Hypoxia is a unique environmental stress, which not only reflects the insufficient oxygen supply of cells and tissues, but also occurs in various physiological and pathological environments. Mitophagy as a selective autophagy can recover and utilize damaged organelles and misfolded proteins to ensure normal cell functions and promote cell survival. Bcl2l13 (B-cell lymphoma-2 like 13) is reported to induce mitophagy as a functional mammalian homolog of Atg32. However, the function of the bcl2l13 gene is still unclear in fish. Here the sequence and structure of the bcl2l13 gene in Megalobrama amblycephala were identified and showed that bcl2l13 contained an open reading frame (ORF) of 1458 bp for encoding 485 aa. Amino acid sequence analysis indicated that Bcl2l13, as a typical anti-apoptotic protein of the Bcl2 family, contained four BH domains, one BHNo domain, and one TM domain. Further study showed that Bcl2l13 was mainly located in the mitochondria, while its localization was changed within the whole cell after the TM domain was deleted. Real-time PCR analysis revealed that bcl2l13 showed higher expression levels in early embryos. After hypoxia treatment, the mRNA levels of the bcl2l13 and autophagy-related genes were significantly up-regulated in most detected tissues, and the bcl2l13 transcription was regulated by Hif-1α mediated pathway. Additionally, the transcription activity of the bcl2l13 promoter was further analyzed using luciferase reporter assays and showed the highest activity in the promoter region from −475 to +111. These results indicated that bcl2l13 may play important roles in embryogenesis and hypoxia mediated autophagy in fish.

The degradation characteristics of aniline and antimony in printing and dyeing wastewater during the micro-oxygenated hydrolytic acidification process and its effect on COD and ammonia nitrogen removal were investigated in this experiment. Firstly, the effects of control factors such as pH, dissolved oxygen (DO), and sludge concentration on COD and ammonia nitrogen removal in the hydrolysis acidification section were optimized. It was recommended that the pH value should be maintained at 6.5; low DO (0–0.5 mg/L) could assist in the conversion of nitrogen for subsequent treatment; the optimum treatment temperature was 25 °C; finally, it was recommended that the sludge concentration should be controlled at 4 mg/L during the operation. Secondly, the effects of aniline and antimony on COD and nitrogen removal were investigated. It was found that when the concentration of aniline was increased from 0.4 mg/L to 5.4 mg/L, the COD concentration in the effluent increased by 96.5%, which indicated that aniline was toxic to anaerobic sludge and obviously inhibited the degradation of COD. When the concentration of antimony was increased from 0.05 mg/L to 2.05 mg/L, the COD removal rate was only 2.9%, which was much lower than that of the water samples with no antimony added. The anaerobic sludge concentration decreased from 5.58 g/L to 3.44 g/L, which indicated that aniline and antimony had a strong inhibitory effect on the activity of anaerobic bacteria and inversely affected COD removal.

Dendritic cells (DCs) are crucial in initiating and shaping both innate and adaptive immune responses. Clinical studies and experimental models have highlighted their significant involvement in various autoimmune diseases, positioning them as promising therapeutic targets. Nicotinamide (NAM), a form of vitamin B3, with its anti-inflammatory properties, has been suggested, while the involvement of NAM in DCs regulation remains elusive. Here, through analyzing publicly available databases, we observe substantial alterations in NAM levels and NAM metabolic pathways during DCs activation. Furthermore, we discover that NAM, but not Nicotinamide Mononucleotide (NMN), significantly inhibits DCs over-activation in vitro and in vivo. The suppression of DCs hyperactivation effectively alleviates symptoms of psoriasis. Mechanistically, NAM impairs DCs activation through a Poly (ADP-ribose) polymerases (PARPs)-NF-κB dependent manner. Notably, phosphoribosyl transferase (NAMPT) and PARPs are significantly upregulated in lipopolysaccharide (LPS)-stimulated DCs and psoriasis patients; elevated NAMPT and PARPs expression in psoriasis patients correlates with higher psoriasis area and severity index (PASI) scores. In summary, our findings underscore the pivotal role of NAM in modulating DCs functions and autoimmune disorders. Targeting the NAMPT-PARP axis emerges as a promising therapeutic approach for DC-related diseases.

This paper introduces a novel method for preparing high-performance, metal-containing carbon foam wave-absorbing materials. The process involves foaming glucose through catalysis by transition metals followed by high-temperature pyrolysis. The resulting carbon foam materials exhibit a highly porous structure, which is essential for their wave-absorption properties. Notably, at a thickness of 2.0 mm, the glucose-derived carbon foam composite catalyzed by Fe and Co (GCF-CoFe) achieved a minimum reflection loss (RLmin) of −51.4 dB at 15.11 GHz, along with an effective absorption bandwidth (EAB) of 5.20 GHz, spanning from 12.80 GHz to 18.00 GHz. These impressive performance metrics indicate that this approach offers a promising pathway for developing low-density, efficient carbon foam materials for wave-absorption applications. This advancement has significant implications for fields requiring effective electromagnetic interference (EMI) shielding, stealth technology, and other related applications, potentially leading to more efficient and lightweight solutions.

As one of the fundamental vehicular perception technologies, millimeter-wave radar’s accuracy in angle measurement affects the decision-making and control of vehicles. In order to enhance the accuracy and efficiency of the Direction of Arrival (DoA) estimation of radar systems, a super-resolution angle measurement strategy based on the Fast Deterministic Maximum Likelihood (FDML) algorithm is proposed in this paper. This strategy sequentially uses Digital Beamforming (DBF) and Deterministic Maximum Likelihood (DML) in the Field of View (FoV) to perform a rough search and precise search, respectively. In a simulation with a signal-to-noise ratio of 20 dB, FDML can accurately determine the target angle in just 16.8 ms, with a positioning error of less than 0.7010. DBF, the Iterative Adaptive Approach (IAA), DML, Fast Iterative Adaptive Approach (FIAA), and FDML are subjected to simulation with two targets, and their performance is compared in this paper. The results demonstrate that under the same angular resolution, FDML reduces computation time by 99.30% and angle measurement error by 87.17% compared with the angular measurement results of two targets. The FDML algorithm significantly improves computational efficiency while ensuring measurement performance. It provides more reliable technical support for autonomous vehicles and lays a solid foundation for the advancement of autonomous driving technology.

Differences in field of view may occur during unmanned aerial remote sensing imaging applications with acousto-optic tunable filter (AOTF) spectral imagers using zoom lenses. These differences may stem from image size deformation caused by the zoom lens, image drift caused by AOTF wavelength switching, and drone platform jitter. However, they can be addressed using hyperspectral image registration. This article proposes a new coarse-to-fine remote sensing image registration framework based on feature and optical flow theory, comparing its performance with that of existing registration algorithms using the same dataset. The proposed method increases the structure similarity index by 5.2 times, reduces the root mean square error by 3.1 times, and increases the mutual information by 1.9 times. To meet the real-time processing requirements of the AOTF spectrometer in remote sensing, a development environment using VS2023+CUDA+OPENCV was established to improve the demons registration algorithm. The registration algorithm for the central processing unit+graphics processing unit (CPU+GPU) achieved an acceleration ratio of ~30 times compared to that of a CPU alone. Finally, the real-time registration effect of spectral data during flight was verified. The proposed method demonstrates that AOTF hyperspectral imagers can be used in real-time remote sensing applications on unmanned aerial vehicles.

The present study aimed to investigate the influence of the mutation abundance of the epidermal growth factor receptor (EGFR) and its co-mutation with TP53 on the therapeutic efficacy of tyrosine kinase inhibitor (TKI) treatment in patients with metastatic lung adenocarcinoma (LUAD). In total, 130 patients (January 2018-September 2022) with metastatic LUAD from the Second Affiliated Hospital of Zhejiang University were included. Kaplan–Meier analysis was performed to measure the duration of drug application (DDA) and the log-rank test was used to compare differences. Univariate and multivariate analyses of Cox proportional hazard regression models were used to evaluate the association between the relevant clinicopathological factors and DDA. Hazard ratios with 95% confidence intervals were also calculated. Among the 130 patients who were treated with first-generation EGFR-TKIs, 86 showed high-EGFR mutation abundance (>22.0%) and 44 showed low-EGFR mutation abundance (≤22.0%). Patients in the high-EGFR group had a greater DDA than those in the low-EGFR group (p < 0.05). The results of the subgroup analysis were consistent with those of the total mutation population (exon19: >18.5% vs. ≤18.5%, 14 months vs. 10 months, p = 0.049; exon21: >22.0% vs. ≤22.0%, 15 months vs. 9 months, p = 0.005). In addition, the mutation abundance of TP53 was negatively correlated with the DDA (p < 0.05). Patients in the combination group had a better DDA than those in the monotherapy group (p < 0.05). Subgroup analysis showed that, among the low mutation abundance of the EGFR exon 21 or 19 cohort, the combination group had a better DDA than the monotherapy group (p < 0.05). An EGFR mutation abundance greater than 22.0% was a positive predictor of DDA in patients with metastatic LUAD. However, a TP53 mutation abundance higher than 32.5% could reverse this situation. Finally, first-line treatment with EGFR-TKIs plus chemotherapy is a potential treatment strategy for patients with low-abundance EGFR mutations.

The coupling processes among the lithosphere, atmosphere, and ionosphere (LAI) during the earthquake preparation phase are still an open scientific debate. Comprehensive LAI coupling effects around the 2022 Ms6.8 Luding earthquake in China are investigated with a multi-parameter and multi-layer approach, including the b-value, revised accelerated moment release, Earth resistivity, ELF magnetic field emissions, atmospheric electric field, surface temperature, foF2 from ionosonde, GNSS TEC, Ne and magnetic field from CSES and Swarm satellites, and energetic electrons from CSES and NOAA satellites. It is found that the anomalies start from the lithospheric parameters as Earth resistivity and b-values 1–2 years before to reflect the local stress loading in the seismic region, then the ionospheric and atmospheric disturbances occur and accelerate −50 days before and −15 days before, and finally the electrons precipitate a few days before. The simultaneous perturbations in LAI illustrate the thermodynamic coupling channel, such as on 24 August, −12 days before. Meanwhile, the abundant developed ionospheric anomalies without atmospheric disturbances demonstrate the electromagnetic coupling way from the lithosphere to the ionosphere directly. Finally, the results demonstrate a two-way model of LAIC: one way is characterized by a slow chain of processes, of thermodynamic nature, starting from the ground and proceeding to the above atmosphere and ionosphere, showing an exponential trend in the cumulative number of anomalies; the second way is characterized by oscillating electromagnetic coupling between the lithosphere and ionosphere, showing intermittent fluctuations in the corresponding cumulative number of anomalies.

In this study, magnetic (Fe)-loaded biochar was successfully prepared by a simple impregnation pyrolysis method. Meanwhile, its degradation capability and mechanism for typical antibiotic metronidazole (MNZ) were systematically investigated under different conditions. The characterization of the synthesized material showed that the specific surface area, pore diameter, and pore volume changed significantly. Also, functional groups and metal element Fe were introduced on the surface of the biochar, leading to its better capability to activate peroxymonosulfate (PMS). The degradation experiments showed that the removal of MNZ in the Fe-BC/PMS system can reach up to 95.3% in 60 min under optimal conditions. Free-radical capture experiments showed that there were several active species of •OH, SO4•−, •O2−, and 1O2 present in the catalyst to synergistically degrade MNZ, among which SO4•− played a major role; it was also found that the material can be easily recycled and was still effective after several uses. Further, the main degradation pathways of MNZ include nitrohydroxylation, hydroxyethyl functional group deletion, carboxylation of the amino functional group of •OH, demethylation, oxidation, and carboxylation. It is obvious that the synthesized magnetic-loaded biochar, Fe-BC, generated from waste rape straw crops, shows high catalytic performance in pollutant degradation, providing an insight into the recycling potential of waste biomass in the catalytic field for pollutant removal.

Food non-denatured processes, such as freeze-drying and grinding, are commonly applied to raw materials with good bioactive functions. Although the functional components are maintained, whether structural and physical changes impact the in vivo function is often ignored in practical situations. Brasenia schreberi mucilage (BSM) has a significant alleviation effect on DSS-induced colitis. This work focused on the influence of non-denatured manufacture on the colonic benefits of BSM-based products. First, three forms of products including fresh mucilage (FM), freeze-dried products (FS), and freeze-dried powder (FP) were prepared. Then, their in vitro physiochemical properties were compared, analyzing their influence on the gut inflammation degree, microbial composition, and SCFA production in mice. The results suggested that the water retention rate of FS and FP was decreased to 34.59 ± 3.85%, and 9.93 ± 1.76%. The viscosity of FM, FS, and FP was 20.14 Pa∙s, 4.92 Pa∙s, and 0.41 Pa∙s, respectively. The freeze-drying and grinding process also damaged the lamellar microstructure of BSM. Then, animal tests showed that colitis mice intervened with FM, FS, and FP had disease activity scores of 2.03, 3.95, and 4.62. Meanwhile, FM notably changed the gut microbial composition and significantly increased propionate and butyrate levels. It seemed that the distinct colitis alleviation efficacy of BSM-based products is attributed to different hydrodynamic properties in the gut. FM had relatively higher viscosity and correspondingly high nutritional density in the gut lumen, which stimulates Firmicutes growth and promotes butyrate production, and thereby exhibited the best efficiency on protecting from colitis.

Purpose: Adhesion between calcium oxalate crystals and renal tubular epithelial cells is a vital cause of renal stone formation; however, the drugs that inhibit crystal adhesion and the mechanism of inhibition have yet to be explored. Methods: The cell injury model was constructed using nano-COM crystals, and changes in oxidative stress levels, endoplasmic reticulum (ER) stress levels, downstream p38 MAPK protein expression, apoptosis, adhesion protein osteopontin expression, and cell–crystal adhesion were examined in the presence of Laminarin polysaccharide (DLP) and sulfated DLP (SDLP) under protected and unprotected conditions. Results: Both DLP and SDLP inhibited nano-COM damage to human kidney proximal tubular epithelial cell (HK-2), increased cell viability, decreased ROS levels, reduced the opening of mitochondrial membrane permeability transition pore, markedly reduced ER Ca2+ ion concentration and adhesion molecule OPN expression, down-regulated the expression of ER stress signature proteins including CHOP, Caspase 12, and p38 MAPK, and decreased the apoptosis rate of cells. SDLP has a better protective effect on cells than DLP. Conclusions: SDLP protects HK-2 cells from nano-COM crystal-induced apoptosis by reducing oxidative and ER stress levels and their downstream factors, thereby reducing crystal–cell adhesion interactions and the risks of kidney stone formation.

Carbon nanomaterials are in high demand owing to their exceptional physical and chemical properties. This study employed a mixture of CH4, H2, and N2 to create carbon nanostructures on a single-crystal diamond using microwave plasma chemical vapor deposition (MPCVD) under high-power conditions. By controlling the substrate surface and nitrogen flow rate, carbon nanowires, carbon nanotubes, and carbon pompons could be selectively deposited. The results obtained from OES, SEM, TEM, and Raman spectroscopy revealed that the nitrogen flow rate and substrate surface conditions were crucial for the growth of carbon nanostructures. The changes in the plasma shape enhanced the etching effect, promoting the growth of carbon pompons. The CN and C2 groups play vital catalytic roles in the formation of carbon nanotubes and nanowires, guiding the precipitation and composite growth of carbon atoms at the interface between the Mo metal catalysts and diamond. This study demonstrated that heterostructures of diamond–carbon nanomaterials could be produced under high-power conditions, offering a new approach to integrating diamond and carbon nanomaterials.

This investigation sought to discern the risk factors for atrial fibrillation within Shanghai’s Chongming District, analyzing data from 678 patients treated at a tertiary hospital in Chongming District, Shanghai, from 2020 to 2023, collecting information on season, C-reactive protein, hypertension, platelets, and other relevant indicators. The researchers introduced a novel dual feature-selection methodology, combining hierarchical clustering with Fisher scores (HC-MFS), to benchmark against four established methods. Through the training of five classification models on a designated dataset, the most effective model was chosen for method performance evaluation, with validation confirmed by test set scores. Impressively, the HC-MFS approach achieved the highest accuracy and the lowest root mean square error in the classification model, at 0.9118 and 0.2970, respectively. This provides a higher performance compared to existing methods, thanks to the combination and interaction of the two methods, which improves the quality of the feature subset. The research identified seasonal changes that were strongly associated with atrial fibrillation (pr = 0.31, FS = 0.11, and DCFS = 0.33, ranked first in terms of correlation); LDL cholesterol, total cholesterol, C-reactive protein, and platelet count, which are associated with inflammatory response and coronary heart disease, also indirectly contribute to atrial fibrillation and are risk factors for AF. Conclusively, this study advocates that machine-learning models can significantly aid clinicians in diagnosing individuals predisposed to atrial fibrillation, which shows a strong correlation with both pathological and climatic elements, especially seasonal variations, in the Chongming District.

The degradation of farmland in China underscores the need for developing and utilizing saline-alkali soil. Soil health relies on microbial activity, which aids in the restoration of the land’s ecosystem, and hence it is important to understand microbial diversity. In the present study, two Gram-stain-positive strains HR 1-10T and J-A-003T were isolated from saline-alkali soil. Preliminary analysis suggested that these strains could be a novel species. Therefore, the taxonomic positions of these strains were evaluated using polyphasic analysis. Phylogenetic and 16S rRNA gene sequence analysis indicated that these strains should be assigned to the genus Halalkalibacter. Cell wall contained meso-2,6-diaminopimelic acid. The polar lipids present in both strains were diphosphatidyl-glycerol, phosphatidylglycerol, and an unidentified phospholipid. The major fatty acids (>10%) were anteiso-C15:0, C16:0 and iso-C15:0. Average nucleotide identity and digital DNA#x2013;DNA hybridization values were below the threshold values (95% and 70%, respectively) for species delineation. Based on the above results, the strains represent two novel species of the genus Halalkalibacter, for which the names Halalkalibacter flavus sp. nov., and Halalkalibacter lacteus sp. nov., are proposed. The type strains are HR 1-10T (=GDMCC 1.2946T = MCCC 1K08312T = JCM 36285T), and J-A-003T (=GDMCC 1.2949T = MCCC 1K08417T = JCM 36286T).

In the period 2022–2023, an analysis of fourteen phenotypic traits was conducted across 192 maize accessions in the Aral region of Xinjiang. The Shannon–Wiener diversity index was employed to quantify the phenotypic diversity among the accessions. Subsequently, a comprehensive evaluation of the index was performed utilizing correlation analysis, principal component analysis (PCA) and cluster analysis. The results highlighted significant findings: (1) A pronounced diversity was evident across the 192 maize accessions, accompanied by complex interrelationships among the traits. (2) The 14 phenotypic traits were transformed into 3 independent indicators through principal component analysis: spike factor, leaf width factor, and number of spikes per plant. (3) The 192 materials were divided into three groups using cluster analysis. The phenotypes in Group III exhibited the best performance, followed by those in Group I, and finally Group II. The selection of the three groups can vary depending on the breeding objectives. This study analysed the diversity of phenotypic traits in maize germplasm resources. Maize germplasm was categorised based on similar phenotypes. These findings provide theoretical insights for the study of maize accessions under analogous climatic conditions in Alar City, which lay the groundwork for the efficient utilization of existing germplasm as well as the development and selection of new varieties.

Our previous retrospective observational study demonstrated the safety of laparoscopically assisted subtotal colectomy with ileorectal anastomosis and preservation of the superior rectal artery (SRA), without instances of leakage, in patients with slow-transit constipation (STC). Thus, we extended the enrollment period and enlarged the sample size to detect the differences in the postoperative complications and surgical and functional outcomes between patients who underwent laparoscopically assisted subtotal colectomy with and without SRA preservation. We conducted a retrospective single-center analysis of patients with STC who underwent laparoscopically assisted subtotal colectomy between 2016 and 2020. The diagnosis of STC was based on the colonic transit and anal functional tests and barium enema to exclude secondary causes. Patients were divided into group A, which underwent surgery with SRA preservation, and group B, which underwent ligation of the SRA during surgery. Outcome assessments for both groups included the incidence of anastomotic breakdown, intraoperative complications, length of hospital stay, estimated blood loss, time to first flatus, and complications. Propensity score matching allocated 34 patients to groups A and B each. Postoperative bowel function, including time to first flatus, stool, and oral intake, recovered better in group A than in group B. Anastomotic leakage, a significant postoperative complication, was less frequent in patients with SRA preservation. In conclusion, preservation of the SRA in patients undergoing laparoscopically assisted subtotal colectomy with ileorectal anastomosis for STC is associated with favorable postoperative bowel function recovery and lower anastomotic leakage rates.

The vanadium redox flow battery (VRFB) is a highly favorable tool for storing renewable energy, and the catalytic activity of electrode materials is crucial for its development. Taurine-functionalized carbon nanotubes (CNTs) were prepared with the aim of augmenting the redox process of vanadium ions and enhancing the efficiency of the VRFB. Sulfonated CNTs were synthesized through a simple modification process in a taurine solution and used as electrocatalysts for redox reactions involving VO2+/VO2+ and V2+/V3+. The SO3H-CNTs modified at 60 °C for 2 h exhibit the best electrocatalytic activity, showing higher redox peak current values compared to pristine carboxylated CNTs (COOH-CNTs). Sulfonic acid groups added to the surface of CNTs increase active sites for redox reactions and act as carriers for mass transfer and bridges for charge transfer, accelerating the rate of the electrode reactions. A battery consisting of SO3H-CNTs as catalysts demonstrates the outstanding charge–discharge performance at a current density of 300 mA∙cm−2. This configuration displays voltage and energy efficiencies of 81.46% and 78.83%, respectively, representing enhancements of 6.15% and 6.12% compared to that equipped with conventional graphite felts (75.31%, 72.71%). This study illustrates that taurine-functionalized carbon nanotubes serve as an efficient and promising catalyst for both the anode and cathode, leading to the improved performance of the VRFB.

The urban utility tunnel is an indispensable part of modern engineering construction. However, the fire risk cannot be ignored due to the narrow space and limited ventilation of the utility tunnel. A study of smoke filling is performed in a 1/8-scaled utility tunnel (25 m × 0.5 m × 0.45 m). Five heat release rates (5, 10, 15, 20 and 25 kW) and four positions of fire sources are used for tests. The initial position of the one-dimensional smoke movement of strong plume is determined. Based on the traditional model, the longitudinal temperature attenuation model of tunnel smoke is established with consideration of radiation and convection heat losses. The theoretical value of the longitudinal temperature rise of smoke is in good agreement with the experimental value. A one-dimensional spreading velocity model is established that coincides well with the experimental value, and the relative error is less than 20%. The spreading velocity of smoke is increased by the heat release rate. The velocity of the smoke spreading at the near end is smaller than that at the center, due to the long spreading route. The current conclusions disclosed in this study provide important guidance for the ventilation design of utility tunnels for fire smoke scenarios.

The primary concerns for individuals using lithium-ion batteries (LIBs) are aging and thermal runaway (TR). This paper focuses on the thermal runaway propagation (TRP) of cycling aged LIB modules. The impacts of state of charge (SOC), state of health, and cyclic aging temperature on TRP in LIB modules are investigated. The analysis includes parameters such as temperature, voltage, and mass of the modules during TRP. It was found that as SOC increases, the maximum increase in temperature and maximum temperature rate of the modules increased, as did the total mass loss and smoke emissions. The average heat transfer between adjacent cells was higher for the lower SOC. Cycle aging reduces the thermal stability of LIBs, leading to a lower maximum temperature and maximum temperature rate, as well as a larger mass loss compared with fresh battery modules. Regarding aging temperature, low-temperature aging reduces the total duration of TRP compared with room temperature, but it increases the maximum temperature rate and causes greater mass loss. Aging also increases the average heat transfer between adjacent cells.

Activity recognition is one of the significant technologies accompanying the development of the Internet of Things (IoT). It can help in recording daily life activities or reporting emergencies, thus improving the user’s quality of life and safety, and even easing the workload of caregivers. This study proposes a human activity recognition (HAR) system based on activity data obtained via the micro-Doppler effect, combining a two-stream one-dimensional convolutional neural network (1D-CNN) with a bidirectional gated recurrent unit (BiGRU). Initially, radar sensor data are used to generate information related to time and frequency responses using short-time Fourier transform (STFT). Subsequently, the magnitudes and phase values are calculated and fed into the 1D-CNN and Bi-GRU models to extract spatial and temporal features for subsequent model training and activity recognition. Additionally, we propose a simple cross-channel operation (CCO) to facilitate the exchange of magnitude and phase features between parallel convolutional layers. An open dataset collected through radar, named Rad-HAR, is employed for model training and performance evaluation. Experimental results demonstrate that the proposed 1D-CNN+CCO-BiGRU model demonstrated superior performance, achieving an impressive accuracy rate of 98.2%. This outperformance of existing systems with the radar sensor underscores the proposed model’s potential applicability in real-world scenarios, marking a significant advancement in the field of HAR within the IoT framework.

To date, most studies have shown that biochar has great potential in carbon sequestration and reduction, as well as soil quality improvement. However, there is limited knowledge of its effect on soil organic carbon (SOC) fractions in tropical farmland. This study aimed to determine the impact of different types and rates of biochar applied in tropical farmlands on so SOC and its active fractions. The SOC, microbial biomass carbon (MBC), dissolved organic carbon (DOC), and soil mineralizable carbon (SMC) in the 0–30 cm soil layers under rice hull (R) and peanut shell (P) biochar treatments were measured. The results showed that the application of R and P biochar increased the contents, stocks, and cumulative stocks of SOC, MBC, and DOC in the 0–10 cm, 10–20 cm, and 20–30 cm soil layers. The contents, stocks, and cumulative stocks increased with increasing biochar application rates. Compared with CK, the ranges of the increased SOC, MBC, and DOC cumulative stocks were 10.76–46.36%, 30.04–195.65%, and 0.02–17.03%, respectively. However, the R60 and P60 had the lowest cumulative stocks of SMC, decreasing by 14.69% and 8.05%, respectively. The biochar treatment of more than 20 t ha−1 reduced the ratio of SMC:SOC and active fractions:SOC. Therefore, it can be inferred that the application of biochar improved the levels of SOC, MBC, and DOC, and the application of more than 20 t ha−1 biochar could decrease soil carbon mineralization, thus improving the stability of SOC in tropical farmlands.

Quantitative evaluation of stress sensitivity of ultra-deep carbonate reservoirs has been one of the challenges in exploration and development, and the problem of permeability loss law in ultra-deep carbonates under variable stress conditions has not been solved so far and further research is urgently needed. Through experimental and numerical simulation methods, the stress-sensitive evaluation equations were established based on matrix-type carbonate and fractured carbonate reservoirs, the stress-sensitive changes under different Young’s modulus were discussed, and the degree of permeability loss under different stresses was evaluated. Finally, the dual-media model of ultra-deep carbonate was established, and the practical application was carried out in the Shunbei area of the Tarim Basin. Studies have shown that (1) under the same effective stress, the stress sensitivity of matrix-type and fracture-type carbonate reservoirs is related to the Young’s modulus of the rock skeleton. In matrix-type carbonate reservoirs, rocks with a larger Young’s modulus have smaller rigidity and stronger stress sensitivity. In fracture-type carbonate reservoirs, the stress sensitivity is relatively weak under a smaller Young’s modulus, and relatively strong under a larger Young’s modulus. (2) Measured under the conditions of 87 MPa of peripheral pressure, 50 MPa of flow pressure, and 120 °C, the effective stress of matrix-type carbonate reservoirs has an exponential relationship with the permeability of reservoirs. The degree of stress sensitivity for fracture-type is generally higher than that of matrix-type reservoirs, and the smaller the Young’s modulus, the larger the difference in stress sensitivity. (3) The stress sensitivity of typical ultra-deep carbonates in the Shunbei area of the Tarim Basin is higher by establishing a dual-porosity model based on the initiating pressure gradient, which supports new evidence for the characteristics of ultra-deep carbonates with high-stress sensitivity. In actual production, the impact of stress sensitivity on the reservoir volume calculation and efficient development of ultra-deep carbonate reservoirs requires critical attention.

Objectives: The effectiveness of colonoscopy to reduce colorectal cancer (CRC) mortality is extrapolated from cohort studies in the absence of randomized controlled trial (RCT) data, whereas flexible sigmoidoscopy is supported by RCT data and may be easier to implement in practice. We characterized the anatomic distribution of CRC to determine the proportion that is visible with sigmoidoscopy. Methods: Patients with a primary diagnosis of colorectal adenocarcinoma were identified in the Surveillance, Epidemiology, and End Results program (2000–2020). Tumors from the rectum to the descending colon were categorized as visible by sigmoidoscopy, whereas more proximal tumors required colonoscopy. Differential prognosis between tumor locations, stratified by age groups and stage, was assessed using the overall restricted mean survival time (RMST) at 2, 5, and 10 years. Results: Among 309,466 patients, 58% had tumors visible by sigmoidoscopy, including 73% of those under age 50 (OR 2.10, 95% CI 2.03–2.16 age < 45, OR 2.20, 95% CI 2.13–2.27 age 45–49 versus age ≥ 50). Male sex (OR 1.54, 95% CI 1.51–1.56) and Asian or Pacific Islander race (OR 1.60, 95% CI 1.56–1.64) were also positively associated with tumors visualizable by sigmoidoscopy. Across age groups, for local disease, RMST was comparable for tumors visible versus not visible on sigmoidoscopy. For regional and metastatic cancer, patients with tumors visible by sigmoidoscopy had improved RMST versus those with more proximal tumors. Conclusions: 58% of CRC arises in locations visible by flexible sigmoidoscopy. Flexible sigmoidoscopy should be considered as a viable option for CRC screening, particularly in younger patients unwilling or unable to undergo colonoscopy.

Nucleic acid modifications play important roles in biological activities and disease occurrences, and have been considered as cancer biomarkers. Due to the relatively low amount of nucleic acid modifications in biological samples, it is necessary to develop sensitive and reliable qualitative and quantitative methods to reveal the content of any modifications. In this review, the key processes affecting the qualitative and quantitative analyses are discussed, such as sample digestion, nucleoside extraction, chemical labeling, chromatographic separation, mass spectrometry detection, and data processing. The improvement of the detection sensitivity and specificity of analytical methods based on mass spectrometry makes it possible to study low-abundance modifications and their biological functions. Some typical nucleic acid modifications and their potential as biomarkers are displayed, and efforts to improve diagnostic accuracy are discussed. Future perspectives are raised for this research field.

Oxyresveratrol is one of the active ingredients derived from mulberry branch with strong anti-inflammatory bioactivity. In this research, we want to explore if oxyresveratrol can improve cognitive impairments and episodic-like memory and its mechanism. In LPS-induced BV-2 cells, 25 μM OXY can significantly inhibit the expression of NO and alter the M1/M2 polarization by regulating M1/M2 phenotype makers. In vivo, OXY (50, 100 mg/kg) significantly reversed cognitive impairments and alleviated neuronal injuries caused by neuroinflammation. According to network pharmacology analysis, OXY alleviated neuroinflammation via the PI3K-Akt pathway. In general, the research revealed that OXY can improve cognitive impairments and episodic-like memory through alleviating LPS-induced neuroinflammation and regulating the PI3K-Akt signaling pathway.

Background: Seafood allergy is a significant global health concern that greatly impacts a patient’s quality of life. The intervention efficacy of oral immunotherapy (OIT), an emerging intervention strategy, for seafood allergy remains controversial. This study aimed to perform a systematic review and meta-analysis to evaluate the efficacy of slightly processed allergen/meat from fish and crustacea in OIT, both in mouse models and clinical patients. Methods: A comprehensive literature search was performed in four mainstream databases and the EBSCOhost database to identify all relevant case–control and cohort studies. The aim was to elucidate the intervention efficacy, encompassing various processing methods and assessing the efficacy of multiple major allergens in OIT. Results: The meta-analysis included five case–control studies on crustacean allergens in mouse models and 11 cohort studies on meat from fish and crustacea in clinical patients for final quantitative assessments. In mouse models, crustacean allergen substantially decreased the anaphylactic score after OIT treatment (mean difference (MD) = −1.30, p < 0.01). Subgroup analyses with low-level heterogeneities provided more reliable results for crab species (MD = −0.63, p < 0.01, I2 = 0), arginine kinase allergen (MD = −0.83, p < 0.01, I2 = 0), and Maillard reaction processing method (MD = −0.65, p < 0.01, I2 = 29%), respectively. In clinical patients, the main meta-analysis showed that the slightly processed meat significantly increased the incidence rate of oral tolerance (OT, incidence rate ratio (IRR) = 2.90, p < 0.01). Subgroup analyses for fish meat (IRR = 2.79, p < 0.01) and a simple cooking treatment (IRR = 2.36, p = 0.01) also demonstrated a substantial increase in the incidence rate of OT. Sensitivity and meta-regression analyses successfully identified specific studies contributing to heterogeneity in mouse models and clinical patients, although these studies did not impact the overall significant pooled effects. Conclusions: This meta-analysis provides preliminary evidence for the high intervention efficacy of slightly processed allergen/meat from fish and crustacea in OIT, both in mouse models and clinical patients. The Maillard reaction and cooking processing methods may emerge as potentially effective approaches to treating allergen/meat in OIT for clinical patients, offering a promising and specific treatment strategy for seafood allergy. However, these findings should be interpreted cautiously, and further supporting evidence is necessary.

The full waveform inversion at this stage still has many problems in the recovery of deep background velocities. Velocity modeling based on end-to-end deep learning usually lacks a generalization capability. The proposed method is a multi-scale convolutional neural network velocity inversion (Ms-CNNVI) that incorporates a multi-scale strategy into the CNN-based velocity inversion algorithm for the first time. This approach improves the accuracy of the inversion by integrating a multi-scale strategy from low-frequency to high-frequency inversion and by incorporating a smoothing strategy in the multi-scale (MS) convolutional neural network (CNN) inversion process. Furthermore, using angle-domain reverse time migration (RTM) for dataset construction in Ms-CNNVI significantly improves the inversion efficiency. Numerical tests showcase the efficacy of the suggested approach.

Brasenia schreberi J.F. Gmel (BS) is rich in mucilage, which has diverse biological activities, and is utilized in the food and pharmaceutical industries due to its nutritional value. Proteomics analysis was employed to investigate the cause of mucilage disappearance in BS and its effect on nutrient accumulation. Among the 2892 proteins identified, 840 differentially expressed proteins (DEPs) were found to be involved in mucilage development. By comparing the expression patterns and functions and pathway enrichment, the DEPs mainly contributed to carbon and energy metabolism, polysaccharide metabolism, and photosynthesis. Our study also revealed positive correlations between mucilage accumulation and tryptophan metabolism, with high levels of indole-3-acetic (IAA) contributing to mucilage accumulation. Furthermore, environmental changes and particularly excessive nutrients were found to be detrimental to mucilage synthesis. Overall, in the absence of various stimuli in the growing environment, BS accumulates more nutrients within the plant itself instead of producing mucilage.

As the prevalence of electric vehicles (EVs) continues to grow, their charging and discharging behaviors pose a challenge to the stable operation of power systems. Therefore, this paper analyzes the charging demand of EV users through GPS trajectory data and proposes an EV-discharging-optimization model based on vehicle-to-grid interaction (V2G). Firstly, the spatial–temporal distribution of EV-charging demand is obtained by cleaning and mining the big data of traveling vehicles, considering dynamic energy consumption theory and users’ willingness; secondly, a probabilistic model of EV users’ participation in V2G-demand response is constructed based on expected utility theory, which both considers the heterogeneity of users and reflects the interactive influence of users’ decisions; finally, a scheduling model of EV discharging in the regional grid is established. The results show that the proposed model can explore the potential of user participation in V2G in the study area, and the V2G response resources can reduce the grid fluctuation and enable users to obtain certain benefits, which achieves a win–win situation between the grid side and the user side.

Austin was first isolated as a novel polyisoprenoid mycotoxin from Aspergillus ustus in 1976. Subsequently, some new austin-type meroterpenoids (ATMTs) have been continually found. This review attempts to give a comprehensive summary of progress on the isolation, chemical structural features, biological activities, and fungal biodiversity of 104 novel ATMTs from 5 genera of terrestrial- and marine-derived fungi reported from October 1976 to January 2023. The genera of Penicillium and Aspergillus are the two dominant producers, producing 63.5% and 30.8% of ATMTs, respectively. Moreover, about 26.9% of ATMTs display various pronounced bioactivities, including insecticidal, anti-inflammatory, cytotoxicity, antibacterial, and PTP1B inhibitory activities. The chemical diversity and potential activities of these novel fungal ATMTs are reviewed for a better understanding, and a relevant summary focusing on the source fungi and their taxonomy is provided to shed light on the future development and research of austin-type meroterpenoids.

Chemical foaming technology is widely used in the preparation of silicone rubber foam and is attributable to its one-step molding capability and eco-friendly production processes. The microrheological properties of silicone rubber play a pivotal role during the foaming process. In this study, Rheolaser Lab (Formulaction, Toulouse, France) was used to conduct in situ examinations for the influence of a crosslinking agent on the microrheological properties of silicone rubber foam for the first time. This study monitors the entire reaction process of silicone rubber foam from liquid to solid, as well as the matching of crosslinking and foaming reactions. Various parameters, including solid–liquid balance, elasticity index, and macroscopic viscosity index, are measured to analyze the microrheological properties of silicone rubber foam. The results show that the silicone rubber foam exhibits good microrheological properties, thereby demonstrating excellent performance at a crosslinking agent content of 2%. Through adjusting the experimental conditions, a sustainable and efficient approach was proposed for better cellular structure control in the industrial preparation of silicone rubber foam.

Oysters that filter feed can accumulate numerous pathogens, including viruses, which can serve as a valuable viral repository. As oyster farming becomes more prevalent, concerns are mounting about diseases that can harm both cultivated and wild oysters. Unfortunately, there is a lack of research on the viruses and other factors that can cause illness in shellfish. This means that it is harder to find ways to prevent these diseases and protect the oysters. This is part of a previously started project, the Dataset of Oyster Virome, in which we further study 30 almost complete genomes of oyster-associated CRESS DNA viruses. The replication-associated proteins and capsid proteins found in CRESS DNA viruses display varying evolutionary rates and frequently undergo recombination. Additionally, some CRESS DNA viruses have the capability for cross-species transmission. A plethora of unclassified CRESS DNA viruses are detectable in transcriptome libraries, exhibiting higher levels of transcriptional activity than those found in metagenome libraries. The study significantly enhances our understanding of the diversity of oyster-associated CRESS DNA viruses, emphasizing the widespread presence of CRESS DNA viruses in the natural environment and the substantial portion of CRESS DNA viruses that remain unidentified. This study’s findings provide a basis for further research on the biological and ecological roles of viruses in oysters and their environment.

Distal outflow bleb-forming procedures in ophthalmic surgery expose subconjunctival tissue to inflammatory cytokines present in the aqueous humor, resulting in impaired outflow and, consequently, increased intraocular pressure. Clinically, this manifests as an increased risk of surgical failure often necessitating revision. This study (1) introduces a novel high-throughput screening platform for testing potential anti-fibrotic compounds and (2) assesses the clinical viability of modulating the transforming growth factor beta-SMAD2/3 pathway as a key contributor to post-operative outflow reduction, using the signal transduction inhibitor verteporfin. Human Tenon’s capsule fibroblasts (HTCFs) were cultured within a 3D collagen matrix in a microfluidic system modelling aqueous humor drainage. The perfusate was augmented with transforming growth factor beta 1 (TGFβ1), and afferent pressure to the tissue-mimetic was continuously monitored to detect treatment-related pressure elevations. Co-treatment with verteporfin was employed to evaluate its capacity to counteract TGFβ1 induced pressure changes. Immunofluorescent studies were conducted on the tissue-mimetic to corroborate the pressure data with cellular changes. Introduction of TGFβ1 induced treatment-related afferent pressure increase in the tissue-mimetic. HTCFs treated with TGFβ1 displayed visibly enlarged cytoskeletons and stress fiber formation, consistent with myofibroblast transformation. Importantly, verteporfin effectively mitigated these changes, reducing both afferent pressure increases and cytoskeletal alterations. In summary, this study models the pathological filtration bleb response to TGFβ1, while demonstrating verteporfin’s effectiveness in ameliorating both functional and cellular changes caused by TGFβ1. These demonstrate modulation of the aforementioned pathway as a potential avenue for addressing post-operative changes and reductions in filtration bleb outflow capacity. Furthermore, the establishment of a high-throughput screening platform offers a valuable pre-animal testing tool for investigating potential compounds to facilitate surgical wound healing.

Six benzophenone derivatives, carneusones A-F (1–6), along with seven known compounds (7–13) were isolated from a strain of sponge-derived marine fungus Aspergillus carneus GXIMD00543. Their chemical structures were elucidated by detailed spectroscopic data and quantum chemical calculations. Compounds 5, 6, and 8 exhibited moderate anti-inflammatory activity on NO secretion using lipopolysaccharide (LPS)-induced RAW 264.7 cells with EC50 values of 34.6 ± 0.9, 20.2 ± 1.8, and 26.8 ± 1.7 μM, while 11 showed potent effect with an EC50 value of 2.9 ± 0.1 μM.

Gait analysis has been studied over the last few decades as the best way to objectively assess the technical outcome of a procedure designed to improve gait. The treating physician can understand the type of gait problem, gain insight into the etiology, and find the best treatment with gait analysis. The gait parameters are the kinematics, including the temporal and spatial parameters, and lack the activity information of skeletal muscles. Thus, the gait analysis measures not only the three-dimensional temporal and spatial graphs of kinematics but also the surface electromyograms (sEMGs) of the lower limbs. Now, the shoe-worn GaitUp Physilog® wearable inertial sensors can easily measure the gait parameters when subjects are walking on the general ground. However, it cannot measure muscle activity. The aim of this study is to measure the gait parameters using the sEMGs of the lower limbs. A self-made wireless device was used to measure the sEMGs from the vastus lateralis and gastrocnemius muscles of the left and right feet. Twenty young female subjects with a skeletal muscle index (SMI) below 5.7 kg/m2 were recruited for this study and examined by the InBody 270 instrument. Four parameters of sEMG were used to estimate 23 gait parameters. They were measured using the GaitUp Physilog® wearable inertial sensors with three machine learning models, including random forest (RF), decision tree (DT), and XGBoost. The results show that 14 gait parameters could be well-estimated, and their correlation coefficients are above 0.800. This study signifies a step towards a more comprehensive analysis of gait with only sEMGs.

In this work, we report a new concept of upconversion-powered photoelectrochemical (PEC) bioanalysis. The proof-of-concept involves a PEC bionanosystem comprising a NaYF4:Yb,Tm@NaYF4 upconversion nanoparticles (UCNPs) reporter, which is confined by DNA hybridization on a CdS quantum dots (QDs)/indium tin oxide (ITO) photoelectrode. The CdS QD-modified ITO electrode was powered by upconversion absorption together with energy transfer effect through UCNPs for a stable photocurrent generation. By measuring the photocurrent change, the target DNA could be detected in a specific and sensitive way with a wide linear range from 10 pM to 1 μM and a low detection limit of 0.1 pM. This work exploited the use of UCNPs as signal reporters and realized upconversion-powered PEC bioanalysis. Given the diversity of UCNPs, we believe it will offer a new perspective for the development of advanced upconversion-powered PEC bioanalysis.

Interleukin-6 (IL-6), a pleiotropic cytokine, plays a crucial role in acute stress induced by bacterial infection and is strongly associated with reactive oxygen species (ROS) production. However, the role of IL-6 in the liver of fish after Aeromonas hydrophila infection remains unclear. Therefore, this study constructed a zebrafish (Danio rerio) il-6 knockout line by CRISPR/Cas9 to investigate the function of IL-6 in the liver post bacterial infection. After infection with A. hydrophila, pathological observation showed that il-6−/− zebrafish exhibited milder liver damage than wild-type (WT) zebrafish. Moreover, liver transcriptome sequencing revealed that 2432 genes were significantly up-regulated and 1706 genes were significantly down-regulated in il-6−/− fish compared with WT fish after A. hydrophila infection. Further, gene ontology (GO) analysis showed that differentially expressed genes (DEGs) were significantly enriched in redox-related terms, including oxidoreductase activity, copper ion transport, etc. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that DEGs were significantly enriched in pathways such as the PPAR signaling pathway, suggesting that il-6 mutation has a significant effect on redox processes in the liver after A. hydrophila infection. Additionally, il-6−/− zebrafish exhibited lower malondialdehyde (MDA) levels and higher superoxide dismutase (SOD) activities in the liver compared with WT zebrafish following A. hydrophila infection, indicating that IL-6 deficiency mitigates oxidative stress induced by A. hydrophila infection in the liver. These findings provide a basis for further studies on the role of IL-6 in regulating oxidative stress in response to bacterial infections.

Extrusion technology is widely utilized in the rubber processing industry, with the extruder serving as the core equipment. As mixed rubber enters the extruder, it undergoes conveyance and plasticization, ultimately forming specific shapes and dimensions upon extrusion. The extruder head is a crucial component, playing a key role in achieving the final product’s required size and shape. Factors such as its structure, materials, and manufacturing processes significantly impact the efficiency, product quality, and sustainability of the extrusion process. However, prolonged operation leads to severe wear of the extruder head, adversely affecting rubber product quality. Additionally, extruder head processing poses challenges, with maintenance and repair being complex procedures. Therefore, exploring a wear-resistant, long-lasting metal material for the extruder head without compromising mixed rubber performance is essential. This study focuses on severely worn extruder head metal materials, comparing wear levels after friction with STELLITE 6 alloy, Hastelloy C-276 alloy, 38CrMoAlA, and tungsten carbide with composite rubber. Results show that compared to the NR/BR composite material after Hastelloy C-276 alloy friction, rubber Payne effect increased by 4.4% (38CrMoAl), 3.2% (STELLITE 6), and 4.6% (tungsten carbide). Similarly, rubber dispersion decreased by 9.4% (38CrMoAl), 4.7% (STELLITE 6), and 9.8% (tungsten carbide). Rolling resistance increased by 18.1% (38CrMoAl), 16% (STELLITE 6), and 23.4% (tungsten carbide). Friction coefficient increased by 3.5% (38CrMoAl), 2.8% (STELLITE 6), and 4.3% (tungsten carbide). Wear volume increased by 39.3% (38CrMoAl), 45.3% (STELLITE 6), and 48.9% (tungsten carbide). Specifically, using Hastelloy C-276 alloy as the extruder head metal material yields the best NR/BR composite material dispersion, highest ten times tear strength, excellent anti-wet skid resistance, and minimum rolling resistance. Conversely, using the other alloys results in varying reductions in the physical and mechanical properties of NR/BR composite materials. This research is crucial for improving rubber product quality and extending extruder head lifespan.

To achieve omnidirectional sensitive detection of partial discharge (PD) in transformers and to avoid missing PD signals, a fiber optic omnidirectional sensing method for PD in transformers combined with the fiber Bragg grating (FBG) and Fabry-Perot (F-P) cavity is proposed. The fiber optic omnidirectional sensor for PD as a triangular prism was developed. The hollow structure of the probe was used to insert a single-mode fiber to form an F-P cavity. In addition, the three sides of the probe were used to form a diaphragm-type FBG sensing structure. The ultrasound sensitization diaphragm was designed based on the frequency characteristics of PD in the transformer and the vibration model of the diaphragm in the liquid environment. The fiber optic sensing system for PD was built and the performance test was conducted. The results show that the resonant frequency of the FBG acoustic diaphragm is around 20 kHz and that of the F-P cavity acoustic diaphragm is 94 kHz. The sensitivity of the developed fiber optic sensor is higher than that of the piezoelectric transducer (PZT). The lower limit of PD detection is 68.72 pC for the FBG sensing part and 47.97 pC for the F-P cavity sensing part. The directional testing of the sensor and its testing within a transformer simulation model indicate that the proposed sensor achieves higher detection sensitivity of PD in all directions. The omnidirectional partial discharge ultrasound sensing method proposed in this paper is expected to reduce the missed detection rate of PD.

This study proposes a three-lens design with a short lens length and explores the curved imaging plane and performs a relative illumination analysis. There are two ways to reduce the lens length: shortening the back focal and lens group lengths. We derived the relevant parameter relationships of three lenses using the first-order geometric optics theory. The optical lens length can be controlled within 2 mm. The shorter the lens length, the larger the angle of the chief ray in the image space, resulting in an increase in the field curvature and astigmatism. Third-order Seidel aberrations can be effectively reduced by a curved image plane. We also derived the equations for relative illuminance, solid angle, surface transmittance, and internal transmittance for the short three-lens design. The optical lens design uses a curved image plane to shorten the distance from the off-axis beam image space to the image plane and reduce the incident angle of the chief ray on the image plane. The formula and design results verified by Code V software (version 11.2) show that both the solid angle and relative contrast of the lens can be increased. For the proposed three-lens design with a short lens length, the semi-field angle is 32°, F/# is 2.7, the effective focal length is 1.984 mm, the image plane area is 2.16 mm × 1.22 mm, and the curvature radius of the concave image plane is 3.726 mm. Moroever, the MTF (100 lp/mm) is larger than 52%, the lateral color aberration is less than 2.12 μm, the optical distortion is less than 2.00%, and the relative illumination is greater than 68%.

Site-specific integration is an important approach used to address the problem of unstable cell lines in industry. In this study, we observed a reduction in the gene copy number and antibody production in a CHOK1 cell line BA03 capable of high antibody expression. We identified a new integration site named locus 7 in the intron region of the parva gene through sequencing, FISH, and genome walking. We demonstrate that the integration of the exogenous gene at this locus does not affect the transcription of the parva and, therefore, has a minimal impact on cell growth. We designed sgRNA and donor vectors to integrate the etanercept-coding gene into locus 7 and obtained a cell line, SSI-4. We performed a passaged stability study on SSI-4 and proved the possibility of the stable, site-specific integration of exogenous genes at this locus in terms of integration site, copy number, expression level, and cell growth. In summary, our study has identified a new integration site suitable for site-specific integration, which lays the foundation for the subsequent development of site-specific integration cell lines.

The linkage mechanism of a cotton bundle fiber strength tester will have an unstable clamping force when clamping fiber bundle samples with uneven thickness, resulting in slippage or damage to the fibers increasing the pectin residue, leading to inaccurate test results and increased maintenance costs. To address this problem, according to the structural principle of the connecting rod-clamping mechanism, through the geometric relationship between the connecting rods to establish a parametric model of the mechanism and the use of the principle of virtual work on the mechanism to solve the force, the proposed new Dynamic Alternative Static Approximate Analysis Method (DASAAM) was based on Adams 2020. The Isight integrated Adams automatic optimization design framework was built. The variance of the change curve of the end force of the mechanism when clamping samples of different thicknesses was used as the evaluation function and the assembly conditions were used as the constraints. The dimensional parameters and angles of the mechanism were optimized using the multi-island genetic algorithm. The simulation results showed that when the thickness of the clamped sample varied in the range of 0–4 mm, the clamping force of the mechanism varied in the range of 8920–8630 N. Finally, the variance of the clamping force measured by the clamping force measurement component was 0.0367. The above results show that the DASAAM provided a new method for solving the static problem of mechanism morphological and position change, and the optimized linkage mechanism had better clamping force stability, which made the strength detection of cotton fiber more accurate, thus improving the quality of textile products.

The severity of liver functional reserve is an important prognostic predictor in hepatocellular carcinoma (HCC). The albumin–bilirubin (ALBI), easy (EZ)-ALBI, platelet-albumin–bilirubin (PALBI), platelet–albumin (PAL) score, and MELD 3.0 score are used to evaluate the severity of liver dysfunction. However, their prognostic role in HCC patients, specifically with renal insufficiency (RI), is unclear. We aimed to investigate the predictive accuracy of the five models in these patients. A total of 1120 newly diagnosed HCC patients with RI were enrolled. A multivariate Cox proportional analysis was used to identify independent predictors associated with survival. In the Cox model, older age, an α-fetoprotein ≥20 ng/mL, vascular invasion, a medium and high tumor burden score, poor performance status, a higher ALBI grade, an EZ-ALBI grade, a PALBI grade, a PAL grade, and MELD 3.0 score were all independently associated with decreased overall survival (all p < 0.001). Among the five liver reserve models, the ALBI grade is the best surrogate marker to represent liver functional reserve in terms of outcome prediction. The albumin-based liver reserve models (ALBI, EZ-ALBI, PALBI, and PAL) and MELD 3.0 are all feasible prognostic markers to indicate liver injury, specifically in HCC patients with RI. Among them, the ALBI grade is the most robust tool for survival prediction in these patients.

Recalcitrant rice blast disease is caused by Magnaporthe oryzae, which has a significant negative economic reverberation on crop productivity. In order to induce the disease onto the host, M. oryzae positively generates many types of small secreted proteins, here named as effectors, to manipulate the host cell for the purpose of stimulating pathogenic infection. In M. oryzae, by engaging with specific receptors on the cell surface, effectors activate signaling channels which control an array of cellular activities, such as proliferation, differentiation and apoptosis. The most recent research on effector identification, classification, function, secretion, and control mechanism has been compiled in this review. In addition, the article also discusses directions and challenges for future research into an effector in M. oryzae.

The genus Carthamus Linnaeus, which belongs to the tribe Cardueae in the Asteraceae family, originated in the Mediterranean region and consists of approximately 20 species worldwide. Understanding the phylogeny of the Carthamus is crucial for the cultivation of C. tinctorius. Although chloroplast genomes are widely used for species identification and evolutionary studies, there have been limited investigations on the chloroplast genomes of Carthamus species. In this study, we assembled the chloroplast genomes of C. persicus, C. tinctorius × C. persicus, and C. lanatus and combined them with the five chloroplast genomes of C. tinctorius for comparative genomic analysis. The sizes of the chloroplast genomes of C. lanatus, C. persicus, and C. tinctorius × C. persicus were 152,602 bp, 153,177 bp, and 153,177 bp, respectively. Comparative analysis showed that the chloroplast genome structures of the four Carthamus species were highly conserved. Additionally, the phylogenomic analysis demonstrated that the plastid genome and angiosperms353 dataset significantly improved the phylogenetic support of Carthamus species. This analysis supported Carthamus as a monophyletic taxon and its internal division into the sect. Carthamus and sect. Atractylis. The Carthamus was closely related to Carduncellus, Femeniasia, Phonus, and Centaurea. In conclusion, this study not only expands our understanding of the cp genomes of Carthamus species but also provides support for more comprehensive phylogenetic studies of Carthamus.

Self-supervised monocular depth estimation, which has attained remarkable progress for outdoor scenes in recent years, often faces greater challenges for indoor scenes. These challenges comprise: (i) non-textured regions: indoor scenes often contain large areas of non-textured regions, such as ceilings, walls, floors, etc., which render the widely adopted photometric loss as ambiguous for self-supervised learning; (ii) camera pose: the sensor is mounted on a moving vehicle in outdoor scenes, whereas it is handheld and moves freely in indoor scenes, which results in complex motions that pose challenges for indoor depth estimation. In this paper, we propose a novel self-supervised indoor depth estimation framework-PMIndoor that addresses these two challenges. We use multiple loss functions to constrain the depth estimation for non-textured regions. We introduce a pose rectified network that only estimates the rotation transformation between two adjacent frames of images for the camera pose problem, and improves the pose estimation results with the pose rectified network loss. We also incorporate a multi-head self-attention module in the depth estimation network to enhance the model’s accuracy. Extensive experiments are conducted on the benchmark indoor dataset NYU Depth V2, demonstrating that our method achieves excellent performance and is better than previous state-of-the-art methods.

Hemoglobin (Hb) usually comprises two α and two β subunits, forming a tetramer responsible for oxygen transportation and storage. Few studies have elucidated fish hemoglobin immune functions. Megalobrama amblycephala is a freshwater-cultured fish prevalent in China. We identified two M. amblycephala hemoglobin subunits and analyzed their expression patterns and antibacterial activities. The respective full-length cDNA sequences of the M. amblycephala Hb α (MaHbα) and β (MaHbβ) subunits were 588 and 603 bp, encoding 143 and 148 amino acids. MaHbα and MaHbβ were highly homologous to hemoglobins from other fish, displaying typical globin-like domains, most heme-binding sites, and tetramer interface regions highly conserved in teleosts. In phylogenetic analyses, the hemoglobin genes from M. amblycephala and other cypriniformes clustered into one branch, and those from other fishes and mammals clustered into other branches, revealing fish hemoglobin conservation. These M. amblycephala Hb subunits exhibit different expression patterns in various tissues and during development. MaHbα is mainly expressed in the blood and brain, while MaHbβ gene expression is highest in the muscle. MaHbα expression was detectable and abundant post-fertilization, with levels fluctuating during the developmental stages. MaHbβ expression began at 3 dph and gradually increased. Expression of both M. amblycephala Hb subunits was down-regulated in most examined tissues and time points post-Aeromonas hydrophila infection, which might be due to red blood cell (RBC) and hematopoietic organ damage. Synthetic MaHbα and MaHbβ peptides showed excellent antimicrobial activities, which could inhibit survival and growth in five aquatic pathogens. Two M. amblycephala hemoglobin subunits were identified, and their expression patterns and antibacterial activities were analyzed, thereby providing a basis for the understanding of evolution and functions of fish hemoglobins.

To improve communication stability, more wireless devices transmit multi-modal signals while operating. The term ‘modal’ refers to signal waveforms or signal types. This poses challenges to traditional specific emitter identification (SEI) systems, e.g., unknown modal signals require extra open-set mode identification; different modes require different radio frequency fingerprint (RFF) extractors and SEI classifiers; and it is hard to collect and label all signals. To address these issues, we propose an enhanced SEI system consisting of a universal RFF extractor, denoted as multiple synchrosqueezed wavelet transformation of energy unified (MSWTEu), and a new generative adversarial network for feature transferring (FTGAN). MSWTEu extracts uniform RFF features for different modal signals, FTGAN transfers different modal features to a recognized distribution in an unsupervised manner, and a novel training strategy is proposed to achieve emitter identification across multi-modal signals using a single clustering method. To evaluate the system, we built a hybrid dataset, which consists of multi-modal signals transmitted by various emitters, and built a complete civil air traffic control radar beacon system (ATCRBS) dataset for airplanes. The experiments show that our enhanced SEI system can resolve the SEI problems associated with crossing signal modes. It directly achieves 86% accuracy in cross-modal emitter identification using an unsupervised classifier, and simultaneously obtains 99% accuracy in open-set recognition of signal mode.
