The Mediterranean fruit fly (medfly), Ceratitis capitata (Wiedemann), one of the most important invasive pests of fresh fruits and vegetables from the coastal Mediterranean habitats, is expanding its current geographic distribution to cooler more temperate areas of Europe. Every year since 2010 the fly is detected in the area of Vienna, Austria. However, whether it can establish permanent populations is not known. In this current paper, the capacity of C. capitata to overwinter in Vienna, Austria (48.1° northern latitude) was studied over 2 consecutive winter seasons (2020–2022). Overwintering trials with different life stages (larva, pupa, and adult) of C. capitata were performed in the open field and in the protected environment of a basement without a heating system. Control flies were kept under constant conditions in a climate chamber (25 °C, 60% RH, 14:10 L:D). Our data showed that no life stage of the Mediterranean fruit fly was able to survive the Austrian winter in the open field. However, in the protected environment C. capitata outlived the winter months in all studied life stages at least in small numbers and several surviving females were able to lay eggs at the time of the following fruiting season. Human African trypanosomiasis (HAT), or sleeping sickness, continues to be a major threat to human health in 36 countries throughout sub-Saharan Africa with up to 60 million people at risk. Over the last decade, there have been several advances in this area, some of which are discussed in this overview. Due to the concerted efforts of several bodies, including better identification and treatment of cases and improved tsetse fly vector control, the number of cases of HAT has declined dramatically. The clinical heterogeneity of HAT has also been increasingly recognized, and the disease, while usually fatal if untreated or inadequately treated, does not always have a uniformly fatal outcome. Improved methods of HAT diagnosis have now been developed including rapid diagnostic tests. Novel drug treatment of HAT has also been developed, notably nifurtimox–eflornithine combination therapy (NECT) for late-stage Trypanosoma brucei gambiense , oral fexinidazole for early and the early component of the late-stage of T.b. gambiense , and the new oral compounds of the oxaborole group, which have shown considerable promise in field trials. Advances in HAT neuropathogenesis have been steady, though largely incremental, with a particular focus on the role of the blood–brain barrier in parasite entry into the central nervous system and the relevant importance of both innate and adaptive immunity. While the World Health Organization goal of elimination of HAT as a public health problem by 2020 has probably been achieved, it remains to be seen whether the second more ambitious goal of interruption of transmission of HAT by 2030 will be attained. Werner syndrome (WS) is a rare genetic disease in humans, caused by mutations in the WRN gene that encodes a protein containing helicase and exonuclease domains. WS is characterized by symptoms of accelerated aging in multiple tissues and organs, involving increased risk of cancer, heart failure, and metabolic dysfunction. These conditions ultimately lead to the premature mortality of patients with WS. In this study, using the null mutant flies (WRNexo Δ ) for the gene WRNexo (CG7670), homologous to the exonuclease domain of WRN in humans, we examined how diets affect the lifespan, stress resistance, and sleep/wake patterns of a Drosophila model of WS. We observed that dietary restriction (DR), one of the most robust nongenetic interventions to extend lifespan in animal models, failed to extend the lifespan of WRNexo Δ mutant flies and even had a detrimental effect in females. Interestingly, the mean lifespan of WRNexo Δ mutant flies was not reduced on a protein-rich diet compared to that of wild-type (WT) flies. Compared to WT control flies, the mutant flies also exhibited altered responses to DR in their resistance to starvation and oxidative stress, as well as changes in sleep/wake patterns. These findings show that the WRN protein is necessary for mediating the effects of DR and suggest that the exonuclease domain of WRN plays an important role in metabolism in addition to its primary role in DNA-repair and genome stability. The Hessian fly, Mayetiola destructor (Say), is one of the most important insect pest plaguing wheat (Triticum aestivum , L) producers across the United States and around the world. Genetic resistance is the stalwart for control of Hessian fly. However, new genotypes (biotypes) arise in deployment of wheat containing resistance genes, so field populations must be evaluated periodically to provide information on the efficacy of those deployed genes. Louisiana (LA), with its diverse agricultural landscape, is not exempt from the challenges posed by this destructive pest. We previously documented the resistance response of wheat lines harboring Hessian fly resistance (H) genes against field populations collected in 2008 from across the southeastern United States, including Iberville Parish, LA. In the spring of 2023, we reevaluated the resistance response of 27 H genes from the field populations collected from Iberville Parish, LA, and compared the results with those observed in 2008. Sixteen H genes showed comparable resistance to the field populations from both years. While 3 of the H genes, H11 , H23 , and H24 , showed a significant decrease in resistance, 2 genes, H16 and H31 , had marked increase in resistance. Furthermore, 6 additional H genes were evaluated in 2023, with 4 showing >70% resistance. Our results clearly identify a total of 20 H genes that are moderate to highly effective against the 2023 Hessian fly population from Iberville Parish, LA. Background: We aimed in this article to assess the likeliness of efavirenz to induce functional senescence in Drosophila melanogaster (fruit fly). Methods: Ten different concentrations of EFV were mixed with fly food and fed to 3-day-old flies orally for a 7 day LC50 calculation. Drug concentrations from LC50 were selected for a 28 day survival to determine the duration of treatment for behavioral and biochemical assays. A 5day feeding plan was used to investigate the effects of the drug on organismal, neuromuscular, reproductive, and metabolic senescence. An in silico study was executed to decipher a molecular interaction of Drosophila enzymes glutathione-s-transferase (GST) or acetylcholinesterase (AChE) with EFV. Results: The calculated LC50 of EFV was 118 mg/10-g fly diet. The test drug induced a significant (P < 0.05) increase in fly mortality, climbing difficulty, and procreative deficits after a 5 day oral exposure. Similarly, there were significant (P < 0.05) biochemical alterations, which suggested in vivo biochemical damage against total thiols (T-SH), SOD (superoxide dismutase), CAT (catalase), GST, AChE, and MDA (malondialdehyde) in the test flies compared to the control groups. In silico study revealed a significantly (P < 0.05) higher binding energy between EFV and the active amino acids of fly AChE and GST when compared to the substrates or standard inhibitors respectively. Conclusion: EFV exhibited ecotoxic potentials evidenced by age-related deficits in the fly's functional integrity such as sluggish movement, procreative deficiency, increased mortality, and oxidant-antioxidant inequality. Determining upper thermal limits of tephritid fly pupae can have practical implications for disinfesting soils and for predicting differential impacts of global warming on flies and their parasites. Here, upper thermal limits of Rhagoletis indifferens Curran (Diptera: Tephritidae) pupae and pteromalid wasps (Hymenoptera: Pteromalidae) inside puparia were determined. Puparia receiving sufficient chill to terminate pupal diapause were exposed to temperatures ramped linearly over 6 h from 21 °C to 47.8, 49.4, 51.1, 55.0, or 60.0 °C for a 0-h hold time. Flies eclosed when pupae were exposed to 47.8 °C but not to 49.4, 51.1, 55.0, or 60.0 °C nor in a separate test to 47.8 °C for 1–3 h hold times. All fly pupae in treatments where no eclosion occurred were dead based on puparial dissections. In contrast, adult wasps eclosed when puparia were exposed to 49.4 and 51.1 °C for 0 h and to 47.8 °C for 1- and 2-h hold times. Despite the greater upper thermal limits of wasps, heat delayed eclosion times of both adult flies and wasps, in 47.8 and 51.1 °C treatments, respectively. In separate tests, longevity of flies exposed as pupae to 47.3–48.6 °C was greater than of control flies, while longevity of control wasps and wasps exposed as immatures to 47.8–51.1 °C did not differ. Flies exposed as pupae to 47.2–48.6 °C produced as many eggs and puparia as control flies. Results suggest heat could be used to disinfest soils of puparia while sparing parasitoids. Insect odorant-binding proteins (OBPs) are members of a rapidly evolving multigene family traditionally thought to facilitate chemosensation. However, studies on Drosophila have shown that members of this family have evolved functions beyond chemosensation, as evident from their expression in reproductive tissues and the brain. Previous studies implicated diverse functions of Obp56h, a member of the largest gene cluster of the D. melanogaster Obp repertoire. Here, we examined the effect of CRISPR/Cas9-mediated deletion of Obp56h on 2 fitness phenotypes, on resistance to starvation stress and heat stress, and on locomotion and sleep phenotypes. Obp56h-/- mutants show a strong sexually dimorphic effect on starvation stress survival, with females being more resistant to starvation stress than the control. In contrast, Obp56h-/- females, but not males, are highly sensitive to heat stress. Both sexes show changes in locomotion and sleep patterns. Transcriptional profiling of RNA from heads of Obp56h-/- flies and the wildtype control reveals differentially expressed genes, including gene products associated with antimicrobial immune responses and members of the Turandot family of stress-induced secreted peptides. In addition, differentially expressed genes of unknown function were identified in both sexes. Genes encoding components of the mitochondrial electron transport chain, cuticular proteins, gene products associated with regulation of feeding behavior (Lst and CCHa2), ribosomal proteins, lncRNAs, snoRNAs, tRNAs, and snRNAs show changes in transcript abundances in Obp56h-/- females. Insects often coevolved with their mutualistic partners such as gut endosymbionts, which play a key in the physiology of host. Studies on such interactions between Bemisia tabaci and its primary and secondary endosymbionts have gained importance due to their indispensable roles in the biology of this insect. Present study reports the predominance of two secondary endosymbionts, Arsenophonus and Cardinium in the Asia II-1 genetic group of whitefly and elucidates their role in the transmission of its vectored Cotton leaf curl virus. Selective elimination of endosymbionts was optimized using serial concentration of ampicillin, chloramphenicol, kanamycin, tetracycline, and rifampicin administered to viruliferous whiteflies through sucrose diet. Primary endosymbiont, Portiera was unresponsive to all the antibiotics, however, rifampicin and tetracycline at 90 μg/ml selectively eliminated Arsenophonus from the whitefly. Elimination of Arsenophonus resulted in significant decrease in virus titer from viruliferous whitefly, further the CLCuV transmission efficiency of these whiteflies was significantly reduced compared to the control flies. Secondary endosymbiont, Cardinium could not be eliminated completely even with higher concentrations of antibiotics. Based on the findings, Arsenophonus plays a key role in the retention and transmission of CLCuV in the Asia II-1 genetic group of B. tabaci, while the role of Cardinium could not be established due to its unresponsiveness to antibiotics. Tennessee and Texas cow-calf producers were surveyed to assess their 2016 expenses for horn fly control methods. Cattle producers who were members of the Texas and Southwestern Cattle Raisers Association and Tennessee cattle producers who have participated in the Tennessee Agricultural Enhancement Program participated in the survey. Average horn fly management costs in Tennessee and Texas were $9.50/head and $12.40/head, respectively. An ordinary least squares regression and quantile regression were estimated to examine how horn fly costs are influenced by producer and farm demographics, seasonality of horn flies, producer horn fly perceptions, and management practices. When controlling for these variables, Tennessee and Texas cattle producers did not spend significantly different amounts on horn fly control methods. Horn fly costs were associated with producer and farm demographics, producer perceptions of horn flies, and management practices. For example, results indicate that horn fly management costs vary depending on a producer's level of education and income. Having Angus cattle and larger herd sizes were associated with lower costs per head spent on horn fly management. Producers who did not consider horn flies to be a problem until greater quantities of flies were present on the animal spent 15% less per head on managing horn flies. In terms of horn fly control methods, feedthrough insecticides increased horn fly costs the most, followed by using ear tags. Horn flies (HF) are one of the most prevalent external parasites on pastured cattle in North America, although they can be found worldwide. HF cause animals severe stress leading to increased cardiac and respiratory rates, and decreased feed efficiency, average daily gain, and milk production. All these detrimental effects caused by HF contribute to over $1 dollars in economic loss in the United States, annually. Current methods of reducing fly numbers, including manure management, feed additives, and insecticides, are being utilized with varying degrees of success. However, these methods require extensive management, are temporary, and are costly to producers. While several studies have investigated the effects of horn flies on growth and milk production traits, there have been very few studies on their potential impacts on reproduction in beef cattle. Insemination and calving data collected on cows and yearling heifers at Northwest Georgia Research and Education Center between 2015 to 2022 was used to determine the effect of HF abundance on fertility. Each animal was exposed to two insemination events. Artificial Insemination (AI) was used except for cows that did not get pregnant from the first AI which were placed on pasture with a proven bull for natural service. The outcomes (success/failure) of each insemination event and calving were recorded. Still births where the dam carried the calf to term, but the calf passed during parturition were recorded as successes. As part of a research project, no fly control methods were carried out on the farms during 2019 and 2022. During these years, images of each animal were taken twice between May and August. The best image of each animal was used to estimate HF count. Animals were placed in three abundance classes (Low: 0-150, Med: 151-400, High: 401+) to analyze the effects of fly abundance on reproduction. For the remaining years (2015-2018, 2020-2021) animals received fly control and were not assessed for fly abundance. As shown Table 1, conception rates of each breeding attempt and calving rates were lower in both years where no horn fly control was used (2019 and 2022) compared with the remaining years where fly control was utilized. The decrease in conception rates when no fly control was used suggests a detrimental effect of HF on reproductive success. Additionally, for the animals measured in 2022, the average fly count for the dams who did not calf successfully was 357 which is significantly higher than the average count for the dams who did successfully calf (290). This suggests that animals with greater fly loads are less likely to carry to term. The sterile insect technique (SIT), used to control different species of tephritid fruit flies (Diptera: Tephritidae), is an important element in sustainable agriculture because of its low negative impact on the environment. In SIT, flies are mass produced and sterilized in the laboratory and then released in a target area. However, once released, laboratory flies may confront harass environments that would reduce their performance and consequently SIT efficiency. Selecting flies that resist stressful conditions may help to improve the efficiency of the SIT by releasing males that resist desiccation, for example, ensuring, thus, their survival in environments with low relative humidity. However, the selection process may affect the resistance of flies to the stress of sterilization, since some life history traits are affected. Here, we studied the effect of irradiation on Anastrepha ludens (Loew) (Diptera: Tephritidae) desiccation resistant flies (DR) compared with nonselected flies (NS). We measured the effect of gamma irradiation dose (0, 20, 40, 60, and 80 Gy) on sterility (males and females) and quality parameters (emergence, flight ability, survival, and male sexual performance) in A. ludens adults of the DR and NS (control) strains. Our results indicate that irradiation affected equally the sterility of adults of both strains. None of the quality parameters differed between strains. The only difference was that DR flies survived longer than control flies. Thus, flies that are resistant to desiccation can be used in the SIT without altering the current process of irradiation and packing. [ABSTRACT FROM AUTHOR]

Stable flies are among the most important pests of livestock throughout much of the world. Their painful bites induce costly behavioral and physiological stress responses and reduce productivity. Stable flies are anthropogenic and their population dynamics vary depending on agricultural and animal husbandry practices. Standardized sampling methods are needed to better identify the factors controlling stable fly populations, test novel control technologies, and determine optimal management strategies. The current study reviewed methods used for a long-term study of stable fly population dynamics in the central Great Plains. An additional study compared the relative size of flies sampled from the general population with that of flies sampled emerging from substrates associated with livestock production. Flies developing in livestock associated substrates are significantly larger than those in the general population indicating that other types of developmental sites are contributing significant numbers of flies to the general population. Because efforts to identify those sites have yet to be successful, we speculate that they may be sites with low densities of developing stable flies, but covering large areas such as croplands and grasslands. Members of the Protein Kinase D (PKD) family are involved in numerous cellular processes in mammals, including cell survival after oxidative stress, polarized transport of Golgi vesicles, as well as cell migration and invasion. PKD proteins belong to the PKC/CAMK class of serine/threonine kinases, and transmit diacylglycerol-regulated signals. Whereas three PKD isoforms are known in mammals, Drosophila melanogaster contains a single PKD homolog. Previous analyses using overexpression and RNAi studies indicated likewise multi-facetted roles for Drosophila PKD, including the regulation of secretory transport and actin-cytoskeletal dynamics. Recently, involvement in growth regulation has been proposed based on the hypomorphic dPKDH allele. We have generated PKD null alleles that are homozygous viable without apparent phenotype. They largely match control flies regarding fertility, developmental timing and weight. Males, but not females, are slightly shorter lived and starvation sensitive. Furthermore, migration of pole cells in embryos and border cells in oocytes appears normal. PKD mutants tolerate heat, cold and osmotic stress like the control but are sensitive to oxidative stress, conforming to the described role for mammalian PKDs. A candidate screen to identify functionally redundant kinases uncovered genetic interactions of PKD with Pkcd, sqa and Drak mutants, further supporting the role of PKD in oxidative stress response, and suggesting its involvement in starvation induced autophagy and regulation of cytoskeletal dynamics. For the first time, the repellent and insecticidal effects of eucalypt essential oil (Eucalyptus globulus) in its free form and associated with different nanostructures (nanoemulsion and nanocapsules) were investigated against Musca domestica (Diptera: Muscidae) and Haemotobia irritans (Diptera: Muscidae) flies. Specimens of M. domestica were collected, separated into groups (n = 10), treated with aspersion of essential oil of eucalypt in its free (1, 5, and 10%) and nanostructured (nanoemulsion or nanoencapsulated at 1, 3, and 5%, respectively) forms. The determination of the insecticidal effect was performed by counting the number of dead flies at intervals of 30, 60, 120, 360 and 750 min after oil aspersion. E. globulus essential oil showed insecticidal effect against M. domestica at concentrations of 1 and 5%. Eucalypt essential oil (10%) caused 100% mortality of M. domestica after 750 min of the treatment. Nanocapsules (3 and 5%) showed efficacy by reducing the number of flies. On the other hand, nanoemulsion did not show insecticidal effect. The repellent action of E. globulus concentrations of 5% was tested against H. irritans on naturally infested cows. The repellent action against H. irritans was verified soon after pulverization. After 24 h, a significant reduction on horn flies' population (83.33 and 66.66%) was observed using free and nanoencapsulated forms tested, respectively. The European cherry fruit fly, Rhagoletis cerasi (L.) (Diptera: Tephritidae), is a highly destructive pest of cherries (Prunus spp.) (Rosaceae) in Europe and Asia. In 2016, R. cerasi was detected in Ontario, Canada, and in 2017 in New York State, USA, the first records of this pest in North America.The initial detections in Canada caused concern for the major cherry-growing states of Michigan, Washington, Oregon, and California in the United States. Establishment of R. cerasi in the United States could restrict cherry exports to other markets and increase costs needed for fly control, but it is unknown if R. cerasi can establish in U.S. commercial cherry regions. Here, we used the CLIMEX ecological niche model to determine the risk of establishment of R. cerasi in the United States and globally. Within the United States under a no-irrigation scenario, R. cerasi would establish in the East and West Coasts; however, under an irrigation scenario, its distribution would expand to the major cherry-growing regions in the interior of central and eastern Washington and in California. Results also showed that if introduced, R. cerasi would likely establish in eastern China, Japan, the Koreas, Australia, New Zealand, South America, South Africa, Mexico, and Canada. Host plant (Prunus spp. and Lonicera spp. [Caprifoliaceae]) presence, although not included in models, would affect fly establishment. The sugarbeet root maggot, Tetanops myopaeformis (von Röder) (Diptera: Ulidiidae), is a major pest of sugar beet Beta vulgaris L. (Carophyllales: Amaranthaceae)in the United States and Canada. Larval feeding on roots can reduce both stand and yield. Current management practices are heavily reliant on chemical control. However, the carbamate and organophosphate insecticides that are commonly used against T. myopaeformis are being phased out of use. Host plant resistance against this pest shows promise, but difficulties with maintaining T. myopaeformis in culture have largely limited such studies to the field. A primary objective of this study was to develop protocols for rearing a laboratory colony of T. myopaeformis that would expedite assays aimed at screening for host plant resistance. Third (final) instar larvae were collected from the field and reared to the adult stage. These laboratory-reared adults laid eggs and ultimately produced a second generation of third-instar larvae in the lab. Adult flies reared from field-collected larvae were used to examine the modality of resistance of a known resistant variety by performing no-choice and paired-choice experiments alongside a susceptible variety in the greenhouse. Paired-choice tests showed no difference in oviposition rates between the two varieties, whereas no-choice tests showed significantly greater feeding damage and abundance of larvae on the susceptible variety. For the resistant variety examined here, we observed evidence of antibiosis, not antixenosis, as the putative modality of resistance. Spotted wing Drosophila, Drosophila suzukii Matsumura (Diptera: Drosophilidae), is an invasive vinegar fly that has become a primary direct pest of berry crops worldwide. We conducted 2 yr of behavioral studies in blueberry plantings to determine how fly activity varied throughout the day. Observations of diurnal activity of adult D. suzukii found the greatest activity in the morning hours between 0600 and 0800 hours, when the majority of flies were on the berries. Flies were also active in the evening mainly between 1800 and 2000 hours; however, this trend was more prominent in 2015, which had cooler and more humid evenings. Experiments examining the effect of irrigation on D. suzukii behavior showed that flies remained active during and after irrigation. The effect of insecticide treatments alone and in combination with irrigation revealed that treatment with spinosad had limited effects on the number of flies per bush, whereas spinetoram reduced the number flying and on the bushes in some cases. Zeta-cypermethrin caused longer and more consistent reduction in D. suzukii flying and on bushes. In all treatments, we observed surviving flies flying near and on treated bushes, indicating that these insecticides do not completely deter fly activity. Irrigation did not influence the effects of zeta-cypermethrin on fly behavior during daily observations up to 3 d after application. Our results highlight that the diurnal patterns of activity of D. suzukii on host plants are flexible and are relatively unaffected by irrigation or insecticide applications. [ABSTRACT FROM AUTHOR]

House flies, Musca domestica L., are pests of poultry facilities and have the ability to develop resistance against different insecticides. This study was conducted to assess the resistance status of house flies to pyrethroid, organophosphate, and novel chemistry insecticides from poultry facilities in Punjab, Pakistan. Five adult house fly populations were studied for their resistance status to selected conventional and novel chemistry insecticides. For four pyrethroids, the range of resistance ratios was 14-55-fold for cypermethrin, 11-45-fold for bifenthrin, 0.84-4.06-fold for deltamethrin, and 4.42-24-fold for lambda-cyhalothrin when compared with a susceptible population. Very low levels of resistance were found to deltamethrin compared with the other pyrethroids. For the three organophosphate insecticides, the range of resistance ratios was 1.70-16-fold for profenofos, 7.50-60-fold for chlorpyrifos, and 4.37-53-fold for triazophos. Very low levels of resistance were found to profenofos compared with the other insecticides. For five novel chemistry insecticides, the range of resistance ratios was 1.20-16.00-fold for fipronil, 3.73-7.16-fold for spinosad, 3.06-23-fold for indoxacarb, 0.96-5.88-fold for abamectin, and 0.56-3.07-fold for emamectin benzoate. Rotation of insecticides with different modes of action showing no or very low resistance may prevent insecticide resistance in house flies. Regular insecticide resistance monitoring and integrated management plans on poultry farms are required to prevent resistance development, field control failures, and environmental pollution. The functional aspects of the adult house fly crop have not been studied even though various human and domestic animal pathogens have been discovered within the crop lumen. The average volume consumed (midgut and crop) by flies starved for 24 h was 3.88 p.1 by feeding both sexes on a sucrose phosphate glutamate buffer. In addition, various volumes of a solution (0.125 M sucrose plus Amaranth dye) were fed to 3-d-old adult female house flies to quantify the crop contraction rate as affected by crop volume. As crop volume increased, the contraction rate increased until it reached a peak at 2 pi, after which it declined. It is hypothesized that the high contraction rate of the crop, which in house fly is almost twice the rate of three other fly species, is one of the factors that makes house fly an excellent vector. Background This study was aimed at examining the potential of non-biting synanthropic filth flies to acquire naturally eggs of human intestinal helminths from unsanitary sites, and its attendant public health importance. Methods Body surface washings and gut contents of flies caught foraging while infected human faeces lay exposed at a garbage dump in Iperu, Ogun State, Nigeria and within 24 hours subsequently after removal of faeces from the dump were examined parasitologically by the formol-ether concentration technique. The viability of helminth eggs isolated from flies was determined by incubation under laboratory conditions. Results A total of 303 flies were examined: Musca domestica (107; 35.3%), Chrysomya megacephala (125; 41.3%) and Musca sorbens (71; 23.4%). Eggs of Ascaris lumbricoides and Trichuris trichiura isolated from exposed human faeces were recovered from the body surfaces and or gut contents of flies caught before (141; 77.5%) and after removal of exposed faeces (44; 36.4%). Eggs of Taenia sp. were isolated only from the gut contents of three C. megacephala flies caught after removal of exposed faeces. Significantly more (p<0.05) eggs were recovered from fly gut contents than body surfaces and from flies caught before than after removal of exposed faeces. 93.1% (552; from exposed faeces) and 57.4% (408; from flies) of A. lumbricoides and T. trichiura eggs were viable. We conducted a series of nine laboratory experiments testing the response of "vinegar flies," Drosophila melanogaster Meigen (Diptera: Drosophilidae), released in bioassay chambers to experimental traps and lures. These experiments showed that an effective trap could be constructed from a clear 225-mi screw-cap jar fitted with a hollow 8-mm-diameter cylindrical cross bridge. Flies could enter the trap from either end of the cylindrical "gate" and in turn could enter the interior chamber of the trap through a cut out portion at mid-span of the cylinder. The experiments also showed that a natural-component lure could be made using a teabag containing freeze-dried banana powder, yeast, and carrageenan gum powder as a humectant. When dipped in water for 10-15s and then placed in the bottom of a trap, the teabag provided effective attraction for at least 7 d. Captured flies were immobilized on a sticky card placed in the trap, allowing them to be easily seen. Unlike other traps that cannot be opened and have liquid lures, the cylindrical-gate trap can be reused repeatedly if the teabag and sticky card are replaced. A final two experiments showed that the prototype operational cylindrical-gate trap with a teabag lure captured 3.3 and 2.3 times more released flies, respectively, than the next best of three commercially available traps. The horn fly, Haematobia irritans (L.) (Diptera: Muscidae), was introduced to Chile in the beginning of the 1990s. Since its introduction, farmers have controlled this pest almost exclusively with insecticides. To understand the consequences of different control strategies on the development of insecticide resistance and their persistence, a field survey was conducted at eight farms in the south of Chile to characterize insecticide resistance in field populations and resistance mechanisms. Horn fly samples were assayed to determine levels of resistance to pyrethroids and diazinon, genotyped for kdr and HiαE7 mutations, and tested for general esterase activity. All field populations, including ones that were not treated with insecticides for the past 5 yr, showed high levels of cypermethrin resistance and high frequencies of the kdr mutation. None of the fly populations demonstrated resistance to diazinon and the HiαE7 mutation was not detected in any of the fly samples. Esterase activities in all populations were comparable to those found in the susceptible reference strain. The findings of high frequencies of homozygous resistant and heterozygous individuals both in insecticide treated horn fly populations and in the untreated fly populations suggests complex interactions among field populations of the horn fly in Chile. Release-recapture studies were conducted with both feral and sterile females of the Caribbean fruit fly, Anastrepha suspensa (Loew) (Diptera: Tephritidae),to determine sampling range for a liquid protein bait (torula yeast/borax) and for a two-component synthetic lure (ammonium acetate and putrescine). Tests were done in a guava, Psidium guajava L., grove and involved releasing flies at a central point and recording the numbers captured after 7 h and 1, 2, 3, and 6 d in an array of 25 Multilure traps located 9-46 m from the release point. In all tests, highest rate of recapture occurred within the first day of release, so estimations of sampling range were based on a 24-h period. Trap distances were grouped into four categories (<10, 10-20,20-30, and >30 m from release point) and relative trapping efficiency (percentage of capture) was determined for each distance group. Effective sampling range was defined as the maximum distance at which relative trapping efficiency was ⩾25%. This corresponded to the area in which 90% of the recaptures occured. Contour analysis was also performed to document spatial distribution of fly dispersal. In tests with sterile flies, immature females dispersed farther and were recovered in higher numbers than mature females, regardless of attractant, and recapture of both cohorts was higher with torula yeast. For mature feral flies, range of the synthetic lure was determined to be 30 m. With sterile females, effective range of both attractants was 20 m. ABSTRACT Field-cage experiments were performed to determine the effectiveness of MultiLure traps (Better World MFG Inc., Fresno, CA) baited with NuLure (Miller Chemical and Fertilizer Corp., Hanover, PA) or BioLure (Suterra LLC, Inc., Bend, OR) in capturing individually marked Mexican fruit fiy,Anastrepha ludens (ew),and West Indian fruit fly,Anastrepha obliqua (Macquart) (Diptera: Tephritidae), of both sexes. Experimental treatments involved wild and laboratory-reared flies of varying ages (2-4 and 15-18 d) and dietary histories (sugar only, open fruit, open fruit plus chicken feces, and hydrolyzed protein mixed with sugar). Data were divided into two parts: total captures over a 24-h period and trap visits/landings, entrances into interior of trap ,and effective captures (i.e., drowning in liquid bait or water) over a 5-h detailed observation period (0600-1100 hours). The response to the two baits varied by fly species, gender, physiological state, age, and strain. Importantly, there were several highly significant interactions among these factors, underlining the complex nature of the response. The two baits differed in attractiveness for A. obliqua but not A. ludens. The effect of strain (wild versus laboratory flies) was significant for A. ludens but not A. obliqua. For effect of dietary history, adults of both species, irrespective of sex, were significantly less responsive to both baits when fed on a mixture of protein and sugar when compared with adults fed the other diets. Finally, we confirmed previous observations indicating that McPhail-type traps are quite inefficient. Considering the total 24-h fly tenure in the cage, and independent of bait treatment and fly type (i.e., strain, adult diet, gender and age), of a total of 2,880 A. ohliqua and 2,880 A. indens adults released into the field cages over the entire study (15 replicates), only 564 (19.6%) and 174 (6%) individuals, respectively, were effectively caught. When only considering the 5-h detailed observation period and independent of bait treatment and fly type, of a total of 785 marked flies that landed on traps (519 females and 266 males, respectively), only 10.3% (144 females and 59 males) and 20.8% (25 females and 18 males) A. obliqua and A. mdens individuals, respectively, ended up being effectively captured. Stable fly, Stomoxys calcitrans (L.) (Diptera: Muscidae), once only a pest of pastured cattle, has become a serious pest of range cattle in the United States. Because of the difficulties associated with stable fly management under range conditions, a pesticide-impregnated cloth target is being developed as a management tool, We conducted studies to determine the influence of weather, time, fabric type, insecticide type, and insecticide concentration on the mortality of stable flies from a susceptible laboratory colony exposed for 30 s to treated cloth targets. We found that 100% of the flies exposed to trigger (Trigger-Royal Box, 65% polyester and 35% cotton) fabric targets that were treated with 0.1% λ-cyhalothrin or 0.1% ζ-cypermethrin and weathered outdoors in Gainesville, FL., for up to 3 mo, were dead within 20 min after a 30-s exposure. Irradiation postharvest phytosanitary treatments are used increasingly and show further promise because of advantages compared with other treatments, Its chief disadvantage is that, unlike all other commercially used treatments, it does not provide acute mortality, although it prevents insects from completing development or reproducing. The objective of this research was to determine to what extent irradiated egg arid early instars of tephritids would develop to later instars that could be found by phytosanitary inspectors or consumers. Mexican fruit fly, Anastrepha indens (Loew), eggs and first instars in grapefruit, Citrus paradisi Macfayden, were irradiated with 70-250 Gy and held at ≈27°C until third instars completed development. The accepted minimum absorbed phytosanitary dose for this pest is 70 Gy, although higher doses may be applied under commercial conditions. The more developed a fruit fly before it was irradiated, the greater the proportion that survived to the third instar. Also, dose was inversely related to developmental success, e.g., a mean of ≈65 and 35%, respectively, of late first instars reached the third instar when irradiated with 70 and 250 Gy. Of those, 65.1 and 23.4%, respectively, pupariated, although no adults emerged. Irradiation may result in a greater frequency of live (albeit incapable of resulting in an infestation) larvae being found than would be expected compared with other treatments that provide acute mortality. The regulatory community should be aware of this and the fact that it does not increase the risk of irradiation phytosanitary treatments resulting in an infestation of quarantine pests. Sticky card captures of houseflies, Musca domestica L. (Diptera: Muscidae), were used to compare efficacy of screen-covered baits containing sugar, sugar and 0.1% (Z)-9-tricosene, sugar and 1.0% (Z)-9-tricosene, Golden Mali-in [1.1% methomyl and 0.049% (Z)-9-tricosene], and QuickBayt 105% imidacloprid and 0.1% (Z) -9-tricosene 1. The QuickBayt treatment caught more flies per hour (mean 116.5) than sugar alone (mean = 81.0), but the addition of (Z)-9-tricosene to sugar did not increase fly capture compared with sugar alone. More males (65% of total) than females were collected on the sticky cards for all treatments. Fly kill by plain sugar (control) and the commercial baits Golden Mali-in, QuikStrike Fly Abatement strips (1.0% nithiazine), and QuickBayt was tested over a 90-mm period. An average of 1.4, 5.6, 363.0, and 1,266.0 flies were killed using sugar, Golden Mali-in, QuikStrike, and QuickBayt, respectively. The similarity between Golden Malrin and plain sugar reflects severe resistance to this once effective methomyl bait. A no-choice feeding assay using lab-reared methomyl-susceptible and methomyl-resistant house flies was conducted with and without (Z)-9-tricosene. Adult mortality was significantly higher in the methomyl-susceptible strain exposed to treatments containing methomyl. The objective of this experiment was to examine the effects of fly control strategies on fly abundance, fly avoidance behaviors, and animal performance in a feedlot setting. Angus bulls (n = 64; BW = 281 ± 36.2 kg) and steers (n = 36; BW = 475 ± 40.4 kg) were blocked by sex and BW and assigned to 1 of 4 experimental treatments in a randomized complete block design (7 pens/treatment; 4 bulls/pen; 3 steers/pen). Treatments included: control (CON; no fly control), abamectin and piperonyl butoxide insecticide tags (FT; XP820®, Y-Tex Corporation, Cody, WY), permethrin and piperonyl butoxide pour on (PO; Permectrin® CDS, Bayer Animal Health, Shawnee Mission, KS), or a garlic-powder top dress (GR) administered at 0.28 g • -1 animal • -1d. The experiment was conducted between June and September. Horn and stable fly abundance (FA) and fly avoidance behaviors (FAB; i.e. tail-flicks, stomping, head-tossing) were recorded by trained personnel. Both FA and FAB were recorded once/wk on days with similar temperatures, humidity, wind speed, and without precipitation based on Oklahoma Mesonet data (Stillwater site). There was a tendency for a treatment × time interaction for horn FA with differences in treatments occurring at wk 1, 2, 8, and 14. There was no treatment × time interaction for stable FA or FAB, however both were affected by time (P < 0.001). There were no differences in final BW (P = 0.34), overall ADG (P = 0.30), overall DMI (P = 0.53), or overall G:F (P = 0.39). There was a tendency (P ≤ 0.10) for GR to have decreased BW on d 28 and decreased ADG from d 0 to 28 compared with FT and PO. There was also a tendency (P = 0.09) for decreased DMI from d 0 to 56 for GR compared to CON and FT. Different larval stages of balsam fir sawfly, Neodiprion abietis (Harris) (Hymenoptera: Diprionidae), were challenged by different concentrations of a nucleopolyhedrovirus in the laboratory to determine larval susceptibility to the virus and to test treatment effects of the virus on the sawfly survivors. The results indicated that younger larvae were more susceptible to the virus than older instars. The speed of larval death depended on the larval age and the virus concentration. Generally, the virus killed second or third instars in ≈5 d and fourth or fifth instars in 10-12 d at concentrations of 107 polyhedral inclusion bodies (PIB)/ml. The virus had profound treatment effects on sawfly survivors. Feeding activity of the survivors was reduced by >40% compared with that of the control group, pupal weight by ≈25%, and adult emergence by >30%. The frequency of the L1014 F kdr mutation was determined in 14 field populations of house flies, Musca domestica L., with resistance factors at LD50 for pyrethrin/piperonyl butoxide and bioresmethrin/piperonyl butoxide from 4 to 29 and 2 to 98, respectively. A polymerase chain reaction test for identifying kdr homo- or heterozygote house flies was used to determine the frequency of kdr. The L1014 F allele was found in all populations tested. The frequency of kdr in the field populations was high and varied from 0.46 to 0.99. Eleven of the populations were in Hardy-Weinberg equilibrium, whereas two strains had higher number of heterozygotes than expected, indicating a possible heterozygote advantage. The frequency of kdr was strongly correlated with the reduced mortality observed in the bioassays with pyrethrum and bioresmethrin synergized by piperonyl butoxide. This indicates that kdr is a major mechanism for pyrethroid resistance in these field populations. Five field populations had resistance factors >25 and >10 for bioresmethrin/piperonyl butoxide and pyrethrin/ piperonyl butoxide, respectively. The frequencies of kdr in these five populations varied from 0.89 to 0.99. The frequencies of kdr in the field populations showing no or a low level of resistance had frequencies of kdr from 0.46 to 0.75, which indicates that the L1014 F kdr allele is a fully recessive genetic trait in house flies. We have shown that the molecular diagnostic PASA method to determine the resistance phenotypes and the frequency of kdr is a powerful tool, which could be used to get information to make recommendations about pest and resistance management. Tachinaephagus zealandicus Ashmead is a gregarious endoparasitoid that attacks third instars of muscoid flies, including house flies, Musca domestica L. A colony of this parasitoid was established from samples collected from a poultry farm in Santa Cruz da Conceição, São Paulo, Brazil. The objective of this study was to evaluate the influence of feeding treatment, host density and temperature on attack rates on T. zealandicus. Parasitoids that were given honey as adults attacked two to three times as many house fly larvae (25 host attacks/female/d) as parasitoids that were given only water or nothing. Host attacks and progeny production by T. zealandicus on house fly and Chrysomyia putoria increased over the range of host:parasitoid ratios tested, reaching a maximum of 21–22 hosts killed and 13 progeny produced/female/d at the highest host density of 32 larvae/female. Host attacks were higher at 22°C than at the other temperatures studied (20–29°C), but differences in attack rates were small over the range of 20–27°C (10–13 host attacks/female). Comparatively few hosts (6.3) were attacked at 29°C. Higher rates of progeny production also were observed among parasitoids tested at lower temperatures (9–11 progeny produced/female at 20–22°C) than at 29°C (1.8 progeny/female). Females of T. zealandicus that were stored at 15°C after emergence had highest rates of host attacks (58–62 hosts killed per group of five female parasitoids) and progeny production (174–261 progeny) after 6–12 d of storage at this temperature; relatively few hosts were attacked or parasitized (6–9 host attacks and progeny/group) after 0 or 1 d at 15°C. This study aims to evaluate effectiveness of two operational management systems for steer gains and fly control. The first strategy evaluated was pasture burn date of March (MAR) or April (APR). The second management strategy was fly control through feeding free-choice mineral with spices (SPICE) or without spices (CON). Eight pastures (n = 281 steers; initial weight 277.7 ± 25.2 kg) were used in a 2 x 2 factorial treatment structure. Steers were weighed individually, randomly assigned to treatment, and grazed for 85 days. Weekly 33% of steers per pasture were photographed and fly numbers counted. ADG was impacted by both burn date and mineral type (P = 0.01). Cattle on the APR-SPICE treatment had a greater ADG than MAR-SPICE and APR-CON with MAR-CON intermediate. Cattle on SPICE gained 4.5 kg (P = 0.12) more than cattle consuming CON mineral. Flies per steer were impacted by burn x mineral x week (P < 0.001). In general, APR-SPICE steers had a greater number of flies weeks 8, 10, and 11 (P < 0.05), corresponding to a time when mineral intake averaged 72% of the formulated intake. The MAR-SPICE steers consumed on average the formulated amount of mineral and through the entire grazing period had lower number of flies than APR-SPICE steers (P < 0.05). The second largest ADG observed with the MAR-CON treatment may be attributed to lower fly numbers as this treatment had one extra week below economic threshold for horn flies. These results are somewhat inconclusive on pasture burn date, but show that the addition of spices to a free-choice complete mineral shows promise as a cost-effective method to increase gains in stocker steers on tallgrass native range. Chlorfenapyr (AC 303630) was found to be 5 times as toxic to pyrethroid-resistant horn flies, Haematobia irritans (L.), as to susceptible. We hypothesize this difference occurs because higher levels of mixed function oxidises in the resistant flies convert more chlorfenapyr (a pro-insecticide) to the insecticidal form. The horn fly, Haematobia irritans (L.), is a significant economic pest of cattle. The economic losses associated with horn fly infestations are attributed to irritation behaviors and blood loss in cattle as well as decreased milk production, decreased weaning weights, and decreased weight gain. Estimates of annual losses due to damage and control costs exceed $1 billion in the U.S. Since the adult horn fly spends almost all its adult life on livestock, the majority of insecticides and various application techniques have been successful in controlling horn flies. However, since they are able to produce multiple generations in a summer season, insecticide resistance in horn fly populations can result in up to a 90% reduction in product efficacy. The objective of this presentation is to share information on the efficacy of commonly used insecticide treatments on horn fly populations. Different control techniques will be shared including the use of insecticidal impregnated ear tags, insecticidal sprays, insecticidal pour-ons, insecticidal capsules and an IGR feedthrough. Reducing fly populations on dairy farms can increase cow comfort and reduce the spread of disease pathogens. The objective of these experiments was to determine the effect of inclusion of diflubenzuron larvicide (ClariFly®) in feeds consumed on fly populations on dairy farms. In 2008 and 2013, twelve dairy farms in Vermont and New York were assigned to 6 blocks by herd size, geographic location and lactating herd housing. On treated dairy farms, ClariFly® was added to all purchased feeds to achieve an intake of 0.10 mg diflubenzuron/kg BW/d. In 2013, ClariFly® was also added to milk or milk replacer fed to calves. Control dairy farms were permitted to use all fly control methods other than "fed-through" products. Each dairy was visited biweekly starting in June. Fly populations were quantified with fly speck count on 3 × 5 notecards, collection of flies on a sticky trap during walk on predetermined path, flies on-calf counts, and observations of fly avoidance behavior. Data were analyzed as a randomized block design with main effects of random block, treatment, period, and treatment by period interaction in MIXED procedure of SAS. Period effects (P < 0.01) reflected normal seasonal changes in fly population. With treatment in 2008 and 2013, fly populations decreased in the lactating herd housing when compared to control (P < 0.05) but an interaction of treatment and period increased difference in summer (P < 0.07). In calf housing, treatment did not decrease fly populations measured in 2008 likely related to limited intake of purchased grain during first weeks of life (P > 0.10). In 2013 with ClariFly® included in milk or milk replacer, fly populations decreased in calf housing (P < 0.001).
