Abstract The analysis of eye movements has proven valuable for understanding brain function and the neuropathology of various disorders. This research aims to utilize eye movement data analysis as a screening tool for differentiation between eight different groups of pathologies, including scholar, neurologic, and postural disorders. Leveraging a dataset from 20 clinical centers, all employing AIDEAL and REMOBI eye movement technologies this study extends prior research by considering a multi-annotation setting, incorporating information from recordings from saccade and vergence eye movement tests, and using contextual information (e.g. target signals and latency of the eye movement relative to the target and confidence level of the quality of eye movement recording) to improve accuracy while reducing noise interference. Additionally, we introduce a novel hybrid architecture that combines the weight-sharing feature of convolution layers with the long-range capabilities of the transformer architecture to improve model efficiency and reduce the computation cost by a factor of 3.36, while still being competitive in terms of macro F1 score. Evaluated on two diverse datasets, our method demonstrates promising results, the most powerful discrimination being Attention & Neurologic; with a macro F1 score of up to 78.8%; disorder. The results indicate the effectiveness of our approach in classifying eye movement data from different pathologies and different clinical centers accurately, thus enabling the creation of an assistant tool in the future.

Abstract Though saccade and vergence eye movements are fundamental for everyday life, the way these movements change as we age has not been sufficiently studied. The present study examines the effect of age on vergence and saccade eye movement characteristics (latency, peak and average velocity, amplitude) and on audiovisual facilitation. We compare the results for horizontal saccades and vergence movements toward visual and audiovisual targets in a young group of 22 participants (mean age 25 ± 2.5) and an elderly group of 45 participants (mean age 65 + 6.9). The results show that, with increased age, latency of all eye movements increases, average velocity decreases, amplitude of vergence decreases, and audiovisual facilitation collapses for vergence eye movements in depth but is preserved for saccades. There is no effect on peak velocity, suggesting that, although the sensory and attentional mechanisms controlling the motor system does age, the motor system itself does not age. The loss of audiovisual facilitation along the depth axis can be attributed to a physiologic decrease in the capacity for sound localization in depth with age, while left/right sound localization coupled with saccades is preserved. The results bring new insight for the effects of aging on multisensory control and attention.

Abstract The kinase haspin phosphorylates histone H3 at threonine-3 (H3T3ph) during mitosis. H3T3ph provides a docking site for the Chromosomal Passenger Complex at the centromere, enabling correction of erratic microtubule-chromosome contacts. Although this mechanism is operational in all dividing cells, haspin-null mice do not exhibit developmental anomalies, apart from aberrant testis architecture. Investigating this problem, we show here that mouse embryonic stem cells that lack or overexpress haspin, albeit prone to chromosome misalignment during metaphase, can still divide, expand and differentiate. RNA sequencing reveals that haspin dosage affects severely the expression levels of several genes that are involved in male gametogenesis. Consistent with a role in testis-specific expression, H3T3ph is detected not only in mitotic spermatogonia and meiotic spermatocytes, but also in non-dividing cells, such as haploid spermatids. Similarly to somatic cells, the mark is erased in the end of meiotic divisions, but re-installed during spermatid maturation, subsequent to methylation of histone H3 at lysine-4 (H3K4me3) and arginine-8 (H3R8me2). These serial modifications are particularly enriched in chromatin domains containing histone H3 trimethylated at lysine-27 (H3K27me3), but devoid of histone H3 trimethylated at lysine-9 (H3K9me3). The unique spatio-temporal pattern of histone H3 modifications implicates haspin in the epigenetic control of spermiogenesis.

Abstract Previous studies suggest vergence and saccade abnormalities in dyslexic adolescents. However, these studies are mainly clinically based and do not provide objective measurements of eye movements, but rather subjectively evaluate vergence using haplosopic conditions in which the two eyes are dissociated (via polarizers, prisms, or intermittent spectacles). Other studies have identified deficits with binocular coordination during reading in dyslexics. Yet, there are few studies that provide objective measurements of eye movements in the dyslexic population to help provide more information regarding if these deficits could be due to an intrinsic motor problem or if they are the consequence of poor reading. 47 dyslexic adolescents (18 female, 29 male; mean age 15.5) and 44 non-dyslexic adolescents (22 female, 22 male; mean age 14.8) wore a head-based eye tracker (PupilCore, Pupil Labs, Berlin) which recorded wide angle saccade and vergence eye movements at 200 Hz. Tests were run using the REMOBI device, which produced a saccade or vergence audiovisual target. Analysis of eye movements was performed with lab-developed software, AIDEAL. The results showed statistically significant abnormalities in vergence and saccades. In vergence, dyslexics displayed a reduced amplitude of the visually driven portion of convergence and a longer duration in the initial phase of divergence. In saccades, dyslexic adolescents demonstrated slower saccades in both directions. They also had an increased disconjugate drift in the first 80 or 160 ms following saccades to the right, suggesting poor binocular coordination. For both vergence and saccades, the peak velocity and time to peak velocity was higher and earlier, respectively, in non-dyslexics compared to dyslexics; yet the average velocity of both movements was lower in dyslexics. Thus, these results indicate peculiar velocity profiles in dyslexics, particularly a slow deceleration phase in both vergence and saccades. The study provides an objective method to diagnose vergence and saccade abnormalities while viewing targets in the real three-dimensional space in a dyslexic population. Vergence abnormalities are demonstrated to be a problem in dyslexics, occurring independently from reading. We hypothesize these disconjugate drifts following saccades are the result of slow vergence capacity. Rehabilitation programs, such as those using REMOBI, should aim to target these deficits in vergence velocity, as this has been shown to improve binocular control.

Abstract Therapeutic targeting of tumor cells with drug nanocarriers relies upon successful interaction with membranes and efficient cell internalization. A further consideration is that engineered nanomaterials should not damage healthy tissues upon contact. A critical factor in this process is the external coating of drug delivery nanodevices. Using in silico, in vitro and in vivo studies, we show for the first time that magnetic nanoparticles coated with polyarabic acid have superior imaging, therapeutic, and biocompatibility properties. We demonstrate that polyarabic acid coating allows for efficient penetration of cell membranes and internalization into breast cancer cells. Polyarabic acid also allows reversible loading of the chemotherapeutic drug Doxorubicin, which upon release suppresses tumor growth in vivo in a mouse model of breast cancer. Furthermore, these nanomaterials provide in vivo contrasting properties, which directly compare with commercial gadolinium-based contrasting agents. Finally, we report excellent biocompatibility, as these nanomaterial cause minimal, if any cytotoxicity in vitro and in vivo. We thus propose that magnetic nanodevices coated with polyarabic acid offer a new avenue for theranostics efforts as efficient drug carriers, while providing excellent contrasting properties due to their ferrous magnetic core, which can help the future design of nanomaterials for cancer imaging and therapy.
