Strong-alkali activation is a prerequisite needed to ensure the full polymerization activity of alkali slag binder and establish excellent mechanical properties; however, it substantially increases the preparation cost. In this study, the effects of both strong and weak alkaline activators on the activation performance of alkali slag were examined, using a combination of X-ray diffraction, scanning electron microscopy, and Fourier-transform infrared spectroscopy analysis methods. The reaction mechanism was analyzed under different alkaline conditions, and the preparation cost could be significantly reduced without significantly degrading mechanical properties. The results indicate that Ca(OH)2 can stimulate the reactivity of slag, resulting in a 40% decrease in compressive strength (compared to NaOH) but a 25–50% reduction in preparation cost. With increasing Ca(OH)2 dosage, the compressive strength first increases and then decreases. The best excitation effect is achieved at a dosage of 40 g Ca(OH)2 per 450 g GBFS. The formation of aluminosilicate is the main driving force for the observed increase in compressive strength. Excessive dosage of Ca(OH)2 will lead to its deposition in the specimen, thus affecting the development of compressive strength.

The objective of this study was to compile the currently available evidence regarding the functional and morphologic outcomes of functional orthodontic therapy for mandibular condyle fracture. We performed searches in PubMed and Google Scholar as well as manually (IOK issues 2008–2019) using the keywords “trauma”, “TMJ”, “activator”, “condylar fracture”, “fracture”, “mandibular condylar fracture”, “occlusal splint” and “functional appliance”. Screening and analysis of study eligibility were performed using the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA). The search strategy identified 198 studies published between 1971 and 2018, with 93 studies remaining after removing duplicate hits. Of the 93 studies, 19 were included in this study, considering the inclusion criteria. There were 12 follow-up, 4 prospective, and 3 purely retrospective studies. Some of the studies showed good functional results of mandibular condyle fracture treatment, in addition to subjective patient satisfaction. The incisal edge difference could be increased to physiological ranges of >35 mm by means of activator therapy. Partial mandibular deviations to the fractured side remained post-therapy, especially after unilateral fractures. Fractures without significant dislocation and luxation showed radiographic changes in shape, described as hypoplastic and ellipsoid, in addition to good morphologic results. One study found that collum length shortened twofold after a luxated fracture compared with fractures without significant dislocation, despite activator therapy. Straightening of the fragment occurred only in the low-dislocated fragments. Overall, children showed a higher remodeling potential than adult patients. Several studies observed an improved clinical outcome for functional therapy after mandibular condyle fracture. The outcome is essentially determined by fracture type, fracture height, and age. Further studies, especially prospective studies, are necessary to improve the evidence of functional orthodontic therapy for mandibular condyle fractures.

In industrial manufacturing, pyrrhotite(Fe1−xS), once depressed, is commonly activated for flotation. However, the replacement of CuSO4 is necessary due to the need for exact control over the dosage during the activation of pyrrhotite, which can pose challenges in industrial settings. This research introduces the use of FeSO4 for the first time to efficiently activate pyrrhotite. The impact of two different activators on pyrrhotite was examined through microflotation experiments and density functional theory (DFT) calculations. Microflotation experiments confirmed that as the CuSO4 dosage increased from 0 to 8 × 10−4 mol/L, the recovery of pyrrhotite initially increased slightly from 71.27% to 87.65% but then sharply decreased to 16.47%. Conversely, when the FeSO4 dosage was increased from 0 to 8 × 10−4 mol/L, pyrrhotite’s recovery rose from 71.27% to 82.37%. These results indicate a higher sensitivity of CuSO4 to dosage variations, suggesting that minor alterations in dosage can significantly impact its efficacy under certain experimental conditions. In contrast, FeSO4 might demonstrate reduced sensitivity to changes in dosage, leading to more consistent performance. Fe ions can chemically adsorb onto the surface of pyrrhotite (001), creating a stable chemical bond, thereby markedly activating pyrrhotite. The addition of butyl xanthate (BX), coupled with the action of Fe2+ on activated pyrrhotite, results in the formation of four Fe-S bonds on Fe2+. The proximity of their atomic distances contributes to the development of a stable double-chelate structure. The S 3p orbital on BX hybridizes with the Fe 3d orbital on pyrrhotite, but the hybrid effect of Fe2+ activation is stronger than that of nonactivation. In addition, the Fe-S bond formed by the addition of activated Fe2+ has a higher Mulliken population, more charge overlap, and stronger covalent bonds. Therefore, Fe2+ is an excellent, efficient, and stable pyrrhotite activator.

To promote the large-scale recycling of solid waste, the hydration characteristics of blended cement with different amounts of GGBS (ground granulated blast-furnace slag) and SS (steel slag) were investigated. The optimum blending amounts of GGBS and SS in cement were 40% and 10% by mass, and the optimum dosage of Na2SO4 in the C50-S40-SS10 (50 wt.% cement–40 wt.% slag–10 wt.% steel slag) system was 2 wt.%. The flexural and compressive strengths of the C50-S40-SS10 system after adding 2 wt.% Na2SO4 are 57.95% and 9.28% higher than that of pure cement at 28 d. XRD, FT-IR and Ca(OH)2 content analysis were chosen to investigate the hydration products of pure cement and blended cement. The results show that GGBS enhanced the hydration of both cement and SS. And GGBS contributed to the generation of calcium silicoaluminate hydrate (C–A–S–H) in the blended cement system. The addition of Na2SO4 promoted the hydration reaction and contributed to the generation of ettringite (AFt) in the ternary system. The hydration heat evolution results showed that GGBS and SS can reduce the hydration heat of cement. Na2SO4 had similar effects and delayed the time of AFt conversion to monosulfide calcium sulphoaluminate (AFm). A mutual activation mechanism of cement–GGBS–SS ternary system mixed with Na2SO4 was proposed in this study.

The SH2-containing inositol polyphosphate 5-phosphatase 1 (SHIP1) enzyme opposes the activity of PI3K and therefore is of interest in the treatment of inflammatory disorders. Recent results also indicate that SHIP1 promotes phagolysosomal degradation of lipids by microglia, suggesting that the enzyme may be a target for the treatment of Alzheimer’s disease. Therefore, small molecules that increase SHIP1 activity may have benefits in these areas. Recently we discovered a bis-sulfonamide that increases the enzymatic activity of SHIP1. A series of similar SHIP1 activators have been synthesized and evaluated to determine structure–activity relationships and improve in vivo stability. Some new analogs have now been found with improved potency. In addition, both the thiophene and the thiomorpholine in the parent structure can be replaced by groups without a low valent sulfur atom, which provides a way to access activators that are less prone to oxidative degradation.

Mycoplasma gallisepticum (MG) is recognized as a principal causative agent of avian chronic respiratory disease, inflicting substantial economic losses upon the poultry industry. However, the extensive use of conventional antibiotics has resulted in the emergence of drug resistance and various challenges in their clinical application. Consequently, there is an urgent need to identify effective therapeutic agents for the prevention and treatment of mycoplasma-induced respiratory disease in avian species. AMP-activated protein kinase (AMPK) holds significant importance as a regulator of cellular energy metabolism and possesses the capacity to exert an anti-inflammatory effect by virtue of its downstream protein, SIRT1. This pathway has shown promise in counteracting the inflammatory responses triggered by pathogenic infections, thus providing a novel target for studying infectious inflammation. Quercetin possesses anti-inflammatory activity and has garnered attention as a potential alternative to antibiotics. However, there exists a gap in knowledge concerning the impact of this activation on MG-induced inflammatory damage. To address this knowledge gap, we employed AlphaFold2 prediction, molecular docking, and kinetic simulation methods to perform a systematic analysis. As expected, we found that both quercetin and the AMPK activator AICAR activate the chicken AMPKγ1 subunit in a similar manner, which was further validated at the cellular level. Our project aims to unravel the underlying mechanisms of quercetin’s action as an agonist of AMPK against the inflammatory damage induced by MG infection. Accordingly, we evaluated the effects of quercetin on the prevention and treatment of air sac injury, lung morphology, immunohistochemistry, AMPK/SIRT1/NF-κB pathway activity, and inflammatory factors in MG-infected chickens. The results confirmed that quercetin effectively inhibits the secretion of pro-inflammatory cytokines such as IL-1β, TNF-α, and IL-6, leading to improved respiratory inflammation injury. Furthermore, quercetin was shown to enhance the levels of phosphorylated AMPK and SIRT1 while reducing the levels of phosphorylated P65 and pro-inflammatory factors. In conclusion, our study identifies the AMPK cascade signaling pathway as a novel cellular mediator responsible for quercetin’s ability to counter MG-induced inflammatory damage. This finding highlights the potential significance of this pathway as an important target for anti-inflammatory drug research in the context of avian respiratory diseases.

Amendments with activators or microorganisms to enhance phytoremediation in toxic-metal-polluted soils have been widely studied. In this research, the production of indoleacetic acid, siderophore, and 1-aminocyclopropane-1-carboxylate (ACC) deaminase by phosphate-solubilizing bacteria was investigated during a pure culture experiment. Pot experiments were performed using Cd-polluted soil with the following treatments: control (CK, only ultrapure water), orange-peel-based activator (OG), and a combination of phosphate-solubilizing bacteria (Acinetobacter pitti) and OG (APOG). Ryegrass plant height and fresh weight, Cd content in ryegrass, total and available Cd soil content, soil enzyme activity, and soil bacterial diversity were determined in this work. The findings showed that the height of ryegrass in OG and APOG increased by 14.78% and 21.23%. In the APOG group, a decreased ratio of Cd was 3.37 times that of CK, and the bioconcentration factor was 1.28 times that of CK. The neutral phosphatase activity of APOG was 1.33 times that of CK and catalase activity was 1.95 times that of CK. The activity of urease was increased by 35.48%. APOG increased the abundance of beneficial bacteria and Proteobacteria was the dominant bacterium, accounting for 57.38% in APOG. Redundancy analysis (RDA) showed that nutrient elements were conducive to the propagation of the dominant bacteria, the secretion of enzymes, and the extraction rate of Cd in the soil. The possible enhancement mechanism of phytoremediation of cadmium by A. pitti combined with OG was that, on the one hand, APOG increased soil nutrient elements and enzyme activities promoted the growth of ryegrass. On the other hand, APOG activated Cd and boosted the movement of Cd from soil to ryegrass. This research offers insight for the combination of phosphate-solubilizing bacteria with an orange-peel-based activator to improve phytoremediation of Cd-contaminated soils and also provides a new way for the resource utilization of fruit residue.

Anthocyanin is a class of water-soluble flavonoids found in Chinese bayberry (Morella rubra) that is not only responsible for the variety of colors visible in nature but also has numerous health-promoting benefits in humans. Through comparative transcriptomics, we isolated and identified a transcription factor (TF) of the R2R3-MYB type, MrMYB9, in order to explore the anthocyanin biosynthesis pathway in red and white Chinese bayberries. MrMYB9 transcript was positively correlated with anthocyanin level and anthocyanin biosynthetic gene expression during Chinese bayberry fruit maturation (R-values in the range 0.54–0.84, p < 0.05). Sequence analysis revealed that MrMYB9 shared a similar R2R3 domain with MYB activators of anthocyanin biosynthesis in other plants. MrMYB9 substantially transactivated promoters of anthocyanin biosynthesis-related EBGs (MrCHI, MrF3’H, and MrANS) and LBGs (MrUFGT) upon co-expression of the AtEGL3 gene. Our findings indicated that MrMYB9 may positively modulate anthocyanin accumulation in Chinese bayberry.

In order to obtain conductive concrete with good electrical conductivity and good mechanical properties, nanographite and magnetite sand excited by different activators and their combinations are added to ordinary concrete to obtain high quality and efficient conductive concrete. The optimal mixture ratio of alkali-excited conductive concrete and the effects of different activators and their combinations on the mechanics and electrical conductivity of concrete were studied. The microstructure of alkali-excited conductive concrete was analyzed by scanning electron microscope (SEM) to study its conductive mechanism. Results show that the conductive concrete obtained by compounding sodium hydroxide, sodium sulfate and calcium hydroxide has optimal mechanical and electrical properties when the graphite is 6% cement, and magnetite sand is 40% fine aggregate. The conductive concrete sample prepared by this method has a flexural strength of 6.84 MPa, a compressive strength of 47.79 MPa and a resistivity of 4805 Ω·cm (28 days). Compared with ordinary concrete (no nanographite and no magnetite sand), the compressive strength of conductive concrete is increased by 122.3%, the bending strength is increased by 116.5%, and the resistivity is reduced by 99.1%. SEM shows that the distribution of conductive materials in concrete is more uniform due to alkali excitation and calcium silicate hydrate (CSH) gel can be formed, which leads to better performance. The research in this paper is only a preliminary exploration of the characteristics of green conductive concrete, and the conductive heating characteristics and electromagnetic wave absorption properties of concrete, along with strength characteristics after adding conductive fillers, need to be further studied. It is suggested that further research should be carried out on the deicing characteristics of conductive concrete and the electromagnetic wave absorption properties used in stealth military engineering.

Steroid hormones play a crucial role in several aspects of human life, and steroidogenesis is the process by which hormones are produced from cholesterol using several enzymes that work in concert to obtain the appropriate levels of each hormone at the right time. Unfortunately, many diseases, such as cancer, endometriosis, and osteoporosis as examples, are caused by an increase in the production of certain hormones. For these diseases, the use of an inhibitor to block the activity of an enzyme and, in doing so, the production of a key hormone is a proven therapeutic strategy whose development continues. This account-type article focuses on seven inhibitors (compounds 1–7) and an activator (compound 8) of six enzymes involved in steroidogenesis, namely steroid sulfatase, aldo-keto reductase 1C3, types 1, 2, 3, and 12 of the 17β-hydroxysteroid dehydrogenases. For these steroid derivatives, three topics will be addressed: (1) Their chemical synthesis from the same starting material, estrone, (2) their structural characterization using nuclear magnetic resonance, and (3) their in vitro or in vivo biological activities. These bioactive molecules constitute potential therapeutic or mechanistic tools that could be used to better understand the role of certain hormones in steroidogenesis.

The technology for obtaining hydrofluoric acid and the peculiarities of its production were studied, and the physical and chemical properties of the waste were examined. Activators that accelerated the hardening of the anhydrite binder were selected. The process of recycling fluorine hydrite waste from the production of hydrofluoric acid at Ulba Metallurgical Plant JSC was studied, and anhydrite unburnt binder with a setting time of 30 min was obtained. On the basis of the obtained data, a technological scheme of anhydrite binder production was developed. The effectiveness of the technological scheme was confirmed experimentally. This work aimed to study the possibility of the integrated use of secondary and anthropogenic raw materials from Ulba Metallurgical Plant, which represents an important means of not only increasing production efficiency and economic benefits and reducing the irrational alienation of land resources, but also protecting against the pollution of water and air basins, as the environmental policy of UMP JSC is nowadays of great importance.

SIRT1, an NAD+-dependent deacetylase, catalyzes the deacetylation of proteins coupled with the breakdown of NAD+ into nicotinamide and 2′-O-acetyl-ADP-ribose (OAADPr). Selective SIRT1 activators have potential clinical applications in atherosclerosis, acute renal injury, and Alzheimer’s disease. Here, we found that the activity of the potent SIRT1 activator CWR is independent of the acetylated substrate. It adopts a novel mechanism to promote SIRT1 activity by covalently bonding to the anomeric C1′ carbon of the ribose ring in OAADPr. In addition, CWR is highly selective for SIRT1, with no effect on SIRT2, SIRT3, SIRT5, or SIRT6. The longer distance between the anomeric C1′ carbon of the ribose ring in OAADPr and Arg274 of SIRT1 (a conserved residue among sirtuins) than that between the anomeric C1′ carbon in OAADPr and the Arg of SIRT2, SIRT3, SIRT5, and SIRT6, should be responsible for the high selectivity of CWR for SIRT1. This was confirmed by site-directed mutagenesis of SIRT3. Consistent with the in vitro assays, the activator also reduced the acetylation levels of p53 in a concentration-dependent manner via SIRT1 in cells. Our study provides a new perspective for designing SIRT1 activators that does not rely on the chemical moiety immediately C-terminal to the acetyl-lysine of the substrate.

To study the key factors that affect the mechanical properties of polymer materials and explore the relationship between mineral crystal formation and strength development, fly ash (FA) polymer samples were prepared using sodium hydroxide, slag, liquid sodium silicate, and hydrated lime as activators. A change in the compressive strength was observed, and X-ray diffraction measurements were carried out to confirm the change. The effects of different types and amounts of activators on the formation and transformation of mineral crystals in FA polymer samples as well as on the development of compressive strength were studied. Moreover, the relationship between the formation and transformation of mineral crystals and the development of compressive strength was established. The results show that the strongly alkaline excitation environment established by sodium hydroxide is the prerequisite for crystal formation and development of compressive strength. Under this strongly alkaline excitation environment, slag, hydrated lime, and liquid sodium silicate can increase the amounts of calcium and silicon, which promote the formation and development of hydrated calcium silicate and hydrated calcium silicoaluminate in polymers and significantly improve the compressive strength.

Protein kinase C (PKC), a family of phospholipid-dependent serine/threonine kinase, is classed into three subfamilies based on their structural and activation characteristics: conventional or classic PKC isozymes (cPKCs; α, βI, βII, and γ), novel or non-classic PKC isozymes (nPKCs; δ, ε, η, and θ), and atypical PKC isozymes (aPKCs; ζ, ι, and λ). PKC inhibitors and activators are used to understand PKC-mediated intracellular signaling pathways and for the diagnosis and treatment of various PKC-associated diseases, such as cancers, neurological diseases, cardiovascular diseases, and infections. Many clinical trials of PKC inhibitors in cancers showed no significant clinical benefits, meaning that there is a limitation to design a cancer therapeutic strategy targeting PKC alone. This review will focus on the activators and inhibitors of PKC and their applications in clinical trials.

Human immunodeficiency virus 1 (HIV-1) is the most prevalent human retrovirus. Recent data show that 34 million people are living with HIV-1 worldwide. HIV-1 infections can lead to AIDS which still causes nearly 20,000 deaths annually in the USA alone. As this retrovirus leads to high morbidity and mortality conditions, more effective therapeutic regimens must be developed to treat these viral infections. A key target for intervention for which there are no current FDA-approved modulators is at the point of proviral transcription. One successful method for identifying novel therapeutics for treating infectious diseases is the repurposing of pharmaceuticals that are approved by the FDA for alternate indications. Major benefits of using FDA-approved drugs include the fact that the compounds have well established toxicity profiles, approved manufacturing processes, and immediate commercial availability to the patients. Here, we demonstrate that pharmaceuticals previously approved for other indications can be utilized to either activate or inhibit HIV-1 proviral transcription. Specifically, we found febuxostat, eltrombopag, and resveratrol to be activators of HIV-1 transcription, while mycophenolate was our lead inhibitor of HIV-1 transcription. Additionally, we observed that the infected cells of lymphoid and myeloid lineage responded differently to our lead transcriptional modulators. Finally, we demonstrated that the use of a multi-dose regimen allowed for enhanced activation with our transcriptional activators.

To study the structure and function of the pH-regulated receptor tyrosine kinase insulin receptor-related receptor (IRR), а member of the insulin receptor family, we obtained six mouse monoclonal antibodies against the recombinant IRR ectodomain. These antibodies were characterized in experiments with exogenously expressed full-length IRR by Western blotting, immunoprecipitation, and immunocytochemistry analyses. Utilizing a previously obtained set of IRR/IR chimeras with swapped small structural domains and point amino acid substitutions, we mapped the binding sites of the obtained antibodies in IRR. Five of them showed specific binding to different IRR domains in the extracellular region, while one failed to react with the full-length receptor. Unexpectedly, we found that 4D5 antibody can activate IRR at neutral pH, and 4C2 antibody can inhibit activation of IRR by alkali. Our study is the first description of the instruments of protein nature that can regulate activity of the orphan receptor IRR and confirms that alkali-induced activation is an intrinsic property of this receptor tyrosine kinase.

Mandibular retrognathism occurs by insufficient mandibular growth and causes several issues, such as respiratory difficulty and diminished masticatory function. At present, functional orthodontic appliances are used for stimulating mandibular growth in pediatric cases. However, the effectiveness of functional appliances is not always stable in daily practices. A more effective, reliable, and safer therapeutic method for mandibular growth promotion would be helpful for growing mandibular retrognathism patients. As we previously discovered that nutritional supplementation of myo-inositol in growing mice specifically increases mandibular endochondral growth, we performed preclinical animal experiments in rabbits in this study. Briefly, six-week-old male Japanese white rabbits were fed with or without myo-inositol supplementation in laboratory chow until 25 weeks old, and 3D image analysis using micro CT data and histological examinations was done. Myo-inositol had no systemic effect, such as femur length, though myo-inositol specifically augmented the mandibular growth. Myo-inositol increased the thickness of mandibular condylar cartilage. We discovered that the nutritional supplementation of myo-inositol during the growth period specifically augmented mandibular growth without any systemic influence, even in rabbits. Our results suggest the possibility of clinical use of myo-inositol for augmentation of the mandibular growth in growing mandibular retrognathism patients in the future.

Flor velum yeast growth activators during biological aging are currently unknown. In this sense, this research focuses on the use of bee pollen as a flor velum activator. Bee pollen influence on viable yeast development, surface hydrophobicity, and yeast assimilable nitrogen has already been studied. Additionally, bee pollen effects on the main compounds related to flor yeast metabolism and wine sensory characteristics have been evaluated. “Fino” (Sherry) wine was supplemented with bee pollen using six different doses ranging from 0.1 to 20 g/L. Its addition in a dose equal or greater than 0.25 g/L can be an effective flor velum activator, increasing yeast populations and its buoyancy due to its content of yeast assimilable nitrogen and fatty acids. Except for the 20 g/L dose, pollen did not induce any significant effect on flor velum metabolism, physicochemical parameters, organic acids, major volatile compounds, or glycerol. Sensory analysis showed that low bee pollen doses increase wine’s biological aging attributes, obtaining the highest score from the tasting panel. Multiflora bee pollen could be a natural oenological tool to enhance flor velum development and wine sensory qualities. This study confirms association between the bee pollen dose applied and the flor velum growth rate. The addition of bee pollen could help winemakers to accelerate or reimplant flor velum in biologically aged wines.

The γ-carbonic anhydrases (CAs, EC 4.2.1.1) present in the Antarctic marine bacteria Pseudoalteromonas haloplanktis and Colwellia psychrerythraea, herein referred to as PhaCA and CpsCA, respectively, were investigated for their activation with a panel of 24 amino acids and amines. Both bacteria are considered Antarctic models for the investigation of photosynthetic and metabolic pathways in organisms adapted to live in cold seawater. PhaCA was much more sensitive to activation by these compounds compared to the genetically related enzyme CpsCA. The most effective PhaCA activators were d-Phe, l-/d-DOPA, l-Tyr and 2-pyridyl-methylamine, with the activation constant KA values of 0.72–3.27 µM. d-His, l-Trp, d-Tyr, histamine, dopamine, serotonin anddicarboxylic amino acids were also effective activators of PhaCA, with KA values of 6.48–9.85 µM. CpsCA was activated by d-Phe, d-DOPA, l-Trp, l-/d-Tyr, 4-amino-l-Phe, histamine, 2-pyridyl-methylamine and l-/d-Glu with KA values of 11.2–24.4 µM. The most effective CpsCA activator was l-DOPA (KA of 4.79 µM). Given that modulators of CAs from Antarctic bacteria have not been identified and investigated in detail for their metabolic roles to date, this research sheds some light on these poorly understood processes.

The β-carbonic anhydrase (CA, EC 4.2.1.1) from the pathogenic protozoan Entamoeba histolytica, EhiCA, was investigated for its activation with a panel of natural and non-natural amino acids and amines. EhiCA was potently activated by D-His, D-Phe, D-DOPA, L- and D-Trp, L- and D-Tyr, 4-amino-L-Tyr, histamine and serotonin, with KAs ranging between 1.07 and 10.1 µM. The best activator was D-Tyr (KA of 1.07 µM). L-Phe, L-DOPA, L-adrenaline, L-Asn, L-Asp, L-Glu and L-Gln showed medium potency activation, with KAs of 16.5⁻25.6 µM. Some heterocyclic- alkyl amines, such as 2-pyridyl-methyl/ethyl-amine and 4-(2-aminoethyl)-morpholine, were devoid of EhiCA activating properties with KAs > 100 µM. As CA activators have poorly been investigated for their interaction with protozoan CAs, our study may be relevant for an improved understanding of the role of this enzyme in the life cycle of E. histolytica.
