- Abstract
Objective To investigate the neuroprotective effect of electro-acupuncture on cerebral isehemia-reperfu-sion injury rats and its mechanism. Methods Ninety adult male SD rats were randomly divided into the sham operation group, model group and electro-acupuncture group, with 30 rats in each. The focal cerebral ischemia-reperfusion injury rat models in the model group and electro-acupuncture group were established by modified thread embolism method; the rats in the shamoperation group were only given blood vessel isolation without blocking blood vessel; the rats in the electro-acupuncture group were given electro-acupuncture at Baihui and Dazhui acupoint immediately after modeling, once every 12 h, 30 min each time, for 5 times. At 6 , 24 and 48 h after reperfusion, Longa method was used to score the neurological deficit. At 48 h after reperfusion, laser Doppler flow meter was used to detect the regional cerebral blood flow and velocity in the ischemic side. Ten rats in each group were randomly selected and decapitated to death, and their brain tissues were taken out quickly, evenly sliced. TTC staining was used to calculate the percentage of cerebral infarction volume; ten rats in each group were randomly selected and decapitated to death, and the cerebral cortex of ischemic side was taken to detect the apoptotic rate of neurons by TUNEL staining. The remaining rats were decapitated and their brain tissues were taken. The expression of Wnt/β-catenin signaling pathway key factor β-catenin protein, apoptosis-related factors Bel-2 and Bax protein in brain tissues were detected by Western blotting. Results Compared with the sham operation group, the cerebral blood flow and velocity decreased significantly, the scores of neurological deficit, the percentage of cerebral infarction area and the apoptotic rate of neurons increased, the expression of Bax protein in the brain tissue increased, and the expression of β-catenin and Bcl-2 protein decreased significantly in the model group ( all P <0.0 5). Compared with the model group, the cerebral blood flow and blood flow velocity increased, the scores of neurological deficit, the percentage of cerebral infarction area, and the apoptotic rate of neurons decreased, the expression of Bax protein in the brain tissues decreased, but the expression of β-catenin and Bcl-2 protein increased significantly in the electro-acupuncture group (all P <0. 05). Conclusion Electro-acupuncture has neuroprotective effect on cerebral ischemia-reperfusion injury rats by activating Wnt/β-catenin signaling pathway, inhibiting Bax protein expression, and promoting Bcl-2 protein expression.
