Solution-state NMR spectroscopy remains the primary method for characterising synthetic supramolecular assemblies. Yet, in their NMR spectra, relaxation interference effects can significantly alter peak intensities hindering interpretation. Here, we present a simple experiment for synthetic chemists to analyse this effect, allowing interpretation of these distorted spectra and validation of spectral assignments. We apply this experiment to synthetic porphyrin oligomers with molecular weights approaching those of protein domains (10 kDa). Our experiment provides a simple means to gain additional structural and dynamical information that will become increasingly useful as chemists create larger molecular architectures.

Bacterial metallo-β-lactamases (MBLs) are involved in resistance to β-lactam antibiotics including cephalosporins. Human SNM1A and SNM1B are MBL superfamily exonucleases that play a key role in the repair of DNA interstrand cross-links, which are induced by antitumour chemotherapeutics, and are therefore targets for cancer chemosensitization. We report that cephalosporins are competitive inhibitors of SNM1A and SNM1B exonuclease activity; both the intact β-lactam and their hydrolysed products are active. This discovery provides a lead for the development of potent and selective SNM1A and SNM1B inhibitors.

Methylation of 2,8-dimethyl-6H,12H-5,11-ethanodibenzo[b,f][1,5]-diazocine (ethano-Tröger's base) with methyl iodide followed by ion metathesis and fluorination with N-fluoro-2,3,4,5,6-pentachloropyridinium triflate affords a new electrophilic N-F reagent, that is more reactive than Selectfluor. 2D (19)F-(15)N HMQC experiments provide (1)JNF coupling constants which are diagnostic for the N-F functional group.

The ability of glycosyldiselenides to act as lectin ligands and their selective detection in plasma by (77)Se NMR is reported.

Metallo-β-lactamases (MBLs) catalyse the hydrolysis of almost all β-lactam antibiotics. We report biophysical and kinetic studies on the São Paulo MBL (SPM-1), which reveal its Zn(ii) ion usage and mechanism as characteristic of the clinically important di-Zn(ii) dependent B1 MBL subfamily. Biophysical analyses employing crystallography, dynamic 19 F NMR and ion mobility mass spectrometry, however, reveal that SPM-1 possesses loop and mobile element regions characteristic of the B2 MBLs. These include a mobile α3 region which is important in catalysis and determining inhibitor selectivity. SPM-1 thus appears to be a hybrid B1/B2 MBL. The results have implications for MBL evolution and inhibitor design.

2-Oxoglutarate and iron dependent oxygenases have potential for the stereoselective hydroxylation of amino acids and related compounds. The biochemical and kinetic properties of recombinant γ-butyrobetaine hydroxylase from human and Pseudomonas sp. AK1 were compared. The results reveal differences between the two BBOXs, including in their stimulation by ascorbate. Despite their closely related sequences, the two enzymes also display different substrate selectivities, including for the production of (di)hydroxylated betaines, implying use of engineered BBOXs for biocatalytic purposes may be productive.

A convenient method for the synthesis of fluoromethylated carnitine biosynthesis intermediates, i.e. fluorinated derivatives of γ-butyrobetaine and trimethyllysine, is described. The fluoromethylated probes were useful in both in vitro and cell based assays employing (19)F NMR and LC-MS analyses.

The trisubstituted enolate- and C-C bond-forming capacities of engineered carboxymethylproline synthases CMPSs are coupled with the malonyl-CoA synthetase MatB to enable stereoselective preparation of 5- and 6-membered N-heterocycles functionalised with alkyl-substituted carboxymethyl side chains, starting from achiral alkyl-substituted malonic acids and L-amino acid semialdehydes. The results illustrate the biocatalytic utility of crotonases in tandem enzyme-catalysed reactions for stereoselective synthesis.

Within cells it is proposed that a major mechanism for the metabolism of formaldehyde is via its reaction with glutathione (GSH) to form S-hydroxymethylglutathione (HMG), which undergoes subsequent oxidation. In addition to HMG and the previously reported (5R,10S)-5-(carboxymethylcarbamoyl)-7-oxo-3-thia-1,6-diazabicyclo[4.4.1]undecane-10-carboxylic acid (BiGF(2)), NMR studies on the reaction of GSH with formaldehyde reveal two previously unassigned monocyclic structures. The results imply that the biologically relevant reactions between aldehydes and peptides/proteins may be more complex than presently perceived.

Unprecedented bicyclic methylene aziridines are prepared by rhodium(II)-catalyzed allene aziridination of buta-2,3-dienyl carbamates. Aspects of their NMR and X-ray data are described and a preliminary reactivity profile is given, including overall S(N)V-mode ring-opening with organometallic reagents.

The utility of wild-type and variant carboxymethylproline synthases for biocatalysis was demonstrated by preparing functionalised 5-, 6- and 7-membered N-heterocycles from amino acid aldehydes and (alkylated) malonyl-coenzyme A derivatives; the N-heterocycles produced were converted to the corresponding bicyclic beta-lactams by a carbapenem synthetase.

Sulfate templation has been used in the synthesis of a novel tris-urea-based triply interlocked capsule, whose structure has been verified by DOSY NMR, mass spectrometry and molecular modelling investigations.

The binding kinetics of disaccharides trehalose and trehalose-6-phosphate to repressor protein TreR have been determined using STD NMR and shed light on the contrasting biological roles of these two sugars.

Treatment of 3-aminocyclohex-1-enes with mCPBA in the presence of trichloroacetic acid gives the corresponding 1,2-anti-2,3-syn-1-trichloroacetoxy-2-hydroxy-3-aminocyclohexane with high levels of diastereoselectivity (90% de). This is consistent with a mechanism of oxidation involving hydrogen-bonded delivery of the oxidant by the allylic ammonium ion formed in situ, followed by highly regioselective ring-opening of the intermediate epoxide by trichloroacetic acid. The effect of conformational constraints upon the oxidation reaction is also examined.

Gamma-turn analogues comprising a modified dipeptide constrained in an eleven-membered ring were prepared by alkene metathesis and analysed by NMR and molecular modelling studies. The results reveal that some of the cyclic analogues form inverse gamma-turns and preferentially adopt conformations determined by the identity of the incorporated amino acid residues and the nature of the constraining linker (E/Z-alkene or alkane).

Highly luminescent inclusion complexes consisting of poly(para-phenylene) (PPP) or poly(4,4'-diphenylene-vinylene) (PDV) in the helical cavity of amylose have been synthesised, structurally characterised by nuclear Overhauser spectroscopy and used to fabricate electroluminescent light-emitting diodes.

The electrophilic fluorodesilylation of enantioenriched allenylsilanes proceeds with efficient transfer of chirality. The silylation-fluorination of propargylic alcohols occurs with overall retention of stereochemistry, a result consistent with a stereospecific anti S(E)2' mechanism for the fluorination step.

Cyclodextrin [2] and [3]rotaxanes have been synthesised by use of a metal-driven ligand formation methodology. The kinetically stable cobalt(III) complexes formed act as either linking or capping groups, the function being selected by manipulation of the reaction conditions to promote the formation of either 2:1 or 1:1 complexes, respectively. In all cases, mixtures of isomers containing different alpha-cyclodextrin orientations were observed. All three orientational isomers of the [3]rotaxane were isolated and structurally characterized by 2D NMR.

A 1 : 1 crystalline complex of lead(IV) tetraacetate and pyridine (LTA-py) has been prepared. The single-crystal X-ray structure, at 296 and 150 K, establishes the presence of a relatively short Pb-N bond (2.307 A) within an intriguing seven-coordinate lead inner sphere consisting of the pyridine ligand and two bidentate and two monodentate acetate ligands. The pyridine occupies a surprising amount of the available coordination space and has induced a dramatic change in coordination compared to the four chelating acetate ligands found in lead tetraacetate (LTA). Thermal measurements (TGA/DSC) indicate the de-coordination of pyridine and its loss from the solid between 360 and 380 K. (207)Pb CP/MAS NMR spectroscopy also demonstrates the existence of the Pb-N bond through observation of (1)J((207)Pb,(14)N)= 63 Hz and a (207)Pb-(14)N dipolar coupling constant, of 149 Hz. The solid-state (207)Pb NMR parameters are used to give insight into the coordination environment of Pb(iv) in LTA-py. In solution, ligand exchange is rapid on chemical shift and J-coupling time scales. A (207)Pb NMR study of the titration of an LTA solution by pyridine yields a stability constant for LTA-py of K = 1.5 M(-1) and predicts it to have a (207)Pb NMR chemical shift essentially identical to that observed by CP/MAS NMR in the solid state. This correlation between the solid state and solution indicates that the seven-coordinate LTA-py structure found in the crystalline state does persist in solution, and this could further explain why the addition of pyridine has such profound effects on lead(IV) carboxylate-mediated organic reactions. Simulations of exchange-broadened line shapes of (13)C CP/MAS NMR spectra in the temperature regime above 280 K indicate local motion of the pyridine rings in the form of 180 degrees jumps (activation energy 72.5 kJ mol(-1)); these are first such ring flips reported for a coordinated pyridine ligand.

Regulation of the hypoxic response in humans is regulated by the post-translational hydroxylation of hypoxia inducible transcription factor; a recombinant form of a human prolyl-4-hydroxylase (PHD2) was characterised and shown to have an unexpectedly high affinity for, and to copurify with endogenous levels of, its Fe(ii) cofactor and 2-oxoglutarate cosubstrate.

The syntheses of potassium 2-(2'-methoxynaphth-1'-yl)-3,4-dimethyl-5-phenylphospholide 4 and eta5-pentamethylcyclopentadienyl(eta5-2-(2-methoxynaphth-1-yl)-3,4-dimethyl-5-phenylphospholyl)-ruthenium(II)5 and -iron(II)6 are described. The barrier to rotation of the naphthyl group (79 kJ mol-1 and 72 kJ mol-1 in CD2Cl2 respectively) characterises 5 and 6 as potential tropos type ligands. Coordination of 5 to [PtCl2(PEt3)] gives two cis and two trans complexes [PtCl2(PEt3)5] wherein rotation about the phospholyl-naphthyl vector is slow.

A semi-selective 2D HMBC experiment is described which yields high-resolution in the indirect carbon-13 dimension by suppressing homonuclear proton coupling modulations and so provides an NMR technique suitable for the structure elucidation of organic compounds which exhibit particularly crowded carbon-13 spectra.

Short oligomeric chains of C-glycosyl beta-D-arabinofuranose configured tetrahydrofuran amino acids (where the C-2 and C-5 substituents of the tetrahydrofuran ring are cis to each other) exhibit a well-defined repeating turn secondary structure stabilised by (i, i - 2) inter-residue hydrogen bonds. This is in contrast to the epimeric alpha-D-arabinofuranose oligomer (where the C-2 and C-5 substituents of the tetrahydrofuran ring are trans to each other) in which there is no indication of any secondary structure in solution.

Evidence is presented for initial oxidation at the C-3 position of the flavonoid C-ring and for two bifurcating steps during catalysis by anthocyanidin synthase.
