Venom systems are complex traits that have independently emerged multiple times in diverse plant and animal phyla. Within each venomous lineage there typically exists interspecific variation in venom composition where several factors have been proposed as drivers of variation, including phylogeny and diet. Understanding these factors is of broad biological interest and has implications for the development of antivenom therapies and venom-based drug discovery. Because of their high species richness and the presence of several major evolutionary prey shifts, venomous marine cone snails (genus Conus) provide an ideal system to investigate drivers of interspecific venom variation. Here, by analyzing the venom gland expression profiles of ∼3,000 toxin genes from 42 species of cone snail, we elucidate the role of prey-specific selection pressures in shaping venom variation. By analyzing overall venom composition and individual toxin structures, we demonstrate that the shifts from vermivory to piscivory in Conus are complemented by distinct changes in venom composition independent of phylogeny. In vivo injections of venom from piscivorous cone snails in fish further showed a higher potency compared with venom of nonpiscivores demonstrating a selective advantage. Together, our findings provide compelling evidence for the role of prey shifts in directing the venom composition of cone snails and expand our understanding of the mechanisms of venom variation and diversification. Lymantria dispar L. is an invasive, non-native defoliating Lepidopteran established in North America that feeds on forest and urban trees. While many products are available to manage L. dispar post-emergence, few exist to prevent egg hatch when applied to egg masses. Here, we present the results of 3 separate experiments aimed at determining the efficacy of pre-emergent insecticides against L. dispar egg hatch. We found that the labeled rate (1:1) of Golden Pest Spray Oil (GPSO; AI: 93% soybean oil) can prevent L. dispar larvae from emerging in both field and lab assays. In large public spaces, we found that this treatment was ineffective at preventing L. dispar emergence or defoliation. Acelepryn (AI: 18.4% chlorantraniliprole) resulted in some suppression of egg hatch at a very low rate (.06 ml/ 3.8 liter) in both lab and field settings and the efficacy of higher rates should be further investigated. We also tested GPSO against Lepitect (97.4% acephate) in a public area that also received a Foray 48B (12.65% Bacillus thuringiensis, subsp. kurstaki) aerial application. On large oak trees in public areas, GPSO and Lepitect were not effective at reducing defoliation. Dormant pesticide applications generally reduce the risk of affecting negatively predator and parasitoid communities and are therefore desirable. Lymantria dispar pre-egg hatch applications will not work in every situation but should be considered as part of an integrated pest management (IPM) strategy for individual homeowner trees where thorough coverage can be obtained. Somatostatin and its related peptides (SSRPs) form an important family of hormones with diverse physiological roles. The ubiquitous presence of SSRPs in vertebrates and several invertebrate deuterostomes suggests an ancient origin of the SSRP signaling system. However, the existence of SSRP genes outside of deuterostomes has not been established, and the evolutionary history of this signaling system remains poorly understood. Our recent discovery of SSRP-like toxins (consomatins) in venomous marine cone snails (Conus) suggested the presence of a related signaling system in mollusks and potentially other protostomes. Here, we identify the molluscan SSRP-like signaling gene that gave rise to the consomatin family. Following recruitment into venom, consomatin genes experienced strong positive selection and repeated gene duplications resulting in the formation of a hyperdiverse family of venom peptides. Intriguingly, the largest number of consomatins was found in worm-hunting species (>400 sequences), indicating a homologous system in annelids, another large protostome phylum. Consistent with this, comprehensive sequence mining enabled the identification of SSRP-like sequences (and their corresponding orphan receptor) in annelids and several other protostome phyla. These results established the existence of SSRP-like peptides in many major branches of bilaterians and challenge the prevailing hypothesis that deuterostome SSRPs and protostome allatostatin-C are orthologous peptide families. Finally, having a large set of predator–prey SSRP sequences available, we show that although the cone snail's signaling SSRP-like genes are under purifying selection, the venom consomatin genes experience rapid directional selection to target receptors in a changing mix of prey. Invasions by insects introduced via international trade continue to cause worldwide impacts. Surveillance programs using traps baited with host volatiles and pheromones can detect incursions of nonnative species. We report on two experiments executed to determine if attractants for several insect species can be combined without compromising trap catches and detection ability of target species. In the first experiment, we tested the effect of bark beetle pheromones (plus α-pinene) and trap contact with foliage on trap catches of the brown marmorated stink bug Halyomorpha halys Stål (Hemiptera: Pentatomidae) in traps baited with a mixture of bisabolenes and methyl (E,E,Z)-2,4,6-decatrienoate. Trap capture of H. halys adults was greater in traps not in contact with foliage, and the bark beetle pheromones ipsenol and ipsdienol did not affect trap capture of H. halys. In the second experiment, we tested the effects of multi-lure interactions among the primary host attractants α-pinene and ethanol, and the pheromones monochamol, ipsenol, ipsdienol, lanierone, and the H. halys compounds, on trap captures of various forest and agricultural insect pests. Specifically, we targeted Monochamus spp. (Coleoptera: Cerambycidae), Ips spp. (Coleoptera: Scolytinae) and H. halys. We found that a combination of all lures did not catch significantly lower numbers of Monochamus carolinensis Olivier, Monochamus scutellatus Say (Coleoptera: Cerambycidae), and Ips pini Say (Coleoptera: Scolytidae) than lure combinations missing components although removal of both lanierone and ipsdienol somewhat increased catches of Ips grandicollis Eichhoff (Coleoptera: Curculionidae). Our results support the use of traps baited with a full combination of these attractants in surveillance programs. Sirex nigricornis F. (Hymenoptera: Siricidae) is an innocuous pine-inhabiting woodwasp native to eastern North America, utilizing dead or dying pine trees as hosts. Although S. nigricornis F. does not cause economic damage, a closely related species, Sirex noctilio, was discovered in New York in 2004 and has continually spread throughout the northeastern United States and southern Canada, threatening the multi-billion-dollar pine timber industry of the southeastern United States and raising interest about potential interactions with native woodwasps and associated mortality agents. A non-sterilizing strain of the biological control agent, Deladenus siricidicola Bedding (Tylenchida: Neotylenchidae), was introduced along with S. noctilio but is not inhibiting the spread or establishment of S. noctilio. A North American congener, Deladenus proximus Bedding, has been recently isolated from S. noctilio and shows promise as a biological control agent. To better understand the potential of D. proximus as a control agent for S. noctilio, we measured and dissected nearly 1,200 S. nigricornis females from Arkansas and Mississippi and evaluated differences among collection location with regard to nematode virulence, woodwasp body size, and egg load. Body size and egg load were related to collection location, and nematode infestation resulted in significantly smaller females who produced significantly fewer eggs. Female woodwasps, especially those collected in Arkansas, were often fully sterilized by nematodes, and a higher percent sterilization was inversely related to body size and fewer eggs. The United States has a rich fauna of native Siricidae (wood wasps), but they are rarely studied because they have limited economic impact. In 2004, a non-native wood-boring pest, Sirex noctilio F., was found established in North America. Because S. noctilio is an economically important pest in pine plantations throughout the Southern Hemisphere, interest in the ecology of American native wood wasp populations has increased. A study was conducted during fall 2011 to investigate the effects of forest stand type and characteristics on native wood wasp abundance, and to describe their flight phenology in northeastern Mississippi. In total, 609 native wood wasps were captured, consisting of 608 Sirex nigricornis F. and one Urocerus cressoni Norton. There were significant treatment and location effects that influenced wood wasp abundance. The flight period of wood wasps captured in our study (October-December) was similar to studies in the southeastern United States, but differed from results in Minnesota and the northeastern United States (June-October). Wood wasp abundance was significantly correlated with higher basal area, smaller tree diameter at breast height, and shorter trees, all indicators of forest stand stress. It appears proper silvicultural management of pine plantations may reduce native wood wasp population abundance in the southeastern United States, as it does to S. noctilio in the Southern Hemisphere. We recently showed that genes at 3 loci account for the majority of variation in canine fur. Allelic variation at genes controlling length of fur, texture, and curl is responsible for the striking phenotypic variety observed among purebred dogs in the United States today. In this paper, we investigate the phenomenon of “improper coat” (IC) or a coat that is not typical of the breed. IC is occasionally observed among specific breeds, such as the Portuguese Water Dog (PWD), and is characterized by short hair on the head, face, and lower legs, rather than a thick and even coat covering the whole body. The IC is reminiscent of that observed on the curly or flat-coated retriever, thus making such dogs unable to compete effectively in conformation events. We have found that the presence of the wild-type allele, rather than the expected variant allele at the R-spondin 2 (RSPO2) gene, accounts for this phenotype. The development of a genetic test that distinguishes these 2 allelic types would allow breeders to easily avoid producing PWD with ICs. [ABSTRACT FROM PUBLISHER]

Traits that have been stringently selected to conform to specific criteria in a closed population are phenotypic stereotypes. In dogs, Canis familiaris, such stereotypes have been produced by breeding for conformation, performance (behaviors), etc. We measured phenotypes on a representative sample to establish breed stereotypes. DNA samples from 147 dog breeds were used to characterize single nucleotide polymorphism allele frequencies for association mapping of breed stereotypes. We identified significant size loci (quantitative trait loci [QTLs]), implicating candidate genes appropriate to regulation of size (e.g., IGF1, IGF2BP2 SMAD2, etc.). Analysis of other morphological stereotypes, also under extreme selection, identified many additional significant loci. Behavioral loci for herding, pointing, and boldness implicated candidate genes appropriate to behavior (e.g., MC2R, DRD1, and PCDH9). Significant loci for longevity, a breed characteristic inversely correlated with breed size, were identified. The power of this approach to identify loci regulating the incidence of specific polygenic diseases is demonstrated by the association of a specific IGF1 haplotype with hip dysplasia, patella luxation, and pacreatitis. Phenotypic stereotypes are traits, often polygenic, that have been stringently selected to conform to specific criteria. In dogs, Canis familiaris, stereotypes result from breed standards set for conformation, performance (behaviors), etc. As a consequence, phenotypic values measured on a few individuals are representative of the breed stereotype. We used DNA samples isolated from 148 dog breeds to associate SNP markers with breed stereotypes. Using size as a trait to test the method, we identified six significant quantitative trait loci (QTL) on five chromosomes that include candidate genes appropriate to regulation of size (e.g., IGEI, IGF2BP2 SMAD2, etc.). Analysis of other morphological stereotypes, also under extreme selection, identified many additional significant loci. Less well-documented data for behavioral stereotypes tentatively identified loci for herding, pointing, boldness, and trainability. Four significant loci were identified for longevity, a breed characteristic not under direct selection, but inversely correlated with breed size. The strengths and limitations of the approach are discussed as well as its potential to identify loci regulating the within-breed incidence of specific polygenic diseases. [ABSTRACT FROM AUTHOR]

Genetic variation in functionally integrated skeletal traits can be maintained over 10 million years despite bottlenecks and stringent selection. Here, we describe an analysis of the genetic architecture of the canid axial skeleton using populations of the Portuguese Water Dog Canis familiaris) and silver fox (Vulpes vulpes). Twenty-one skeletal metrics taken from radiographs of the forelimbs and hind limbs of the fox and dog were used to construct separate anatomical principal component (PC) matrices of the two species. In both species, 15 of the 21 PCs exhibited significant heritability, ranging from 25% to 70%. The second PC, in both species, represents a trade-off in which limb-bone width is inversely correlated with limb-bone length. PC2 accounts for approximately 15% of the observed skeletal variation, ∼30% of the variation in shape. Many of the other significant PCs affect very small amounts of variation (e.g., 0.2-2%) along trade-off axes that partition function between the forelimbs and hind limbs. These PCs represent shape axes in which an increase in size of an element of the forelimb is associated with a decrease in size of an element of the hind limb and vice versa. In most cases, these trade-offs are heritable in both species and genetic loci have been identified in the Portuguese Water Dog for many of these. These PCs, present in both the dog and the fox, include ones that affect lengths of the forelimb versus the hind limb, length of the forefoot versus that of the hind foot, muscle moment (i.e., lever) arms of the forelimb versus hind limb, and cortical thickness of the bones of the forelimb versus hind limb. These inverse relationships suggest that genetic regulation of the axial skeleton results, in part, from the action of genes that influence suites of functionally integrated traits. Their presence in both dogs and foxes suggests that the genes controlling the regulation of these PCs of the forelimb versus hind limb may be found in other tetrapod taxa. [ABSTRACT FROM AUTHOR]

Four erythrocyte variables (erythrocyte count, hemoglobin, mean cell volume, packed cell volume), 14 serum variables (alanine transferase, albumin, alkaline phosphatase, calcium, chloride, cholesterol, creatinine, glucose, phosphorus, potassium, sodium, total protein, triglycerides, urea nitrogen), and 7 venous acid base variables (base excess, bicarbonate, carbon dioxide partial pressure, oxygen partial pressure, oxygen saturation, pH, and total carbon dioxide) were evaluated for heritability in domestic cats (Felis catus). Values used for individual cats were expressed as the mean over all lifetime measurements, using 444-530 animals for clinical chemistry, 629 animals for acid-base, and 564 animals for erythrocyte metrics. Gender and age at death (where applicable) also were evaluated for correlation with variables. Heritabilities for clinical chemistry, acid-base, and erythrocyte variables ranged, respectively, from 0.13 to 0.78, from 0.23 to 0.59, and from 0.41 to 0.69 (P < 0.05). This result indicates that serum variability has a genetic basis and is segregating in this feline population. These findings may have important implications in both research and clinical medicine. The first genetic transcript map of the soybean genome was created by mapping one SNP in each of 1141 genes in one or more of three recombinant inbred line mapping populations, thus providing a picture of the distribution of genic sequences across the mapped portion of the genome. Single-nucleotide polymorphisms (SNPs) were discovered via the resequencing of sequence-tagged sites (STSs) developed from expressed sequence tag (EST) sequence. From an initial set of 9459 polymerase chain reaction primer sets designed to a diverse set of genes, 4240 STSs were amplified and sequenced in each of six diverse soybean genotypes. In the resulting 2.44 Mbp of aligned sequence, a total of 5551 SNPs were discovered, including 4712 single-base changes and 839 indels for an average nucleotide diversity of θ = 0.000997. The analysis of the observed genetic distances between adjacent genes vs. the theoretical distribution based upon the assumption of a random distribution of genes across the 20 soybean linkage groups clearly indicated that genes were clustered. Of the 1141 genes, 291 mapped to 72 of the 112 gaps of 5-10 cM in the preexisting simple sequence repeat (SSR)-based map, while 111 genes mapped in 19 of the 26 gaps >10 cM. The addition of 1141 sequence-based genic markers to the soybean genome map will provide an important resource to soybean geneticists for quantitative trait locus discovery and map-based cloning, as well as to soybean breeders who increasingly depend upon marker-assisted selection in cultivar improvement.
