Plasmodial resistance to artemisinin-based combination therapies emphasizes the need for new drug development to control malaria. This paper describes the antiplasmodial activity of metabolites produced by endophytic fungi of three Cameroonian plants. Ethyl acetate extracts of fungi cultivated on three different media were tested against Plasmodium falciparum chloroquine-sensitive (Pf3D7) and chloroquine-resistant (PfINDO) strains using the SYBR green florescence assay. Selected endophytes were further grown in potato dextrose broth supplemented with small organic elicitors and their extracts tested for activity. The effect of elicitors on de novo metabolite synthesis was assessed by reverse-phase HPLC. Activity screening of 81 extracts indicated that Aspergillus niger 58 (IC 50 2.25-6.69 μg/mL, Pf3D7), Fusarium sp. N240 (IC 50 1.62-4.38 μg/mL, Pf3D7), Phomopsis sp. N114 (IC 50 0.34-7.26 μg/mL, Pf3D7), and Xylaria sp. N120 (IC 50 2.69-6.77 μg/mL, Pf3D7) produced potent extracts when grown in all three media. Further culture of these endophytes in potato dextrose broth supplemented with each of the eight small organic elicitors and subsequent extracts screening indicated the extract of Phomopsis sp. N114 grown with 1% 1-butanol to be highly selective and extremely potent (IC 50 0.20-0.33 μg/mL; SI > 666). RPHPLC profiles of extracts of Phomopsis sp. N114 grown with or without 1-butanol showed some peaks of enhanced intensities in the former without any qualitative change in the chromatograms. This study showed the ability of selected endophytes to produce potent and selective antiplasmodial metabolites in varied culture conditions. It also showed how the production of desired metabolites can be enhanced by use of small molecular weight elicitors.

For screening bilobalide (BB)-producing endophytic fungi from medicinal plant Ginkgo biloba, a total of 57 fungal isolates were isolated from the internal stem, root, leaf, and bark of the plant G. biloba. Fermentation processes using BB-producing fungi other than G. biloba may become a novel way to produce BB, which is a terpene trilactones exhibiting neuroprotective effects. In this study, a BB-producing endophytic fungal strain GZUYX13 was isolated from the leaves of G. biloba grown in the campus of Guizhou University, Guiyang city, Guizhou province, China. The strain produced BB when grown in potato dextrose liquid medium. The amount of BB produced by this endophytic fungus was quantified to be 106 μg/L via high-performance liquid chromatography (HPLC), substantially lower than that produced by the host tissue. The fungal BB which was analyzed by thin layer chromatography (TLC) and HPLC was proven to be identical to authentic BB. The strain GZUYX13 was identified as Pestalotiopsis uvicola via morphology and ITS rDNA phylogeny. To the best of our knowledge, this is the first report concerning the isolation and identification of endophytic BB-producing Pestalotiopsis spp. from the host plant, which further proved that endophytic fungi have the potential to produce bioactive compounds.

A new species of Neopestalotiopsis based on both morphological and molecular characteristics is described. Neopestalotiopsis iranensis sp. nov. isolated from rotted strawberry (Fragaria ananassa) fruits as well as from stolon and leaf lesions in Kurdistan province, Iran. Initially, light tan and sunken spots developed on fruits and resulted in a soft decay of the fruit flesh. The new species is morphologically distinguished from similar species with different conidium size and by possessing longer apical appendages, as well as some knobbed basal appendages. Phylogenetic analyses (Bayesian inference, maximum likelihood and maximum parsimony analyses) based on internal transcribed spacer, β-tubulin, and partial translation elongation factor 1-alpha combined gene sequences also indicated that this species is phylogenetically distinct from others. Moreover, strawberry crop is introduced here as a new host for N. mesopotamica.

An endophytic Xylaria sp., having broad antimicrobial activity, was isolated and characterized from Ginkgo biloba L. From the culture extracts of this fungus, a bioactive compound P3 was isolated by bioactivity-guided fractionation and identified as 7-amino-4-methylcoumarin by nuclear magnetic resonance, infrared, and mass spectrometry spectral data. The compound showed strong antibacterial and antifungal activities in vitro against Staphylococcus aureus [minimal inhibitory concentrations (MIC) 16 microg.ml(-1)], Escherichia coli (MIC, 10 microg.ml(-1)), Salmonella typhia (MIC, 20 microg.ml(-1)), Salmonella typhimurium (MIC, 15 microg.ml(-1)), Salmonella enteritidis (MIC, 8.5 microg.ml(-1)), Aeromonas hydrophila (MIC, 4 microg.ml(-1)), Yersinia sp. (MIC, 12.5 microg.ml(-1)), Vibrio anguillarum (MIC, 25 microg.ml(-1)), Shigella sp. (MIC, 6.3 microg.ml(-1)), Vibrio parahaemolyticus (MIC, 12.5 microg.ml(-1)), Candida albicans (MIC, 15 microg.ml(-1)), Penicillium expansum (MIC, 40 microg.ml(-1)), and Aspergillus niger (MIC, 25 microg.ml(-1)). This is the first report of 7-amino-4-methylcoumarin in fungus and of the antimicrobial activity of this metabolite. The obtained results provide promising baseline information for the potential use of this unusual endophytic fungus and its components in the control of food spoilage and food-borne diseases.

Two wood-dwelling ascomycetes, Xylaria hypoxylon and Xylaria polymorpha, were isolated from rotting beech wood. Lignin degradation was studied following the mineralization of a synthetic [formula: see text]-labelled lignin in solid and liquid media. Approximately 9% of the synthetic lignin was mineralized by X. polymorpha during the growth on beech wood meal, and the major fraction (65.5%) was polymerized into water- and dioxan-insoluble material. Both fungi produced laccase (up to 1,200 U l-1) in an agitated complex medium based on tomato juice; peroxidase activity (<80 U l-1) was only detected for X. polymorpha in soybean meal suspension. The enzymatic attack of X. polymorpha on beech wood resulted in the formation of three fractions of water-soluble lignocellulose fragments with molecular masses of 200, 30 (major fraction) and 3 kDa, as demonstrated by high-performance size exclusion chromatography. This fragment pattern differs considerably from that of the white-rot fungus Bjerkandera adusta, which preferentially released smaller lignocellulose fragments (0.8 kDa). The finding that X. polymorpha produced large lignocellulose fragments, along with the fact that high levels of hydrolytic enzymes (esterase 630 U l-1, xylanase 120 U l-1) were detected, indicates the cleavage of bonds between the lignin and hemicellulose moieties.

A mitochondrial DNA plasmid was detected in an isolate of Ophiostoma novo-ulmi with a degenerative disease. The DNA plasmid was shown to be derived from the mitochondrial DNA and to map to a region corresponding to the large ribosomal RNA coding region. The DNA plasmid was not transmitted into sexual (ascospore) progeny, irrespective of whether the diseased isolate acted as the female or male parent. Transmission of the disease to healthy, plasmid-free, "recipient" isolates by hyphal anastomosis was not accompanied by transfer of mitochondrial DNA or DNA plasmid from the diseased "donor" isolate, but resulted in de-novo generation of different plasmids, derived from the recipient's mitochondrial DNA.
