Heavy metal contamination is an urgent ecological governance problem in mining areas. In order to seek for a green and environmentally friendly reagent with better plant restoration effect to solve the problem of low efficiency in plant restoration in heavy metal pollution soil. In this study, we evaluated the effects of three biodegradable chelating agents, namely citric acid (CA), fulvic acid (FA) and polyaspartic acid (PASP), on the physicochemical properties of copper tailings, growth of ryegrass (Lolium perenne L.) and heavy metal accumulation therein. The results showed that the chelating agent application improved the physicochemical properties of copper tailings, increased the biomass of ryegrass and enriched more Cu and Cd in copper tailings. In the control group, the main existing forms of Cu and Cd were oxidizable state, followed by residual, weak acid soluble and reducible states. After the CA, FA or PASP application, Cu and Cd were converted from the residual and oxidizable states to the reducible and weak acid soluble states, whose bioavailability in copper tailings were thus enhanced. Besides, the chelating agent incorporation improved the Cu and Cd extraction efficiencies of ryegrass from copper tailings, as manifested by increased root and stem contents of Cu and Cd by 30.29–103.42%, 11.43–74.29%, 2.98–110.98% and 11.11–111.11%, respectively, in comparison with the control group. In the presence of multiple heavy metals, CA, FA or PASP showed selectivity regarding the ryegrass extraction of heavy metals from copper tailings. PCA analysis revealed that the CA-4 and PASP-7 treatment had great remediation potentials against Cu and Cd in copper tailings, respectively, as manifested by increases in Cu and Cd contents in ryegrass by 90.98% and 74.29% compared to the CK group. Accurately monitoring Canopy Nitrogen Concentration (CNC) is a prerequisite for precision nitrogen (N) fertiliser management at the farm scale with carbon and N budgeting across the landscape and ecosystems. While many spectral indices have been proposed for CNC monitoring, their applicability and accuracy are often adversely affected by confounding factors such as aboveground biomass (AGB), crop type, growth stages, and environmental conditions, limiting their broader application and adoption; with AGB being one of the most dominant signals and confounding factors at canopy scale. The confounding effect can become more challenging as AGB is also physiologically linked with CNC across the growth stages. Additionally, the interplay between index form, selection of optimal wavebands and their bandwidths remains poorly understood for CNC index design. This study proposes robust and cost-effective 2- and 4-waveband multispectral (MS) CNC indices applicable across a wide range of crop conditions. We collected 449 canopy reflectance spectra (400–980 nm) together with corresponding CNC and AGB measurements across four growth stages of ryegrass (winter and summer), and five growth stages of barley (winter-spring) in Victoria, Australia, in 2018 and 2019. All possible waveband (400–980 nm) combinations revealed that the best combination varied between seasons and crop types. However, the visible spectrum, particularly the blue region, presented high and consistent performance. Bandwidths of 10–40 nm outperformed either very narrow (2 nm) or very broad bandwidths (80 nm). The newly developed 2-waveband index (416 and 442 nm with 10-nm bandwidth; R2 = 0.75 and NRMSE = 0.2) and 4-waveband index (512, 440, 414 and 588 nm with 40-nm bandwidth; R2 = 0.81 and NRMSE = 0.17) exhibited the best performance, while validation with an independent dataset (from a different growing period to those used in the model development) obtained NRMSE values of 0.25 and 0.24, respectively. Crop irrigation requirements are usually estimated based on crop evapotranspiration (ETc) as determined by the reference evapotranspiration (ETo) and crop coefficient (Kc). There is a lack of knowledge on the irrigation requirements of tropical forage crops in Brazil, contrasting with the increasing use of irrigation in pastures. The effort of this study was to investigate what would be the water needs of tropical forages in Southern Brazil, based on a robust experimental database. The study was carried out in São Paulo State-Brazil using different forages species and their combinations [Guinea grass (GG); Guinea grass + black oat + ryegrass (GOR); Bermuda grass (BG), and Bermuda + black oat + ryegrass (BOR)]. The experimental fields were fully irrigated, and the Kc values were derived from ETc measurements on lysimeters; ETo was estimated using daily data from a nearby weather station and the standard FAO56 parameterization. Mean daily ETc values for GG, GOR, BG and BOR were 4.1, 2.9, 3.6, and 3.4 mm, respectively, and respective mean Kc values were 0.99, 0.90, 1.0, and 0.94. Average Kc values for all plots decreased as ETo increased, producing a negative Kc-ETo relationship, mainly when ETo reached values greater than 5 mm d−1. This was most likely due to internal plant stomatal resistance to vapor release from the leaves diffusing to the atmosphere at high ETo. So, the time-based Kc curves described by FAO 56 manual should be adjusted for the analyzed crops considering different ranges of ETo to improve the required irrigation depth. [ABSTRACT FROM AUTHOR]

Climate change is anticipated to lead to an increase in the occurrence and intensity of drought and fluctuations in insect cycles that will challenge modern pasture systems. Feeding by root aphids such as Aploneura lentisci Pass. can be a significant challenge to pastures. These below-ground living aphids are commonly found in New Zealand and Australia, feeding year-round on the roots of graminaceous plants such as perennial ryegrass (Lolium perenne L.). Some strains of the fungal endophyte Epichloë festucae var. lolii in perennial ryegrass can provide protection against root aphids and greater resilience under drought, contributing to higher persistency and growth than endophyte-free plants. However, the interaction between insect pressure and drought is not understood. This study examined the effect of drought on root aphid populations and plant performance in perennial ryegrass plants relative to endophyte status (±) and endophyte strain (AR37, NZCT) in a glasshouse experiment. Plants were cloned across the drought and well-watered treatments, and half of the plants were inoculated with root aphids, whilst half of the plants were treated with insecticide. Endophyte infection with strain NZCT and AR37 reduced root aphid numbers. Aphid populations were significantly higher in drought-stressed than in well-watered plants in both endophyte-infected and endophyte-free treatments. Under drought conditions, root aphid populations were increased 4-fold in NZCT and 8-fold in AR37 and endophyte-free plants in comparison with their well-watered counterparts. Root aphids reduced shoot dry weight by 16% in drought-exposed and 26% in well-watered plants in comparison with their insecticide-treated counterparts whilst reducing root biomass by 49%. Our results suggest that root aphids are likely able to exploit the higher availability of amino acids in the plant sap of drought-exposed plants. This study provides evidence that climate change-mediated impacts of root aphids could reduce production in perennial ryegrass-dominant pastures. Green manuring is an established strategy for soil nutrient enrichment; however, its effects on the soil microbiome remain unclear. In this study, we assessed the impact of green manuring on soil microbial diversity, functional profiles, and enzymatic activities in a pot experiment involving tobacco rotation with three types of green manure: smooth vetch, ryegrass, and radish. Our data revealed distinct temporal shifts in bacterial communities under diverse tillage while fungi communities showed obvious clustering between fallow and green manuring. Moreover, green manure-sensitive bacterial and fungal taxa constitute a greater proportion of total sequences than their fallow-sensitive counterparts, indicating a more pronounced influence of green manuring on microbial communities. Functionally, green manuring augmented the relative abundance of N-cycle-related genes and was strongly associated with elevated N-acetyl-glucosaminidase activity. Notably, a subset of green manure-sensitive microbes exhibited significant antagonistic interactions with the fungal pathogens Fusarium spp. and Fusarium solani, and synergistic interactions with the beneficial bacteria Pseudomonas spp. Collectively, these findings suggest that green manuring promotes an optimized soil environment characterized by efficient nutrient cycling and diminished pathogenicity, thereby providing a scientific basis for targeted microbiota manipulation in agricultural systems. Outcomes of weed biological control projects are highly variable, but a mechanistic understanding of how top-down and bottom-up factors influence the success of weed biological control is often lacking. We grew Rumex obtusifolius, the most prominent native weed in European grasslands, in the presence and absence of competition from the grass Lolium perenne and subjected it to herbivory through targeted inoculation with root-boring Pyropteron spp. To explore whether the interactive effects of competition and inundative biological control were size-dependent, R. obtusifolius was planted covering a large range of plant sizes found in managed grasslands. Overall, competition from the grass sward reduced aboveground biomass and final root mass of R. obtusifolius about 62- and 7.5-fold, respectively, and increased root decay of R. obtusifolius from 14 to 58%. Herbivory alone increased only root decay. However, grass competition significantly enhanced infestation by Pyropteron spp. and, as a consequence, enhanced the impact of herbivory on aboveground biomass and final root mass. The synergistic effect was so strong that R. obtusifolius plants grown from initially smaller roots did no longer develop. Inoculating R. obtusifolius with Pyropteron species in grasslands should be further pursued as a promising inundative biological control strategy in the weed's native range. [ABSTRACT FROM AUTHOR]

Background and aims: The intake of selenium, an essential element for animals and humans, in ruminants is largely determined by selenium concentration in ingested forages, which take up selenium mainly from soil. Ruminant excreta is a common source of organic fertilizer, which provides both nutrients and organic matter. This study aims to unentangle the unclear effect of applying different types of ruminant excreta in soils of different organic matter contents on selenium uptake by forage. Methods: Perennial ryegrass (Lolium perenne) was grown in soils of different organic matter contents. Urine and/or feces collected from sheep fed with organic or inorganic mineral supplements, including selenium, were applied to the soils. The selenium in the collected samples were analyzed using ICP-MS. The associated biogeochemical reactions were scrutinized by wet chemistry. Results: The application of urine and/or feces resulted in either the same or lower selenium concentrations in perennial ryegrass. The excreta type did not affect total selenium accumulation in grass grown in low organic matter soil, whereas in high organic matter soil, feces resulted in significantly lower total selenium accumulation than urine, which was attributed to a possible interaction of selenium sorption in soil and microbial reduction of Se. Conclusion: This one-time excreta application did not increase, but further decrease in some treatments, selenium concentration and accumulation in the perennial ryegrass. Consequently, to increase ruminant selenium intake, supplementing selenium directly to animals is more recommended than applying animal manure to soil, which might drive selenium reduction and decrease selenium uptake by grass. Perennial ryegrass is a grass species used to establish lawns in urban areas where pollution is a major environmental problems. Cadmium (Cd) and nickel (Ni) contribute significantly to these pollutants and may cause photosynthetic limitation. The main objective of this work was to perform a comprehensive analysis of photosynthetic efficiency of perennial ryegrass seedlings under Cd and Ni stress. Some of the main indices of photosynthetic efficiency (prompt and delayed chlorophyll-a fluorescence signals and modulated reflectance at 820 nm) were compared with growth parameters. Two cultivars were tested: 'Niga' and 'Nira'. A decrease in photosystem (PS) II and PSI activity was observed. This was due to an increase in nonradiative dissipation of the PSII antenna, a decrease in PSII antenna size, or a decrease in the number of photosynthetic complexes with fully closed PSII RCs. Efficiency of electron transport was decreased. The effect on the modulated reflectance signal could indicate a restriction in electron flow from PSII to PSI. The correlation between photosynthetic efficiency parameters, such as Area, Fo, Fm, and Fv, and growth parameters, confirmed that some photosynthetic efficiency parameters can be used as indicators for early detection of heavy metal effects. Increasing productivity and resource efficiency through sustainable intensification of grazing systems impose new challenges on crops and livestock. The sustainability of world agriculture is threatened by the degradation of ecosystem services anthropogenic interventions such as reduced biological diversity, water and air pollution, and loss of soil quality. The search for technologies that increase animal productivity with economic and environmental sustainability is necessary. The aim was to determine whether productive systems based on African Bermuda grass, overseeded in rows with annual ryegrass and black oat, would present better beef cattle performance by irrigation and/or inclusion of forage legumes in the grazing systems. We used 72 steers, 24 in each of the 3 years of evaluation, distributed in 12 paddocks under rotational stocking. Each paddock had four sub-paddocks. The experiment was completely randomized in a 2 × 2 factorial arrangement, using irrigation and legumes as factors. Production and chemical composition of pastures and animal performance were evaluated for 3 years in winter, spring, and summer. The irrigation provided promising results in spring, with a higher stocking rate (2194 vs 1254 kg/ha) and live weight gain per area (5.24 vs 2.81 kg/ha/day) in the second year due to water restriction. The forage legumes with irrigation provided a higher average daily gain in the summer than the non-irrigated pasture with legumes. Here we show for the first time that in long-term results (3 years), the pastures with legumes showed a high live weight gain per area (1556 vs 1452 kg/ha/year), and in the third year, the highest difference for the stocking rate in the summer (4134 vs 3311 kg/ha). Legumes or irrigation do not change the pasture's nutritional compounds. The use of forage legumes benefits the grazing system with long-term results. Purpose: The incorporation of ryegrass (Lolium multiflorum Lam.; RG), a winter manure, could partly replace chemical N and reduce N loss during the succeeding rice seasons, but little is known about its impact on greenhouse gas emission. This study investigated the effect of different RG-urea substitution ratios (0% RG and 100% urea, 25% RG and 75% urea, 50% RG and 50% urea, 75% RG and 25% urea, 100% RG and 0% urea,) on net C and N mineralization, CH4 and N2O emissions in a paddy soil. Methods: Gas samples for CH4 and N2O fluxes were collected by using a closed chamber and determined by chromatograph method. Net C and N mineralization from the incorporated RG residue were tested by a mesh bag method. Results: Net C and N mineralization from RG followed a single exponential decay model, with 95.5%-97.8% of the original C and 98.7%-99.3% of N released during 192 days. The RG-urea substitution ratio increased CH4 emission, but was negatively correlated with N2O emission. In comparison with 0% substitution, global warming potential (GWP) and greenhouse gas intensity (GHGI) were not significantly different for the 25% and 50% RG substitutions, but were significantly higher for the 75% and 100% substitutions (P < 0.05). Soil redox, C and N remaining in litter residue were key characteristics explaining CH4 emission, while NH4+-N and NO3−-N were correlated with N2O emission. Conclusion: The increased GWP by CH4 emission after RG incorporation could be offset by N2O reduction when RG-urea substitution ratio was 50% or less. [ABSTRACT FROM AUTHOR]

In summer 2018, brown stripe symptoms were observed on leaf blades of perennial ryegrass (Lolium perenne L.) used as a turfgrass in an athletic field in Saitama Prefecture, Japan. Bacterial ooze exuded from lesions, from which a pathogenic bacterium was isolated. Inoculation of blades of perennial ryegrass with the isolates reproduced the same symptoms. Pathogenicity and bacteriological properties of the isolates were almost identical to those of Acidovorax avenae, and the identification was supported by phylogenetic analysis. This is the first report of the brown stripe on perennial ryegrass caused by A. avenae (= A. avenae subsp. avenae) in Japan. [ABSTRACT FROM AUTHOR]

Purpose : Nitrogen (N) transfer from white clover (Trifolium repens cv.) to ryegrass (Lolium perenne cv.) has the potential to meet ryegrass N requirements. This study aimed to quantify N transfer in a mixed pasture and investigate the influence of the microbial community and land management on N transfer. Methods: Split root 15N-labelling of clover quantified N transfer to ryegrass via exudation, microbial assimilation, decomposition, defoliation and soil biota. Incorporation into the microbial protein pool was determined using compound-specific 15N-stable isotope probing approaches. Results: N transfer to ryegrass and soil microbial protein in the model system was relatively small, with one-third arising from root exudation. N transfer to ryegrass increased with no microbial competition but soil microbes also increased N transfer via shoot decomposition. Addition of mycorrhizal fungi did not alter N transfer, due to the source-sink nature of this pathway, whilst weevil grazing on roots decreased microbial N transfer. N transfer was bidirectional, and comparable on a short-term scale. Conclusions: N transfer was low in a model young pasture established from soil from a permanent grassland with long-term N fertilisation. Root exudation and decomposition were major N transfer pathways. N transfer was influenced by soil biota (weevils, mycorrhizae) and land management (e.g. grazing). Previous land management and the role of the microbial community in N transfer must be considered when determining the potential for N transfer to ryegrass. [ABSTRACT FROM AUTHOR]

Irrigation with mine water not only improves water resource utilization rates and alleviates water shortages but can also promote crop growth and yields. However, long-term irrigation with mine water can significantly change the physicochemical properties of soil due to its unique mineral content. In this study, two groups of experiments were conducted (pot experiments and soilless cultivation) using mine water from the Fushun mining area to explore its effects on the physiological and photosynthetic characteristics of ryegrass, as well as soil properties. Mine water irrigation inhibited all of the indicators evaluated in this study, whereas a mixture of clean water and mine water had a stimulatory effect. Interestingly, this stimulatory effect was weakened as the proportion of mine water increased but reached its maximum when the ratio was 2:1. Moreover, the inhibitory effect of the irrigation water was weakened as the proportion of clean water increased. The contents of K+, Na+, Ca2+ and Mg2+ in soil were higher than those in the soil matrix, and the content of the nutrient elements N, P and K, and metal cations increased gradually as the mine water ratio increased, and the electrical conductivity increased significantly. Moreover, the pH of the soil decreased steadily (i.e. acidity increased) with increased soil salinity. Our findings indicated that a mixture of mine water from Fushun mining area and clean water at a 1:2 ratio could improve the physiological, growth, and photosynthetic characteristics of ryegrass by enhancing soil quality. Our study thus provides an experimental precedent for the utilization of mine water in ecological restoration and agricultural irrigation, and could therefore serve as a basis for the development of novel strategies for environmental restoration and the utilization of water resources. The addition of exogenous organic matter (EOM) generally stimulates microbial activity and shapes soil structure, likely with stronger effects at a higher application dose. It is not known, however, if and how the application dose of EOM would impact its own mineralisation and that of native soil organic carbon (SOC) by such mediation of soil pore network structure. In 90-day soil incubation experiments, we compared mineralisation of ± 2 cm pieces of 13C-labelled ryegrass as EOM reference at two doses (0.5 g kg−1 vs. 5 g kg−1) and mineralisation of SOC in sandy loam and silt loam soils that were initially subjected to different degrees of disturbance, namely soil sieving at < 2 mm and < 10 mm. We found that the high EOM dose-induced mesoporosity (i.e. vol% increase of pore neck class > 300 μm class by 130% vs. unamended control) and macroporosity (i.e. EOM doses 0.5 g kg−1 and 5 g kg−1 increased the vol% of the 60–100 μm class by 80 and 120% vs. unamended control) formation in the sandy loam soil, whereas the porosity in the silt loam soil remained unaffected. The % of EOM mineralised was independent of EOM dose and therefore did not relate to the changes in soil pore network structure. However, the observed development of meso- and macropores in the sandy loam soil appeared to be associated with stronger stimulation of SOC mineralisation at increasing EOM dose (30% and 71% more SOC mineralisation at low and high dose vs. unamended control). In the silt loam soil, only the high EOM dose-stimulated SOC mineralisation vs. unamended control (28%) but soil pore network structure remained unresponsive to EOM dose. This suggests that stimulation of SOC mineralisation following EOM addition might also be in part linked to the formation of larger pores in the sandy loam soil, and that this stimulation is not always proportional to EOM dose. Information regarding the germination and seedling growth behavior of a potential weed species is an important tool to manage weeds without the use of agricultural chemicals that cause harmful effects on human health and the environment. A series of experiments were directed to investigate the influence of different environmental factors (temperature, pH, NaCl, moisture stress, and seed burial depth) on germination and seedling emergence of perennial ryegrass (Lolium perenne L.) under controlled conditions. Results suggested that 25 °C is the optimum temperature for maximum germination (95%) and seedling growth of perennial ryegrass, however, a quick decline was observed at 35 °C. Seed germination was unaffected by pH levels ranging from 5 to 10. The 92% seed germination was recorded where no salt stress was applied and germination was reduced by 87% at 250 mMNaCl concentration. Seed germination was unaffected by osmotic potential ranges from 0 to − 0.4 MPa thereafter declined and completely inhibited at − 0.8 or − 1.0 MPa. No seed emerged at the soil surface or a soil depth of 6 or 7 cm and 90% emergence occurred at 1 cmsoil depth. The germination and seedlings parameters like time to initial germination, mean germination time, time taken to 50% germination and germination index, root and shoot length, and fresh and dry weight of root and shoot are significantly affected with the environmental factors. The information obtained in this study will be helpful to develop better management strategies for germination and the emergence of perennial ryegrass in areas where it has the ability to rapidly colonize. Aims: The aim of this work was to evaluate the fertilizing effect of three anaerobic co-digestates on the growth of the herbaceous plant Lolium rigidum. Methods: Nine treatments, combining different nutritional solutions (organic and inorganic) and number of fertilizations (one or two) were evaluated. Organic nutritive solution: plants grown with different olive mill solid waste (OMSW) -microalgae co-digestates: 75% OMSW-25% Raphidocelis subcapitata, volatile solids (VS) basis (OMSW-Rs); 50% OMSW- 50% Chlamydomonas Reinhardtii, VS basis (OMSW-Chl); and 75% OMSW-25% Secenedesmus quadricauda, VS basis (OMSW-Sq). Inorganic nutritive solution (INS): plants grown with inorganic Hoagland nutrient solution at 50%. After 60 days of experimentation, biometric and nutritional characteristics and photosynthetic activity were measured. Results: The results showed a favourable growth, development and nutritional quality of L. rigidum plants when digestates obtained from the anaerobic co-digestion of OMSW-microalgae are used as organic nutritional solutions as opposed to INS ones. The highest total biomass of L. rigidum was obtained with the treatments that involved two fertilizations. No inhibition due to excess nutrients was observed. A higher root/shoot ratio was achieved with the digestates of OMSW-Rs and OMSW-Ch as compared to that obtained with OMSW-Sq (F = 17.23 p ≤ 0.001). The nitrogen shoot biomass obtained after the organic treatments with the above-mentioned co-digestates was higher than that obtained after the inorganic treatment. Net photosynthesis rates did not present differences in the co-digestates treatments, being equal or superior to the INS treatments. Ryegrass (Lolium perenne L.), a high-quality forage grass, is a good nutrient source for herbivorous livestock. However, improving nitrogen use efficiency and avoiding nitrate toxicity caused by excessive nitrogen are continual challenges in ryegrass production. The molecular mechanism underlying the response of ryegrass to nitrogen, especially excessive nitrogen, remains unclear. In this study, the transcriptomic changes under different nitrogen levels were investigated in perennial ryegrass by high-throughput next-generation RNA sequencing. Phenotypic characterization showed that treatment with half of the standard N concentration (N0.5) led to a better growth state than the other three treatments. The treatments with the standard N concentration (N1) and treatments with ten times higher than the standard N concentration (N10) contained excessive nitrogen, which placed stress on plant growth. Analysis of differentially expressed genes indicated that 345 and 104 genes are involved in the regulation of nitrogen utilization and excessive nitrogen stress, respectively. KEGG enrichment analysis suggested that "photosynthesis-antenna proteins" may respond positively to appropriate nitrogen conditions, whereas "steroid biosynthesis", "carotenoid biosynthesis" and "C5-branched dibasic acid metabolism" were identified as the top significantly enriched pathways in response to excessive nitrogen. Additionally, 21 transcription factors (TFs) related to nitrogen utilization were classified into 10 families, especially the AP2-EREBP and MYB TF families. Four TFs related to excessive nitrogen stress were identified, including LOBs, NACs, AP2-EREBPs and HBs. The expression patterns of these selected genes were also analyzed. Legume-based forage plant mixtures are known to increase biomass production over the mixture species grown as pure stands (overyielding), which has partly been attributed to enhanced nitrogen availability by legumes. However, the relative importance of underlying processes of these positive diversity effects and their drivers are not fully understood. Here we assessed if outcome and causes of diversity effects depend on the legume-species genetic identity. Over five years, we cultivated different white clover (Trifolium repens) populations, a grass and forb species in pure stands and clover-based mixtures and recorded biomass yield. Complementarity and selection effects of mixtures and relative yields of mixture species were calculated based on both unfertilized and nitrogen-fertilized non-leguminous pure stands. Results showed that the clover population altered the overall strength of diversity effects as well as the direction and magnitude of their temporal trends, at least for the grass component of mixtures. Differences in diversity effects between clover populations diminished when fertilized instead of unfertilized non-leguminous pure stands were considered. Hence, a part of these differences likely results from dissimilar effects of clover populations on nitrogen availability. Argentine stem weevil adults (ASW, Listronotus bonariensis) feed on the leaves of agricultural grasses and their larvae mine the pseudostem, causing extensive damage that can result in plant death. Plants emit volatiles that serve as signals to host-searching insects and these odours can be altered by both herbivory and fungal endophyte-infection. This study investigated whether ASW adults utilise olfaction to identify their host plants, perennial ryegrass (Lolium perenne), and if conspecific herbivory or the presence of Epichloë festucae var. lolii fungal endophytes (strain wild-type or AR1) influenced such responses. Results from olfactometer bioassays established that ASW adults were able to utilise their olfactory response to orient towards volatiles released by perennial ryegrass and further, the weevils displayed a preference for plants previously damaged by conspecific weevils. However, there was no evidence that weevils had the ability to distinguish between endophyte-infected and endophyte-free plants using olfaction alone. Using a push–pull extraction technique, thirteen volatile compounds were identified in the blend released by perennial ryegrass. Endophyte and herbivory were found to alter these volatile compounds and quantities emitted by this forage grass. This study suggests that despite observing differences in the plant volatile blend, ASW do not perceive endophyte (wild-type and AR1) using olfaction alone and must rely on other cues, e.g. contact chemoreception or post-ingestional malaise, to detect the presence of a bioactive endophyte in an otherwise acceptable host plant. Nematodes play an important role in agricultural soil in terms of their pest status and their contribution to soil quality, where they may serve as a valuable indicator of soil health. The aim of the study was to assess changes occurring in nematode community structure, as affected by soil tillage applied to high N-input and irrigated pastures in South Africa. Such practices were investigated by means of analysing diversity and ecosystem function indices, as well as the faunal profile, to enable analysis of their effect on soil quality. The pre-treatment soil analyses showed a low overall mean nematode density. No clear and predictable pattern was present in the nematode community structure in relation to tillage. The lack of indicative information leading from the nematode community structure to the tillage disturbance types might be linked to the fact that such systems were able to recover from soil disturbance within the relatively short period of less than a year. The popularized application of annual ryegrass—maize rotation (ARMR) in southern China has been proposed to fully utilize the farmlands and to increase forage yield and quality. Herein, one growth cycle of ARMR was conducted and soil bacteria were analyzed by 16S rRNA sequencing for control (CK), after the preceding crop (monoculture, or mixed sowing of annual ryegrass and oat) and the successive crop (maize). Our results indicated that the α-diversity of soil bacteria was changed in the ARMR system, which was related to the activity of urease and available phosphatase. The mixed sowing of annual ryegrass and oat in preceding crop could improve the yield and quality, while it was accompanied by unbalanced soil community. With the increased sowing proportion of oat to annual ryegrass, the soil pH increased while the soil available phosphatase decreased. The ARMR system was found to benefit the soil microenvironment by increasing the beneficial soil bacteria and enzyme activity or decreasing the harmful soil bacteria. Considering the soil bacteria α-diversity index and physicochemical properties comprehensively, the recommended sowing regime is the mixed sowing of M2 (22.5 kg·hm−2 annual ryegrass with 75 kg·hm−2 oat). Paenibacillus species are Gram-positive bacteria that have been isolated from a diverse array of plant species and soils, with some species exhibiting plant growth-promoting (PGP) activities. Here we report two strains (S02 and S25) of a novel Paenibacillus sp. that were isolated from perennial ryegrass (Lolium perenne) seeds. Comparative genomics analyses showed this novel species was closely related to P. polymyxa. Genomic analyses revealed that strains S02 and S25 possess PGP genes associated with biological nitrogen fixation, phosphate solubilisation and assimilation, as well as auxin production and transportation. Moreover, secondary metabolite gene cluster analyses identified 13 clusters that are shared by both strains and three clusters unique to S25. In vitro assays demonstrated strong bioprotection activity against phytopathogens (Colletotrichum graminicola and Fusarium verticillioides), particularly for strain S02. A transcriptomics analysis evaluating nitrogen fixation activity showed both strains carry an expressed nif operon, but strain S02 was more active than strain S25 in nitrogen-free media. Another transcriptomics analysis evaluating the interaction of strains with F. verticillioides showed strain S02 had increased expression of core genes of secondary metabolite clusters (fusaricidin, paenilan, tridecaptin and polymyxin) when F. verticillioides was present and absent, compared to S25. Increasing the efficiency of current forage breeding programs through adoption of new technologies, such as genomic selection (GS) and phenomics (Ph), is challenging without proof of concept demonstrating cost effective genetic gain (∆G). This paper uses decision support software DeltaGen (tactical tool) and QU-GENE (strategic tool), to model and assess relative efficiency of five breeding methods. The effect on ∆G and cost ($) of integrating GS and Ph into an among half-sib (HS) family phenotypic selection breeding strategy was investigated. Deterministic and stochastic modelling were conducted using mock data sets of 200 and 1000 perennial ryegrass HS families using year-by-season-by-location dry matter (DM) yield data and in silico generated data, respectively. Results demonstrated short (deterministic)- and long-term (stochastic) impacts of breeding strategy and integration of key technologies, GS and Ph, on ∆G. These technologies offer substantial improvements in the rate of ∆G, and in some cases improved cost-efficiency. Applying 1% within HS family GS, predicted a 6.35 and 8.10% ∆G per cycle for DM yield from the 200 HS and 1000 HS, respectively. The application of GS in both among and within HS selection provided a significant boost to total annual ∆G, even at low GS accuracy rA of 0.12. Despite some reduction in ∆G, using Ph to assess seasonal DM yield clearly demonstrated its impact by reducing cost per percentage ∆G relative to standard DM cuts. Regarding to the super-beneficial effects of arbuscular mycorrhizal fungi (AMF) on host plant in response to environmental stresses, there is a great interest for understanding the mechanisms that enable mycorrhizal plant tolerate the stress conditions. In the present study, some physiological and molecular responses induced by arbuscular mycorrhizal fungus Funneliformis fasciculatum in Ni-stressed Lolium perenne were investigated. Since heavy-metal stress is reported to alter the water relations of plant, expression of two aquaporin (AQP)-coding genes (Lptip1;1 and Lptip1;2) were studied as a molecular mechanism which may be involved in Ni-stress tolerance. The results demonstrated a pronounced positive effect of AMF on Lolium growth, water content and photosynthetic pigments content under Ni stress condition, probably through an avoidance strategy which led to the decrease in Ni accumulation in plant tissues as well as decreasing Ni translocation to the shoots. The results also indicated the mycorrhizal-induced expression of both Lptip1;1 and Lptip1;2 genes in plant roots and shoots under Ni stress. Background and aims: Incorporation of biochar into the soil sequesters C for millennia, but the concomitant effects on plant rhizodepositions and nutrient (e.g., nitrogen; N) trade-offs via interactions of heterotrophic microbiota, might offset this sequestration. Methods: Ryegrass (Lolium perenne L.) with and without biochar amendment were pulse labelled in a 13CO2 atmosphere and 15N fertilizer added. Ryegrass and soils were destructively sampled at 16 and 30 days after seedling emergence. Isotope analysis was coupled with MiSeq sequencing of bacterial (16s rRNA) and fungal (ITS) genes to identify the effect of biochar on the associated microbiota involved in 13C allocation into soil aggregates and promotion of 15N uptake by L. perenne. Results: Biochar increased root biomass and 15N uptake but decreased rhizodeposited-13C recovery from large and small macroaggregates (by 12–57% and 57–72%, respectively). These changes in 13C flow and 15N uptake were accompanied by an increase in microbial biomass, and enhanced negative correlations between bacteria and fungi. O2PLS indicated members of seventeen genera that were correlated with soil stabilization of rhizodeposits in soil and plant N-uptake. For instance, Xanthomonadales (Proteobacteria) and RB41 (Acidobacteria), previously reported to be plant growth promoting rhizobacteria, were found to be positively correlated with 15N uptake by L. perenne. Conclusions: Our research explored the genera associated with biochar-modified 15N uptake by Lolium perenne and photosynthate 13C allocation into soil aggregates. Future research with SIP is required to fully assess microbial turnover, the ubiquity of similar rhizosphere microbiota and their fundamental importance for sequestration in the plant-soil-microbe-biochar systems. Ryegrass was pulse-labeled with enriched 13CO2 for 18 h, followed by dynamic photosynthetic-carbon (13C) quantification in the plant (shoot, root), soil aggregates (three size classes), and microbial phospholipids fatty acids (PLFA-SIP) in soil amended with or without 700 °C-pyrolyzed biochar. We observed that biochar led to no difference of 13C allocation in shoot or root but reduced 88.7% of total 13C in soil, with decreased incorporation by 92.8% (macroaggregates), 94.5% (microaggregates), and 84.1% (silt-clays), respectively, compared to biochar-unamended soil. Meanwhile, biochar exerted negative effects on fungal relative abundance but led to positive impacts on that of bacteria, e.g., it reduced root-associated fungi (i.e., 16:1ω5c) and fungal-assimilated 13C (from averagely 71.2 ng C g−1 soil to 26.3 ng C g−1 soil after biochar application). The enhanced bacteria/fungi could be driven by biochar-mediated pH increase that relieved acid stress to bacteria. Co-occurrence network confirmed that biochar addition favored bacteria to compete with fungi, leading to decreased aggregation and stability (indicated by reduced normalized mean weight diameter) due to less fungal entangling with aggregates, thus exposing the rhizodeposits to bacterial (i.e., actinomycetes) decomposition. The correlation analysis further evidenced that fungal abundance was associated with 13C accumulation in soil aggregates, while bacterial relative abundance especially that of actinomycetes was negatively correlated with 13C accumulation. Random forest modeling (RF) supported the contributions of fungi to 13C-sequestration compared to bacteria. Taken together, we concluded that less stabilization of rhizodeposits in the biochar-amended soil was due to changes in microbial community, particularly the balance of fungi-bacteria and their interactions with soil physicochemical properties, i.e., aggregation and pH. We constructed a mining soil restoration system combining plant, complex substrate and microbe. Sludge was added to reconstructed mine substrates (RMS) to accelerate the reclamation process. The effect of sludge on plant growth, microbial activity, soil aggregate stability, and aggregation-associated soil characteristics was monitored during 10 years of reclamation. Results show that the height and total biomass of ryegrass increases with reclamation time. Sludge amendment increases the aggregate binding agent content and soil aggregate stability. Soil organic carbon (SOC) and light-fraction SOC (LFOC) in the RMS increase by 151% and 247% compared with those of the control, respectively. A similar trend was observed for the glomalin-related soil protein (GRSP). Stable soil aggregate indexes increase until the seventh year. In short, the variables of RMS determined after 3–7 years insignificantly differ from those of the untreated sample in the tenth-year. Furthermore, significant positive correlations between the GRSP and SOC and GRSP and soil structure-related variables were observed in RMS. Biological stimulation of the SOC and GRSP accelerates the recovery of the soil structure and ecosystem function. Pasture management is highly dependent on accurate biomass estimation. Usually, such activity is neglected as current methods are time-consuming and frequently perceived as inaccurate. Conversely, spectral data is a promising technique to automate and improve the accuracy and precision of estimates. Historically, spectral vegetation indices have been widely adopted and large numbers have been proposed. The selection of the optimal index or satisfactory subset of indices to accurately estimate biomass is not trivial and can influence the design of new sensors. This study aimed to compare a canopy-based technique (rising plate meter) with spectral vegetation indices. It examined 97 vegetation indices and 11,026 combinations of normalized ratio indices paired with different regression techniques on 900 pasture biomass data points of perennial ryegrass (Lolium perenne) collected throughout a 1-year period. The analyses demonstrated that the canopy-based technique is superior to the standard normalized difference vegetation index (∆, 115.1 kg DM ha−1 RMSE), equivalent to the best performing normalized ratio index and less accurate than four selected vegetation indices deployed with different regression techniques (maximum ∆, 231.1 kg DM ha−1). When employing the four selected vegetation indices, random forests was the best performing regression technique, followed by support vector machines, multivariate adaptive regression splines and linear regression. Estimate precision was improved through model stacking. In summary, this study demonstrated a series of achievable improvements in both accuracy and precision of pasture biomass estimation, while comparing different numbers of inputs and regression techniques and providing a benchmark against standard techniques of precision agriculture and pasture management. Comparative evaluations were conducted to assess the effects of different pH levels, NaCl-induced salt stress, and PEG-induced drought stress on the mycelial growth of Xerocomus badius. The results showed that X. badius mycelium grew well at a wide pH range of 5.00 ~ 9.00. Although the mycelium remained viable, mycelial growth of X. badius was significantly inhibited with increasing salt and drought stresses. Furthermore, a soilless experiment in Petri dishes was performed to investigate the potential of X. badius to induce beneficial effects on seed germination and seedling growth of annual ryegrass (Lolium multiflorum Lam.) under salt and drought stresses. Seed priming with X. badius enhanced the seedling growth of L. multiflorum Lam. under NaCl-induced salt stress and PEG-induced drought stress. However, X. badius did not significantly improve the seed germination under non-stress and mild stress conditions. It suggested that X. badius inoculation with seeds was not essential for seed germination under non-stress and mild stress conditions, but contributed highly to seedling growth under severe stress conditions. Therefore, seed priming with X. badius on ryegrass could be an effective approach to enhance plant tolerance against drought and salt stresses. Italian ryegrass (Lolium multiflorum Lam.) is widely cultivated in temperate regions owing to its excellent producibility and digestibility. Nitrogen is an essential nutrient for plant growth; however, the overuse of nitrogen fertilizers can lead to excessive accumulation of nitrate-nitrogen in Italian ryegrass, which can prove to be lethal for ruminants. To study the genetic control of the nitrate-nitrogen content in Italian ryegrass, an Italian ryegrass line LNG8 (with low 0.12% dry matter (DM) of nitrate-nitrogen) and a second line JFIR12 (with a "normal" 0.20% DM of nitrate-nitrogen) were pair-crossed and 190 F2 individuals derived from these were used to construct an SSR-based linkage map for conducting QTL analysis. A linkage map was constructed based on 212 Lolium SSR markers. The female parental map consisted of seven linkage groups (LGs) and had a total length of 714.1 cM with 178 loci and an average map density of 4.0 cM. The male parental map consisted of 7 LGs and had a total length of 737.4 cM with 199 loci and an average map density of 3.7 cM. We measured the nitrate-nitrogen content of the mapping population in three replicates (including the main stem and two tillers separated from the main stem) at the six-leaf stage. The nitrate-nitrogen content of the mapping population followed a normal distribution with a marginal skewness toward the lower parent. Our analysis revealed the presence of six QTLs on three LGs. A major QTL on LG1 was detected in all three replicates in both two-parental maps, which could explain 19.4%–29.9% of the phenotypic variation. Furthermore, based on the information of model plant species such as rice and Arabidopsis, we cloned six nitrogen metabolism-related genes and attempted to locate these on our linkage maps; however, none of these six genes were located in the demarcated QTL regions. These results will be useful for understanding the genetic bases of nitrate-nitrogen content in Italian ryegrass and the molecular cloning of QTLs for determination of nitrate-nitrogen content. Purpose: Regulatory and economic conditions drive the application of the concept of homologous groups on therapeutic and diagnostic formulations. In this context, a possible reduction of allergens in the skin prick test panel in the clinical routine should be investigated for the grass, birch and Dermatophagoides homologous group. Methods: Grass and cultivated rye skin prick test results of 1101 patients, birch/hazel/alder skin prick test results of 1131 patients and 1068 Dermatophagoides pteronyssinus and farinae skin prick tests were retrospectively evaluated with regard to sensitivity and false-negative rate detecting sensitization against cultivated rye with grass skin prick test, sensitization against hazel and/or alder with birch skin prick test and sensitization towards Dermatophagoides farinae with Dermatophagoides pteronyssinus skin prick test. Results: Skin prick test results in all groups highly correlated in Spearman Rank Order correlation. Sensitivity of grass skin prick test detecting sensitization against cultivated rye was highest with 98.9%, sensitivity of birch skin prick test detecting sensitization toward hazel and alder was 95.6 and 90.0%, respectively. Sensitivity of skin prick test with Dermatophagoides pteronyssinus detecting sensitization towards Dermatophagoides farinae was 93.6%. Only 26 patients with positive rye, hazel, alder or Dermatophagoides farinae skin prick test that were overlooked by grass, birch or Dermatophagoides pteronyssinus skin prick test reported allergic symptoms. Conclusion: In routine clinical practice, sensitization against cultivated rye can safely be detected by skin prick test with grass allergen. In addition, the sensitivity of birch skin prick test is high in terms of sensitization towards hazel, but lower for alder. Sensitivity of Dermatophagoides pteronyssinus skin prick test also is high to detect sensitization towards Dermatophagoides farinae. Further research will indicate if several skin prick test allergens will disappear and be completely replaced by a single skin prick test based on the principle of homologous groups. Aims: An Italian ryegrass cultivar (Lolium multiflorum Lam. cv. Bb2067), selected and bred for increased leaf magnesium (Mg) concentration in the 1970s, reduced the incidence of hypomagnesaemia in sheep under experimental grazings. Here, we report evidence from archival experiments showing that cv. Bb2067 had consistently greater Mg concentrations at multiple sites. We also aimed to quantify variation in leaf Mg concentration among modern perennial ryegrass (Lolium perenne L.), hybrid ryegrass (Lolium perenne × multiflorum), and tall fescue (Festuca arundinacea Shreb.) cultivars. Methods: Data are reported from unpublished 1980s field-plot experiments for cv. Bb2067 and contemporaneous reference cultivars, sown at two UK locations in 1983 and 1984, and from 397 cultivars of modern forage grass in 13 UK-based breeding experiments sampled in spring 2013. Results: Across sites, years and cuts, cv. Bb2067 had a consistently greater leaf Mg concentration and lower potassium (K) concentration and forage tetany index (FTI) than reference cultivars in the 1980s experiments. Seasonal variation in leaf Mg and K concentrations and FTI were observed in the 1980s experiments, with K concentrations being generally greatest and Mg concentrations smallest in spring. Among modern forage grasses, there was large variation in leaf Mg concentration (up to 6.3-fold) and FTI (up to 2.1-fold), both within and between species, which varied independently of yield. Among a subset of hybrid ryegrass cultivars, there is evidence that the high Mg trait is already present in some modern breeding lines, albeit previously unreported. Conclusions: The variation in leaf Mg concentration and FTI among old and new cultivars shows there is considerable potential to breed forages with improved mineral quality to improve livestock health, potentially without compromising yield or other nutritional traits. Global food wastage equates to about 1.3 billion tons per year, which causes serious environmental impacts. The objective of this study was to evaluate the influences of addition of digestate from food waste in comparison to a synthetic liquid urea ammonium nitrate solution on plant growth, rhizosphere bacterial community composition and diversity, and hyphal abundance of arbuscular mycorrhizal (AM) fungi. Plant and soil samples were collected at 25, 50, and 75 days after seedling emergence. Annual ryegrass growth was significantly increased by both liquid urea ammonium nitrate and digestate, and digestate was just as effective as liquid urea ammonium nitrate. Additionally, digestate (50 kg N ha−1) significantly increased AM fungal hyphae density. Liquid urea ammonium nitrate (50 kg N ha−1) significantly decreased AM fungal hyphae density compared with liquid urea ammonium nitrate (25 kg N ha−1) at DAE 75. Digestate and liquid urea ammonium nitrate applications significantly shifted the bacterial community composition and OTU richness and changed the abundance of microbial C and N-cycling genes, while application rates had no significant effect. Structural equation modeling showed that digestate and UAN addition both directly and indirectly affected bacterial, C and N cycling genes community composition; the indirect effects were related to increased soil NO3− content and reduced pH. This study showed that the use of digestate as a soil amendment can be environmentally effective and can provide a sustainable supply of nutrients that increases soil organic C. Moreover, the use of digestate can readily be incorporated into agricultural practices with potentially less impact on soil microflora diversity and function than conventional fertilizers. Background and aims: Interactions between functional groups of soil fauna and plants are poorly explored although they drive functional processes such as nutrient availability and therefore plant performance. Here, we investigated the separated and combined effects of two collembolan functional groups on soil properties and growth of Lolium perenne, a typical grass species from temperate grasslands. Methods: Under microcosm conditions, we established four treatments based on the presence and combination of two collembolan functional groups: (1) control without Collembola; (2) epedaphic species; (3) euedaphic species; (4) epedaphic + euedaphic species. After five months of experiment, we measured the effect of those treatments on both nutritional and morphological variables of L. perenne and on soil properties. Results: Collembolan presence stimulated plant performance. Individuals of L. perenne growing with euedaphic species presented higher numbers of leaves and nutrient contents compared with individuals growing with epedaphic species. Further, the combination of both collembolan functional groups enhanced plant performance and soil nutrient availability, demonstrating that a functionally diversified soil fauna assemblage cause overyielding of ecological processes. Grass endophytes have been shown to confer enhanced environmental resilience to symbiont cultivars with reports of modified growth. If inoculating with an endophyte (E+) made an accession morphologically distinct from its registered endophyte free (E−) accession, there could be protection and ownership issues for testing authorities and breeders. This study investigated if, in official Plant Breeders Rights (PBR) field trials, the morphological characteristics of E+and E− accessions of perennial ryegrass and tall fescue cultivars were sufficiently modified to designate them as mutually distinct and also distinct from their definitive accessions (Def), held by the testing authorities. Testing perennial ryegrass on 17 characters at 2 sites generated 48,960 observations and for tall fescue on 9 characters at 1 site, 12,960 observations (each for 3 accessions of 4 cultivars × 60 plants × 2 growing cycles). Distinctness required a p < 0.01 difference in a single character from the combined over years analysis (COYD). A few significant differences were recorded between E− and E+accessions. Cultivar Carn E+ was smaller than Carn E− for Infloresence Length (p < 0.01) in both years but COYD analysis (p < 0.05) was insufficient to declare distinctiveness. Overall, the number of observed differences between E−/E+ accessions was less or similar to the number expected purely by chance. In contrast, comparisons between Def and E− or E+ accessions showed a number of significant differences that were substantially more numerous than expected by chance. These results showed no conclusive evidence of endophyte inclusion creating false PBR distinctions but unexpectedly, several E− and E+ accessions were distinguished from their official definitive stock. The symbiotic Epichloë festucae var. lolii endophyte produces alkaloids which can provide its host grass, perennial ryegrass (Lolium perenne L), with a selective advantage in both natural and agricultural managed ecosystems. This study focuses on understanding the alkaloid concentrations that occur in endophyte-infected perennial ryegrass during the early establishment phase. In a glasshouse experiment fungal alkaloid concentrations (peramine, lolitrem B, ergovaline, and epoxy-janthitrems) were measured in perennial ryegrass seedlings infected with E. festucae var. lolii proprietary strains AR1, AR37, NEA2, and NZ common toxic for 69 days after sowing. The endophyte becomes metabolically active, starting alkaloid production, as early as 6 days after sowing. Alkaloid concentrations peaked in 8- to 10- day-old seedlings due to a seedling growth slowdown. This study provides data showing that the loss of insect protection in endophyte-infected seedlings is linked to a reduction in chemical defence after seed-stored, maternally synthesised alkaloids are diluted by seedling dry matter accumulation. Perennial ryegrass (Lolium perenne), one of the most widely used forage and cool-season turfgrass worldwide, has a breeding history of more than 100 years. However, the current draft genome annotation and transcriptome characterization are incomplete mainly because of the enormous difficulty in obtaining full-length transcripts. To explore the complete structure of the mRNA and improve the current draft genome, we performed PacBio single-molecule long-read sequencing for full-length transcriptome sequencing in perennial ryegrass. We generated 29,175 high-confidence non-redundant transcripts from 15,893 genetic loci, among which more than 66.88% of transcripts and 24.99% of genetic loci were not previously annotated in the current reference genome. The re-annotated 18,327 transcripts enriched the reference transcriptome. Particularly, 6709 alternative splicing events and 23,789 alternative polyadenylation sites were detected, providing a comprehensive landscape of the post-transcriptional regulation network. Furthermore, we identified 218 long non-coding RNAs and 478 fusion genes. Finally, the transcriptional regulation mechanism of perennial ryegrass in response to drought stress based on the newly updated reference transcriptome sequences was explored, providing new information on the underlying transcriptional regulation network. Taken together, we analyzed the full-length transcriptome of perennial ryegrass by PacBio single-molecule long-read sequencing. Perennial ryegrass (Lolium perenne L.) provides a cost-effective forage species for New Zealand farms. Vegetative persistence (maintained herbage growth and survival without reseeding) is an important trait for decreasing costs of pasture establishment. Breeding for vegetative persistence is difficult because of a deficiency of long-term trials to observe the complex interactions between plant genotype and environment. Over time in a long-term trial, a shift in the genetic mean of a cultivar could occur as plants with genetically enhanced persistence traits survive. This study aimed to identify a shift in the genetic mean of two cultivars sown in a long-term trial in Hawkes Bay, New Zealand. Plots of 1 m rows were assessed in a trial for 13 months to compare changes in additive genetic variation ( σ a 2 ), narrow-sense heritability ( h n 2 ), and trait associations of half-sib (HS) populations generated from "Persistent" plants and commercially bought seed of "Original" cultivars Grasslands Samson and Commando. Significant (P < 0.05) σ a 2 and high levels of h n 2 for some populations were identified for scores of herbage growth, leaf width, plant habit and aftermath heading. This study found genetic shifts over time were cultivar specific. Traits that were different between Persistent and Original populations were previously associated with decreased pasture intake. The bioeconomic models developed in Weed Science consider the effect of a species on the crop. Bioeconomic models accounting for species interactions within the weed community might represent more realistic approximation to the economic outcome of weed control options in Mediterranean cereal crops. A bioeconomic model, based in community dynamics, competition and economic submodels, was developed and used to investigate the long-term agronomic, economic and environmental consequences of using herbicide-based strategies for the control of winter wild oats (Avena sterilis) and rigid ryegrass (Lolium rigidum) in winter wheat. The results of simulations indicated that different winner strategies can be postulated depending on selection criterion. Based on purely economic outcome, the best strategy was the application of half-dose herbicide to both species whereas if the goal is to achieve a long-term economically viable and environmental friendly decision making, then the strategy chosen would be the one based on a full-dose herbicide application for the control of L. rigidum and half-dose for control of A. sterilis. The sensitivity of the model to variation in economic and competition parameters was checked. The most sensitive model parameters were the potential yield and the wheat price. The gray field slug, Deroceras reticulatum, is a key pest of seed crops. There is an increasing interest in developing new chemical controls due to limited management options. Essential oils, being non-toxic to humans and exempt from pesticide registration and residue tolerance requirements under US federal law (Sect. 25(b) of the Federal Insecticide, Fungicide, and Rodenticide Act), could be safe and easily implementable alternatives. In this study, the most toxic essential oils to D. reticulatum adults of 13 plant-derived essential oils and one synthetic toxin (caffeine) were determined based on Lethal Concentration 50 values produced in a laboratory Petri dish bioassay. Thyme, spearmint, and pine oil were the most lethal, causing 50% mortality of D. reticulatum at concentrations of 0.148, 0.153, and 0.176% (v/v), respectively. Thyme and spearmint oil were then tested in a greenhouse microcosm experiment and a separate phytotoxicity assessment. In the greenhouse, slugs were added to containers planted with annual ryegrass (Lolium multiflorum) and the containers were sprayed with either 0.5% (v/v) essential oil emulsion, Slug-Fest (industry standard molluscicide), surfactant control at 1% (v/v), or water control. Both oils caused 97.5% mortality of slugs, performing comparably to metaldehyde. Phytotoxic effects were assessed by spraying oils on seedlings and adult plants of two cultivars each of perennial ryegrass (Lolium perenne) and tall fescue (Festuca arundinacea). No definitive signs of phytotoxicity were observed upon visual inspection, and there were no differences in chlorophyll content or biomass between treated and untreated plants. Background and aims: Low temperature stress is a common hazard during plant growth. Endophyte infection has been shown to increase cold tolerance in host plants. Many Lolium perenne cultivars contain low to moderate levels of endophyte. This study was done to explore cultivar improvement by segregation of endophyte containing individuals from the original cultivar to create a high endophyte subpopulation. Methods: Endophyte-infected plants were segregated over the first 3 years to produce high-endophyte subpopulation, and field and greenhouse experiments were carried out in the forth and fifth to determine the cold tolerance of the L. perenne subpopulation with high endophyte infection rates (N), the parent (F), the control endophyte-free subpopulation (E) and the control local variety (L). Results: (1) After 3 years of screening, high endophyte infection rates in the tillers and seeds of plants were still observed (96.5%), and agronomic traits (crown width, plant height, panicle number, withering, regreen-up, the growth cycle and the over-wintering rate) was also improved with increased Epichloë colonization of host plant. (2) The subpopulation with high endophyte infection rates and improved agronomic traits had better cold tolerance than the parent, the control endophyte-free subpopulation and the control local variety. The possible mechanisms by which high endophyte infection enhances cold resistance in the field include increased root system, increased the over-wintering rate, reduced regrowth periods with the sowing date being October 15th. (3) The high-endophyte subpopulation significantly increased SOD, POD, CAT, and APX activities at 0, 5, and 10 °C by 11.8%–44.6%, compared with the parent population. Conclusions: The subpopulation had a high endophyte infection rate, improved agronomic traits and higher enzymatic activities. Aims: Deep-rooted agricultural crops can potentially utilize deep soil moisture to reduce periods where growth is water limited. Chicory (Cichorium intybus L.) is a deep-rooted species, but the benefits of deep roots to water uptake has not been studied. The aim of this study was to investigate the value of deep roots (>2 m) under topsoil water limitation. Methods: Chicory grown in 4 m deep soil-filled rhizotrons was exposed to either topsoil drought or resource competition from the shallow-rooted species ryegrass (Lolium perenne L.) and black medic (Medicago lupulina L.). The effect on deep water uptake was assessed using non-destructive measurements of roots, soil water and tracers. Results: Water uptake occurred below 1.7 m depth in 2016, and below 2.3 m depth in 2017 and contributed significantly to chicory water use. However, neither surface soil drying nor intercropping increased deep water uptake to relieve water deficit in the shoots. Conclusion: Chicory benefits from deep-roots during drought events, as it acceses deep soil moisture unavailable to more shallow rooted species, yet deep water uptake was unable to compensate for the reduced topsoil water uptake due to soil drying or crop competition. Aims: We tested if chicory acquires nutrients from soil layers down to 3.5 m depth and whether the deep nutrient uptake increases as a result of drought or intercropping with ryegrass and black medic. We also tested whether application of the trace elements Cs, Li, Rb, Sr and Se, as tracers, result in similar uptake rates. Methods: The methodological tests were carried out in a pilot experiment where the tracers were applied to 1 m depth in lucerne and red beet grown in tube rhizotrons. The dynamics of deep nutrient uptake in chicory was studied in large 4 m deep rhizoboxes. A drought was induced when roots had reached around 2 m depth. Results: Chicory acquired 15N from 3.5 m depth and trace element tracers from 2.3 m depth. We found no compensatory tracer uptake with depth during drought. We found some indications of a compensatory tracer uptake from 2.3 and 2.9 m depth in intercropped chicory. Application of equimolar amounts of trace elements resulted in similar excess tracer concentrations within species. Conclusion: Chicory demonstrates nutrient uptake from below 3 m but does not increase deep nutrient uptake as a response to limited topsoil nutrient availability induced by drought or intercropping. Defoliation often has little effect on the percent plant root length colonised by arbuscular mycorrhizal (AM) fungi, and this has been interpreted as a lack of support for the carbon limitation hypothesis. We performed an experiment with three levels of repeated defoliation (none, every 3 weeks and weekly) of Lolium rigidum growing in pasture soil, and assessed colonisation of roots by naturally occurring AM fungi over 4 months in the glasshouse. Surprisingly, the percent root length colonised by AM fungi increased with defoliation. We also assessed root mass and the length of colonised root to obtain an estimate of the quantity of mycorrhizal root. As expected, both root mass and length of mycorrhizal root decreased with defoliation, as did soluble sugars in the roots. Thus, increasing the frequency of defoliation reduced mycorrhizas consistent with the carbon-limitation hypothesis. Such a decline in mass of mycorrhizal root could contribute to reducing soil biological fertility in pastures over time. [ABSTRACT FROM AUTHOR]

Aims: Roots are vital organs for plants, but the assessment of root traits is difficult, particularly in deep soil layers under natural field conditions. A popular technique to investigate root growth under field or semi-field conditions is the use of minirhizotrons. However, the subsequent manual quantification process is time-consuming and prone to error. Methods: We developed a multispectral minirhizotron imaging system and a subsequent image analysis strategy for automated root detection. Five wavelengths in the visible (VIS) and near-infrared (NIR) spectrum are used to enhance living roots by a multivariate grouping of pixels based on differences in reflectance; background noise is suppressed by a vesselness enhancement filter. The system was tested against manual analysis of grid intersections for both spring barley (Hordeum vulgare L.) and perennial ryegrass (Lolium perenne L.) cultivars at two time-points. The images of living roots were captured in wet subsoil conditions with dead roots present from a previous crop. Results: Under the soil conditions used in the study, NIR reflectance (940 nm), provided limited ability to separate between rhizosphere components, compared to reflectance in the violet and blue light spectrum (405 nm and 450 nm). Multivariate image analysis of the spectral data, combined with vesselness enhancement and thresholding allowed for automated detection of living roots. Automated image analysis largely replicated the root intensity found during manual grid intersect analysis of the same images. Although some misclassification occurred, caused by elongated structures of dew and chalkstone with similar reflectance pattern as living root, the system provided similar or in some cases improved detection of genotypic differences in the total root length within each tube. Conclusion: The multispectral imaging system allows for automated detection of living roots in minirhizotron studies. The system requires considerably less time than traditional manual recording using grid intersections. Background and aims: Microbial denitrification is the primary driver of nitrogen losses from the plant-soil system and the key process for the closure of the global N cycle. All major controls of denitrification might be directly or indirectly affected by plants. However, there is a lack of research of the direct effects of plants on soil denitrification and how this effect might be mediated by soil properties. This study assesses the effect of three common crop species and two agricultural soils on denitrification potentials. Methods: We conducted a factorial experiment under controlled conditions to analyze the effects of (1) different plant species (barley, wheat or ryegrass), (2) two different soils (texture/ SOC) and (3) two different soil moisture levels on Denitrification Enzyme Activity (DEA) in bulk and rhizosphere soil. Results: The SOC richer clay loam soil showed on average higher DEA (+81%) compared to the SOC poorer silty loam soil. All three plants were found to stimulate denitrification with significant differences between certain species: rye grass (+92% ± 14%) ≥ barley (+75% ± 26%) ≥ wheat (+50% ± 19%). Conclusions: DEA in agricultural soils is interactively controlled by plant species and soil type with an overall stimulating effect of plants on the denitrification potential. Laser-induced breakdown spectroscopy (LIBS) is an analytical technique that can be used to facilitate variable rate fertilizer application, potentially increasing yield, reducing costs and reducing environmental side effects of nutrient loss. LIBS can give real-time information about macro and micro nutrients with little to no sample preparation. The study reported in this paper investigated whether LIBS can predict nutrient levels of fresh and dried pelletized pasture and what the limitations are. Spectra were acquired in air and under argon. Partial least square regression was used to build models for each macro and micro nutrient. The best results were for potassium, sodium and manganese with root mean square errors of cross-validation of 0.20, 0.029 and 0.0008 wt%, respectively, coefficient of determination of 0.92, 0.93 and 0.90, limits of detection of 0.99, 0.11 and 0.0027 wt%, and precisions of 0.30, 0.042 and 0.0012 wt%. LIBS can be used to assess nutrient levels of fresh pasture. Reducing the shot-to-shot variation will lead to improved calibrations. [ABSTRACT FROM AUTHOR]

Key message: Exploitation of data from a ryegrass breeding program has enabled rapid development and implementation of genomic selection for sward-based biomass yield with a twofold-to-threefold increase in genetic gain.Abstract: Genomic selection, which uses genome-wide sequence polymorphism data and quantitative genetics techniques to predict plant performance, has large potential for the improvement in pasture plants. Major factors influencing the accuracy of genomic selection include the size of reference populations, trait heritability values and the genetic diversity of breeding populations. Global diversity of the important forage species perennial ryegrass is high and so would require a large reference population in order to achieve moderate accuracies of genomic selection. However, diversity of germplasm within a breeding program is likely to be lower. In addition, de novo construction and characterisation of reference populations are a logistically complex process. Consequently, historical phenotypic records for seasonal biomass yield and heading date over a 18-year period within a commercial perennial ryegrass breeding program have been accessed, and target populations have been characterised with a high-density transcriptome-based genotyping-by-sequencing assay. Ability to predict observed phenotypic performance in each successive year was assessed by using all synthetic populations from previous years as a reference population. Moderate and high accuracies were achieved for the two traits, respectively, consistent with broad-sense heritability values. The present study represents the first demonstration and validation of genomic selection for seasonal biomass yield within a diverse commercial breeding program across multiple years. These results, supported by previous simulation studies, demonstrate the ability to predict sward-based phenotypic performance early in the process of individual plant selection, so shortening the breeding cycle, increasing the rate of genetic gain and allowing rapid adoption in ryegrass improvement programs. Aims: Belowground legume nitrogen (N) composed of roots and rhizodeposition is an important N input to soils, but published data of belowground N vary broadly, probably due to extrapolation from short-term experiments and dissimilar growing conditions. We quantified belowground N inputs of red clover (Trifolium pratense L.) during two consecutive years in a clover-grass sward along a soil nutrient availability gradient.Methods: We established a red clover-perennial ryegrass (Lolium perenne L.) model sward in microplots located in field plots of the DOK experiment, which has a 33-year history of organic and conventional cropping, resulting in a soil nutrient availability gradient. Four treatments were examined: the zero fertilisation control, bio-organic with half and full dose manure application, and the conventional system with mineral fertilisation at full dose. We studied the development of clover aboveground and belowground N using multiple pulse 15N urea leaf labelling.Results: Belowground clover N increased over time and with rising nutrient availability and was proportional to aboveground clover N at all times. Belowground clover N amounted to 40% of aboveground clover N during two consecutive years, irrespective of the nutrient availability status. Belowground clover N development was initially dominated by fast root growth, followed by enhanced root turnover during the second year. Potassium availability limited clover growth and total N accumulation in treatments with low nutrient availability.Conclusions: Belowground red clover N inputs could be estimated from aboveground N by a constant factor of 0.4, regardless of the nutrient availability and cultivation time. Root turnover led to a distinct absolute increase of N rhizodeposition over time.
