Abstract

The previous findings that the group I and III mite allergens, and amylase present in mite faeces are hydrolytic enzymes has prompted a study to determine whether this material contains other enzymes which could be allergenic. Thus, spent growth medium devoid of whole Dermatophagoides pteronyssinus mites was shown to contain glucoamylase, lipase and lysozyme in addition to the cysteine protease, serine protease and amylase activities associated with the above allergens, respectively. All of these enzymes are probably associated with mite digestive processes. They were rapidly solubilised, heterogeneous with regard to charge (pI in the range 4–8) and demonstrated maximum biochemical activity in the neutral pH range. Three serine proteases were detected and comprised a chymotrypsin-like, a trypsin-like and an unclassified enzyme with pI of 4.1 and 5.3, 8.5 and 7.1, respectively. Only one cysteine protease was observed, which paralleled immunochemically identified Der p I in a variety of assays. It was shown to cleave at lysyl residues and could be inhibited by the specific cysteine protease inhibitor, E-64. The remaining serine proteases, glucoamylase, lipase and lysozyme represent potential allergens.

Abstract

A lambda gt11 library made with cDNA from the house dust mite Dermatophagoides pteronyssinus was screened with human allergic serum by IgE plaque radioimmunoassay. This resulted in the isolation of clones coding for the major allergen Der p II. The cDNA coded for a 129-residue protein of 14,131 daltons with no N-glycosylation sites. No sequence homology with other proteins was evident. The Der p II expressed in Escherichia coli reacted with IgE in 14 of 17 sera from mite-allergic patients giving clonal evidence for its designation as a major allergen. This, along with previous work, has resulted in the cloning of the two major mite allergens.

Abstract

The allergen Dpt 12, from the house dust mite Dermatophagoides pteronyssinus, was isolated from spent growth medium by chromatofocusing. The allergen was isolated in the pI range 5.86–6.95, indicating heterogeneity with regard to pI. However, on sodium dodecylsulphate – polyacrylamide gel electrophoresis the allergen appeared homogeneous with an apparent molecular weight of 27,000 daltons. Radioallergosorbent scores utilizing paper discs coupled with Dpt 12 correlated significantly (p < 0.0005) with those obtained using discs coupled with whole mite extract, although the corresponding titres were higher with Dpt 12 (p < 0.0005).

Abstract

An extract of Ascaris suum perienteric fluid was fractionated on Sephadex G-200 and the resulting high molecular weight (approx. 600,000 daltons) and low molecular weight (approx. 16,000 daltons) fractions examined for allergenicity using human sera. Radioallergosorbent (RAST) and RAST-inhibition assays showed that the high molecular weight fraction was 15- to 40-fold more potent than the low molecular weight fraction. Crossed radio-immunoelectrophoretic analysis showed that the high molecular weight fraction contained 10 allergens in contrast to the 6 comprising the low molecular weight fraction. Polyacrylamide gel electrophoretic studies in combination with RAST indicated that two high molecular weight glycoprotein-staining components may be major allergens. A. suum cross-reacted allergenically with Toxocara canis and various life cycle stages of Nippostrongylus brasilliensis but not with Strongyloides ratti or Schistosoma mattheei. The cross-reacting allergens were predominantly of high molecular weight and the molecular weight distribution pattern obtained for A. suum, T. canis and N. brasilliensis paralleled that obtained for the distribution of the carrier of the hapten phosphorylcholine (Pc). However, it was shown that the hapten per se was not allergenic. In contrast, analysis of the allergens from N. brasilliensis using sera from rats infected with the same parasite showed that the homologous allergens were predominantly of low molecular weight (approx. 20,000 daltons) and did not contain Pc.

Abstract

The prevalence of asthma in a population of the South Fore linguistic group of the Eastern Highlands Province of Papua New Guinea has shown a remarkable increase in the last decade and is now 7.3% in adults and 0.6% in children. Allergy to house dust mite appears to play a major role in the disease pathogenesis. The low childhood prevalence may be associated with endemic parasitism which operates by suppressing production of IgE antibodies to ubiquitous allergens. This suppression is significantly correlated with age in boys of 6–20 years, but not for girls who retain low serum titres throughout the entire age range. Bronchial reactivity, assessed by histamine inhalation test, does not identify asymptomatic children who may be potential asthmatic subjects suggesting that, in this population, bronchial hyperreactivity follows the development of immunological hypersensitivity.

Abstract

Six candidate extracts of Lolium perenne (rye-grass) pollen have been studied in 6 laboratories using a variety of immunochemical and physicochemical techniques. Radioallergosorbent test inhibition, crossed immunoelectrophoresis, crossed radio-immunoelectrophoresis, sodium dodecyl sulphate-polyacrylamide gel electrophoresis combined with immunoblot, thin-layer isoelectric focusing and enzyme-linked immunosorbent assay inhibition were used to evaluate each of the coded extracts. The source materials were also studied for identity and possible contamination by light microscopy. On the basis of these data, the Rye-Grass Working Party recommended to the Steering Committee of the Allergen Standardization Subcommittee of the International Union of Immunological Societies that the extract coded C be chosen as the candidate international reference preparation.

Abstract

A cDNA clone coding for the major house dust mite allergen Der p 1 was isolated from a lambda gt 11 library. Its sequence correlates with known amino acid sequences of Der p 1 and it produces a fusion protein which reacts with rabbit anti-Der p 1 antiserum.
