We report on high-sensitivity neutral hydrogen observations toward the gas-rich interacting galaxies NGC 3395/3396 with the Five-hundred-meter Aperture Spherical radio Telescope (FAST). Compared to previous observations carried out by the Very Large Array (VLA) and the Westerbork Synthesis Radio Telescope (WSRT), a more extended H  i envelope around this system has been detected. The total H  i gas mass of the NGC 3395/3396 system is estimated to be 7.8 × 109 M |$_\odot$|⁠. This value is 2.7 times more than that reported based on the VLA interferometric maps. Previous observations found a large H  i tail extending to the south-west and a minor tail emerging from the north of this peculiar galaxy pair. Based on the high-sensitivity observations of FAST, an extended H  i plume to the north-west and a gas plume to the north-east have been detected for the first time. Neutral hydrogen of the two smaller galaxies IC 2604 and IC 2608 in the south of the system have also been detected. We discuss the origins of these extra gas and possible tidal interactions between these galaxies. The most prominent tidal feature of NGC 3395/3396, the south-west tail combined with the new detected north-west plume, behaves like a large ring. We suggest the ring might be formed by the previous fly-by interaction between NGC 3395 and 3396, which happened 500 Myr ago. Our study shows that high-sensitivity H  i observations are important in revealing low column density gas, which is crucial to a deeper understanding of this interacting system. Muography is a promising and rapidly developing physical prospecting technique based on the attenuation of muon flux. The feasibility and potential of applying muography to mining were presented in a small number of previous case studies. In this work, we aimed to address three challenges that limit the applicability and efficiency of muography in mineral exploration: (1) application to low-density-contrast ore body exploration, (2) analysis of altitudinal impacts on measurements and (3) precise and efficient reconstruction. We conducted the first case of applying muography to the exploration for low-density-contrast ore bodies. Six muon imaging systems were placed underground to collect surviving muons for roughly half a year. We analysed the altitudinal impact on the data measurements and proposed a simplified method to eliminate it. We also developed the seed algorithm, a novel 3-D reconstruction algorithm, that can reconstruct anomalies located far away from the detectors and avoid their elongation along the observed directions. Benefitting from the seed algorithm, a low-density-contrast ore body and a limonitic siliceous slate structure were reconstructed, demonstrating the sensitivity of this technique in density distinction; discoveries of several mined-out areas are important for accident avoidance; and reconstruction of the stope and scarps served as strong circumstantial evidence of the reliability of the result. The success of this experiment shows the great value of muography in the economic, research and safety aspects of mineral exploration and inspection. We present the results of our new H  i observations for the nearby galaxy NGC 4449 using the Five-hundred-meter Aperture Spherical radio Telescope (FAST). More diffuse H  i gas was detected by FAST compared with previous studies. The measured extent of H  i gas is more than ∼19 arcmin (20 kpc) larger than that in the Very Large Array map, and the H  i gas disc is ∼22 times the optical diameter of NGC 4449. The total H  i mass detected for NGC 4449 is 3.07 |$\times 10^9\, \rm M_{\odot }$|⁠ , which is in agreement with that detected by the Effelsberg telescope. We found one H  i cloud from the channel map that is in superposition with the optical dwarf galaxy NGC 4449B. This cloud is slightly offset from the NGC 4449B disc, suggesting that it could be gas stripped from NGC 4449B's optical body when it was disrupted and pulled into the nuclear region of NGC 4449. We also find a region in the outer part of the H  i envelope with a peculiar velocity field and double-peaked spectral profiles, whose velocity distribution is consistent with the nearby companion dwarf galaxy DDO 125. This implies that NGC 4449 may have stripped gas out of DDO 125. We suggest that the irregularity of NGC 4449's huge H  i gas halo might originate from interactions with NGC 4449B and DDO 125. We observed the classical interacting galaxy M51 with FAST and obtain high-sensitivity H  i image with column density down to 3.8 × 1018 cm−2. In the image we can see a diffuse extended envelope around the system and several new tidal features. We also get a deeper look at M51b's probable gas, which has an approximated velocity range of 560 to 740 km s−1 and a flux of 7.5 Jy km s−1. Compared to the VLA image, we observe more complete structures of the Southeast Tail, Northeast Cloud, and Northwest Plume, as well as new features of the Northwest Cloud and Southwest Plume. M51's most prominent tidal feature, the Southeast Tail, looks very long and broad, in addition to the two small detached clouds at the periphery. Due to the presence of optical and simulated counterparts, the Northwest Cloud appears to be the tail of M51a, while the Northwest Plume is more likely a tidal tail of M51b. The large mass of the Northwest Plume suggests that M51b may have been as gas-rich as M51a before the interaction. In addition, the formation process of the Northeast Cloud and Southwest Plume is obscured by the lack of optical and simulated counterparts. These novel tidal features, together with M51b's probable gas, will inspire future simulations and provide a deeper understanding of the evolution of this interacting system. [ABSTRACT FROM AUTHOR]

With the aim to study the extended H  i environment around dwarf galaxies in the local universe, we performed a high-sensitivity H  i observation of the DDO 168 and DDO 167 pair using the Five-hundred-meter Aperture Spherical Radio Telescope (FAST). We chose this pair as previous studies indicate that DDO 168 might have undergone an interaction with a local companion. Based on this deep observation, no new extra gas was found around DDO 167 and it was hardly resolved by FAST. On the other hand, on the north side of DDO 168 we discovered an extended plume of H  i gas which has never been detected before. The plume has a ring-like structure with a radius of about 5.0 kpc and its gas mass is estimated to be 5.2 × 107 M⊙. The expanding velocity of the plume is about 30 km s−1, corresponding to a dynamical age of about 200 Myr. The position–velocity (PV) diagrams cut through the two galaxies show a short and pronounced 'spur' pointing to DDO 167 from DDO 168, indicating a tidal interaction between the two galaxies. We also found an H  i knot in the plume. The gas mass of the knot is about 9.5 × 106 M⊙. This value is much higher than its virial mass, indicating the knot is gravitationally unstable and star formation will take place there in the future. Our study suggests that the ring-like structure of the plume might be caused by the passage of DDO 167 through the outer disc of DDO 168 about 200 Myr ago. Background Allele-specific expression (ASE) refers to the preferential expression of one allele over the other and contributes to adaptive phenotypic plasticity. Here, we used a reciprocal cross-model between phenotypically divergent European Berkshire and Asian Tibetan pigs to characterize 2 ASE classes: imprinting (i.e. the unequal expression between parental alleles) and sequence dependent (i.e. unequal expression between breed-specific alleles). We examined 3 transcript types, including protein-coding genes (PCGs), long noncoding RNAs, and transcripts of unknown coding potential, across 7 representative somatic tissues from hybrid pigs generated by reciprocal crosses. Results We identified a total of 92 putative imprinted transcripts, 69 (75.00%) of which are described here for the first time. By combining the transcriptome from purebred Berkshire and Tibetan pigs, we found ∼6.59% of PCGs are differentially expressed between breeds that are regulated by trans- elements (e.g. transcriptional factors), while only ∼1.35% are attributable to cis (e.g. promoters). The higher prevalence of trans- PCGs indicates the dominated effects of trans -regulation in driving expression differences and shaping adaptive phenotypic plasticity between breeds, which were supported by functional enrichment analysis. We also found strong evidence that expression changes mediated by cis- effects were associated with accumulated variants in promoters. Island species and their ecosystems play an important role in global biodiversity preservation, and many vulnerable island species are conservation priorities. Although insular habitat likely facilitates the species diversification process, it may also aggravate the fragility of these species with high risk of inbreeding. The Shedao pit-viper Gloydius shedaoensis is an island endemic species with an extremely high population density, which has been categorized as vulnerable in the IUCN (International Union for the Conservation of Nature and Natural Resources) Red List. We collected 13,148 SNP (Single Nucleotide Polymorphism) from across its genome and examined its genetic diversity and demographic history. The Shedao pit-viper has a low genetic diversity but shows no sign of inbreeding. Furthermore, population genetic structure analysis, including the neighbor-joining tree, principal coordinate analysis, clustering, and spatial autocorrelation, revealed a general lack of spatial structure. Only the isolation by distance residues suggested a weak patchiness. Overall, the population is nearly panmictic and gene flow is evenly distributed across the island. A large number of individuals, small size of the island, and the lack of population structure likely all contribute to the lack of inbreeding in this species. We also detected signs of male-biased dispersal, which likely is another inbreeding avoidance strategy. Historical demographic analysis suggested that the historical population size and distribution of the species are much larger than their current ones. The multiple transgressive–regressive events since the Late Pleistocene are likely the main cause of the population size changes. In the modern poultry industry, newly hatched chicks are unavoidably transported from the hatching to the rearing foster. Stress caused by multiple physical and psychological stressors during transportation is particularly harmful to the liver. Astragalus polysaccharide (APS) possesses multiple benefits against hepatic metabolic disorders. Given that transport stress could disturb hepatic glucolipid metabolism and the role of APS in metabolic regulation, we speculated that APS could antagonize transport stress-induced disorder of hepatic glucolipid metabolism. Firstly, newly hatched chicks were transported for 0, 2, 4, and 8 h, respectively. Subsequently, to further investigate the effects of APS on transport stress-induced hepatic glucolipid metabolism disturbance, chicks were pretreated with water or APS and then subjected to transport treatment. Our study suggested that APS could relieve transport stress-induced lipid deposition in liver. Meanwhile, transport stress also induced disturbances in glucose metabolism, reflected by augmented mRNA expression of key molecules in gluconeogenesis and glycogenolysis. Surprisingly, APS could simultaneously alleviate these alterations via peroxisome proliferator-activated receptor gamma coactivator 1 alpha (PGC-1α)/Sirtuin 1 (SIRT1)/AMP-activated protein kinase (AMPK) pathway. Moreover, APS treatment regulated the level of peroxisome proliferator-activated receptor alpha (PPARα) and peroxisome proliferator-activated receptor gamma (PPARγ), thereby alleviating transport stress-induced alterations of VLDL synthesis, cholesterol metabolism, lipid oxidation, synthesis, and transport-related molecules. These findings indicated that APS could prevent the potential against transport stress-induced hepatic glucolipid metabolism disorders via PGC-1α/SIRT1/AMPK/PPARα/PPARγ signaling system. There are many species of Chinese traditional leguminosae family plants that are well known for their medicinal applications, such as Astragalus membranaceus , Catsia tora , Glycyrrhiza uralensis , Sophora flavescens and Albacia acacia. Their unique bioactive composition and internal phenological environment contribute to the formation of specific and unique endophytic fungal communities, which are important resources for new compounds used in a variety of pharmacological activities. Nonetheless, they have not been systematically studied. In the last decade, nearly 64 genera and thousands of species of endophytic fungi have been discovered from leguminosae plants, as well as 138 secondary metabolites (with 34 new compounds) including flavonoid, alkaloids, phenol, anthraquinone, macrolide, terpenoid, phytohormone and many more. These were shown to have diverse applications and benefits, such as antibacterial, antitumor, antioxidative, immunoregulatory and neuroprotective properties. Here, we provide a summarized overview with the aim of raising awareness of endophytic fungi from medicinal leguminosae plants and providing a comprehensive review of the discoveries of new natural products that may be of medicinal and pharmaceutical importance. Background The hard-shelled mussel (Mytilus coruscus) is widely distributed in the temperate seas of East Asia and is an important commercial bivalve in China. Chromosome-level genome information of this species will contribute not only to the development of hard-shelled mussel genetic breeding but also to studies on larval ecology, climate change biology, marine biology, aquaculture, biofouling, and antifouling. Findings We applied a combination of Illumina sequencing, Oxford Nanopore Technologies sequencing, and high-throughput chromosome conformation capture technologies to construct a chromosome-level genome of the hard-shelled mussel, with a total length of 1.57 Gb and a median contig length of 1.49 Mb. Approximately 90.9% of the assemblies were anchored to 14 linkage groups. We assayed the genome completeness using BUSCO. In the metazoan dataset, the present assemblies have 89.4% complete, 1.9% incomplete, and 8.7% missing BUSCOs. Gene modeling enabled the annotation of 37,478 protein-coding genes and 26,917 non-coding RNA loci. Phylogenetic analysis showed that M. coruscus is the sister taxon to the clade including Modiolus philippinarum and Bathymodiolus platifrons. Conserved chromosome synteny was observed between hard-shelled mussel and king scallop, suggesting that this is shared ancestrally. Transcriptomic profiling indicated that the pathways of catecholamine biosynthesis and adrenergic signaling in cardiomyocytes might be involved in metamorphosis. Conclusions The chromosome-level assembly of the hard-shelled mussel genome will provide novel insights into mussel genome evolution and serve as a fundamental platform for studies regarding the planktonic-sessile transition, genetic diversity, and genomic breeding of this bivalve. Although the governing equations of many systems, when derived from first principles, may be viewed as known, it is often too expensive to numerically simulate all the interactions they describe. Therefore, researchers often seek simpler descriptions that describe complex phenomena without numerically resolving all the interacting components. Stochastic differential equations (SDEs) arise naturally as models in this context. The growth in data acquisition, both through experiment and through simulations, provides an opportunity for the systematic derivation of SDE models in many disciplines. However, inconsistencies between SDEs and real data at short time scales often cause problems, when standard statistical methodology is applied to parameter estimation. The incompatibility between SDEs and real data can be addressed by deriving sufficient statistics from the time-series data and learning parameters of SDEs based on these. Here, we study sufficient statistics computed from time averages, an approach that we demonstrate to lead to sufficient statistics on a variety of problems and that has the secondary benefit of obviating the need to match trajectories. Following this approach, we formulate the fitting of SDEs to sufficient statistics from real data as an inverse problem and demonstrate that this inverse problem can be solved by using ensemble Kalman inversion. Furthermore, we create a framework for non-parametric learning of drift and diffusion terms by introducing hierarchical, refinable parameterizations of unknown functions, using Gaussian process regression. We demonstrate the proposed methodology for the fitting of SDE models, first in a simulation study with a noisy Lorenz '63 model, and then in other applications, including dimension reduction in deterministic chaotic systems arising in the atmospheric sciences, large-scale pattern modeling in climate dynamics and simplified models for key observables arising in molecular dynamics. Background: The frequent movement of population between countries brings an increasing number of travel-related infections. This study aims to define the spectrum and dynamics of imported infections observed from international travel in the Chinese mainland.Methods: Sick travellers were screened by inbound sentinel surveillance and post-travel clinic visits from 2014 to 18. The infections were classified as respiratory, gastrointestinal, vector-borne, blood/sexually transmitted and mucocutaneous. The analysed variables included the place of origin of the travellers (Chinese or foreign) and the time when travel-related infection was present (at the time of return, during travel and post-travel visits to the clinic).Results: In total, 58 677 cases were identified amongst 1 409 265 253 travellers, with an incidence of 41.64/million, comprising during-travel incidence of 27.44/million and a post-travel incidence of 14.20/million. Respiratory infections constituted the highest proportion of illnesses during travel (81.19%, 31 393 of 38 667), which mainly came from Asian countries and tourists; with influenza virus and rhinovirus infections being mainly diagnosed. Vector-borne diseases constituted the highest proportion of post-travel illnesses (98.14%, 19 638 of 20 010), which were mainly diagnosed from African countries and labourers; with malaria and dengue fever being mainly diagnosed. The differential infection spectrum varied in terms of the traveller's demography, travel destination and travel purpose. As such, a higher proportion of foreign travellers had blood/sexually transmitted diseases (89.85%, 2832 of 3152), while Chinese citizens had a higher prevalence of vector-borne diseases (85.98%, 19 247 of 22 387) and gastrointestinal diseases (79.36%, 1115 of 1405). The highest incidence rate was observed amongst travellers arriving from Africa, while the lowest was observed amongst travellers arriving from Europe.Conclusions: The findings might help in preparing recommendations for travellers and also aid in primary care or other clinics that prepare travellers before trips abroad. Background The domestic goose is an economically important and scientifically valuable waterfowl; however, a lack of high-quality genomic data has hindered research concerning its genome, genetics, and breeding. As domestic geese breeds derive from both the swan goose (Anser cygnoides) and the graylag goose (Anser anser), we selected a female Tianfu goose for genome sequencing. We generated a chromosome-level goose genome assembly by adopting a hybrid de novo assembly approach that combined Pacific Biosciences single-molecule real-time sequencing, high-throughput chromatin conformation capture mapping, and Illumina short-read sequencing. Findings We generated a 1.11-Gb goose genome with contig and scaffold N50 values of 1.85 and 33.12 Mb, respectively. The assembly contains 39 pseudo-chromosomes (2n = 78) accounting for ∼88.36% of the goose genome. Compared with previous goose assemblies, our assembly has more continuity, completeness, and accuracy; the annotation of core eukaryotic genes and universal single-copy orthologs has also been improved. We have identified 17,568 protein-coding genes and a repeat content of 8.67% (96.57 Mb) in this genome assembly. We also explored the spatial organization of chromatin and gene expression in the goose liver tissues, in terms of inter-pseudo-chromosomal interaction patterns, compartments, topologically associating domains, and promoter-enhancer interactions. Under acute hypoxia, multiple ion channels on the cell membrane are activated, causing cell swelling and eventually necrosis. LRRC8A is an indispensable protein of the volume-regulated anion channel (VRAC), which participates in swelling and the acceleration of cell necrosis. In this study, we revealed a dynamic change in the expression level of the LRRC8 family during hypoxia in 3T3-L1 cells. The disruption of LRRC8A in 3T3-L1 cells was also associated with a significant anti-necrotic phenotype upon hypoxia accompanied by the reduced expression of necrosis-related genes. In vivo, differential expression of LRRC8 family members was also identified between high-altitude pigs and their low-altitude relatives. Taken these findings together, this study demonstrates the involvement of LRRC8A in hypoxia-induced cell necrosis. The transition of terrestrial snakes to marine life ∼10 Ma is ideal for exploring adaptive evolution. Sea snakes possess phenotype specializations including laterally compressed bodies, paddle-shaped tails, valvular nostrils, cutaneous respiration, elongated lungs, and salt glands, yet, knowledge on the genetic underpinnings of the transition remains limited. Herein, we report the first genome of Shaw's sea snake (Hydrophis curtus) and use it to investigate sea snake secondary marine adaptation. A hybrid assembly strategy obtains a high-quality genome. Gene family analyses date a pulsed coding-gene expansion to ∼20 Ma, and these genes associate strongly with adaptations to marine environments. Analyses of selection pressure and convergent evolution discover the rapid evolution of protein-coding genes, and some convergent features. Additionally, 108 conserved noncoding elements appear to have evolved quickly, and these may underpin the phenotypic changes. Transposon elements may contribute to adaptive specializations by inserting into genomic regions around functionally related coding genes. The integration of genomic and transcriptomic analyses indicates independent origins and different components in sea snake and terrestrial snake venom; the venom gland of the sea snake harbors the highest PLA2 (17.23%) expression in selected elapids and these genes may organize tandemly in the genome. We investigated the chemical evolution of HC3N in six dense molecular clouds, using archival available data from the Herschel infrared Galactic Plane Survey (Hi-GAL) and the Millimeter Astronomy Legacy Team Survey at 90 GHz (MALT90). Radio sky surveys of the Multi-Array Galactic Plane Imaging Survey (MAGPIS) and the Sydney University Molonglo Sky Survey (SUMSS) indicate these dense molecular clouds are associated with ultracompact H  ii (UCH  ii) regions and/or classical H  ii regions. We find that in dense molecular clouds associated with normal classical H  ii regions, the abundance of HC3N begins to decrease or reaches a plateau when the dust temperature gets hot. This implies UV photons could destroy the molecule of HC3N. On the other hand, in the other dense molecular clouds associated with UCH  ii regions, we find the abundance of HC3N increases with dust temperature monotonously, implying HC3N prefers to be formed in warm gas. We also find that the spectra of HC3N (10-9) in G12.804−0.199 and RCW 97 show wing emissions, and the abundance of HC3N in these two regions increases with its non-thermal velocity width, indicating HC3N might be a shock origin species. We further investigated the evolutionary trend of N (N2H+)/ N (HC3N) column density ratio, and found this ratio could be used as a chemical evolutionary indicator of cloud evolution after the massive star formation is started. The strong spatial confinement of a nanocavity plasmonic field has made it possible to visualize the inner structure of a single molecule and even to distinguish its vibrational modes in real space. With such ever-improved spatial resolution, it is anticipated that full vibrational imaging of a molecule could be achieved to reveal molecular structural details. Here we demonstrate full Raman images of individual vibrational modes at the ångström level for a single Mg-porphine molecule, revealing distinct characteristics of each vibrational mode in real space. Furthermore, by exploiting the underlying interference effect and Raman fingerprint database, we propose a new methodology for structural determination, which we have called 'scanning Raman picoscopy', to show how such ultrahigh-resolution spectromicroscopic vibrational images can be used to visually assemble the chemical structure of a single molecule through a simple Lego-like building process. Objective Growing evidence indicates that microRNAs (miRNA) play a critical role in the pathogenesis of OA, and overexpressing or silencing miRNA expression in OA models can contribute to the development of miRNA-based therapeutics. The objective of this study was to determine whether intra-articular injection of miRNA can inhibit OA progression. Methods The miRNA expression profile was determined in OA cartilage tissues and controls. Functional analysis of the miRNAs on extracellular matrix degradation was performed after miRNA mimic or inhibitor transfection. Luciferase reporter assays and western blotting were employed to determine miRNA targets. To investigate the functional mechanism of miR-21-5p in OA development, miR-21-5pfl/flCol2a1-CreER and wild-type mice were subject to surgical destabilization of the medial meniscus. Therapeutically, wild-type mice undergoing surgical destabilization of the medial meniscus were treated with intra-articular injection of agomir- and antagomir-21-5p. Results We found that expression of miR-21-5p was significantly up-regulated in OA cartilage tissues. The articular cartilage degradation of miR-21-5p conditional knockout mice was significantly alleviated compared with that of wild-type mice in spontaneous and destabilization of the medial meniscus models. Through gain-of-function and loss-of-function studies, miR-21-5p was shown to significantly affect matrix synthesis genes expression, and chondrocyte proliferation and apoptosis. Further, fibroblast growth factor 18 (FGF18) was identified as a target of miR-21-5p. Intra-articular injection of antagomir-21-5p significantly attenuated the severity of experimental OA. Clinically, FGF18 expression level was correlated with miR-21-5p expression and a modified Mankin scale. Conclusion Our findings reveal a miRNA functional pathway important for OA development, highlighting miRNA-21-5p silencing as an attractive therapeutic regimen in future clinical trials involving patients with OA. Purpose: We assessed the discrimination of lean mass estimates that have been adjusted for adiposity for physical functioning deficits and prediction of incident disability.Methods: Included were 2,846 participants from the Health, Aging and Body Composition Study with available whole-body dual energy absorptiometry measures of appendicular lean mass index (ALMI, kg/m2) and fat mass index (FMI, kg/m2). Age-, sex-, and race-specific Z-Scores and T-Scores were determined by comparison to published reference ranges. ALMI values were adjusted for FMI (ALMIFMI) using a novel published method. Sex-stratified analyses assessed associations between lean mass estimates and the physical performance score, ability to complete a 400-meter walk, grip strength, and incident disability. Dichotomized definitions of low lean for age and sarcopenia were examined and their performance compared to the ALM-to-BMI ratio.Results: Compared to ALMI T-Scores and Z-Scores, the ALMIFMI scores demonstrated stronger associations with physical functioning, and were similarly associated with grip strength. Greater FMI Z-Scores and T-Scores were associated with poor physical functioning and incident disability. Definitions of low lean for age and sarcopenia using ALMIFMI (compared to ALMI) better discriminated those with poor physical functioning and a greater risk of incident disability. The ALM-to-BMI ratio was modestly associated with grip strength and physical performance, but was not associated with completion of the 400-meter walk or incident disability, independent of adiposity and height.Conclusion: Estimation of skeletal muscle mass relative to adiposity improves correlations with physical performance and prediction of incident disability suggesting it is an informative outcome for clinical studies. Abstract: Brain sizes vary substantially across vertebrate taxa, yet, the evolution of brain size appears tightly linked to the evolution of life histories. For example, larger brained species generally live longer than smaller brained species. A larger brain requires more time to grow and develop at a cost of exceeded gestation period and delayed weaning age. The cost of slower development may be compensated by better homeostasis control and increased cognitive abilities, both of which should increase survival probabilities and hence life span. To date, this relationship between life span and brain size seems well established in homoeothermic animals, especially in mammals. Whether this pattern occurs also in other clades of vertebrates remains enigmatic. Here, we undertake the first comparative test of the relationship between life span and brain size in an ectothermic vertebrate group, the anuran amphibians. After controlling for the effects of shared ancestry and body size, we find a positive correlation between brain size, age at sexual maturation, and life span across 40 species of frogs. Moreover, we also find that the ventral brain regions, including the olfactory bulbs, are larger in long‐lived species. Coptotermes suzhouensis (Isoptera: Rhinotermitidae) is a significant subterranean termite pest of wooden structures and is widely distributed in southeastern China. The complete mitochondrial DNA sequence of C. suzhouensis was analyzed in this study. The mitogenome was a circular molecule of 15,764 bp in length, which contained 13 protein-coding genes (PCGs), 22 transfer RNA genes, two ribosomal RNA genes, and an A+T-rich region with a gene arrangement typical of Isoptera mitogenomes. All PCGs were initiated by ATN codons and terminated by complete termination codons (TAA), except COX2, ND5, and Cytb, which ended with an incomplete termination codon T. All tRNAs displayed a typical clover-leaf structure, except for tRNASer(AGN), which did not contain the stemloop structure in the DHU arm. The A+T content (69.23%) of the A+T-rich region (949 bp) was higher than that of the entire mitogenome (65.60%), and two different sets of repeat units (A+B) were distributed in this region. Comparison of complete mitogenome sequences with those of Coptotermes formosanus indicated that the two taxa have very high genetic similarity. Forty-one representative termite species were used to construct phylogenetic trees by maximum likelihood, maximum parsimony, and Bayesian inference methods. The phylogenetic analyses also strongly supported (BPP, MLBP, and MPBP = 100%) that all C. suzhouensis and C. formosanus samples gathered into one clade with genetic distances between 0.000 and 0.002. Adipogenesis involves a highly orchestrated series of complex events in which microRNAs (miRNAs) may play an essential role. In this study, we found that the miR-185 expression increased gradually during 3T3-L1 cells differentiation. To explore the role of miR-185 in adipogenesis, miRNA agomirs and antagomirs were used to perform miR-185 overexpression and knockdown, respectively. Overexpression of miR-185 dramatically reduced the mRNA expression of the adipogenic markers,PPARγ,FABP4,FAS, andLPL, and the protein level of PPARγand FAS. MiR-185 overexpression also led to a notable reduction in lipid accumulation. In contrast, miR-185 inhibition promoted differentiation of 3T3-L1 cells. By target gene prediction and luciferase reporter assay, we demonstrated that sterol regulatory element binding protein 1 (SREBP-1) may be the target of miR-185. These results indicate that miR-185 negatively regulates the differentiation of 3T3-L1 cells by targeting SREBP-1, further highlighting the importance of miRNAs in adipogenesis. Sexual selection theory predicts a trade-off between premating (ornaments and armaments) and postmating (testes and ejaculates) sexual traits, assuming that growing and maintaining these traits is costly and that total reproductive investments are limited. The number of males in competition, the reproductive gains from investing in premating sexual traits, and the level of sperm competition are all predicted to influence how males allocate their finite resources to these traits. Yet, empirical examination of these predictions is currently scarce. Here, we studied relative expenditure on pre- and postmating sexual traits among frog species varying in their population density, operational sex ratio, and the number of competing males for each clutch of eggs. We found that the intensifying struggle to monopolize fertilizations as more and more males clasp the same female to fertilize her eggs shifts male reproductive investment toward sperm production and away from male weaponry. This shift, which is mediated by population density and the associated level of male-male competition, likely also explains the trade-off between pre- and postmating sexual traits in our much broader sample of anuran species. Objectives. To determine whether a novel measure of appendicular lean mass relative to fat mass is associated with physical functioning in RA. Methods. In a cross-sectional design, three independent RA cohorts were retrospectively analysed. Wholebody DXA measures of appendicular lean mass index (ALMI, kg/m²) and fat mass index (FMI, kg/m²) were converted to age, sex and race-specific Z-scores using published National Health and Nutrition Examination Survey reference ranges. Adiposity-adjusted ALMI Z-scores (ALMIFMI) were determined using a published method to adjust for normal associations between ALMI and FMI Z-scores. Associations between ALMI Z-scores, ALMIFMI Z-scores and physical functioning were assessed after adjusting for age, sex and study. Functional outcomes assessed included the HAQ, Valued Life Activities assessment and Short Physical Performance Battery. Low lean for age was defined as a Z-score of -1 or less. Results. Our sample consisted of 442 patients with RA. The combined cohort had a mean ALMI Z-score of-0.51 (1.08) and a mean ALMIFMI Z-score of -0.58 (1.53), suggesting muscle mass deficits compared with a nationally representative sample. Greater ALMIFMI Z-scores demonstrated stronger associations with better functional outcomes compared with ALMI Z-scores. Associations were not attenuated with adjustment for systemic inflammation or pain. The FMI Z-score was independently associated with physical functioning, with a stronger association seen among patients with greater FMI Z-score. Adiposityadjusted definitions of low lean mass more clearly identified those with functional impairment. Conclusion. Background. The prognosis of patients with hepatocellular carcinoma (HCC) without portal vein tumor thrombosis (PVTT) after curative resection is at variance. We identified the risk factors of poor postoperative prognosis and consequently developed prognostic nomograms generating individual risk of death and recurrence for this subgroup of patients with HCC. Methods. The risk factors were identified and nomograms were developed based on a retrospective study of 734 patients in the primary cohort who underwent curative resection for HCC from 2010 to 2012. The predictive accuracy and discriminative ability of the nomograms were determined by concordance index (Cindex) and calibration curve and compared with traditional staging systems of HCC. The results were validated in an independent cohort of 349 patients operated at the same institution in 2007. Results. All of the independent factors for survival in multivariate analysis in the primary cohort were selected into the nomograms. The calibration curve for probability of survival showed good agreement between prediction by nomograms and actual observation. The C-indices of the nomograms for predicting overall survival and recurrence-free survival were 0.755 (95% confidence interval [CI], 0.752-0.758) and 0.665 (95% CI, 0.662-0.668), respectively, which were statistically higher than the C-indices of other HCC prognostic models. The results were further confirmed in the validation cohort. Conclusion. Increasing evidence suggests that micro (mi)RNAs play important roles in various biological process. To evaluate the roles of miRNA in the porcine liver, we investigated the dynamic profiles of microRNAomes using liver tissue from pigs during the embryonic period (embryonic day 90), weaning stage (postnatal day 30), and adult stage (7 years old). A total of 186 unique miRNAs were differentially expressed during liver development. We also identified that 17, 13, and 6 miRNAs were specifically abundant at embryonic day 90, postnatal day 30, and at 7 years, respectively. Besides regulating basic cellular roles in development, miRNAs expressed at the three developmental stages also participated in regulating “embryonic liver development,” “early hepatic growth and generating a functioning liver,” and “energy metabolic processes,” respectively. Background: Fatigue is a common symptom in patients with heart failure that is easy to ignore. In addition, fatigue may affect patients’ physical function and psychosocial conditions that can impair their quality of life. An effective nursing care programme is required to alleviate patients’ fatigue and improve their quality of life. Aim: To investigate the effects of a supportive educational nursing care programme on fatigue and quality of life in patients with heart failure. Methods: A randomised controlled trial design was used. Ninety-two patients with heart failure were randomly assigned to an intervention group (n=47) or a control group (n=45). The patients in the intervention group participated in 12 weeks of a supportive educational nursing care programme including fatigue assessment, education, coaching self-care and evaluation. The intervention was conducted by a cardiac nurse during four face-to-face interviews and three follow-up telephone interviews. Fatigue and quality of life were assessed at the baseline and 4 weeks, 8 weeks and 12 weeks after enrollment in both groups. Results: The participants in the intervention group exhibited a significant decrease in the level of fatigue after 12 weeks, whereas those in the control group exhibited no significant changes. Compared with the control group, the intervention group exhibited a significantly greater decrease in the level of fatigue and significantly greater improvement in quality of life after 12 weeks of intervention. Aims Deserts are one of the ecosystems most sensitive to global climate change. However, there are few studies examining how changing abiotic and biotic factors under climate change will affect plant species diversity in the temperate deserts of Asia. This study aimed to: (i) characterize species distributions and diversity patterns in an Asian temperate desert; and (ii) to quantify the effects of spatial and environment variables on plant species diversity. Methods We surveyed 61 sites to examine the relationship between plant species diversity and several spatial/environmental variables in the Gurbantunggut Desert. Spatial and environmental variables were used to predict plant species diversity in separate multiple regression and ordination models. Variation in species responses to spatial and environmental conditions was partitioned by combining these variables in a redundancy analysis (RDA) and by creating multivariate regression trees (MRT). Important Findings We found 92 plant species across the 61 sites. Elevation and geographic location were the dominant environmental factors underlying variation in site species richness. A RDA indicated that 93% of the variance in the species-environment relationships was explained by altitude, latitude, longitude, precipitation and slope position. Precipitation and topographic heterogeneity, through their effects on water availability, were more important than soil chemistry in determining the distribution of species. MRT analyses categorized communities into four groups based on latitude, soil pH and elevation, explaining 42.3% of the standardized species variance. Soil pH strongly influenced community composition within homogeneous geographic areas. Our findings suggest that precipitation and topographic heterogeneity, rather than edaphic heterogeneity, are more closely correlated to the number of species and their distributions in the temperate desert. The VD4 region from the Chlamydia trachomatis major outer membrane protein contains important neutralizing B-cell epitopes of relevance for antibody-mediated protection against genital tract infection. We developed a multivalent vaccine construct based on VD4s and their surrounding constant segments from serovars D, E, and F. Adjuvanted with cationic liposomes, this construct promoted strong immune responses to serovar-specific epitopes, the conserved LNPTIAG epitope and neutralized serovars D, E, and F. Vaccinated mice were protected against challenge, with protection defined as reduced bacterial numbers in vagina and prevention of pathological changes in the upper genital tract. Adoptive transfer of serumand T-cell depletion experiments demonstrated a dominant role for antibodies and CD4+ T cells in the protective immune response. Integrating a multivalent VD4 construct into the sequence of the major outer membrane protein resulted in a protective and broadly neutralizing vaccine. Aims Exploring flowering patterns and detecting processes are essential when probing into the nature of reproductive traits during the life history and the interactions among different evolutionary clades. Such patterns are believed to be influenced by many factors, but quantifying these impacts at the community-level remains poorly understood. Methods We investigated the flowering patterns based on long-term herbarium records in a given area from subtropical forest regions in southern China. We obtained 5258 herbarium voucher specimens collected from the Dinghushan National Nature Reserve (DNNR) belonging to 166 families, 943 genera and 2059 species and examined the month when each species was flowering during the period 1920-2007. Important Findings The results showed that plants flowered sequentially almost throughout the whole year, showing the characteristics of subtropical evergreen broad-leaved forests. Flowering spectrums of the entire flora and the four life forms exhibited a clear unimodality that is probably typical of subtropical forest communities. Flowering patterns of the DNNR were positively correlated with mean rainfall, mean air temperature and mean sunshine duration. Median flowering dates of the 38 large species-rich families ranged from early April to late August; 25 families exhibited significant unimodal distribution patterns, whereas the remaining families were unclear or bimodal. Median flowering dates of the 10 most species-rich genera ranged from middle May to later July. While the results are consistent with climatic factors playing a general role in flowering patterns, median flowering dates varied significantly among species-rich families and genera, suggesting that phylogenies could provide specific constraints in subtropical forests. This study aims to characterize antimicrobial resistance and antimicrobial resistance genetic determinants of an Escherichia coli clinical isolate HD0149 from China in 2012. This strain displayed high-level resistance to cephalosporins, carbapenems, fluoroquinolones, aminoglycosides and fosfomycin. A range of antimicrobial resistance genes was detected responsible for its multiple antimicrobial resistances, involving the blaCMY-2, blaCTX-M-65, blaNDM-1, blaSFO-1, blaTEM-1, fosA3, rmtB, sul1 and sul2 genes. Four amino acid substitutions were detected in the quinolone resistance-determining regions (QRDRs) of GyrA (S83L and D87N), ParC (S80I) and ParE (S458A). Conjugation experiments revealed two multiresistance plasmids present in E. coli HD0149. The blaSFO-1 gene associated with blaNDM-1 gene was located in a 190 kb IncA/C plasmid and the blaCTX-M-65, fosA3 and rmtB genes were located in a 110 kb IncF plasmid. This is the first identification of the blaSFO-1 gene in an E. coli isolate and on a conjugative IncA/C plasmid. This may dramatically enhance the international prevalence and dissemination of blaSFO-1 among Enterobacteriaceae. [ABSTRACT FROM AUTHOR]

The physical, chemical and kinetic characteristics of 12 northern infrared dark clouds (IRDCs) are systematic studied using the 13CO (1–0) and C18O (1–0) lines, observed with the Purple Mountain Observatory 13.7 m radio telescope, the 1.1 mm Bolocam Galactic Plane Survey data and Galactic Legacy Infrared Mid-Plane Survey Extraordinaire Spitzer Infrared Array Camera 8 μm data. The molecular lines emission and 1.1 mm continuum emission almost coincide in morphology for each IRDC and both are associated well with the IRDCs. 10 IRDCs present the filamentary structure and substructures. Totally, 41 IRDC cores are identified, and a statistic research for them shows that the northern IRDC cores have a typical excitation temperature of 8–10 K, an integrated intensity ratio of 13CO to C18O of 3–6 and the column density (1–6) × 1022 cm−2. About 57.5 per cent of the IRDC cores are gravitationally bound, which are more compact, warmer and denser. In addition, we study the mass distribution functions of the whole IRDC cores as well as the gravitationally bound cores, finding that they almost have the same power-law indices. This indicates that the evolution of the IRDC cores almost has no effect on the mass spectrum of the molecular cores and thus can be used to study the stellar initial mass function. Moreover, three IRDC cores G24.00-3, G31.38-1 and G34.43-4 are detected to have large-scaled infall motions. Broad-spectrum disease resistance is a highly valuable trait in plant breeding and attracts special attention in research. The Arabidopsis gene locus RESISTANCE TO POWDERY MILDEW 8 (RPW8) contains two adjacent homologous genes, RPW8.1 and RPW8.2, and confers broad-spectrum resistance to powdery mildew. Remarkably, the RPW8.2 protein is specifically localized to the extrahaustorial membrane (EHM) encasing the feeding structure of powdery mildew whereby RPW8.2 activates haustorium-targeted defenses. Here, we show that ectopic expression of the yellow fluorescent protein (YFP)-tagged RPW8.1 from the native promoter leads to unique cell death lesions and enhances resistance to virulent fungal and oomycete pathogens that cause powdery mildew and downy mildew diseases, respectively. In powdery mildew-infected plants, RPW8.1–YFP accumulates at higher levels in the mesophyll cells underneath the infected epidermal cells where RPW8.2–YFP is mainly expressed. This cell type-preferential protein accumulation pattern largely correlates with that of H2O2 accumulation, suggesting that RPW8.1 may spatially collaborate with RPW8.2 in activation of resistance to powdery mildew. Interestingly, when ectopically expressed from the RPW8.2 promoter, RPW8.1–YFP is also targeted to the EHM of powdery mildew and the transgenic plants display resistance to both powdery mildew and downy mildew. Using YFP as a reporter, we further reveal that the RPW8.1 promoter is constitutively active but induced to higher levels in cells at the infection site, whereas the RPW8.2 promoter is activated specifically in cells at the infection site. Taken together, our results suggest that RPW8.1 (and its promoter) is functionally distinct from RPW8.2 and may have a higher potential in engineering broad-spectrum resistance in plants. Mutation of the gene cl7(t), a novel allele of dep2, increases the force required to open the lemma and palea, and results in weak swelling ability in the lodicules, leading to cleistogamy.Gene transformation is an important method for improvement of plants into elite varieties. However, the possibility of gene flow between genetically modified (GM) crops and similar species is a serious public issue that may potentially endanger ecological stability. Cleistogamy is expected to be an ideal genetic tool for preventing transgene propagation from GM crops. A rice mutant, cl7(t), was created by ethyl methanesulfonate mutagenesis. The mutant exhibited cleistogamy, and had closed spikelets, reduced plant height, and altered morphology of the leaves, panicle, and seeds. Anatomical investigations revealed that the cl7(t) mutant contained more vascular bundles and thicker stems than the wild type, which increased the mechanical strength of its internodes, and anti-lodging ability. Further studies demonstrated that the force required to open the lemma and palea was higher in the cl7(t) mutant, and there was weak swelling ability in the lodicules, which leads to cleistogamy. Allelic analyses and complementation tests indicated that cl7(t) was a novel allele of dep2, a mutant that was previously reported to have similar panicle morphology. Sequence analysis showed that cl7(t) had a single nucleotide substitution (C to A) in the third exon that leads to a Ser substitution with a stop codon, giving a truncated DEP2 protein. Quantitative RT-PCR and in situ hybridization tests demonstrated that there was lower CL7(t) expression level in the spikelets and weaker CL7(t) signals in the lodicules of the cl7(t) mutant compared with wild type, which implies that CL7(t) might participate in the development of lodicules. To improve the agronomic traits of cl7(t) to fit the needs of field production, the cl7(t) mutant was crossed with an intermediate-type rice variety named Guanghui102, which bears some important agronomic traits, including increased grain numbers and high rate of seed setting. Through multi-generational pedigree selection, cleistogamy lines with improved economic traits were obtained, which can be used for the selection of ecologically safe GM rice varieties.
