Background: Hemodialysis solutions are liquid concentrates used during hemodialysis sessions. They are commonly used as a renal replacement therapy., Objective: This study aimed to develop a method to determine Na, K, Ca, and Mg in hemodialysis solutions by high-resolution continuum source flame atomic absorption spectrometry (HR-CS F AAS) using Design of Experiments (DOE)., Methods: The combination of the Doehlert Matrix with the desirability function was used to establish a compromise condition in the optimization of the significant variables studied: 60 L/h for acetylene flow rate, 8 mm for burner height, and 0.25% (w/v) and 0.60% (w/v) for Cs and La concentrations. The method was validated by following the International Council for Harmonisation of Technical Requirements for Pharmaceuticals for Human Use (ICH) guidelines and parameters, and, as such, the selectivity, linearity, precision, accuracy, and robustness were evaluated., Results: Calibration curves with the ranges of 25-130 mg/L for Na, 1.0-5.0 mg/L for K and Ca, and 0.30-0.70 mg/L for Mg were employed. The method proved to be precise (RSD lower than 2.7%) and accurate (mean recovery data, contemplating the three levels added, were 103.0% for Na, 100.5% for K, 101.3% for Ca, and 101.5% for Mg)., Conclusions: The developed method enables the sequential multielement determination of Na, K, Ca, and Mg in a single run. Background and Aims: Soil endemics have long fascinated botanists owing to the insights they can provide about plant ecology and evolution. Often, these species have unique foliar nutrient composition patterns that reflect potential physiological adaptations to these harsh soil types. However, understanding global nutritional patterns to unique soil types can be complicated by the influence of recent and ancient evolutionary events. Our goal was to understand whether plant specialization to unique soils is a stronger determinant of nutrient composition of plants than climate or evolutionary constraints., Methods: We worked on gypsum soils. We analysed whole-plant nutrient composition (leaves, stems, coarse roots and fine roots) of 36 native species of gypsophilous lineages from the Chihuahuan Desert (North America) and the Iberian Peninsula (Europe) regions, including widely distributed gypsum endemics, as specialists, and narrowly distributed endemics and non-endemics, as non-specialists. We evaluated the impact of evolutionary events and soil composition on the whole-plant composition, comparing the three categories of gypsum plants., Key Results: Our findings reveal nutritional convergence of widely distributed gypsum endemics. These taxa displayed higher foliar sulphur and higher whole-plant magnesium than their non-endemic relatives, irrespective of geographical location or phylogenetic history. Sulphur and magnesium concentrations were mainly explained by non-phylogenetic variation among species related to gypsum specialization. Other nutrient concentrations were determined by more ancient evolutionary events. For example, Caryophyllales usually displayed high foliar calcium, whereas Poaceae did not. In contrast, plant concentrations of phosphorus were mainly explained by species-specific physiology not related to gypsum specialization or evolutionary constraints., Conclusions: Plant specialization to a unique soil can strongly influence plant nutritional strategies, as we described for gypsophilous lineages. Taking a whole-plant perspective (all organs) within a phylogenetic framework has enabled us to gain a better understanding of plant adaptation to unique soils when studying taxa from distinct regions., (© The Author(s) 2024. Kidney stones, as typical biominerals produced within the human body, pose a significant threat to human health, affecting over 12% of the global population. However, the exact mechanisms underlying their formation are not fully understood. Recent metal isotopic analysis provides a new way to study the roles of metal cations in biological processes within organisms. Here, we report the Mg isotope ratios of human kidney stones for the first time. The total range of measured values for δ26Mg in kidney stones is 1.05‰, from -1.12‰ to -0.07‰. Our data exhibit a significant 24Mg enrichment compared with the values calculated from density functional theory. We suggest that the Mg-isotopic fractionations in vivo are linked to active Mg transport mediated by proteins during intestinal absorption and preferential renal reabsorption of ionized Mg2+ via tight junctional proteins. Our results indicate that the inhibitory effect of Mg on kidney stones is related to the kink-blocking mechanism, and the incorporation of hydrated Mg lessens the extent of inhibition and the magnitude of isotope discrimination. Background and Aims: The duckweeds (Lemnaceae) consist of 36 species exhibiting impressive phenotypic variation, including the progressive evolutionary loss of a fundamental plant organ, the root. Loss of roots and reduction of vascular tissues in recently derived taxa occur in concert with genome expansions of ≤14-fold. Given the paired loss of roots and reduction in structural complexity in derived taxa, we focus on the evolution of the ionome (whole-plant elemental contents) in the context of these fundamental changes in body plan. We expect that progressive vestigiality and eventual loss of roots might have both adaptive and maladaptive consequences that are hitherto unknown., Methods: We quantified the ionomes of 34 accessions in 21 species across all duckweed genera, spanning 70 Myr in this rapidly cycling plant (doubling times are as rapid as ~24 h). We related both micro- and macroevolutionary ionome contrasts to body plan remodelling and showed nimble microevolutionary shifts in elemental accumulation and exclusion in novel accessions., Key Results: We observed a robust directional trend in calcium and magnesium levels, decreasing from the ancestral representative Spirodela genus towards the derived rootless Wolffia, with the latter also accumulating cadmium. We also identified abundant within-species variation and hyperaccumulators of specific elements, with this extensive variation at the fine (as opposed to broad) scale., Conclusions: These data underscore the impact of root loss and reveal the very fine scale of microevolutionary variation in hyperaccumulation and exclusion of a wide range of elements. Background: Low and high concentrations of plasma magnesium are associated with increased risk of future all-cause dementia; however, the underlying reasons remain elusive. The magnesium ion is an important electrolyte serving as a cofactor in many enzymatic processes in the human organism. Magnesium affects both neuronal and vascular functions. We investigated the associations of plasma concentrations of magnesium associate with common subtypes of dementia as Alzheimer dementia and non-Alzheimer dementia, and potential pathways by which magnesium may affect risk of dementia., Methods: Plasma concentrations of magnesium were measured in 102 648 individuals from the Copenhagen General Population Study. Cox regression and natural effects mediation analyses evaluated associations with either Alzheimer dementia or non-Alzheimer dementia., Results: Multifactorially adjusted hazard ratios for non-Alzheimer dementia were 1.50(95% confidence interval (CI):1.21-1.87) for the lowest and 1.34(1.07-1.69) for the highest vs the fourth quintile (reference) of plasma magnesium concentrations. Diabetes, cumulated smoking, stroke, and systolic blood pressure mediated 10.4%(3.1-22.8%), 6.8%(1.2-14.0%), 1.3%(0.1-3.6%), and 1.0%(0.2-2.6%), respectively, in the lowest quintile, whereas stroke mediated 3.2%(0.4-11.9%) in the highest quintile. No associations were observed for Alzheimer dementia., Conclusions: Low and high plasma magnesium concentrations were associated with high risk of vascular-related non-Alzheimer dementia, with the lowest risk observed at a concentration of 2.07 mg/dL (0.85 mmol/L). No association was observed for Alzheimer dementia. Mediation analysis suggested that diabetes may be in the causal pathway between low plasma magnesium concentrations and high risk of non-Alzheimer dementia, while cumulated smoking, stroke, and systolic blood pressure played minor mediating roles., (© American Association for Clinical Chemistry 2021. In this work, the initial results of the type testing of the LiF:Mg,Cu,P (TLD-100H) whole-body personal dosemeters are presented. An assessment of reproducibility, linearity of the response, the residual signal as a function of the dose, energy and angular dependence of the response was performed. In general, the dosemeters show good reproducibility for different dose values and a linear behaviour for a range between 0.1 and 300 mSv. The detection limits obtained are lower than 50 μSv. Riboswitches are cis-acting regulatory RNA biosensors that rival the efficiency of those found in proteins. At the heart of their regulatory function is the formation of a highly specific aptamer-ligand complex. Understanding how these RNAs recognize the ligand to regulate gene expression at physiological concentrations of Mg2+ ions and ligand is critical given their broad impact on bacterial gene expression and their potential as antibiotic targets. In this work, we used single-molecule FRET and biochemical techniques to demonstrate that Mg2+ ions act as fine-tuning elements of the amino acid-sensing lysC aptamer's ligand-free structure in the mesophile Bacillus subtilis. Mg2+ interactions with the aptamer produce encounter complexes with strikingly different sensitivities to the ligand in different, yet equally accessible, physiological ionic conditions. Our results demonstrate that the aptamer adapts its structure and folding landscape on a Mg2+-tunable scale to efficiently respond to changes in intracellular lysine of more than two orders of magnitude. The remarkable tunability of the lysC aptamer by sub-millimolar variations in the physiological concentration of Mg2+ ions suggests that some single-aptamer riboswitches have exploited the coupling of cellular levels of ligand and divalent metal ions to tightly control gene expression., (© The Author(s) 2019. Many agrowastes are being used for energy production by combustion in power plants. This process generates huge amounts of ash, which has a potential pozzolanic activity for blending with Portland cement or hydrated lime. In this paper, the ash obtained from elephant grass (Pennisetum purpureum Schum var. purple) leaves (EGLs) was studied, including the silicon content and its distribution, the presence of other compounds, and in addition, the presence of silica bodies (phytoliths). Combustion temperatures of 450 and 650°C produced an unaltered inorganic skeleton (spodogram), whereas at 850°C, there is a sintering process because of high potassium content in the ash. Phytoliths and different types of hairs were identified, and they contained high percentages of silica. Magnesium (mainly as periclase) was distributed in the most porous parts in the interior of the leaves. The silica can react with calcium hydroxide (pozzolanic reaction) forming calcium silicate hydrates (observed by field-emission scanning electron microscopy and thermogravimetric analysis). Fixed lime percentages at 28 curing days (63%) indicated the high reactivity of EGL ashes in calcium hydroxide pastes due to the pozzolanic reaction. This study demonstrates the possibility of the reuse of ashes from EGLs for the production of environmental-friendly cements.

Excessive expansion of Staphylococcus aureus is associated with several skin diseases, including atopic dermatitis (AD). Recently, we have demonstrated that washing skins with ultra-pure soft water containing little bivalent metal ions improved skin conditions of atopic subjects. In this study, we investigated the roles of calcium or magnesium on the proliferation of S. aureus both in vitro and in vivo. Depletion of calcium and magnesium in the culture medium significantly suppressed the expansion of S. aureus growth. When S. aureus, diluted with water containing calcium/magnesium at the concentration of medium-hard water (83·0 mg l -1 as CaCO 3 ) or the one that contains little calcium/magnesium, was applied onto the tape-stripped skin of Hos:HR-1 mice, growth of S. aureus in water without those minerals on the skin was suppressed. These results suggest that depletion of both calcium and magnesium abrogate the proliferation of S. aureus not only in the culture system but also on the skin surface of mice. Since colonization of S. aureus on the skin is well-known to exacerbate AD symptoms, usage of ultra-pure soft water containing less calcium and magnesium may improve the skin condition through the suppression of S. aureus growth on the skin of patients with skin problems., Significance and Impact of the Study: This study demonstrates the importance of calcium and magnesium for the colonization and growth of Staphylococcus aureus by using both in vitro culture systems and in vivo experiments on the murine skin. Our results indicate that the removal of these metal ions is probably beneficial for protecting the skin from S. aureus. Thus, using ultra-pure soft water without metal ions may improve the skin condition of patients with skin problems through the protection from S. aureus colonization., (© 2018 The Society for Applied Microbiology.)

Background: Linear relationships are commonly observed between shoot magnesium ([Mg]shoot) and shoot calcium ([Ca]shoot) concentrations among angiosperm species growing in the same environment., Scope and Conclusions: This article argues that, in plants that do not exhibit 'luxury' accumulation of Mg or Ca, (1) distinct stoichiometric relationships between [Mg]shoot and [Ca]shoot are exhibited by at least three groups of angiosperm species, namely commelinid monocots, eudicots excluding Caryophyllales, and Caryophyllales species; (2) these relationships are determined by cell wall chemistry and the Mg/Ca mass quotients in their cell walls; (3) differences between species in [Mg]shoot and [Ca]shoot within each group are associated with differences in the cation exchange capacity (CEC) of the cell walls of different species; and (4) Caryophyllales constitutively accumulate more Mg in their vacuoles than other angiosperm species when grown without a supra-sufficient Mg supply.

The negative health effects of inadequate magnesium intake are well established, but the extent of the problem of deficiency warrants further exploration. This review explores the dietary factors, such as changes in agricultural practices and dietary patterns, that affect magnesium consumption over time and examines the current adequacy of magnesium intake among adults in the United States. Large, cross-sectional, population-based data sets confirm over half the adult population in the United States does not consume adequate amounts of magnesium, although recent population-based studies show a steady and consistent recovery in magnesium consumption over the last several decades. Because there is no simple, rapid, accurate test to determine whole-body magnesium status, continued monitoring of magnesium consumption is essential to determine whether the trend of increasing magnesium consumption will continue. In the meantime, since the clinical consequences of inadequate magnesium status are well established, there are few reasons not to encourage increased magnesium intake in adults, especially since magnesium is found in healthy foods that should be consumed more often and there are no reported cases of hypermagnesemia from food alone.

To mitigate the effects of heat and drought stress, a better understanding of the genetic control of physiological responses to these environmental conditions is needed. To this end, we evaluated an upland cotton ( Gossypium hirsutum L.) mapping population under water-limited and well-watered conditions in a hot, arid environment. The elemental concentrations (ionome) of seed samples from the population were profiled in addition to those of soil samples taken from throughout the field site to better model environmental variation. The elements profiled in seeds exhibited moderate to high heritabilities, as well as strong phenotypic and genotypic correlations between elements that were not altered by the imposed irrigation regimes. Quantitative trait loci (QTL) mapping results from a Bayesian classification method identified multiple genomic regions where QTL for individual elements colocalized, suggesting that genetic control of the ionome is highly interrelated. To more fully explore this genetic architecture, multivariate QTL mapping was implemented among groups of biochemically related elements. This analysis revealed both additional and pleiotropic QTL responsible for coordinated control of phenotypic variation for elemental accumulation. Machine learning algorithms that utilized only ionomic data predicted the irrigation regime under which genotypes were evaluated with very high accuracy. Wide and common applications of ionising radiation require continuous improvement of radiation safety and dosimetry methods. The thermoluminescent (TL) method is well known and very popular. Apart from its advantages, it also carries certain disadvantages. The erasure of the TL signal on detector readout and the resulting impossibility of post-readout dose reassessment is one of them. At the Institute of Nuclear Physics, Polish Academy of Sciences (IFJ PAN), a method for dose reassessment based on phototransferred thermoluminescence (PTTL) has been developed. This method has been applied for dose reassessment to MTS-N (LiF:Mg, Ti) detectors used in individual whole-body dosemeters and adjusted to an automatic reader used in routine measurements. The next step was to extend and adapt this method for extremity dosemeters. With the use of the PTTL method, it is possible to reassess relatively high doses measured on individual whole-body and extremity dosemeters collected from our customers after routine use. The influence of PTTL background has greater impact in extremity dosemeters where it is only one and thinner MTS-N detector compared with four MTS-N detectors in whole-body dosemeters. The minimum dose was determined as 5 mSv for extremity dosimetry. Below 5 mSv, the impact of PTTL background is comparable to the signal, and the estimated uncertainty of reassessed dose is at the level of reassessed dose., (© The Author 2016. Thermoluminescence dosemeters are widely used in individual and environmental monitoring. The aim of this work was to compare the thermal stability of dosemeters of the Ext-Rad and whole-body card types with LiF:Mg,Ti and LiF:Mg,Cu,P detectors stored at different temperatures and periods. The dosemeters were stored at 0°C, room temperature and 40°C for periods that lasted 8, 30, 45, 90 and 120 d. In general, TLD-100H detectors present higher TL signal stability than TLD-100 detectors. The intensity of the signal remained constant for both materials for storage periods at 0°C. At RT the same results was observed for TLD-100H. For TLD-100 detectors, a maximum variation of 22 % was registered for the longest period. At 40°C the TL signal decreased with storage time for both detectors. The TL signal of TLD-100H detectors presented maximum variations of 12 % whereas for TLD-100 detectors, larger variations of 25 % were observed., (© The Author 2015. In this research, the effect of magnesium (Mg), copper (Cu) and phosphorus (P) impurities on dosimetry response of LiF:Mg,Cu,P phosphors is studied experimentally and by the simulation procedure. In the experimental procedure, LiF:Mg,Cu,P phosphors in the powder form were synthesised by chemical co-precipitation method. After annealing at 250°C for 10 min, known amounts of powder were exposed to gamma doses from 0.2 to 1 Gy. The activation energy of the electronic traps for the dosimetric peak at 150°C in LiF:Mg,Cu,P crystalline lattice obtained was 0.69 eV. In the simulation study, the role of stated dopants on electronic and structural properties of LiF crystalline lattice is investigated with the WIEN2 K Code. The activation energies of the electronic and hole traps for the dosimetric peak at the same temperature in LiF:Mg,Cu,P crystalline lattice obtained are 0.75 and 3.1 eV, respectively. Application of whole-body personal TL dosemeters based on a high-sensitivity LiF:Mg,Cu,P (MCP-N) in mixed field beta-gamma radiation has been characterised. The measurements were carried out with (90)Sr/(90)Y, (85)Kr and (137)Cs point sources to calculate the energy response and linearity of the TLD response in a dose range of 0.1-30 mSv. From the result, calibration curves were obtained, enabling the readout of individual dose equivalent Hp(10) from gamma radiation and Hp(0.07) from beta radiation in mixed field beta-gamma. Outdoor gamma radiation measurements in Cluj County, Romania have been performed using solid-state thermoluminescent detectors in order to develop a high-resolution database for natural gamma dose rates. Integrated measurements have been carried out for an exposure time of minimum 3 weeks. According to European Union requirements, the territory has been divided into 69 grids of 10 × 10 km. The cells were monitored using LiF:Mg,Cu,P detectors. For two locations the results were 136 ± 7 and 150 ± 7 nGy h(-1), respectively. These results can be explained by the existent geological substrate. The values ranged from 56 ± 4 to 150 ± 7 nGy h(-1), with an average value of 91 ± 2 nGy h(-1), being in agreement with the 2008 United Nations Scientific Committee on the Effects of Atomic Radiation (UNSCEAR) Report. A single-laboratory validation study was conducted for the simultaneous determination of arsenic, cadmium, calcium, cobalt, copper, chromium, iron, lead, magnesium, manganese, molybdenum, nickel, selenium, and zinc in all major types of commercial fertilizer products by microwave digestion and inductively coupled plasma-optical emission spectroscopy analysis. This validation study proposes an extension and modification of AOAC 2006.03. The extension is the inclusion of calcium, copper, iron, magnesium, manganese, and zinc, and the modification is incorporation of hydrochloric acid in the digestion system. This dual acid digestion utilizes both hydrochloric and nitric acids in a 3 to 9 mL volume ratio/100 mL. In addition to 15 of the 30 original validation materials used in the 2006.03 collaborative study, National Institute of Standards and Technology Standard Reference Material 695 and Magruder 2009-06 were incorporated as accuracy materials. The main benefits of this proposed method are a significant increase in laboratory efficiency when compared to the use of both AOAC Methods 965.09 and 2006.03 to achieve the same objective and an enhanced recovery of several metals.

Volatile cues released by plants play an important role in plant-insect interactions and are influenced by pests or soil conditions affecting plant metabolism. Field microcosm experiments were used to characterize arthropod spontaneous assemblies in homogenous unstressed wheat patches exposed to volatile cues coming from wheat plants with different levels of stress. The design was a factorial completely randomized block design with three replications. Source wheat pots combined two stress factors: 1) soil degradation level: high and low, and 2) aphid herbivory: with (A) and without (B). Eighteen experimental units consisted of source stressed wheat pots, connected by tubes conducting the volatile cues to sink wheat patches. These patches were located at the end of the tubes placed in a flowering wheat field. Arthropod assemblies on wheat sinks were different between years and they were associated to the source cues. Soil condition was the main discriminating factor among arthropods when a clear contrast between high and low soil degradation was observed, whereas aphid herbivory was the main discriminating factor when soil condition effects were absent. Main soil properties related with arthropods assembly were Mg and K in the first year and cation exchange capacity, total nitrogen, and pH in the second year of experiment. According to this study, spontaneous arthropod distributions in the homogeneous, unstressed wheat patch responded to the volatile cues coming from wheat sources growing in particular soil conditions. It is possible to suggest that soil-plant-herbivore interactions change wheat cues and this phenomenon produces significant differences in neighboring arthropod community structure.

Germination and seedling establishment are among the most critical phases in the development of plants, and seed vigour has become an important trait for the selection of robust crop cultivars. Little is known about the potentially limiting role of mineral nutrients in early metabolic and developmental processes during germination. Therefore, we assessed the ionome and relative distribution of mineral elements in different seed and seedling tissues of oilseed rape (Brassica napus L.) and monitored the internal allocation of nutrients during germination. In seeds, cotyledons harboured the main pool of K, P, S, Mg, Fe, Mn and Zn, whereas the seed coat contained most of the Ca, Na, B, Cu and Mo. Although the early root and hypocotyl tissue expanded first, concentrations of most elements were initially low. Re-allocation of elements to the root/hypocotyl tissue from other pools set in two days after seed imbibition and was most rapid for K. Relative to the critical deficiency levels of vegetative tissues, seed tissues were particularly low in B, K and Fe. Further analyses of the ionome of seeds and seedlings, grouped according to their germination efficiency, indicated that in particular low S, Mg and Ca coincided with germination failure. This study documents highly dynamic changes in the ionome of seed and seedling tissues and provides evidence for potentially limiting elements during early germination and seedling establishment in rapeseed.

The beneficial effects of dietary glucosylceramide on the barrier function of the skin have been increasingly reported, but the entire mechanism has not been clarified. By DNA microarray, we investigated changes in gene expression in hairless mouse skin when a damage-inducing AD diet and a glucosylceramide diet (GluCer) were imposed. GluCer administration potentially suppressed the upregulation of six genes and the downregulation of four genes in the AD group. Examination of the epidermal and/or dermal expression of Npr3, Cyp17a1, Col1a1, S100a9, Sprr2f, Apol7a, Tppp, and Scd3 revealed responses of various parts of the skin to the diets. In normal hairless mice, GluCer administration induced an increase in the dermal expression of Cyp17a1 and the epidermal expression of Tppp, and a decrease in the epidermal expression of S100a9. Our results provide information on gene expression not only in whole skin but also in the epidermis and dermis that should prove useful in the search for the mechanisms underlying the effects of GluCer on damaged and normal skin.

Intra-annual nutrient (nitrogen, phosphorus, potassium, calcium and magnesium) flux was quantified for Pinus taeda L. at a nutrient-poor, well-drained sandy site in Scotland County, NC, USA where a 2 × 2 factorial of irrigation and nutrition was applied in four replications in a 10-year-old stand with 1200 stems ha(-1). Treatments were applied with the goal of providing optimum nutrition (no nutritional deficiencies) and water availability. Component (foliage, branch, stem and root) nutrient content was estimated monthly for 2 years using nutrient concentration and phenology assessments combined with destructive harvests. Positive flux values indicated nutrient accumulation in the trees while negative values indicated nutrient loss from the trees. Fertilization significantly increased nitrogen, phosphorus, potassium, calcium and magnesium flux 140%, on average, over non-fertilized. Irrigation significantly increased calcium flux 28% while there was no significant irrigation effect on nitrogen, phosphorus, potassium or magnesium. Maximum nutrient fluxes (kg ha(-1) day(-1)) for non-fertilized and fertilized stands were 0.36 and 1.05 for nitrogen, 0.042 and 0.095 for phosphorus, 0.13 and 0.51 for potassium, 0.27 and 0.42 for calcium, and 0.04 and 0.12 for magnesium, respectively. Maximum flux was coincident with ephemeral tissue (foliage and fine root) development and likely would be higher in stands with more foliage than those observed in this study (projected leaf area indices were 1.5 and 3.0 for the non-fertilized and fertilized stands). Minimum nutrient fluxes (kg ha(-1) day(-1)) for non-fertilized and fertilized stands were -0.18 and -0.42 for nitrogen, -0.029 and -0.070 for phosphorus, -0.05 and -0.18 for potassium, -0.04 and -0.05 for calcium, and -0.02 and -0.03 for magnesium, respectively. Minimum fluxes were typically observed in the dormant season and were linked to foliage senescence and branch death. Foliage and branch component nutrient contents were out of phase for nitrogen, phosphorus, potassium and magnesium, indicating nutrient retranslocation and storage in branches prior to foliage development and after foliage senescence. In contrast to current operational fertilizer programs which often target winter application these data suggest the best application times would be during foliage development.

International normalized ratio (INR) discrepancies were noted between clinical laboratories using various prothrombin time (PT) systems. We studied the influence of different commercial blood collection tubes and different PT systems on INR measurements. INRs of fresh patient samples were determined by 3 laboratories, each using different PT systems. In the first part of the study, samples were drawn with Vacutainer tubes and in the second part with Monovette tubes. In the first part of the study, the maximum bias for all patients amounted to 0.46 INR (14%), and in the second part, to 0.14 INR (4.9%). The maximum bias for all patients could be reduced further by local system calibration using frozen pooled plasma specimens. The sodium citrate solutions in the blood collection tubes were contaminated with magnesium ions (approximately 2.7 mmol/L and 0.3 mmol/L in the Vacutainer and Monovette, respectively). INR discrepancies could be explained largely by this influence of blood collection tubes. The maximum allowable magnesium contamination in sodium citrate anticoagulant solutions should be less than 1 mmol/L.

Production of ATP by the glycolytic pathway in the mammalian pathogenic stage of protists from the genus Trypanosoma is required for the survival of the parasites. Cofactor-independent phosphoglycerate mutase (iPGAM) is particularly attractive as a drug target because it shows no similarity to the corresponding enzyme in humans, and has also been genetically validated as a target by RNAi experiments. It has previously been shown that trypanosomatid iPGAMs require Co(2+) to reach maximal activity, but the biologically relevant metal has remained unclear. In this paper the metal content in the cytosol of procyclic and bloodstream-form T. brucei (analysed by inductively coupled plasma-optical emission spectroscopy) shows that Mg(2+), Zn(2+) and Fe(2+) were the most abundant, whereas Co(2+) was below the limit of detection (<0.035 μM). The low concentration indicates that Co(2+) is unlikely to be the biologically relevant metal, but that instead, Mg(2+) and/or Zn(2+) may assume this role. Results from metal analysis of purified Leishmania mexicana iPGAM by inductively coupled plasma-mass spectrometry also show high concentrations of Mg(2+) and Zn(2+), and are consistent with this proposal. Our data suggest that in vivo cellular conditions lacking Co(2+) are unable to support the maximal activity of iPGAM, but instead maintain its activity at a relatively low level by using Mg(2+) and/or Zn(2+). The physiological significance of these observations is being pursued by structural, biochemical and biophysical studies.

Aims: To examine the mechanism of ozone-induced damage to cytoplasmic membrane and cell ultrastructure of Pseudomonas aeruginosa ATCC27853., Methods and Results: Cell suspensions of Ps. aeruginosa ATCC27853 were treated with ozonated water. The leakages of cellular potassium (K⁺), magnesium (Mg²⁺) and adenosine triphosphate (ATP), determined by inductively coupled plasma/mass spectrometry (ICP/MS) and a commercial bioluminescence assay kit, were to assess ozone-induced damage to the cytoplasmic membrane. Maximum leakages of K⁺ and Mg²⁺ were attained, respectively, at 0·53 mg l⁻¹ ozone after 0·5 and 2 min with > 99% inactivation of culturable bacteria, while that of ATP was achieved at 0·67 mg l⁻¹ ozone after 1 min. Transmission electron microscopy (TEM) and scanning electron microscopy (SEM) revealed that treated cells retained intact shapes and cytoplasm agglutinations and vacuoles occurred., Conclusions: Ozone inactivates Ps. aeruginosa ATCC27853 by the combined results of increased cytoplasmic membrane permeability and cytoplasm coagulation, rather than by severe membrane disruption and cell lysis., Significance and Impact of the Study: Pseudomonas aeruginosa is a common water-related pathogen. In this paper, the results aimed at assessing the performance of two varieties of LiF detectors (LiF:Mg,Ti and LiF:Mg,Cu,P) in photon fields relatively to reproducibility, detection threshold and angular dependence as defined in the ISO 12794 standard are presented. The fading properties and the limit of detection were also investigated for both materials. The results suggest that both LiF varieties are well suited for extremity monitoring. However, better fading properties of LiF:Mg,Cu,P when compared with LiF:Mg,Ti, combined with previous results relatively to energy dependence suggests that LiF:Mg,Cu,P dosemeters are better suited for extremity monitoring.

LiF is a well-known thermoluminescent (TL) material used in individual monitoring, and its fading characteristics have been studied for years. In the present study, the fading characteristics (for a period of 150 d) of various commercial LiF materials with different dopants have been evaluated. The materials used in the study are those used in routine procedures by the Personal Dosimetry Department of Greek Atomic Energy Commission and in particular, LiF:Mg,Ti (MTS-N, TL Poland), LiF:Mg,Cu,P (MCP-N, TL Poland), LiF:Mg,Cu,P (MCP-Ns, thin active layer detector, TL Poland) and LiF:Mg,Cu,P (TLD100H, Harshaw). The study showed that there is a sensitivity loss in signal of up to 20 % for the MTS-N material for a 150-d period in the pre-irradiation fading phase. The MCP-N has a stable behaviour in the pre-irradiation fading phase, but this also depends on the readout system. As far as the post-irradiation fading effect is concerned, a decrease of up to 20 % for the MTS-N material is observed for the same time period. On the other hand, the LiF:Mg,Cu,P material presents a stable behaviour within ± 5 %. These results show that the fading effect is different for each material and should be taken into account when estimating doses from dosemeters that are in use for >2 months.

Recent epidemiological studies suggest a rather low-dose threshold (<0.5 Gy) for the induction of a cataract of the eye lens. Some other studies even assume that there is no threshold at all. Therefore, protection measures have to be optimised and current dose limits for the eye lens may be reduced in the future. ICRP Publication 103 on H(p)(d), in §(136), reads that '… a depth d = 3 mm has been proposed for the rare case of monitoring the dose to the lens of the eye. In practice, however, H(p)(3) has rarely been monitored and H(p)(0.07) can be used for the same monitoring purpose… '. As recommended on the EU 'Technical recommendations for monitoring individuals occupationally exposed to external radiation', a test on the ENEA TL extremity dosemeter is herein reported. The results within the actual EU founded Optimization of RAdiation protection for MEDical staff (ORAMED) Project, whose WP2 is aimed at the quantity H(p)(3) and eye lens dosimetry in practice, are taken into account. The paper summarises the main aspects of the study carried out at ENEA-Radiation Protection Institute (Bologna, Italy) to provide practical solutions (in the use and the design) to evaluate the response of the ENEA TL extremity dosemeter in terms of H(p)(3).

The sensitivity of Harshaw™ magnesium/copper/phosphorus-doped lithium fluoride is shown to be stable over long time periods, even when, as is often the case in practical operations, the TLDs are read out only three times a year.

Amongst the dosemeters offered by the Health Protection Agency (HPA) Personal Dosimetry Service are finger rings, which use the Harshaw™ DXTRAD element. This paper describes restricted-type testing carried out by HPA on the previously untried combination of the LiF:Mg,Cu,P material and a thicker filter (42 mg cm(-2)). The tests were based on ISO 12794 [International Organization for Standardization. ISO 12794:2000 nuclear energy--radiation protection--individual thermoluminescence dosemeters for the extremities and eyes. ISO (2000)] and included energy and angle dependence of response for photons. The conclusion was, for photon dosimetry applications the thicker filter is acceptable for use in the HPA service.

The phototransferred thermoluminescence (PTTL) technique is applied to the Harshaw LiF:Mg,Cu,P material. It is demonstrated that using 254-nm UV light, dose levels as low as 0.2 mGy can be re-estimated. The PTTL efficiency was found to be ∼ 6 % in the dose range of 0.2 mGy-1 Gy, and it appears to be dose-independent. This implies that a simple calibration factor could be applied to the PTTL data for the re-estimation of dose levels. It was demonstrated that with a proper choice of the TL readout parameters, and the UV-light irradiation conditions, dose levels that are relevant to personal or environmental dosimetry can be re-estimated.

On the basis of the newly discovered behaviour of LiF:Mg,Cu,P detectors at high and ultra-high doses, a new method of thermoluminescence (TL) measurement of radiation doses ranging from micrograys up to a megagray, has been recently developed at the Institute of Nuclear Physics (IFJ). The method is based on the relationship between the TL signal, integrated in the given temperature range and dose. It is quantified by a parameter called the 'ultra-high temperature ratio'. It has been demonstrated that this new method can measure radiation doses in the range of about 1 µGy to 1 MGy, using a single LiF:Mg,Cu,P detector. This method was recently successfully blindly tested for 10 MeV electrons up to doses of 200 kGy. It can be used for dosimetry in high-energy accelerators, especially in the Large Hadron Collider at CERN, and has great potential for accident dosimetry in particular.

The photon energy response of different RADOS (Mirion Technologies) personal dosemeters with MTS-N (LiF:Mg,Ti) and MCP-N (LiF:Mg,Cu,P) thermoluminescence (TL) detectors was investigated. Three types of badges were applied. The irradiation with reference photon radiation qualities N (the narrow spectrum series), and S-Cs and S-Co nuclide radiation qualities, specified in ISO 4037 [International Organization for Standardization (ISO). X and gamma reference radiations for calibrating dosemeters and doserate meters and for determining their response as a function of photon energy. ISO 4037. Part 1-4 (1999)], in the energy range of 16-1250 keV, were performed at the Dosimetry Laboratory Seibersdorf. The results demonstrated that a readout of a single MTS-N or MCP-N detector under the Al filter can be used to determine Hp(10) according to requirements of IEC 61066 [International Electrotechnical Commission (IEC). Thermoluminescence dosimetry systems for personal and environmental monitoring. International Standard IEC 61066 (2006)] for TL systems for personal dosimetry. The new RADOS badge with the experimental type of a holder (i.e. Cu/Al filters) is a very good tool for identifying the radiation quality (photon energy).

Unsealed beta-gamma-emitting sources are used (15 GBq (90)Y each session) in nuclear medicine therapeutic procedures. Inside the manipulation cell and while giving the injection to the patient, the skin exposure is very high; electron radiation field is not homogeneous and thus the exposure of the extremities is not uniform. Particular individual monitoring is adopted: single thermoluminescence dosemeter, wrapped in polyethylene film and placed on an adhesive tape, is positioned on the tip of the fingers; 6-10 dosemeters are assigned to each operator per session. The energy and angle response is studied for X-ray spectra, (90)Sr/Y and (204)Tl--a unique mean calibration factor is calculated in order to estimate H(p)(0.07). Performance of dosemeter is analysed according to ISO 62387-1(2007) and the combined uncertainty (calculated using the Monte Carlo method) results lie in the order of 11 %. This method reveals the critical step of manipulation and administration and ensures that dose limits are not exceeded.

The effects of UV-induced bleaching of deep traps on Harshaw thermoluminescent (TL) LiF:Mg,Cu,P and LiF:Mg,Ti materials were investigated. During a normal heating cycle, LiF:Mg,Cu,P is limited to a maximum temperature of 240 °C. LiF:Mg,Ti can be read to higher temperatures; however, encapsulation in polytetrafluoroethylene limits the maximum readout temperature to 300 °C. Generally, for both materials, these respective temperatures are sufficient for emptying traps corresponding to the main dosemetric peaks. However, when the dosemeters are subjected to a high dose level, such as 1 Gy (much higher than individual monitoring dose levels), higher temperature traps are filled that cannot be emptied without exceeding the above-mentioned maximum temperatures. These high temperature traps tend to be unstable during normal readout and can significantly increase the residual TL signal. The purpose of this study was to investigate the applicability of a UV-induced bleaching technique for emptying higher temperature traps following high-dose applications. In addition, in the case of LiF:Mg,Cu,P, where the maximum readout temperature is significantly lower, we investigated the possibility of reducing the residual signal using the application of repeated readout cycles. The optical bleaching approach was found to be effective in the case of LiF:Mg,Ti; however, for LiF:Mg,Cu,P, no reduction in the residual signal was observed. For this latter material, the application of repeatable readout cycles is very effective and residual signals equivalent to dose levels as low as 0.01 mGy were observed following an initial dose of 5 Gy. To the best of our knowledge, this work is the first attempt to apply an 'optical annealing' technique to the Harshaw thermoluminescent dosemeter (TLD) materials.

Background: Numerous studies have suggested an inverse relationship between drinking water hardness and cardiovascular disease. However, the weight of evidence is insufficient for the WHO to implement a health-based guideline for water hardness. This study followed WHO recommendations to assess the feasibility of using ecological time series data from areas exposed to step changes in water hardness to investigate this issue., Method: Monthly time series of cardiovascular mortality data, subdivided by age and sex, were systematically collected from areas reported to have undergone step changes in water hardness, calcium and magnesium in England and Wales between 1981 and 2005. Time series methods were used to investigate the effect of water hardness changes on mortality., Results: No evidence was found of an association between step changes in drinking water hardness or drinking water calcium and cardiovascular mortality. The lack of areas with large populations and a reasonable change in magnesium levels precludes a definitive conclusion about the impact of this cation. We use our results on the variability of the series to consider the data requirements (size of population, time of water hardness change) for such a study to have sufficient power. Only data from areas with large populations (>500,000) are likely to be able to detect a change of the size suggested by previous studies (rate ratio of 1.06)., Conclusion: Ecological time series studies of populations exposed to changes in drinking water hardness may not be able to provide conclusive evidence on the links between water hardness and cardiovascular mortality unless very large populations are studied. Investigations of individuals may be more informative.

Tobacco smoking constitutes a significant source of indoor air pollution. Various chemical compounds that are emitted during tobacco smoking can have a direct cytotoxic effect on spermatozoa by damaging DNA. There is some evidence that tobacco smoking in men could affect male fertility. The goals of this study were to find relationships between thiocyanates (as biomarkers of environmental tobacco smoke exposure) and other inorganic ions in human semen samples and present the effectiveness of the proposed sample preparation procedure combined with ion chromatography technique for the determination of inorganic ions, especially thiocyanates, in human semen samples collected from heavy, moderate, and passive smokers, as well as nonsmoking individuals.

This paper describes a validation process in compliance with the NFIEN ISO/IEC 17025 standard for the determination of the macrominerals calcium, magnesium, sodium, and potassium in foodstuffs by microsampling with flame atomic absorption spectrometry after closed-vessel microwave digestion. The French Standards Commission (Agence Francaise de Normalisation) standards NF V03-110, NF EN V03-115, and XP T-90-210 were used to evaluate this method. The method was validated in the context of an analysis of the 1322 food samples of the second French Total Diet Study (TDS). Several performance criteria (linearity, LOQ, specificity, trueness, precision under repeatability conditions, and intermediate precision reproducibility) were evaluated. Furthermore, the method was monitored by several internal quality controls. The LOQ values obtained (25, 5, 8.3, and 8.3 mg/kg for Ca, Mg, Na, and K, respectively) were in compliance with the needs of the TDS. The method provided accurate results as demonstrated by a repeatability CV (CVr) of < 7% and a reproducibility CV (CVR) of < 12% for all the elements. Therefore, the results indicated that this method could be used in the laboratory for the routine determination of these four elements in foodstuffs with acceptable analytical performance.

Aims: The ability to transform Vibrio spp. is limited by the extracellular nuclease that their cells secrete. The reported transformation efficiency of this organism is 10(2)-10(5) transformants per microgram DNA. We tried different buffers and conditions, aiming to elevate its transformation efficiency., Methods and Results: MgCl(2) and sucrose are often included in the washing and/or electroporation buffers to stabilize the cell membrane. However, Mg(2+) is required for production and activity of the extracellular nuclease. A simple electroporation buffer lacking Mg(2+) was found to increase transformation efficiency dramatically, to levels 50-fold more than the buffers containing Mg(2+). To maintain the stability of the cell membranes, Mg(2+) was replaced with high concentrations of sucrose, from 272 to 408 mmol l(-1). With the new buffers, the transformation efficiency of Vibrio parahaemolyticus was increased to 2.2 x 10(6) transformants per microgram DNA., Conclusions: Mg(2+) in the buffer adversely affected transformation of V. parahaemolyticus by electroporation. The cell membranes of vibrio can be stabilized by high concentration of sucrose when Mg(2+) is absent., Significance and Impact of the Study: A greater transformation efficiency can facilitate the genetic analysis of an organism and its pathogenicity. Buffers lacking Mg(2+) can be used for other nuclease-producing organisms.

Jaggery, brown sugar, white sugar, and molasses collected from the local sugarcane industry of Pakistan have been analyzed for essential elements in order to estimate their nutritional adequacy. Instrumental neutron activation analysis was used to determine Ca, CI, Co, Cr, Fe, K, Mg, Mn, Na, and Zn through sequential, short, medium, and long irradiation times. Maximum concentrations for most of these elements were determined in molasses, with lower concentrations determined in jaggery and brown sugar; white sugar contained trace amounts of all essential elements. Contributions to the weekly Recommended Dietary Allowance (RDA) values for the elements were estimated only for jaggery, brown sugar, and white sugar because molasses in Pakistan is not consumed as a dietary item. Jaggery contributes the highest percentages of Cr, Mg, Mn, and Zn, whereas the highest percentages of Cl, Fe, K, and Na can be acquired from brown sugar. The contribution of white sugar to the weekly RDAs for these elements is negligible, indicating that white sugar is a poor source of the essential elements. However, the introduction of molasses to the diet can contribute to an adequate intake of these elements.

Background and Aims: Plants from gypsum habitats are classified as gypsophiles and gypsovags. The former include both narrow endemics limited to small gypsum areas and regionally dominant gypsophiles growing in gypsum areas of large regions, whereas gypsovags are plants that can grow both in gypsum and non-gypsum soils. Factors controlling the distribution of gypsum plants are still not fully understood., Methods: To assess how the different types of gypsum plants deal with the stressful conditions of gypsum substrates, comparisons were made of the leaf chemical composition of four gypsovags, five regionally dominant gypsophiles and four narrow gypsum endemics growing in two massive gypsum areas of the Iberian Peninsula., Key Results: The chemical composition of gypsovags was clearly different from regionally dominant gypsophiles, while the chemical composition of narrow-gypsophile endemics was more similar to the chemical composition of gypsovags than to that of regionally dominant gypsophiles. Regionally dominant gypsophiles showed higher concentrations of ash, Ca, S, N, Mg P and Na, whereas gypsovags and local gypsophile endemics displayed higher concentrations of C and greater C : N ratios., Conclusions: Such differences suggest that the three groups of gypsum plants follow diverse ecological strategies. It is suggested that regionally dominant gypsophiles might fit the 'specialist' model, being species specifically adapted to gypsum, whereas both gypsovags and narrow-gypsophile endemics might fit the 'refuge' model, being stress-tolerant species that find refuge on gypsum soils from competition. The analysis of the leaf chemical composition could be a good predictor of the degree of plants specialization to gypsum soils.

Aims: This study was developed to purify and to characterize a keratinolytic protease from the bacterium Microbacterium sp. strain kr10., Methods and Results: Enzyme purification was carried out by sequential liquid chromatography on Sephadex G-100 and Q-Sepharose columns. The purification was about 255-fold, with a yield of 34%, as determined with azocasein as substrate. The molecular weight of the enzyme was estimated as 42,000 Da by SDS-PAGE. The enzyme had pH and temperature optima of 7.5 and 50 degrees C respectively. This keratinase was inhibited by EDTA and 1,10-phenanthroline, and analysis of metal content indicates that Zn(2+) and Mg(2+) are present. A 2(2) factorial design was developed to investigate the effect of keratinase and mercaptoacetate concentration on feather keratinolysis. Statistical analysis showed that both variables have a significant effect on hydrolysis of keratin., Conclusions: A new keratinase produced by Microbacterium sp. was purified and characterized., Significance and Impact of the Study: This keratinolytic enzyme offers an interesting potential for the hydrolysis of keratin wastes to be used as feed supplement or bioconversion to added-value products.

In KOH, the Mg(II)-bromopyrogallol red (BPR) complex produced a very sensitive polarographic wave at -1.30 V. The wave height was linear with the concentration of Mg(II) in the range of 0.05 to 2 microg/mL. The detection limit was 0.01 microg/mL. The electrochemical behavior of Mg(II)-BPR was studied by electrochemical and spectrophotometric methods. Experiments proved that the polarographic wave of Mg(II)-BPR was due to the reduction of BPR in the Mg(II)-BPR complex. The method, which was sensitive, selective, and simple to perform, was used to determine magnesium in foods, and the results were consistent with those obtained by atomic absorption spectroscopy.

Aims: To investigate the chemical characterization of the mucilage sheath produced by Chlorella sorokiniana., Methods and Results: Algal mucilage sheath was hydrolysed with NaOH, containing EDTA. The purity of the hydrolysed sheath was determined by an ATP assay. The composition of polysaccharide in the sheath was investigated by high-performance anion-exchange chromatography with pulsed amperometric detection. Sucrose, galacturonic acid, xylitol, inositol, ribose, mannose, arabinose, galactose, rhamnose and fructose were detected in the sheath as sugar components. Magnesium was detected in the sheath as a divalent cation using inductively coupled argon plasma. The sheath matrix also contained protein., Conclusions: It appears that the sheath is composed of sugars and metals. Mucilage sheath contains many kinds of saccharides that are produced as photosynthetic metabolites and divalent cations that are contained in the culture medium., Significance and Impact of the Study: This is the first report on chemical characterization of the sheath matrix produced by C. sorokiniana.

An energy dispersive x-ray microanalysis study was performed throughout the total length of cementum on five impacted human teeth. Mineral content of calcium, phosphorous, and magnesium were determined with an electron probe from the cemento-enamel junction to the root apex on the external surface of the cementum. The concentration profiles for calcium, phosphorous, and magnesium were compared by using Ca/P and Mg/Ca atomic percent ratio. Our findings demonstrated that the Ca/P ratio at the cemento-enamel junction showed the highest values (1.8-2.2). However, the area corresponding to the acellular extrinsic fiber cementum (AEFC) usually located on the coronal one-third of the root surface showed a Ca/P media value of 1.65. Nevertheless, on the area representing the fulcrum of the root there is an abrupt change in the Ca/P ratio, which decreases to 1.3. Our results revealed that Mg(2+) distribution throughout the length of human cementum reached its maximum Mg/Ca ratio value of 1.3-1.4 at.% around the fulcrum of the root and an average value of 0.03%. A remarkable finding was that the Mg/Ca ratio pattern distribution showed that in the region where the Ca/P ratio showed a decreasing tendency, the Mg/Ca ratio reached its maximum value, showing a negative correlation. In conclusion, this study has established that clear compositional differences exist between AEFC and cellular mixed stratified cementum varieties and adds new knowledge about Mg(2+) distribution and suggests its provocative role regulating human cementum metabolism.

The Mg2+ fluorescent dye mag-fura 2, entrapped in cells or organelles, has frequently been used for dual excitation ratio-metric determinations of free ionic Mg2+ concentrations in eukaryotic, mostly mammalian cells. Here we report its successful application to measure free Mg2+ concentrations ([Mg2+]i) in Salmonella enterica cells. When kept in nominally Mg2+ free buffer (resting conditions), the [Mg2+]i of wild-type cells has been determined to be 0.9 mM. An increase in the external Mg2+ concentration ([Mg2+]e) resulted in a rapid increase of [Mg2+]i, saturating within a few seconds at about 1.5 mM with [Mg2+]e of 20 mM. In contrast, cells lacking the Mg2+ transport proteins CorA, MgtA, MgtB failed to show this rapid increase. Instead, their [Mg2+]i increased steadily over extended periods of time and saturated at concentrations below those of wild-type cells. Mg2+ uptake rates increased more than 15-fold when corA was overexpressed in these mutant cells. Uptake of Mg2+ into corA expressing cells was strongly stimulated by nigericin, which increased the membrane potential DeltaPsi at the expense of DeltapH, and drastically reduced by valinomycin, which decreased the membrane potential DeltaPsi. These results reveal mag-fura 2 as a useful indicator to measure steady-state [Mg2+]i values in resting bacterial cells and to determine Mg2+ uptake rates. They confirm the role of CorA as the major Mg2+ transport protein and reveal the membrane potential as driving force for Mg2+ uptake into S. enterica cells.

Crystal structure of nano-scale precipitates in age-hardening aluminum alloys is a challenge to crystallography. The utility of selected area electron diffraction intensities from embedded precipitates is limited by double scattering via matrix reflections. This effect can be signally reduced by the precession technique, which we have used to collect extensive intensity data from the semicoherent, metastable eta-precipitate in the Al-Zn-Mg alloy system. A structure model in the space group P-62c is proposed from high-resolution microscopy and electron diffraction intensities. The advantages of using the precession technique for quantitative electron diffraction is discussed.

The distribution of minerals in quinoa (Chenopodium quinoa Willd.) seed was examined using energy dispersive X-ray microanalysis (EDX) in combination with scanning electron microscopy (SEM). Phosphorus, K, and Mg coincided in localization in embryonic tissue. Since phytin globoids have been known to localize in protein bodies in embryonic cells of quinoa seed, it is thought that P is attributed to phytic acid and that K and Mg form to phytate. Calcium and K were present in the pericarp, where the cell wall is thickly developed, suggesting that these minerals are associated with pectin. Sulfur occurred in embryonic tissues, which would be derived from sulfur amino acid residues of storage proteins concentrated in the tissues. Abrasion of quinoa seeds resulted particularly in decrease in Ca content.

The aim of this prospective controlled study was to ascertain the effect of hyperthyroidism on sperm quality and composition. We studied 23 thyrotoxic male patients, aged 43.8 +/- 2.4 yr (mean +/- SEM), and 15 healthy male controls of approximately the same age (42.2 +/- 2.2 yr). Two semen analyses at intervals of 2-3 wk were obtained before and about 5 months after euthyroidism was achieved either by methimazole alone (14 patients) or (131)I plus methimazole (9 patients). Total fructose, zinc (Zn), and magnesium (Mg) were also measured in seminal plasma in 16 patients, because 7 had semen volume less than 2 ml. In the control group semen analysis was performed only once. Mean (+/-SEM) semen volume was within normal range both in patients (3.3 +/- 0.2 ml) and controls (3.5 +/- 0.4 ml; P = NS). Mean sperm density was lower in patients, although the difference compared with controls did not reach statistical significance (35.7 +/- 5.3 vs. 51.5 +/- 6.1 x 10(6)/ml; P = 0.062). The same was found with sperm morphology (68 +/- 7% vs. 78 +/- 8%; P = NS). Finally, mean motility was lower in thyrotoxic males than in controls (28 +/- 8% vs. 57 +/- 7%; P < 0.01). After treatment, sperm density and motility improved [35.7 +/- 5.3 vs. 43.3 +/- 6.5 x 10(6)/ml (P = NS) and 28 +/- 8% vs. 45 +/- 7% (P < 0.05), respectively], but sperm morphology did not change (68 +/- 7% vs. 70 +/- 6%; P = NS). Mean values for fructose, Zn, and Mg did not differ between controls and patients either before or after achievement of euthyroidism [9.2 +/- 0.7, 3.0 +/- 0.5, and 4.2 +/- 0.7 nmol/liter vs. 8.6 +/- 0.9, 3.0 +/- 0.5, and 4.7 +/- 0.8 nmol/liter (patients before) and 9.1 +/- 0.7, 3.1 +/- 0.6, and 4.5 +/- 0.9 nmol/liter (patients after treatment) for fructose, Zn, and Mg, respectively]. Moreover, according to the treatment given, no statistically significant differences were found before or after treatment. Finally, seminal plasma fructose, Zn, and Mg levels did not correlate with sperm parameters or with pretreatment thyroid hormone levels. In conclusion, the results of our study indicate that male patients with hyperthyroidism have abnormalities in seminal parameters, mainly sperm motility. These abnormalities improve or normalize when the patients become euthyroid. Restoration of sperm parameters was independent of the treatment provided for the hyperthyroid syndrome. Moreover, seminal plasma elements, such as fructose, Zn, and Mg, did not correlate with sperm density, motility, or morphology.
