With the increasing market demand for high-quality aquatic products, the application of industrialized aquaculture facilities may get more attention. In order to improve the poor performance of thermal insulation, the accuracy of the numerical model was verified in this study through actual measured data. The model verification results shown that the average relative errors of the measured and calculated values of indoor air temperature, water temperature and roof inner surface temperature in the industrialized aquaculture workshop is within 2.5%, it suggested that the numerical calculation results are accurate. Furthermore, the thermal environment and thermal insulation performance of industrialized aquaculture facilities in winter were conducted based on the numerical calculations. After optimized the thermophysical parameters of the workshop enclosure structure, we found that the water body temperature could reach 21°C (which was close to the breeding temperature of grouper (Epinephelinae). Therefore, the numerical calculation method was further used to analyze the energy consumption of aquaculture water in January of a typical year in this area by heating to three constant temperatures (22, 25, and 28°C). When the aquaculture water was heated to the three constant temperature states, it needed to consume 8.56×105, 1.02×106 and 1.22×106 MJ of energy respectively, which were equal to the amount of energy released by the complete combustion of 29.3, 35.1 and 41.8 t standard coal. Moreover, it is concluded that the artificial temperature increase in winter maintains the temperature in the range of 22~25°C to provide the highest heating efficiency. Water–salt stress and nutrient limitation may affect leaf economic spectrum of halophytes and confuse our understanding on plant physiological principles in a changing world. In this study, three halophytic plant communities of Phragmites australis, Suaeda salsa, and Tamarix chinensis, were selected in two sites (sites 1 and 2) on the west coast of Bohai Sea. The net photosynthetic rate (Pn), transpiration rate (Tr), stomatal conductance (Gs), leaf vapor pressure deficit (VPDleaf) and their influencing factors were studied to test the possible carbon assimilation strategies of the halophytes. P. australis had higher Pn, Tr, and Gs than S. salsa and T. chinensis in both sites. Similar trends were found for leaf P and photosynthetic N and P efficiency (PNUE and PPUE, respectively) in one or both sites. By contrast, the leaf dry mass per area (LMA) increased in the order of P. australis < S. salsa < T. chinensis in both sites. For identical species in different sites, Pn, leaf P, and PNUE were lower but Tr, VPDleaf, leaf N, leaf N:P, and PPUE were higher in site 1 than in site 2 for one or more halophytes. Although soil physicochemical properties in different sites explained several variations among the halophytes, two-way ANOVA indicated that the species can explain most of the leaf traits compared with the site. LMA also had significant nonlinear relationships with Pn, Tr, Gs, and VPDleaf. PNUE and PPUE showed positive correlation with Pn in both sites, but they decreased in the power-law function with increasing LMA. Overall, the redundancy analysis showed that the gas exchange capacity of the halophytic plant communities was significantly affected by PPUE (60.0% of explanation), PNUE (57.1%), LMA (35.0%), leaf P (22.0%), and soil N (15.8%). The giant pandas’ (Ailuropoda melanoleuca) reproductive strategy is unique among mammals. Yet there are characteristics of giant panda behaviour we do not fully understand. Probably one of the least understood is the assumption that in captivity virtually all giant panda females rear only one cub when twins are born and abandon the other if given the chance. So far, only two females have raised twins simultaneously, but just with intensive human assistance. This case-study marks the first successful rearing of giant panda twins in captivity entirely by the mother. Using video data for detailed behavioural observations, we provide the first behavioural assessment of a giant panda female raising two cubs simultaneously without direct human assistance or disturbance. We compared the maternal behaviour during the denning period of twin cubs raised in 2016 with two singleton cubs born 2007 and 2010. YANG YANG, the dam, rested less and interacted more with the twins than with the singletons in the first month postpartum and invested a greater part of her daily time budget on rearing the twins. Reproductive monitoring for captive breeding in giant pandas is based on behavioural observation and non-invasive hormone analysis. In urine, interpretation of results requires normalisation due to an animal’s changing hydration. Correction of urinary concentrations based on creatinine is the gold standard. In this study, a largely unexplored, easy-to-perform normalisation technique, based on urinary specific gravity (USpG), was examined and compared to creatinine. To this extent, six cycles from two female pandas (SB741(1) and SB569(5)) were monitored through urine analysis for oestrogen, progesterone, ceruloplasmin and 13,14-dihydro-15-keto-PGF2a (PGFM). The Pearson’s correlation between creatinine and USpG was high (r = 0.805–0.894; p < 0.01), indicative for a similar performance of both normalisation methods. However, generally lower values were observed during pro-oestrus and primary (progesterone) rise. This could be associated with huge shifts in appetite, monitored by faecal output (kg) with an averaged > 50% decrease during oestrus and >50% increase during primary progesterone rise. In parallel, respectively highest and lowest creatinine and USpG levels, were measured, with creatinine obviously more affected as a result of linkage with muscle tissue metabolism affected by reproductive hormones. As a consequence, metabolite levels were significantly different between both corrected datasets with significantly higher oestrogen peak levels during oestrus ranging from 2.13–86.93 and 31.61–306.45 ng/mL (USpG correction) versus 2.33–31.20 and 36.36–249.05 ng/mL Cr (creatinine correction) for SB569 and SB741 respectively, and significant lower progesterone levels during primary progesterone rise ranging from 0.35–3.21 and 0.85–6.80 ng/mL (USpG correction) versus 0.52–10.31 and 2.10–272.74 ng/mL Cr (creatinine correction) for SB569 and SB741 respectively. Consequently, USpG correction rendered unbiased profiles, less subject to variation and metabolic artefacts and therefore allowed a more straightforward identification of peak oestrogen and onset of secondary progesterone rise, being potentially advantageous for future studies unravelling key giant panda reproductive events, including (delayed) implantation. Baylisascaris schroederi is a common parasite of captive giant pandas. The diagnosis of this ascariasis is normally carried out by a sedimentation-floatation method or PCR to detect eggs in feces, but neither method is suitable for early diagnosis. Fatty acid-binding protein (FABP) and galectin (GAL) exist in various animals and participate in important biology of parasites. Because of their good immunogenicity, they are seen as potential antigens for the diagnosis of parasitic diseases. In this study, we cloned and expressed recombinant FABP and GAL from B. schroederi (rBs-FABP and rBs-GAL) and developed indirect enzyme-linked immunosorbent assays (ELISAs) to evaluate their potential for diagnosing ascariasis in giant pandas. Immunolocalization showed that Bs-FABP and Bs-GAL were widely distributed in adult worms. The ELISA based on rBs-FABP showed sensitivity of 95.8% (23/24) and specificity of 100% (12/12), and that based on rBs-GAL had sensitivity of 91.7% (22/24) and specificity of 100% (12/12). Variations of plant C: N: P stoichiometry could be affected by both some environmental fluctuations and plant physiological processes. However, the trade-off mechanism between them and their influencial factors were not understood completely. In this study, C, N, P contents and their stoichiometry of S. salsa’s plant organs (leaves, stems, and roots), together with their environmental factors including salinity, pH, soil N and soil P, were examined in the intertidal and supratidal habitats of coastal wetlands during the different sampling times (May, July, September, November). The results showed that both plant organ and sampling times affected C, N, and P and stoichiometry of S. salsa in the intertidal and supratidal habitats, however, their influencial conditions and mechanisms were different. In the intertidal habitat, the different slopes of C-P and N-P within interspecific organs suggested that plant P, C:P and N:P of S. salsa were modulated by P concentrations that allocated in the specific organs. However, the slopes of C-N were found to be not significant within interspecific organs, but during the sampling times. These differences of plant N and C:N were related with the physiological demand for N in the specific life history stage. In the supratidal habitat, no significant differences were found in the slopes of C-N, C-P, and N-P within interspecific organs. However, different slopes of C-N among the sampling times also indicated a self-regulation strategy for plant N and C:N of S. salsa in different ontogenetic stages. In contrast to the intertidal habitat, seasonal variations of P, C:P and N:P ratios within interspecific organs reflected the soil P characteristics in the supratidal habitat. In the giant panda, adaptation to a high-fiber environment is a first step for the adequate functioning of intestinal bacteria, as the high cellulose content of the gut due to the panda's vegetarian appetite results in a harsh environment. As an excellent producer of several enzymes and vitamins, Bacillus subtilis imparts various advantages to animals. In our previous study, we determined that several strains of B. subtilis isolated from pandas exhibited good cellulose decomposition ability, and we hypothesized that this bacterial species can survive in and adapt well to a high-fiber environment. To evaluate this hypothesis, we employed RNA-Seq technology to analyze the differentially expressed genes of the selected strain B. subtilis HH2, which demonstrates significant cellulose hydrolysis of different carbon sources (cellulose and glucose). In addition, we used bioinformatics software and resources to analyze the functions and pathways of differentially expressed genes. Interestingly, comparison of the cellulose and glucose groups revealed that the up-regulated genes were involved in amino acid and lipid metabolism or transmembrane transport, both of which are involved in cellulose utilization. Conversely, the down-regulated genes were involved in non-essential functions for bacterial life, such as toxin and bacteriocin secretion, possibly to conserve energy for environmental adaptation. The results indicate that B. subtilis HH2 triggered a series of adaptive mechanisms at the transcriptional level, which suggests that this bacterium could act as a probiotic for pandas fed a high-fiber diet, despite the fact that cellulose is not a very suitable carbon source for this bacterial species.
