The oral gingival barrier is a constantly stimulated and dynamic environment where homeostasis is often disrupted, resulting in inflammatory periodontal diseases. Type 2 diabetes (T2D) has been reported to be associated with gingival barrier dysfunction, but the effect and underlying mechanism are inconclusive. Herein, we performed single-cell RNA sequencing (scRNA-seq) of gingiva from leptin receptor-deficient mice (db/db) to examine the gingival heterogeneity in the context of T2D. Periodontal health of control mice is characterized by populations of Krt14+-expressing epithelial cells and Col1a1+-fibroblasts mediating immune homeostasis primarily through the enrichment of innate lymphoid cells. The db/db gingiva exhibited decreased epithelial/stromal ratio and dysfunctional barrier. We further observed stromal, particularly fibroblast immune hyperresponsiveness, linked to the recruitment of myeloid-derived cells at the db/db gingiva. Both scRNA-seq and histological analysis suggested the inflammatory signaling between fibroblasts and neutrophils as a potential driver of diabetes-induced periodontal damage. Notably, the "immune-like" stromal cells were wired toward the induction of gingival IL-17A hyperresponsiveness in db/db mice. Our work reveals that the "immune-like" fibroblasts with transcriptional diversity are involved in the innate immune homeostasis at the diabetic gingiva. Objective: C1q/TNF-related protein 3 (CTRP3), a member of CTRP family, has been found to have neuroprotective effect. In the current study, we investigated the protective role of CTRP3 in hippocampal neurons exposed to oxygen-glucose deprivation/reperfusion (OGD/R). Materials and methods: The mRNA and protein levels of CTRP3 in OGD/R-stimulated hippocampal neurons were measured using qRT-PCR and western blot analysis, respectively. CCK-8 assay was performed to assess cell viability. ROS production was measured using the fluorescence probe 2′,7′-dichlorofluorescein diacetate (H2DCFDA). The activities of SOD and GPx were determined using ELISA. Cell apoptosis was assessed. Luciferase reporter assay was carried out to assess the activation of ARE). The levels of p-AMPK and Nrf2 were measured using western blot. Results: Our results showed that the expression of CTRP3 was significantly downregulated in hippocampal neuronal cells exposed to OGD/R. Overexpression of CTRP3 improved cell viability of OGD/R-induced hippocampal neurons. In addition, overexpression of CTRP3 attenuated the OGD/R-caused oxidative stress with decreased ROS production and increased activities of SOD and GPx. Moreover, CTRP3 caused a significant increase in bcl-2 expression and decreases in bax expression and caspase-3 activity. Furthermore, CTRP3 overexpression significantly upregulated the levels of p-AMPK and Nrf2, as well induced the activation of ARE in OGD-R-induced hippocampal neurons. CTRP3 upregulated the mRNA expression levels of HO-1, NQO-1 and GPx-3. Additionally, treatment with the inhibitor of AMPK partially reversed the neuroprotective effect of CTRP3 in OGD/R-exposed neurons. Background Chronic thromboembolic pulmonary hypertension (CTEPH) is a rare and life-threatening condition with poor prognosis in patients with antiphospholipid syndrome (APS). Pulmonary thromboendarterectomy (PTE) is the optimal surgical option for CTEPH. Objectives This retrospective cohort study aimed to evaluate the efficacy and risk of PTE in patients with APS-associated CTEPH. Methods Consecutive patients with APS-associated CTEPH diagnosed between January 2012 and September 2017 at Peking Union Medical College Hospital were retrospectively evaluated. Demographics, clinical manifestations, antiphospholipid antibody (aPL) profiles, and pulmonary arterial hypertension–targeted medications were collected. Deterioration of cardiac function and death were chosen as the endpoints, in order to assess the effect of PTE on short-term and long-term prognoses (evaluated by the change of cardiac function after treatment and cardiac deterioration or death in the follow-up, respectively). Results A total of 20 patients with APS-associated CTEPH were enrolled, and eight patients underwent PTE. Chi-square test (p = 0.01) and Kaplan–Meier curves (log rank test, p = 0.04) showed that there were statistically significant differences in both short-term and long-term prognoses between patients with and without PTE. Conclusion These results provide strong evidence that PTE is a curative resolution in patients with APS-associated CTEPH. Following a full specialized and multidisciplinary risk-benefit evaluation to limit the risk of thrombosis or bleeding and to manage possible thrombocytopenia, PTE is at least a temporal curative resolution for CTEPH complicated with APS. The aim of this study was to provide a clear description of the course, precise branching pattern and distribution of the deep branch of the ulnar nerve. A total of 36 hands from 18 preserved cadavers were dissected. The vertical distance from the pisoscaphoid line to the crossing points between the deep branch of the ulnar nerve and each metacarpal was about 4 cm. The deep branch of the ulnar nerve gave off two types of muscular branches: (1) trunks that innervate more than two intrinsic hand muscles; and (2) multiple separate branches innervating only a single muscle. A novel process was developed for scalable fabrication of micro-fine spherical Ti–6Al–4V alloy powders. The hydrogenation-treated Ti–6Al–4V alloy ingot was mechanically crushed into particles and then sieved into three size grades. The powders were separately sent through the radio frequency (RF) argon plasma system for spheroidisation. The fabrication process and powder characteristics were investigated. The results indicate the alloy ingot upon hydrogenation treatment can be efficiently crushed into fine particles with size of 5–76 μm. During RF plasma processing, the powders are found to be greatly refined due to hydrogen decrepitation with subsequent transformation into spherical morphology. The effect of hydrogen decrepitation on particle refinement is impaired with decreasing particle size of feed powders. The spherical powders exhibit a narrow particle size distribution and the average size is in the range of 8.2–27.9 μm. The spheroidised powders mainly consist of β-Ti and TiH1.5. Background: Chemokine receptor 4(CXCR4) plays an important role in the potential growth of pancreatic tumor and its ability to develop metastasis. Ultrasmall superparamagnetic iron oxide (USPIO) nanoparticles offer strong contrast-enhancing effect for MR imaging of pancreatic tissue.Purpose: To establish a biomarker-targeted nanoparticulate contrast agent CXCR4-USPIO for pancreatic cancer cell MR imaging and to investigate the relationship between in vitro MR T2 enhancement ratio, ΔR2 values, and the cellular CXCR4 expression levels.Material and Methods: The CXCR4 monoclonal antibody and bovine serum albumin (BSA) were bioconjugated with USPIO using carbodiimide. The T2 and T2* values of CXCR4-USPIO, BSA-USPIO, and USPIO were evaluated at the same iron concentration levels. After incubating four pancreatic cancer cell lines (AsPC-1, BxPC-3, CFPAC-1, PANC-1) with CXCR4-USPIO and BSA-USPIO, respectively, changes of T2 values were measured with a clinical 1.5-T MRI scanner. Western blot and immunofluorescence tests were applied to semi-quantitatively analyze the expression levels of CXCR4 in the four cell lines.Results: MR imaging revealed a linear correlation between ΔR2 and ΔR2* values of CXCR4-USPIO nanoparticles and the iron concentrations. The T2 enhancement ratio and ΔR2 values of AsPC-1, BxPC-3, CFPAC-1, PANC-1 cell lines in the CXCR4-USPIO group exhibited strong correlation with the CXCR4 peptide relative grey values measured by western blot (r = 0.976, P = 0.024; r = 0.959, P = 0.041, respectively) and the mean fluorescence signal intensity detected by laser scanning confocal microscopy (r = 0.996, P = 0.004; r = 0.962, P = 0.038, respectively).Conclusion: The targeted probe CXCR4-USPIO was created for MR molecular imaging of pancreatic cancer cell lines. The hot cracking behaviour in the diode laser braze joint of 13Cr–4Ni stainless steel using Au–18Ni, Ag–10Pd and Ag–21Cu–25Pd filler metals has been investigated. The types of joint investigated were the T fillet joint and the L fillet joint which simulated the second braze bead in the T fillet joint. Tandem beam brazing was also carried out in order to prevent the hot cracking by post-heating treatment with a trailing beam. A centreline crack, characterised as a ductility dip crack, occurred in the second braze bead of the T fillet joint using Au–Ni filler metal. On the other hand, no cracks occurred in either the first or the second braze bead in T fillet braze joints using Ag–Pd and Ag–Cu–Pd filler metals. The hot cracking susceptibility of the Au–Ni braze metal was evaluated by the spot Varestraint test. Most of the cracks observed in the spot Varestraint test specimen were also characterised as ductility dip cracks, and the susceptibility to such cracking increased with increasing the augmented strain. The ductility dip temperature range (DTR) was estimated from the crack position and length to be 1000–1250 K at strains over 0·4%. Numerical analyses of the thermal stress and strain revealed that the plastic strain–temperature curve intersected the DTR in the single beam brazing, but did not intersect the DTR with post-heating during the laser brazing. The effect of post-heating on the crack prevention was verified by tandem beam brazing of L fillet joint with a trailing beam. No cracks occurred in the braze bead made with a trailing beam at laser powers of 200–300 W. MoS2 and MoS2/Zr coatings were deposited on the surface of YT15 carbide by medium frequency magnetron sputtering together with multiarc ion plating. Microstructural and fundamental properties of these coatings were examined. Sliding wear tests against hardened steel using the ring block method were carried out with these coated materials. The friction coefficient and wear rates were measured. The wear surface features of the coatings were examined by SEM. Results showed that the MS-1 specimen (with MoS2/Zr composite coating) has the higher hardness and adhesion load for the coating to the substrate compared with that of the MS-2 specimen (only with MoS2 coating). The friction coefficient of MS-1 specimen was depended on the applied load, and was quite insensitive to the sliding speed. The MS-1 specimen exhibits improved friction behaviour to that of the MS-2 specimens, and the antiwear lifetime of the MoS2 coatings can be prolonged through adding Zr additives. The diode laser brazing of Ni base heat resistant alloy with precious filler metals has been conducted using the tandem beam for preheating and brazing. A couple of 1 mm thick plates of alloy 600 (Inconel 600) were butt brazed using Au–18Ni, Ag–10Pd and Ag–21Cu–25Pd filler metals of 0·5 mm diameter with a brazing flux. Sound butt joints which were free from brazing defects such as porosity and lack of penetration could be obtained at brazing clearances of 0·1–1·5 mm. The tensile strength of the braze joint produced using Ag–Pd filler metal increased with decreasing brazing clearance and reached ∼70% of the base metal strength at a brazing clearance of 0·1 mm while those obtained by using Au–Ni and Ag–Cu–Pd filler metals were comparable with the base metal strength at any clearances between 0·1 and 1·5 mm. Purpose: To examine the imaging characteristics of osteoblastic osteosarcoma in older patients, we compared them with those in adolescents because the radiological features of osteosarcoma can be atypical in elderly patients. Material and Methods: 29 cases of adolescent patients and 12 patients older than 40 years of age were reviewed. All cases were pathologically confirmed as osteoblastic osteosarcoma. The comparative factors were tumor location, plain radiological features including periosteal reactions, and the degree of soft tissue mass with magnetic resonance (MR) imaging. Results: Older patients demonstrated frequent osteolytic findings (83.3%), and none of the tumors showed a significant major periosteal reaction. Seven (58.3%) of the 12 tumors had no soft tissue mass (grade 0) and only three (25%) had a grade 3 mass. The differences in the two groups were statistically significant (P<0.001). Conclusion: In older patients, primary osteosarcoma should be considered when making a differential diagnosis of osteolytic lesions without any periosteal reactions and small soft-tissue extensions. In cases with atypical findings, the morphology of specimens (aspiration, core, or surgical biopsy) from the lesion should be recommended when making an accurate diagnosis. [ABSTRACT FROM AUTHOR]

Tumors of the oral cavity are highly vascularized malignancies. Disruption of neovascular networks was shown to limit the access of nutrients and oxygen to tumor cells and inhibit tumor progression. Here, we evaluated the effect of the activation of an artificial death switch (iCaspase-9) expressed in neovascular endothelial cells on the progression of oral tumors. We used biodegradable scaffolds to co-implant human dermal microvascular endothelial cells stably expressing iCaspase-9 (HDMEC-iCasp9) with oral cancer cells expressing luciferase (OSCC3-luc or UM-SCC-17B-luc) in immunodeficient mice. Alternatively, untransduced HDMEC were co-implanted with oral cancer cells, and a transcriptionaly targeted adenovirus (Ad-VEGFR2-iCasp-9) was injected locally to deliver iCaspase-9 to neovascular endothelial cells. In vivo bioluminescence demonstrated that tumor progression was inhibited, and immunohisto-chemistry showed that microvessel density was decreased, when iCaspase-9 was activated in tumor-associated microvessels. The universality and sufficiency of the five-factor model in the Chinese context were investigated. In Study 1, analysis of the Revised NEO Personality Inventory (NEO-PI-R) and the Chinese Personality Assessment Inventory (CPAI) taken by Chinese students showed four joint factors similar to the domains of the NEO-PI-R. Two unique factors were obtained. The Interpersonal Relatedness factor was defined only by CPAI scales. The Openness domain, however, was not represented in the CPAI scales. In Study 2, involving Chinese managers, the robustness of the Interpersonal Relatedness factor was demonstrated. In Study 3, the six-factor model was confirmed with Hawaiian students. Further analyses showed that the six-factor models were superior to the five-factor models and that the Interpersonal Relatedness scales could not be consistently explained by a combination of the Big Five factors. Implications for the universality of the five-factor model and the cross-cultural relevance of the CPAI Interpersonal Relatedness factor are discussed. [ABSTRACT FROM AUTHOR]
