osseointegration, leading to biofilm formation on the implant surface. This study focuses on addressing these issues by developing a biomaterial for implant coatings. 45S5 bioglass® has been widely used in tissue engineering due to its ability to form a hydroxyapatite layer, ensuring a strong bond between the hard tissue and the bioglass. In this context, 45S5 bioglasses®, modified by the incorporation of different amounts of copper oxide, from 0 to 8 mol%, were synthesized by the melt–quenching technique. The incorporation of Cu ions did not show a significant change in the glass structure. Since the bioglass exhibited the capacity for being polarized, thereby promoting the osseointegration effectiveness, the electrical properties of the prepared samples were studied using the impedance spectroscopy method, in the frequency range of 102–106 Hz and temperature range of 200–400 K. The effects of CuO on charge transport mobility were investigated. Additionally, the bioactivity of the modified bioglasses was evaluated through immersion tests in simulated body fluid. The results revealed the initiation of a Ca–P-rich layer formation on the surface within 24 h, indicating the potential of the bioglasses to enhance the bone regeneration process.

Without physically intensive building, modern infrastructure development would be impossible. Musculoskeletal diseases (MSDs) and other occupational health issues may arise in such a demanding environment. Construction workers often develop MSDs from repeated actions, uncomfortable postures, and heavy lifting. Musculoskeletal disorders may damage muscles, bones, tendons, ligaments, etc. The effect of MSDs is well known; occupational health studies increasingly include gender-specific aspects. Despite being in the minority, the number of female construction employees is growing. However, physiological variations and occupational activities and environments may provide distinct obstacles. Thus, identifying gender-specific MSDs in construction is essential for worker safety. This research proposes a gender-specific machine learning (ML)-based musculoskeletal disorder detection framework (GS-ML-MSD2F) in the construction industry. A simple random selection procedure chose 250 female and 250 male rebar workers with at least six months of experience for the dataset. In January and June 2023, face-to-face interviews and ergonomic evaluations were undertaken. The data were analyzed using different machine learning methods, and the effectiveness of the methods was studied. The data showed that 60% of female rebar workers had MSD symptoms. The lower back and shoulders accounted for 40% of cases. Multiple machine learning methods revealed two significant factors related to musculoskeletal disorders: lengthy working hours and uncomfortable postures, and long working hours had an adjusted odds ratio of 8.5%, whereas awkward posture had an adjusted odds ratio of 42.5%. These results emphasize the relevance of working hours and posture in MSD prevention for female rebar workers in the construction sector.

Oral biofilms are considered the principal etiological agent in the development of periodontitis. Novel species that may contribute to periodontitis and dysbiosis have been identified recently. The study aims to evaluate the presence of F. alocis and D. pneumosintes in healthy and diseased patients and their association with clinical parameters and with red complex bacteria. The study included 60 subjects, with 30 patients each in the healthy and periodontitis groups. The clinical parameters were noted, and samples were subjected to DNA extraction followed by a polymerase chain reaction. Statistical analysis was performed using the Graph Pad Prism software. Results: F. alocis and D. pneumosintes were detected at a significantly higher percentage in the periodontitis group compared to the healthy group (p < 0.05). D. pneumosintes was significantly associated with T. forsythia in the periodontitis group (p < 0.05). Both of these organisms were present in sites with higher clinical attachment loss (p < 0.05). This study demonstrated that both F. alocis and D. pneumosintes were detected at a significantly higher percentage in periodontitis subjects and were detected more frequently in sites with a greater clinical attachment loss. It was also evident that both F. alocis and D. pneumosintes can be present independently of other putative periodontal pathogens.

Bacillus anthracis is the bacterium responsible for causing the zoonotic disease called anthrax. The disease presents itself in different forms like gastrointestinal, inhalation, and cutaneous. Bacterial spores are tremendously adaptable, can persist for extended periods and occasionally endanger human health. The Anthrax Toxin Receptor-2 (ANTXR2) gene acts as membrane receptor and facilitates the entry of the anthrax toxin into host cells. Additionally, mutations in the ANTXR2 gene have been linked to various autoimmune diseases, including Hyaline Fibromatosis Syndrome (HFS), Ankylosing Spondylitis (AS), Juvenile Hyaline Fibromatosis (JHF), and Infantile Systemic Hyalinosis (ISH). This study delves into the genetic landscape of ANTXR2, aiming to comprehend its associations with diverse disorders, elucidate the impacts of its mutations, and pinpoint minimal non-pathogenic mutations capable of reducing the binding affinity of the ANTXR2 gene with the protective antigen. Recognizing the pivotal role of single-nucleotide polymorphisms (SNPs) in shaping genetic diversity, we conducted computational analyses to discern highly deleterious and tolerated non-synonymous SNPs (nsSNPs) in the ANTXR2 gene. The Mutpred2 server determined that the Arg465Trp alteration in the ANTXR2 gene leads to altered DNA binding (p = 0.22) with a probability of a deleterious mutation of 0.808; notably, among the identified deleterious SNPs, rs368288611 (Arg465Trp) stands out due to its significant impact on altering the DNA-binding ability of ANTXR2. We propose these SNPs as potential candidates for hypertension linked to the ANTXR2 gene, which is implicated in blood pressure regulation. Noteworthy among the tolerated substitutions is rs200536829 (Ala33Ser), recognized as less pathogenic; this highlights its potential as a valuable biomarker, potentially reducing side effects on the host while also reducing binding with the protective antigen protein. Investigating these SNPs holds the potential to correlate with several autoimmune disorders and mitigate the impact of anthrax disease in humans.

Bone regeneration poses a significant challenge in the field of tissue engineering, prompting ongoing research to explore innovative strategies for effective bone healing. The integration of stem cells and nanomaterial scaffolds has emerged as a promising approach, offering the potential to enhance regenerative outcomes. This study focuses on the application of a stem cell-laden nanomaterial scaffold designed for bone regeneration in rabbits. The in vivo study was conducted on thirty-six healthy skeletally mature New Zealand white rabbits that were randomly allocated into six groups. Group A was considered the control, wherein a 15 mm critical-sized defect was created and left as such without any treatment. In group B, this defect was filled with a polycaprolactone–hydroxyapatite (PCL + HAP) scaffold, whereas in group C, a PCL + HAP-carboxylated multiwalled carbon nanotube (PCL + HAP + MWCNT-COOH) scaffold was used. In group D, a PCL + HAP + MWCNT-COOH scaffold was used with local injection of bone morphogenetic protein-2 (BMP-2) on postoperative days 30, 45, and 60. The rabbit bone marrow-derived mesenchymal stem cells (rBMSCs) were seeded onto the PCL + HAP + MWCNT-COOH scaffold by the centrifugal method. In group E, an rBMSC-seeded PCL + HAP + MWCNT-COOH scaffold was used along with the local injection of rBMSC on postoperative days 7, 14, and 21. For group F, in addition to the treatment given to group E, BMP-2 was administered locally on postoperative days 30, 45, and 60. Gross observations, radiological observation, scanning electron microscopic assessment, and histological evaluation study showed that group F displayed the best healing properties, followed by group E, group D, group C, and B. Group A showed no healing with ends blunting minimal fibrous tissue. Incorporating growth factor BMP-2 in tissue-engineered rBMSC-loaded nanocomposite PCL + HAP + MWCNT-COOH construct can augment the osteoinductive and osteoconductive properties, thereby enhancing the healing in a critical-sized bone defect. This novel stem cell composite could prove worthy in the treatment of non-union and delayed union fractures in the near future.
