Introduction: Sjögren's syndrome (SS) presents complex diagnostic challenges due to its multi-organ involvement, often leading to misdiagnosis, which can result in unnecessary treatments, elevated healthcare costs, and significant impacts on patient quality of life. Accurate diagnosis is therefore critical, utilising ACR/EULAR criteria that include both labial minor salivary gland (LMSG) biopsy and anti-SSA antibodies., Methods: This retrospective study analysed medical records of 87 adults suspected of primary SS, who underwent both anti-SSA serology and LMSG biopsy. We evaluated the diagnostic accuracy of these tests under existing ACR/EULAR criteria and a newly proposed 'modified ACR/EULAR criteria - ClinDx'. Statistical analysis included Pearson's chi-square test for the association between test results and disease status and receiver operating characteristic (ROC) curves to assess the sensitivity and specificity of the diagnostic models., Results: Utilising ACR/EULAR criteria, 40 patients were diagnosed with SS, while 47 were categorised as non-diseased. The ClinDx criteria application resulted in 32 SS diagnoses and 55 non-diseased classifications, highlighting discrepancies in patients with low anti-SSA titers (< 200 MFU). Statistical analysis confirmed a significant association (p < 0.001) between test results and disease status, indicating the robustness of the modified criteria in enhancing diagnostic accuracy., Conclusions: This study underscores the utility of integrating serological tests and histological biopsies in diagnosing SS. While anti-SSA antibodies provide a good preliminary screening tool, the specificity of LMSG biopsies is indispensable. In recent years, heterogeneous photocatalysis has emerged as a promising alternative for the treatment of organic pollutants. This technique offers several advantages, such as low cost and ease of operation. However, finding a semiconductor material that is both operationally viable and highly active under solar irradiation remains a challenge, often requiring materials of nanometric size. Furthermore, in many processes, photocatalysts are suspended in the solution, requiring additional steps to remove them. This can render the technique economically unviable, especially for nanosized catalysts. This work demonstrated the feasibility of using a structured photocatalyst (ZnO, g-C 3 N 4 , and carbon xerogel) optimized for this photodegradation process. The synthesized materials were characterized by nitrogen adsorption and desorption, X-ray diffraction (XRD), and diffuse reflectance spectroscopy (DRS). Adhesion testing demonstrated the efficiency of the deposition technique, with film adhesion exceeding 90%. The photocatalytic evaluation was performed using a mixture of three textile dyes in a recycle photoreactor, varying pH (4.7 and 10), recycle flow rate (2, 4, and 6 L h -1 ), immobilized mass (1, 2, and 3 mg cm -2 ), monolith height (1.5, 3.0, and 4.5 cm), and type of radiation (solar and visible artificials; and natural solar). The structured photocatalyst degraded over 99% of the dye mixture under artificial radiation. The solar energy results are highly promising, achieving a degradation efficiency of approximately 74%. Furthermore, it was possible to regenerate the structured photocatalyst up to seven consecutive times using exclusively natural solar light and maintain a degradation rate of around 70%. Background: There is much debate over the occurrence of biliary reflux to the gastric pouch after one anastomosis gastric bypass (OAGB) and its potential risks., Objective: To assess endoscopic and histopathological findings following a standardized protocol of biopsy collection two years after OAGB., Methods: A historical cohort study was conducted, based on a prospectively collected database, which involved 39 participants who underwent OAGB. Participants underwent clinical evaluation and esophagogastroduodenoscopy at the time of surgery and 24 months afterward. Post-operatively, biopsy specimens in esophagogastric junction, pouch, and anastomosis were systematically collected., Results: 92.3% of the participants were female and the mean age was 37 ± 8.5 years. The mean body mass index (BMI) significantly decreased from 37.6 ± 5.7 kg/m 2 to 27 ± 4.1 kg/m 2 after 2 years (p < 0.001). The mean %TWL was 27.2 ± 10.5%. The prevalence of non-erosive gastritis significantly increased from 25.6 to 51.3% (p = 0.02). Erosive gastritis significantly decreased from 28.2 to 10.3% (p = 0.04). Four cases of marginal ulcers were identified (10.3%). The commonest histopathological finding was mild inflammation in 74.3% (esophagogastric junction), 58.9% (pouch), and 71.8% (anastomosis). There was one case of focal intestinal metaplasia in each site of interest and no cases of dysplasia or severe inflammation., Conclusions: Using a standardized protocol of post-operative biopsy collection, low rates of severe endoscopic and histopathological abnormalities were observed two years after OAGB. Nevertheless, as most patients have histologically proven inflammation, bile in the gastric pouch, and endoscopic gastritis, long-term surveillance is essential because of the uncertain risk of these abnormalities., (© 2023. The objective of this study was to estimate the genetic parameters for feed efficiency-related traits and their genetic correlations with growth, male fertility, and carcass traits using multi-trait analysis in Guzerat cattle. Further, it aimed to predict the direct and correlated responses for feed efficiency traits when selection was applied for growth, male fertility, and carcass traits. The evaluated traits were adjusted weight at 120 (W120), 210 (W210), 365 (W365), and 450 days of age (W450), adjusted scrotal circumference at 365 days of age (SC365) and at 450 days of age (SC450), scrotal circumference, ribeye area (REA), backfat thickness (BFT), rump fat thickness (RFT), residual feed intake (RFI), and dry matter intake (DMI). The genetic parameters were obtained by the restricted maximum likelihood method (REML), using an animal model in multi-trait analyses. The heritability estimates for W120, W210, W365, W450, SC365, and SC450 varied from low to high (0.17 to 0.39). The carcass traits, REA, BFT, and RFT, displayed low to moderate heritability estimates, 0.27, 0.10, and 0.31, respectively. The heritability estimates for RFI (0.15) and DMI (0.23) were low and moderate, respectively. The RFI showed low genetic correlations with growth traits, ranging from - 0.07 to 0.22, from 0.03 to 0.05 for scrotal circumference, and from - 0.35 to 0.16 for carcass, except for DMI, which ranged from 0.42 to 0.46. The RFI and DMI presented enough additive genetic variability to be used as selection criteria in Guzerat breed genetic improvement program. Additionally, the response to selection for RFI would be higher when selection is performed directly for this trait. The selection for residual feed intake would not promote unfavorable correlated responses for scrotal circumference, carcass (yield and finish), and growth traits. Therefore, the selection for more efficient animals would not compromise the productive, reproductive, and carcass performance, contributing to reduce the production costs, increasing the profitability and sustainability of beef cattle production in tropical areas., (© 2023. This study aimed to estimate prediction ability and genetic parameters for residual feed intake (RFI) calculated using a regression equation for each test (RFI test ) and for the whole population (RFI pop ) in Nellore beef cattle. It also aimed to evaluate the correlations between RFI pop and RFI test with growth, reproductive, and carcass traits. Genotypic and phenotypic records from 8354 animals were used. An analysis of variance (ANOVA) was performed to verify the adequacy of the regression equations applied to estimate the RFI test and RFI pop . The (co)variance components were obtained using the single-step genomic best linear unbiased prediction under single and two-trait animal model analyses. The genetic and phenotypic correlations between RFI test and RFI pop with dry matter intake, frame, growth, reproduction, and carcass-related traits were evaluated. The prediction ability and bias were estimated to compare the RFI test and RFI pop genomic breeding values (GEBV). The RFI pop ANOVA showed a higher significance level (p < 0.0001) than did the RFI test for the fixed effects. The RFI pop displayed higher additive genetic variance estimated than the RFI test , although the RFI pop and RFI test displayed similar heritabilities. Overall, the RFI test showed higher residual correlations with growth, reproductive, and carcass traits, while the RFI pop displayed higher genetic correlations with such traits. The GEBV for the RFI test was slightly biased than GEBV RFI pop . The approach to calculate the RFI influenced the decomposition and estimation of variance components and genomic prediction for RFI. The application of RFI pop would be more appropriate for genetic evaluation purpose to adjust or correct for non-genetic effects and to decrease the prediction bias for RFI., (© 2022. The objective of this study was to evaluate the performance, feed intake, carcass traits, and economic aspects of cattle fed different levels of supplementation during the growing phase (mineral supplementation-MS; low protein supplementation-PS, at 0.15% of body weight (BW); and a high protein-energy supplementation-PES at 0.40% of BW) and finished on the pasture with concentrate supplementation at 1.6% of BW. Ninety bulls were distributed in a completely randomized design and allocated into paddocks predominantly composed of Urochloa brizantha cv. Marandu grass. During the finishing phase, cattle were managed in paddocks of Urochloa decumbens grass. The average daily gain (ADG) and total weight gain during the growing phase were not different between animals fed PS (0.670 kg/day; 57.1 kg, respectively) and PES (0.730 kg/day; 62.4 kg, respectively), but both differed significantly from those supplemented with MS (0.540 kg/day; 45.1 kg). There was no difference between groups for average daily gain and total weight gain during the finishing phase (0.600 kg/day; 48.3 kg) and the same response was observed for carcass yield (55.18%, on average). However, bulls fed PES had higher hot carcass weight compared with those supplemented with MS and PS. Protein supplementation (0.15% of BW) and protein-energy supplementation (0.40% of BW) improved beef cattle performance during the growing phase. However, supplementation during the growing phase has a small impact on cattle performance in the finishing phase, although protein-energy supplementation at the growing phase improves the hot carcass weight and carcass traits during the finishing phase., (© 2022. In Nellore beef cattle, studies addressing genetic correlations between ultrasound marbling content and other economically important traits are still incipient. Therefore, this work aimed to estimate heritability and genetic correlations between ultrasound marbling content in the longissimus dorsi muscle (MARB) and growth, reproductive, feed efficiency, and carcass-related traits in a Nellore beef cattle population from Brazil. Phenotypic records of 614,395 Nellore animals were used and included adjusted weight at 210 (W210) and 450 (W450) days of age, adult cow weight (AW), early heifer pregnancy (EH), stayability (STAY), adjusted scrotal circumference at 365 days of age (SC365), ribeye area (REA), subcutaneous backfat thickness (BF), rump fat thickness (RF), and marbling (MARB). The genetic parameters for all traits but EH and STAY were estimated considering a linear animal model, whereas for those two nonlinear traits, a threshold animal model was used. The direct and correlated response to selection for MARB versus the other traits, and the relative efficiency of selection, were also calculated. The heritability estimate for MARB was 0.31 and for the other conventional evaluated traits was low to moderate, with values ranging from 0.14 to 0.41. The genetic correlations between MARB and growth, reproductive, feed efficiency, and carcass-related trait were very low, with values close to zero, with similar correlated responses. The MARB displayed adequate genetic variability to respond to selection and crossbreeding programs looking forward to higher meat quality and differential market standards for the Nellore beef. The selection for growth, reproductive, feed efficiency, and carcass-related traits would not affect MARB in Nellore beef cattle and vice versa. The objective of this review was to assess and evaluate whether the published diagnostic accuracy studies provide evidence to sustain the current diagnostic guidelines put forth by ACR/EULAR used for patients with suspected Sjögren's syndrome (SS). Literature databases, including Medline, Embase, and EBM Reviews, were searched for relevant studies on the correlation between ACR/EULAR criteria, particularly those with a direct comparison between their accuracy in diagnosing Sjögren's syndrome. We followed Cochrane, QUADAS-2, and STARD guidelines and the four-phase flow diagram by the PRISMA Statement. Reports in several languages, but only human studies were considered. Three studies assessed the accuracy of the current diagnostic tests, and these did not present adequate designs that would allow a well-supported conclusion with a high level of certainty. Due to significant clinical and methodological heterogeneity, a meta-analysis was not performed. A qualitative review of the papers was undertaken. Neither the comparative nor the non-comparative study designs permit conclusive recommendations regarding an alternative diagnostic pathway for SS. Well-designed studies of the diagnostic accuracy of SS tests are needed to validate current guidelines or to suggest changes to the current guidelines., (© 2021. As more uses for biomarkers are sought after for an increasing number of disease targets, single-target biomarkers are slowly giving way for biomarker panels. These panels incorporate various sources of biomolecular and clinical data to guarantee a higher robustness and power of separation for a clinical test. Multifactorial diseases such as psychiatric disorders show great potential for clinical use, assisting medical professionals during the analysis of risk and predisposition, disease diagnosis and prognosis, and treatment applicability and efficacy. More specific tests are also being developed to assist in ruling out, distinguishing between, and confirming suspicions of multifactorial diseases, as well as to predict which therapy option may be the best option for a given patient's biochemical profile. As more complex datasets are entering the field, involving multi-omic approaches, systems biology has stepped in to facilitate the discovery and validation steps during biomarker panel generation. Key Message: The tomato mutant Never ripe (Nr), a loss-of-function for the ethylene receptor SlETR3, shows enhanced growth, associated with increased carbon assimilation and a rewiring of the central metabolism. Compelling evidence has demonstrated the importance of ethylene during tomato fruit development, yet its role on leaf central metabolism and plant growth remains elusive. Here, we performed a detailed characterization of Never ripe (Nr) tomato, a loss-of-function mutant for the ethylene receptor SlETR3, known for its fruits which never ripe. However, besides fruits, the Nr gene is also constitutively expressed in vegetative tissues. Nr mutant showed a growth enhancement during both the vegetative and reproductive stage, without an earlier onset of leaf senescence, with Nr plants exhibiting a higher number of leaves and an increased dry weight of leaves, stems, roots, and fruits. At metabolic level, Nr also plays a significant role with the mutant showing changes in carbon assimilation, carbohydrates turnover, and an exquisite reprogramming of a large number of metabolite levels. Notably, the expression of genes related to ethylene signaling and biosynthesis are not altered in Nr. We assess our results in the context of those previously published for tomato fruits and of current models of ethylene signal transduction, and conclude that ethylene insensitivity mediated by Nr impacts the whole central metabolism at vegetative stage, leading to increased growth rates., (© 2020. The objective of this study was to characterize and typify dairy farms and to evaluate society's perception of dairy production in rural settlements in the Brazilian semi-arid region. A questionnaire containing questions related to socioeconomic, livestock, physical, herd management, and production indicators was applied to 17 milk farmers for typological analysis. A second questionnaire was subsequently applied to the same 17 farmers for the holistic perception of dairy farming in rural settlements, in addition to 47 technicians in the area of animal science and 140 laypeople. Milk samples were collected in the settlement's community cooling tank, while the water used for milking management was collected in the milking parlor at the 17 farms. Multivariate analysis techniques and thematic analysis were performed. Three dairy production systems were identified: low production group (0-25 L), medium production group (25-50 L), and high production group (50-100 L). The physicochemical properties (composition, density, and cryoscopy) of milk attended the Brazilian regulation values. The microbiological parameters of milk and water used for milking are at odds with regulations. Regarding the typology, the indicators, livestock, and productivity showed discriminatory power (P < 0.05) between the productive systems. The responses from the segments of society showed different views on dairy production in rural settlements. It is concluded that the dairy farmers have low education and the cleaning product and water used in milking are out of regulation. The livestock and production indicators typified mainly dairy farms, while the holistic perception showed a lack of efficiency in rural extension, ignorance of the potential of the productive system. In the 2010-2014 period, the mean prevalence of congenital anomalies (CA) in the world was estimated at 398/10,000 births. CA are an important cause of mortality, disability, and comorbidity. Thus, the present study aims to describe the geographical and temporal distributions of live births and infant mortality (IM) due CA (IM-CA) in Brazil, from 2012 to 2017. The data used in this study is available at the Department of Informatics of the Unified Health System (DATASUS). The prevalence of CA at birth was 81.67/10,000 (95% CI 80.46-82.88), and the IM-CA rate was 27.97/10,000 (95% CI 27.95-28.00) in the studied period. The five CA with the highest rates were polydactyly (9.66/10,000, 95% CI 6.10-9.82), Down syndrome (3.40/10,000, 95% CI 3.41-5.99), microcephaly (2.92/10,000, 95% CI 2.91-3.12), hydrocephalus (2.72/10,000, 95% CI 2.65-2.90), and spina bifida (2.44/10,000, 95% CI 2.43-2.64). São Paulo was the Brazilian state with the highest CA birth rate (119.3/10,000), and Amazonas was the state with the highest IM-CA rate (33.8/10,000). The description and data analyses such as those performed in this work are relevant for healthcare systems and can be very useful in the formulation of public health campaigns and policies, as well as informing and educating professionals and the population. The management of clinical actions should consider all social, economic, geographic, and epidemiological factors.

Goats are critical in mixed smallholder agricultural systems in lower and middle-income countries, while fleas are important human and animal health concerns around the world. Convenience sampling was used to describe and consider risk factors for flea infestations of peri-urban goats, with the aim of informing the iterative development of animal husbandry and management based control strategies. Seven hundred and ninety-two goats were examined in 228 households across 10 peri-urban communities surrounding Blantyre in southern Malawi. The prevalence of Ctenocephalides felis fleas was 18.3, 37.1 and 100% at the levels of individual goats, households and communities, respectively, highlighting a neglected human and animal health concern. Constant introduction of new livestock coupled to a lack of biosecurity within communities, the ubiquitous presence of dog and cat hosts for C. felis, the frequency and thoroughness of cleaning overnight goat accommodation, and goat age less than 12 months old were identified as risk factors for flea infestation. This focal cross-sectional study highlights the significance of fleas in peri-urban communities and uncovers trends and commonalities that are needed to inform sustainable disease management. The majority of the peri-urban goat keepers were female, had resided in the same community throughout their whole life and had primary level education. Advice on the planned management of fleas in livestock needs to be tailored towards this demographic group. This approach affords an opportunity to promote public health measures to address household flea infestations and zoonotic disease spread.

Key Message: Linkage and genome-wide association analyses using high-throughput SNP genotyping revealed different loci controlling resistance to different isolates of race 65 of Colletotrichum lindemuthianum in common bean. Development of varieties with durable resistance to anthracnose is a major challenge in common bean breeding programs because of the extensive virulence diversity of Colletotrichum lindemuthianum fungus. We used linkage and genome-wide association analyses to tap the genomic regions associated with resistance to different isolates of race 65. Linkage mapping was done using an F 2 population derived from the cross between the Mesoamerican common beans BRS Estilo x Ouro Vermelho, inoculated with two different isolates of race 65. Association genetics relied on a diversity common bean panel containing 189 common bean accessions inoculated with five different isolates of race 65 as an attempt to validate the linkage analysis findings and, eventually, identify other genomic regions associated with resistance to race 65. The F 2 population and diversity panel were genotyped with the BARCBean6K&#95;3 Illumina BeadChip containing 5398 SNP markers. Both linkage and genome-wide association analyses identified different loci controlling resistance to different isolates of race 65 on linkage group Pv04. Genome-wide association analysis also detected loci on Pv05, Pv10 and Pv11 associated with resistance to race 65. These findings indicate that resistance to race 65 can be overcome by the virulence diversity among different isolates of the same race and could lead to the loss of resistance after cultivar release. We identified 25 resistant common bean cultivars to all five isolates of race 65 in the diversity panel. The accessions should be useful to develop cultivars combining different resistance genes that favor durable resistance to anthracnose in common bean.

This study aimed to estimate the genetic parameters of milk (305-day milk yield (MY305)), growth (weaning weight (WW), yearling weight (YW), and weight at 550 days (W550)), and reproductive (age at first calving (AFC)) traits in Guzerá cattle by using Bayesian multiple-trait models. Systematic effects included sex and age at calving for the growth and milk traits, respectively. The additive genetic and contemporary groups (herd and year and season of birth) were included as random effects. Additionally, maternal genetic and permanent effects were also included as random effects for the WW. The heritability estimates were 0.29 (MY305), 0.42 (WW), 0.49 (YW), 0.56 (W550), and 0.25 (AFC). The genetic correlations among the growth traits were higher than 0.83; between the MY305 and WW, MY305 and YW, and MY305 and W550, the genetic correlations were 0.25, 0.32, and 0.36, respectively. The AFC was negatively correlated with the milk and growth traits. These results suggest the viability and potential of the joint selection for milk, beef, and reproductive traits in Guzerá cattle.

The objective was to evaluate the intake, digestibility, milk yield and composition of cows fed mesquite pod meal (MPM). Five crossbred dairy cows, with an average body weight of 450 ± 5 kg and producing 10 ± 1 kg of milk/day were distributed in a 5 × 5 Latin square arrangement with five periods of 20 days each, comprised of 15 days for adaptation to the experimental diets (T1 = 0%, T2 = 1.5%, T3 = 3.0%, T4 = 4.5% and T5 = 6.0% MPM) and 5 days for data collection. Variables were assessed by analysis of variance and regression at P < 0.01 and P < 0.05 of significance, using the Student's test. Animals were kept on pastures and the concentrates were provided at 6 kg/animal divided in two meals per day. The inclusion of 3.0% mesquite pod meal did not affect the intake, digestibility of nutrients and milk yield, but it changed milk composition. The dietary inclusion of 3.0% mesquite pod meal as a palatability enhancer increases the percentage of milk solids, lactose, fat and protein. Therefore, mesquite pod meal can be included in diets for dairy cows to improve the nutritive value of milk.

Despite the existence of many preclinical studies, scientific evidence is lacking on the clinical use of alpha-lipoic acid (ALA) for central nervous system disorders. Therefore, we aimed at revising the literature concerning the use of ALA for the treatment of psychiatric and neurological conditions and to point out what is missing for the introduction of this antioxidant to this purpose. For this systematic review we performed a search using PubMed and SCOPUS databases with the following keywords: "alpha-Lipoic Acid AND central nervous system OR psychiatric disorders OR neurological disorders OR mood disorders OR anxiety OR psychosis OR Alzheimer OR Parkinson OR stroke". The total number of references found after automatically and manually excluding duplicates was 1061. After primary and secondary screening 32 articles were selected. Regarding psychiatric disorders, the studies of ALA in schizophrenia are advanced being ALA administration related to the improvement of schizophrenia symptoms and side effects of antipsychotic medication. In neurological disorders, ALA as a supplement was effective in the prevention of Alzheimer disease progression. For stroke, the use of the supplement ALAnerv® (containing 300 mg ALA) presented important results, since it was observed a reversal of clinical parameters and oxidative imbalance in these patients. For other neurological conditions, such as encephalopathy, multiple sclerosis, traumatic brain injury, mitochondrial disorders and migraine, the results are still preliminary. Overall, there is a need of well-designed clinical trials to enhance the clinical evidences of ALA effects for the treatment of neurological and psychiatric conditions.

Farmers often resort to an occasional tillage (strategic tillage (ST)) operation to combat constraints of no-tillage (NT) farming systems. There are conflicting reports regarding impacts of ST and a lack of knowledge around when, where and how ST is implemented to maximise its benefits without impacting negatively on soil and environment. We established 14 experiments during 2012-2015 on farms with long-term history of continuous NT to (i) quantify the associated risks and benefits to crop productivity, soil and environmental health and (ii) explore key factors that need to be considered in decisions to implement ST in an otherwise NT system. Results showed that introduction of ST reduced weed populations and improved crop productivity and profitability in the first year after tillage, with no impact in subsequent 4 years. Soil properties were not impacted in Vertosols; however, Sodosols and Dermosols suffered short-term negative soil health impacts (e.g. increased bulk density). A Sodosol and a Dermosol also posed higher risks of runoff and associated loss of nutrients and sediment during intense rainfall after ST. The ST reduced plant available water in the short term, which could result in unreliable sowing opportunities for the following crop especially in semi-arid climate that prevails in north-eastern Australia. The results show that generally, there were no significant differences in crop productivity and soil health between tillage implements and tillage frequencies between ST and NT. The study suggests that ST can be a viable strategy to manage constraints of NT systems, with few short-term soil and environmental costs and some benefits such as short-term farm productivity and profitability and reduced reliance on herbicides.

Key Message: Overexpression of a tomato TCTP impacts plant biomass production and performance under stress. These phenotypic alterations were associated with the up-regulation of genes mainly related to photosynthesis, fatty acid metabolism and water transport. The translationally controlled tumor protein (TCTP) is a multifaceted and highly conserved eukaryotic protein. In plants, despite the existence of functional data implicating this protein in cell proliferation and growth, the detailed physiological roles of many plant TCTPs remain poorly understood. Here we focused on a yet uncharacterized TCTP from tomato (SlTCTP). We show that, when overexpressed in tobacco, SlTCTP may promote plant biomass production and affect performance under salt and osmotic stress. Transcriptomic analysis of the transgenic plants revealed the up-regulation of genes mainly related to photosynthesis, fatty acid metabolism and water transport. This induced photosynthetic gene expression was paralleled by an increase in the photosynthetic rate and stomatal conductance of the transgenic plants. Moreover, the transcriptional modulation of genes involved in ABA-mediated regulation of stomatal movement was detected. On the other hand, genes playing a pivotal role in ethylene biosynthesis were found to be down-regulated in the transgenic lines, thus suggesting deregulated ethylene accumulation in these plants. Overall, these results point to a role of TCTP in photosynthesis and hormone signaling.

Glutamine has received attention due to its ability to ameliorate the immune system response. Once conventional liposomes are readily recognized and captured by immune system cells, the encapsulation of glutamine into those nanosystems could be an alternative to reduce glutamine dosage and target then to neutrophils. Our goals were to nanoencapsulate glutamine into conventional liposomes (Gln-L), develop an analytical high-performance liquid chromatography (HPLC) method for its quantification, and evaluate the viability of neutrophils treated with Gln-L. Liposomes were prepared using the thin-film hydration technique followed by sonication and characterized according to pH, mean size, zeta potential, and drug encapsulation efficiency (EE%). We also aimed to study the effect of liposomal constituent concentrations on liposomal characteristics. The viability of neutrophils was assessed using flow cytometry after intraperitoneal administration of free glutamine (Gln), Gln-L, unloaded-liposome (UL), and saline solution as control (C) in healthy Wistar rats. The selected liposomal formulation had a mean vesicle size of 114.65 ± 1.82 nm with a polydispersity index of 0.30 ± 0.00, a positive surface charge of 36.30 ± 1.38 mV, and an EE% of 39.49 ± 0.74%. The developed chromatographic method was efficient for the quantification of encapsulated glutamine, with a retention time at 3.8 min. A greater viability was observed in the group treated with glutamine encapsulated compared to the control group (17%), although neutrophils remain viable in all groups. Thus, glutamine encapsulated into liposomes was able to increase the number of viable neutrophils at low doses, thereby representing a promising strategy for the treatment of immunodeficiency conditions.

This study aimed to assess the mechanisms of innate immune function responses to silica-coated iron oxide nanoparticle functionalized with dithiocarbamate groups (IONP) exposure alone and its associated mercury (Hg) in European eel (Anguilla anguilla L.) phagocytes isolated from peritoneum (P-phagocytes), gill (G-phagocytes), head kidney (HK-phagocytes) and spleen (S-phagocytes). The study evaluated viability, phagocytosis, oxidative burst activity (OBA) and lipid peroxidation (LPO). Four groups were made: (1) 2 × 10(6) phagocytes + RPMI-1640 (control), (2) 2 × 10(6) phagocytes + IONP (2.5 mg L(-1)), (3) 2 × 10(6) phagocytes + Hg (50 μg L(-1)) and (4) 2 × 10(6) phagocytes + IONP + Hg. Samplings were performed at 0, 2, 4, 8, 16, 24, 48 and 72 h of exposure. A. anguilla P-, G-, HK- and S-phagocytes in vitro exposure to IONP alone revealed either increased (except HK-phagocytes at 16 h) or no change in viability, suggesting that the cells are metabolically active and resistant to IONP exposure alone. In terms of phagocytes overactivation and reactive oxygen species (ROS) production as an indirect mechanism of immunotoxicity, the phagocytes responded in the following manner: P- > S- > HK- = G-phagocytes for IONP exposure alone, S- > HK- > P- = G-phagocytes for Hg exposure alone and HK- > G- = S- > P-phagocytes for concomitant exposure. Overall, considering Hg as a surrogate for metals and its association with IONP, as well as the likelihood that it could pose a serious threat to aquatic organisms by modulating their immune defense mechanisms if accidentally discharged into the aquatic environment, current results suggest that the step of IONP-metal complex removal must not be underrated and should be processed without any more ado.

Background: Depression is the most widely prevalent psychiatric disorder in dialysis and can result in a lower quality of life (QoL) and probably a worse nutritional status. The aim of this study was to evaluate the associations among depression, QoL and nutritional status in hemodialysis., Methods: This study included 104 patients on dialysis longer than 3 months, aged 18-65 years. Their nutritional status was assessed according to body mass index (BMI), serum albumin and phase angle; QoL was assessed using the SF-36 questionnaire, and their risk of depression was measured using the Beck Depression Inventory (BDI). Associations among depression, QoL and nutritional status were assessed using Fisher's exact test, and correlations between continuous variables were examined by Pearson's coefficient., Results: Depression was detected in 65.3% of patients, and all of the dimensions of QoL were compromised, with the worst values observed for role-functioning physical (RP) and general health (GH). A negative correlation between the dimensions of QoL and BDI score was observed. Serum albumin was significantly correlated with general health (GH), and phase angle with physical functioning (PF)., Conclusion: A high prevalence of depression was detected, as well as reduced mean scores for QoL. BDI score was significantly correlated with both physical and mental components of QoL. Phase angle was associated with physical functioning (PF) and physical component summary scales (PCS), whereas no nutritional marker was associated with the BDI score. The periodic monitoring of depression should be implemented in hemodialysis, in addition to monitoring QoL and nutritional status. A better understanding of these relationships could result in a better efficiency of dialysis treatment.
