Concerns are repeatedly raised about possible adverse respiratory effects of wearing filtering face pieces (FFP) during physical activity. This study compared the impact of FFP type 2 (NF95) on pulmonary function, blood gas values, metabolism and discomfort during light, moderate and vigorous physical activity. Healthy adults (n = 13; 6 females, 7 males; mean 31.3, SD 5.5 years) participated in this randomized two-armed (Ergometer cycling with a FFP type 2 vs. no mask) crossover trial. Baseline cardiopulmonary exercise testing and two interventions (masked and unmasked ergometer cycling 40%, 50% and 70% VO2max, 10 min each) were separated by 48 h washout periods. Spiroergometric data (End tidal carbon dioxide partial pressure PetCO2; breathing frequency; inspiration time), blood gas analysis outcomes (capillary carbon dioxide partial pressure, pCO2) and subjective response (Breathing effort and perceived exertion) were contrasted between conditions using ANOVAs. All participants completed the crossover trial, seven started with the FFP2 condition (No adverse events or side effects). FFP2 decreased breathing frequency, prolonged inspiration time, increased perceived breathing effort and PetCO2 (p <.05). Blood pCO2 in millimetres mercury increased during exercise with 50%VO2max (mean 36.67, SD 3.19 vs. mean 38.46, SD 2.57; p <.05) and 70%VO2max (35.04, 2.84 vs. 38.17, 3.43; p <.05) but not during exercise with 40%VO2max (36.55, 2.73 vs. 38.70). Perceived exertion was not affected (p > 0.05) by mask wearing. Conclusion: Mask-induced breathing resistance decreased respiratory performance and limited pulmonary gas exchange. While FFP2 affected subjective breathing effort per se, invasive diagnostics showed that statistically significant metabolic effects are induced from moderate intensity upwards. Background: The leaf epidermis plays an important role in the transmission of light and the regulation of water and gas exchange, which influences the photosynthesis of mesophyll cells. Small papillae (SP) are one of the important structural elements of the leaf epidermis. The mechanism of the effect that small papillae have on rice leaf photosynthetic performance remains unclear. Results: In this study, a small papilla deficient 25 (spd25) mutant was isolated from japonica rice Longjin1. Small papillae were absent on the adaxial and abaxial leaf surfaces of the spd25 mutant and the silicon and cuticular wax content in the spd25 mutant leaves decreased. Map-based cloning and functional analysis revealed that SPD25, encoding a guanine nucleotide exchange factor for Rop, is a novel allele of OsRopGEF10. The spd25 mutant showed an increased water loss rate and reduced relative water content. The lower stomatal conductance in the spd25 mutant prevented water loss but decreased the intercellular CO2 concentration and net assimilation rate. The fluorescence parameters showed that the inhibited CO2 assimilation reaction feedback regulated the photochemical electron-transfer reaction, but the performance of Photosystem II was stable. Further analysis indicated that the excess light energy absorbed by the spd25 mutant was dissipated in the form of non-photochemical quenching to avoid photodamage through the optical properties of small papillae. Conclusions: SPD25 regulates the development of small papillae on the surface of rice leaves, which play an important role in balancing photosynthetic gas exchange and water loss. Drought timing determines the degree to which dry events impact ecosystems, with the ability of key processes to withstand change differing between drought periods. Findings indicate that drought timing effects vary across ecosystems, with few studies focusing on alpine grasslands. We conducted a mesocosm experiment using small grassland monoliths collected in September from the high Alps and left to overwinter at 0 °C until the experiment began in lowland Italy under late-winter outdoor conditions. Together with watered controls, we imposed three different drought treatments (zero precipitation): (1) one-month early-drought immediately after simulated snowmelt; (2) one-month mid-drought a month after melt-out; and (3) continuous two-month drought across the entire experimental period. Ecosystem responses were assessed by measuring CO2 fluxes, while vegetation responses were investigated by measuring aboveground net primary production (ANPP) of graminoids and forbs and post-harvest resprouting after one-month rehydration. We found that ecosystem respiration and gross ecosystem production (GEP) during the day were more negatively affected by mid-season drought compared to drought starting early in the season. By the end of treatments, GEP reduction under mid-season drought was similar to that of a continuous two-month drought. ANPP reduction was similar in early- and mid-drought treatments, showing a greater decrease under an enforced two-month period without precipitation. Plant resprouting, however, was only reduced in full- and mid-season drought pots, with forbs more negatively affected than graminoids. Seasonal soil moisture variation can account for these patterns: remaining winter moisture allowed almost full canopy development during the first month of the season, despite precipitation being withheld, while soil moisture depletion in the second month, resulting from higher temperatures and greater biomass, caused a collapse of gas exchange and diminished plant resprouting. Plant susceptibility to salt depends on several factors from its genetic makeup to modifiable physiological and biochemical status. We used lemongrass (Cymbopogon flexuosus) plants as a relevant medicinal and aromatic cash crop to assess the potential benefits of chitosan oligomers (COS) on plant growth and essential oil productivity during salinity stress (160 and 240 mM NaCl). Five foliar sprays of 120 mg L−1 of COS were applied weekly. Several aspects of photosynthesis, gas exchange, cellular defence, and essential oil productivity of lemongrass were traced. The obtained data indicated that 120 mg L−1 COS alleviated photosynthetic constraints and raised the enzymatic antioxidant defence including superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) activities that minimised salt-induced oxidative damage. Further, stomatal conductance (gs) and photosynthetic CO2 assimilation (A) were improved to support overall plant development. The same treatment increased geraniol dehydrogenase (GeDH) activity and lemongrass essential oil production. COS-induced salt resilience suggests that COS could become a useful biotechnological tool in reclaiming saline soil for improved crop productivity, especially when such soil is unfit for leading food crops. Increased soil salinization is among the main factors that limits safe rice production. Arbuscular mycorrhizal fungi (AMF) have been shown to alleviate the toxic effects of salt stress in plants. However, more studies on AMF combined with other functional microorganisms are needed to further improve salt tolerance in rice. Therefore, the compound inoculum Funneliformis mosseae (Fm) together with two functional microorganisms, Piriformospora indica (Pi) and Agrobacterium rhizogenes (Ar) was evaluated for their effect on the rice growth, photosynthetic gas exchange parameters, ion homeostasis, and the expression of salt tolerance-related genes under 0, 80, 120 and 160 mM salt stress conditions. The results showed that: (1) the rice seedling biomass of the AMF compound inoculant treatment group was significantly higher than that of the non-inoculation treatment group (P < 0.05); (2) under NaCl stress, inoculation with AMF compound inoculants can activate the rice antioxidant enzyme system and improve osmoregulation ability; (3) AMF compound inoculants can increase the concentration of K+ in the plant and inhibit the transfer of Na+ to rice leaves, maintaining a high K+/Na+; and (4) AMF compound inoculants could induce and regulate the overexpression of genes related to salt tolerance, photosynthesis and ion homeostasis in rice, and improve the tolerance of rice under salt stress. Our study showed that AMF compound inoculants could improve the adaptability of rice under NaCl stress and promote plant growth by regulating the photosynthetic gas exchange parameter, reactive oxygen species (ROS) scavenging ability, and ion homeostasis of plants. Production of sugarcane and more recently of energy cane strengthen renewable bioenergy production capacity. However, droughts resulting from climate change have limited the production of these crops. One of the strategies to attenuate water deficit damage in these crops is the use of silicate, which contributes to plant physiology. This strategy is likely to increase water use efficiency, thus promoting crop sustainability. Notwithstanding, studies on this issue are still incipient. This study assesses whether Si applied via fertigation and foliar spraying in the seedling production phase and as a complement after seedling transplanting to the soil is efficient in attenuating water deficit in sugarcane and energy cane. The study further elucidates physiological and biochemical mechanisms involved in this process. For this, the authors conducted two experiments: one with sugarcane and the other with energy cane. Treatments were arranged in randomized blocks with 5 replications, in a 2 × 2 factorial scheme. Factors consisted of the absence (-Si) and presence of Si (+ Si) applied via fertigation and foliar spraying; and two water regimes: 70% (without water deficit) and 30% (severe water deficit) of the soil water retention capacity. Silicon was supplied during the formation phase of presprouted seedlings and during the transplanting of seedlings to pots filled with samples of Entisol (Quartzipsamment). In these pots, water regimes were induced from 7 to 160 days after transplanting. Severe water deficit reduced the water content and water potential of plants. This situation induced oxidative stress and impaired gas exchange and photosynthetic water use efficiency, reducing plant growth. Silicon supply via fertigation in association with foliar spraying in the seedling formation phase with complementation after transplanting was efficient in increasing Si accumulation in the plants. Silicon was effective in attenuating severe water deficit damage up to initial culm formation through mechanisms that maintain water and physiological balance by favoring the antioxidant defense system in sugarcane and energy cane plants. Ammopiptanthus mongolicus is the only evergreen broad-leaved shrub present in arid areas of Northwest China and plays an important role in maintaining the stability of the local desert ecosystem. It can survive under extreme temperatures (e.g., extreme low temperature: − 24.8 °C and extreme high temperature: 37.7 °C). To understand the gene expression-physiological regulation network of A. mongolicus in extreme temperature environments, we monitored the changes in gene expression and photosynthetic traits of the leaves. The results showed that at low temperatures, the net photosynthetic rates (A), Fv'/Fm' and electron transport rate (ETR) decreased, the Fv/Fm ratio was only 0.32, and the proportion of nonregulatory heat dissipation Y(NO) increased. Based on a KEGG analysis of the differentially expressed genes, 15 significantly enriched KEGG pathways were identified, which were mainly related to circadian rhythm, photosynthesis, lipid metabolism, carbohydrate metabolism, plant hormones and other life activities. At high temperatures, the A value increased, and the proportion of regulatory energy dissipation Y(NPQ) increased. The KEGG analysis identified 24 significantly enriched KEGG pathways, which are mainly related to circadian rhythm, carbon sequestration of photosynthesis, carotenoid biosynthesis, secondary metabolites, cofactors and vitamin metabolism. In general, at the expense of photosynthesis, A. mongolicus can ensure the survival of leaves by increasing Y(NO) levels, regulating the circadian rhythm, increasing the synthesis of unsaturated fatty acids and changing the role of plant hormones. Under high-temperature stress, a high photosynthetic capacity was maintained by adjusting the stomatal conductance (gsw), increasing Y(NPQ), consuming excess light energy, continuously assembling and maintaining PSII, and changing the production of antioxidants. Changes in root growth and metabolism of P in tomato cultivars are necessary in acidic soils in tropical and subtropical regions in response to P deficiency. This increase in the efficiency of phosphorus absorption by tomatoes can significantly reduce the doses of phosphate fertilizers used, as well as, possibly, the more immediate use of P fixed in the soil matrix, with favorable effects on agricultural sustainability, promoting the use of marginal areas in terms of soil fertility, and the national fertilizer economy. The tested hypothesis was that there would be no difference in the uptake and utilization of P by tomato cultivars; therefore, this study investigated the variation in the dynamics of absorption and efficiency of P-use through changes in the root, stem, leaf, gas exchange, and P-use efficiency in tomato cultivars contrasting P-absorption. The experimental design comprised a factorial scheme consisting of two cultivars that were tolerant and sensitive to P deficiency and three P concentrations (control, moderate deficiency, and severe deficiency). P limitation in the tolerant cultivar promoted high dry matter concentration (root, stem, and leaf), leaf area, root volume, nutrient translocation, rate of leaf gas exchange, and efficiency under P-deficiency stress. It was concluded from the research that the variation in the dynamics of absorption and efficiency of P use of the tolerant cultivar increased the production of roots, leaves, and leaf gas exchange under P stress conditions. Stomata—small pores generally found on the leaves of plants—control gas exchange between plant and the atmosphere. Elucidating the mechanism that underlies such control through the regulation of stomatal opening/closing is important to understand how plants regulate photosynthesis and tolerate against drought. However, up-to-date, molecular components and their function involved in stomatal regulation are not fully understood. We challenged such problem through a chemical genetic approach by isolating and characterizing synthetic molecules that influence stomatal movement. Here, we describe that a small chemical collection, prepared during the development of C–H amination reactions, lead to the discovery of a Stomata Influencing Molecule (SIM); namely, a sulfonimidated oxazole that inhibits stomatal opening. The starting molecule SIM1 was initially isolated from screening of compounds that inhibit light induced opening of dayflower stomata. A range of SIM molecules were rapidly accessed using our state-of-the-art C–H amination technologies. This enabled an efficient structure–activity relationship (SAR) study, culminating in the discovery of a sulfonamidated oxazole derivative (SIM*) having higher activity and enhanced specificity against stomatal regulation. Biological assay results have shed some light on the mode of action of SIM molecules within the cell, which may ultimately lead to drought tolerance-conferring agrochemicals through the control of stomatal movement. Stomata are epidermal pores formed by pairs of specialized guard cells, which regulate gas exchanges between the plant and the atmosphere. Modulation of transcription has emerged as an important level of regulation of stomatal activity. The AtMYB60 transcription factor was previously identified as a positive regulator of stomatal opening, although the details of its function remain unknown. Here, we propose a role for AtMYB60 as a negative modulator of oxylipins synthesis in stomata. The atmyb60-1 mutant shows reduced stomatal opening and accumulates increased levels of 12-oxo-phytodienoic acid (12-OPDA), jasmonic acid (JA) and jasmonoyl-l-isoleucine (JA-Ile) in guard cells. We provide evidence that 12-OPDA triggers stomatal closure independently of JA and cooperatively with abscisic acid (ABA) in atmyb60-1. Our study highlights the relevance of oxylipins metabolism in stomatal regulation and indicates AtMYB60 as transcriptional integrator of ABA and oxylipins responses in guard cells. The importance of perioperative respiration monitoring is highlighted by high incidences of postoperative respiratory complications unrelated to the original disease. The objectives of this pilot study were to (1) simultaneously acquire respiration rate (RR), tidal volume (TV), minute ventilation (MV), SpO2 and PetCO2 from patients in post-anesthesia care unit (PACU) and (2) identify a practical continuous respiration monitoring method by analyzing the acquired data in terms of their ability and reliability in assessing a patient's respiratory status. Thirteen non-intubated patients completed this observational study. A portable electrical impedance tomography (EIT) device was used to acquire RREIT, TV and MV, while PetCO2, RRCap and SpO2 were measured by a Capnostream35. Hypoventilation and respiratory events, e.g., apnea and hypopnea, could be detected reliably using RREIT, TV and MV. PetCO2 and SpO2 provided the gas exchange information, but were unable to detect hypoventilation in a timely fashion. Although SpO2 was stable, the sidestream capnography using the oronasal cannula was often unstable and produced fluctuating PetCO2 values. The coefficient of determination (R2) value between RREIT and RRCap was 0.65 with a percentage error of 52.5%. Based on our results, we identified RR, TV, MV and SpO2 as a set of respiratory parameters for robust continuous respiration monitoring of non-intubated patients. Such a respiration monitor with both ventilation and gas exchange parameters would be reliable and could be useful not only for respiration monitoring, but in making PACU discharge decisions and adjusting opioid dosage on general hospital floor. The present study investigated the interactive effects of three environmental stress factors elevated CO2, temperature, and drought stress on soybean growth and yield. Experiments were conducted in the sunlit, controlled environment Soil–Plant–Atmosphere–Research chambers under two-level of irrigation (WW-well water and WS-water stress-35%WW) and CO2 (aCO2-ambient 400 µmol mol−1 and eCO2-elevated 800 µmol mol−1) and each at the three day/night temperature regimes of 24/18 °C (MLT-moderately low), 28/22 °C (OT-optimum), and 32/26 °C (MHT-moderately high). Results showed the greatest negative impact of WS on plant traits such as canopy photosynthesis (PCnet), total dry weight (TDwt), and seed yield. The decreases in these traits under WS ranged between 40 and 70% averaged across temperature regimes with a greater detrimental impact in plants grown under aCO2 than eCO2. The MHT had an increased PCnet, TDwt, and seed yield primarily under eCO2, with a greater increase under WW than WS conditions. The eCO2 stimulated PCnet, TDwt, and seed yield more under WS than WW. For instance, on average across T regimes, eCO2 stimulated around 25% and 90% dry mass under WW and WS, respectively, relative to aCO2. Overall, eCO2 appears to benefit soybean productivity, at least partially, under WS and the moderately warmer temperature of this study. [ABSTRACT FROM AUTHOR]

Flux-footprint modelling is commonly used for determining source areas of land–atmosphere gas-exchange observations from a single meteorological tower to determine the relative flux contribution of different areas within the station fetch. In doing so, a conversion from a continuous function to a two-dimensional spatial representation takes place prior to the interpretation of results. This interpreted result is commonly used to validate satellite regional-scale, remote-sensing-based, gas-exchange models. However, little attention has been paid to exactly how the conversion is performed. The conversion to a raster data model can result in the loss of information. This loss can be cell size dependent. Therefore, it is important that the spatial-conversion method used is appropriate for the specific application. We present here an assessment of the influence of spatial resolution (i.e., cell size) and conversion method on the accuracy of modelled flux footprints using tower-based flux instrumentation. Five methods for aggregating flux footprints to a two-dimensional grid are presented and a sensitivity analysis is performed. Two well-validated flux-footprint models are used at spatial resolutions ranging from 10 m to 1000 m. Results indicate significant data loss, change of apparent maximum location, and change in the representative fetch of the point measurement. Furthermore, these results are measurement site dependent. This often-overlooked step in the conversion of a flux-footprint model to a grid is important. These results provide guidelines for those utilizing a flux-footprint model to validate spatial models for assessing gas exchange. [ABSTRACT FROM AUTHOR]

Background: The environmental stresses caused by climate change have become more severe in recent decades, affecting tree growth and physiology. Tropical forests have great potential for global carbon sequestration. However, they suffer from heavy rainfall and prolonged dry periods due to climate change. Swietenia macrophylla King and Melia azedarach L. are economically valuable trees that are widely planted in southern Taiwan. Plantations are exposed to either prolonged dry periods or heavy rainfall within the seasons of tropical monsoon areas. Photo-physiological comparisons may provide information that can improve management of S. macrophylla and M. azedarach plantations in tropical regions. Results: Both species exhibited a midday depression in leaf photosynthesis regardless of the season. The net photosynthetic rate ( P ), stomatal conductance ( g ), and transpiration rate ( E) in the dry season all significantly decreased in both tree species. In addition, M. azedarach used water more efficiently than did S. macrophylla during the dry season, but S. macrophylla had higher P compared with that in M. azedarach during the wet season. Temperature and vapor pressure deficit influenced P variation in S. macrophylla and M. azedarach, respectively. Our data suggested that the P and g of M. azedarach, but not of S. macrophylla, were linearly correlated during the dry season. The reduction of the leaf area was more sever in M. azedarach than in S. macrophylla, thus preventing water loss more efficiently. Conclusions: M. azedarach adapted to drought by reducing total leaf area and maintaining higher P , g , E, and WUE compared with those measured in S. macrophylla during the dry season. M. azedarach is more drought adaptation and more suitable for both humid and semi-humid areas than S. macrophylla, whereas the latter should be limited to more humid areas. [ABSTRACT FROM AUTHOR]

Given the crucial role of land surface processes in global and regional climates, there is a pressing need to test and verify the performance of land surface models via comparisons to observations. In this study, the eddy covariance measurements from 20 FLUXNET sites spanning more than 100 site-years were utilized to evaluate the performance of the Common Land Model (CoLM) over different vegetation types in various climate zones. A decomposition method was employed to separate both the observed and simulated energy fluxes, i.e., the sensible heat flux, latent heat flux, net radiation, and ground heat flux, at three timescales ranging from stepwise (30 min) to monthly. A comparison between the simulations and observations indicated that CoLM produced satisfactory simulations of all four energy fluxes, although the different indexes did not exhibit consistent results among the different fluxes. A strong agreement between the simulations and observations was found for the seasonal cycles at the 20 sites, whereas CoLM underestimated the latent heat flux at the sites with distinct dry and wet seasons, which might be associated with its weakness in simulating soil water during the dry season. CoLM cannot explicitly simulate the midday depression of leaf gas exchange, which may explain why CoLM also has a maximum diurnal bias at noon in the summer. Oxygen flux between aquatic ecosystems and the water column is a measure of ecosystem metabolism. However, the oxygen flux varies during the day in a 'hysteretic' pattern: there is higher net oxygen production at a given irradiance in the morning than in the afternoon. In this study, we investigated the mechanism responsible for the hysteresis in oxygen flux by measuring the daily pattern of oxygen flux, light, and temperature in a seagrass ecosystem ( Zostera muelleri in Swansea Shoals, Australia) at three depths. We hypothesised that the oxygen flux pattern could be due to diel variations in either gross primary production or respiration in response to light history or temperature. Hysteresis in oxygen flux was clearly observed at all three depths. We compared this data to mathematical models, and found that the modification of ecosystem respiration by light history is the best explanation for the hysteresis in oxygen flux. Light history-dependent respiration might be due to diel variations in seagrass respiration or the dependence of bacterial production on dissolved organic carbon exudates. Our results indicate that the daily variation in respiration rate may be as important as the daily changes of photosynthetic characteristics in determining the metabolic status of aquatic ecosystems. Background: In West Africa, natural ecosystems such as woodlands are the main source for energy, building poles and livestock fodder. They probably behave like net carbon sinks, but there are only few studies focusing on their carbon exchange with the atmosphere. Here, we have analyzed CO2 fluxes measured for 17 months by an eddycovariance system over a degraded woodland in northern Benin. Specially, temporal evolution of the fluxes and their relationships with the main environmental factors were investigated between the seasons. Results: This study shows a clear response of CO2 absorption to photosynthetic photon flux density (Qp), but it varies according to the seasons. After a significant and long dry period, the ecosystem respiration (R) has increased immediately to the first significant rains. No clear dependency of ecosystem respiration on temperature has been observed. The degraded woodlands are probably the 'carbon neutral' at the annual scale. The net ecosystem exchange (NEE) was negative during wet season and positive during dry season, and its annual accumulation was equal to +29±16 g C m-2. The ecosystem appears to be more efficient in the morning and during the wet season than in the afternoon and during the dry season. Conclusions: This study shows diurnal and seasonal contrasted variations in the CO2 fluxes in relation to the alternation between dry and wet seasons. The Nangatchori site is close to the equilibrium state according to its carbon exchanges with the atmosphere. The length of the observation period was too short to justify the hypothesis about the 'carbon neutrality' of the degraded woodlands at the annual scale in West Africa. Besides, the annual net ecosystem exchange depends on the intensity of disturbances due to the site management system. Biological soil crusts (BSC) are the dominant functional vegetation unit in some of the harshest habitats in the world. We assessed BSC response to stress through changes in biotic composition, CO2 gas exchange and carbon allocation in three lichen-dominated BSC from habitats with different stress levels, two more extreme sites in Antarctica and one moderate site in Germany. Maximal net photosynthesis (NP) was identical, whereas the water content to achieve maximal NP was substantially lower in the Antarctic sites, this apparently being achieved by changes in biomass allocation. Optimal NP temperatures reflected local climate. The Antarctic BSC allocated fixed carbon (tracked using 14CO2) mostly to the alcohol soluble pool (low-molecular weight sugars, sugar alcohols), which has an important role in desiccation and freezing resistance and antioxidant protection. In contrast, BSC at the moderate site showed greater carbon allocation into the polysaccharide pool, indicating a tendency towards growth. The results indicate that the BSC of the more stressed Antarctic sites emphasise survival rather than growth. Changes in BSC are adaptive and at multiple levels and we identify benefits and risks attached to changing life traits, as well as describing the ecophysiological mechanisms that underlie them. Environmental stimuli, including elevated carbon dioxide levels, regulate stomatal development; however, the key mechanisms mediating the perception and relay of the CO2 signal to the stomatal development machinery remain elusive. To adapt CO2 intake to water loss, plants regulate the development of stomatal gas exchange pores in the aerial epidermis. A diverse range of plant species show a decrease in stomatal density in response to the continuing rise in atmospheric CO2 (ref. 4). To date, one mutant that exhibits deregulation of this CO2-controlled stomatal development response, hic (which is defective in cell-wall wax biosynthesis, ref. 5), has been identified. Here we show that recently isolated Arabidopsis thaliana β-carbonic anhydrase double mutants (ca1 ca4) exhibit an inversion in their response to elevated CO2, showing increased stomatal development at elevated CO2 levels. We characterized the mechanisms mediating this response and identified an extracellular signalling pathway involved in the regulation of CO2-controlled stomatal development by carbonic anhydrases. RNA-seq analyses of transcripts show that the extracellular pro-peptide-encoding gene EPIDERMAL PATTERNING FACTOR 2 (EPF2), but not EPF1 (ref. 9), is induced in wild-type leaves but not in ca1 ca4 mutant leaves at elevated CO2 levels. Moreover, EPF2 is essential for CO2 control of stomatal development. Using cell-wall proteomic analyses and CO2-dependent transcriptomic analyses, we identified a novel CO2-induced extracellular protease, CRSP (CO2 RESPONSE SECRETED PROTEASE), as a mediator of CO2-controlled stomatal development. Our results identify mechanisms and genes that function in the repression of stomatal development in leaves during atmospheric CO2 elevation, including the carbonic-anhydrase-encoding genes CA1 and CA4 and the secreted protease CRSP, which cleaves the pro-peptide EPF2, in turn repressing stomatal development. Elucidation of these mechanisms advances the understanding of how plants perceive and relay the elevated CO2 signal and provides a framework to guide future research into how environmental challenges can modulate gas exchange in plants. A split-split-plot design with three replications in two years of 2009 and 2010 was conducted to investigate the effect of different levels of irrigation water (main plot), salinity of irrigation water (sub-plot) and nitrogen fertilizer rate (sub-subplot) on maize growth rate and gas exchange. Irrigation treatments were I2 (1.0 crop evapotranspiration (ETc)+0.25ETc as leaching), I2 (0.75I1) and I3 (0.5I1) applied at 7-day intervals. The salinity treatments of irrigation were 0.6 (fresh water), 2.0 and 4.0 dS m-1. There were also three nitrogen (N) treatments including 0, 150 and 300 kg N ha-1. Results showed that vegetative growth stage of maize in salinity stress lasted 5% more than that in water stress. The most sensitive trait under water, salinity and nitrogen stress was grain yield (GY). The optimum treatment for maize production is full fresh water application by 150 kg N ha-1. Results also showed that crop growth rate (CGR) was statistically higher in I1 and I2 as 58 and 34% relative to I3 treatment, respectively. Furthermore, CGR was statistically lower in S2 and S3 as 10 and 18% relative to S1, respectively. Besides, N application significantly increased CGR by an average of 15% as compared with no N rate. The net assimilation rate (NAR) reached its maximum value in I2, S2 and N2 relative to other treatments indicating that NAR did not necessarily occurred at maximum LAI conditions. In general, maize had statistically greater NAR in pollination and filling stages relative to other growth stages. Results of gas exchange for maize as a sensitive crop to water deficit, showed that photosynthesis rate (An) and stomatal conductance (gs) were statistically decreased in water deficit by an average of 30 and 43% as compared to full irrigation treatment, respectively. However, reduction in An and gs in salinity conditions was the same as 13% compared to no salinity treatment. Transpiration rate (T) was statistically lower under water and salinity stress by an average of 75 and 26% as compared to no water and salinity stress, respectively. The ratio of An/gs in I2 and I3 was statistically higher as 23% than that obtained in I1 treatment indicating that maximum amount of intrinsic water use efficiency could be achieved by water saving strategies. This result confirmed by transpiration efficiency (An/T) which was statistically higher as 40 and 14% in I3 and S3 relative to full and fresh water irrigation treatment, respectively. In general, there was a decreasing trend in An and gs toward the end of growing season, mainly due to leaves ageing. Similar pattern also occurred in measurements during a day from morning to afternoon. Furthermore, An and gs decreased as vapor pressure deficit (VPD) increased. The relationships between An and gs vs VPD in different growth stages implied that the plant ability to regulate An and gs in response to VPD at leaf level declined in the last growth stages. Aims: There is still limited information about the dynamics of potassium (K) uptake and use over the growth cycle in cereals, but quantification of such relationships is necessary to model crop response to K and improve K fertiliser practice. This study examined the effects of varying K supply on K use efficiency, leaf gas exchange, root and shoot growth at successive stages of plant phenological development, and on seed yield in wheat ( Triticum aestivum L.). Methods: The study was conducted with a low-K loamy sand in a naturally-lit glasshouse. Six rates of soil K were applied as 15, 22.5, 30, 45, 75, 135 mg K kg −1, and six harvests were made to assess growth responses at different stages. Results: Tiller development, shoot dry weight, leaf photosynthesis and transpiration efficiency were closely related to soil K supply with time. Plants with 15 to 45 mg K kg −1 had lower root-to-shoot ratio than those supplied with 75 or 135 mg K kg −1. Plants showed maximal K accumulation in shoots prior to anthesis regardless of K rates, but K use efficiency differed during growth among the treatments. Adequate K supply enhanced seed yield by increasing ear numbers, single seed weight and harvest index. Conclusions: The rates of K supply affect K use efficiency, the root-to-shoot ratio and seed yield in wheat with a key feature of growth on a low-K sand as active K uptake during vegetative growth and subsequent reliance on K redistribution for reproductive growth. In plant ecophysiology, functional leaf traits are generally not assessed in relation to phenological phase of the canopy. Leaf traits measured in deciduous perennial species are known to vary between spring and summer seasons, but there is a knowledge gap relating to the late-summer phase marked by growth cessation and bud set occurring well before fall leaf senescence. The effects of phenology on canopy physiology were tested using a common garden of over 2,000 black cottonwood ( Populus trichocarpa) individuals originating from a wide geographical range (44-60ºN). Annual phenological events and 12 leaf-based functional trait measurements were collected spanning the entire summer season prior to, and following, bud set. Patterns of seasonal trait change emerged by synchronizing trees using their date of bud set. In particular, photosynthetic, mass, and N-based traits increased substantially following bud set. Most traits were significantly different between pre-bud set and post-bud set phase trees, with many traits showing at least 25 % alteration in mean value. Post-bud set, both the significance and direction of trait-trait relationships could be modified, with many relating directly to changes in leaf mass. In Populus, these dynamics in leaf traits throughout the summer season reflected a shift in whole plant physiology, but occurred long before the onset of leaf senescence. The marked shifts in measured trait values following bud set underscores the necessity to include phenology in trait-based ecological studies or large-scale phenotyping efforts, both at the local level and larger geographical scale. Aims: Roots need to be in good contact with the soil to take up water and nutrients. However, when the soil dries and roots shrink, air-filled gaps form at the root-soil interface. Do gaps actually limit the root water uptake, or do they form after water flow in soil is already limiting? Methods: Four white lupins were grown in cylinders of 20 cm height and 8 cm diameter. The dynamics of root and soil structure were recorded using X-ray CT at regular intervals during one drying/wetting cycle. Tensiometers were inserted at 5 and 18 cm depth to measure soil matric potential. Transpiration rate was monitored by continuously weighing the columns and gas exchange measurements. Results: Transpiration started to decrease at soil matric potential ψ between −5 kPa and −10 kPa. Air-filled gaps appeared along tap roots between ψ = −10 kPa and ψ = −20 kPa. As ψ decreased below −40 kPa, roots further shrank and gaps expanded to 0.1 to 0.35 mm. Gaps around lateral roots were smaller, but a higher resolution is required to estimate their size. Conclusions: Gaps formed after the transpiration rate decreased. We conclude that gaps are not the cause but a consequence of reduced water availability for lupins. [ABSTRACT FROM AUTHOR]

Plant stomata display a wide range of short-term behavioural and long-term morphological responses to atmospheric carbon dioxide concentration ([CO]). The diversity of responses suggests that plants may have different strategies for controlling gas exchange, yet it is not known whether these strategies are co-ordinated in some way. Here, we test the hypothesis that there is co-ordination of physiological (via aperture change) and morphological (via stomatal density change) control of gas exchange by plants. We examined the response of stomatal conductance ( G) to instantaneous changes in external [CO] ( C) in an evolutionary cross-section of vascular plants grown in atmospheres of elevated [CO] (1,500 ppm) and sub-ambient [O] (13.0 %) compared to control conditions (380 ppm CO, 20.9 % O). We found that active control of stomatal aperture to [CO] above current ambient levels was not restricted to angiosperms, occurring in the gymnosperms Lepidozamia peroffskyana and Nageia nagi. The angiosperm species analysed appeared to possess a greater respiratory demand for stomatal movement than gymnosperm species displaying active stomatal control. Those species with little or no control of stomatal aperture (termed passive) to C were more likely to exhibit a reduction in stomatal density than species with active stomatal control when grown in atmospheres of elevated [CO]. The relationship between the degree of stomatal aperture control to C above ambient and the extent of any reduction in stomatal density may suggest the co-ordination of physiological and morphological responses of stomata to [CO] in the optimisation of water use efficiency. This study evaluates acclimation of photosynthesis and stomatal conductance in three evolutionary lineages of C, C-C intermediate, and C species grown in the low CO and hot conditions proposed to favo r the evolution of C photosynthesis. Closely related C, C-C, and C species in the genera Flaveria, Heliotropium, and Alternanthera were grown near 380 and 180 μmol CO mol air and day/night temperatures of 37/29°C. Growth CO had no effect on photosynthetic capacity or nitrogen allocation to Rubisco and electron transport in any of the species. There was also no effect of growth CO on photosynthetic and stomatal responses to intercellular CO concentration. These results demonstrate little ability to acclimate to low CO growth conditions in closely related C and C-C species, indicating that, during past episodes of low CO, individual C plants had little ability to adjust their photosynthetic physiology to compensate for carbon starvation. This deficiency could have favored selection for more efficient modes of carbon assimilation, such as C-C intermediacy. The C-C species had approximately 50% greater rates of net CO assimilation than the C species when measured at the growth conditions of 180 μmol mol and 37°C, demonstrating the superiority of the C-C pathway in low atmospheric CO and hot climates of recent geological time. Crassulacean acid metabolism (CAM) and the capacity to store large quantities of water are thought to confer high water use efficiency (WUE) and survival of succulent plants in warm desert environments. Yet the highly variable precipitation, temperature and humidity conditions in these environments likely have unique impacts on underlying processes regulating photosynthetic gas exchange and WUE, limiting our ability to predict growth and survival responses of desert CAM plants to climate change. We monitored net CO assimilation ( A), stomatal conductance ( g), and transpiration ( E) rates periodically over 2 years in a natural population of the giant columnar cactus Carnegiea gigantea (saguaro) near Tucson, Arizona USA to investigate environmental and physiological controls over carbon gain and water loss in this ecologically important plant. We hypothesized that seasonal changes in daily integrated water use efficiency (WUE) in this constitutive CAM species would be driven largely by stomatal regulation of nighttime transpiration and CO uptake responding to shifts in nighttime air temperature and humidity. The lowest WUE occurred during time periods with extreme high and low air vapor pressure deficit ( D). The diurnal with the highest D had low WUE due to minimal net carbon gain across the 24 h period. Low WUE was also observed under conditions of low D; however, it was due to significant transpiration losses. Gas exchange measurements on potted saguaro plants exposed to experimental changes in D confirmed the relationship between D and g. Our results suggest that climatic changes involving shifts in air temperature and humidity will have large impacts on the water and carbon economy of the giant saguaro and potentially other succulent CAM plants of warm desert environments. Immediate leaf functional responses to drought, such as stomatal closure and photosynthetic rate reduction, are already known from short-term studies. We tested the hypothesis that long-term acclimation of leaves to drought includes hydraulic and stomatal anatomical changes and that gas exchange and nitrogen allocation patterns are inevitably adjusted to the new structural status. 26 structural and functional traits in one sensitive cultivar (Simeto) and two drought resistant landraces (Ntopia Heraklion 184, Kontopouli 17) of field- grown wheat ( Triticum turgidum L. var. durum) were examined under four water shortage levels. Drought acclimation responses were more intense in Simeto than in the two landraces. In accordance to the working hypothesis, drought-acclimated leaves showed lower hydraulic conductance due to narrower vessels and higher stomatal and vein densities than the control leaves, resulting in a safe mode of water transfer and consumption which is essential for the survival in water limiting conditions.Irrespectively of genotype and water regime, significant correlations among structural (hydraulic characteristics, stomatal and vein patterns) and functional (gas exchange, nitrogen content) parameters were found, indicating the functional adjustment to the new structural status. The Principal Component Analysis showed that these structure-function interactions reflected the trade-off between growth and protection against water losses (Axis 1), as well as the competition between different sinks (carbon gain vs structural reinforcement and reproductive effort) in N allocation (Axis 2). Drought acclimation in wheat leaf is integrally processed by the coordination of structural and functional parameters in order to compensate for the adverse effects of water shortage. This structure-function network that regulates the transition from normal growth-mode to protection- mode, includes at least two important 'nodal points': xylem conducting efficiency (water entry) and stomatal function (water exit). Water stress causes defoliation, which can reduce yield and root quality of sugar beets (Beta vulgaris L.) through altered gas exchange characteristics of the leaves. In a two-year experiment, three sugar beet cultivars (Europa, Rival and Corsica) were subjected to three defoliation levels (control-C, moderate-MD, severe-SD) and re-watering after their exposition to drought for a month. Leaf physiological traits including net photosynthesis (A), transpiration rate (E), stomatal conductance (gs), intracellular CO2 (Ci), water use efficiency (WUEL-A/E and WUEi-A/gs), leaf N concentration, petiole NO3-N concentration, specific leaf area (SLA), leaf water potential (WP) and leaf water content (LWC), were determined before defoliation and 15, 30 and 40 days after defoliation (DAD). On contrary to previous reports, water-stressed cultivars differed significantly in their leaf physiology; the late-season cultivar Corsica had the lowest E and gs values without any significant reduction in A. Thus, Corsica was the most water-conservative cultivar. Re-watering rapidly restored leaf physiology but a gradual decline, with the progress of DAD, was evident for A, E, gs and Ci. After re-growth, cultivars differed only in WP and LWC with Europa, the early-harvested cultivar, to have the highest values. Thus, the better response (higher yield increase and lower root quality degradation) of Corsica to re-watering and the subsequent re-growth, as reported by Tsialtas et al. (2009), could be ascribed to its better LAI maintenance compared to Europa and Rival rather than to a better leaf physiological response. The SD plants showed the highest leaf physiological activity since they had the highest A, E, gs and leaf N concentration. mmonia (NH) fluxes between beech leaves ( Fagus sylvatica) and the atmosphere were investigated in a 90-year-old forest canopy and related to leaf nitrogen (N) pools and glutamine synthetase (GS) activities. The stomatal ammonia compensation point, χ, was measured by both a twig cuvette and bioassay techniques involving measurements of pH and ammonium (NH) concentration in the leaf apoplastic solution. The χ determined on the basis of the gas exchange measurements followed a seasonal variation with early-season peaks during leaf expansion (9.6 nmol NH mol air) and late-season peaks during leaf senescence (7.3 nmol NH mol air). In the mid-season, the χ of mature green leaves was much lower (around 3 nmol NH mol air) and dropped below the NH concentration in the ambient atmosphere. For comparison, χ obtained by the apoplastic bioassay were 7.0, 3.7 and 6.4 nmol NH mol air in early-, mid-, and late -season, thus agreeing reasonably well with χ values derived from the gas exchange measurements. Potential NH emission fluxes during early and late season were 1.31 and 0.51 nmol m leaf surface area s, respectively, while leaves were a sink for NH during mid-season. During leaf establishment and senescence, both apoplastic and bulk tissue NH concentrations were relatively high coinciding with low activities of glutamine synthetase, which is a key enzyme in leaf N metabolism. In conclusion, the exchange of NH between beech leaves and the atmosphere followed a seasonal variation with NH emission peaks being related to N mobilization during early leaf establishment and remobilization during late leaf senescence. Drought stress is known to limit plant performance in Mediterranean-type ecosystems. We have investigated the dynamics of the hydraulics, gas exchange and morphology of six co-existing Mediterranean woody species growing under natural field conditions during a drought that continued during the entire summer. Based on the observed minimum leaf water potentials, our results suggest that the six co-existing species cover a range of plant hydraulic strategies, from isohydric to anisohydric. These differences are remarkable since the selected individuals grow within several meters of each other, sharing the same environment. Surprisingly, whatever the leaf water potentials were at the end of the dry period, stomatal conductance, photosynthesis and transpiration rates were relatively similar and low across species. This result contradicts the classic view that anisohydric species are able to maintain gas exchange for longer periods of time during drought stress. None of the plants showed the expected structural acclimation response to the increasing drought (reduction of leaf-to-sapwood area ratio), thereby rejecting the functional equilibrium hypothesis for our study system. Instead, three of the six species increased photosynthetic area at the branch level. The observed dissimilar patterns of gas exchange, hydraulics and morphology across species seem to be equally successful given that photosynthesis at the leaf level was maintained at similar rates over the whole dry period. mesoscale meteorological model (FOOT3DK) is coupled with a gas exchange model to simulate surface fluxes of CO and HO under field conditions. The gas exchange model consists of a C3 single leaf photosynthesis sub-model and an extended big leaf (sun/shade) sub-model that divides the canopy into sunlit and shaded fractions. Simulated CO fluxes of the stand-alone version of the gas exchange model correspond well to eddy-covariance measurements at a test site in a rural area in the west of Germany. The coupled FOOT3DK/gas exchange model is validated for the diurnal cycle at singular grid points, and delivers realistic fluxes with respect to their order of magnitude and to the general daily course. Compared to the Jarvis-based big leaf scheme, simulations of latent heat fluxes with a photosynthesis-based scheme for stomatal conductance are more realistic. As expected, flux averages are strongly influenced by the underlying land cover. While the simulated net ecosystem exchange is highly correlated with leaf area index, this correlation is much weaker for the latent heat flux. Photosynthetic CO uptake is associated with transpirational water loss via the stomata, and the resulting opposing surface fluxes of CO and HO are reproduced with the model approach. Over vegetated surfaces it is shown that the coupling of a photosynthesis-based gas exchange model with the land-surface scheme of a mesoscale model results in more realistic simulated latent heat fluxes. Grafting is an easier and faster approach than plant breeding to take advantage of both existing resistant plants, especially wild cultivars, and high-bred cultivars. The aim of this study was to investigate the changes in leaf photosynthetic capacity of a hybrid eggplant Solanum melongena L., cv. Rima (R), widely used in Greece] when grafted on tomato rootstocks known for their resistance to nematodes and diseases. For this purpose, a hybrid egg-plant has been used as a control and has been self-grafted (grafting of a scion on its own roots used as rootstock) RR and also as a scion on two hybrid tomatoes rootstocks, as follows: Primavera (RP) and Heman (RH). Leaf photosynthetic capacity was estimated by measuring the leaf gas exchanges under several light levels at ambient CO2 concentration to approximate the leaf light response curve. The measurements performed control plant Rand self-grafted eggplants RR show that the leaf respiration per unit leaf area is not altered by the scion/rootstock combination. These elements point out a scion controlled respiration, independent of the rootstock. The leaf photosynthetic capacities of the R and RR treatments were not different, while one of the scion/rootstock combinations (RH) showed a significant change with respect to the control treatments. The rootstock therefore seems to be able to modify the scion leaf photosynthetic capacity, but this may not be true for some scion/rootstock combinations (RP in our case). Leaf area index (LAI) is an important ecophysiological variable because leaves are the organs responsible for gas exchange between plants and the atmosphere. This variable can be calculated from primary values of leaf area assessed by destructive or non-destructive methods, which is relatively easy when crop species are investigated, but is not the case when the focus is on natural wood plants communities. In this paper, we analyze the seasonality of LAI estimated by three different methods in the Amazonia-savannah transitional forest, located 50 km north-east of Sinop city, Mato Grosso, Brazil. In the first method, we combine Monsi and Saekis' original method [Monsi M, Saeki T () Jpn J Bot 14:22-52], which measures LAI using the Beer-Lambert extinction law, and the proposition of Goudriaan [Goudriaan J () Agric For Meteorol 43:155-169] to estimate the extinction coefficient from solar height. The second method differed from the first only in the way in which the daily fraction of intercepted photosynthetic active radiation (FPAR) was calculated, as proposed by Charles-Edwards and Lawn (Charles-Edwards DA, Lawn RJ () Plant Cell Environ 7:247-251]. In the third method, we used a remote sensing technique [MOD15_BU-collection 4, produced and distributed by EROS Data Center Distributed Active Archive Center (EDC DAAC)]. We found that the first and the second methods revealed the expected LAI dynamics, which increased during the dry-wet transition and wet season, and decreased during the wet-dry transition and dry season. From 20 randomly distributed sets in a 1.0 ha area, only 3 showed significant differences in LAI estimated from the first two methods; conversely, LAI was overestimated by the third method. urately representing complex land-surface processes balancing complexity and realism remains one challenge that the weather modelling community is facing nowadays. In this study, a photosynthesis-based Gas-exchange Evapotranspiration Model (GEM) is integrated into the Noah land-surface model replacing the traditional Jarvis scheme for estimating the canopy resistance and transpiration. Using 18-month simulations from the High Resolution Land Data Assimilation System (HRLDAS), the impact of the photosynthesis-based approach on the simulated canopy resistance, surface heat fluxes, soil moisture, and soil temperature over different vegetation types is evaluated using data from the Atmospheric Radiation Measurement (ARM) site, Oklahoma Mesonet, 2002 International HO Project (IHOP_2002), and three Ameriflux sites. Incorporation of GEM into Noah improves the surface energy fluxes as well as the associated diurnal cycle of soil moisture and soil temperature during both wet and dry periods. An analysis of midday, average canopy resistance shows similar day-to-day trends in the model fields as seen in observed patterns. Bias and standard deviation analyses for soil temperature and surface fluxes show that GEM responds somewhat better than the Jarvis scheme, mainly because the Jarvis approach relies on a parametrised minimum canopy resistance and meteorological variables such as air temperature and incident radiation. The analyses suggest that adding a photosynthesis-based transpiration scheme such as GEM improves the ability of the land-data assimilation system to simulate evaporation and transpiration under a range of soil and vegetation conditions. Sky conditions play an important role in the Earth's climate system and CO uptake by plants. We used eddy covariance and meteorological data, including global and diffuse photosynthetic photon flux density (PPFD), recorded over the 2008 and 2009 growing season at two Sitka spruce [ Picea sitchensis ( Bong.) Carr.] forest sites in northern Britain, in order to establish relationships between physiological properties under diverse sky conditions, i.e. (1) sunny, (2) cloudy, and (3) overcast, and several canopy activity-related properties. These properties are: (1) response to PPFD, (2) photosynthetic light use efficiency, and (3) canopy stomatal conductance. We found that Sitka spruce forests utilise PPFD in a more efficient way when solar radiation is dominated by diffuse radiation. Furthermore, our results show that diffuse radiation enhances canopy stomatal conductance, an effect which may be the result of both blue light enrichment within the canopy and the reduction in vapour pressure deficit during cloudy and overcast weather. Meconopsis horridula var. racemesa Prain. is a famous alpine flower and medicinal plant native to high elevations in the Himalayas, but cultivating it at lower altitudes presents great challenges. The photosynthetic gas exchange and chlorophyll fluorescence of M. horridula were investigated at three temperatures to evaluate its photosynthetic performance and the relative importance of biochemical limitation, stomatal limitation, and mesophyll limitation to photosynthesis under different temperature regimes. Meconopsis horridula grown at 20°C could obtain the highest photosynthetic rate and photochemical efficiency among the three temperatures, and photosynthetic performance at low temperature was better than at high temperature. Non-photochemical quenching was an important mechanism protecting the photosynthetic apparatus of M. horridula under temperature stress conditions. Although mesophyll conductance was the dominant factor for limiting photosynthesis of M. horridula both at low temperature and high temperature, the photosynthesis at high temperature was also limited by stomatal conductance and biochemical efficiency. Indigenous broadleaf plantations are increasingly developing as a prospective silvicultural management approach for substituting in place of large pure conifer plantations in subtropical China. However, little information is known about the effects of tree species conversion on soil-atmosphere greenhouse gas (GHG) exchanges. Four adjacent monospecific plantations were selected in subtropical China to examine the effects of tree species on soil-atmosphere exchanges of NO, CH and CO. One coniferous plantation was composed of Pinus massoniana (PM), and the three broadleaf plantations were Castanopsis hystrix (CH), Michelia macclurei (MM) and Mytilaria laosensis (ML). We found that mean soil NO and CO emissions in the PM plantation were 4.34 μg N m h and 43.25 mg C m h, respectively, lower than those in the broadleaf plantations (>5.25 μg N m h and >56.38 mg C m h). The PM plantation soil had higher mean CH uptake (39.03 μg C m h) than the broadleaf plantation soils (<32.67 μg C m h). Variations in soil NO emissions among tree species could be primarily explained by the differences in litter C:N ratio and soil total N stock. Differences in soil CH uptake among tree species could be mostly attributed to the differences in mean soil CO flux and water filled pore space (WFPS). Litter C:N ratio could largely account for variations in soil CO emissions among tree species. This study confirms that there is no GHG benefit of converting PM plantation to broadleaf plantations in subtropical China. Therefore, the future strategy of tree species selection for substituting in place of large coniferous plantations in subtropical China needs to consider the potential effects of tree species on soil-atmosphere GHG exchanges. Stomata in the epidermal tissues of leaves are valves through which passes CO2, and as such they influence the global carbon cycle. The two-dimensional pattern and density of stomata in the leaf epidermis are genetically and environmentally regulated to optimize gas exchange. Two putative intercellular signalling factors, EPF1 and EPF2, function as negative regulators of stomatal development in Arabidopsis, possibly by interacting with the receptor-like protein TMM. One or more positive intercellular signalling factors are assumed to be involved in stomatal development, but their identities are unknown. Here we show that a novel secretory peptide, which we designate as stomagen, is a positive intercellular signalling factor that is conserved among vascular plants. Stomagen is a 45-amino--rich peptide that is generated from a 102-amino-acid precursor protein designated as STOMAGEN. Both an in planta analysis and a semi-in-vitro analysis with recombinant and chemically synthesized stomagen peptides showed that stomagen has stomata-inducing activity in a dose-dependent manner. A genetic analysis showed that TMM is epistatic to STOMAGEN (At4g12970), suggesting that stomatal development is finely regulated by competitive binding of positive and negative regulators to the same receptor. Notably, STOMAGEN is expressed in inner tissues (the mesophyll) of immature leaves but not in the epidermal tissues where stomata develop. This study provides evidence of a mesophyll-derived positive regulator of stomatal density. Our findings provide a conceptual advancement in understanding stomatal development: inner photosynthetic tissues optimize their function by regulating stomatal density in the epidermis for efficient uptake of CO2. The duration and amount of water captured on leaves and its functional significance is highly varied. Leaf surface wettability influences water absorption, gas exchange, pathogen infection, nutrient leaching, contamination by pollutants, self-cleaning properties and in freezing environments the probability of extrinsic ice nucleation. To test the impact of environment on the development of leaf wettability, this functional trait was measured in 227 dominant plant species along an extreme altitudinal environment gradient (186–5,268 m) on the wet and dry slopes of the Nepalese Himalayas. Plants from the understorey and open places in woodlands were also compared. Leaf wettability was assessed by droplet contact angle (θ), retention and leaf inclination measurement. With increasing altitude leaf wettability decreased significantly parallel to the observed atmospheric temperature decrease (0.5 K/100 m). Leaves from non-freezing tropical and subtropical origins were highly wettable (θ < 90°). Temperate leaves were non-wettable (110° < θ < 130°). Subalpine and alpine leaves were highly non-wettable (130° < θ < 150°) and adaxial pubescence occurred more frequently. Leaves taken from the understorey were more wettable but had a better droplet run off than leaves sampled in open places. In the semi-arid northern slopes (temperate to alpine) of the Himalayas leaf wettability was decreased in comparison to the southern humid side. The majority of the leaves had a low droplet retention <20°; higher values were linked to high non-wettability (θ > 130°) which was more often observed at high altitude. Good droplet run off at ±10° inclination was found in highly wettable leaves (θ < 90°) of tropical and subtropical origin and on leaves from the forest understorey. Structural properties for low wettability are developed in cold and dry environments and open sites with frequent dew formation as it appears to be an important functional trait to prevent a number of the negative effects adhering surface water may have. Barnacle fouling has shown to impede gas exchange ability of mangroves. Fouled mangrove plants may therefore obtain less carbon dioxide and water for photosynthesis, resulting in reduced food and chloroplasts production, but such hypothesis remains untested. The objective of the present study compared the stomata density (essential for obtaining carbon dioxide and water molecules) and leaf chlorophyll concentration (essential for photosynthesis) of fouled and non-fouled (control) of seedlings, juveniles and adults of the mangroves Kandelia obovata, in Hong Kong and Taiwan. The seedlings and juveniles of the dominant mangrove plant species, Kandelia obovata, in Hong Kong and Taiwan had a higher density of stomata but a lower chlorophyll concentration in the leaves, when the trunks and twigs were fouled by the barnacle Fistulobalanus albicostatus. Fouled K. obovata appears to develop more stomata in the leaves to compensate the blocking effect of the lenticel from barnacle fouling. As fouling impacts the gaseous exchange ability of mangroves, fouled plants could obtain less carbon dioxide and water for photosynthesis, resulting in reduced food and chloroplasts production. Fouled adult plants, however, had variable responses in leaf chlorophyll concentrations among the study sites, suggesting adults were more tolerant of barnacle fouling. The present study reveals seedling and juvenile mangrove plants are very susceptible to barnacle fouling, which impedes the gaseous exchange mechanism and food production, which can subsequently result in reduced growth, fitness and survival. Leaf water 18O enrichment (Δo) influences the isotopic composition of both gas exchange and organic matter, with Δo values responding to changes in atmospheric parameters. In order to examine possible influences of plant parameters on Δo dynamics, we measured oxygen isotope ratios (δ18O) of leaf and stem water on plant species representing different life forms in Amazonia forest and pasture ecosystems. We conducted two field experiments: one in March (wet season) and another in September (dry season) 2004. In each experiment, leaf and stem samples were collected at 2-h intervals at night and hourly during the day for 50 h from eight species including upper-canopy forest trees, upper-canopy forest lianas, and lower-canopy forest trees, a C4 pasture grass and a C3 pasture shrub. Significant life form-related differences were detected in 18O leaf water values. Initial modeling efforts to explain these observations over-predicted nighttime Δo values by as much as 10‰. Across all species, errors associated with measured values of the δ18O of atmospheric water vapor (δv) appeared to be largely responsible for the over-predictions of nighttime Δo observations. We could not eliminate collection or storage of water vapor samples as a possible error and therefore developed an alternative, plant-based method for estimating the daily average δv value in the absence of direct (reliable) measurements. This approach differs from the common assumption that isotopic equilibrium exists between water vapor and precipitation water, by including transpiration-based contributions from local vegetation through 18O measurements of bulk leaf water. Inclusion of both modified δv and non-steady state features resulted in model predictions that more reliably predicted both the magnitude and temporal patterns observed in the data. The influence of life form-specific patterns of Δo was incorporated through changes in the effective path length, an important but little known parameter associated with the Péclet effect. The banana ( Musa sp.) plant is one of the largest monocotyledoneous terrestrial herbaceous plants in the world. The measurement of transpiration (Tr) for a whole banana plant is always difficult to perform due to its size. However, the sap flow (SF) of the plant has been successfully measured by using the thermal dissipation probe (TDP) or ‘Granier’ method in the corm of the banana plant (Lu et al. J Exp Bot 53:1771–1779, ). The present study aimed to validate their method using a sizable number of banana plants in a greenhouse in Israel. The SF data was compared to the gravimetric measurement of transpiration. The lag time of SF behind Tr was also analyzed. Results showed that the daily SF agreed with the daily Tr when the effective radius for sap flow in the corm was taken as 0.63 R, where R is the radius of the corm. The SF lagged 45 min behind the Tr from 0630 to 1040 hours. Whereas the Tr was not statistically ( P>0.05) different from the SF between 1330 and 1700 hours. The reduction in water capacity of banana plant due to SF lag was about 10.5% of the daily Tr, and it recovered gradually in the afternoon. American elm ( Ulmus americana) seedlings were either non-inoculated or inoculated with Hebeloma crustuliniforme, Laccaria bicolor and a mixture of the two fungi to study the effects of ectomycorrhizal associations on seedling responses to soil compaction and salinity. The seedlings were grown in the greenhouse in pots containing non-compacted (0.4 g cm−3 bulk density) and compacted (0.6 g cm−3 bulk density) soil and subjected to 60 mM NaCl or 0 mM NaCl (control) treatments for 3 weeks. All three fungal inocula had similar effects on the responses of elm seedlings to soil compaction and salt treatment. In non-compacted soil, ectomycorrhizal fungi reduced plant dry weights, root hydraulic conductance, but did not affect leaf hydraulic conductance and net photosynthesis. When treated with 60 mM NaCl, ectomycorrhizal seedlings had several-fold lower leaf concentrations of Na+ compared with the non-inoculated plants. Soil compaction reduced Na+ leaf concentrations in non-ectomycorrhizal plants and decreased dry weights, gas exchange and root hydraulic conductance. However, in ectomycorrhizal plants, soil compaction had little effect on the leaf Na+ concentrations and on other measured growth and physiological parameters. A range of traits, including metabolic costs, biomass allocation and seed reserves, may contribute to interspecific variation in the shade tolerance of tree seedlings. In addition, shade tolerance may be affected by differential responses of species to soil resource availability at low light. We used a custom-built whole-plant gas-exchange chamber to quantify instantaneous whole-plant light compensation point (WPLCP) and to parameterize whole-plant daily C gain models for seedlings of eight temperate deciduous tree species. We examined the relationship of WPLCP to growth, biomass allocation and gas-exchange under high and low light and nutrient availabilities and compared it to WPCLP of naturally recruited saplings. For species showing a response, both increased light and nutrient availability resulted in increased WPLCP. However, species’ responses to resource availability did not correspond closely with shade tolerance as has generally been predicted. Variation in WPLCP within species was best predicted by whole-plant dark respiration rates, leaf-level light compensation point and leaf mass per area. Among species, seed size was a strong negative correlate of WPLCP, explaining 66% of the variation. Species with the lowest WPLCP maintained lower growth rates across treatments but greater biomass in the low-light treatment compared with more light-demanding species. These data suggest that a number of traits, in particular metabolic costs and seed size, contribute to WPLCP. However, gas-exchange-based WPLCP was 1.5–3.5 times lower than corresponding growth-based field estimates of WPLCP, suggesting that other factors such as biotic interactions or ontogenetic shifts in whole-plant light requirements may substantially increase species’ WPLCP under natural conditions. Dissolved O2 is an important aquatic ecosystem health indicator. Metabolic and gas exchange ( G) rates, which control O2 concentration, are affected by nutrient loading and other environmental factors. Traditionally, aquatic metabolism has been reported as primary production:community respiration ( P: R) ratios using diel measurements and interpretations of dissolved O2 and/or CO2 concentrations, and recently using stable isotopes (δ18O, Δ17O) and steady state assumptions. Aquatic ecosystems, such as rivers and ponds, are not at steady state and exhibit diel changes, so steady state approaches are often inappropriate. A dynamic O2 stable isotope model (photosynthesis–respiration–gas exchange; PoRGy) is presented here, requiring a minimum of parameters to quantify daily averaged P, R, and G rates under transient field conditions. Unlike steady state approaches, PoRGy can address scenarios with 100% O2 saturation but with δ18O-O2 values that are not at air equilibrium. PoRGy successfully accounts for isotopic G when applied to an oxygen isotope equilibration laboratory experiment. PoRGy model results closely matched the diel O2 and δ18O-O2 data from three field sites with different P: R: G ratios and various P, R and G rates. PoRGy provides a new research tool to assess ecosystem health and to pose environmental impact-driven questions. Females and males of sexually dimorphic species have distinct resource demands due to differential allocation to reproduction. Sexual allocation theory predicts that functional traits will diverge between sexes to support these demands. However, such dimorphism may be masked by the impact of current reproduction on source-sink interactions between vegetative and reproductive organs. We ask whether natural selection has led to genetic dimorphism in homologous physiological traits between sexes of the dioecious willow shrub, Salix glauca. In a common garden experiment we compared physiological responses to drought stress by male and female ramets in the absence of confounding demands from reproductive structures. Ramets experienced similar pre-dawn leaf water status (Ψl) as parental genets in flower within the natural population, indicating that experimental dry-down mirrored environmental conditions in nature. Male and female ramets achieved similar instantaneous water use efficiency, based on the ratio of carbon gain to water loss, under wet and dry conditions. However, female ramets experienced greater water stress (i.e., more negative Ψl) than males under dry conditions. Lower Ψl for female ramets may partly reflect the maintenance of conductance under drought; males, in contrast, maintain Ψl under drought by reducing conductance. Differences between sexes in terms of conductance and leaf water status of the vegetative ramets were absent in a concomitant comparison of parental flowering plants. Our results show (1) genetic divergence in physiology between sexes of S. glauca occurs in the absence of gender-specific reproductive sinks, (2) males are the more physiologically plastic sex with respect to water use, and (3) paradoxically, divergence in water relations between sexes is not detectable at sexual maturity under natural conditions. Irrigation effects were investigated on an 8-year-old olive ( Olea europaea L., cv. Cobrançosa) commercial orchard located in northeast Portugal. Trees were subjected to a rainfed control (T0) and three treatments (T1, T2, T3) that received a seasonal water amount equivalent to 30%, 60% and 100% of the estimated local evaporative demand by a drip irrigation system. Irrigation increases the photosynthetic activity of olive trees, in association with increases in water status, and reduces the midday and afternoon depression in gas exchange. The closely association between photosynthetic rate ( A) and stomatal conductance ( g s) revealed that the decline in net photosynthesis over the course of the day was largely a consequence of stomatal limitation. However, the ratio of intercellular to atmospheric CO2 concentration increased markedly from morning to midday in non-irrigated plants, in spite of lower g s, suggesting that non-stomatal limitations of photosynthesis also occur when environmental conditions become more stressful. The occurrence of perturbations at chloroplastic level in rainfed plants was demonstrated by a lower maximum photochemical efficiency of photosystem II during the afternoon. Chlorophyll fluorescence measurements also revealed the occurrence of a dynamic photoinhibition in irrigated trees, mainly in T2 and T3, which seemed to be effective in protecting the photosynthetic apparatus from photodamage. Irrigation enhances antioxidant protection and decreases the oxidative damage at leaf level. Leaves grown under rainfed conditions revealed symptoms of oxidative stress, like the reduction (14%) in chlorophyll concentration and the increased levels (57%) of lipid peroxidation. We also found that the scavenging function of superoxide dismutase was impaired in rainfed plants. In contrast, the low thiobarbituric acid reactive substances concentration in T3 indicates that irrigation enhances the repairing mechanisms and decreases the oxidative damage by lipid peroxidation. Accordingly, leaves in T3 treatment had high levels of –SH compounds and the highest antioxidant potential. Meanwhile, the finding that guaiacol peroxidase activity increased in rainfed plants, associated with the appearance of oxidative damage, suggests that this enzyme has no major antioxidative function in olive. During two intensive field campaigns in summer and autumn 2004 nitrogen (N2O, NO/NO2) and carbon (CO2, CH4) trace gas exchange between soil and the atmosphere was measured in a sessile oak ( Quercus petraea (Matt.) Liebl.) forest in Hungary. The climate can be described as continental temperate. Fluxes were measured with a fully automatic measuring system allowing for high temporal resolution. Mean N2O emission rates were 1.5 µg N m−2 h−1 in summer and 3.4 µg N m−2 h−1 in autumn, respectively. Also mean NO emission rates were higher in autumn (8.4 µg N m−2 h−1) as compared to summer (6.0 µg N m−2 h−1). However, as NO2 deposition rates continuously exceeded NO emission rates (−9.7 µg N m−2 h−1 in summer and −18.3 µg N m−2 h−1 in autumn), the forest soil always acted as a net NO x sink. The mean value of CO2 fluxes showed only little seasonal differences between summer (81.1 mg C m−2 h−1) and autumn (74.2 mg C m−2 h−1) measurements, likewise CH4uptake (summer: −52.6 µg C m−2 h−1; autumn: −56.5 µg C m−2 h−1). In addition, the microbial soil processes net/gross N mineralization, net/gross nitrification and heterotrophic soil respiration as well as inorganic soil nitrogen concentrations and N2O/CH4 soil air concentrations in different soil depths were determined. The respiratory quotient (ΔCO2 resp ΔO) for the uppermost mineral soil, which is needed for the calculation of gross nitrification via the Barometric Process Separation (BaPS) technique, was 0.8978 ± 0.008. The mean value of gross nitrification rates showed only little seasonal differences between summer (0.99 µg N kg−1 SDW d−1) and autumn measurements (0.89 µg N kg−1 SDW d−1). Gross rates of N mineralization were highest in the organic layer (20.1–137.9 µg N kg−1 SDW d−1) and significantly lower in the uppermost mineral layer (1.3–2.9 µg N kg−1 SDW d−1). Only for the organic layer seasonality in gross N mineralization rates could be demonstrated, with highest mean values in autumn, most likely caused by fresh litter decomposition. Gross mineralization rates of the organic layer were positively correlated with N2O emissions and negatively correlated with CH4 uptake, whereas soil CO2 emissions were positively correlated with heterotrophic respiration in the uppermost mineral soil layer. The most important abiotic factor influencing C and N trace gas fluxes was soil moisture, while the influence of soil temperature on trace gas exchange rates was high only in autumn. Lotus corniculatus L. and Lotus glaber Mill. are warm-season legume species adapted to many kinds of environmental stress, including flooding conditions, whereas other popular forage legumes, like alfalfa or white clover, cannot thrive. This study evaluates the relationship between root aerenchyma, water relations and leaf gas exchange and the differential tolerance to soil flooding of L. corniculatus and L. glaber. Adult plants of these species, established independently in grasslands mesocosms, were subjected to 40 days of early spring flooding at a water depth of 6 cm. Both species presented constitutive aerenchyma tissue in the roots. Under flooding conditions, this parameter was 26.2% in L. glaber and 15.3% in L. corniculatus. In addition, flooded plants of L. glaber presented a leaf biomass 47.5% higher above water while L. corniculatus showed a leaf biomass 59.6% lower in the same layer, in comparison to control plants. Flooded plants of L. glaber maintained the stomatal conductance ( g s) and transpiration rate ( E) for 25 days, although these parameters reduce slightly to 40–60% in comparison to controls after 40 days of flooding. In this species, a reduction in photosynthesis ( A) in flooding conditions was detected only on the last day of measurement. In L. corniculatus, the same parameters ( g s, E and A) were affected by flooding since day 18 of treatment, and values reached 25–40% in comparison to control plants after 40 days of flooding. Flooding did not affect above-ground biomass in L. glaber; while in L. corniculatus, above-ground biomass was 35% lower than in control plants. Our results confirmed that L. glaber is more able to cope with flooding stress than L. corniculatus, even in the presence of natural competitors. On the whole, this experiment provides information that can aid in the identification of anatomical and physiological parameters associated with flood-tolerance in this forage legume species, with economic potential for the agricultural areas subject to periodic flooding. The carbon isotope signature (δ13C) of foliar cellulose from sunlit tops of trees typically becomes enriched as trees of the same species in similar environments grow taller, indicative of size-related changes in leaf gas exchange. However, direct measurements of gas exchange in common environmental conditions do not always reveal size-related differences, even when there is a distinct size-related trend in δ13C of the very foliage used for the gas exchange measurements. Since δ13C of foliage predominately reflects gas exchange during spring when carbon is incorporated into leaf cellulose, this implies that gas exchange differences in different-sized trees are most likely to occur in favorable environmental conditions during spring. If gas exchange differs with tree size during wet but not dry conditions, then this further implies that environmental sensitivity of leaf gas exchange varies as a function of tree size. These implications are consistent with theoretical relationships among height, hydraulic conductance and gas exchange. We investigated the environmental sensitivity of gas exchange in different-sized Douglas-fir (Pseudotsuga menziesii) via a detailed process model that specifically incorporates size-related hydraulic conductance [soil–plant–atmosphere (SPA)], and empirical measurements from both wet and dry periods. SPA predicted, and the empirical measurements verified, that differences in gas exchange associated with tree size are greatest in wet and mild environmental conditions and minimal during drought. The results support the hypothesis that annual net carbon assimilation and transpiration of trees are limited by hydraulic capacity as tree size increases, even though at particular points in time there may be no difference in gas exchange between different-sized trees. Maximum net ecosystem exchange occurs in spring in Pacific Northwest forests; therefore, the presence of hydraulic limitations during this period may play a large role in carbon uptake differences with stand-age.
