Ultrasound micromolding technology has been applied to get microporous polylactide scaffolds from the subsequent leaching of incorporated NaCl salts. A small amount of water-soluble polyethylene glycol (PEG) was required in order to improve the leaching process and get compact pieces with interconnected pores. Distribution of polymers in the processed specimens was quite homogeneous due to the small PEG content, although it was more concentrated in the regions close to the feeding channels due to its higher viscosity. Hydrophobic drugs like triclosan could be incorporated causing a minimum degradation during ultrasound processing and suffering an insignificant solubilization during the leaching step. Final scaffolds showed clear bactericide or bacteriostatic effects before and after 10 h of exposure. Cell proliferation of MDCK epithelial cells was higher for TCS loaded porous scaffolds (200%) than for unloaded samples (170%) and non-porous polylactide (PLA) specimens (100%, control). Micrographs showed the absence of non-inhibition areas in both the specimens and the container, confirming the biocompatibility of PLA specimens.

A technology based on the application of ultrasound as an energy source was applied to get polylactide (PLA) micropieces with minimum degradation and processing time. This requirement could be even shorter than 1.5 s. The ultrasound technology was also demonstrated to be efficient for the incorporation of drugs with a pharmacological activity. Thus, the loading of two representative bactericide agents (i.e., triclosan (TCS), and chlorhexidine (CHX)), having differentiated chemical properties was evaluated. Typical physicochemical characterization included mechanical and thermal properties together with the evaluation of molecular degradation during processing for both unloaded and loaded specimens. Results pointed out that the thermally stable TCS could be loaded into the specimens without any problem, but cautions should be taken into account for CHX. Nevertheless, degradation could in this case be avoided when the drug load was lower than 3 wt-%, a result that contrasts with the significant decomposition attained by using conventional melting processes, which required long processing times at high temperatures. Morphologic analyses of loaded specimens did not reveal significant defects, while spectroscopic analyses showed that a good dispersion of drugs inside pieces could be attained. Drugs were slowly released from micropieces with a rate that was dependent on their hydrophilic character. Thus, release in a phosphate buffered saline (PBS)-ethanol medium (70% of PBS) followed a first order kinetics with constants of 0.0356 h&minus, 1 and 0.027 h&minus, 1 for CHX and TCS, respectively. A clear bactericide effect against both Gram-positive and Gram-negative bacteria was achieved at the beginning of exposure to the corresponding culture media, while a bacteriostatic effect was interestingly still detected after long exposure times. In fact, bacterial growth could be reduced to near 20% when micropieces were loaded with only 3 wt-% of any of the selected CHX and TCS drugs.

Biodegradable polyester fibers have been loaded with two engineered analogues of gramicidin soviet. In these cyclic peptide derivatives, which were designed in a previous work to stabilize the bioactive conformation while enhancing the antimicrobial activity, the D-Phe was replaced by D-Pro, and the L-Pro was changed by 1-aminocyclopropanecarboxylic acid (Ac3c) or by an Ac3c derivative with two vicinal phenyl substituents in a trans relative disposition (S,S-c3diPhe). The diameter, topography, thermal stability and wettability of the polyester fibers, which have been obtained by electrospinning, strongly depend on the molecular constraints and stability of the loaded peptides. More specifically, unloaded and linear gramicidin-loaded fibers (used as control) are hydrophobic, rough and micrometric, while fibers loaded with the cyclic peptides are hydrophilic, ultra-smooth, nanometric and less thermally stable. The activity of the two cyclic peptides increases when loaded into polyester fibers, suggesting that the polymeric matrix stabilizes the bioactive β-sheet structure. Hybrid materials constituted by peptides and synthetic polymers have nowadays a great interest since they can combine the properties and functions of each constitutive block, being also possible to modify the final characteristics by using different topologies. Poly(L-lactide-b-L-phenylalanine) copolymers with various block lengths were synthesized by sequential ring-opening polymerization of L-lactide and the N-carboxyanhydride of L-phenylalanine. The resulting block copolymers were characterized by NMR spectrometry, IR spectroscopy, gel permeation chromatography, MALDI-TOF and UV-vis, revealing the successful incorporation of the polyphenylalanine (PPhe) peptide into the previously formed poly(L-lactide) (PLLA) polymer chain. X-ray diffraction and DSC data also suggested that the copolymers were phase-separated in domains containing either crystalline PLLA or PPhe phases. A peculiar thermal behavior was also found by thermogravimetric analysis when polyphenylalanine blocks were incorporated into polylactide.

The aim of this work was to prepare and characterize polymer–ceramic composite material for dental applications, which must resist fracture and wear under extreme forces. It must also be compatible with the hostile environment of the oral cavity. The most common restorative and biocompatible copolymer, 2,2-bis(p-(2′-2-hydroxy-3′-methacryloxypropoxy)phenyl)propane and triethyleneglycol dimethacrylate, was combined with 3D-printed yttria-stabilized tetragonal zirconia scaffolds with a 50% infill. The proper scaffold deposition and morphology of samples with 50% zirconia infill were studied by means of X-ray computed microtomography and scanning electron microscopy. Samples that were infiltrated with copolymer were observed under compression stress, and the structure’s failure was recorded using an Infrared Vic 2DTM camera, in comparison with empty scaffolds. The biocompatibility of the composite material was ascertained with an MG-63 cell viability assay. The microtomography proves the homogeneous distribution of pores throughout the whole sample, whereas the presence of the biocompatible copolymer among the ceramic filaments, referred to as a polymer-infiltrated ceramic network (PICN), results in a safety “damper”, preventing crack propagation and securing the desired material flexibility, as observed by an infrared camera in real time. The study represents a challenge for future dental implant applications, demonstrating that it is possible to combine the fast robocasting of ceramic paste and covalent bonding of polymer adhesive for hybrid material stabilization.

The impact of respiratory coinfections in COVID-19 is still not well understood despite the growing evidence that consider coinfections greater than expected. A total of 295 patients older than 18 years of age, hospitalized with a confirmed diagnosis of moderate/severe pneumonia due to SARS-CoV-2 infection (according to definitions established by the Ministry of Health of Peru) were enrolled during the study period. A coinfection with one or more respiratory pathogens was detected in 154 (52.2%) patients at hospital admission. The most common coinfections were Mycoplasma pneumoniae (28.1%), Chlamydia pneumoniae (8.8%) and with both bacteria (11.5%), followed by Adenovirus (1.7%), Mycoplasma pneumoniae/Adenovirus (0.7%), Chlamydia pneumoniae/Adenovirus (0.7%), RSV-B/Chlamydia pneumoniae (0.3%), and Mycoplasma pneumoniae/Chlamydia pneumoniae/Adenovirus (0.3%). Expectoration was less frequent in coinfected individuals compared to non-coinfected (5.8% vs. 12.8%). Sepsis was more frequent among coinfected patients than non-coinfected individuals (33.1% vs. 20.6%), and 41% of the patients who received macrolides empirically were PCR-positive for Mycoplasma pneumoniae and Chlamydia pneumoniae.
