Although an autonomous underwater vehicle (AUV) is noted for its good autonomy, concealment and anti-interference ability, errors in its inertial navigation system (INS) inevitably increase over time, leading to positional failure during long-term voyages. Terrain-assisted navigation can help the INS to correct its position. The traditional iterative closest contour point (ICCP) achieves high matching accuracy when the initial position error of the INS is small, but is prone to mismatching when the initial error is large. This study combines ICCP with particle swarm optimization (PSO) to overcome this problem. First, the global optimization ability of PSO is improved by changing the acceleration factor and introducing an artificial bee colony (ABC) onlooker bee greedy search (ABC- ω APSO). Second, the Euclidean distance of ICCP is replaced by the Mahalanobis distance to abate the influence of system error on the matching accuracy. Finally, the initial position error is reduced by rough matching using the ABC- ω APSO, which has global optimization capability. Next, fine matching is performed by ICCP. This two-step process resolves the sensitivity problem of ICCP to the initial position error. The experimental results revealed a good matching effect after the double-matching procedure. When the initial INS errors were 0.55′ to the east and 0.55′ to the north, the matching error was reduced to 89.3 m, suggesting that the approach can realize autonomous passive navigation of AUVs. There are problems in the special pastoral environment, including large changes in target size and serious interference from light and environmental factors. To solve the above problems, an enhanced YOLOv4-tiny target detection network is proposed in this study. This network first solves the problem of livestock size fluctuation in pastoral areas, uses a pyramid network with multiscale feature fusion, and considers shallow local detail features and deep semantic information. Subsequently, a novel compound multichannel attention mechanism is proposed to increase the accuracy of the target detection network for the pastoral environment. The problem of poor accuracy of target detection network is solved. The algorithm is ported to Jetson AGX embedded platform for validation to examine the real-time performance of the algorithm. As revealed by the experimental results, enhanced YOLOv4-tiny achieves 89.77% detection accuracy and 30 frames/second detection speed, which increases the average detection accuracy by 11.67% compared with the conventional YOLOv4-tiny while maintaining almost the same detection rate. Senescence-associated alterations in microglia may have profound impact on cerebral homeostasis and stroke outcomes. However, the lack of a transcriptome-wide comparison between young and aged microglia in the context of ischemia limits our understanding of aging-related mechanisms. Herein, we performed RNA sequencing analysis of microglia purified from cerebral hemispheres of young adult (10-week-old) and aged (18-month-old) mice five days after distal middle cerebral artery occlusion or after sham operation. Considerable transcriptional differences were observed between young and aged microglia in healthy brains, indicating heightened chronic inflammation in aged microglia. Following stroke, the overall transcriptional activation was more robust (>13-fold in the number of genes upregulated) in young microglia than in aged microglia. Gene clusters with functional implications in immune inflammatory responses, immune cell chemotaxis, tissue remodeling, and cell-cell interactions were markedly activated in microglia of young but not aged stroke mice. Consistent with the genomic profiling predictions, post-stroke cerebral infiltration of peripheral immune cells was markedly decreased in aged mice compared to young mice. Moreover, post-ischemic aged microglia demonstrated reduced interaction with neighboring neurons and diminished polarity toward the infarct lesion. These alterations in microglial gene response and behavior may contribute to aging-driven vulnerability and poorer recovery after ischemic stroke. [ABSTRACT FROM AUTHOR]

A series of novel 1,4-bis(alkynyl)benzene derivatives were synthesized from trimethylsilyl-substituted alkynes by the mediation of zirconocene with excellent regioselectivity in high yields. The 3,6-bis(trimethylsilyl)-4,5-dialkylphthalic acid dimethyl esters were prepared by cycloaddition of 2,5-bis(trimethylsilyl)zirconacyclopentadienes to dimethyl acetylenedicarboxylate. After iodination with iodine monochloride, 3,6-diiodo-4,5-dialkylphthalic acid dimethyl esters reacted with terminal alkynes to prepare the corresponding 1,4-bis(alkynyl)benzene derivatives by Sonogashira coupling reactions. After removal of trimethylsilyl, 4,5-dibutyl-3,6-bis(ethynyl)phthalic acid dimethyl ester (compound 3) reacted with 4-iodobenzoic acid ethyl ester and 2-iodothiophene, respectively, to obtain the corresponding products 4a and 4c. Compound 3 can be extended to higher oligomers, which reacted with 1-bromo-4-iodobenzene and phenylacetylene in a stepwise manner under Sonogashira conditions to give the phenylene-ethynylene oligomer 5 in an isolated yield of 85%. The structures of the products were confirmed by 1H NMR spectroscopy, 13C NMR spectroscopy, and MS. The optical properties of the 1,4-bis(alkynyl)benzene derivatives were studied by UV-Vis spectroscopy and fluorescence spectra. Ischemic injury can be alleviated by the judicious use of hypothermia. However, the optimal regimens and the temporal kinetics of post-stroke neurovascular responses to hypothermic intervention have not been systematically studied. These gaps slow the clinical translation of hypothermia as an anti-stroke therapy. Here, we characterized the effects of transient selective brain hypothermia (TSBH) from the hyperacute to chronic stages of focal ischemia/reperfusion brain injury induced by transient middle cerebral artery occlusion in mice. A simple cooling device was used to induce TSBH during cerebral ischemia. This treatment reduced mortality from 31.8% to 0% and improved neurological outcomes for at least 35 days post-injury. TSBH mitigated blood–brain barrier leakage during the hyperacute and acute injury stages (1–23 h post-reperfusion). This early protection of the blood–brain barrier was associated with anti-inflammatory phenotypic polarization of microglia/macrophages, reduced production of pro-inflammatory cytokines, and less brain infiltration of neutrophils and macrophages during the subacute injury stage (three days post-reperfusion). TSBH elicited enduring protective effects on both grey and white matter for at least 35 days post-injury and preserved the long-term electrophysiological function of fiber tracts. In conclusion, TSBH ameliorates ischemia/reperfusion injury in the neurovascular unit from hyperacute to chronic injury stages after experimental stroke. [ABSTRACT FROM AUTHOR]

This is a prospective non-randomized cohort study of 113 consecutive patients to investigate the safety and efficacy of a short-duration intraarterial selective cooling infusion (IA-SCI) targeted into an ischemic territory combined with mechanical thrombectomy (MT) in patients with large vessel occlusion-induced acute ischemic stroke (AIS); 45/113 patients underwent IA-SCI with 350 ml 0.9% saline at 4℃ for 15 min at the discretion of the interventionalist. Key parameters such as vital signs and key laboratory values, symptomatic and any intracranial hemorrhage, coagulation abnormalities, pneumonia, urinary tract infections and mortality were not significantly different between the two groups. Final infarct volume (FIV) was assessed on noncontrast CT performed at three to seven days. After an adjusted regression analysis, the between-group difference in FIV (19.1 ml; 95% confidence interval (CI) 3.2 to 25.2; P = 0.038) significantly favored the IA-SCI group. At 90 days, no differences were found in the proportion of patients who achieved functional independence (mRS 0–2) (51.1% versus. 41.2%, adjusted odd ratio (aOR) 1.9, 95% CI 0.8–2.6, P = 0.192).
