A new two-site ELISA was validated for ovine plasma and used to measure circulating inhibin-A concentrations during a synchronized oestrous cycle in four ewes and throughout pregnancy in six ewes. Inhibin A concentrations were also determined in four ewes during chronic treatment with a GnRH agonist and after subsequent exposure to pregnant mares' serum gonadotrophin (PMSG) to stimulate ovarian follicular development. Concentrations of FSH, LH, oestradiol and progesterone were determined by radioimmunoassay. The detection limit of the inhibin-A ELISA was approximately 50 pg ml-1 and no significant crossreaction was observed with a range of related molecules including activin-A, inhibin-B, activin-B, follistatin and alpha 2-macroglobulin. Inhibin-A concentrations were below the detection limit in plasma from hypophysectomized and ovariectomized ewes. During the oestrous cycle, plasma inhibin-A concentrations (approximately 0.3-0.4 ng ml-1) did not vary during the follicular phase whereas plasma oestradiol increased approximately tenfold. After the preovulatory LH/FSH surge, inhibin-A fell to a nadir (approximately 0.15 ng ml-1) coincident with the peak of the postovulatory FSH rise. During the next 2 days, FSH concentrations fell to basal values as inhibin-A concentrations increased (P < 0.05) to a peak (approximately 0.5 ng ml-1) 3 days after the preovulatory LH/FSH surge. Over the following 3 days, FSH values increased again (P < 0.05) as inhibin-A concentrations fell to approximately 0.25 ng ml-1 (P < 0.05). Chronic GnRH agonist treatment suppressed FSH concentrations by about 50%, while inhibin-A and oestradiol concentrations fell below detection limits. Within 2 days after the PMSG injection, concentrations of inhibin-A (approximately 4.5 ng ml-1) and oestradiol (approximately 20 pg ml-1) had increased to very high values, while FSH concentrations had been reduced by a further 50%. Plasma concentrations of inhibin-A and FSH were similar to those in nonpregnant ewes during the first 60 days of gestation, but inhibin-A values fell markedly (sevenfold; P < 0.01) between days 60 and 90, coincident with a twofold decrease in FSH (P < 0.05). Inhibin A and FSH concentrations remained low for the remainder of gestation and were positively correlated throughout pregnancy (r = 0.48; P < 0.005). These observations support an endocrine feedback role for ovarian inhibin-A and oestradiol in controlling the secondary (postovulatory) FSH surge in ewes, but indicate that an increase in oestradiol is responsible for the characteristic reduction in FSH during the early to mid-follicular phase. The reduced secretion of FSH from mid- to late pregnancy cannot be attributed to increased inhibin-A secretion by the feto-placental unit, but most likely reflects increased steroid secretion from this source. more...

To investigate the mechanism controlling the fall in maternal pituitary responsiveness to GnRH, LH synthesis and pituitary GnRH receptor content during pregnancy, maternal pituitaries were collected from sheep on days 35, 45, 60, 90, 110, 125 and 135 of pregnancy. Circulating steroids and gonadotrophins were determined in blood samples collected from these ewes immediately before death. Pituitary blocks from each ewe were perifused with either medium alone (control) or medium supplemented with oestradiol, oestradiol plus progesterone or oestradiol plus RU486, for 150 min before administration of two 15 s GnRH pulses 90 min apart. The amounts of mRNA encoding LH beta and GnRH receptor were determined in pituitary tissue fragments snap-frozen in liquid N2 at the time of collection from the ewes. While basal LH secretion fell during pregnancy, pituitary responsiveness to GnRH remained high (up to seven times basal LH concentrations). After day 90, the first GnRH pulse elicited LH peaks equivalent to the LH peaks produced by the second GnRH pulse. Therefore, GnRH self-priming was not evident possibly because the pituitaries were constantly primed by increased concentrations of maternal oestradiol. Around day 90, circulating concentrations of progesterone rose from 7.8 +/- 1.5 to 12.2 +/- 3.8 ng ml-1. Up to day 60, oestradiol in the perifusion buffer had stimulatory effects on LH secretion although this was reduced by RU486. By day 125, the content of mRNA encoding LH beta had declined during pregnancy to 7% of the content on day 35, although the content of mRNA encoding GnRH receptor remained unchanged. From these data, there appears to be a transitional period at around day 90 of gestation when pituitary sensitivity to steroids in vitro is lost together with detectable GnRH self-priming. In conclusion, the marked decline in pituitary amounts of mRNA encoding LH beta, but not in GnRH responsiveness or expression of GnRH receptor, after day 45 of pregnancy suggests that the principal effect of pregnancy on gonadotroph function is mediated via a mechanism other than reduced pituitary amounts of GnRH receptors. Two possible mechanisms are (1) a reduction in GnRH output leading to lowered LH synthesis, or (2) the presence of an inhibitory factor with a short half-life in the maternal circulation. more...

It has been suggested that in superovulated women the endogenous LH surge is attenuated by a non-steroidal factor, called gonadotrophin surge-attenuating factor (GnSAF), which reduces gonadotrophin secretion in response to LHRH. To determine whether human follicular fluid (hFF) from superovulated women contains GnSAF activity, the secretion of LH and FSH by cultured sheep pituitaries was studied. After charcoal extraction of steroids, hFF was treated by heparin/Sepharose chromatography, which reversibly binds inhibin. The effects of whole hFF and the bound and unbound fractions on basal and LHRH-induced gonadotrophin secretion were then assessed. Steroid-free hFF significantly reduced basal FSH, but not basal LH, secretion, and significantly attenuated the LH and FSH responses to LHRH. The bound (inhibin) fraction significantly decreased both basal and LHRH-induced FSH secretion but did not affect LH release. The unbound fraction had no effect on basal LH or FSH secretion, but significantly attenuated LHRH-induced secretion of both LH and FSH. We conclude that the unbound fraction of hFF from superovulated women contains GnSAF. A technique is described for the in-vivo determination of mammary gland size and gross composition in goats by using nuclear magnetic resonance imaging (MRI). The volume of test objects determined with MRI had an error of +0.4 +/- 1.6% of the actual volume. In lactating goats the in-vivo MRI estimate of mammary parenchymal volume was significantly greater than, but highly significantly correlated with, the weight of parenchyma determined post mortem (for the whole udder, r = 0.88, P less than 0.001; for individual glands, r = 0.85, P less than 0.001). MRI-determined estimates of the volume of fluid within the mammary gland were within 1.2% of the volume of milk removed from the udders after imaging. The spin-lattice (T1) relaxation time of the whole udder correlated closely with the volume of fluid within the udder. Mammary development and regression were measured in goats in vivo using magnetic resonance imaging (MRI). Measurements were made during the first and second cycles of pregnancy, lactation and involution. In primiparous goats, and exponential pattern of growth was evident during gestation and for the first 2 weeks of lactation. Parenchyma volume correlated significantly with milk yield across goats during early lactation, and across stage of lactation within goats. Milking was discontinued in Week 26 of the first lactation. Involution was characterized by an initial accumulation of fluid (over 2 days) followed by reabsorption; parenchyma volume did not decrease significantly until the 3rd week of involution, which was also the time at which these goats were mated to start their second gestation. Their udders still contained significant quantities of fluid (40-60% of the gross volume), but parenchyma volume was also greater (by 4.7-fold) than in goats beginning their first gestation. By Week 15 of gestation there was no longer a parity difference in parenchyma; the udders of first-gestation goats had grown significantly, but those of second-gestation goats had not. Conversely, between gestation Week 15 and lactation Week 2 mammary growth was significantly more rapid in the second cycle, such that the udder was larger at the start of the second lactation. more...

Simultaneous telemetry of the body and testis temperatures of 8 hedgehogs was carried out during hibernation and during sexual reactivation in spring. Between October and January, when the testes were involuted, the body/testis temperature differential was variable, with mean daily testis temperatures up to 1 degrees C warmer than body temperatures. From mid-February onwards, when plasma testosterone approached maximal concentrations, mean testicular temperatures stabilized 1.4 +/- 0.2 degrees C below body temperatures. During spermatogenesis testicular temperature of hedgehogs was significantly lower than body temperature. Over the euthermic body temperature range of 34.7-36.2 degrees C, testicular temperatures varied from 34.0 to 34.9 degrees C. Only at body temperatures over 36.2 degrees C did testicular temperature reach 35 degrees C. During spermatogenesis hedgehog testis temperatures are similar to those of many scrotal mammals. more...

Adult hedgehogs, maintained in captivity under natural environmental conditions of photoperiod and ambient temperature, were bled monthly. Plasma was assayed for melatonin, testosterone, prolactin, thyroxin, beta-endorphin and both plasma and urine for cortisol. Melatonin concentrations followed a circannual pattern, maximal between November and February at photoperiods less than 10L:14D, which is suggested as a key photoperiod. beta-Endorphin concentrations were maximal between March and September, coinciding with the reproductively active season of the hedgehog. Prolactin values were elevated during hibernation, indicating continued hypothalamo-pituitary axis activity. Testosterone and thyroxin levels were high between February and July and February and August respectively. During spring thyroxin concentrations rose 1 month later in females than males, reflecting the earlier arousal of the males from hibernation. There were no marked seasonal cycles of plasma or urinary cortisol. The results indicate photoperiod as the main factor in regulating hedgehog seasonality, with melatonin, beta-endorphin and prolactin important in the timing of reactivation of reproduction. Sexual differences in hedgehogs suggests environmental fine tuning of endogenous cycles, males being ready to inseminate females early in spring, while females only begin full breeding activity when conditions are suitable. more...

The body temperatures of 3 breeding and 7 non-breeding female hedgehogs were monitored during 3 years. Lactating females had significantly higher and less variable body temperatures than did pregnant and non-breeding hedgehogs. Breeding females had circadian body temperature cycles that were more biphasic than those of non-breeders. Females maintaining higher mean daily body temperatures showed less variability about the mean than those exhibiting lower mean daily body temperatures. The rate at which body temperatures became more variable as mean daily body temperatures fell was 4.6 times more rapid in non-breeding females than in pregnant and lactating hedgehogs. With the exception of a single female in late gestation, hedgehogs readily became torpid under conditions of low ambient temperature and short photoperiod during summer, particularly when their food intake was restricted. more...
