Extreme climate and weather events, such as a drought, hurricanes, or ice storms, can strongly imprint ecosystem processing and may alter ecosystem structure. Ecosystems in extreme environments are particularly vulnerable because of their adaptation to severe limitations in energy, water, or nutrients. The vulnerability can be expressed as a relatively long-lasting ecosystem response to a small or brief change in environmental conditions. Such an event occurred in Antarctica and affected two vastly different ecosystems: a marine-dominated coastal system and a terrestrial polar desert. Both sites experienced winds that warmed air temperatures above the 0°C threshold, resulting in extensive snow and ice melt and triggering a series of cascading effects through the ecosystems that are continuing to play out more than a decade later. We examined levels of genetic variability within and among populations of three Antarctic springtail species (Arthropoda: Collembola) and tested the hypothesis that genetic divergences occur among glacially-isolated habitats. The study was conducted in southern Victoria Land, Ross Dependency, Antarctica, and samples were collected from locations in the vicinity of the Mackay Glacier. We analyzed mt DNA ( cytochrome c oxidase subunit I; COI) sequence variability for 97 individuals representing three species ( Gomphiocephalus hodgsoni, N = 67; Cryptopygus nivicolus, N = 20; and Antarcticinella monoculata, N = 8). Haplotype diversity and genetic divergences were calculated and used to indicate population variability and also to infer divergence times of isolated populations using molecular clock estimates. Two of the three species showed high levels of genetic divergence. Gomphiocephalus hodgsoni, a widespread and common species, showed 7.6% sequence divergence on opposite sides of the Mackay Glacier. The more range restricted C. nivicolus showed 4.0% divergence among populations. The third species, A. monoculata, was found in only one location. Molecular clock estimates based on sequence divergences suggest that populations separated within the last 4 Mya. We conclude that habitat fragmentation resulting from Pliocene (5 Mya) and Pleistocene (2 Mya to 10 Kya) glaciations has promoted and maintained high levels of diversity among isolated springtail populations on relatively small spatial scales. The region surrounding the Mackay Glacier is likely to have provided refugia for springtail populations during glacial maxima and remains an area of high genetic and species diversity for Collembola within the Ross Sea region. The pecan weevil, Curculio caryae (Horn), is a major pest of pecans in the Southeastern United States. Entomopathogenic nematodes and fungi are potential alternatives to chemical insecticides for C. caryae control. Our objective was to survey pecan orchards in the southeastern United States for entomopathogenic nematodes and fungi and determine the virulence of tile new isolates to C. caryae larvae. Soil was collected from 105 sites in 21 orchards in Arkansas, Georgia, Louisiana, and Mississippi. Entomopathogens were isolated by exposing soil to C. caryae and greater wax moth larvae, Galleria mellonella, (L.). We isolated entomopathogenic fungi and nematodes from 16 and 6 of the 21 orchards surveyed, respectively. The entomopathogenie fungi included Beauveria bassiana (Balsamo) Vuillemin and Metarhizium anisopliae (Metschnikoff) Sorokin, and nematodes included Heterorhabditis bacteriophora Poinar, Steinernema carpocapsae (Weiser), Steinernema glaseri (Steiner), and Steinernema rarum (Doucet). This is the first report of Steinernema rarum in the United States. Soil characteristics in orchards were analyzed for pH, organic matter, and nutrients; we detected a negative relationship between fungal occurrence and manganese levels in soil and a positive relationship between M. anisopliae occurrence and calcium or magnesium levels. In laboratory assays, virulence of 15 nematode and 22 fungal isolates to C. caryae larvae was tested in small plastic cups containing soil. Results indicated poor susceptibility of the C. caryae larvae to entomopathogenic nematodes. Several fungal isolates that caused significantly higher mortality in C. caryae larvae than other strains (including a commercial strain or B. bassiana) should be investigated further as potential control agents of C. caryae. The potential for entomopathogenic nematodes to control flies in cattle feedlots was determined by screening 40 strains representing 8 species of Heterorhabditis Poinar and 5 species of Steinernenw Travassos for virulence toward 3rd-instar house flies (maggots), Musca domestica L. None of the 22 strains of Heterorhabditis infecting maggots caused significant levels of mortality in a filter paper assay. Ten strains of Steinernema infected maggots, of which 7 strains (4 S. carpocapsae (Weiser), 2 S. feltiae (Filipjev), and 1 S. scapterisci Nguyen & Smart) caused significant mortality. Ten Heterorhabditis strains and 10 Steinernema strains successfully reproduced for =/>2 generationsin maggots. No difference was observed between 72-h survival of maggots and adult emergence. Six strains of Steinernema were selected for10 generations on maggots and then compared with unselected lines. No difference in pathogenicity between selected and unselected lines was observed. Two strains of S. feltiae, SN and UNK-36, and 2 of the best Heterorhabditis strains, H. bacteriophora Poinar OSWEGO and H. megidis Poinar, Jackson & Klein HF-85 were tested in a fresh bovine manure substrate. All 4 strains produced significant fly mortality in the manure substrate, although the S. feltiae strains had significantlylower LC50 values than did the Heterorhabditis spp. The most promising strain, S. feltiae SN, gave LC50 and LC99 values of 4 and 82 infective juveniles per maggot respectively. These doses were equivalent to 2.7 and 55 infective juveniles pergram of manure and 5.1 and 104 infective juveniles per square centimeter of surface area. Plectus murrayi is one of the most common and locally abundant invertebrates of continental Antarctic ecosystems. Because it is readily cultured on artificial medium in the laboratory and highly tolerant to an extremely harsh environment, P. murrayi is emerging as a model organism for understanding the evolutionary origin and maintenance of adaptive responses to multiple environmental stressors, including freezing and desiccation. The de novo assembled genome of P. murrayi contains 225.741 million base pairs and a total of 14,689 predicted genes. Compared to Caenorhabditis elegans, the architectural components of P. murrayi are characterized by a lower number of protein-coding genes, fewer transposable elements, but more exons, than closely related taxa from less harsh environments. We compared the transcriptomes of lab-reared P. murrayi with wild-caught P. murrayi and found genes involved in growth and cellular processing were up-regulated in lab-cultured P. murrayi, while a few genes associated with cellular metabolism and freeze tolerance were expressed at relatively lower levels. Preliminary comparative genomic and transcriptomic analyses suggest that the observed constraints on P. murrayi genome architecture and functional gene expression, including genome decay and intron retention, may be an adaptive response to persisting in a biotically simplified, yet consistently physically harsh environment.
