Stinging nettles (Urtica dioica) have a long history of association with human civilization, having been used as a source of textile fibers, food and medicine. Here, we present a chromosome-level, phased genome assembly for a diploid female clone of Urtica dioica from Romania. Using a combination of PacBio HiFi, Oxford Nanopore, and Illumina sequencing, as well as Hi-C long-range interaction data (using a novel Hi-C protocol presented here), we assembled two haplotypes of 574.9 Mbp (contig N50 = 10.9 Mbp, scaffold N50 = 44.0 Mbp) and 521.2 Mbp (contig N50 = 13.5 Mbp, scaffold N50 = 48.0 Mbp), with assembly BUSCO scores of 92.6% and 92.2%. We annotated 20,333 and 20,140 genes for each haplotype, covering over 90% of the complete BUSCO genes and including two copies of a gene putatively encoding the neurotoxic peptide urthionin, which could contribute to nettle’s characteristic sting. Despite its relatively small size, the nettle genome displays very high levels of repetitiveness, with transposable elements comprising more than 60% of the genome, as well as considerable structural variation. This genome assembly represents an important resource for the nettle community and will enable the investigation of the genetic basis of the many interesting characteristics of this species.

Heavy metal pollution is one of the most devastating abiotic factors, significantly damaging crops and human health. One of the serious problems it causes is a rise in cadmium (Cd) toxicity. Cd is a highly toxic metal with a negative biological role, and it enters plants via the soil–plant system. Cd stress induces a series of disorders in plants’ morphological, physiological, and biochemical processes and initiates the inhibition of seed germination, ultimately resulting in reduced growth. Fiber crops such as kenaf, jute, hemp, cotton, and flax have high industrial importance and often face the issue of Cd toxicity. Various techniques have been introduced to counter the rising threats of Cd toxicity, including reducing Cd content in the soil, mitigating the effects of Cd stress, and genetic improvements in plant tolerance against this stress. For decades, plant breeders have been trying to develop Cd-tolerant fiber crops through the identification and transformation of novel genes. Still, the complex mechanism of Cd tolerance has hindered the progress of genetic breeding. These crops are ideal candidates for the phytoremediation of heavy metals in contaminated soils. Hence, increased Cd uptake, accumulation, and translocation in below-ground parts (roots) and above-ground parts (shoots, leaves, and stems) can help clean agricultural lands for safe use for food crops. Earlier studies indicated that reducing Cd uptake, detoxification, reducing the effects of Cd stress, and developing plant tolerance to these stresses through the identification of novel genes are fruitful approaches. This review aims to highlight the role of some conventional and molecular techniques in reducing the threats of Cd stress in some key fiber crops. Molecular techniques mainly involve QTL mapping and GWAS. However, more focus has been given to the use of transcriptome and TFs analysis to explore the potential genomic regions involved in Cd tolerance in these crops. This review will serve as a source of valuable genetic information on key fiber crops, allowing for further in-depth analyses of Cd tolerance to identify the critical genes for molecular breeding, like genetic engineering and CRISPR/Cas9.

Hibiscus cannabinus (kenaf) is an annual fiber crop grown in warm seasons and known for its remarkable productivity; it has been cultivated worldwide for thousands of years as a fiber source. While every part of the plant can be utilized for some purpose, its primary significance lies in the diverse applications of its cellulosic fiber. Kenaf features a blend of long bast and short core fibers, rendering it suitable for various industrial uses. Initially utilized for cordage and livestock feed, kenaf’s applications have expanded over the last century to encompass its utilization as paper pulp, biocomposites, textiles, biomass energy, seed oil, filtration aids, industrial absorbents, and even as a component of potting medium or as a potential source of medicine. Although traditionally a niche crop, the discovery of its diverse applications positions kenaf for rapid expansion in production in the upcoming decades. This article aims to explore the manifold applications of kenaf, highlighting those with the greatest future potential and discussing those that hold promise for commercial-level application with additional research.

Jute (Corchorus spp.) belongs to the Malvaceae family, and there are two species of jute, C. capsularis and C. olitorious. It is the second-largest natural bast fiber in the world according to production, which has diverse uses not only as a fiber but also as multiple industrial materials. Because of climate change, plants experience various stressors such as salt, drought, heat, cold, metal/metalloid toxicity, and flooding. Although jute is particularly adapted to grow in hot and humid climates, it is grown under a wide variety of climatic conditions and is relatively tolerant to some environmental adversities. However, abiotic stress often restricts its growth, yield, and quality significantly. Abiotic stress negatively affects the metabolic activities, growth, physiology, and fiber yield of jute. One of the major consequences of abiotic stress on the jute plant is the generation of reactive oxygen species, which lead to oxidative stress that damages its cellular organelles and biomolecules. However, jute’s responses to abiotic stress mainly depend on the plant’s age and type and duration of stress. Therefore, understanding the abiotic stress responses and the tolerance mechanism would help plant biologists and agronomists in developing climate-smart jute varieties and suitable cultivation packages for adverse environmental conditions. In this review, we summarized the best possible recent literature on the plant abiotic stress factors and their influence on jute plants. We described the possible approaches for stress tolerance mechanisms based on the available literature.

Fiber crops are an important group of economic plants. Traditionally cultivated for fiber, fiber crops have also become sources of other materials such as food, animal feed, cosmetics and medicine. Asia and America are the two main production areas of fiber crops in the world. However, oomycete diseases have become an important factor limiting their yield and quality, causing devastating consequences for the production of fiber crops in many regions. To effectively control oomycete pathogens and reduce their negative impacts on these crops, it is very important to have fast and accurate detection systems, especially in the early stages of infection. With the rapid development of molecular biology, the diagnosis of plant pathogens has progressed from relying on traditional morphological features to the increasing use of molecular methods. The objective of this paper was to review the current status of research on molecular diagnosis of oomycete pathogens on fiber crops. Our search of PubMed identified nearly 30 species or subspecies of oomycetes on fiber crops, among which the top three species were Phytophthora boehmeriae, Phytophthora nicotianae and Pythium ultimum. The gene regions that have been used for molecular identifications of these pathogens include the internal transcribed spacer (ITS) regions of the nuclear ribosomal RNA gene cluster, and genes coding for translation elongation factor 1α (EF-1α) and mitochondrial cytochrome c oxidase subunits I and II (Cox 1, Cox 2), etc. We summarize the molecular assays that have been used to identify these pathogens and discuss potential areas of future development for fast, specific, and accurate diagnosis of oomycetes on fiber crops.

Industrial hemp (Cannabis sativa L.) production is increasing dramatically in the US due to recent changes which lift restrictions on the growth and sale of hemp products; however, due to the decades-long prohibition of hemp, there is a lack of current research with respect to varieties and best agricultural practices for the many uses of this versatile crop. Natural fiber production relies on retting, a microbially-mediated process necessary for the separation of fibers from the plant which can occur unevenly in the field environment and result in inconsistent fiber quality and lower processing efficiency. In this study, the microbiome of hemp stalks is investigated throughout the retting process using 16S rRNA gene amplicon sequencing using the Illumina MiSeq platform. Field retting conditions were simulated in a controlled greenhouse environment in order to determine the effects of different moisture levels and soil contact on the retting process. Samples were taken over six time points, reflecting the community of freshly cut stalks to optimally-retted material, and finally over-retted material showing degraded fibers. The results show a very consistent population throughout retting, dominated primarily by Proteobacteria, but showing an increase in the abundance of the Bacteroidetes, namely Chryseobacterium, in time points corresponding to optimally-retted and over-retted stalks in treatments receiving higher moisture levels, but not in the low-moisture treatment. Soil application did not appear to influence the microbial community throughout retting, indicating a resilient population present in and on the hemp stalks at harvest.

Plant fibers find wide application in various fields that demand specific parameters of fiber quality. To develop approaches for the improvement of yield and quality of bast fibers, the knowledge of the fiber developmental stages and of the key molecular players that are responsible for a certain parameter, are vitally important. In the present review the key stages of fiber development, such as initiation, intrusive growth, and formation of thickened cell wall layers (secondary and tertiary cell walls) are considered, as well as the impact of each of these stages on the final parameters of fiber yield and quality. The problems and perspectives of crop quality regulation are discussed.

The development of flax (Linum usitatissimum L.) fibers was studied to obtain better insight on the progression of their high mechanical performances during plant growth. Fibers at two steps of plant development were studied, namely the end of the fast growth period and at plant maturity, each time at three plant heights. The indentation modulus of the fiber cell wall was characterized by atomic force microscopy (AFM) using peak-force quantitative nano-mechanical property mapping (PF-QNM). Changes in the cell wall modulus with the cell wall thickening were highlighted. For growing plants, fibers from top and middle heights show a loose inner Gn layer with a lower indentation modulus than mature fibers, which exhibit thickened homogeneous cell walls made only of a G layer. The influence of these changes in the fiber cell wall on the mechanical performances of extracted elementary fibers was also emphasized by tensile tests. In addition, Raman spectra were recorded on samples from both growing and mature plants. The results suggest that, for the fiber cell wall, the cellulose contribution increases with fiber maturity, leading to a greater cell wall modulus of flax fibers.
