Objective: To investigate the expression of long-chain non-coding RNA LINC00472 in renal fibrotic cell models and its functional role in renal tubular epithelial cell fibrosis and epithelial-mesenchymal transition (EMT) induced by TGF-β1. Methods: Recombinant human TGF-β1 was used to induce fibrosis and EMT in human renal tubular epithelial cells (HK-2). Real-time fluorescent quantitative PCR was used to detect the expression levels of fibrosis-related genes and LINC00472 mRNA in the cells, and Western blot was used to Western blot. Method was used to detect the protein expression level of fibrosis-related genes in cells, and the effect of LINC00472 expression on the migration ability of HK-2 cells was evaluated by scratch experiment. Results: TGF-β1 can successfully induce fibrosis and EMT in HK-2 cells, and inhibit the expression of LINC00472 in a dose- and time-dependent manner (P<0.05). Inhibition of LINC00472 further promoted the up-regulation of Fibronectin and Vimentin induced by TGF-β1, and the down-regulation of E-cadherin (P<0.05); while overexpression of LINC00472 could reverse the inducing effect of TGF-β1 on fibrosis-related genes (P<0.05). In addition, inhibiting LINC00472 can further enhance the HK-2 cell migration induced by TGF-β1 (P<0.05), while upregulating LINC00472 can inhibit cell migration (P<0.05). Conclusion: LINC00472 has a low expression in TGF-β1 induced renal fibrosis cell model, and the decrease of its expression level can promote cell fibrosis and EMT process. [ABSTRACT FROM AUTHOR]
