Macrosaccus morrisella (Fitch) (Lepidoptera: Gracillariidae) is a leaf-mining microlepidopteran native to North America recently reported feeding on soybean in the United States and Canada. Control methods for the management of this pest remain unknown. The insecticides Agri-Mek SC (abamectin) and Endigo ZXC (lambda-cyhalothrin + thiamethoxam) were evaluated for: (i) effects against immature stages of M. morrisella in potted soybean plants; (ii) effects against adults of M. morrisella through different routes of exposure; and (iii) efficacy of field applications against M. morrisella. In the experiment with immature stages, when either insecticide was applied to potted plants with M. morrisella in serpentine or blotch mines, significantly fewer adults emerged compared to the untreated plants. In contrast, adult emergence from potted plants treated with either insecticide when M. morrisella was in tentiform mines did not differ from untreated plants. In the experiment with adults, the survival of M. morrisella exposed to either insecticide through oral exposure or residual contact + oral exposure was significantly lower than that for residual contact exposure alone, which in turn was lower than the untreated control. Within each exposure route, the response to the 2 insecticides did not differ. In the field experiment, at 21 days after insecticide application, the mean percentage of leaf area mined was significantly lower in plots treated with Endigo ZCX compared to plots treated with Agri-Mek SC or those left untreated. Foxglove aphid, Aulacorthum solani Kaltenbach (Hemiptera: Aphididae), is a major pest worldwide. It can infest various crops, including soybean, and reduce yields. The use of insect-resistant cultivars can effectively manage pests. Dowling is a soybean cultivar that can control Aphis glycines through antibiosis and antixenosis. In this study, we investigated the feeding behavior of A. solani using electropenetrography (EPG), and its life-history traits using an age-stage, two-sex life table, and its settling preferences on Dowling (resistant) and Heinong 51 (HN51, susceptible) soybean cultivars. The Dowling cultivar showed strong antibiosis against A. solani. Aphids feeding on Dowling exhibited significantly reduced survival, fecundity, and longevity, and increased nymph duration. Moreover, Dowling had negative impacts on the demographic parameters of the aphids. The number of A. solani individuals was significantly less on Dowling than on HN51, indicating the antixenotic effects of Dowling. As shown using EPG, the mechanical blocking wave (F) of A. solani was significantly longer after feeding on Dowling than after feeding on HN51, indicating that Dowling has a higher mechanical resistance. In conclusion, Dowling exhibited strong resistance to A. solani. These results are beneficial for integrated pest management in soybean fields and breeding programs., (© The Author(s) 2024. Drought stress causes the greatest soybean [Glycine max (L.) Merr.] yield losses among the abiotic stresses in rain-fed U.S. growing areas. Because less than 10% of U.S. soybean hectares are irrigated, combating this stress requires soybean plants which possess physiological mechanisms to tolerate drought for a period of time. Phenotyping for these mechanisms is challenging, and the genetic architecture for these traits is poorly understood. A morphological trait, slow or delayed canopy wilting, has been observed in a few exotic plant introductions (PIs), and may lead to yield improvement in drought stressed fields. In this study, we visually scored wilting during stress for a panel of 162 genetically diverse maturity group VI-VIII soybean lines genotyped with the SoySNP50K iSelect BeadChip. Field evaluation of canopy wilting was conducted under rain-fed conditions at two locations (Athens, GA and Salina, KS) in 2015 and 2016. Substantial variation in canopy wilting was observed among the genotypes. Using a genome-wide association mapping approach, 45 unique SNPs that tagged 44 loci were associated with canopy wilting in at least one environment with one region identified in a single environment and data from across all environments. Several new soybean accessions were identified with canopy wilting superior to those of check genotypes. The germplasm and genomic regions identified can be used to better understand the slow canopy wilting trait and be incorporated into elite germplasm to improve drought tolerance in soybean.

The soybean pod borer Leguminivora glycinivorella (Matsumura) is one of the most important soybean pests and often causes serious damage to Glycine max (L.) Merr., a leading source of dietary protein and oil in animal feed. However, the potential distribution patterns of this economically important pest and its driving factors require further investigation. Here, we used the optimized MaxEnt model to predict the potential distribution of this pest with multiple variables associated with climate, land use, and host plant, at its recorded range and a globe scale. Based on 4 variable combinations, the results show that the current suitable habitats of L. glycinivorella are primarily distributed in most of China, the Korean Peninsula, and Japan. Whereas no suitable area is present in other continents. In future projections, the suitable region shows a slight northward expansion compared with the result predicted with current climatic conditions, and the suitable areas of almost all future projections were stable in size. Among the 9 bioclimatic factors, BIO03 (isothermality) consistently highly contributes to the predictions, indicating that temperature may be a key factor influencing the habitat distribution of L. glycinivorella. Comparative analyses of projections further show that non-climatic factors are informative in the modeling as routinely used bioclimate variables. The spatio-temporal distribution patterns of suitable habitats and the regulatory factors predicted in this study could provide important guidance for L. glycinivorella management., (© The Author(s) 2024. Improving soybean (Glycine max) seed composition by increasing the protein and oil components will add significant value to the crop and enhance environmental sustainability. Diacylglycerol acyltransferase (DGAT) catalyzes the final rate-limiting step in triacylglycerol biosynthesis and has a major impact on seed oil accumulation. We previously identified a soybean DGAT1b variant modified with 14 amino acid substitutions (GmDGAT1b-MOD) that increases total oil content by 3 percentage points when overexpressed in soybean seeds. In the present study, additional GmDGAT1b variants were generated to further increase oil with a reduced number of substitutions. Variants with one to four amino acid substitutions were screened in the model systems Saccharomyces cerevisiae and transient Nicotiana benthamiana leaf. Promising GmDGAT1b variants resulting in high oil accumulation in the model systems were selected for overexpression in soybeans. One GmDGAT1b variant with three novel amino acid substitutions (GmDGAT1b-3aa) increased total soybean oil to levels near the previously discovered GmDGAT1b-MOD variant. In a multiple location field trial, GmDGAT1b-3aa transgenic events had significantly increased oil and protein by up to 2.3 and 0.6 percentage points, respectively. The modeling of the GmDGAT1b-3aa protein structure provided insights into the potential function of the three substitutions. Insecticides prevent or reduce insect crop damage, maintaining crop quality and quantity. Physiological traits, such as an insect's feeding behavior, influence the way insecticides are absorbed and processed in the body (toxicokinetics), which can be exploited to improve species selectivity. To fully understand the uptake of insecticides, it is essential to study their total uptake and toxicokinetics independent of their toxic effects on insects. We studied the toxicokinetics (TK) of insecticidally inactive test compounds incorporating agro-like structural motifs in larvae of the Egyptian cotton leafworm (Spodoptera littoralis, Lepidoptera), and their distribution across all biological matrices, using laboratory experiments and modeling. We measured Spodoptera larval behavior and temporal changes of whole-body concentrations of test compounds during feeding on treated soybean leaf disks and throughout a subsequent depuration period. Differences in the distribution of the total quantities of compounds were found between the biological matrices leaf, larva, and feces. Rate constants for uptake and elimination of test compounds were derived by calibrating a toxicokinetic model to the whole-body concentrations. Uptake and elimination rate constants depended on the physicochemical properties of the test compounds. Increasing hydrophobicity increased the bioaccumulation potential of test compounds. Incomplete quantities in larval matrices indicated that some compounds may undergo biotransformation. As fecal excretion was a major elimination pathway, the variable time of release and number of feces pellets led to a high variability in the body burden. During ongoing proteomic analysis of the soybean (Glycine max (L.) Merr) germplasm collection, PI 603408 was identified as a landrace whose seeds lack accumulation of one of the major seed storage glycinin protein subunits. Whole genomic resequencing was used to identify a two-base deletion affecting glycinin 5. The newly discovered deletion was confirmed to be causative through immunological, genetic, and proteomic analysis, and no significant differences in total seed protein content were found to be due to the glycinin 5 loss-of-function mutation per se. In addition to focused studies on this one specific glycinin subunit-encoding gene, a total of 1,858,185 nucleotide variants were identified, of which 39,344 were predicted to affect protein coding regions. In order to semiautomate analysis of a large number of soybean gene variants, a new SIFT 4G (Sorting Intolerant From Tolerated 4 Genomes) database was designed to predict the impact of nonsynonymous single nucleotide soybean gene variants, potentially enabling more rapid analysis of soybean resequencing data in the future.

Seedcorn maggot, Delia platura (Meigen) (Diptera: Anthomyiidae), is an economically important early-season pest of corn and soybean in the United States. Adult seedcorn maggot is attracted to decomposing plant residues for oviposition, creating potential management issues where growers typically use tillage to incorporate fertility amendments and to create a seedbed. The use of growing degree-day models to time planting dates is an important tool for effectively managing this pest, but their use has not been examined in organic crop production. Here, we report the results of experiments to determine the effects of cover crops, tillage, and relative planting date on seedcorn maggot in corn and soybean in 2 experiments: The first during the transition to organic from conventional management and the second during the 3 yr following organic certification in central Pennsylvania, United States. Overall, delaying the planting date by 1-2 wk reduced fly emergence in corn, but not in soybean in both experiments. Seedcorn maggot emergence was also consistently greater in corn than in soybean, with 6 times more flies in corn than in soy. About 15 times more seedcorn maggot flies emerged from corn in treatments in which cover crops were managed with tillage compared to treatments in which cover crops were terminated with a roller-crimper followed by no-till planting of corn. Fly emergence was negatively related to the proportion of legumes in the cover crop mixture preceding corn. Cold winter temperatures govern the distribution and abundance of many insect species, but refugia that provide microclimates can moderate temperature-driven mortality. Winter temperatures have been implicated in limiting the survival and range of Piezodorus guildinii (Westwood) (Hemiptera: Pentatomidae; redbanded stink bug), an economically damaging invasive pest in the southeastern United States, but the role of refugia in overwintering survival of this pest is poorly understood. We conducted 2 studies in successive years to evaluate how leaf litter from hardwoods, pines, and soybeans modulate overwintering site selection and survival of P. guildinii. In the second-year study, we also quantified the buffering effect of the 3 leaf litter types compared to ambient conditions and assessed diapause. In the first-year study, we found that stink bugs preferentially dispersed into leaf litter compared with remaining unsheltered on bare soil; no clear preference among leaf litter types was found. In the second year, however, no clear differences were found among leaf litter types and bare soil. Means of daily minimum temperatures under leaf litter were at least 3.0 ± 0.9 °C (SE) warmer and generally less variable than ambient conditions. While high mortality in both studies illustrates that more work must be done to fully understand overwintering survival, limited survival through potentially lethal conditions in the first-year study nonetheless emphasizes the possibility of populations persisting and rebounding in the following spring. Drought tolerance is a complex trait in soybean that is controlled by polygenetic quantitative trait loci (QTLs). In this study, wilting score, days-to-wilting, leaf relative water content, and leaf relative conductivity were used to identify QTLs associated with drought tolerance in recombinant inbred lines derived from a cross between a drought-sensitive variety, Lin, and a drought-tolerant variety, Meng. A total of 33 drought-tolerance QTLs were detected. Of these 17 were major QTLs. In addition, 15 were novel drought-tolerance QTLs. The most predominant QTL was on chromosome 11. This was detected in at least three environments. The overlapped mapping interval of the four measured traits was 0.2 cM in genetic distance (about 220 kb in physical length). Glyma.11g143500 (designated as GmUAA6), which encodes a UDP-N-acetylglucosamine transporter, was identified as the most likely candidate gene. The allele of GmUAA6 from Lin (GmUAA6Lin) was associated with improved soybean drought tolerance. Overexpression of GmUAA6Lin in Arabidopsis and soybean hairy roots enhanced drought tolerance. Furthermore, a 3-bp insertion/deletion (InDel) in the coding sequence of GmUAA6 explained up to 49.9% of the phenotypic variation in drought tolerance-related traits, suggesting that this InDel might be used in future marker-assisted selection of drought-tolerant lines in soybean breeding programs., (© The Author(s) 2024. Soybean oil is highly unsaturated but oxidatively unstable, rendering it nonideal for food applications. Until recently, the majority of soybean oil underwent partial chemical hydrogenation, which produces trans fats as an unavoidable consequence. Dietary intake of trans fats and most saturated fats are conclusively linked to negative impacts on cholesterol levels and cardiovascular health. Two major soybean oil breeding targets are: (1) to reduce or eliminate the need for chemical hydrogenation, and (2) to replace the functional properties of partially hydrogenated soybean oil. One potential solution is the elevation of seed stearic acid, a saturated fat which has no negative impacts on cardiovascular health, from 3 to 4% in typical cultivars to > 20% of the seed oil. We performed QTL analysis of a population developed by crossing two mutant lines, one with a missense mutation affecting a stearoyl-acyl-carrier protein desaturase gene resulting in ∼11% seed stearic acid crossed to another mutant, A6, which has 24–28% seed stearic acid. Genotyping-by-sequencing (GBS)-based QTL mapping identified 21 minor and major effect QTL for six seed oil related traits and plant height. The inheritance of a large genomic deletion affecting chromosome 14 is the basis for largest effect QTL, resulting in ∼18% seed stearic acid. This deletion contains SACPD-C and another gene(s); loss of both genes boosts seed stearic acid levels to ≥ 18%. Unfortunately, this genomic deletion has been shown in previous studies to be inextricably correlated with reduced seed yield. Our results will help inform and guide ongoing breeding efforts to improve soybean oil oxidative stability.

The green-belly stink bug (Diceraeus melacanthus Dallas (Hemiptera: Pentatomidae)) is a key pest of corn-soybean crop systems and the management of this pest is difficult due to behavioral characteristics. However, products that alter its behavior, such as arrestants, dislodgers, and phagostimulants, have been used in an attempt to improve the effectiveness of chemical control. In this study, the effect of these products on the walking behavior of nymphs and adults of D. melacanthus was initially evaluated through computational behavioral tracking (Ethovision system). Adults of D. melacanthus exposed to the dislodgers Creolin Pearson and Quimifol S450 (sulfur) significantly increased the distance covered and the walking speed. On the other hand, the only treatment that significantly affected the walking behavior of nymphs was the treatment with soy milk (phagostimulant). The physical-chemical analyses indicated considerable changes in the pH and electrical conductivity of mixtures of such products with insecticides from different chemical groups, as well as lack of homogeneity (physical incompatibility). Nevertheless, none of the products tested improved the control efficacy of an insecticide based on imidacloprid + beta-cyfluthrin, either in laboratory tests (contact bioassay) or in tests conducted in corn crops during 2 crop harvests. Thus, the association of these products in a tank mixture does not increase the control levels of D. melacanthus in post-emergence of maize and may have an antagonistic effect in some associations., (© The Author(s) 2023. The plant microbiome and plant-associated bacteria are known to support plant health, but there are limited studies on seed and seedling microbiome to reveal how seed-associated bacteria may confer disease resistance. In this study, the application of antibiotics on soybean seedlings indicated that seed-associated bacteria were involved in the seed rot resistance against a soil-borne pathogen Calonectria ilicicola, but this resistance cannot be carried to withstand root rot. Using PacBio 16S rRNA gene full-length sequencing and microbiome analyses, 14 amplicon sequence variants (ASVs) including 2 ASVs matching to Bacillus altitudinis were found to be more abundant in the four most resistant varieties versus the four most susceptible varieties. Culture-dependent isolation obtained two B. altitudinis isolates that both exhibit antagonistic capability against six fungal pathogens. Application of B. altitudinis on the most resistant and susceptible soybean varieties revealed different colonization compatibility, and the seed rot resistance was restored in the five varieties showing higher bacterial colonization. Moreover, quantitative PCR confirmed the persistence of B. altitudinis on apical shoots till 21 days post-inoculation (dpi), but 9 dpi on roots of the resistant variety TN5. As for the susceptible variety HC, the persistence of B. altitudinis was only detected before 6 dpi on both shoots and roots. The short-term colonization of B. altitudinis on roots may explain the absence of root rot resistance. Collectively, this study advances the insight of B. altitudinis conferring soybean seed rot resistance and highlights the importance of considering bacterial compatibility with plant varieties and colonization persistence on plant tissues., (© The Author(s) 2024. Solar-induced chlorophyll fluorescence (SIF) provides an opportunity to rapidly and non-destructively investigate how plants respond to stress. Here, we explored the potential of SIF to detect the effects of elevated O3 on soybean in the field where soybean was subjected to ambient and elevated O3 throughout the growing season in 2021. Exposure to elevated O3 resulted in a significant decrease in canopy SIF at 760 nm (SIF760), with a larger decrease in the late growing season (36%) compared with the middle growing season (13%). Elevated O3 significantly decreased the fraction of absorbed photosynthetically active radiation by 8-15% in the middle growing season and by 35% in the late growing stage. SIF760 escape ratio (fesc) was significantly increased under elevated O3 by 5-12% in the late growth stage due to a decrease of leaf chlorophyll content and leaf area index. Fluorescence yield of the canopy was reduced by 5-11% in the late growing season depending on the fesc estimation method, during which leaf maximum carboxylation rate and maximum electron transport were significantly reduced by 29% and 20% under elevated O3. These results demonstrated that SIF could capture the elevated O3 effect on canopy structure and acceleration of senescence in soybean and provide empirical support for using SIF for soybean stress detection and phenotyping., (© The Author(s) 2023. The soybean gall midge, Resseliella maxima Gagné (Diptera: Cecidomyiidae), is a pest that injures soybeans in the Midwest United States. Little is known about the natural enemies of R. maxima or the potential for biological control. Therefore, we performed a 2-yr survey in Minnesota to examine the predator community associated with R. maxima infestations. We found that Orius insidiosus (Say) (Heteroptera: Anthocoridae) and Pterostichus melanarius (Illiger) (Coleoptera: Carabidae) were the most common foliar- and ground-foraging predators, respectively. Some of the commonly encountered predator species were tested in laboratory predation experiments. Harmonia axyridis (Pallas) (Coleoptera: Coccinellidae) and O. insidiosus represented the foliar-foraging predators tested, and H. axyridis consumed significantly more R. maxima larvae than O. insidiosus. Among the ground-foraging predators, 4 carabids were tested. Poecilus lucublandus (Say) (Coleoptera: Carabidae) and Pt. melanarius consumed significantly more R. maxima larvae than Poecilus chalcites (Say) (Coleoptera: Carabidae) and Bembidion quadrimaculatum oppositum (Say) (Coleoptera: Carabidae). We conclude that Pt. melanarius should receive further attention as a potential biological control agent of R. maxima, due to its high abundance in the soybean fields in this study, temporal overlap with the pest, and high propensity to feed on the pest., (© The Author(s) 2023. Soybean gall midge, Resseliella maxima Gagné, was recently identified as a new species causing significant injury to soybean and is currently found in 164 counties across 7 midwestern states (NE, IA, SD, MN, MO, ND, and KS). Infestation of soybean begins in late spring, when adults emerge from last year's soybean field. Infestation of a new soybean crop depends on the presence of fissures which start to form at the base of the soybean plant around the V2 stage. Field observations indicate that these fissures are only present below the cotyledonary nodes or in the area within 3-5 cm above the soil surface. To determine the importance of these fissures for R. maxima infestation and plant injury, hilling or the movement of the soil to cover the base of soybean plants at the V2-V3 stage was compared with the standard practice (no-hilling). Field studies were conducted at 3 sites in east-central Nebraska during the 2021 growing season. The results showed a significant reduction in the frequency of infested plants, larval number per plant, and plant injury for hilled compared to no-hill treatment. This reduction in the presence of larvae and plant injury corresponded with a significantly greater yield for hilled compared to the no-hill treatment. These results highlight the importance of fissures on soybean for R. maxima adult infestation as well as the potential for hilling to be used as a management strategy for R. maxima., (© The Author(s) 2023. Lipid droplets and membranes in radicle cells from desiccated embryonic axes of soybean (Glycine max) seeds were examined by a recently developed correlative light and electron microscopy system, which has been designed to facilitate the observation of identical locations using an upright reflected light microscope and compact SEM successively with minimum time lapse. Lipids are major components of membranes and are also stored in numerous lipid droplets lining plasma membranes in many seed cells. Fluorescently stained lipid droplets and membranes in the desiccated radicle cells were mainly located along the surface of shrunk protoplasm and around presumptive protein bodies, which will turn into vacuoles and increase their volume for radicle protrusion. Co-localization of lipid droplets and membranes suggests the presence of a membrane protection mechanism during desiccation and rehydration processes that ensures prompt elongation of radicle cells during germination., (© The Author(s) 2023. Proteins and peptides derived from various food sources are used in a variety of applications, including functional foods, pharmaceuticals, and cosmetics. The three-dimensional structure of proteins provides useful insights into their functions and essential information for the creation of proteins with new functions. In this review, a series of functional conversion technologies based on protein structural information derived from foods traditionally consumed in Japan, such as natto (fermented soybeans) and rice, are introduced. For natto, we first identified 2 types of Bacillus subtilis-derived endolytic and exolytic enzymes with different modes of action on soybean cell wall polysaccharides and then focused on the technology used to create an endolytic enzyme from an exolytic enzyme. By applying this technology, a method for creating novel bioactive peptides from rice seed proteins was established. Height is a critical component of plant architecture, significantly affecting crop yield. The genetic basis of this trait in soybean remains unclear. In this study, we report the characterization of the Compact mutant of soybean, which has short internodes. The candidate gene was mapped to chromosome 17, and the interval containing the causative mutation was further delineated using biparental mapping. Whole-genome sequencing of the mutant revealed an 8.7 kb deletion in the promoter of the Glyma.17g145200 gene, which encodes a member of the class III gibberellin (GA) 2-oxidases. The mutation has a dominant effect, likely via increased expression of the GA 2-oxidase transcript observed in green tissue, as a result of the deletion in the promoter of Glyma.17g145200. We further demonstrate that levels of GA precursors are altered in the Compact mutant, supporting a role in GA metabolism, and that the mutant phenotype can be rescued with exogenous GA3. We also determined that overexpression of Glyma.17g145200 in Arabidopsis results in dwarfed plants. Thus, gain of promoter activity in the Compact mutant leads to a short internode phenotype in soybean through altered metabolism of gibberellin precursors. Conservation agriculture practices such as eliminating tillage and planting high residue cover crops are becoming increasingly important in field crop systems in the US Mid-Atlantic. However, these practices have sometimes been associated with an increase in moderate to severe damage to field crops by slugs. Conserving natural enemy populations is a desirable way to manage slug infestations because remedial control measures are limited. Here, we tested the effects of conservation practices, weather, and natural enemies on slug activity-density measured by tile traps placed among 41 corn and soybean fields during the spring of 2018 and 2019 in the Northern Shenandoah Valley, Virginia, USA. We found that a positive effect of cover crops on slug activity-density was reduced by tillage and that slug activity-density declined with increasing ground beetle activity-density. Slug activity-density also declined with decreasing rainfall and increasing average temperature. Weather was the only significant predictor of ground beetle activity-density, which was reduced in sites and weeks that were relatively hot and dry or that were cool and wet. However, we also found a marginally significant negative effect of pre-plant insecticides on ground beetles. We suggest that the observed interacting effects of cover crops and tillage reflect favorable conditions for slugs provided by increased small grain crop residue that can be mitigated to some extent by even low levels of tillage. This study aimed to characterize and correlate the stylet penetration behaviors of nymphs of the Neotropical brown stink bug, Euschistus heros (F.), on immature soybean pods. Waveforms were obtained using electropenetrography (EPG). The findings revealed that the nymphs exploited the xylem vessels and the seed tegument or endosperm. Primarily 4 phases were characterized: nonfeeding, pathway, salivation, and ingestion. The waveforms of each phase were similar in appearance across instars. The biological meanings of waveforms were based on visual observations, comparison with waveforms of adults, and histological studies. Np represents the insect resting or walking on soybean pod surface. Eh1 represents the first contact between the mouthparts (stylets) and plant tissue. Eh2 represents xylem sap ingestion, and Eh3 represents seed activities (including tegument and endosperm). The number of waveform events did not differ among instars for all waveforms. However, for Eh3, fifth instars performed more activities than other instars. The second instars had the smallest value, and third and fourth instars had intermediate values. For total duration, all waveforms differed among instars. Np duration was shorter for third compared with second and fourth instars and intermediate for fifth instar. For Eh1, second and third instars had the longest duration (1.5× to 2× greater) compared with fourth and fifth instars. For Eh2 and Eh3, the second-instar showed the longest (~2× greater) and shortest durations, respectively. The Neotropical brown stink bug, Euschistus heros (Fabricius, 1798) (Hemiptera: Pentatomidae), is one of the major insect pests in soybean crops, causing severe damage; however, some aspects of its biology that are essential for management remain unknown. To aid in the management of E. heros, the present study investigated the fertility life table of this species at 7 temperatures (18, 20, 22, 25, 28, 30, and 32 ± 1 °C) and 4 relative-humidity levels (30, 50, 70, and 90 ± 10%). Based on the net reproductive rate, R0, we developed an ecological zoning for this pest in Brazil, to identify climatically favorable areas for population growth. Our results indicated that the most favorable range is between 25 and 28 °C and above 70% RH. The ecological zoning indicated that farmers should be more concerned in the northern and Midwest regions, which include the state of Mato Grosso, the largest soybean and corn producer in Brazil. Background and Aims: Leaf shape in crops can impact light distribution and carbon capture at the whole plant and canopy level. Given similar leaf inclination, narrow leaves can allow a greater fraction of incident light to pass through to lower canopy leaves by reducing leaf area index, which can potentially increase canopy-scale photosynthesis. Soybean has natural variation in leaf shape which can be utilized to optimize canopy architecture. However, the anatomical and physiological differences underlying variation in leaf shape remain largely unexplored., Methods: In this study, we selected 28 diverse soybean lines with leaf length to width ratios (leaf ratio) ranging between 1.1 and 3.2. We made leaf cross-sectional, gas exchange, vein density and hydraulic measurements and studied their interrelationships among these lines., Key Results: Our study shows that narrow leaves tend to be thicker, with an ~30 µm increase in leaf thickness for every unit increase in leaf ratio. Interestingly, thicker leaves had a greater proportion of spongy mesophyll while the proportions of palisade and paraveinal mesophyll decreased. In addition, narrow and thicker leaves had greater photosynthesis and stomatal conductance per unit area along with greater leaf hydraulic conductance., Conclusions: Our results suggest that selecting for narrow leaves can improve photosynthetic performance and potentially provide a yield advantage in soybean., (Published by Oxford University Press on behalf of the Annals of Botany Company 2022.)

The larval parasitoid Diadegma hiraii (Kusigemati) was evaluated as a potential biological control agent of the soybean pod borer, Leguminivora glycinivorella (Matsumura). The timing of adult emergence after overwintering was ascertained, and land-use factors that enhance population density were analyzed. Host cocoons were collected and exposed to different temperatures and photoperiod regimes. Subsequently, the emergence of parasitoid was monitored. Land-use types were categorized into 4 land-use types (Poaceae, Fabaceae, Brassicaceae, and forest). Adult parasitoid emergence was dependent on temperature, but largely unaffected by photoperiod. The estimated emergence time of parasitoid was 3 months before the occurrence of the host, suggesting that the overwintered generation may lay eggs in alternate hosts. Parasitism rate was positively correlated with the area covered by Poaceae plants within a 500-m radius of the soybean field. Based on the results of the overwintering ecology and landscape analysis, D. hiraii probably completes its life cycle in the agroecosystems. The parasitoid's effectiveness as a biological control agent may be influenced by the arrangement of land-use types in the agroecosystems surrounding soybean fields. However, the pest control provided by D. hiraii is limited because of approximately 30% of parasitism rate. The redbanded stink bug, Piezodorus guildinii (Westwood, 1837), is a highly destructive soybean pest native to the Neotropical Region. In the past 60 yr, P. guildinii has been observed to expand its distribution in North and South America, causing significant soybean yield losses. In order to predict the future distribution direction of P. guildinii and create an effective pest control strategy, we projected the potential global distribution of P. guildinii using 2 different emission scenarios, Shared Socioeconomic Pathways 126 and 585, and 3 Earth system models, with the maximum entropy niche model (MaxEnt). Then, the predicted distribution areas of P. guildinii were jointly analyzed with the main soybean-producing areas to assess the impact for different soybean region. Our results showed that temperature is the main environmental factor limiting the distribution of P. guildinii. Under present climate conditions, all continents except Antarctica have suitable habitat for P. guildinii. These suitable habitats overlap with approximately 45.11% of the total global cultivated soybean areas. Moreover, P. guildinii was predicted to expand its range in the future, particularly into higher latitudes in the Northern hemisphere. Countries, in particular the United States, where soybean is widely available, would face a management challenge under global warming. In addition, China and India are also high-risk countries that may be invaded and should take strict quarantine measures. Recombination allows for the exchange of genetic material between two parents, which plant breeders exploit to make improved cultivars. This recombination is not distributed evenly across the chromosome. Recombination mostly occurs in euchromatic regions of the genome and even then, recombination is focused into clusters of crossovers termed recombination hotspots. Understanding the distribution of these hotspots along with the sequence motifs associated with them may lead to methods that enable breeders to better exploit recombination in breeding. To map recombination hotspots and identify sequence motifs associated with hotspots in soybean [Glycine max (L.) Merr.], two biparental recombinant inbred lines populations were genotyped with the SoySNP50k Illumina Infinium assay. A total of 451 recombination hotspots were identified in the two populations. Despite being half-sib populations, only 18 hotspots were in common between the two populations. While pericentromeric regions did exhibit extreme suppression of recombination, 27% of the detected hotspots were located in the pericentromeric regions of the chromosomes. Two genomic motifs associated with hotspots are similar to human, dog, rice, wheat, drosophila, and arabidopsis. These motifs were a CCN repeat motif and a poly-A motif. Genomic regions spanning other hotspots were significantly enriched with the tourist family of mini-inverted-repeat transposable elements that resides in <0.34% of the soybean genome. Plant specialized metabolites (PSMs) are often stored as glycosides within cells and released from the roots with some chemical modifications. While isoflavones are known to function as symbiotic signals with rhizobia and to modulate the soybean rhizosphere microbiome, the underlying mechanisms of root-to-soil delivery are poorly understood. In addition to transporter-mediated secretion, the hydrolysis of isoflavone glycosides in the apoplast by an isoflavone conjugate-hydrolyzing β-glucosidase (ICHG) has been proposed but not yet verified. To clarify the role of ICHG in isoflavone supply to the rhizosphere, we have isolated two independent mutants defective in ICHG activity from a soybean high-density mutant library. In the root apoplastic fraction of ichg mutants, the isoflavone glycoside contents were significantly increased, while isoflavone aglycone contents were decreased, indicating that ICHG hydrolyzes isoflavone glycosides into aglycones in the root apoplast. When grown in a field, the lack of ICHG activity considerably reduced isoflavone aglycone contents in roots and the rhizosphere soil, although the transcriptomes showed no distinct differences between the ichg mutants and wild-types (WTs). Despite the change in isoflavone contents and composition of the root and rhizosphere of the mutants, root and rhizosphere bacterial communities were not distinctive from those of the WTs. Root bacterial communities and nodulation capacities of the ichg mutants did not differ from the WTs under nitrogen-deficient conditions either. Sampling plans are an essential part of integrated pest management programs. Sequential sampling plans enable rapid and low-cost assessment of pest densities. Thrips are emerging pests in soybean crops, and the main method used in pest control is chemical. In soybean crops, insecticides are applied mainly using tractors or airplanes. Thus, this work aimed to determine sequential sampling plans for thrips in soybean crops with insecticide applications using a tractor or airplane. Data were collected in 56 soybean fields, and each field was 20 ha. Sampling plans were determined and validated. The lower (m0) and upper (m1) limits of the sequential sampling plans were: m0 = 1.72 and m1 = 3.43 (by tractor applications) and, m0 = 2.27 and m1 = 4.53 thrips. sample-1 (by airplane applications). The slope (S) and the lower (h0) and upper (h1) intercepts of the sequential sampling plans were: S = 2.42, h0 = -5.79, and h1 = 5.79 (by tractor applications) and, S = 3.19, h0 = -6.83, and h1 = 6.83 (by airplane applications). Sequential sampling plans allowed for correct decisions to be made in all situations using a maximum of 10 samples. The sequential plan reduced the sampling effort by over 87% compared to conventional sampling plans. Improvements in genetics, technology, and agricultural intensification have increased soybean yields; however, adverse climate conditions may prevent these gains from being fully realized in the future. Higher growing season temperatures reduce soybean yields in key production regions including the US Midwest, and better understanding of the developmental and physiological mechanisms that constrain soybean yield under high temperature conditions is needed. This study tested the response of two soybean cultivars to four elevated temperature treatments (+1.7, +2.6, +3.6, and +4.8 °C) in the field over three growing seasons and identified threshold temperatures for response and linear versus non-linear trait responses to temperature. Yield declined non-linearly to temperature, with decreases apparent when canopy temperature exceeded 20.9 °C for the locally adapted cultivar and 22.7°C for a cultivar adapted to more southern locations. While stem node number increased with increasing temperature, leaf area index decreased substantially. Pod production, seed size, and harvest index significantly decreased with increasing temperature. The seasonal average temperature of even the mildest treatment exceeded the threshold temperatures for yield loss, emphasizing the importance of improving temperature tolerance in soybean germplasm with intensifying climate change., (Published by Oxford University Press on behalf of the Society for Experimental Biology 2022.)

Aim: The aims of this study were to characterize the causal agent of soybean leaf spot and determine the pathogenicity of the pathogen to the main crops in Northeast China and detect the sensitivity of pathogens to the main chemical fungicides and the potted control effect., Methods and Results: In 2020 and 2021, an uncommon leaf spot was observed in Harbin, Heilongjiang Province (125°42'-130°10'E, 44°04'-46°40'N), China. The pathogen can infect soybean leaves and cause leaf spot. We collected diseased soybean leaves and isolated four pathogen organisms, all of which were identified as Alternaria alternata through morphological and molecular identification. Koch's postulates were used to confirm pathogenicity. To the best of our knowledge, this is the first report on soybean leaf spot disease caused by A. alternata in northeast China. Moreover, A. alternata had a broad host range and caused leaf spot in most legumes. However, it did not infect medicated lentil (Dolicho Lablab L.) or tobacco (Nicotiana tabacum L.). Assessment of A. alternaria susceptibility to fungicides by spore germination method, isolates of A. alternata were most sensitive to flusilazole, with EC50 values of 0.0040-0.0053 μg ml-1. Through two pot experiments, the average control efficacy of 0.1 mg ml-1 flusilazole on soybean leaf spot caused by A. alternata was 80.7%., Conclusions: The work reported that A. alternata is the pathogen organism that causes soybean leaf spot in northern China. The pathogen organism can infect a variety of leguminous plants. Lactulose is commonly used in pharmacy for constipation and hepatic encephalopathy treatment. The prebiotic effect of lactulose is also often mentioned. However, its cryoprotective effect in combination with lecithin on the main representatives of probiotics has not been tested yet. The 12 taxa of bifidobacteria and Lactobacillaceae members were used for the purpose. These were mixed in a ratio of 1:1 with lactulose + lecithin (finally 5.0% and 1.25%, respectively; LL). The 25% glycerol (G+) solution and cultures themselves were applied as positive and negative controls, respectively. Bacterial suspensions were stored at a mild freezing temperature (-20°C) until the end of the experiment (210th day). The LL solution had a comparable (insignificant difference at the P-value = 0.05) cryoprotective effect as the positive control in five of six bifidobacteria and in three of six representatives of Lactobacillaceae. The better cryoprotective effect was revealed in other Lactobacillaceae. At the end of the experiment, the generally accepted therapeutic minimum (>107 Colony Forming Units/mL) was determined in LL solution in five bifidobacteria and four Lactobacillaceae strains. Increased root secretion of H+ is a known strategy in plant adaption to low phosphorus (P) stress as it enhances mobilization of sparingly soluble P sources in the soil. However, our knowledge of the full effects induced by this enhanced acidification of the rhizosphere remains incomplete. In this study we found that P deficiency increased the net H+ flux rate from soybean (Glycine max) roots. Among the eight H+-pyrophosphatase (GmVP) genes in the soybean genome, GmVP2 showed the highest expression level under low P conditions. Transient expression of a GmVP2-GFP construct in tobacco (Nicotiana tabacum) leaves, together with functional characterization of GmVP2 in transgenic soybean hairy roots demonstrated that it encodes a plasma-membrane transporter that mediates H+ exudation. Overexpression of GmVP2 in Arabidopsis resulted in enhanced root H+ exudation, promoted root growth, and improved the utilization of sparingly soluble Ca-P. The improved root growth caused by GmVP2-overexpression might be due to the differential expression of genes related to hormone and flavonoid metabolism, and to root development. Overexpression of GmVP2 also changed the structure of the rhizospheric microbial community, as reflected by a preferential accumulation of Acidobacteria. Overall, our results suggest that GmVP2 mediates H+ exudation in the root response to Pi starvation, and that this influences plant growth, the mobilization sparingly soluble P-sources, and the structure of the microbial community in a coordinated manner., (© The Author(s) 2022. In this review, we describe the genomic and physiological features of the yeast species predominantly isolated from Nuruk, a starter for traditional Korean rice wines, and Jang, a traditional Korean fermented soy product. Nuruk and Jang have several prevalent yeast species, including Saccharomycopsis fibuligera, Hyphopichia burtonii, and Debaryomyces hansenii complex, which belong to the CUG clade showing high osmotic tolerance. Comparative genomics revealed that the interspecies hybridization within yeast species for generating heterozygous diploid genomes occurs frequently as an evolutional strategy in the fermentation environment of Nuruk and Jang. Through gene inventory analysis based on the high-quality reference genome of S. fibuligera, new genes involved in cellulose degradation and volatile aroma biosynthesis and applicable to the production of novel valuable enzymes and chemicals can be discovered. The integrated genomic and transcriptomic analysis of Hyphopichia yeasts, which exhibit strong halotolerance, provides insights into the novel mechanisms of salt and osmo-stress tolerance for survival in fermentation environments with a low-water activity and high-concentration salts. Ontogenic changes in soybean radiation use efficiency (RUE) have been attributed to variation in specific leaf nitrogen (SLN) based only on data collected during seed filling. We evaluated this hypothesis using data on leaf area, absorbed radiation (ARAD), aboveground dry matter (ADM), and plant nitrogen (N) concentration collected during the entire crop season from seven field experiments conducted in a stress-free environment. Each experiment included a full-N treatment that received ample N fertilizer and a zero-N treatment that relied on N fixation and soil N mineralization. We estimated RUE based on changes in ADM between sampling times and associated ARAD, accounting for changes in biomass composition. The RUE and SLN exhibited different seasonal patterns: a bell-shaped pattern with a peak around the beginning of seed filling, and a convex pattern followed by an abrupt decline during late seed filling, respectively. Changes in SLN explained the decline in RUE during seed filling but failed to predict changes in RUE in earlier stages and underestimated the maximum RUE observed during pod setting. Comparison between observed and simulated RUE using a process-based crop simulation model revealed similar discrepancies. The decoupling between RUE and SLN during early crop stages suggests that leaf N is above that needed to maximize crop growth but may play a role in storing N that can be used in later reproductive stages to meet the large seed N demand associated with high-yielding crops., (© The Author(s) 2022. Saponins are amphipathic glycosides with detergent properties present in vegetables. These compounds, when ingested, can cause difficulties in absorbing nutrients from food and even induce inflammatory processes in the intestine. There is already some evidence that saponins can be degraded by β-glucosidases of the GH3 family. In the present study, we evaluated, through computational tools, the possibility of a β-glucosidase (AMBGL17) obtained from a metagenomic analysis of the Amazonian soil, to catalytically interact with a saponin present in soybean. For this, the amino acid sequence of AMBGL17 was used in a phylogenetic analysis to estimate its origin and to determine its three-dimensional structure. The 3D structure of the enzyme was used in a molecular docking analysis to evaluate its interaction with soy saponin as a ligand. The results of the phylogenetic analysis showed that AMBGL17 comes from a microorganism of the phylum Chloroflexi, probably related to species of the order Aggregatinales. Molecular docking showed that soybean saponin can interact with the catalytic site of AMBGL17, with the amino acid GLY345 being important in this catalytic interaction, especially with a β-1,2 glycosidic bond present in the carbohydrate portion of saponin. Chemical nitrogen fixation by the Haber-Bosch method permitted industrial-scale fertilizer production that supported global population growth, but simultaneously released reactive nitrogen into the environment. This minireview highlights the potential for bacterial nitrogen fixation and mitigation of greenhouse gas (GHG) emissions from soybean and rice fields. Nitrous oxide (N2O), a GHG, is mainly emitted from agricultural use of nitrogen fertilizer and symbiotic nitrogen fixation. Some rhizobia have a denitrifying enzyme system that includes an N2O reductase and are able to mitigate N2O emission from the rhizosphere of leguminous plants. Type II methane (CH4)-oxidizing bacteria (methanotrophs) are endophytes in paddy rice roots and fix N2 using CH4 (a GHG) as an energy source, mitigating the emission of CH4 and reducing nitrogen fertilizer usage. The kudzu bug, Megacopta cribraria (Fabricius) (Hemiptera: Plataspidae), is a major economic pest of soybean in the southeastern United States. With climate warming, this pest is expected to move northward and cause additional crop damage. Parasitoid biocontrol is a potential method of integrated pest management for kudzu bug. Two species of egg parasitoid wasps have been observed emerging from kudzu bug egg masses in the southeastern United States: Paratelenomus saccharalis (Dodd) (Hymenoptera: Platygastridae) and Ooencyrtus nezarae (Ishii) (Hymenoptera: Encyrtidae). This paper used egg mass emergence data collected between 2018 and 2020 in Alabama soybean fields and compared the data to weather indices. Indices included the number of days with minimum temperatures below zero, accumulated rainfall (mm m-2), as well as species specific metrics of accumulated growing degree days, accumulated daily minimum temperature (°C), and accumulated daily maximum temperature (°C). Emergence of the generalist parasitoid, O. nezarae, was highly correlated with kudzu bug nymph abundance, accumulated degree day, accumulated daily temperatures, and precipitation. Ooencyrtus nezarae emergence was predicted in a stepwise regression equation by aggregated degree day and date of collection, which indicates that seasonality may be a predictor of its presence. In contrast, collections of the specialist parasitoid, P. saccharalis, were near-zero throughout the collection period, suggesting that this species may no longer be a usable biocontrol agent in the southeastern United States as a result of external limiting factors., (© The Author(s) 2022. The local temperature in the Harbin region of northeast China has been rising, with temperatures above 27°C often being recorded. The soybean aphid, Aphis glycines (Matsumura) (Hemiptera: Aphididae), and foxglove aphid, Aulacorthum solani (Kaltenbach) (Hemiptera: Aphididae), are important pests in soybeans in the region, which face great survival pressure due to rising temperatures. In this study, A. glycines and A. solani were reared at thermoperiods with diurnal temperatures of 25, 27, 29, 31, 33, 35, and 37°C, nocturnal temperature of 20 °C, 70% ± 5% relative humidity, and a photoperiod of 14:10 (L: D) hr. Diurnal thermoperiod 25°C was used as control. Some life characteristics of the aphids were assessed. The adaptation of A. solani to high diurnal temperatures was found to be worse than that of A. glycines. Aphis glycines could develop and reproduce successfully at diurnal temperatures of 27-35°C, whereas A. solani could only develop and reproduce at 27-29°C. At diurnal thermoperiods at 25°C, adult lifespan of A. solani was as long as that of A. glycines. When diurnal temperatures increased from 27 to 31°C, adult lifespan of A. solani were shorter than those of A. glycines. At diurnal thermoperiods at 25 and 27°C, the body sizes of A. solani adults were larger than those of A. glycines. At diurnal thermoperiods at 29 and 31°C, the body size of A. solani adults were as large as those of A. glycines. Our results provide important information on the survival, development, and reproduction of A. glycines and A. solani exposed to temperatures above 27°C, which is important for evaluating the adaptability of aphids to high temperatures and predicting their dynamics in soybeans in northeast China, where the local environmental temperature continues to rise., (© The Author(s) 2022. Fitness costs associated with insect resistance to insecticides can be exploited to implement resistance management programs. However, most of these studies are restricted to evaluating biological traits on artificial diets. Here, we investigated the fitness cost associated with chlorantraniliprole in Spodoptera frugiperda (J.E. Smith) feeding on corn, soybean, and cotton plants. We used a near-isogenic strain of S. frugiperda resistant to chlorantraniliprole (Iso-RR), a susceptible strain (SS), and heterozygotes strains (H1 and H2) to evaluate several biological and population growth parameters. Larval survival of the Iso-RR strain was on average 90% on corn, 65% on soybean, and 57% on cotton plants. Development time of the larval stage also differed among host plants, Iso-RR strain took on average 14, 17, and 26 days to reach the pupal stage on corn, soybean, and cotton plants respectively. Net reproductive rate, intrinsic rate of population increase, and finite rate of population increase were higher for Iso-RR strain feeding on corn plants than other host plants. The relative fitness, based on the intrinsic rate of population increase, of S. frugiperda resistant strain on corn, soybean, and cotton plants were 1.04, 0.85, and 0.88, respectively. Therefore, no fitness cost was observed for S. frugiperda feeding on corn plants, but a significant fitness cost was observed when this pest fed on soybean and cotton plants. We showed that the food source influences the fitness cost of S. frugiperda resistant to diamide. Hemp (Cannabis sativa L.) is a reemerging crop in the United States with increasing outdoor acreage in many states. This crop offers a potential host for polyphagous, defoliating lepidopteran pests currently present in Louisiana. The ability of soybean looper [Chrysodeixis includens (Walker)] (Lepidoptera: Noctuidae), fall armyworm [Spodoptera frugiperda (J.E. Smith)] (Lepidoptera: Noctuidae), and beet armyworm [Spodoptera exigua (Hübner)] to develop and reproduce on hemp was investigated in this study. Insects were reared on two hemp varieties, Maverick and Pipeline, as well as documented host plants soybean [Glycine max (L.) Merr.] (Fabales: Fabaceae) variety UA5414RR and cowpea [Vigna unguiculata (L.)] (Fabales: Fabaceae) variety Quickpick Pinkeye. Larvae of all three species reared on 'Maverick' had significantly faster preadult developmental times compared to the other hosts. Chrysodeixis includens larvae fed excised leaves of 'Maverick' and 'Pipeline' experienced higher intrinsic and finite rates of increase, higher net reproductive rates, and faster mean generation and doubling times. Spodoptera frugiperda larvae reared on 'Maverick', 'Pipeline', and 'UA5414RR' had higher intrinsic and finite rates of increase, higher net reproductive rates, and faster mean generation and doubling times compared to 'Quickpick Pinkeye'. Spodoptera exigua larvae had the highest survivorship on 'Maverick' and similar, positive lifetable statistics when reared on 'Maverick' and 'Pipeline'. Large-scale soybean [Glycine max (L.) Merr.] cultivation has substantially transformed the Midwestern landscape in recent decades. Floral nectar produced by immense fields of soybeans has the potential to influence foraging ecology and resource accumulation of honey bee (Apis mellifera L.) colonies. In this study, we combined microscopic and molecular pollen analysis of honey samples with waggle dance inference of spatial foraging patterns to demonstrate that honey bees routinely forage on soybeans in Ohio. In analyzing honey samples from across the state, we found ubiquitous presence of soybean pollen in honey collected from agricultural lands during soybean bloom. The abundance of soybean pollen in honey increased with the amount of soybean fields surrounding the apiaries. Honey bee waggle dances recorded during soybean bloom revealed that honey bees preferred soybean fields for foraging over other habitat types. With these results, future research efforts aimed at enhancing mutual interactions between soybeans and honey bees may represent an unexplored pathway for increasing soybean production while supporting honey bees and other pollinators in the surrounding landscape., (© The Author(s) 2022. The majority of plant protein in the world's food supply is derived from soybean (Glycine max). Soybean is a key protein source for global animal feed and is incorporated into plant-based foods for people, including meat alternatives. Soybean protein content is genetically variable and is usually inversely related to seed oil content. ABI3-interacting protein 2 (AIP2) is an E3-RING ubiquitin ligase that targets the seed-specific transcription factor ABI3. Silencing both soybean AIP2 genes (AIP2a and AIP2b) by RNAi enhanced seed protein content by up to seven percentage points, with no significant decrease in seed oil content. The protein content enhancement did not alter the composition of the seed storage proteins. Inactivation of either AIP2a or AIP2b by a CRISPR-Cas9-mediated mutation increased seed protein content, and this effect was greater when both genes were inactivated. Transactivation assays in transfected soybean hypocotyl protoplasts indicated that ABI3 changes the expression of glycinin, conglycinin, 2S albumin, and oleosin genes, indicating that AIP2 depletion increased seed protein content by regulating activity of the ABI3 transcription factor protein. These results provide an example of a gene-editing prototype directed to improve global food security and protein availability in soybean that may also be applicable to other protein-source crops., (© The Author(s) 2022. Spectral remote sensing has the potential to improve scouting and management of soybean aphid (Aphis glycines Matsumura), which can cause yield losses of over 40% in the North Central Region of the United States. We used linear support vector machines (SVMs) to determine 1) whether hyperspectral samples could be classified into treat/no-treat classes based on the economic threshold (250 aphids per plant) and 2) how many wavelengths or features are needed to generate an accurate model without overfitting the data. A range of aphid infestation levels on soybean was created using caged field plots in 2013, 2014, 2017, and 2018 in Minnesota and in 2017 and 2018 in Iowa. Hyperspectral measurements of soybean canopies in each plot were recorded with a spectroradiometer. SVM training and testing were performed using 15 combinations of normalized canopy reflectance at wavelengths of 720, 750, 780, and 1,010 nm. Pairwise Bonferroni-adjusted t-tests of Cohen's kappa values showed four wavelength combinations were optimal, namely model 1 (780 nm), model 2 (780 and 1,010 nm), model 3 (780, 1,010, and 720 nm), and model 4 (780, 1,010, 720, and 750 nm). Model 2 showed the best overall performance, with an accuracy of 89.4%, a sensitivity of 81.2%, and a specificity of 91.6%. Phytophthora effector PSR1 suppresses small RNA (sRNA)-mediated immunity in plants, but the underlying mechanism remains unknown. Here, we show that Phytophthora suppressor of RNA silencing 1 (PSR1) contributes to the pathogenicity of Phytophthora sojae and specifically binds to three conserved C-terminal domains of the eukaryotic PSR1-Interacting Protein 1 (PINP1). PINP1 encodes PRP16, a core pre-mRNA splicing factor that unwinds RNA duplexes and binds to primary microRNA transcripts and general RNAs. Intriguingly, PSR1 decreased both RNA helicase and RNA-binding activity of PINP1, thereby dampening sRNA biogenesis and RNA metabolism. The PSR1-PINP1 interaction caused global changes in alternative splicing (AS). A total of 5,135 genes simultaneously exhibited mis-splicing in both PSR1-overexpressing and PINP1-silenced plants. AS upregulated many mRNA transcripts that had their introns retained. The high occurrence of intron retention in AS-induced transcripts significantly promoted Phytophthora pathogen infection in Nicotiana benthamiana, and this might be caused by the production of truncated proteins. Aim: The objective of this research was to screen fungal isolates originally isolated from cotton plants and measure their effects on the interactions between soybean and two aboveground pests (cabbage looper; Trichoplusia ni and soybean looper; Chrysodeixis includens) as well as a belowground pest (soybean cyst nematode; Heterodera glycines)., Methods and Results: For aboveground pests, we measured the leaf area consumed and larval weight. For our belowground pest tests, we measured shoot height, shoot fresh weight, root fresh weight and number of cysts. Out of the 50 fungal isolates tested, we tested 30 fungi in the interaction with cabbage looper, 36 for soybean looper, 41 for soybean cyst nematode. We tested 23 isolates against all pests and identified multiple isolates that significantly changed the response of pests on inoculated soybean plants versus controls., Conclusions: We identified three fungal isolates that significantly reduced both leaf area consumed aboveground by caterpillars and number of cysts produced belowground by nematodes. These isolates were an Epicoccum italicum, a Chaetomium undulatum and a Stemphylium majusculum., Significance and Impact of Study: Overall, this study provides important insights into plant-fungal interactions and their effect on both above- and belowground pests. Winter annual cover crops can be planted before soybean in Mississippi for many agronomic reasons. Incorporating winter annual cover crops into soybean production changes the seasonal hosts within fields. Some studies suggest that reducing tillage and using diverse species of cover crops can increase arthropod diversity and predator activity. Neonicotinoid seed treatments are often implemented to combat early season insect pests in soybean that follow cover crops, but negative effects on the environment such as reductions in biodiversity are often attributed to these compounds. We conducted an experiment to measure the effects on the diversity of the soybean epigeal and foliar communities when incorporating cover crops as well as insecticidal seed treatments into Mississippi soybean growing systems. Our results showed that legume cover crops had significant impacts on the epigeal community diversity of soybean planted behind them. These cover crops, especially hairy vetch, supported a more diverse foliar community before termination. To prevent increases in herbivorous arthropods, neonicotinoid seed treatments can be used without affecting epigeal predators such as beetles, ants, and spiders. The neonicotinoid seed treatments affected arthropod diversity, but the reductions were mainly caused by decreases in herbivorous pest insects that fed on treated soybean plants., (© The Author(s) 2022.
