A simple and specific liquid chromatographic method was developed for determination of clopidol in chicken tissues. Samples were extracted with acetonitrile. The extracts were cleaned up on an alumina column and an anion exchange column. The clopidol was separated on a column (30 cm × 3.9 mm) of μBondapak C18 (10 μm) by using acetonitrile–water (20 + 80, v/v) as mobile phase, and determined quantitatively at 270 nm. Recoveries were 86.0–97.6%, with relative standard deviations of 2.14–9.42% at 0.010–2.0 mg/kg from 4 spiked matrixes of chicken muscle, egg, liver, and kidney. The limit of detection was 0.005 mg/kg. Compared with the modified AOAC gas chromatographic method, the present method is simple and fast to operate. Its results are accurate and reliable, making it favorable for environmental protection and meeting requirements for human safety. Thus, it is suitable for routine analysis of large quantities of samples.

The white-rot fungus Marasmius quercophilus C30 is able to produce several laccases. The proportion of the enzymes produced depends on culture conditions. On malt medium, LAC1 was produced continuously over the 14 days of the cultivation period and was the only activity detectable. Copper increased total laccase activity by a factor 10 and induced the transient expression of one or more extra laccases in the culture medium. A combination of copper and p-hydroxybenzoic acid made it possible to extend the expression of induced laccase activities over the cultivation period and to reach a maximum activity 30 times higher than in non-induced culture. Extracellular laccases produced in this last condition were eluted as four peaks on an anion exchange column and were partially characterized.

Quantitation of o- and p-sulfamoylbenzoic acid residues in saccharin and its sodium salt is achieved by a method comprising methanolic extraction and high-performance ion exchange chromatography. A commercially available anion exchange column was employed with an aqueous buffered (pH 9.2) mobile phase. As little as 80 ppm of the ortho-isomer and 25 ppm of the para-isomer can be accurately determined. The levels of detectability (2 times noise) are estimated as 8 ppm (0.16 μg on column) and 2.5 ppm (0.05 μg on column), respectively. Recoveries from saccharin ranged from 92.7 to 96.5% (ortho) and from 92.2 to 103.3% (para). Recoveries from the sodium salt ranged from 93.1 to 104.4% (ortho) and from 93.5 to 97.8% (para). Of 9 other potential saccharin impurities tested separately, only one was found to interfere slightly in the chromatographic part of the procedure.

A rapid procedure is described for determining total iron-binding capacity (TIBC) by use of a small, disposable resin column. Serum is saturated with excess ferric citrate, and iron not bound to protein is then removed by passing the mixture through the column of anion-exchanger. Transferrin-bound iron in the effluent, the TIBC, is determined colorimetrically (AutoAnalyzer), with 2,4,6-tripyridyl-1,3,5-triazine as color reagent. The percentage saturation with iron may be obtained from colorimetric measurement of original serum iron and the TIBC, or by radioactive counting if 59Fe is incorporated in the ferric citrate used to saturate the serum. Our method is compared with the MgCO3 precipitation technique for the removal of excess iron after saturation and to an ultrafiltration technique in which protein-bound iron is retained by a membrane and the filtrate analyzed for unbound iron, thus obviating absorbents in the determination of TIBC. Results from the resin-column and ultrafiltration procedures correlated well, whereas those from the MgCO3 method were consistently lower and more erratic.

The miniature anion-exchange/centrifugation (AEC) technique, developed originally for the detection of low parasitaemias in laboratory rodents, was adapted to field use for the diagnosis of trypanosomiasis in man in Africa and was tested in The Gambia. During this field study it was found that microfilariae of Dipetalonema perstans could also pass through the anion exchange column and appear in the centrifugate as 'medusa heads'. One locality-group (Mansafa Bolon) showed a generally higher prevalence and prevalences in women over 40 years old were higher than in the corresponding male groups in every locality. The potential usefulness of this technique in epidemiological studies of filariasis and ways of improving the accuracy of numerical estimates are briefly discussed.
