This comprehensive review investigates the potential of cyanobacteria, particularly nitrogen-fixing strains, in addressing global challenges pertaining to plastic pollution and carbon emissions. By analyzing the distinctive characteristics of cyanobacteria, including their minimal growth requirements, high photosynthetic efficiency, and rapid growth rates, this study elucidates their crucial role in transforming carbon sequestration, biofuel generation, and biodegradable plastic production. The investigation emphasizes cyanobacteria’s efficiency in photosynthesis, positioning them as optimal candidates for cost-effective bioplastic production with minimized land usage. Furthermore, the study explores their unconventional yet promising utilization in biodiesel production, mitigating environmental concerns such as sulfur emissions and the presence of aromatic hydrocarbons. The resulting biodiesel exhibits significant combustion potential, establishing cyanobacteria as a viable option for sustainable biofuel production. Through a comprehensive assessment of both achievements and challenges encountered during the commercialization process, this review offers valuable insights into the diverse contributions of cyanobacteria. Its objective is to provide guidance to researchers, policymakers, and industries interested in harnessing bio-inspired approaches for structural and sustainable applications, thereby advancing global efforts towards environmentally conscious plastic and biofuel production.

In electric vehicles (EVs), the batteries are arranged in the battery pack (BP), which has a small layout space and difficulty in dissipating heat. Therefore, in EVs, the battery thermal management systems (BTMSs) are critical to managing heat to ensure safety and performance, particularly under higher operating temperatures and longer discharge conditions. To solve this problem, in this article, the thermal analysis models of a 3-battery-cell BP were created, including scenarios (1) natural air cooling without a BTMS; (2) natural air cooling with water cooling hybrid BTMS; and (3) forced air cooling plus water cooling composite BTMS. The thermal performances of the pack-level BPs were simulated and analyzed based on computational fluid dynamics (CFD). A variety of boundary conditions and working parameters, such as ambient temperature, inlet coolant flow rate and initial temperature, discharge rate, air flow rate, and initial temperature, were considered. The results show that without a BTMS (Scenario 1), the maximum temperature in the BP rises rapidly and continuously to reach 63.8 °C, much higher than the upper bound of the recommended operating temperature range (ROTR between +20 °C to +35 °C) under the extreme discharge rate of 3 C and even if the discharge rate is 2 C. With a hybrid BTMS (Scenario 2), the maximum temperature in BP rises to about 38.7 °C, slightly above the upper bound of the ROTR. Lowering the coolant (water) initial temperature can effectively lower the temperature up to 5.7 °C in BP, but the water flow rate cannot since the turbulence model. While with a composite BTMS (Scenario 3), the temperature can be further lowered up to 1.5 °C under the extreme discharge rate of 3C, just reaching the upper bound of the ROTR. In addition, lowering the initial coolant temperature or air temperature can effectively decrease the temperatures up to 5.1 and 1.0 °C, respectively, in BP, but the coolant flow rate (due to the turbulence model) and the air flow rate cannot. Finally, the thermal performances of the different battery cells in the BP with different cooling systems and at the different positions of the BP were compared and analyzed. The present work may contribute to the design of BTMSs in the EV industry.

The sophisticated, elegant protein-polymers designed by nature can serve as inspiration to redesign and biomanufacture protein-based materials using synthetic biology. Historically, petro-based polymeric materials have dominated industrial activities, consequently transforming our way of living. While this benefits humans, the fabrication and disposal of these materials causes environmental sustainability challenges. Fortunately, protein-based biopolymers can compete with and potentially surpass the performance of petro-based polymers because they can be biologically produced and degraded in an environmentally friendly fashion. This paper reviews four groups of protein-based polymers, including fibrous proteins (collagen, silk fibroin, fibrillin, and keratin), elastomeric proteins (elastin, resilin, and wheat glutenin), adhesive/matrix proteins (spongin and conchiolin), and cyanophycin. We discuss the connection between protein sequence, structure, function, and biomimetic applications. Protein engineering techniques, such as directed evolution and rational design, can be used to improve the functionality of natural protein-based materials. For example, the inclusion of specific protein domains, particularly those observed in structural proteins, such as silk and collagen, enables the creation of novel biomimetic materials with exceptional mechanical properties and adaptability. This review also discusses recent advancements in the production and application of new protein-based materials through the approach of synthetic biology combined biomimetics, providing insight for future research and development of cutting-edge bio-inspired products. Protein-based polymers that utilize nature’s designs as a base, then modified by advancements at the intersection of biology and engineering, may provide mankind with more sustainable products.

The ever-increasing requirements for structural performance drive the research and development of lighter, stronger, tougher, and multifunctional composite materials, especially, the lattice structures, heterogeneities, or hybrid compositions have attracted great interest from the materials research community. If it is pushed to the extreme, these concepts can consist of highly controlled lattice structures subject to biomimetic material design and topology optimization (TO). However, the strong coupling among the composition and the topology of the porous microstructure hinders the conventional trial-and-error approaches. In this work, discontinuous carbon fiber-reinforced polymer matrix composite materials were adopted for structural design. A three-dimensional (3D) periodic lattice block inspired by cuttlefish bone combined with computer modeling-based topology optimization was proposed. Through computer modeling, complex 3D periodic lattice blocks with various porosities were topologically optimized and realized, and the mechanical properties of the topology-optimized lattice structures were characterized by computer modeling. The results of this work were compared with other similar designs and experiments to validate the effectiveness of the proposed method. The proposed approach provides a design tool for more affordable and higher-performance structural materials.

The yeast Saccharomyces cerevisiae has been widely applied in fermentation industries, chemical industries and biological research and it is widespread in different environments, especially in sugar-rich environments. However, little is known about the occurrence, distribution and roles of S. cerevisiae in marine environments. In this study, only 10 strains among all the yeasts isolated from different marine environments belonged to S. cerevisiae. It was found that most of the strains of S. cerevisiae in marine environments occurred in guts, the surface of marine fish and mangrove trees. In contrast, they were not found in seawater and sediments. All the strains of S. cerevisiae isolated from the marine environments had a lower ability to produce ethanol than the highly alcohol-producing yeast Saccharomyces sp. W0 isolated from fermented rice, but the strains 2E00400, 2E00558, 2E00498, 2E00723, 2E00724 could produce higher concentrations of ethanol than any other marine-derived strains of S. cerevisiae obtained in this study. However, some of them had higher ethanol tolerance and higher trehalose content than Saccharomyces sp. W0. In particular, ethanol tolerance of the yeast strain 2E00498 was higher than that of Saccharomyces sp. W0. This may be related to the harsh marine environments from which they were isolated. Such yeast strains with higher alcohol tolerance could be used to further improve the alcohol tolerance of Saccharomyces sp. W0.

Four new ansamycins, named divergolides T–W (1–4), along with two known analogs were isolated from the fermentation broth of the mangrove-derived actinomycete Streptomyces sp. KFD18. The structures of the compounds, including the absolute configurations of their stereogenic carbons, were determined by spectroscopic data and single-crystal X-ray diffraction analysis. Compounds 1–4 showed cytotoxic activity against the human gastric cancer cell line SGC-7901, the human leukemic cell line K562, the HeLa cell line, and the human lung carcinoma cell line A549, with 1 being the most active while compounds 5 and 6 were inactive against all the tested cell lines. Compounds 1 and 3 showed very potent and specific cytotoxic activities (IC50 2.8 and 4.7 µM, respectively) against the SGC-7901 cells. Further, the apoptosis-inducing effect of 1 and 3 against SGC-7901 cells was demonstrated by two kinds of staining methods for the first time.

Neoagaro-oligosaccharides (NAOS) have health benefits that are related to their amount and degree of polymerization (DP). However, the current methods that are used to quantify enzymatically released NAOS are un-specific and time-consuming. Agar has been extracted from Gelidium amansii and has been degraded by AgaXa (a recombinant β-agarase). Polysaccharide analysis using carbohydrate gel electrophoresis (PACE) has been adapted in order to quantify NAOS. In addition, the anti-oxidant activity of the degraded samples has been assessed. We have found that the PACE method provided sensitive, precise, and accurate quantification for each of the six NAOS samples. PACE has revealed that the DP of the enzymatic products from the AgaXa digestion were mainly neoagaro-octaose and neoagaro-decaose. The degraded samples exhibited increased radical-scavenging activity towards 2,2-diphenyl-1-picrylhydrazyl and 2,2-azino-bis(3-ethylbenzothiazoline sulfonic acid) radicals. While the anti-oxidant activity may have been from NAOS activity and contributions from neoagaro-octaose and neoagaro-decaose. The adapted PACE method that has been presented here is promising for large sample analysis during quality control and for characterizing novel β-agarase degradation mechanisms.

This study seeks to assess the impact of varying concentrations of lysophospholipids on the antioxidant capacity, digestive performance, and intestinal microbiota of L. vannamei. A total of 840 shrimp, with an average initial weight of 2.22 ± 0.11 g, were randomly distributed across 28 aquaculture tanks (30 shrimp per tank), organized into 7 distinct treatment groups, each comprising 4 replicates. The control group (DL2) was administered a basal diet formula supplemented with 2% soy lecithin, whereas the experimental groups received lysophospholipids at varying concentrations of 0% (RL0), 0.1% (RL0.1), 0.5% (RL0.5), 1% (RL1), 1.5% (RL1.5), and 2% (RL2). The results revealed that the total antioxidant capacity (T-AOC) level in the RL0.1 group was significantly elevated compared to the DL2 and RL2 groups (p < 0.05). Furthermore, glutathione peroxidase (GSH-Px) and catalase (CAT) activities were markedly higher in the RL1 and RL1.5 groups relative to other groups (p < 0.05). Superoxide dismutase (SOD) activity exhibited a significant increase across all lysophospholipid-supplemented groups when compared to the DL2 group (p < 0.05). Malondialdehyde (MDA) levels were notably elevated in the RL0.5, RL1.5, and RL2 groups relative to the other groups (p < 0.05). Regarding antioxidant-related genes in the hemocytes, the relative expression levels of Nrf1, Nrf2, GPx, SOD, CAT, and Hippo were significantly higher in the RL0.5 and RL1 groups compared to the DL2 group (p < 0.05). Additionally, the relative expression levels of GPx and SOD were significantly elevated in the RL2 group compared to the DL2 group (p < 0.05). In the intestinal tract, the activities of trypsin and α-amylase (AMS) were significantly elevated in the RL0.1, RL0.5, and RL1 groups compared to the DL2 group (p < 0.05). Additionally, the RL0.1 group demonstrated significantly higher lipase (LPS) activity than the other groups supplemented with lysophospholipids (p < 0.05). Furthermore, the relative expression levels of the trypsin and α-amylase genes were significantly increased in the RL1 and RL1.5 groups in comparison to the DL2 group (p < 0.05). Sequencing analysis of the intestinal microbiota indicated that the incorporation of lysophospholipids resulted in an enhancement of the composition and structure of the intestinal microbiota. The functional abundance of the intestinal microbiome was primarily enriched in metabolic pathways. Overall, the incorporation of lysophospholipids into the diet exerted a beneficial effect on the antioxidant capacity, digestive performance, and intestinal microbiota of L. vannamei, especially with the supplementation of 0.1% lysophospholipids.

This study was conducted to investigate the impacts of the dietary addition of taurine and enzymatic cottonseed protein concentrate (ECPC) in low-fishmeal diet on the growth performance, plasma biochemical indices, hepatic antioxidant capacity, intestinal anti-inflammatory capacity, intestinal microflora, and muscle quality of golden pompano (Trachinotus ovatus). A total of three isonitrogenous diets were given to 225 golden pompanos (5.6 ± 0.14 g). They were randomly divided into nine cages (1.0 m × 1.0 m × 1.5 m; three cages per treatment) with equal stocking numbers of twenty-five fish per cage. The results indicated that the CSM-TC group significantly increased the growth performance of juvenile T. ovatus (p < 0.05). The results indicated that compared with other groups, the addition of 1% ECPC and 0.25% taurine has been found to enhance the WGR (weight gain rate), SGR (specific growth rate), and CF (condition factor). Compared with other groups, the relative expressions of GH, GHR1, GHR2, IGF1, IGF2, and MyoG were significantly higher in fish fed with CSM-TC. The results showed that CSM-TC significantly increased the activities of alkaline phosphatase, complement 3, and complement 4 enzymes (p < 0.05). The results showed that dietary CSM-TC increased the activities of hepatic superoxide dismutase and total antioxidant capacity enzymes. Compared with other groups, the hepatic relative expressions of Nrf2, HO-1, and GSH-Px were significantly higher in fish fed with CSM-TC. The results showed that dietary CSM-TC increased the activities of intestinal chymotrypsin, lipase, and α-amylase enzymes. A CSM-TC diet significantly increased the relative expressions of IL-10, ZO-1, Occludin, Claudin-3, and Claudin-15 (p < 0.05). The results showed that CSM-C significantly increased the index of Ace and Chao1 (p < 0.05). In conclusion, a high-fermented cottonseed meal diet can have detrimental effects on physiological health in golden pompano, while adding 1% ECPC and 0.25% taurine can improve hepatic and intestinal health via attenuating inflammation and oxidative stress.

Poliovirus (PV) is on the brink of eradication due to global vaccination programs utilizing live-attenuated oral and inactivated polio vaccines. Recombinant PV virus-like particles (VLPs) are emerging as a safe next-generation vaccine candidate for the impending polio-free era. In this study, we investigate the production, antigenicity, thermostability, immunogenicity, and structures of VLPs derived from PV serotype 2 (PV2) wildtype strain and thermally stabilized mutant (wtVLP and sVLP, respectively). Both PV2 wtVLP and sVLP are efficiently produced in Pichia pastoris yeast. The PV2 sVLP displays higher levels of D-antigen and significantly enhanced thermostability than the wtVLP. Unlike the wtVLP, the sVLP elicits neutralizing antibodies in mice at levels comparable to those induced by inactivated polio vaccine. The addition of an aluminum hydroxide adjuvant to sVLP results in faster induction and a higher magnitude of neutralizing antibodies. Furthermore, our cryo-EM structural study of both sVLP and wtVLP reveals a native conformation for the sVLP and a non-native expanded conformation for the wtVLP. Our work not only validates the yeast-produced PV2 sVLP as a promising vaccine candidate with high production potential but also sheds light on the structural mechanisms that underpin the assembly and immunogenicity of the PV2 sVLP. These findings may expedite the development of sVLP-based PV vaccines.

Several exogenous probiotics are applicable in fish culture; however, challenges in isolation and verification have hindered the full utilization of numerous host probiotics. Therefore, this study aimed to apply the host probiotic Exiguobacterium acetylicum G1-33 to hybrid grouper (Epinephelus fuscoguttatus ♀ × Epinephelus lanceolatus ♂) cultures and explore its mechanism of action. In total, 360 hybrid grouper were divided into four groups, which were fed the following for 60 days: three received commercial feed with varying concentrations of E. acetylicum G1-33 (106, 108, and 1010 CFU/g), while a control group received commercial feed. The results showed that supplementation with 106 and 108 CFU/g of E. acetylicum G1-33 enhanced gut morphology, upregulated growth-related genes (ghr1, igf-2, s6k1, tor), and promoted growth, with supplementation with 108 CFU/g resulting in the most notable enhancement. However, supplementation with 1010 CFU/g inhibited growth, possibly because of changes in intestinal morphology. Additionally, supplementation with E. acetylicum G1-33 upregulated the expression of immune-related genes (c3, myd88, Cu/Zn-sod, tlr3, and tnf2) in the liver and head kidney but led to an increase in malondialdehyde content, as well as a decrease in alkaline phosphatase and acid phosphatase activities, in the liver and serum, indicating increased oxidative stress. Moreover, supplementation with 106 and 108 CFU/g E. acetylicum G1-33 enhanced the widespread expression of immune-related genes in the head kidney and liver, respectively, and improved resistance to Vibrio harveyi, whereas supplementation with 1010 CFU/g weakened this resistance. In conclusion, E. acetylicum G1-33, particularly at 108 CFU/g, emerged as an effective probiotic, optimizing growth performance and immunity in hybrid grouper. This research is pioneering in its application of E. acetylicum in mariculture, potentially broadening the range of probiotic strategies in aquaculture.

The development of wind power has brought about increasing challenges in decommissioning, among which DWTBs (decommissioned wind turbine blades) are the most difficult component to deal with. To enable the cost-effective, energy-efficient, and environmentally friendly large-scale utilization of DWTBs, an experimental study on thermogravimetric and pyrolysis characteristics of DWTBs was carried out. A new process involving recycling glass fiber with pyrolysis gas re-combustion and flue gas recirculation as the pyrolysis medium was innovatively proposed, and the simulation calculation was carried out. Thermogravimetric experiments indicated that glass fiber reinforced composite (GFRC) was the main heat-generating part in the heat utilization process of blades, and the blade material could basically complete pyrolysis at 600 °C. As the heating rate increased, the formation temperature, peak concentration, and proportion of combustible gas in the pyrolysis gas also increased. The highest peak concentration of CO gas was observed, with CO2 and C3H6 reaching their peaks at 700 °C. The solid product obtained from pyrolysis at 600 °C could be oxidized at 550 °C for 40 min to obtain clean glass fiber. And the pyrolysis temperature increased with the increase in the proportion of recirculation flue gas. When the proportion of recirculation flue gas was 66%, the pyrolysis temperature could reach 600 °C, meeting the necessary pyrolysis temperature for wind turbine blade materials. The above research provided fundamental data support for further exploration on high-value-added recycling of DWTBs.

Cable-driven parallel robots (CDPRs) have been gaining much attention due to their many advantages over traditional parallel robots or serial robots, such as their markedly large workspace and lightweight design. However, one of the main issues that needs to be urgently solved is the tension in the distribution of CDPRs due to two reasons. The first is that a cable can only be stretched but not compressed, and the other is the redundancy of the parallel robot. To address the problem, an optimization method for tension distribution is proposed in the paper. The structural design of the parallel robot is first discussed. The dynamics model of the parallel robot is established by the Newton–Euler method. Based on the minimum variance of cables’ tension, an optimization method of tension distribution is presented for the parallel robot. Furthermore, the tension extreme average term is introduced in the optimization method, and the firefly algorithm is applied to obtain the optimal solution for tension distribution. Finally, the proposed approach is tested in the simulation case where the end-effector of the robot moves in a circular motion. Simulation results demonstrate that the uniformity and continuity of tension are both outstanding for the proposed method. In contrast with traditional solving methods, the efficiency of this method is largely improved.

This paper investigates the bumpless transfer control of linear switched positive systems based on state and disturbance observers. First, state and disturbance observers are designed for linear switched positive systems to estimate the state and the disturbance. By combining the designed state observer, the disturbance observer, and the output, a new controller is constructed for the systems. All gain matrices are described in the form of linear programming. By using co-positive Lyapunov functions, the positivity and stability of the closed-loop system can be ensured. In order to achieve the bumpless transfer property, some additional sufficient conditions are imposed on the control conditions. The novelties of this paper lie in that (i) a novel framework is presented for positive disturbance observer, (ii) double observers are constructed for linear switched positive systems, and (iii) a bumpless transfer controller is proposed in terms of linear programming. Finally, two examples are given to illustrate the effectiveness of the proposed results.

This research sought to assess the effects of dietary supplements with Gracilaria lichenoides and Bacillus amyloliquefaciens, either individually or combined, on the growth performance, antioxidant capacity, and intestinal function of Penaeus monodon. A total of 840 shrimps were randomly assigned to 28 tanks with an average initial weight of (1.04 ± 0.03) g (30 shrimp per tank) with 7 different treatment groups and 4 replicates per treatment. The control treatment (C) consisted of a basal diet; in contrast, the experimental groups were complement with varying levels of G. lichenoides (3% or 8%), either alone (S3 and S8) or in combination with B.amyloliquefaciens at different concentrations (3% G. lichenoides and 109 CFU/g—S3B9; 8% G. lichenoides and 1011 CFU/g B. amyloliquefaciens—S8B11; 109 CFU/g B. amyloliquefaciens—S9; 1011 CFU/g B. amyloliquefaciens—B11). The results indicated that the maximum values of final body weight (FBW) (10.49 ± 0.90) g, weight gain rate (WGR) (908.94 ± 33.58) g, and specific growth rate (SGR) (4.20 ± 0.06) g were perceived in the 3% G. lichenoide diet treatment, and compared with the control group, the difference was significant (p < 0.05). The whole-body lipid content of shrimp in the B9 group was significantly higher than that in the B11 group (p < 0.05), but no significant difference was observed when compared with shrimp fed other diets (p > 0.05). The ash content of shrimp in the B9 group was found to be significantly higher than that in the S3B9 group (p < 0.05). Furthermore, the lipase activity in the stomach and intestines of the experimental groups exhibited a statistically significantly increase compared to the control (p < 0.05). In comparison to the control group, the hepatopancreas of the S3 group exhibited a significant increase in the activities of glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and antioxidant genes [SOD, catalase (CAT), GSH-Px, thioredoxin (Trx), Hippo, and NF-E2-related factor 2 (Nrf2)] expression levels (p < 0.05). Additionally, the activities of total antioxidant capacity (T-AOC), SOD, peroxidase (POD), and antioxidant genes (CAT, GSH-Px, Trx, and Hippo) in the S3B9 treatment of hepatopancreas showed significant improvement (p < 0.05). The inclusion of dietary G. lichenoides and B. amyloliquefaciens resulted in enhanced relative expression of intestinal lipid metabolism genes (fatty acid synthetase (FAS), lipophorin receptor (LR), fatty acid transport protein 1 (FATP1)) and suppressed the expression of the long-chain fatty acid-CoA ligase 4 (LCL4) gene. Analysis of microbiota sequencing indicated improvements in composition and structure, with notable increases in Firmicutes at the phylum level and Vibrio at the genus level in the S3 group, as well as an increase in Tenericutes at the genus level in the S8B11 group. Overall, the inclusion of dietary G. lichenoides and B. amyloliquefaciens positively impacted the growth, antioxidant capacity, and microbial composition of shrimp, with particular enhancement observed in shrimp fed a supplementary 3% G. lichenoides diet.

The rabbitfish, Siganus oramin, is a commercially important table fish in southeastern China. However, there have been few studies on its gonad development and reproduction regulation. Comparative transcriptome analysis was first performed on adult male and female gonads of S. oramin. In total, 47,070 unigenes were successfully assembled and 22,737 unigenes were successfully annotated. Through comparative transcriptome analysis of male and female gonads, a total of 6722 differentially expressed genes were successfully identified, with 3528 upregulated genes and 3154 downregulated genes in the testes. In addition, 39 differentially expressed reproduction-related genes were identified. Finally, quantitative real-time PCR was used to validate the expression levels of several differentially expressed genes. These results provide important data for further studying the function of reproduction-related genes and the molecular mechanism regulating gonad development and reproduction in S. oramin.

Coilia nasus is a typical anadromous migratory fish found in the lower reaches of the Yangtze River. Every year, C. nasus clusters offshore and swims upstream along the Yangtze River into the tributaries and lakes in the middle and lower reaches of the Yangtze River to breed. In this study, female C. nasus were collected as study subjects from the Chongming section of Shanghai, the Taizhou section of Jiangsu, and the Anqing section of Anhui. Their ovaries were used to examine tissue sections and investigate gene expression, including the follicle-stimulating hormone receptor (fshr), the luteinizing hormone receptor (lhr), kisspeptin-1 (kiss1), and forkhead box l2 (foxl2), which are related to reproductive development, while the serum levels of estrogen (including estradiol, E2) and progestins (including 17α,20β-dihydroxy-4-pregenen-3-one, 17α,20β-DHP) were also analyzed. Our results showed that, first, the growth period of the oocytes was small in stage II of ovarian development, in which both E2 and 17α,20β-DHP levels and gene expression were low. Then, in stage III, the growth period of the oocytes became large, and the yolk granules and oil droplets began to appear. Simultaneously, E2 and the expression of kiss1 and foxl2 were significantly elevated. Finally, stage IV was the period of a large amount of accumulation of nutrients in the oocytes, and 17α,20β-DHP levels and the expression of fshr and lhr were significantly elevated. These results enrich the theoretical study of ovarian development in the natural population of C. nasus, supplementing the biological basis of C. nasus reproduction and scientifically supporting the study of C. nasus population ecology and resource conservation.

The Yangtze finless porpoises (Neophocaena asiaeorientalis asiaeorientalis) living in different environments display significant differences in behavior and physiology. To compare and analyze gene expression differences between an ex situ population and a controlled environment population of the Yangtze finless porpoise, we sequenced the transcriptome of blood tissues living in a semi-natural reserve and an artificial facility, respectively. We identified 6860 differentially expressed genes (DEGs), of which 6603 were up-regulated and 257 were down-regulated in the controlled environment vs ex situ comparison. GO and KEGG enrichment analysis showed that the up-regulated genes in the controlled environment population were significantly associated with glucose metabolism, amino acid metabolism, and the nervous system, while those up-regulated in the ex situ population were significantly associated with energy supply and biosynthesis. Further analysis showed that metabolic and hearing-related genes were significantly affected by changes in the environment, and key metabolic genes such as HK, PFK, IDH, and GLS and key hearing-related genes such as OTOA, OTOF, SLC38A1, and GABBR2 were identified. These results suggest that the controlled environment population may have enhanced glucose metabolic ability via activation of glycolysis/gluconeogenesis, the TCA cycle, and inositol phosphate metabolism, while the ex situ population may meet higher energy requirements via enhancement of the amino acid metabolism of the liver and muscle and oxidative phosphorylation. Additionally, the acoustic behavior and auditory-related genes of Yangtze finless porpoise may show responsive changes and differential expression under different environment conditions, and thus the auditory sensitivity may also show corresponding adaptive characteristics. This study provides a new perspective for further exploration of the responsive changes of the two populations to various environments and provides a theoretical reference for further improvements in conservation practices for the Yangtze finless porpoise.

Neurological diseases can lead to the denervation of brain regions caused by demyelination, traumatic injury or cell death. The molecular and structural mechanisms underlying lesion-induced reorganization of denervated brain regions, however, are a matter of ongoing investigation. In order to address this issue, we performed an entorhinal cortex lesion (ECL) in mouse organotypic entorhino-hippocampal tissue cultures of both sexes and studied denervation-induced plasticity of mossy fiber synapses, which connect dentate granule cells (dGCs) with CA3 pyramidal cells (CA3-PCs) and play important roles in learning and memory formation. Partial denervation caused a strengthening of excitatory neurotransmission in dGCs, CA3-PCs and their direct synaptic connections, as revealed by paired recordings (dGC-to-CA3-PC). These functional changes were accompanied by ultrastructural reorganization of mossy fiber synapses, which regularly contain the plasticity-regulating protein synaptopodin and the spine apparatus organelle. We demonstrate that the spine apparatus organelle and synaptopodin are related to ribosomes in close proximity to synaptic sites and reveal a synaptopodin-related transcriptome. Notably, synaptopodin-deficient tissue preparations that lack the spine apparatus organelle failed to express lesion-induced synaptic adjustments. Hence, synaptopodin and the spine apparatus organelle play a crucial role in regulating lesion-induced synaptic plasticity at hippocampal mossy fiber synapses.

With the rapid development of the new energy vehicle industry, the number of power battery decommissioning is increasing year by year. The recycling of power batteries is of great significance for protecting the ecological environment, improving the efficiency of resource utilization, and ensuring the sustainable and healthy development of the new energy automobile industry. In this study, the chemical compositions of power batteries were introduced, the technical path and development status of the echelon utilization of decommissioned power batteries were discussed, and the specific steps and challenges of regenerative utilization of decommissioned power batteries were described in detail from two aspects of pyrometallurgy and hydrometallurgy. Combined with the relevant research results, the main methods of the direct regeneration of positive electrode materials were analyzed. Finally, the main development direction and related suggestions for the resource utilization of decommissioned power batteries were put forward.

The Yangtze finless porpoise (Neophocaena asiaeorientalis asiaeorientalis, YFP) is an endangered species endemic to the Yangtze River in China, and it is the only freshwater whale in the genus Neophocaena. In terms of protection, three effective conservation strategies exist: in situ conservation, ex situ conservation, and artificial breeding, all of which have been implemented by the Chinese government. Of these, ex situ conservation involves the relocation of Yangtze finless porpoises to semi-natural waters with less human interference, and artificial breeding involves the relocation of Yangtze finless porpoises to a controlled environment that is more strictly managed. To compare and analyze the responsive changes in gene expression of the YFPs between the ex situ and controlled environments, we performed the RNA sequencing of blood tissues from these YFPs. A total of 1201 differentially expressed genes (DEGs) were identified, of which 423 were up-regulated in the ex situ population and 778 were up-regulated in the controlled-environment population. Gene enrichment analysis showed that 1201 DEGs between the ex situ and controlled-environment populations were generally enriched for vision-, digestion- and immune-system-related pathways. Further analysis revealed that several key immune system pathways, such as the chemokine signaling pathway and B cell receptor signal pathway, were activated in the ex situ population. In addition, the key pathways related to vision, including phototransduction and the inflammatory mediator regulation of TRP channels, as well as the pathways related to the digestive system, such as protein digestion and absorption and salivary secretion, were activated in the controlled-environment population. These results suggest that the ex situ populations may respond to complex environmental conditions in semi-natural waters by enhancing their immune function through the increased expression of immune-related genes and that the visual function and protein digestion of the YFPs were improved compared to those of the ex situ population based on the conditions of artificial feeding, such as the higher transparency of the water and regular feeding. This study provides clues for evaluating the adaptability of YFPs to different environments and is a useful reference for future ex situ conservation and artificial breeding.

The present study was conducted to investigate the effects of dietary fermented cottonseed meal (FCSM) substitution for fish meal on intestinal enzymatic activity, inflammatory and physical-barrier-related gene expression, and intestinal microflora of juvenile golden pompano. The 375 golden pompanos were divided into 15 groups of 25 fish each, with three replicates for each experimental group. The fish were fed five experimental diets (0 (FM), 12.5% (CSM12.5), 25% (CSM25), 50% (CSM50), and 100% (CSM100) substitution levels) for 8 weeks. The fish were reared and fed the experimental diets under a natural-day light cycle. Compared with the control group, the activities of AMY (amylase) enzymes in the CSM12.5 group and all other groups were elevated (p < 0.05). The CSM25 group exhibited a considerable up-regulation of IL-10 (Interleukin-10) expression relative to the FM group (p < 0.05). With an increase in dietary FM substitution with FCSM from 0 to 25%, the relative expressions of NF-κB (Nuclear factor kappa-B), IL-1β (Interleukin-1 beta), and IL-8 (Interleukin-8) were down-regulated. In this study, the relative expressions of ZO-1 (zonula occluden-1) and Occludin were up-regulated, and those of Claudin-3 and Claudin-15 significantly up-regulated, when the FCSM substitution ratio was 25%. The results of high-throughput sequencing of the intestinal microflora showed that ACE indices the lowest in the CSM25 group, which was significantly different from those in the CSM100 group (p < 0.05). The CSM50 group had the highest Shannon and Simpson indices and the highest community diversity. In addition, replacing a high percentage of fish meal with FCSM can negatively affect the intestinal flora of fish. In this study, the 25% substitution ratio improved nutrient absorption, reduced intestinal inflammation, improved intestinal physical barrier damage, did not affect intestinal microecology, and had no adverse effects on fish. However, substitution of a high proportion of FM with FCSM negatively affects the intestinal microflora and nutrient absorption capacity of fish.
