The Bacillus pumilus SG isolated from soil samples at the Persian Gulf was analyzed for its ability to produce biosurfactant. Various screening techniques were used for evaluating biosurfactant production and confirming biosurfactant presence in the culture supernatant. Most n-alkanes in the bacterial culture media were effectively degraded in the presence of biosurfactant acquired from the bacteria. The highest interfacial tension (IT) reduction (42 mN/m) was obtained at 24-h fermentation time (exponential phase) and did not change significantly afterwards. The glycolipid structure of the biosurfactant was revealed through NMR and FTIR spectroscopy analysis. Two-level factorial design was then applied for optimization of biosurfactant production, where a maximal reduction of culture broth IT (30 mN/m) acquired in the presence of crude oil (0.5%, v/v), NaNO 3 (1 g/L), yeast extract (1 g/L), peptone (2 g/L) and temperature of 25 °C. The produced biosurfactant that exhibited a critical micelle concentration of 0.1 mg/ml was thermally stable. The glycolipid biosurfactant also displayed significant antibacterial activities against both Gram-positive and Gram-negative bacteria. The maximum inhibition of glycolipids biosurfactant was found against Acinetobacter strains (zone of inhibition, 45 mm). In addition, antibiofilm activities with a 50-90% biofilm reduction percent were indicated by the glycolipid biosurfactant. In conclusion, the glycolipid biosurfactant produced by B. pumilus SG revealed a wide range of functional properties and was verified as a good candidate for biomedical application. Evidence shows that nanoparticles exert lower toxicity, improved targeting, and enhanced bioactivity, and provide versatile means to control the release profile of the encapsulated moiety. Among different NPs, inorganic nanoparticles (Ag, Au, Ce, Fe, Se, Te, Zn, etc.) possess a considerable place owing to their unique bioactivities in nanoforms. Selenium, an essential trace element, played a vital role in the growth and development of living organisms. It has attracted great interest as a therapeutic factor without significant adverse effects in medicine at recommended dose. Selenium nanoparticles can be fabricated by physical, biological, and chemical approaches. The biosynthesis of nanoparticles is shown an advance compared to other procedures, because it is environmentally friendly, relatively reproducible, easily accessible, biodegradable, and often results in more stable materials. The effect of size, shape, and synthesis methods on their applications in biological systems investigated by several studies. This review focused on the procedures for the synthesis of selenium nanoparticles, in particular the biogenesis of selenium nanoparticles and their biomedical characteristics, such as antibacterial, antiviral, antifungal, and antiparasitic properties. Wild-type microorganisms have become tolerant to higher antibiotic and antimicrobial agent concentrations due to the global increase in antibiotic consumption. Green-synthesized nanoparticles (NPs) have been proposed as potential antimicrobial agents to overcome the problem. This research prepared cadmium nanoparticles (Cd NPs) using Artemisia persica extract. To clarify the biological behavior of Cd NPs and Cd (NO 3 ) 2 , cytotoxicity, antibacterial, anti-biofilm, and biocompatible experiments were performed. Since Cd toxicity is associated with liver, kidney damage, and other deficits, HepG2 and HUVEC cell lines were employed as the in vitro cytotoxicity models. Cd NPs had a lower cytotoxic effect than Cd (NO 3 ) 2 against both HepG2 and HUVEC cells. The Cd NPs exhibited no hemolysis activity. The antibacterial and anti-biofilm studies were conducted using gram-positive Staphylococcus aureus and gram-negative Proteus mirabilis and Pseudomonas aeruginosa with the ability to form severe adherent biofilms. The antibacterial activity of Cd NPs against clinically isolated S. aureus, P. mirabilis, and P. aeruginosa was above 2560 µg mL - 1 . The Cd NPs (640 µg mL - 1 ) decreased the biofilm formation of S. aureus, P. mirabilis, and P. aeruginosa by 24.6%, 31.6%, and 26.4%, respectively.Moreover, adding Cd NPs (100 µg/disc) to antibiotic discs increased the antibacterial activity of vancomycin, gentamicin, tetracycline, streptomycin, meropenem, and kanamycin against Methicillin-resistant S. aureus, significantly. Due to the emergence of resistant microorganisms, Cd NPs can be used as an exciting material to counterattack global health problems. In this study, platinum nanoparticles (Pt NPs) were synthesized by a green method using an aqueous extract of Eucalyptus camaldulensis with assistance of microwave irradiation (850 W) and their physicochemical characteristics were studied by UV-visible spectroscopy, scanning electron microscopy (SEM), energy-dispersive X-ray spectroscopy (EDS), X-ray diffraction (XRD), and Fourier transform infrared spectroscopy (FTIR) analyses. Antioxidant activities, hemocompatibility, and cytotoxic effects of the prepared Pt NPs were then evaluated. The attained results showed that the newly formed Pt NPs possess a size range between 7.4 and 11.2 nm. These spherical-shaped NPs were slightly aggregated and held various functional groups on their surface. The antioxidant activity of Pt nanostructures was comparable to that of butylated hydroxyl anisole at concentrations higher than 320 µg/mL. At the same concentration of 640 μg/mL, the scavenging activities were 3.36 ± 0.9% (hexachloroplatinic acid) and 52.13 ± 0.43% (Pt NPs). The results of hemolytic assay revealed satisfactory hemocompatibility of the Pt NPs even at the concentration as high as 4 mg/mL (hemolysis percent equal to 3.5 ± 1.3%). The cytotoxicity studies revealed that MCF-7, A549, and 3T3 cell lines treated with hexachloroplatinic acid and cisplatin for 24 h and 48 h showed a higher percentage of cell death compared with the Pt NPs. After 24 h, for A549, 3T3, and MCF-7 cells exposed to Pt NPs, the cell viability was measured to be 80 ± 3.2%, 96 ± 1%, and 89 ± 2.6%, respectively, at concentration of 640 µg/mL. In the present study, multiwalled carbon nanotubes (MWCNTs) were functionalized with glycyrrhizin and Tween 80 and applied for immobilization of Pseudomonas cepacia lipase ( Pc L). Characterization of f-MWCNTs was performed through Fourier-transform infrared spectroscopy, thermal gravimetric, field emission scanning electron microscopy, and energy-dispersive X-ray spectroscopy analysis. The optimum specific activity of immobilized Pc L (studied by Plackett-Burman statistical design) occurred at 0.3 mg/mL of f-MWCNTs, 25 mM of phosphate buffer (pH 6.0), 15 min sonication time, 8 U/mL of enzyme concentration, and 24 h immobilization time at 4 °C in the absence of glutaraldehyde. In these conditions, the specific activity was 16.57 ± 0.71 U/mg, which was very close to the predicted amount (16.62 ± 0.64 U/mg). The results of thermal and pH stability showed that the stability of immobilized Pc L was higher than that of the free Pc L. The activity of immobilized Pc L on f-MWCNTs held 93% after being incubated for 60 min at 70 °C. Moreover, the immobilized Pc L on f-MWCNTs retained about 65% of its initial activity after 30 days of storage at 25 °C. In addition, about 50% of initial activity of immobilized Pc L retained after 10 cycles of uses. The present study was designed to propose a simple, cost-effective, and efficient method for the preparation of a biocompatible composite made from magnetic diatomaceous earth (mDE) coated by aminopropyltriethoxysilane (APTES) and its application for immobilization of porcine pancreatic lipase (PPL). The produced mDE-APTES was instrumentally characterized and the obtained results of FTIR analysis and scanning electron microscopy equipped by energy-dispersive X-ray spectroscopy (SEM-EDS) showed successful coating of APTES on mDE surface. PPL was then immobilized onto mDE to obtain the biocatalyst of PPL@mDE (immobilization yield and efficiency of 78.0 ± 0.3% and 80.1 ± 0.6, respectively) and the presence of enzyme was confirmed by EDS method. The attained results of the reusability of PPL@mDE revealed that 57% of the initial activity was retained after 11 cycles of biocatalyst application. PPL@mDE demonstrated higher storage stability than the free enzyme at 4 °C, 25 °C, and 37 °C. The apparent K m (2.35 ± 0.12 mM) and V max (13.01 ± 0.64 µmol/min) values for the immobilized enzyme were considerably altered compared to those of the free enzyme ( p  > 0.05). PPL@mDE was subsequently employed for the synthesis of banana flavor (isoamyl acetate) in n -hexane, which yields an esterification percentage of 100 at 37 °C after 3 h. Extremophilic microbial derived lipases have been widely applied in different biotechnological processes due to their resistance to harsh conditions such as high salt concentration, elevated temperature, and extreme acidic or alkaline pH. The present study was designed to overproduce the halophilic, thermoalkalophilic lipase of Bacillus atrophaeus FSHM2 through chemically induced random mutagenesis and optimization of cultural medium components assisted by statistical experimental design. At first, improvement of lipase production ability of B. atrophaeus FSHM2 was performed through exposure of the wild bacterial strain to ethidium bromide for 5-90 min to obtain a suitable mutant of lipase producer (designated as EB-5, 4301.1 U/l). Afterwards, Plackett-Burman experimental design augmented to D-optimal design was employed to optimize medium components (olive oil, maltose, glucose, sucrose, tryptone, urea, (NH 4 ) 2 SO 4 , NaCl, CaCl 2 , and ZnSO 4 ) for lipase production by the EB-5 mutant. A maximum lipase production of 14,824.3 U/l was predicted in the optimum medium containing 5% of olive oil, 0.5% of glucose, 0.5% of sucrose, 2% of maltose, 2.5 g/l of yeast extract, 1.75 g/l of urea, 1.75 g/l of (NH 4 ) 2 SO 4 , 2.5 g/l of tryptone, 2 g/l of NaCl, 1 g/l of CaCl 2 , and 1 g/l of ZnSO 4 . Lignocellulose bioconversion is a harsh process requiring the use of surfactants and organic solvents. Consequently, the incorporation of laccases in this bioconversion requires the bioprospecting of enzymes that can remain stable under extreme conditions. An extracellular, highly stable laccase was produced by the halophilic isolate Aquisalibacillus elongatus in submerged liquid culture fermentation. Statistical and non-statistical strategies gave the highest enzymatic activity (8.02 U mL -1 ) following addition of glucose (1.7 g L -1 ), copper sulfate (0.8 g L -1 ), urea (15 g L -1 ), and CaCl 2 (0.8 g L -1 ). The enzyme, after purification using a synthetic affinity support, delignified a peanut shell substrate by 45%. A pH of 8.0 and a temperature of 35 °C were optimal for delignification of this bio-waste material. Addition of [Bmim][PF 6 ], 1,4-dioxane, acetone, and HBT promoted this bio-waste delignification. Bio-treatment in the presence of 50% [Bmim][PF 6 ] gave a maximal lignin removal of 87%. The surfactants tested had no significant effects on the delignification yield. The laccase also detoxified the toxic phenols found in peanut shell waste. The high catalytic efficiency of this enzyme against a lignocellulosic sample under extreme conditions suggests the suitability of this laccase for industrial applications.

Background: Enzymatic elimination of synthetic dyes, one of the most environmentally hazardous chemicals, has gained a great interest during the two last decades. The present study was performed to evaluate the decolorization and detoxification potential of the purified laccase of Paraconiothyrium variabile in both non-assisted and hydroxybenzotriazole-aided form against six azo dyes., Results: The obtained results showed that Acid Orange 67, Disperse Yellow 79, Basic Yellow 28, Basic Red 18, Direct Yellow 107, and Direct Black 166 were decolorized up to 65.3, 53.3, 46.7, 40.7, 34, and 26.2 %, respectively, after 1 h treatment with laccase (0.5 U/mL). Addition of HBT up to 5 mM enhanced decolorization percent of all the investigated dyes. The results of kinetic study introduced the monoazo dye of Acid Orange 67 as the most suitable substrate for laccase with K m of 0.49 mM and V max of 189 mmol/min/mg. Evaluation the toxic effect of laccase-treated dye sample based on the growth inhibition of standard bacterial strains revealed decrease in toxicity of all applied dyes after treatment by laccase., Conclusions: Application of the P. variabile laccase as biocatalyst efficiently decreased the toxicity of all studied synthetic azo dyes.

Background: In recent years, enzymatic-assisted removal of hazardous dyes has been considered as an alternative and eco-friendly method compared to those of physicochemical techniques. The present study was designed in order to obtain the optimal condition for laccase-mediated (purified from the ascomycete Paraconiothyrium variabile) decolorization of Acid Blue 92; a monoazo dye, using response surface methodology (RSM). So, a D-optimal design with three variables, including pH, enzyme activity, and dye concentration, was applied to optimize the decolorization process. In addition, the kinetic and energetic parameters of the above mentioned enzymatic removal of Acid Blue 92 was investigated., Results: Decolorization of Acid Blue 92 was maximally (94.1% ± 2.61) occurred at pH 8.0, laccase activity of 2.5 U/mL, and dye concentration of 75 mg/mL. The obtained results of kinetic and energetic studies introduced the laccase-catalyzed decolorization of Acid Blue 92 as an endothermic reaction (Ea, 39 kJ/mol; ΔS, 131 J/mol K; and ΔH, 40 kJ/mol) with K m and V max values of 0.48 mM and 227 mM/min mg, respectively. Furthermore, the results of microtoxicity study revealed that the toxicity of laccase-treated dye was significantly reduced compared to the untreated dye., Conclusions: To sum up, the present investigation introduced the Paraconiothyrium variabile laccase as an efficient biocatalyst for decolorization of synthetic dye Acid Blue 92.

Background: Elimination of hazardous phenolic compounds using laccases has gained attention during recent decades. The present study was designed to evaluate the ability of the purified laccase from Paraconiothyrium variabile (PvL) for elimination of phenol and the endocrine disrupting chemical bisphenol A. Effect of laccase activity, pH, and temperature on the enzymatic removal of the mentioned pollutants were also investigated., Results: After 30 min treatment of the applied phenolic pollutants in the presence of PvL (5 U/mL), 80% of phenol and 59.7% of bisphenol A was removed. Increasing of laccase activity enhanced the removal percentage of both pollutants. The acidic pH of 5 was found to be the best pH for elimination of both phenol and bisphenol A. Increasing of reaction temperature up to 50°C enhanced the removal percentage of phenol and bisphenol A to 96.3% and 88.3%, respectively., Conclusions: To sum up, the present work introduced the purified laccase of P. variabile as an efficient biocatalyst for removal of one of the most hazardous endocrine disruptor bisphenol A.

Background: Decolorization of hazardous synthetic dyes using laccases in both free and immobilized form has gained attention during the last decades. The present study was designed to prepare immobilized laccase (purified from Paraconiothyrium variabile) on porous silica beads followed by evaluation of both free and immobilized laccases for decolorization of two synthetic dyes of Acid Blue 25 and Acid Orange 7. Effects of laccase concentration, pH and temperature alteration, and presence of 1-hydroxybenzotriazole (HBT) as laccase mediator on decolorization pattern were also studied. In addition, the kinetic parameters (Km and Vmax) of the free and immobilized laccases for each synthetic dye were calculated., Results: Immobilized laccase represented higher temperature and pH stability compare to free one. 39% and 35% of Acid Blue 25 and Acid Orange 7 was decolorized, respectively after 65 min incubation in presence of the free laccase. In the case of immobilized laccase decolorization percent was found to be 76% and 64% for Acid Blue 25 and Acid Orange 7, respectively at the same time. Increasing of laccase activity enhanced decolorization percent using free and immobilized laccases. Relative decolorization of both applied dyes was increased after treatment by laccase-HBT system. After nine cycles of decolorization by immobilized laccase, 26% and 31% of relative activity were lost in the case of Acid Blue 25 and Acid Orange 7, respectively., Conclusions: To sum up, the present investigation introduced the immobilized laccase of P. variabile on porous beads as an efficient biocatalyst for decolorization of synthetic dyes.
