Purpose Of the Study: Genome-wide association studies have revealed an association of ADAMTS7 polymorphisms with the risk of cardiovascular diseases. Nonetheless, the role of ADAMTS7 polymorphisms on myocardial infarction (MI) risk remains poorly understood. Here, we aim to evaluate the effect of ADAMTS7 tag single nucleotide polymorphisms (SNPs) on individual susceptibility to MI.Study Design: Genotyping of the four tagSNPs (rs1994016, rs3825807, rs4380028 and rs7173743) was performed in 232 MI cases and 661 control subjects using PCR-ligase detection reaction (LDR) method. The association of these four tagSNPs with MI risk was performed with SPSS software.Results: Multivariate logistic regression analysis showed that ADAMTS7 tagSNP rs3825807 exhibited a significant effect on MI risk. Compared with the TT homozygotes, the CT genotype (OR1.93, 95% CI1.30to 2.85, Pc=0.004) and the combined CC/CT genotypes (OR1.70, 95% CI1.16 to 2.50, Pc=0.028) were statistically significantly associated with the increased risk for MI. Further stratified analysis revealed a more significant association with MI risk among older subjects, hypertensives, non-diabetics and patients with hyperlipidaemia. Consistently, the haplotype rs1994016T-rs3825807C containing rs3825807 C allele exhibited increased MI risk (OR1.52, 95% CI1.10 to 2.10, p=0.010). However, we did not detect any association of the other three tagSNPs with MI risk.Conclusions: Our finding suggest that ADAMTS7 tagSNP rs3825807 contributes to MI susceptibility in the Chinese Han population. During geophysical surveys, water layers may interfere with the detection of oil layers. In order to distinguish between oil and water layers in porous cracked media, research on the properties of the cracks, the oil and water layers, and their relation to pulsed neutron logging characteristics is essential. Using Hudson's crack theory, we simulated oil and water layers in a cracked porous medium with different crack parameters corresponding to the well log responses. We found that, in a cracked medium with medium-angle (40°-50°) cracks, the thermal neutron count peak value is higher and more sensitive than those in low-angle and high-angle crack environments; in addition, the thermal neutron density distribution shows more minimum values than in other cases. Further, the thermal neutron count and the rate of change for the oil layer are greater than those of the water layer, and the time spectrum count peak value for the water layer in middle-high-angle (40°-70°) cracked environments is higher than that of the oil layer. The thermal neutron density distribution sensitivity is higher in the water layer with a range of small crack angles (0°-30°) than in the oil layer with the same range of angles. In comparing the thermal neutron density distribution, thermal neutron count peak, thermal neutron density distribution sensitivity, and time spectrum maximum in the oil and water layers, we find that neutrons in medium-angle (40°-50°) cracked reservoirs aremore sensitive to deceleration and absorption than those in water layers; neutrons in approximately horizontal (0°-30°) cracked water layers are more sensitive to deceleration than those in reservoirs. Study Question: What are the effects of high-glucose concentrations on DNA methylation of human oocytes?Summary Answer: High-glucose concentrations altered DNA methylation levels of Peg3 and Adiponectin in human in vitro maturation oocytes.What Is Known Already: Maternal diabetes has a detrimental influence on oocyte quality including epigenetic modifications, as shown in non-human mammalian species.Study Design, Size, Duration: Immature metaphase I (MI) stage oocytes of good quality were retrieved from patients who had normal ovarian potential and who underwent ICSI in the Reproductive Medicine Center of People's Hospital of Zhengzhou University. MI oocytes were cultured in medium with different glucose concentrations (control, 10 mM and 15 mM) in vitro and 48 h later, oocytes with first polar body extrusion were collected to check the DNA methylation levels.Participants/materials, Setting, Methods: MI oocytes underwent in vitro maturation (IVM) at 37°C with 5% mixed gas for 48 h. Then the mature oocytes were treated with bisulfite buffer. Target sequences were amplified using nested or half-nested PCR and the DNA methylation status was tested using combined bisulfite restriction analysis (COBRA) and bisulfite sequencing (BS).Main Results and the Role Of Chance: High-glucose concentrations significantly decreased the first polar body extrusion rate. Compared to controls, the DNA methylation levels of Peg3 in human IVM oocytes were significantly higher in 10 mM (P < 0.001) and 15 mM (P < 0.001) concentrations of glucose. But the DNA methylation level of H19 was not affected by high-glucose concentrations in human IVM oocytes. RNA interference has been used to study insects' gene function and regulation. Glycogen synthase (GS) and glycogen phosphorylase (GP) are two key enzymes in carbohydrates' conversion in insects. Glycogen content and GP and GS gene expression in several tissues and developmental stages of the Brown planthopper Nilaparvata lugens Stål (Hemiptera: Delphacidae) were analyzed in the present study, using quantitative reverse-transcription polymerase chain reaction to determine their response to double-stranded trehalases (dsTREs), trehalose-6-phosphate synthases (dsTPSs), and validamycin injection. The highest expression of both genes was detected in the wing bud, followed by leg and head tissues, and different expression patterns were shown across the developmental stages analyzed. Glycogen content significantly decreased 48 and 72h after dsTPSs injection and 48h after dsTREs injection. GP expression increased 48h after dsTREs and dsTPSs injection and significantly decreased 72h after dsTPSs, dsTRE1-1, and dsTRE1-2 injection. GS expression significantly decreased 48h after dsTPS2 and dsTRE2 injection and 72h after dsTRE1-1 and dsTRE1-2 injection. GP and GS expression and glycogen content significantly decreased 48 h after validamycin injection. The GP activity significantly decreased 48h after validamycin injection, while GS activities of dsTPS1 and dsTRE2 injection groups were significantly higher than that of double-stranded GFP (dsGFP) 48h after injection, respectively. A rapid and sensitive high performance liquid chromatography method was developed and validated for the determination of enantiomeric purity of 1-styrene-3-hexyl propynol. Baseline separation with resolution greater than 2.0 was achieved within 17 min on a CHIRALCEL OD-H (25 cm × 0.46 cm; particle size 5 µm) column using n-hexane:isopropanol (95:5 v/v) as mobile phase at a flow rate of 1mL·min-1 and 25°C as column temperature. The analytes were detected by UV absorbance at 210 nm. The effects of column, composition of mobile phase, temperature and flow rate on the chromatography were investigated. The method was extensively validated and proved to be robust. The recoveries were in the range of 99.6-102.6% with relative standard \deviation <0.32%. The limit of detection and limit of quantitation for (S)-1-styrene-3-hexyl propynol were 0.0041 and 0.22 µg·mL-1, and for (R)-1-styrene-3-hexyl propynol were 0.0036 and 0.19 µg·mL-1, respectively. This method was proved to be accurate, stable, rapid and sensitive for the determination of enantiomeric purity of 1-styrene-3-hexyl propynol and its homologs. [ABSTRACT FROM AUTHOR]

Background: This randomized, open trial compared regimens including 2 doses (2D) of human papillomavirus (HPV) 16/18 AS04-adjuvanted vaccine in girls aged 9-14 years with one including 3 doses (3D) in women aged 15-25 years.Methods: Girls aged 9-14 years were randomized to receive 2D at months 0 and 6 (M0,6; (n = 550) or months 0 and 12 (M0,12; n = 415), and women aged 15-25 years received 3D at months 0, 1, and 6 (n = 482). End points included noninferiority of HPV-16/18 antibodies by enzyme-linked immunosorbent assay for 2D (M0,6) versus 3D (primary), 2D (M0,12) versus 3D, and 2D (M0,6) versus 2D (M0,12); neutralizing antibodies; cell-mediated immunity; reactogenicity; and safety. Limits of noninferiority were predefined as <5% difference in seroconversion rate and <2-fold difference in geometric mean antibody titer ratio.Results: One month after the last dose, both 2D regimens in girls aged 9-14 years were noninferior to 3D in women aged 15-25 years and 2D (M0,12) was noninferior to 2D (M0,6). Geometric mean antibody titer ratios (3D/2D) for HPV-16 and HPV-18 were 1.09 (95% confidence interval, .97-1.22) and 0.85 (.76-.95) for 2D (M0,6) versus 3D and 0.89 (.79-1.01) and 0.75 (.67-.85) for 2D (M0,12) versus 3D. The safety profile was clinically acceptable in all groups.Conclusions: The 2D regimens for the HPV-16/18 AS04-adjuvanted vaccine in girls aged 9-14 years (M0,6 or M0,12) elicited HPV-16/18 immune responses that were noninferior to 3D in women aged 15-25 years.Clinical Trials Registration: NCT01381575. Background. MicroRNAs (miRNAs) are small, noncoding RNAs that regulate gene expression at posttranscriptional level. H. pylori is a major human pathogenic bacterium in gastric mucosa. To date, the role of miRNAs in response to H. pylori infection has not been explored. Methods. The expression profile of cellular miRNAs during H. pylori infection was analyzed by usingmicroarray and quantitative reverse-transcriptase polymerase chain reaction. The potential target of miR-155 was identified by luciferase assay and Western blot. Promoter analysis and inhibitor experiment were used to investigate the pathway involved in the induction of miR-155. Examination of miR-155 function was performed by overexpression and inhibition of miR-155. Results. H. pylori was able to increase the miR-155 expression in gastric epithelial cell lines and gastric mucosal tissues, and nuclear factor-κB (NF-κB) and activator protein-1 (AP-1) pathway were required for the induction of miR-155. miR-155 may down-regulate IκB kinase ϵ, Sma- and Mad-related protein 2 (SMAD2), and Fasassociated death domain protein. Furthermore, the overexpression of miR-155 negatively regulated the release of interleukin-8 and growth-related oncogene-α. Conclusions. This study provides the first description of increased expression of miR-155 in H. pylori infection, and miR-155 may function as novel negative regulator that help to fine-tune the inflammation response of H. pylori infection. We examined the epidemiological profile of rotavirus infection among children hospitalized for diarrhea in Taiwan, to assess the burden of this disease. From 1 April 2001 through 31 March 2003, children <5 years old with gastroenteritis admitted to 4 sentinel hospitals were enrolled in a surveillance study and had stool specimens tested for the presence of rotavirus, enteric adenovirus, and the bacterial pathogens for which routine screening is performed. For 52% of patients, a recognized enteric pathogen was identified, including rotavirus (43% of patients), bacteria (11%), enteric adenovirus (2.5%), and a mixture of pathogens (3.9%). Rotavirus was detected year-round, but great month-to-month variability made it difficult to identify a distinct seasonal pattern. Rotavirus disease was most common among children 7-23 months old, but the rate of rotavirus detection varied little between the youngest and oldest age groups. The novel strain P[8]G9 was detected most commonly (37% of strains), followed by strains P[8]G1 (31%), P[4]G2 (10%), P[8]G3 (9.3%), and P[8]G4 (3.7%). Rotavirus infection is the most important cause of diarrhea among hospitalized children in Taiwan, and a rotavirus vaccination program for young children might significantly reduce this problem.
