The complex field environment under conservation tillage aggravates the vibration during a planter’s operation, affecting the sowing quality and fertilization depth. Studying its vibration characteristics can help to realize active vibration reduction control of planter row units. To this end, this paper took a four-row no-till planter as the research object. By establishing a field vibration model of the planter row unit, the factors affecting the vibration of the unit were clarified, and stubble height, working speed and the additional weight of the planter were used as experimental factors in carrying out field orthogonal experiments. In our experiment, we collected and analyzed vibration data on the four-row planter row units and the frame at different positions to explore the influence of various factors on the vibration characteristics of the planter. The experimental results showed that the working speed was the most important factor affecting the vibration of the planter, and the impact of stubble height and additional weight on the amplitude of the planter was more significant at low speed (1.5 m/s) than that at high speed (2.5 m/s). The difference in amplitude of each planter unit in the lateral direction was the largest, the average amplitude range of which was 1.898 m/s2. The vibration energy of each planter row unit under different working conditions was mainly concentrated in the range of 10–50 Hz. However, the three-point hitch of the planter transmitted the vibration excitation of the tractor, causing 110–120 Hz high-frequency vibration of the inner row units, while the outer row units were less affected, with the vibration energy, in the range above 100 Hz, being 2.5 dB smaller than that on the inner side. The right ground wheel transmission device was abnormal, which worked together with the excitation transmitted by the three-point hitch, making the average vibration acceleration amplitude of the planter row units on the right side in the lateral direction more than 0.522 m/s2 higher than that of the units on the left side. Therefore, different vibration reduction forces need to be applied according to the position of the planter row unit, so that the units can avoid the natural frequency of the frame (115 Hz) when vibrating. This study can provide a reference for active vibration reduction control and improvements in sowing quality for high-speed no-till planters.

In response to the problems of low control accuracy, single detection of operating parameters, and insufficient collaborative control of unmanned fertilization in field fertilization operations, this paper proposes an adaptive control strategy for fertilizer discharging shaft speed based on segmented linear interpolation method. By constructing a relationship model between fertilizer discharging shaft speed and motor control signals in different speed ranges, the on-site self-calibration of fertilizer discharging shaft speed and the precise control of the fertilization rate is achieved. At the same time, real-time detection and warning technology for multiple working condition parameters were integrated, and a bus communication protocol between unmanned tractors and variable-rate fertilizer applicators was developed. A variable-ratefertilization monitoring system based on unmanned tractors was developed, and actual experimental tests were conducted to test the system’s performance. Among them, the calibration test results of fertilization rate showed that the discharging rate of the fertilizer apparatuses (p) was negatively correlated with the rotation speed of the fertilizer discharging shaft, and that the installation position of the fertilizer apparatuses affected the uniformity of fertilization between the rows of the fertilizer applicator. The speed response test of the fertilizer discharging shaft showed that the average response time (Ts) of the fertilizer discharging shaft speed controlled by the self-calibration model was 0.40 s, the average steady-state error (ess) was 0.13 r/min, and the average overshoot (σ) was 7.33%. Compared with the original linear model, the ess was reduced by 0.23 r/min, and the σ was reduced by 1.54 percentage points. The results of the fertilization status detection tests showed that the system can achieve real-time detection of different operating parameters and states, as well as collaborative control of tractors and fertilizer applicators. The results of the fertilization rate control accuracy test showed that the average fertilization control error of the system was 1.91% under different target fertilization rate, which meets the requirements of variable-rate fertilization field operations. This study can serve as a technical reference for the design and development of fertilization robots in the context of unmanned farm development.

For the mechanized technical mode of total wheat straw returning to field, there are problems such as large vibration during the operation of the straw returning machine that, in turn, affect the effect of stubble breaking. This study took the Tongtian 1-JHY-220 straw returning machine as the research object to conduct field experiments, with wheat stubble height, forward velocity, and PTO speed as experimental parameters. And the vibration characteristics at different positions of the machine and the final stubble breaking rate were used as evaluation indicators. Combined with the orthogonal experiment and response surface analysis method, this article analyzes and discusses the influence of various parameters on vibration characteristics and operational effectiveness. The results show that PTO speed and wheat stubble height were the main factors affecting the vibration and operation quality of the straw returning machine. Low PTO speed and high stubble height can improve the stubble breaking rate of the straw returning machine and reduce its operation vibration. Furthermore, the multi-objective optimization results show that when the forward velocity in the range of 8.5–9 km/h, the PTO speed is 540 r/min, and the stubble height is in the range of 200–250 mm, the stubble breaking rate of the straw returning machine is greater than 86%. At this time, the total vibration of the straw returning machine and tractor rear axle is relatively small. This study can lay a foundation for further studying the impact of the vibration of the straw returning machine on the stubble breaking effect and provide a reference for the preparation of high-quality seedbed under conservation tillage.

Zinc finger proteins (ZFPs) are essential transcription factors in eukaryotes, particularly the extensively studied C2H2 family, which is known for its involvement in various biological processes. This research provides a thorough examination and analysis of the C2H2-ZFP gene family in Flammulina filiformis. Using bioinformatics tools, 58 FfC2H2-ZFP genes spread across 11 chromosomes were identified and scrutinized in detail for their gene structures, protein characteristics, and phylogenetic relationships. The study of phylogenetics and synteny sheds light on the evolutionary relationships among C2H2-ZFPs in F. filiformis and other fungi, revealing a complex evolutionary past. The identification of conserved cis-regulatory elements in the gene promoter regions suggests intricate functionalities, particularly in the developmental and stress response pathways. By utilizing RNA-seq and qRT-PCR techniques, the expression patterns of these genes were explored across different developmental stages and tissues of F. filiformis, unveiling distinct expression profiles. Notably, significant expression variations were observed in the stipe elongation region and pilei of various sizes, indicating potential roles in fruiting body morphogenesis. This study enhances our knowledge of the C2H2-ZFP gene family in F. filiformis and lays the groundwork for future investigations into their regulatory mechanisms and applications in fungal biology and biotechnology.

A good planting bed is a prerequisite for improving planting quality, while complex ground excitation often leads to machine bouncing and operation vibration, which then affects the operation effect. In order to improve the quality of rotary tillage operations, it is necessary to study the effects of various vibration excitations on the unit during tractor rotary tillage operations and analyze the vibration interaction relationship among the tractor, the three-point suspension mechanism, and the rotary tiller. For this purpose, multiple three-way acceleration sensors were installed at different positions on the rotary tiller unit of a Lexing LS1004 tractor(Lexing Agricultural Equipment Co. Ltd., Qingdao, China) to collect vibration data at different operating speeds and conduct vibration characteristic analysis between different components. The test results showed that when the unit moved forward at 2.1 km/h, 3.6 km/h, and 4.5 km/h, respectively, the vibration acceleration of the tractor, the three-point suspension mechanism, and the rotary tiller increased with the increase in speed, and there was indeed interaction between them. The vertical acceleration change during the test in the three-point suspension mechanism was the most significant (5.914 m/s2) and was related to the increase in the speed of the vehicle and the vibration transfer of the rotary tiller. Meanwhile, the vertical vibration acceleration of the tractor’s symmetrical structure was not similar, suggesting the existence of structural assembly problems. From the perspective of frequency domain analysis, the resonant frequency at the cab of the tractor was reduced in a vertical vibration environment, with relatively low frequencies (0~80 Hz) and small magnitudes, which might be beneficial to the driver’s health. The rotary tillage group resonated around 350 Hz, and this characteristic can be used to appropriately increase the vibration of the rotary tiller to reduce resistance. The tractor cab resonated around 280 Hz, which must be avoided during field operations to ensure driver health and reduce machine wear. The research results can provide a reference for reducing vibration and resistance during tractor rotary tillage operations, as well as optimizing and improving the structure of rotary tillers and tractors.

Coniella vitis is a dominant phytopathogen of grape white rot in China, significantly impacting grape yield and quality. Previous studies showed that the growth and pathogenicity of C. vitis were affected by the environmental pH. Arrestin-like protein PalF plays a key role in mediating the activation of an intracellular-signaling cascade in response to alkaline ambient. However, it remains unclear whether palF affects the growth, development, and virulence of C. vitis during the sensing of environmental pH changes. In this study, we identified a homologous gene of PalF/Rim8 in C. vitis and constructed CvpalF-silenced strains via RNA interference. CvpalF-silenced strains exhibited impaired fungal growth at neutral/alkaline pH, accompanied by reduced pathogenicity compared to the wild-type (WT) and empty vector control (CK) strains. The distance between the hyphal branches was significantly increased in the CvpalF-silenced strains. Additionally, CvpalF-silenced strains showed increased sensitivity to NaCl, H2O2, and Congo red, and decreased sensitive to CaSO4. RT-qPCR analysis demonstrated that the expression level of genes related to pectinase and cellulase were significantly down-regulated in CvpalF-silenced strains compared to WT and CK strains. Moreover, the expression of PacC, PalA/B/C/F/H/I was directly or indirectly affected by silencing CvpalF. Additionally, the expression of genes related to plant cell wall-degrading enzymes, which are key virulence factors for plant pathogenic fungi, was regulated by CvpalF. Our results indicate the important roles of CvpalF in growth, osmotolerance, and pathogenicity in C. vitis.

Most previously studies had considered that plant fungal disease spread widely and quickly by airborne fungi spore. However, little is known about the release dynamics, aerodynamic diameter, and pathogenicity threshold of fungi spore in air of the greenhouse environment. Grape gray mold is caused by Botrytis cinerea; the disease spreads in greenhouses by spores in the air and the spore attaches to the leaf and infects plant through the orifice. In this study, 120 μmol/L propidium monoazide (PMA) were suitable for treatment and quantitation viable spore by quantitative real-time PCR, with a limit detection of 8 spores/mL in spore suspension. In total, 93 strains of B. cinerea with high pathogenicity were isolated and identified from the air samples of grapevines greenhouses by a portable sampler. The particle size of B. cinerea aerosol ranged predominately from 0.65–3.3 μm, accounting for 71.77% of the total amount. The B. cinerea spore aerosols were infective to healthy grape plants, with the lowest concentration that could cause disease being 42 spores/m3. Botrytis cinerea spores collected form six greenhouse in Shandong Province were quantified by PMA-qPCR, with a higher concentration (1182.89 spores/m3) in May and June and a lower concentration in July and August (6.30 spores/m3). This study suggested that spore dispersal in aerosol is an important route for the epidemiology of plant fungal disease, and these data will contribute to the development of new strategies for the effective alleviation and control of plant diseases.

Nucleotide substitutions have played an important role in molecular evolution, and understanding their dynamics would contribute to genetic studies. Related research with defined DNA sequences lasted for decades until whole-genome sequencing arose. UV radiation (UVR) can generate base changes and other genetic variations in a short period of time, so it would be more meaningful to explore mutations caused by UVR from a genomic perspective. The monokaryon enoki strain WT583 was selected as the experimental material in this study because it can spontaneously produce large amounts of oidia on PDA plates, and the monokaryons originating from oidia have the same genotype as their mother monokaryon. After exposure to UV radiation, 100 randomly selected mutants, with WT583 as the reference genome, were sent for genome sequencing. BWA, samtools, and GATK software were employed for SNP calling, and the R package CMplot was used to visualize the distribution of the SNPs on the contigs of the reference genome. Furthermore, a k-mer-based method was used to detect DNA fragment deletion. Moreover, the non-synonymous genes were functionally annotated. A total of 3707 single-base substitutions and 228 tandem mutations were analyzed. The immediate adjacent bases showed different effects on the mutation frequencies of adenine and cytosine. For adenine, the overall effects of the immediate 5′-side and 3′-side bases were T > A > C > G and A > T > G > C, respectively; for cytosine, the overall effects of the immediate 5′-side and 3′-side bases were T > C > A > G and C > T > A > G, respectively. Regarding tandem mutations, the mutation frequencies of double-transition, double-transversion, 3′-side transition, and 5′-side transition were 131, 8, 72, and 17, respectively. Transitions at the 3′-side with a high mutation frequency shared a common feature, where they held transversions at the 5′-side of A→T or T→A without covalent bond changes, suggesting that the sequence context of tandem motifs might be related to their mutation frequency. In total, 3707 mutation sites were non-randomly distributed on the contigs of the reference genome. In addition, pyrimidines at the 3′-side of adenine promoted its transversion frequency, and UVR generated DNA fragment deletions over 200 bp with a low frequency in the enoki genome. The functional annotation of the genes with non-synonymous mutation indicated that UVR could produce abundant mutations in a short period of time.

Low temperatures are among the most important abiotic stresses, severely affecting vine growth and development. In order to overwinter safely, grape growers pay high labor costs, which are unsustainable in modern agriculture. Biodegradable liquid film (BLF) is an environmentally friendly material that assists grapevines to withstand the cold. In this experiment, Cabernet Sauvignon from Junding Winery was used as material for field trials. The regulatory effect of BLF on the cold resistance of grapevines was analyzed, and its mechanism of action was explored using metabolomics. The results showed that BLF significantly increased the activity of superoxide dismutase, the content of proline and ascorbic acid, reducing sugar, sucrose, and soluble sugar, and also decreased the content of malondialdehyde. In addition, BLF significantly increased the activity of invertase and decreased the activity of pectinase and phosphofructokinase. Metabolomics data showed that BLF improved cold resistance in grapevines by affecting lipids, flavonoids, amino acids, and nucleotide metabolic pathways.

Traffic flow forecasting is an important function of intelligent transportation systems. With the rise of deep learning, building traffic flow prediction models based on deep neural networks has become a current research hotspot. Most of the current traffic flow prediction methods are designed from the perspective of model architectures, using only the traffic features of future moments as supervision signals to guide the models to learn the spatiotemporal dependence in traffic flow. However, traffic flow data themselves contain rich spatiotemporal features, and it is feasible to obtain additional self-supervised signals from the data to assist the model to further explore the underlying spatiotemporal dependence. Therefore, we propose a self-supervised traffic flow prediction method based on a spatiotemporal masking strategy. A framework consisting of symmetric backbone models with asymmetric task heads were applied to learn both prediction and spatiotemporal context features. Specifically, a spatiotemporal context mask reconstruction task was designed to force the model to reconstruct the masked features via spatiotemporal context information, so as to assist the model to better understand the spatiotemporal contextual associations in the data. In order to avoid the model simply making inferences based on the local smoothness in the data without truly learning the spatiotemporal dependence, we performed a temporal shift operation on the features to be reconstructed. The experimental results showed that the model based on the spatiotemporal context masking strategy achieved an average prediction performance improvement of 1.56% and a maximum of 7.72% for longer prediction horizons of more than 30 min compared with the backbone models.

We propose a novel approach to cluster hierarchical time series (HTS) for efficient forecasting and data analysis. Inspired by a practically important but unstudied problem, we found that leveraging local information when clustering HTS leads to a better performance. The clustering procedure we proposed can cope with massive HTS with arbitrary lengths and structures. In addition to providing better insights, this method can also speed up the forecasting process for a large number of HTS. Each time series is first assigned the forecast from its cluster representative, which can be considered as “prior shrinkage” for the set of time series it represents. Then, the base forecast can be efficiently adjusted to accommodate the specific attributes of the time series. We empirically show that our method substantially improves performance for large-scale clustering and forecasting tasks involving HTS.

Traffic flow forecasting is a basic function of intelligent transportation systems, and the accuracy of prediction is of great significance for traffic management and urban planning. The main difficulty of traffic flow predictions is that there is complex underlying spatiotemporal dependence in traffic flow; thus, the existing spatiotemporal graph neural network (STGNN) models need to model both temporal dependence and spatial dependence. Graph neural networks (GNNs) are adopted to capture the spatial dependence in traffic flow, which can model the symmetric or asymmetric spatial relations between nodes in the traffic network. The transmission process of traffic features in GNNs is guided by the node-to-node relationship (e.g., adjacency or spatial distance) between nodes, ignoring the spatial dependence caused by local topological constraints in the road network. To further consider the influence of local topology on the spatial dependence of road networks, in this paper, we introduce Ollivier–Ricci curvature information between connected edges in the road network, which is based on optimal transport theory and makes comprehensive use of the neighborhood-to-neighborhood relationship to guide the transmission process of traffic features between nodes in STGNNs. Experiments on real-world traffic datasets show that the models with Ollivier–Ricci curvature information outperforms those based on only node-to-node relationships between nodes by ten percent on average in the RMSE metric. This study indicates that by utilizing complex topological features in road networks, spatial dependence can be captured more sufficiently, further improving the predictive ability of traffic forecasting models.

Due to the influence of the optical proximity effect (OPE), it is easy for a pattern of photoresistance to be inconsistent with a design pattern, thus damaging the performance of a SAW resonator. To solve this problem, this paper proposes an optimization method for SAW filters based on optical proximity correction (OPC). This method can avoid the tip discharge problem of SAW filters by suppressing the problem of rounding and shrinking of dummy electrode and electrode tail caused by OPE. This method increases the quality factor (Q) of the SAW resonator and thus decreases the insertion loss of the SAW filter. The filter increases the bandwidth by 1.8 MHz at −1.5 dB after applying the OPC method. Additionally, it improves the stability of the filter under high power conditions.

Stipe gradient elongation is an important and remarkable feature in the development of most mushroom fruiting bodies. However, its molecular mechanism has rarely been described. Here, the decreasing trend of stipe elongation and increasing trend of cell length in a gradient from the top to the base of the stipe were determined in a model basidiomycete mushroom: Flammulina filiformis. According to RNA-seq results, 1409 differentially expressed genes (DEGs) were identified among elongation region (ER), transition region (TR), and stable region (SR) samples, including 26 transcription factors (TFs). Based on Short Time-series Expression Miner (STEM) clustering of DEGs, clusters 1 and 3, with obvious expression trends that were consistent with or in contrast to the elongation rate, were screened. The cluster 1 DEGs were mainly involved in the GO cellular component category and KEGG genetic information processing class; however, the cluster 3 DEGs were mainly involved in metabolic processes. Furthermore, qRT-PCR confirmed that key genes of the long-chain fatty acid synthesis pathway were involved in stipe gradient elongation and regulated by NADPH oxidase-derived ROS signaling molecules. These findings provide an essential basis for understanding the molecular mechanism of stipe gradient elongation.

Downy mildew is a major disease that severely restricts the healthy and sustainable development of the global grape and wine industry, so there is significant interest in breeding high-quality disease-resistant varieties. In this study, hybridization was carried out between two disease-resistant and four high-quality varieties for eight hybrid combinations. The downy mildew resistance of 6 hybrid parents and 350 hybrid F1 progeny was determined by the leaf disc inoculation test, and the frequency distribution of the disease resistance grades was analyzed for the different populations. Agarose gel electrophoresis and disease resistance identification were used to screen SSR molecular markers of the tested hybrids for resistance to downy mildew. The results showed that minor disease-resistance genes in V. vinifera varieties were the main determinant of the variation in disease susceptibility among different varieties. V. vinifera resistance to downy mildew is inherited by a quantitative trait controlled by multiple genes. Intraspecific hybridization of V. vinifera can be applied to breed suitable intermediate materials or new types of disease resistance. The screening and verification of simple-sequence repeats (SSR) as molecular markers revealed that UDV-304, UDV-305, and UDV-737 could be used as standard markers for the identification of downy mildew resistance of hybrid progeny of Ecolly and Cabernet Sauvignon, UDV-305 could be used for the hybrid progeny of Meili and Cabernet Sauvignon, and VMC8g9 could be used for the hybrid progeny of Ecolly or Meili and Dunkelfelder. The disease-resistant progeny selected in this study can be used as intermediate materials for the breeding of new high-quality, disease-resistant varieties by intraspecific recurrent selection in V. vinifera. The screened standard markers can be utilized for rapid batch identification of hybrid progeny of different hybrid combinations to facilitate molecular marker-assisted breeding.

The mushroom stipe raises the pileus above the substrate into a suitable position for dispersing spores. The stipe elongates at different speeds along its length, with the rate of elongation decreasing in a gradient from the top to the base. However, the molecular mechanisms underlying stipe gradient elongation are largely unknown. Here, we used the model basidiomycete mushroom Flammulina filiformis to investigate the mechanism of mushroom stipe elongation and the role of reactive oxygen species (ROS) signaling in this process. Our results show that O2− and H2O2 exhibit opposite gradient distributions in the stipe, with higher O2− levels in the elongation region (ER), and higher H2O2 levels in the stable region (SR). Moreover, NADPH-oxidase-encoding genes are up-regulated in the ER, have a function in producing O2−, and positively regulate stipe elongation. Genes encoding manganese superoxide dismutase (MnSOD) are up-regulated in the SR, have a function in producing H2O2, and negatively regulate stipe elongation. Altogether, our data demonstrate that ROS (O2−/H2O2) redistribution mediated by NADPH oxidase and MnSODs is linked to the gradient elongation of the F. filiformis stipe.

Biodegradable liquid mulch film (LF), which can be degraded naturally without harming the environment, is a new type of covering material that provides an environmentally friendly alternative to plastic mulch film (PF). In this study, the effects of LF and PF (ploughing (CK) used as a control) on the soil and fruit quality of Hutai-8 were evaluated through an experiment, and several soil physicochemical properties and indicators of fruit quality were measured. In-row mulching significantly increased the content of total potassium, available phosphorus, and available potassium in the topsoil (0–20 cm), the ripeness of the grape berries, and the content of phenolics in the skin. The effects were consistent between the two years. The effect of LF was more pronounced in the same year, indicating that LF is an effective alternative to PF. Therefore, LF can be used as an environmentally friendly substitute for PF to improve soil and fruit quality and incorporated into cultivation management plans. Correlation analysis revealed that the content of reducing sugars, flavonoids, total phenols, flavan-3-ols, and anthocyanins, as well as fruit ripeness, increased as the content of total potassium, available phosphorus, and available potassium in the soil increased.

Biodegradable liquid film (BLF) improves soil structure and increases plant freezing tolerance after spraying on the surface of soil and plant. In this study, the effects of BLF on grape composition and volatile compounds in Cabernet sauvignon (Vitis vinifera L.) grapes were determined by spraying BLF during the dormant periods over three years. The aim of the study was to evaluate the potential impact of BLF as an overwintering protection measure on grape fruit quality. In 2020 and 2021, BLF spraying increased reducing sugar content and 100-berry weight, decreased titratable acid content, and improved the maturity factor. Compared with the vines not sprayed with BLF, the content of total phenols and total anthocyanins in grape skins showed an increase over the three-year period, with the largest increases of 31.92% and 48.38%, respectively, and the content of total tannins and total flavan-3-ols increased in 2020 and 2021. BLF treatment also increased the total phenolic content in seeds for all three years, reaching a significant level in 2019, 16.38% higher than control treatment (CK). HPLC analysis showed that BLF treatment affected the content and composition of monomeric anthocyanins in grape skins, especially in 2021, BLF treatment significantly increased the content of nine monomeric anthocyanins, and the proportion of acetylated and coumaroylated anthocyanins. However, GC-MS analysis indicated that BLF had little effect on volatile compounds. These results suggest that BLF can be used as an overwintering protection measure in cold regions to promote the accumulation of sugars and polyphenolics, thereby improving overall grape quality.

Intraspecific recurrent selection in V. vinifera is an effective method for breeding of high quality, disease-, cold-, and drought-resistance grapes. Exploring the optimal treatment methods for grape (V. vinifera) seeds can help to accelerate the process of intraspecific recurrent selection and improve breeding efficiency. In this study, seeds of six V. vinifera varieties were used as experimental materials, and the germination and seedling formation characteristics were studied by single factor treatment and orthogonal compound treatment, respectively. To do this, stratification, chemical substances, beak cutting, and pre-germination treatments were tested, and the optimal treatment combination was determined for each variety. The results indicated that the optimal conditions obtained in the orthogonal experiments were not completely consistent with those in the single-factor experiments. Single factor experiment results demonstrated that two stratification methods (chilling gauze-storage and chilling sand-storage) and two pre-germination methods (pre-germination in petri dishes and pre-germination in a bean sprouter) vary in effectiveness for different varieties. gibberellin acid (GA3) soaking and beak-cutting promote the germination and seedling rate of the tested varieties. Orthogonal test results demonstrate that, for Dunkelfelder and Cabernet Sauvignon, the optimal treatment combination was chilling sand-storage + GA3 soaking seed + beak cutting + pre-germination in petri dishes. For Meili, the optimal treatment combination was chilling sand-storage + acetic acid (HAc) soaking seed + beak cutting + pre-germination in petri dishes. For Ecolly, the optimal treatment combination was chilling sand-storage + GA3 soaking seed + beak cutting + pre-germination in a bean sprouter. For Garanior, the optimal treatment combination was chilling sand-storage + HAc soaking seed + no beak cutting + pre-germination in petri dishes. For Marselan, the optimal treatment combination was chilling gauze-storage + GA3 soaking seed + beak cutting + pre-germination in a bean sprouter. This study identified the optimal conditions for seed germination and seedling formation of six grape varieties, which will facilitate future work to characterize the seed germination and seedling formation of seeds obtained by intraspecific hybridization of these varieties. This work also provides a reference for addressing problems of low seed germination rate and suboptimal seedling formation for better utilization of grape germplasms.

With the extreme changes of the global climate, winter freezing injury has become an important limiting factor for the development of the global grape industry. Therefore, there is a significant need for the screening of cold-resistant wine grape germplasms and cold regionalization for cold-resistant breeding and the development of grapevine cultivation in cold regions. In this study, the low-temperature half-lethal temperature (LT50) values were determined for the annual dormant branches of 124 wine grape germplasms (V. vinifera) to evaluate their cold resistance. The LT50 values of the 124 tested germplasms ranged from −22.01 °C to −13.18 °C, with six cold-resistant germplasms below −20 °C. Based on the LT50 values, the 124 germplasms were clustered into four types, with cold resistance from strong to weak in the order of type Ⅱ > type Ⅰ > type Ⅳ > type Ⅲ, corresponding to the four cold hardiness zones. Zones 1, 2, 3, and 4 included 6, 22, 68, and 28 germplasms, respectively, with decreasing cold resistance. The number of germplasms in different hardiness zones followed a normal distribution, with the most in zone 3. In Type Ⅱ, the fruit skin color of germplasms was positively correlated with cold hardiness, while the temperature of origin was negatively correlated with cold hardiness. The average LT50 of germplasms in different origin regions ranged from −17.44 °C to −16.26 °C, with differences among some regions. The cold regionalization analysis resulted in the distribution of 124 germplasms in four temperature regions in China with six germplasms in region A (−22 °C ≤ LT50 ≤ −20 °C), 30 germplasms in region B (−20°C ≤ LT50 ≤ −18°C), 71 germplasms in region C (−18 °C ≤ LT50 ≤ −15 °C), and 17 germplasms in region D (−15 °C ≤ LT50 ≤ −13 °C). Strong cold-resistant wine grape germplasms (V. vinifera) were identified, and these could be used as parental material for cold-resistant breeding. In some areas in China, soil-burial over-wintering strategies are used, but our results suggest that some wine grapes could be cultivated without requiring winter burial during overwintering. The results of this study should provide guidance for the selection of promising strains for cold-resistant breeding for expanded cultivation of improved varieties for wine grape production in China.

As a representative economic crop with the potential for long-term sustainable development, grapevines (Vitis vinifera L.) bring direct economic benefits to the people, and the indirect ecological benefits produced by grapevines cannot be ignored. This study utilized relevant statistical data from the government sectors and data obtained at vineyards and wineries through interviews and surveys. The value of the ecosystem service of the grape industry in soil-burial over-wintering areas was first evaluated from four first-level indicators types and 10 s-level indicators types, using Hongsibu of the eastern foothill of Helan Mountain, Ningxia, as an example. The results show that the total current benefits of the grape industry ecosystem service function in Hongsibu are ¥ 1067 million. The direct economic benefits are ¥ 660 million and indirect ecological benefits are ¥ 407 million. The value ranking of the first-level function types is as follows: supply services > cultural services > support services > regulatory services. The ranking of the value of second-level function types is as follows: food production > ecological poverty alleviation > biodiversity > carbon fixation and oxygen release > ecotourism > air purification > nutrient accumulation > water conservation and efficiency > water holding > soil conservation. Cultivation measures (such as burial-free cultivation technology, inter-row grass technology, and suspending grape branches after winter pruning technology) increased the value of the service function of the grape ecosystem and explored some potential benefits. The results indicated the necessity of assessing ecosystem service value of the grape industry, which could enable people to realize the direct economic benefits as well as the ecological benefits. Meanwhile, the positive effects of cultivation techniques on the value of ecosystem service function were demonstrated in soil-burial over-wintering areas. This study provides important reference value and comprehensive guidance for the healthy and sustainable development of the grape industry.

Due to the particularity of the continental monsoon climate in China, more than 90% of the wine grape cultivation areas require vines to be buried in winter for a burial period that can extend to half a year. Additionally, traditional vine cultivation practices can expose the surface of the soil during winter, easily leading to soil erosion. To meet the restrictive factors for viticulture in the Chinese cold climate zone, a new sustainable viticulture strategy called crawled cordon mode (CCM) has been developed. CCM includes crawled cordon training (CCT), physical methods of flower and fruit thinning, winter suspension of shoots, the use of a biodegradable liquid film, and covering of grass and branches for simplified management of vineyards. This article summarizes the specific implementation methods of the main measures of CCM and their significant effects on the quality of grapes and wine, ecological environment, and costs, and aims to provide inspiration for the study of sustainable and eco-friendly cultivation measures for vineyards in other cold climate zones.

Visual sorting of express parcels in complex scenes has always been a key issue in intelligent logistics sorting systems. With existing methods, it is still difficult to achieve fast and accurate sorting of disorderly stacked parcels. In order to achieve accurate detection and efficient sorting of disorderly stacked express parcels, we propose a robot sorting method based on multi-task deep learning. Firstly, a lightweight object detection network model is proposed to improve the real-time performance of the system. A scale variable and the joint weights of the network are used to sparsify the model and automatically identify unimportant channels. Pruning strategies are used to reduce the model size and increase the speed of detection without losing accuracy. Then, an optimal sorting position and pose estimation network model based on multi-task deep learning is proposed. Using an end-to-end network structure, the optimal sorting positions and poses of express parcels are estimated in real time by combining pose and position information for joint training. It is proved that this model can further improve the sorting accuracy. Finally, the accuracy and real-time performance of this method are verified by robotic sorting experiments.

In this work, closed-cell aluminum foams with 4 wt.% contents of short-cut basalt fibers (BFs) were successful prepared by using the modified melt-foaming method. The pore size of BF-containing aluminum foam and commercially pure aluminum foam was counted. The distribution of BF and its effect on the compressive properties of closed-cell aluminum foams were investigated. The results showed that the pore size of BF-containing aluminum foams was more uniform and smaller. BF mainly existed in three different forms: Some were totally embedded in the cell walls, some protruded from the cell walls, and others penetrated through the cells. Meanwhile, under the present condition, BF-containing aluminum foams possessed higher compressive strength and energy absorption characteristics than commercially pure aluminum foams, and the reasons were discussed.

Wogonin is a natural anticancer candidate. The purpose of this study was to explore the pharmacokinetic profiles, tissue distribution, excretion and plasma protein binding of wogonin in Sprague—Dawley rats. A rapid, sensitive, and specific LC-MS/MS method has been developed for the determination of wogonin in different rat biological samples. After i.v. dosing of wogonin at different levels (10, 20 and 40 mg/kg) the elimination half-life was approximately 14 min, the AUC0-∞ increased in a dose disproportional manner from 112.13 mg/L·min for 10 mg/kg to 758.19 mg/L·min for 40 mg/kg, indicating a non linear pharmacokinetic profile. After i.g. dosing at 100 mg/kg, plasma levels of wogonin peaked at 28 min with a Cmax value of 300 ng/mL and a very low oral bioavailability (1.10%). Following i.v. single dose (20 mg/kg), wogonin was detected in all examined tissues (including testis) with the highest levels in kidney and liver. Approximately 21% of the administered dose was excreted as unchanged drug (mainly via non-biliairy fecal route (16.33%). Equilibrium dialysis was used to evaluate plasma protein binding of wogonin at three concentrations (0.1, 0.5 and 2 µg/mL). Results indicated a very high protein binding degree (over 90%), reducing substantially the free fraction of the compound.

Patellofemoral pain syndrome (PFPS) ranks among the most prevalent factors causing anterior knee pain. Quadriceps exercises such as forward lunges are important to treat PFPS. Aim: We investigated whether there is a difference in muscle activity between the vastus medialis oblique (VMO) and vastus lateralis (VL) muscles and the center of pressure (CoP) among three hip positions, namely, hip adduction forward lunge (HADF), hip neutral forward lunge (HNEF), and hip abduction forward lunge (HABF), in healthy subjects. Method: This was a randomized controlled pilot study that included twenty healthy (age: 23.7 ± 2.51) volunteers. The CoP was measured using a “Wii Balance Board” from Nintendo, and VMO and VL muscle activity were measured via wireless surface electromyography. Results: The CoP outcomes showed significant differences among the three positions in terms of position (p < 0.001), sum distance (p < 0.001), range (p < 0.001), and max distance (p < 0.001). VMO (p < 0.000), and VL (p < 0.005) muscle activity and the ratio of VMO/VL (p < 0.000) significantly differed among the three positions. Conclusions: Although VMO and VL muscle activity increased in the HADF and HABF, excessive changes in the CoP occurred in the two positions. The change in the CoP during the HADF and HABF may result in valgus, varus, and loading of the knee joint, which may worsen PFPS. The HNEF is recommended for quadriceps strengthening in healthy subjects and PFPS patients.

The purpose of this study was to compare the effects of a complex shoulder exercise program including respiratory training with those of a similar program including thorax-stabilization exercises on muscle activity and posture in individuals exhibiting scapular internal rotation. The subjects of this study were 32 adults with scapular internal rotation. The exercise group performed the complex shoulder exercise program with respiratory training (EG), while the control group performed the complex shoulder exercise program with thorax-stabilization exercises (CG). After 4 weeks of training, the changes in muscle activity and posture were measured. The differences between the two groups were assessed through comparative analysis using an independent t-test. The paired t-test revealed significant disparities in craniovertebral angle (CVA) and sagittal shoulder angle (SSA) posture between the measurements before and after the programs (p < 0.05). Additionally, the paired t-test indicated notable differences in muscle activity of the upper trapezius (UT), middle trapezius (MT), lower trapezius (LT), and serratus anterior (SA) muscles (p < 0.05). According to the findings, both intervention groups presented significant improvements in cervical and shoulder posture and muscle activity. Consequently, combination of the complex shoulder exercise program with either respiratory training or thorax-stabilization exercises had a favorable impact on posture and muscle activity among individuals exhibiting scapular internal rotation.

Pregabalin is a medication primarily used in the treatment of neuropathic pain and anxiety disorders, owing to its gabapentinoid properties. Pregabalin monotherapy faces limitations due to its variable efficacy and dose-dependent adverse reactions. In this study, we conducted a comprehensive investigation into the potentiation of pregabalin’s analgesic effects by dexborneol, a neuroprotective bicyclic monoterpenoid compound. We performed animal experiments where pain models were induced using two methods: peripheral nerve injury, involving axotomy and ligation of the tibial and common peroneal nerves, and incisional pain through a longitudinal incision in the hind paw, while employing a multifaceted methodology that integrates behavioral pharmacology, molecular biology, neuromorphology, and lipidomics to delve into the mechanisms behind this potentiation. Dexborneol was found to enhance pregabalin’s efficacy by promoting its transportation to the central nervous system, disrupting self-amplifying vicious cycles via the reduction of HMGB1 and ATP release, and exerting significant anti-oxidative effects through modulation of central lipid metabolism. This combination therapy not only boosted pregabalin’s analgesic property but also notably decreased its side effects. Moreover, this therapeutic cocktail exceeded basic pain relief, effectively reducing neuroinflammation and glial cell activation—key factors contributing to persistent and chronic pain. This study paves the way for more tolerable and effective analgesic options, highlighting the potential of dexborneol as an adjuvant to pregabalin therapy.

Dendrobium huoshanense is a famous edible and medicinal herb, and polysaccharides are the main bioactive component in it. In this study, response surface methodology (RSM) combined with a Box–Behnken design (BBD) was used to optimize the enzyme-assisted extraction (EAE), ultrasound–microwave–assisted extraction (UMAE), and hot water extraction (HWE) conditions and obtain the polysaccharides named DHP-E, DHP-UM, and DHP-H. The effects of different extraction methods on the physicochemical properties, structure characteristics, and bioactivity of polysaccharides were compared. The differential thermogravimetric curves indicated that DHP-E showed a broader temperature range during thermal degradation compared with DHP-UM and DHP-H. The SEM results showed that DHP-E displayed an irregular granular structure, but DHP-UM and DHP-H were sponge-like. The results of absolute molecular weight indicated that polysaccharides with higher molecular weight detected in DHP-H and DHP-UM did not appear in DHP-E due to enzymatic degradation. The monosaccharide composition showed that DHPs were all composed of Man, Glc, and Gal but with different proportions. Finally, the glycosidic bond types, which have a significant effect on bioactivity, were decoded with methylation analysis. The results showed that DHPs contained four glycosidic bond types, including Glcp-(1→, →4)-Manp-(1→, →4)-Glcp-(1→, and →4,6)-Manp-(1→ with different ratios. Furthermore, DHP-E exhibited better DPPH and ABTS radical scavenging activities. These findings could provide scientific foundations for selecting appropriate extraction methods to obtain desired bioactivities for applications in the pharmaceutical and functional food industries.

Background and Objectives: To evaluate the efficacy of bladder-prostatic muscle reconstruction and bladder neck eversion anastomosis in the recovery of urinary continence after robot-assisted radical prostatectomy (RARP). Materials and Methods: From January 2020 to May 2022, 69 patients who underwent RARP in our hospital were recruited. Thirty-seven patients underwent RARP with the Veil of Aphrodite technique (control group). On the basis of the control group, 32 patients underwent bladder-prostatic muscle reconstruction and bladder neck eversion anastomosis during RARP (observation group). The recovery of urinary continence was followed up at 24 h and 1, 4, 12, and 24 weeks after catheter removal. Results: There were no significant differences in operative time (127.76 ± 21.23 min vs. 118.85 ± 24.71 min), blood loss (118.27 ± 16.75 mL vs. 110.77 ± 19.63 mL), rate of leakage (3.13% vs. 2.70%), rate of positive surgical margin (6.25% vs. 10.81%), or postoperative Gleason score [7 (6–8) vs. 7 (7–8)] between the observation group and the control group (p > 0.05). After catheter removal, the rates of urinary continence at 24 h, 1 week, 4 weeks, 12 weeks, and 24 weeks were 46.88%, 68.75%, 84.38%, 90.63%, and 93.75% in the observation group, respectively. Meanwhile, the rates of urinary continence in the control group were 21.62%, 37.84%, 62.16%, 86.49%, and 91.89%, respectively. There was a significant difference between the two groups (p = 0.034), especially at 24 h, 1 week, and 4 weeks after catheter removal (p < 0.05). Conclusions: Bladder-prostatic muscle reconstruction and bladder neck eversion anastomosis were beneficial to the recovery of urinary continence after RARP, especially early urinary continence.

Gingerols and shogaols are recognized as active ingredients in ginger and exhibit diverse pharmacological activities. The preclinical pharmacokinetics and tissue distribution investigations of gingerols and shogaols in rats remain less explored, especially for the simultaneous analysis of multi-components. In this study, a rapid, sensitive, selective, and reliable method using an Ultra-Performance Liquid Chromatography Q-Exactive High-Resolution Mass Spectrometer (UPLC-Q-Exactive⁻HRMS) was established and validated for simultaneous determination of eight compounds, including 6-gingerol, 6-shogaol, 8-gingerol, 8-shogaol, 10-gingerol, 10-shogaol, Zingerone, and 6-isodehydrogingenone in plasma and tissues of rats. The analytes were separated on a Syncronis C18 column (100 × 2.1 mm, 1.7 µm) using a gradient elution of acetonitrile and 0.1% formic acid in water at a flow rate of 0.25 mL/min at 30 °C. The method was linear for each ingredient over the investigated range with all correlation coefficients greater than 0.9910. The lowest Lower Limit of quantitation (LLOQ) was 1.0 ng/mL. The intra- and inter-day precisions (Relative Standard Deviation, RSD%) were less than 12.2% and the accuracy (relative error, RE%) ranged from −8.7% to 8.7%. Extraction recovery was 91.4⁻107.4% and the matrix effect was 86.3⁻113.4%. The validated method was successfully applied to investigate the pharmacokinetics and tissue distribution of eight components after oral administration of ginger extract to rats. These results provide useful information about the pharmacokinetics and biodistribution of the multi-component bioactive ingredients of ginger in rats and will contribute to clinical practice and the evaluation of the safety of a Chinese herbal medicine.

Soybean production is significantly impacted by Phytophthora root rot (PRR), which is caused by Phytophthora sojae . The nucleotide-binding leucine-rich repeat (NLR) gene family plays a crucial role in plant disease resistance. However, current understanding of the function of soybean NLR genes in resistance to PRR is limited. To address this knowledge gap, transgenic soybean plants overexpressing the NLR gene ( Glyma.18g283200 ) were generated to elucidate the molecular mechanism of resistance. Here, transcript changes and metabolic differences were investigated at three time points (12, 24, and 36 h) after P. sojae infection in hypocotyls of two soybean lines, Dongnong 50 (susceptible line, WT) and Glyma.18g283200 overexpression line (resistant line, OE). Based on the changes in differentially expressed genes (DEGs) in response to P. sojae infection in different lines and at different time points, it was speculated that HOPZ-ACTIVATED RESISTANCE 1 (ZAR1), valine, leucine, and isoleucine degradation, and phytohormone signaling may be involved in the defense response of soybean to P. sojae at the transcriptome level by GO term and KEGG pathway enrichment analysis. Differentially accumulated metabolites (DAMs) analysis revealed that a total of 223 and 210 differential metabolites were identified in the positive ion (POS) and negative ion (NEG) modes, respectively. An integrated pathway-level analysis of transcriptomics (obtained by RNA-seq) and metabolomics data revealed that isoflavone biosynthesis was associated with disease resistance. This work provides valuable insights that can be used in breeding programs aiming to enhance soybean resistance against PRR.
