As our planet is entering into the "global boiling" era, understanding regional climate change becomes imperative. Effective downscaling methods that provide localized insights are crucial for this target. Traditional approaches, including computationally-demanding regional dynamical models or statistical downscaling frameworks, are often susceptible to the influence of downscaling uncertainty. Here, we address these limitations by introducing a diffusion probabilistic downscaling model (DPDM) into the meteorological field. This model can efficiently transform data from 1° to 0.1° resolution. Compared with deterministic downscaling schemes, it not only has more accurate local details, but also can generate a large number of ensemble members based on probability distribution sampling to evaluate the uncertainty of downscaling. Additionally, we apply the model to generate a 180-year dataset of monthly surface variables in East Asia, offering a more detailed perspective for understanding local scale climate change over the past centuries. In the ALDH2 rs671 variant, a guanine changes to an adenine, resulting in a dramatic decrease in the catalytic activity of the enzyme. Population-based data are contradictory about whether this variant increases the risk of Alzheimer's disease. In East Asian populations, the prevalence of the ALDH2 rs671 variant is 30–50%, making the National Human Brain Bank for Development and Function (the largest brain bank in East Asia) an important resource to explore the link between the ALDH2 rs671 polymorphism and Alzheimer's disease pathology. Here, using 469 postmortem brains, we find that while the ALDH2 rs671 variant is associated with increased plaque deposits and a higher Aβ40/42 ratio, it is not an independent risk factor for Alzheimer's disease. Mechanistically, we show that lower ALDH2 activity leads to 4-HNE accumulation in the brain. The (R)−4-HNE enantiomer adducts to residue Lys53 of C99, favoring Aβ40 generation in the Golgi apparatus. Decreased ALDH2 activity also lowers inflammatory factor secretion, as well as amyloid β phagocytosis and spread in brains of patients with Alzheimer's disease. We thus define the relationship between the ALDH2 rs671 polymorphism and amyloid β pathology, and find that ALDH2 rs671 is a key regulator of Aβ40 or Aβ42 generation. Here, Wang et al. report that the ALDH2 rs671 variant exacerbates amyloid-β pathology in the human brain. Aquaculture has suffered significant financial losses as a result of the infection of zoonotic Aeromonas hydrophila, which has a high level of resistance to classic antibiotics. In this study, we isolated an A. hydrophila strain B3 from diseased soft-shelled turtle (Pelodiscus sinensis), which is one of the most commercially significant freshwater farmed reptiles in East Asia, and found that A. hydrophila was its dominant pathogen. To better understand the inhibition effect and action mechanism of Chinese herbs on A. hydrophila, we conducted Chinese herbs screening and found that Lonicera japonica had a significant antibacterial effect on A. hydrophila B3. Experimental therapeutics of L. japonica on soft-shelled turtle showed that the supplement of 1% L. japonica to diet could significantly upregulate the immunity-related gene expression of soft-shelled turtle and protect soft-shelled turtle against A. hydrophila infection. Histopathological section results validated the protective effect of L. japonica. As the major effective component of L. japonica, chlorogenic acid demonstrated significant inhibitory effect on the growth of A. hydrophila with MIC at 6.4 mg/mL. The in vitro assay suggested that chlorogenic acid could inhibit the hemolysin/protease production and biofilm formation of A. hydrophila and significantly decrease the expression of quorum sensing, biofilm formation, and hemolysin-related genes in A. hydrophila. Our results showed that the Chinese herb L. japonica would be a promising candidate for the treatment of A. hydrophila infections in aquaculture, and it not only improves the immune response of aquatic animals but also inhibits the virulence factor (such as biofilm formation) expression of A. hydrophila. East Asian floods and droughts in summer show a typical dipole pattern with a north-south oscillation centered near 30°N, called the southern drought–northern flood (SDNF) pattern, which has caused significant economic losses and casualties in the past three decades. However, effective explanations and predictions are still challenging, making suitable disaster prevention more difficult. Here, we find that a key predictor of this dipole pattern is the Quasi-Biennial Oscillation (QBO, tropical winds above 10 km). The QBO can modulate precipitation in East Asia, contributing the largest explained variation of this dipole pattern. A QBO-included statistical model can effectively predict summer floods and droughts at least three months in advance and explain at least 75.8% of precipitation variation. More than 30% of the SDNF pattern is attributed to the QBO in July-August 2020 and 2021. This result suggests a good prospect for using the tropical mid- to upper atmosphere in seasonal forecasts for summer. Planted forests are critical to climate change mitigation and constitute a major supplier of timber/non-timber products and other ecosystem services. Globally, approximately 36% of planted forest area is located in East Asia. However, reliable records of the geographic distribution and tree species composition of these planted forests remain very limited. Here, based on extensive in situ and remote sensing data, as well as an ensemble modeling approach, we present the first spatial database of planted forests for East Asia, which consists of maps of the geographic distribution of planted forests and associated dominant tree genera. Of the predicted planted forest areas in East Asia (948,863 km2), China contributed 87%, most of which is located in the lowland tropical/subtropical regions, and Sichuan Basin. With 95% accuracy and an F1 score of 0.77, our spatially-continuous maps of planted forests enable accurate quantification of the role of planted forests in climate change mitigation. Background: Facial-onset sensory and motor neuronopathy (FOSMN) syndrome is a rare clinical syndrome in which the etiopathogenesis and disease-causing genes remain unknown. In addition, clinical and molecular pathological studies have rarely been evaluated in a large case series.Methods: In this study, we present the clinical features and electrodiagnostic findings of the largest cohort of six patients with FOSMN in East Asia to date. Immunofluorescence assessment of TAR DNA-binding protein (TDP)-43 in muscle and skin fibroblasts, detection of GGC trinucleotide repeat expansions in NOTCH2NLC gene, and GGGGCC hexanucleotide repeat expansions in the C9orf72 gene were also performed.Results: All patients exhibited typical symptoms and signs of FOSMN syndrome. Almost all patients showed a delayed or absent blink reflex. Neurogenic damage was found in five patients by electromyography. Two of the five patients with muscle and skin biopsies showed TDP-43-positive inclusions in both the nucleus and cytoplasm of muscular tissue and skin fibroblasts. There were no repeat expansions in the C9orf72 or NOTCH2NLC genes in any of the six patients.Conclusions: To date, this is the largest FOSMN cohort in East Asia. TDP-43-positive cytoplasmic inclusions in muscle and skin fibroblasts may be a pathologic feature of the disease. The patient's dynamic mutation test showed no GGC trinucleotide repeat expansions in the NOTCH2NLC and GGGGCC hexanucleotide repeat expansions in the C9orf72 gene. In the late 1920's, a mega-drought in China resulted in widespread crop failure and famine. Sufficient evidence suggests that this drought belonged to a dry period ranging from approximately 1922 to 1932. To understand the characteristics and the cause of this persistent drought period, we combined various data, including observations, tree ring proxy data, reanalysis data, simulation results of the Fifth Phase of the Coupled Model Intercomparison Project and numerical downscaling simulations. The results show that during 1922–1932, most regions in eastern China suffered from a persistent drought that lasted for six years, and the maximum negative precipitation anomaly reached −1.5 times the standard deviation. Given its spatial coverage, duration, and strength, the 1920s drought was unique for the 20th century. The 1920s drought was primarily caused by internal variability. Strong easterlies in lower latitudes, strong monsoon circulation, and abnormally high geopotential heights at middle and upper levels were responsible for the 1920s drought conditions in eastern China; these drought conditions could be further attributed to the joint impact of the Atlantic Multidecadal Oscillation, Pacific Decadal Oscillation and Indian Ocean Basin Mode. Although alpine ecosystems have been commonly recognized as sensitive to recent climate change, few studies have examined its impact on the long-term productivity of vegetation and soil erosion. Using paleoecological records, these two aspects were examined in the alpine zone of the Taibai Mountains (elevation, 3767 m) in monsoon-dominated East Asia since the middle Holocene. Proxies for the productivity of vegetation and severity of soil erosion from high-resolution alpine lacustrine records show that the productivity was closely related to mean annual temperature and soil erosion, to summer precipitation from the East Asian Summer Monsoon (EASM). Specifically, when the mean annual temperature was low and precipitation was abundant, during 5800–4000 calendar years before the present (cal. yr BP), the alpine ecosystem was characterized by low vegetation productivity and severe soil erosion. However, the productivity increased and soil erosion decreased from 4000 cal. yr BP onwards. These results highlight the role of paleoecological evidence in studying ecosystem services on longer time scales, which is significant in making policies for sustainable development under climate change in regions for which such long-term monitoring data are not available. Due to the formation of the Qiongzhou Strait by climate change and marine transition, Hainan island was isolated from the mainland southern China during the Last Glacial Maximum. Hainan island, located at the southernmost part of China and separated from the Leizhou Peninsula by the Qiongzhou Strait, laid on one of the modern human northward migration routes from Southeast Asia to East Asia. The Hlai language-speaking Li minority, the second largest population after Han Chinese in Hainan island, is the direct descendants of the initial migrants in Hainan island and has unique ethnic properties and derived characteristics; however, the forensic-associated studies on Hainan Li population are still insufficient. Hence, 136 Hainan Li individuals were genotyped in this study using the MPS-based ForenSeq™ DNA Signature Prep Kit (DNA Primer Set A, DPMA) to characterize the forensic genetic polymorphism landscape, and DNA profiles were obtained from 152 different molecular genetic markers (27 autosomal STRs, 24 Y-STRs, 7 X-STRs, and 94 iiSNPs). A total of 419 distinct length variants and 586 repeat sequence sub-variants, with 31 novel alleles (at 17 loci), were identified across the 58 STR loci from the DNA profiles of Hainan Li population. We evaluated the forensic characteristics and efficiencies of DPMA, demonstrating that the STRs and iiSNPs in DPMA were highly polymorphic in Hainan Li population and could be employed in forensic applications. In addition, we set up three datasets, which included the genetic data of (i) iiSNPs (27 populations, 2640 individuals), (ii) Y-STRs (42 populations, 8281 individuals), and (iii) Y haplogroups (123 populations, 4837 individuals) along with the population ancestries and language families, to perform population genetic analyses separately from different perspectives. In conclusion, the phylogenetic analyses indicated that Hainan Li, with a southern East Asia origin and Tai-Kadai language-speaking language, is an isolated population relatively. But the genetic pool of Hainan Li influenced by the limited gene flows from other Tai-Kadai populations and Hainan populations. Furthermore, the establishment of isolated population models will be beneficial to clarify the exquisite population structures and develop specific genetic markers for subpopulations in forensic genetic fields. El Niño profoundly impacts precipitation in high-population regions. This demands an advanced understanding of the changes in El Niño-induced precipitation under the future global warming scenario. However, thus far, consensus is lacking regarding future changes in mid-latitude precipitation influenced by El Niño. Here, by analyzing the Coupled Model Intercomparison Project simulations, we show that future precipitation changes are tightly linked to the response of each type of El Niño to the tropical Pacific mean sea surface temperature (SST) change. A La Niña-like mean SST change intensifies basin-wide El Niño events causing approximately 20% more precipitation over East Asia and North America via enhancing moisture transport. Meanwhile, an El Niño-like mean SST change generates more frequent eastern Pacific El Niño events, enhancing precipitation in North American. Our findings highlight the importance of the mean SST projection in selectively influencing the types of El Niño and their remote impact on precipitation. El Niño-driven precipitation profoundly affects the mid-latitudes, but how this impact changes in the future is uncertain. This study revisits the impacts of different types of El Niño on East Asian summer precipitation (EASP) with observed and re-analyzed data from 1958 to 2015. It focuses on the boreal summer preceding peaks of El Niño when prominent SST anomalies are mainly situated in the tropical Pacific. Distinct responses of EASP are obtained for the three identified types of SST anomalies noted as summer EP (Eastern Pacific) El Niño, summer MP (Mixed-type Pacific) El Niño, and summer CP (Central Pacific) El Niño. In the case of summer EP El Niño, there is a robust south-north rainfall dipole with anomalous dry conditions in North China and wet conditions in Southeast China. Such a rainfall pattern is mainly attributed to a low-pressure anomaly over Northeast Asia. For summer MP El Niño, it presents a tripolar pattern of precipitation anomalies with wetness along the Yangtze-Huaihe River valley and dryness in both South and North China. This is due to the presence of low-pressure anomalies over Northeast China and a westward shift of the Western Pacific Subtropical High. Summer CP El Niño shows a triangle-pattern of precipitation anomalies with wet conditions in both southern and northern China, but dry conditions in the Jianghuai region (north of the Yangtze River in East China), caused by low-pressure anomalies over central-north China. Further study investigates how different types of summer El Niño induce different changes of the Walker circulation and associated large-scale convergence/divergence outside the tropics, and ultimately provoke different responses of atmospheric circulation. For summer EP El Niño, tropical perturbations over the east-central tropical Pacific and over North India can enter into the subtropical jet and make a mid-latitude low pressure belt response at upper levels of the atmosphere. For summer MP El Niño, the perturbation over the east-central tropical Pacific seems ineffective in transmitting atmospheric anomalies to mid-latitudes. As a result, they are mainly confined to the tropics and lead to a general rise of the upper-level geopotential height over the entire tropics. However, the perturbation over Northwest Africa can excite northeastward propagating Rossby waves in mid-high latitudes. For summer CP El Niño, the induced fluctuant wave train within the subtropical jet is likely the competing effect of perturbations over the east-central tropical Pacific, North India and North Africa. Furthermore, for all the three types of summer El Niño, perturbations can trigger a similar low-level meridional wave train along the East Asian coast through the monsoon trough. Objectives: Our aim was to systematically review published evidence on the construct validity, test-retest reliability and responsiveness of generic preference-based measures (PBMs) used in East and South-East Asia.Methods: This systematic review was guided by the COSMIN guideline. A literature search on the MEDLINE, EMBASE, PsycINFO and PubMed databases up to August 2019 was conducted for measurement properties validation papers of the EuroQol-5 Dimensions (EQ-5D), Short Form-6 Dimensions (SF-6D), Health Utilities Index (HUI), Quality of Well-Being (QWB), 15-Dimensional (15D) and Assessment of Quality of Life (AQOL) in East and South-East Asian countries. Included papers were disaggregated into individual studies whose results and quality of design were rated separately. The population-specific measurement properties (construct validity, test-retest reliability and responsiveness) of each PBM were assessed separately using relevant studies. The overall methodological quality of the studies used in each of the assessments was also rated.Results: A total of 79 papers containing 1504 studies were included in this systematic review. The methodological quality was 'very good' or 'adequate' for the majority of the construct validity studies (99%) and responsiveness studies (61%), but for only a small portion of the test-retest reliability studies (23%). EQ-5D was most widely assessed and was found to have 'sufficient' construct validity and responsiveness in many populations, while the SF-6D and EuroQol-Visual Analog Scale (EQ-VAS) exhibited 'inconsistent' construct validity in some populations. Scarce evidence was available on HUI and QWB, but current evidence supported the use of HUI.Conclusions: This systematic review provides a summary of the quality of existing generic PBMs in Asian populations. The current evidence supports the use of EQ-5D as the preferred choice when a generic PBM is needed, and continuous testing of all PBMs in the region. [ABSTRACT FROM AUTHOR]

Convection-permitting regional climate models have been shown to improve precipitation simulation in many aspects, such as the diurnal cycle, precipitation frequency, intensity and extremes in many studies over several geographical regions of the world, but their skill in reproducing the warm-season precipitation characteristics over the East Asia has not been robustly tested yet. Motivated by recent advances in computing power, model physics and high-resolution reanalysis, we use the convection-permitting weather research and forecasting (WRF) model configured with 3 km grid spacing to simulate the warm-season precipitation in eastern China for 10 seasons (2008–2017). The hourly 31-km-resolution ERA5 reanalysis data are used to provide initial and boundary conditions for the simulations. The objectives are (1) to evaluate the model skill in simulating warm-season precipitation climatology in the East Asian monsoon region, (2) to identify the promises and problems of the convection-permitting simulation, and (3) to investigate solutions for the model deficiencies. Results demonstrate that the 3-km-resolution WRF model reasonably reproduces the spatial characteristics of seasonal and sub-seasonal precipitation, the seasonal meridional migration associated with the summer monsoon activity, the diurnal variation phase and amplitude, and the propagating convection east of the Tibetan Plateau. The major deficiency is that the model overestimates precipitation amount, especially in the afternoon. Analysis and sensitivity experiments suggest that improved treatment of sub-grid cloud fraction and the aerosol effects may help to suppress the oft-reported high precipitation bias. This paper studies the impacts of El Niño-Southern Oscillation (ENSO) on wintertime extreme precipitation in China from 1961 to 2017, and possibly different influences of two El Niño types are also examined. We find that ENSO poses profound influences on extreme precipitation in many portions of China. El Niño (La Niña) intensifies (weakens) the winter precipitation extremes in southeastern China (SEC) and slightly weakens (intensifies) those in central-north China, mainly by changing the frequency rather than the intensity of extreme precipitation. Further analysis suggests that the East Asian winter monsoon tends to be weaker during El Niño winters, suppressing the southward invasion of cold dry air. Via the weakened Walker circulation, El Niño also triggers anomalously descending motion and anticyclone over the western North Pacific (WNP), which exhibits southwesterly anomalies over SEC and is thus conducive to the transport of sufficient moisture into that area. The changes over WNP are also accompanied by ascending motions over East Asia via a local meridional circulation alike the Pacific–Japan pattern. Meanwhile, El Niño induces a southward-displaced East Asian jet stream, as characterized by intensified westerly over southern China, corresponding to anomalously ascending motion there. Together, these changes facilitate the updrafts of circulations and the condensation of water vapor, thus increasing the occurrence of extreme precipitation events in SEC. Moreover, we find that different Niño types exhibit distinct subregional influences. In particular, the canonical eastern Pacific El Niño significantly increases the frequency of extreme precipitation in South China, whereas the Modoki central Pacific El Niño mainly impacts that in East China. Binary markers of insertion and deletion (InDel) play an important role in forensic personal identification, parentage testing, and individual ancestry inference. We first genotyped 30 InDels included in the Investigator DIPplex in 403 unrelated healthy Zunyi Miao people and analyzed the genetic polymorphisms, as well as explored the genetic relationship between Miao and 32 Chinese reference populations. No departures from the HWE were observed. The combined power of discrimination and the combined probability of exclusion were 0.99999999998 and 0.9884, respectively. Forensic parameters demonstrated that 30 markers are polymorphic and informative in the Zunyi Miao population and can be used as a tool for forensic personal identification and parentage testing. Allele frequency divergence analysis found that 12 out of 30 displaying high allele frequency difference between Turkic-speaking populations and other Chinese populations can be used as candidates of ancestry informative markers for ancestry inference of sub-population in East Asia. Population genetic parameters in the comprehensive population comparison among 33 Chinese populations indicated that our studied Hmong-Mien-speaking Miao has a close genetic relationship with geographically adjacent Enshi Tujia and genetically differentiate from Turkic-speaking populations. Skillful seasonal prediction of the number of extremely cold day (NECD) has considerable benefits for climate risk management and economic planning. Yet, predictability of NECD associated with East Asia winter monsoon remains largely unexplored. The present work estimates the NECD predictability in temperate East Asia (TEA, 30°-50°N, 110°-140°E) where the current dynamical models exhibit limited prediction skill. We show that about 50 % of the total variance of the NECD in TEA region is likely predictable, which is estimated by using a physics-based empirical (P-E) model with three consequential autumn predictors, i.e., developing El Niño/La Niña, Eurasian Arctic Ocean temperature anomalies, and geopotential height anomalies over northern and eastern Asia. We find that the barotropic geopotential height anomaly over Asia can persist from autumn to winter, thereby serving as a predictor for winter NECD. Further analysis reveals that the sources of the NECD predictability and the physical basis for prediction of NECD are essentially the same as those for prediction of winter mean temperature over the same region. This finding implies that forecasting seasonal mean temperature can provide useful information for prediction of extreme cold events. High prevalence of cholecystolithiasis in parts of East Asia has been postulated to be associated with Clonorchis sinensis infection. This study describes the development of a TaqMan-based real-time PCR assay for the detection of C. sinensis DNA in gallbladder bile and stone samples from patients with cholecystolithiasis. Primers and probe targeting the cytochrome c oxidase subunit 1 gene of mitochondrial DNA proved to be highly specific for C. sinensis and did not amplify other related heterogeneous DNA samples. The detection limit of this assay was 0.1 pg of adult C. sinensis genomic DNA. All of the egg-positive samples determined by microscopy yielded positive results by real-time PCR assay and that genetic testing of gallbladder stones using real-time PCR was considered as the most effective means for assessing C. sinensis infection status. This assay not only contributes to a greater understanding of stone pathogenesis but also benefits patients with cholecystolithiasis by facilitating effective diagnosis, treatment, and relapse prevention. Quantitative real-time reverse transcription PCR (qRT-PCR) is a rapid, sensitive, and reliable technique for gene expression studies. The accuracy and reliability of qRT-PCR results depend on the stability of the reference genes used for gene normalization. Therefore, a systematic process of reference gene evaluation is needed. Ganoderma lucidum is a famous medicinal mushroom in East Asia. In the current study, 10 potential reference genes were selected from the G. lucidum genomic data. The sequences of these genes were manually curated, and primers were designed following strict criteria. The experiment was conducted using qRT-PCR, and the stability of each candidate gene was assessed using four commonly used statistical programs-geNorm, NormFinder, BestKeeper, and RefFinder. According to our results, PP2A was expressed at the most stable levels under different fermentation conditions, and RPL4 was the most stably expressed gene in different tissues. RPL4, PP2A, and β-tubulin are the most commonly recommended reference genes for normalizing gene expression in the entire sample set. While Economic and Business Ethics has already attracted increasing attention in East Asia, a comprehensive survey of Economic and Business Ethics has never been done in this region. This study investigates the current status of Economic and Business Ethics as field of teaching, training and research in the East Asia region, particularly in China, Japan, and Korea. Based on multiple approaches that include questionnaire surveys, desktop analysis, and personal observation, this article reports on the current state of training, teaching, and research in the field of Economic and Business Ethics in the three countries.
