The closely related bacterial species Bacillus cereus and Bacillus weihenstephanensis are adapted to the mesophilic and the psychrotrophic temperature range, respectively. While B. cereus strains are associated with foodborne diseases, B. weihenstephanensis strains are so far not, although similar virulence genes are found in both species. Our investigations show that both species were virulent in the insect model, Galleria mellonella, following infection via oral and haemocoel routes. However, virulence of B. weihenstephanensis was much higher at 15 °C than at 37 °C. Furthermore, a temperature-dependent difference between the species was seen in a cell culture cytotoxicity assay. In summary, our results demonstrate for the first time virulence of B. weihenstephanensis strains in an in vivo model. In addition, we found that G. mellonella is a useful model for studies of the psychrotolerant species of the B. cereus group, suggesting that insects might be an ecological growth niche for several members of this bacterial group.

Individual components (P51 and P42) from the crystal toxin (Bin) of Bacillus sphaericus were used for in vitro binding competition experiments with brush border membranes (BBMFs) from Culex pipiens and Anopheles gambiae larval midguts. P51 competed for the Bin binding site with a similar affinity to the Bin toxin, on both BBMFs. For C. pipiens, P42 bound non-specifically until P51 was added with maximum binding of P42 at a molar ratio of each component. The binding of P42 was much greater on A. gambiae BBMFs and the presence of either P51 or P42 enhanced the binding of the other component, with highest binding when a mole to mole ratio of each protein was supplied.

A field-collected population of Crlex pipietls quinquefasciatus larvae from an urban area of Recife (Brazil), which has been treated for 2 yr with Bacillus sphaericus, was found to be ≈10-fold less susceptible than control field populations. Comparative in vitro binding experiments between the radiolabeled toxin from B. sphaericus and isolated midgut brush border membranes from the treated and the untreated larval populations, indicated no change in the affinity of the midgut receptor of the treated population for the toxin, and only a slight decrease in the receptor concentration.
