We hypothesized that lactic acid bacteria (LAB) present in sediments at aquaculture sites can be employed as an indicator for the early prediction of deterioration, as the concentration of organic acids controls hydrogen sulfide production via sulfate-reducing bacteria. We selected two aquaculture sites with different acid volatile sulfide (AVS-S) values, St. OJ (average AVS-S = 0.24 mg S/g dry mud) and St. UM (average AVS-S = 1.16 mg S/g dry mud), which were less and critically deteriorated, respectively, and examined our hypothesis by performing a 3-year-long survey in Tanabe Bay, Wakayama Prefecture, Japan. In St. UM, the bacterial community showed positive correlations with AVS-S values and water contents. With AVS-S accumulation at the site, the abundances of LAB decreased below the detection limit, suggesting that LAB viable counts may be unsuitable for predicting early deterioration at sites with severe AVS-S accumulation. In St. OJ, the LAB viable counts, organic acid content, and AVS-S values increased after the beginning of sea bream aquaculture, and the bacterial community showed high correlations with the LAB counts, succinic and total organic acid concentrations, and the abundance of the class Bacilli. Understanding the dynamic change in abundance of both fecal and opportunistic waterborne pathogens in urban surface water under different abiotic and biotic factors helps the prediction of microbiological water quality and protection of public health during recreational activities, such as swimming. However, a comprehensive understanding of the interaction among various factors on pathogen behavior in surface water is missing. In this study, the effect of salinity, light, and temperature and the presence of indigenous microbiota, on the decay/persistence of Escherichia coli and Pseudomonas aeruginosa in Rhine River water were tested during 7 days of incubation with varying salinity (0.4, 5.4, 9.4, and 15.4 ppt), with light under a light/dark regime (light/dark) and without light (dark), temperature (3, 12, and 20 °C), and presence/absence of indigenous microbiota. The results demonstrated that light, indigenous microbiota, and temperature significantly impacted the decay of E. coli. Moreover, a significant (p<0.01) four-factor interactive impact of these four environmental conditions on E. coli decay was observed. However, for P. aeruginosa, temperature and indigenous microbiota were two determinate factors on the decay or growth. A significant three-factor interactive impact between indigenous microbiota, temperature, and salinity (p<0.01); indigenous microbiota, light, and temperature (p<0.01); and light, temperature, and salinity (p<0.05) on the decay of P. aeruginosa was found. Due to these interactive effects, caution should be taken when predicting decay/persistence of E. coli and P. aeruginosa in surface water based on a single environmental condition. In addition, the different response of E. coli and P. aeruginosa to the environmental conditions highlights that E. coli monitoring alone underestimates health risks of surface water by non-fecal opportunistic pathogens, such as P. aeruginosa. When combined with conducive atmospheric conditions, air pollution caused by fossil fuel consumption associated with transportation, industry and electricity production for households, create and sustain continuous pollution over the megalopolis of Tehran, Iran. In conjunction with daily meteorological forecasts, remote sensing can be used to identify and predict days with hazardous levels of air pollution, providing an opportunity for air quality alert systems to be triggered and warnings circulated to reduce health risks to citizens of Tehran province. Combining remotely sensed ozone column density (OCD) data from the Sentinel-5 TROPOMI sensor with NASA Giovanni data concerning meteorological parameters (temperature (T), wind speed (WS) and specific humidity (SH)), geographical parameters and population data, this study considers the drivers and effects of ozone pollution on the urban climate and vegetation condition (normalized difference vegetation index (NDVI)) of 16 counties in Tehran province, Iran during 12 months (i.e., January 2021 to December 2021). Future monthly forecasts of the OCD, climatic and terrestrial factors in 2022 are also presented. Google Earth Engine and the NASA Giovanni platforms were employed for the processing and analysis of data using an interpolation technique. Additionally, a Box–Jenkins ARIMA and Exponential Smoothing (ETS) models were compared and tailored to generate monthly forecasts of OCD, T, WS, SH and NDVI. The highest and lowest OCD was obtained in June and December 2021, with a concentration of 0.14277 mol/m2 and 0.12383 mol/m2, respectively. However, the annual average OCD was higher in the cities of Shahriar and Pakdasht in March, with values of 0.13237 mol/m2 and 0.13244 mol/m2, respectively. The lowest OCD recorded was 0.13105 mol/m2, in Shemiranat city, in the north of Tehran. The results indicate a positive correlation between OCD and NDVI, and a negative correlation between OCD, SH, WS and T. A strong seasonal trend in OCD was identified for all cities, but across the entire province, altitude and population size were the most significant explanatory variables for spatial variations in OCD. This research demonstrates that an effective OCD monitoring and forecasting model may be generated from remote sensing and meteorological variables. The implementation and utilization of these models are of paramount importance as they offer vital information to authorities for continuous air quality monitoring and strategic planning, particularly for days with hazardous air pollution. The traditional monitoring method of water resources environment mainly relies on manual sampling and laboratory analysis, which is limited in time and space and can not realize the comprehensive monitoring of water resources environment. With the continuous development of computer technology and network communication technology, real-time data monitoring method of water resources environment based on computer remote data acquisition and image analysis has gradually become a new solution. This paper aims to propose a real-time data monitoring method of water resources environment based on computer remote data acquisition and image analysis, in order to achieve accurate and efficient monitoring of water resources environment, timely acquisition and analysis of key data of water resources environment, and provide scientific basis for relevant decisions. In this paper, the key parameters of water quality, water quantity and water ecology are collected by using computer remote data acquisition and image analysis. Using computer image analysis technology, the collected image data are processed and analyzed, and the related characteristics and indicators of water resources environment are extracted. The results show that the real-time data monitoring method of water resources environment based on computer remote data acquisition and image analysis can monitor the data changes of water resources environment accurately and efficiently. Computer vision (CV) technologies have been applied extensively in turbid water assessment. However, the accuracies of CV to detect turbidity is limited by several factors such as inferior image quality and adoptation of traditional machine learning (MsL). Several past studies have shown that the accuracy of CV to detect turbidity can be enhanced by employing image pre-processing and artificial intelligence (AI). Therefore, this study proposes a combined CV and convolution neural network (CNN) approach for turbid water classification in accordance with national water quality standards. A total of 71 turbid water samples in the range of 0–100 nephelometry turbidity units (NTU) were prepared from a mixture of 1000 NTU stock Formazine solution and distilled water. Digital images of water samples were acquired with the Asus Zenfone Go smartphone and nephelometry principle. Synthetic minority oversampling technique (SMOTE) was employed for image pre-processing, synthesis, and augmentation. Classification models were developed with Keras CNN architecture and TensorFlow framework. Two national water standards referred to were Malaysia National Water Quality Standard Class I (NWQS-C1) and France Système D'évaluation de la Qualité des cours d'EAU bleu aptitude (SEQ-EAU-b). The proposed CV-CNN approach was successfully implemented with 94.34–98.42% accuracy. The accuracy of CNN was slightly higher when trained with color images (98.42%) than with grayscale images (94.34%). This study aimed to investigate the effects of different dosages of adenine on intestinal microorganisms and enzyme activities, laying the experimental groundwork for subsequent exploration of the microbial mechanisms underlying diarrhea with kidney yang deficiency syndrome. Twenty-four mice were assigned to the following four groups: the control (NC) group, low-dosage adenine (NML) group, middle-dosage adenine (NMM) group, and high-dosage adenine (NMH) group. Mice in the NML, NMM, and NMH groups received 25 mg/(kg·d), 50 mg/(kg·d), and 100 mg/(kg·d) of adenine, respectively, 0.4 mL/each, once a day for 14 days. The NC group received 0.4 mL sterile water. Parameters including body weight, rectal temperature, intestinal microorganisms, enzyme activities, and microbial activity were measured. Results indicated that mice in the experimental group displayed signs of a poor mental state, curled up with their backs arched, and felt sleepy and lazy, with sparse fur that was easily shed, and damp bedding. Some mice showed fecal adhesion contamination in the perianal and tail areas. Dosage-dependent effects were observed, with decreased food intake, body weight, rectal temperature, and microbial activity and increased water intake and fecal water content. Enzyme activity analyses revealed significantly higher activities of protease, sucrase, amylase, and cellulase in intestinal contents and lactase, sucrase, amylase, and cellulase in the mucosa of the NMM group compared to those of other groups. Ultimately, the higher adenine dosage was associated with more pronounced symptoms of kidney yang deficiency syndrome, with 50 mg/kg adenine exhibiting the most substantial impact on the number of intestinal microbial colonies and enzyme activities. Dissolved organic matter (DOM) and its composition in aquatic ecosystems is a key indicator of ecosystem function and an important component of the global carbon cycle. Tropical rainforest headwaters play an important role in global carbon cycling. However, there is a large uncertainty on how DOM sources interact during mobilisation and the potential fate of associated carbon and nutrients. Using field techniques to measure dissolved organic carbon (DOC) concentration and composition, changes in DOM source from headwaters to larger downstream rivers were observed. This study shows that the hydrological connectivity, developed during the transition from dry to wet seasons, changes the DOM supply and transport across a tropical river catchment. The observed variability in the DOC-river discharge relationship provides further evidence of the changes in the DOM supply in a small headwater. This novel insight into the seasonal changes of the dynamics of DOM supply to the river helps understanding the mobilization of terrestrial DOM to tropical headwaters and its export from smaller to larger rivers. The study aims to assess variations in spatio-temporal characteristics of water quality parameters from three tropical estuaries, namely Muri-Ganga, Saptamukhi, and Hooghly, in the western portion of the Indian Sundarbans. Reliable retrieval of near-surface concentration of water quality parameters such as Chlorophyll-a, SST & TSM from diverse aquatic ecosystems with broad ranges of tropical requirements has always remained a complex issue. In this study, application of Case 2 Regional Colour Correction (C2RCC) processor has been tested for its accuracy across different bio-optical regimes in both inland and coastal waters. Satellite images for the same period were also collected and analysed using the C2RCC processing sequence to retrieve parameters like the depth of water, surface reflectance, water temperature, inherent optical properties (IOPs), chlorophyll-a, salinity, total suspended matter (TSM), etc., using the SNAP software. In situ sampling from specific locations within these estuaries and water quality analysis were conducted for the period 2017–2019. The OLCI retrieved datasets were compared and corroborated with field survey datasets. It was observed that the highest amount of TSM was recorded at Diamond Harbour during the 2018 pre-monsoon season (301.40 mg/L field-based value and 308.54 mg/L estimated value). Similarly, chlorophyll-a had higher concentrations throughout the monsoon season (3.03 mg m−3, (field survey), and 2.96 mg m−3, (estimated) at Fraserganj and Sagar south points. A very good correlation was observed for all seasons for Chl-a (r = 0.829) and TSM (r = 0.924) between the OLCI data and in situ measurements. Higher correlation and significant 'r' values highlight the importance of having both field-based as well as remotely-sensed information in understanding any dynamic system in a sustained manner. Results also confirm that the water quality model using OLCI Chl-a and TSM products outperforms conventional techniques. The study demonstrates the efficacy of using Sentinel 3 OCLI data for shallow marine and estuarine remote sensing applications, especially for monitoring TSM and Chl-a concentrations. Dissolved oxygen (DO) is a critical factor in maintaining healthy aquatic ecosystems, including aquaculture ponds. Low DO levels can lead to hypoxia conditions, which are detrimental to fish health and productivity. To deal with this issue, we intend for a smart monitoring system that predicts hypoxia conditions due to low DO levels in aquaculture ponds. The proposed system collects water quality data using Internet of things (IoT) devices and segments it into different categories based on water quality parameters, with a particular focus on low DO levels. By detecting hypoxia conditions early, fish farmers can take corrective measures to prevent fish mortality and improve fish health. To achieve this, our proposed system uses a lightweight Spatially Shared Attention Long Short-Term Memory (SSA-LSTM) model that captures both temporal and spatial dependencies of DO content in water, enabling accurate prediction of hypoxia conditions. Our model outperforms traditional LSTM models and other existing state-of-the-art models, achieving 99.8% accuracy. The proposed system provides a reliable and efficient solution to monitor hypoxia conditions in aquaculture systems and help fish farmers make informed decisions for optimal fish health and productivity. Fecal microbiota transplantation (FMT) has been shown to improve gut dysbiosis in dogs; however, it has not completely been understood in police dogs. This study aimed to investigate the effects of FMT on performance and gut microflora in Kunming police dogs. Twenty Wolf Cyan dogs were randomly assigned to receive physiological saline or fecal suspension at low, medium, or high doses through oral gavage for 14 days. Growth performance, police performance, serum biochemical profiling, and gut microflora were determined 2-week post-FMT. Dogs after FMT treatment were also subjected to an hour road transportation and then were evaluated for serum stress indicators. Overall, FMT enhanced the growth performance and alleviated diarrhea rate in Kunming dogs with the greatest effects occurring in the low dose FMT (KML) group. The improvement of FMT on police performance was also determined. These above alterations were accompanied by changed serum biochemical parameters as indicated by elevated total protein and albumin and reduced total cholesterol and glycerol. Furthermore, the serum stress indicators after road transportation in dog post-FMT significantly decreased. Increased bacterial diversity and modified bacterial composition were found in the feces of dogs receiving FMT. The fecal samples from FMT dogs were characterized by higher abundances of the genera Lactobacillus, Prevotella, and Fusobacterium and lower concentrations of Cetobacterium, Allobaculum, Bifidobacterium, and Streptococcus. The present study supports a potential benefit of FMT on police performance in Kunming dogs. The study evaluated microbial and Potentially Toxic Elements—PTEs risks in high Andean river water in Peru using Monte Carlo simulation. A total of 144 water samples were collected from four rivers and evaluated for physicochemical parameters, PTEs and bacterial pathogens. The microbial risk analysis for exposure to pathogens present in the water was based on the probability of occurrence of diseases associated with Escherichia coli, Pseudomonas aeruginosa and enterococci. PTEs risk analysis was performed using a Monte Carlo simulation approach. The results showed that the highest microbial risk due to exposure to water contaminated by E. coli, P. aeruginosa and enterococci was recorded in the Miraflores and Chia rivers. Meanwhile, the analysis of carcinogenic and non-carcinogenic risk by PTEs in adults and children revealed that the Chia river presents a high risk of contamination by PTEs, especially the carcinogenic risk for children. The Monte Carlo simulation indicated a 56.16% and 94.85% probability of exceeding the limit value of 0.0001 for carcinogenic risk in adults and children, respectively. Rapid urbanization, population growth, agricultural practices, and industrial activities have led to widespread groundwater contamination. This study evaluated heavy metal contamination in residential drinking water in Shiraz, Iran (2021). The analysis involved 80 groundwater samples collected across wet and dry seasons. Water quality was comprehensively assessed using several indices, including the heavy metals evaluation index (HEI), heavy metal pollution index (HPI), contamination degree (CD), and metal index (MI). Carcinogenic and non-carcinogenic risk assessments were conducted using deterministic and probabilistic approaches for exposed populations. In the non-carcinogenic risk assessment, the chronic daily intake (CDI), hazard quotient (HQ), and hazard index (HI) are employed. The precision of risk assessment was bolstered through the utilization of Monte Carlo simulation, executed using the R software platform. Based on the results, in both wet and dry seasons, Zinc (Zn) consistently demonstrates the highest mean concentration, followed by Manganese (Mn) and Chromium (Cr). During the wet and dry seasons, 25% and 40% of the regions exhibited high CD, respectively. According to non-carcinogenic risk assessment, Cr presents the highest CDI and HQ in children and adults, followed by Mn, As and HI values, indicating elevated risk for children. The highest carcinogenic risk was for Cr in adults, while the lowest was for Cd in children. The sensitivity analysis found that heavy metal concentration and ingestion rate significantly impact both carcinogenic and non-carcinogenic risks. Heparin (HP) and heparan sulfate (HS) are multifunctional polysaccharides widely used in clinical therapy. Heparinases (Hepases) are enzymes that specifically catalyse HP and HS degradation, and they are valuable tools for studying the structure and function of these polysaccharides and for preparing low molecular weight heparins. In this study, by searching the NCBI database, a novel enzyme named PCHepII was discovered in the genome of the marine bacterium Puteibacter caeruleilacuae. Heterologously expressed PCHepII in Escherichia coli (BL21) has high expression levels and good solubility, active in sodium phosphate buffer (pH 7.0) at 20°C. PCHepII exhibits an enzyme activity of 254 mU/mg towards HP and shows weak degradation capacity for HS. More importantly, PCHepII prefers to catalyse the high-sulfated regions of HP and HS rather than the low-sulfated regions. Although PCHepII functions primarily as an endolytic Hepase, it mainly generates disaccharide products during the degradation of HP substrates over time. Investigations reveal that PCHepII exhibits a preference for catalysing the degradation of small substrates, especially HP tetrasaccharides. The catalytic sites of PCHepII include the residues His199, Tyr254, and His403, which play crucial roles in the catalytic process. While most pathogenic bacteria are efficiently removed from wastewater during biological treatment, some pathogens, notably Arcobacter, may be abundant in the purified water. Using 16S rRNA gene profiling, the composition of microbial communities of municipal wastewater in the city of Moscow was studied before and after biological purification at the Lyubertsy wastewater treatment plant. Fecal contaminants of the genera Acinetobacter, Aeromonas, Arcobacter, Bacteroides, Streptococcus, and Veillonella, which include human pathogens, predominated in the influent wastewater. After treatment, the relative abundance of these bacteria decreased by 50‒100 times. Predominant organisms in the microbiome of the effluent water were bacteria characteristic of activated sludge, including the nitrifiers of the genera Nitrospira and Nitrosomonas, as well as phosphate- and glycogen-accumulating microorganisms. To explore the efficacy and safety of fecal microbiota transplantation (FMT) as a treatment approach for ulcerative colitis (UC), a comprehensive systematic review and meta-analysis of randomized controlled trials was conducted. To collect and evaluate randomized controlled trials of high quality on FMT for UC, we searched a number of databases, including PubMed, Web of Science, Cochrane, Embase, and Medline, for studies published between the establishment of the databases and March 2023. We conducted a meta-analysis of the studies using Review Manager software (version 5.4.1) to determine the differences in rates of remission and adverse reactions between the FMT group and the control group, utilizing the risk ratio (RR) and 95% confidence interval (CI) to combine our findings. A total of 13 randomized controlled trials (RCTs) on the efficacy of FMT in patients with UC were included in the study, in which 580 patients participated, including 293 patients treated with FMT and 287 control subjects. Meta-analysis revealed that clinical remission was significantly better in the FMT group than in the control group [RR = 1.73; 95% CI = (1.41, 2.12); P < 0.00001]; endoscopic remission was significantly better in the FMT group than in the control group [RR = 1.74; 95% CI = (1.24, 2.44); P = 0.001]. Additionally, there were no significant differences in the incidence of adverse reactions between the two groups [RR = 1.00; 95% CI = (0.86, 1.15); P = 0.96]. Fecal microbiota transplantation has shown potential as a therapeutic intervention for inducing clinical remission in ulcerative colitis UC; nevertheless, the attainment of endoscopic remission and the maintenance of long-term remission continue to present challenges. The excessive utilization of antibiotics has led to significant water contamination and posed severe threats to human well-being. Consequently, the pressing imperative to identify antibiotics in the environment arises. In this study, we have successfully synthesized a hollow PCN-222 MOF distinguished by its substantial surface area and abundant functional groups, particularly the porphyrin cores. To augment the electrical conductivity of the hollow PCN-222 (HPCN-222), gold (Au) particles were incorporated within the porphyrin core using a fundamental hydrothermal method. This modification facilitated the effective immobilization of aptamer strands through π-π stacking and electrostatic interactions. As a result, the Au@HPCN-222 composite demonstrated exceptional efficacy as a substrate for immobilizing the aptamer (Apt) onto the GCE surface. By employing differential pulse voltammetry (DPV) we successfully achieved the detection of chloramphenicol (CAP) with a remarkably low limit of detection of 0.0138 ng mL−1 and the peak DPV currents at 0.18 V (vs. Ag/AgCl) were used for calibration. Humans are significantly impacting riverine systems worldwide, prompting us to investigate the effects of water pollution on the gut microbiome of Cyprinus carpio (common carp). Using 16S rRNA gene sequencing, we compared the gut microbiomes of common carp from two sites along river Yamuna with different pollution levels. Water pollution significantly altered the fish gut microbiome structure and microbial composition. Proteobacteria dominated in both sampling sites, while Bacteroidota prevailed in polluted water samples, indicating sewage and fecal contamination. Less polluted samples exhibited Verrucomicrobiae and Planctomycetes, negatively correlated with pollution levels. The polluted site had higher prevalence of potentially pathogenic and heavy metal-resistant bacteria, as well as microbial communities associated with wastewater treatment systems. Functional prediction highlighted the significant role of the gut microbiome in digestion and metabolism, with active enzymes for breaking down various organic substances. Biosynthetic pathways for leucine, valine, and isoleucine were present in both sites, known to be involved fish immunity. The host maintained a stable and diverse bacterial consortium, while microbial diversity became more specialized due to human activities, adapting to anthropogenic stress and selection pressures. The emergence of the plasmid-mediated tigecycline resistance gene tetX family in pig farms has attracted worldwide attention. The use of tetracycline antibiotics in pig farms has a facilitating effect on the prevalence of the tetX family, but the relationship among its presence, expression, and resistance phenotype in resistant bacteria is unknown. In this study, the presence and expression characteristics of tetracycline resistance genes (TRGs) in 89 strains of doxycycline-resistant E. coli (DRE) isolated from pig manure samples from 20 pig farms under low concentrations of doxycycline stress (2 μg/mL) were analyzed. The detection rate of tetO was 96.63%, which is higher than those of other TRGs, such as tetA (94.38%), tetX (76.40%), tetB (73.03%), and tet(X4) (69.66%). At least three TRG types were present in DRE strains, which thus showed extensive resistance to tetracycline antibiotics, and 37% of these strains were resistant to tigecycline. In the presence of a low concentration of doxycycline, tetA played an important role, and the expression and existence ratio of TRGs indicated low expression of TRGs. Furthermore, the doxycycline resistance of DRE was jointly determined by the total absolute abundance of TRGs, and the absolute abundance of tetX and tet(X4) was significantly positively associated with tigecycline resistance in DRE (P < 0.05). Overall, DRE isolated from swine manure is an important reservoir of the tetX family, which suggests that DRE in swine manure has a high risk of tigecycline resistance, poses a potential threat to human health, and should be of public concern. Milkfish (Chanos chanos) belongs to the family Chanidae and it is a potential candidate species for aquaculture with the best biological characteristics. This study investigates the genetic diversity and population structure of C. chanos along the Indian coast using cytochrome b (cyt b) sequences of mitochondrial DNA (mtDNA). A total of 90 samples collected from five different locations across the Indian coast were sequenced for analysis using cyt b. The sequencing of a 1100-bp cyt b mtDNA fragment revealed the presence of 38 haplotypes with a haplotype diversity value of 0.835 and a nucleotide diversity value of 0.00400. The variation within and among populations accounted for about 97.33% and 2.67%, respectively. The fixation index analysis indicated that there is no significant genetic divergence among the populations from different geographical areas. Neighbour-joining tree analysis of the haplotype data showed no distinct patterns of phylogeographic structure. Results from this study indicated that there is a lack of genetic divergence between the populations of C. chanos along the Indian coast. The haplotype network showed star-like geneology which indicated the demographic expansion of the C. chanos population in these locations. The recent demographic expansion of the C. chanos population was also supported by the results of Tajima's D statistics. To describe the spatiotemporal variations characteristics and future trends of urban air quality in China, this study evaluates the spatiotemporal evolution features and linkages between the air quality index (AQI) and six primary pollution indicators, using air quality monitoring data from 2014 to 2022. Seasonal autoregressive integrated moving average (SARIMA) and random forest (RF) models are created to forecast air quality. (1) The study's findings indicate that pollution levels and air quality index values in Chinese cities decline annually, following a "U"-shaped pattern with a monthly variation. The pollutant levels are high in winter and low in spring, and low in summer and rising in the fall (O3 shows the opposite). (2) The spatial distribution of air quality in Chinese cities is low in the southeast and high in the northwest, and low in the coastal areas and higher in the inland areas. The correlation coefficients between AQI and the pollutant concentrations are as follows: fine particulate matter (PM2.5), inhalable particulate matter (PM10), carbon monoxide (CO), nitrogen dioxide (NO2), sulfur dioxide (SO2), and ozone (O3) values are correlated at 0.89, 0.84, 0.54, 0.54, 0.32, and 0.056, respectively. (3) In terms of short-term AQI predictions, the RF model performs better than the SARIMA model. The long-term forecast indicates that the average AQI value in Chinese cities is expected to decrease by 0.32 points in 2032 compared to the 2022 level of 52.95. If a dead body is discovered in water, it nearly always raises the question about the cause of death, often associated with the persistent problem to differentiate between a drowning incident and post-mortem immersion. In numerous cases, a reliable confirmation of death by drowning is often only possible by a combination of diagnoses obtained from autopsy and additional investigations. As to the latter, the use of diatoms has been suggested (and debated) since decades. Based on the consideration that diatoms are present in almost every natural waterbody and are unavoidably incorporated when water is inhaled, their presence in the lung and other tissues can provide evidence of drowning. However, the traditional diatom test methods are still subject of controversial discussion and suspected of erroneous outcome, predominantly through contamination. A promising alternative to minimize the risk of erroneous outcome seems to be disclosed by the recently suggested MD-VF-Auto SEM technique. Especially the establishment of a new diagnostic marker (L/D ratio), which represents the factorial proportion between the diatom concentration in lung tissue and the drowning medium, allows for clearer distinction of drowning and post-mortal immersion and is largely robust to contamination. However, this highly elaborated technique requires specific devices which are frequently unavailable. We therefore developed a modified method of SEM-based diatom testing to enable the use on more routinely available equipment. Process steps such as digestion, filtration, and image acquisition were thoroughly broken down, optimized, and ultimately validated in five confirmed drowning cases. Taking certain limitations into consideration, L/D ratio analysis provided promising results, even in cases of advanced decomposition. Purpose: The objective of this study was to investigate the co-effect of long-term exposure to atmospheric particulate matter PM2.5 and single nucleotide polymorphisms on schizophrenia relapse. Methods: A total of 332 patients with schizophrenia were recruited. Genotyping of eight SNPs for five genes along the neurotrophin signaling pathway was performed by the Sequenom Massarray technology platform. Based on the data from the monitoring stations, the PM2.5 level of each patient's residence was assessed by the inverse distance weighting method using Arc GIS software. Cox regression analysis was used to determine independent risk factors. The relationship between PM2.5 levels and the risk of schizophrenia relapse was evaluated using the restricted cubic spline (RCS) method. Results: In this study, a total of 191 of 332 patients with schizophrenia relapsed with hospitalization. The risk of schizophrenia relapse was 13.62 (95% CI 8.29 to 22.37) in areas with PM2.5 concentrations of 48.43 to 75.35 µg/m3. The risk of schizophrenia relapse was 5.81 (95% CI 3.58–9.42, p < 0.001) and 13.62 (95% CI 8.29–22.37, p < 0.001) in the exposure categories Q3 and Q4, respectively, compared with Q1, and non-linear relationship between cumulative PM2.5 exposure and risk of schizophrenia relapse. A greater association was observed in the YWHAB gene polymorphic locus rs6031849 genotype TG (Hazard ratio 16.62, 95% CI 5.73 to 48.24). Conclusions: PM2.5 levels, YWHAB gene polymorphism locus rs6031849, and gender jointly influenced schizophrenia relapse, with long-term exposure to high levels of PM2.5 having the greatest effect on schizophrenia relapse. Numerous human activities and poor sanitation management cause public health concern, particularly in rural communities without reliable water supply systems and resources for the monitoring of the quality of their water sources. This study assessed the relationship between observed sanitary risks and hydrogen sulphide (H2S) strip test results in the identification of faecal contamination of various water sources used at household level in rural areas of the Vhembe District Municipality. The highest percentage sanitary risk scores ranging from 50 to 100% were recorded for both river and dam water commonly used by the households for multiple purposes, including drinking. All the surface water samples (100%) also tested positive for H2S production, which is linked to the contamination of water sources by bacteria of faecal origin. The overall results showed a significant and positive correlation (r = 0.623, p = 0.003 in the wet season and r = 0.504, p = 0.017 in the dry season) between sanitary risk scores and H2S strip test results. In low resource settings, the use of sanitary inspections combined with the inexpensive and easy-to-use H2S strip tests can be effective as drinking water quality management tools to raise an awareness among community members of the faecal contamination of their water sources. Groundwater, which appears on the surface in the form of springs, is an important ecologically significant component of the aquatic ecosystem, sensitive to changes in environmental conditions. The anthropogenic impact associated with urbanization leads to a change in the characteristics of groundwater, which, in turn, affects the composition of microbial communities in spring waters. Using high-throughput sequencing of the 16S ribosomal RNA gene fragments, we characterized the composition of microbial communities in five natural springs in the city of Moscow in the spring, summer, and winter seasons. The microbial communities of each spring in different seasons were similar to each other and clearly differed from the microbiomes of the other springs. Among the Archaea, which averaged about 20% of the microbial communities, ammonium-oxidizing Crenarchaeota predominated, as well as Nanoarchaeota. Most of the Bacteria belonged to the phyla Proteobacteria, Patescibacteria, Verrucomicrobiota, Chloroflexi, and Bacteroidota. Autotrophic bacteria, including iron-oxidizing bacteria of the family Gallionellaceae and nitrifiers, as well as methanotrophs, accounted for significant proportions in the microbial communities of the springs with a presumably deeper water source. Chemical and molecular analyses did not reveal contamination of spring waters with toxic substances and oil-derived products, as well as the presence of pathogenic microorganisms and indicators of fecal pollution. However, during the spring season, the proportions of halophilic and hydrocarbon-oxidizing bacteria increased in water microbiomes, which may reflect the entry into groundwater after snow thawing of deicing reagents and hydrocarbons, which are successfully biodegraded in the soil. In this study, genomic and plasmid characteristics of Escherichia coli were determined with the aim of deducing how mcr genes may have spread on a colistin withdrawn pig farm. Whole genome hybrid sequencing was applied to six mcr-positive E. coli (MCRPE) strains isolated from pigs, a farmworker and wastewater collected between 2017 and 2019. Among these, mcr-1.1 genes were identified on IncI2 plasmids from a pig and wastewater, and on IncX4 from the human isolate, whereas mcr-3 genes were found on plasmids IncFII and IncHI2 in two porcine strains. The MCRPE isolates exhibited genotypic and phenotypic multidrug resistance (MDR) traits as well as heavy metal and antiseptic resistance genes. The mcr-1.1-IncI2 and IncX4 plasmids carried only colistin resistance genes. Whereas, the mcr-3.5-IncHI2 plasmid presented MDR region, with several mobile genetic elements. Despite the MCRPE strains belonged to different E. coli lineages, mcr-carrying plasmids with high similarities were found in isolates from pigs and wastewater recovered in different years. This study highlighted that several factors, including the resistomic profile of the host bacteria, co-selection via adjunct antibiotic resistance genes, antiseptics, and/or disinfectants, and plasmid-host fitness adaptation may encourage the maintenance of plasmids carrying mcr genes in E. coli. A synthesis of statistical inference and machine learning (ML) tools has been employed to establish a comprehensive insight of a coarse data. Water components' data for 16 central distributing locations of Lahore, the capital of second most populated province of Pakistan, has been analyzed to gauge current water stature of the city. Moreover, a classification of surplus-response variables through tolerance manipulation was incorporated to debrief dimension aspect of the data. By the same token, the influence of supererogatory variables' renouncement through identification of clustering movement of constituents is inquired. The approach of building a spectrum of colluding results through application of comparable methods has been experimented. To test the propriety of each statistical method prior to its execution on a huge data, a faction of ML schemes have been proposed. The supervised learning tools pca, factoran and clusterdata were implemented to establish an elemental character of water at elected locations. A location 'LAH-13' was highlighted for containing an out of normal range Total Dissolved Solids (TDS) concentration in the water. The classification of lower and higher variability parameters carried out by Sample Mean (XBAR) control identified a set of least correlated variables pH, As, Total Coliforms and E. Coli. The analysis provided four locations LAH-06, LAH-10, LAH-13 and LAH-14 for extreme concentration propensity. An execution of factoran demonstrated that specific tolerance of independent variability '0.005' could be employed to reduce dimension of a system without loss of fundamental data information. A higher value of cophenetic coefficient, c = 0.9582 provided the validation for an accurate cluster division of similar characteristics' variables. The current approach of mutually validating ML and SA (statistical analysis) schemes will assist in preparing the groundwork for state of the art analysis (SOTA) analysis. The advantage of our approach can be examined through the fact that the related SOTA will further refine the predictive precision between two comparable methods, unlike the SOTA analysis between two random ML methods. Recent studies revealed that gut microbiota modulates the response to cancer immunotherapy and fecal microbiota transplantation has clinical benefits in melanoma patients during treatment. Understanding how microbiota affects individual responses is crucial for precision oncology. However, it is challenging to identify key microbial taxa with limited data as statistical and machine learning models often lose their generalizability. In this study, DeepGeni, a deep generalized interpretable autoencoder, is proposed to improve the generalizability and interpretability of microbiome profiles by augmenting data and by introducing interpretable links in the autoencoder. DeepGeni-based machine learning classifier outperforms state-of-the-art classifier in the microbiome-driven prediction of responsiveness of melanoma patients treated with immune checkpoint inhibitors. Moreover, the interpretable links of DeepGeni elucidate the most informative microbiota associated with cancer immunotherapy response. DeepGeni not only improves microbiome-driven prediction of immune checkpoint inhibitor responsiveness but also suggests potential microbial targets for fecal microbiota transplant or probiotics improving the outcome of cancer immunotherapy. Escherichia coli (E. coli) cells are present in fecal materials that can be the main source for disease‐causing agents in water. As a result, E. coli is recommended as a water quality indicator. We have developed an innovative platform to detect E. coli for monitoring water quality on-site by integrating paper-based sample preparation with nucleic acid isothermal amplification. The platform carries out bacterial lysis and DNA enrichment onto a paper pad through ball-based valves for fluid control, with no need of laboratory equipment, followed by loop-mediated isothermal amplification (LAMP) in a battery-operated coffee mug, and colorimetric detection. We have used the platform to detect E. coli in environmental water samples in about 1 h, with a limit of quantitation of 0.2 CFU/mL, and 3 copies per reaction. The platform was confirmed for detecting multiple E. coli strains, and for water samples of different salt concentrations. We validated the functions of the platform by analyzing recreational water samples collected near the Atlantic Ocean that contain different concentrations of salt and bacteria. Contamination of public parks by dogs is a potential source of conflict among park users, causing "tragedy of the commons" problems. Besides the social conflict, feces can pose serious health risks to both dogs and humans. In this study we analyzed the extent and patterns of the distribution of dog feces in the urban parks of the City of Calgary. We collected dog feces from randomly selected locations in the urban parks. The average density of dog feces by the different dog leash policies of the parks and the distribution pattern of the fecal density within the parks were assessed, and the total contamination of the public parks for the entire city was estimated. We found off-leash parks to be significantly more contaminated than other types of parks. We estimated 127.23 g/ha of dog feces are left unpicked in city parks in total every week. Dog feces were found more often and in greater amount in off-leash parks, and near park entrances and parking lots, than in on-leash parks and away from the park entrances. These results suggest that public park visitors, especially those visiting off-leash parks, are likely to be exposed to large amounts of dog feces. Abstract—The results of a bacterioplankton study (total abundance; saprophytic, hydrocarbon oxidizing, and phenol resistant bacteria; total microbial count; and coliform bacteria) in an urbanized tributary of Lake Onego in 2015 are presented. The lower reaches of the Lososinka River flow over the territory of the city of Petrozavodsk (Karelia) and receive 14 untreated storm runoff streams from the municipal storm sewers. Differences in the temporal abundance dynamics of bacteria, which are indicators of oil pollution, the sanitary condition, and saprobity, were found between the reference and urban parts of the river. These variations are not related to the hydrological and thermal regime of the river and reflect the impact of the pollutant discharge from the urban area on the formation of the bacterial community of the river. The study revealed a significant increase in all the studied indicators of the quantitative development of bacterioplankton in the urban part of the river as compared to the reference part. The number of heterotrophic bacteria of various ecological–trophic groups decreases in the lake water by a factor of 8–16 already at a distance of 500 m from the mouth of the river due to dilution. A significant decrease in the numbers of hydrocarbon oxidizing bacteria in the river water and an increase in sanitary-indicative microorganisms are shown to have occurred over four years. Within the city limits of Petrozavodsk, the Lososinka River has been assessed as polluted with petroleum products, as well as organic substances of fecal origin and unfit for recreation based on the level of the indicator bacteria. In contrast to old-growth forests, early-successional stands remain understudied despite potentially harbouring species of conservation interest. With this work, focused on hazel grouse Tetrastes bonasia, a cryptic and indicator species known to select for close-to-natural forests, we evaluated winter densities, home range, microhabitat selection and diet, combining DNA-based mark-recapture and metabarcoding from faecal samples. In total, 216 droppings, collected over 2 years along forest transects in the Italian Alps, were successfully genotyped and 43 individuals were identified. Density estimates were similar to values reported by other studies in the Alps with an average of 4.5 and 2.4 individuals/km2 in the first and second study year, respectively, and mean home ranges estimated at 0.95 km2. According to habitat selection models and eDNA-based diet analysis, hazel grouse selected early-succession secondary-growth forests formed after the abandonment of traditional agropastoral activities. These forests, mostly composed of hazel Corylus avellana, Norway spruce Picea abies and Sorbus spp., provided winter food resources and shelter. The diet analysis also highlighted forest arthropods as a non-negligible source of food. Birds avoided areas subject to intensive browsing by ungulates; small forest roads seasonally closed to traffic had positive influence on hazel grouse (i.e. higher abundance of droppings), while roads open to traffic had no effect. Importantly, despite the high coverage of mature forest habitats of Community Interest (53% of our study area), droppings were more abundant in non-listed early-succession secondary forests with similar plant composition. Our results suggest that forest succession after agropastoral abandonment may be beneficial for some forest birds of conservation interest, while acknowledging its negative effects on the previous grassland biodiversity. Karst aquifers are important resources for drinking water supply and are very vulnerable to contamination. Microbial concentrations at karst springs, in particular, often vary quickly over a short period of time. In this study, the response of microbial water quality and particle-size distribution of two alpine karst springs to rainfall events was investigated to test and validate parameters that can be used as early-warning systems for fecal contamination. At both investigated karst springs, total organic carbon, particle-size distribution (especially small particle fractions), and particle load show a good correlation to the fecal indicator bacteria E. coli and can therefore be used as a real-time indicator of fecal contamination at the investigated springs. In addition to conventional bacterial determination methods, the β-D-glucuronidase activity, which can be measured in near real-time, was used as a novel indicator parameter for fecal contamination. At the event scale, the β-D-glucuronidase (GLUC) activity shows a good correlation to E. coli and can be used as an additional real-time indicator of fecal contamination. For the studied springs, when they show two peaks in turbidity and small particles, these two parameters are suitable for an early warning system because the bacterial contamination occurs during the secondary peak of these parameters. These results highlight the vulnerability of karst aquifers and demonstrate the applicability of advanced measurement techniques in detecting fecal contamination in real-time, which is especially important given the time-consuming nature of conventional bacterial detection methods. Free-living amoebae (FLA) are cosmopolitan microorganisms known to be pathogenic to humans who often have a history of contact with contaminated water. Swimming pools and recreational waters are among the environments where the greatest human exposure to FLA occurs. This study aimed to determine the prevalence of FLA in swimming pools and recreational waters, through a systematic review and meta-analysis that included studies published between 1977 and 2022. A total of 106 studies were included and an overall prevalence of FLA in swimming pools and recreational waters of 44.34% (95% CI = 38.57–50.18) was found. Considering the studies published up to 2010 (1977–2010), between 2010 and 2015, and those published after 2010 (> 2010–2022), the prevalence was 53.09% (95% CI = 43.33–62.73) and 37.07% (95% CI = 28.87–45.66) and 45.40% (95% CI = 35.48–55.51), respectively. The highest prevalence was found in the American continent (63.99%), in Mexico (98.35%), and in indoor hot swimming pools (52.27%). The prevalence varied with the variation of FLA detection methods, morphology (57.21%), PCR (25.78%), and simultaneously morphology and PCR (43.16%). The global prevalence by genera was Vahlkampfia spp. (54.20%), Acanthamoeba spp. (33.47%), Naegleria spp. (30.95%), Hartmannella spp./Vermamoeba spp. (20.73%), Stenamoeba spp. (12.05%), and Vannella spp. (10.75%). There is considerable risk of FLA infection in swimming pools and recreational waters. Recreational water safety needs to be routinely monitored and, in case of risk, locations need to be identified with warning signs and users need to be educated. Swimming pools and artificial recreational water should be properly disinfected. Composite or polymeric insulators have gained significant attention in recent years compared to conventional ceramic disc insulators used in power transmission. The aim of this study is to investigate the performance of ceramic disc and polymeric ceramic 11 kV outdoor products under industrial pollution using laboratory-based pollution performance tests. The insulator performance tests are conducted at various contaminants levels of cement to assess its effectiveness. Statistical analysis of the flashover voltage is investigated through survival functions, hazard functions, and withstand voltages at different failure probability levels. This is an indicator of the level of pollution in the insulator. In this study, normal and Weibull distribution-based statistical models are employed to examine the performance of insulators. The experimental results reported that the ceramic disc insulator and polymeric insulator proved to be more reliable and compatible as insulators in both normal and industrial pollution conditions based on statistical survival functions, hazard functions, and voltage withstand predictions. Based on the statistical analysis of both insulator flashover characteristics and withstand characteristics, the experimental results showed that the normal polymeric insulators and the polluted polymeric insulators have an overall functional reliability performance that is much better than ceramic disc insulators. The plant pathogen pepper mild mottle virus (PMMoV) has recently been proposed as a water quality indicator, it is a RNA virus belonging to the genus Tobamovirus in the family Virgoviridae that causes harm to the pepper crops. After consuming processed food products containing infected peppers, such as hot sauces, PMMoV is excreted in high concentrations in feces; therefore, this is the most common RNA virus, constantly found in the feces of humans. The fecal–oral pathway is emerging as an environmental problem. The presence of high concentrations of pathogens associated with human excreta in environmental waters or water reuse supplies poses a threat to public health. Due to the difficulty in determining the presence of pathogens effectively in water, attempts to monitor microbial water quality often use surrogates or indicator organisms that can be easily detected; therefore, PMMoV is used as a viral surrogate in aquatic environment. This paper describes the incidence and persistence of PMMoV in aquatic environments and in waste treatment plants and its usefulness for quantifying virus reductions by advanced water treatment technologies. In recent research, SARS-CoV-2 was reported to be found in wastewater and utilized for the purpose of monitoring coronavirus illness outbreaks. Since PMMoV is readily identified in the human feces and can also serve as an indicator of human waste, the determined PMMoV concentrations may be utilized to give the normalized report of the SARS-CoV-2 concentration, so that, the amount of human waste found in the wastewater can be taken into consideration. Obesity in dogs is an emerging issue that affects canine health and well-being. Its development is ascribed to several factors, including genetic predisposition and dietary management, and recent evidence suggests that intestinal microbiota may be involved as well. Previous works have shown obesity to be linked to significant changes in gut microbiota composition in humans and mice, but only limited information is available on the role played by canine gut microbiota. The aim of this exploratory study was to investigate whether composition of canine faecal microbiota may be influenced by overweight condition and breed. All the enrolled companion dogs were young adults, intact, healthy, and fed commercial extruded pet food; none had received antibiotics, probiotics or immunosuppressant drugs in the previous six months. Labrador Retriever (LR) and Border Collie (BC) were chosen as reference breeds and Body Condition Score (BCS) on a 9-point scale as reference method for evaluating body fat. The faecal microbial communities of 15 lean (BCS 4–5/9; 7 LRs and 8 BCs) and 14 overweight (BCS > 5/9; 8 LRs and 6 BCs) family dogs were analysed using 16S rRNA gene sequencing. Moreover, for each dog, the daily intake of energy (kcal/d) and dietary macronutrients (g/d) were calculated according to an accurate feeding history collection. Firmicutes and Bacteroidetes resulted the predominant phyla (51.5 ± 10.0% and 33.4 ± 8.5%, respectively) in all dogs. Bioinformatic and statistical analysis revealed that no bacterial taxon differed significantly based on body condition, except for genus Allisonella (p < 0.05); BC gut microbiota was richer (p < 0.05) in bacteria belonging to phyla Actinobacteria (family Coriobacteriaceae in particular) and Firmicutes (Allobaculum and Roseburia genera). No remarkable differences were recorded either for diversity indices (i.e., alpha diversity, p > 0.10) or for divergence within the sample set (i.e., beta diversity, p > 0.05). PERMANOVA tests performed on single factors demonstrated the tendency of dietary protein to influence the recruited dogs' microbiota beta-diversity at amplicon sequence variant level (p = 0.08). In conclusion, the faecal microbiota of dogs involved in this exploratory study showed no major variations based on body condition. Animal diet and health influence fitness, making individual variation in these markers essential for understanding how individuals and populations respond to their environments. Faecal and hair samples provide a record of this information and can be non-invasively collected from animals in the field. However, physiology, diet, and susceptibility to parasitic infections vary within individuals, requiring repeated samples from individuals. We developed a technique using biotelemetry data for individual identification of non-invasive faecal material and hair sampled from female elk (Cervus canadensis). We non-invasively collected individually genotyped faecal and hair samples from resting sites, then compared the accuracy of supervised machine learning models to predict the individual identities of the samples. We found both the tightness of global positioning system point clusters and activity level surrounding the sample allowed us to positively identify samples belonging to specific individuals with 77% accuracy. Our approach can be applied to other populations for which biotelemetry data are available and is potentially adaptable for other species. Furthermore, application of our approach will reduce the need for individual identification of non-invasive samples using genetic analysis, which is costly and prone to low recovery success. Increased access to physiological, dietary, and health information obtainable from individual non-invasive samples will strengthen our understanding of animal responses to their environments. Over the last decade, AMD waters have gained more attention as a potential source of metals due to the emerging need to recover or recycle metals from secondary resources. Metals recovery supports sustainability and the development of a circular economy with benefits for resource conservation and the environment. In this study, five extractants (Acorga M5640, LIX 54, LIX 622, LIX 622 N, and LIX 864) diluted (15% (v/v)) in Shell GTL with 2.5% (v/v) octanol were compared and evaluated for Cu recovery from an extreme AMD sample (5.3 ± 0.3 g/L Cu) collected at the inactive São Domingos Mine in the Iberian Pyrite Belt of Portugal. Of the five extractants, Acorga M5640 showed the best selective efficiency. Further tests showed that 30% (v/v) of this extractant was able to selectively extract ≈ 96.0% of the Cu from the AMD in one extraction step and all of the remaining Cu (to below detection) in three steps. Among the different stripping agents tested, 2 M sulfuric acid was the most efficient, with ≈ 99% of the Cu stripped, and the recyclability of the organic phase was confirmed in five successive cycles of extraction and stripping. Furthermore, contact time tests revealed that the extraction kinetics allows the transfer of ≈ 97% of the Cu in 15 min, and aqueous to organic phase ratios tests demonstrated a maximum loading capacity of ≈ 16 g/L Cu in the organic phase. Raising the concentration of Cu in the stripping solution (2 M sulfuric acid) to ≈ 46 g/L through successive striping steps showed the potential to recover elemental Cu using traditional electrowinning. Finally, a biological approach for Cu recovery from the stripping solution was evaluated by adding the supernatant of a sulfate-reducing bacteria culture to make different molar ratios of biogenic sulfide to copper; ratios over 1.75 resulted in precipitation of more than 95% of the Cu as covellite nanoparticles. Freshwater ecosystems are under multiple threats in modern times such as water extraction for human consumption, industries and agricultural activities, water contamination and habitat destruction for example. At the same time the biodiversity of these ecosystems are often poorly studied, especially in arid countries such as Iran. In this work, we study one of the ecologically important members of Iranian freshwater fauna, freshwater crab species of the genus Potamon. Here, we barcoded the different populations occurring in the country and delimited the species to allow for a better understanding of their distribution and taxonomy. In this study, we evaluated the taxonomical statues of Potamon species in Iran using genetic data. Food safety and foodborne diseases are significant global public health concerns. Meat and poultry carcasses can be contaminated by pathogens like E. coli and salmonella, by contact with animal fecal matter and ingesta during slaughter and processing. Since fecal matter and ingesta can host these pathogens, detection, and excision of contaminated regions on meat surfaces is crucial. Fluorescence imaging has proven its potential for the detection of fecal residue but requires expertise to interpret. In order to be used by meat cutters without special training, automated detection is needed. This study used fluorescence imaging and deep learning algorithms to automatically detect and segment areas of fecal matter in carcass images using EfficientNet-B0 to determine which meat surface images showed fecal contamination and then U-Net to precisely segment the areas of contamination. The EfficientNet-B0 model achieved a 97.32% accuracy (precision 97.66%, recall 97.06%, specificity 97.59%, F-score 97.35%) for discriminating clean and contaminated areas on carcasses. U-Net segmented areas with fecal residue with an intersection over union (IoU) score of 89.34% (precision 92.95%, recall 95.84%, specificity 99.79%, F-score 94.37%, and AUC 99.54%). These results demonstrate that the combination of deep learning and fluorescence imaging techniques can improve food safety assurance by allowing the industry to use CSI-D fluorescence imaging to train employees in trimming carcasses as part of their Hazard Analysis Critical Control Point zero-tolerance plan. The human gut microbiome plays a central role in health and disease. Environmental factors, such as lifestyle and diet, are known to shape the gut microbiome as well as the reservoir of resistance genes that these microbes harbour; the resistome. In this study we assessed whether long-term dietary habits within a single geographical region (the Netherlands) impact the human gut resistome. Faecal samples from Dutch omnivores, pescatarians, vegetarians and vegans were analysed by metagenomic shotgun sequencing (MSS) (n = 149) and resistome capture sequencing approach (ResCap) (n = 64). Among all diet groups, 119 and 145 unique antibiotic resistance genes (ARGs) were detected by MSS or ResCap, respectively. Five or fifteen ARGs were shared between all diet groups, based on MSS and ResCap, respectively. The total number of detected ARGs by MSS or ResCap was not significantly different between the groups. MSS also revealed that vegans have a distinct microbiome composition, compared to other diet groups. Vegans had a lower abundance of Streptococcus thermophilus and Lactococcus lactis compared to pescatarians and a lower abundance of S. thermophilus when compared to omnivores. Oxazepam, one of the most frequently prescribed anxiolytic drugs, is not completely removed from wastewater with conventional treatment processes. It can thus be found at trace levels in environmental water, with human urine constituting the major source of contamination. This study focused on the development and characterization of molecularly imprinted polymers (MIPs) for the selective solid-phase extraction of oxazepam at trace levels from environmental water and human urine samples. Two MIPs were synthesized, and their selectivity in pure organic and aqueous media were assayed. After optimizing the extraction procedure adapted to a large sample volume to reach a high enrichment factor, the most promising MIP was applied to the selective extraction of oxazepam from environmental water. Extraction recoveries of 83 ± 12, 92 ± 4 and 89 ± 10% were obtained using the MIP for tap, mineral and river water, respectively, while a recovery close to 40% was obtained on the corresponding non-imprinted polymer (NIP). Thanks to the high enrichment factors, a limit of quantification (LOQ) of 4.5 ng L−1 was obtained for river water. A selective extraction procedure was also developed for urine samples and gave rise to extraction recoveries close to 95% for the MIP and only 23% for the NIP. Using the MIP, a LOQ of 357 ng L−1 was obtained for oxazepam in urine. The use of the MIP also helped to limit the matrix effects encountered for the quantification of oxazepam in environmental samples and in human urine samples after extraction on an Oasis HLB sorbent. Carbapenems, as the "last line of defense" against Gram-negative bacteria, are increasingly being challenged by drug-resistant bacteria, especially Enterobacteriaceae. In this study, a carbapenem-resistant Gram-negative bacterium, named AH001, was isolated from hospital sewage, and a modified Hodge test confirmed that this bacterium can produce carbapenemase. Further analysis revealed that this bacterium exhibits multidrug resistance against an additional seven antibiotics. Whole-genome sequencing and analysis showed that AH001 could not be classified by existing MLST, and its serotype could not be distinguished among O9, O89 or O168 according to O antigen prediction. Anthropogenic contamination of coastal-marine water is responsible for introducing multidrug-resistant bacteria such as the pNDM-harbouring Escherichia coli into the seafood chain. This study was conducted to understand the survivability of a multidrug-resistant, the New Delhi Metallo-β-lactamase-producing E. coli (AS-EC121) in tropical seawater at room temperature (28–32 °C) compared to E. coli K12 strain. The experimental and control strains were inoculated at 6 log CFU/ml level into seawater. After an initial sharp decline in counts, AS-EC121 and K12 strains showed a gradual loss of viability after week-1 of inoculation. AS-EC121 was undetectable after day-56, while K12 colonies disappeared a week later, from day-63. The conjugation experiment revealed that pNDM was transferable to a recipient E. coli strain in seawater. This study suggests that the multidrug-resistant, pNDM-harbouring E. coli is able to survive in seawater for over 2 months stably maintaining the resistance plasmid. The resistance genotypes do not seem to compromise the survivability of MDR E. coli and the stability of plasmid provides ample opportunities for dissemination of plasmids among co-inhabiting bacteria in the coastal–marine environments. The routine evaluation of water environments is necessary to manage enteric virus-mediated fecal contamination and the possible emergence of novel variants. Here, we detected human rotavirus A (HRVA) circulating in two wastewater treatment plants, two lakes, irrigation water and a wastewater landfill located in Riyadh. VP7-derived surface protein sequences were assessed by phylogenetic analyses and inspection of thermotolerance-mediated secondary structure and seasonal variation. HRVA was most prevalent at An-Nazim wastewater landfill (AN-WWLF; 63.89%). Phylogenetic analyzes revealed the predominance of HRVA G2 lineage for the first time in Saudi Arabia. Moreover, a single HRVA sequence (2B64I-ANLF3/2018) was recovered at 45 °C from AN-WWLF; secondary structure prediction indicated that this sequence was thermotolerant with a high hydrophobicity, an absence of Ramachandran outliers, and a higher content of proline patches on the protein surface. Varied relationships were significantly observed between sampling areas influenced by temperature ranges (p < 0.05). HRVA prevalence was influenced by seasonal variations, favoring moderate temperatures in late autumn and early winter in all locations. However, a significant temperature impact was detected in Wadi-Hanifah Lake (p = 0.01). Our study extends the knowledge of currently circulating HRVA genotypes, and indicates the probable emergence of thermotolerant strains and seasonally mediated HRVA prevalence. Long-term changes, from 1984 to 2010, in the indicators of microbial pollution (total viable count, coliforms, Escherichia coli, enterococci, and Clostridium perfringens) are analysed in the Riga Hydropower Plant Reservoir, an essential source of drinking water for Riga, the capital of Latvia. Counts in microbial indicators fluctuated seasonally and were related to physicochemical parameters (nitrogen compounds, turbidity, temperature, and pH). The changes in microbial pollution were brought about by two major socio-economic developments. Firstly, Latvia's independence from the USSR in 1991 which facilitated a distinct reduction in most microorganism counts due to a sharp decline in industrial and agricultural production. This resulted in a significant drop in point and nonpoint pollution in the river basin. A further development was Latvia joining the European Union in 2004. The corresponding focus on water management, including wastewater treatment, was a major priority of environmental investment and lead to improvements in microbial water quality. Blastocystis sp. is an intestinal protist parasite commonly found in the feces of humans and animals worldwide. Blastocystis exhibits extensive genetic diversity and has been identified in humans and a variety of animals including other mammals and birds. Blastocystis subtypes do not exhibit strict host specificity which raises the possibility of zoonotic transmission through either direct contact or fecal contamination of food or water. However, reports detailing the subtypes and prevalence of Blastocystis in avian species are limited. Therefore, this study investigated the presence of Blastocystis in chickens by molecular characterization of the small subunit rRNA (SSU rRNA) gene. Fecal samples from 130 chickens were collected from local markets in Uberlândia and Belo Horizonte in the state of Minas Gerais, Brazil. To detect and identify subtypes of Blastocystis, a next-generation amplicon sequencing protocol was used. Forty-four of the 130 (33.8%) chickens examined were positive for Blastocystis. Blastocystis subtypes ST6 (23/130; 17.7%), ST7 (43/130; 33.1%), ST10 (1/130; 0.8%), ST14 (5/130; 3.8%), ST25 (1/130; 0.8%), and a novel subtype (ST29) (2/130; 1.5%) were observed. A nanopore sequencing strategy was used to obtain the near full-length SSU rRNA gene nucleotide sequence and validate novel subtype ST29. Mixed infections containing multiple subtypes were common and identified in 63.6% of Blastocystis-positive chickens. All positive samples contained one or both potentially zoonotic subtypes ST6 and ST7. The prevalence of Blastocystis in chickens was high, and molecular characterization mostly identified subtypes previously found in humans. The novel coronavirus disease COVID-19 has infected over 46 million people in 219 countries and territories. Following evidence of viral loadings and infectivity of feces of infected individuals, public health authorities have suggested to take precautions on the transmission of COVID-19 via fecal-associated routes. Recent discussions on fecal transmission of COVID-19 have mainly focused on municipal sewage. Yet, a widely neglected aspect in containing the virus is that a major part of the population in developing regions do not have access to private, clean sanitary facilities. Therefore, we hypothesize that open defecation and the prevalent use of squat toilets are additional risk factors in those communities. Here, we review fecal transmission of COVID-19, the practices of open defecation, and the resultant routes of transmission of fecal pathogens. Also, we highlight the open design of common squat toilets and the potential exposure to fecal droplets and residues. We observed that at least 20 countries reporting more than 10,000 confirmed infections have 5–26% of their population practicing open defecation. We illustrate the potential routes of transmission of COVID-19 and other fecal pathogens via human feces in communities practicing open defecation. Here, poor hand hygiene, contaminated shoes and objects, mechanical vectors, and outdoor human activities can all contribute to fecal transmission.
