Abstract Genomic variation impacts on cellular networks by affecting the abundance (e.g., protein levels) and the functional states (e.g., protein phosphorylation) of their components. Previous work has focused on the former, while in this context, the functional states of proteins have largely remained neglected. Here, we generated high‐quality transcriptome, proteome, and phosphoproteome data for a panel of 112 genomically well‐defined yeast strains. Genetic effects on transcripts were generally transmitted to the protein layer, but specific gene groups, such as ribosomal proteins, showed diverging effects on protein levels compared with RNA levels. Phosphorylation states proved crucial to unravel genetic effects on signaling networks. Correspondingly, genetic variants that cause phosphorylation changes were mostly different from those causing abundance changes in the respective proteins. Underscoring their relevance for cell physiology, phosphorylation traits were more strongly correlated with cell physiological traits such as chemical compound resistance or cell morphology, compared with transcript or protein abundance. This study demonstrates how molecular networks mediate the effects of genomic variants to cellular traits and highlights the particular importance of protein phosphorylation.

Abstract Action-oriented, transformative, and transdisciplinary approaches are increasingly heralded as promising tools that enable researchers not only to produce new knowledge about a situation, but also to actively engage in tackling it; however, such approaches raise critical questions about the methodological implications and conflicts involved in pursuing multiple objectives concurrently. This article seeks to advance this debate by examining the methodological implications of pursuing both knowledge production-oriented (epistemic) and action-oriented (transformative) objectives in urgent and uncertain situations. It asks how far a transformative mixed methods research design can leverage the potential of research to achieve multiple objectives in times of crisis. This methodological inquiry is based on a transformative mixed methods study on housing conditions and well-being which was initiated during the first COVID-19 lockdown in Switzerland. The study was composed of a country-wide survey, mobile crowdsourcing, interviews and Citizen Think Tanks. The results of this methodological reflection illustrate how this research design made it possible to (i) explore the effects of the crisis on domestic living spaces, (ii) engage with the crisis in a joint research space, and (iii) stimulate empowerment through reflection and mutual learning. These insights suggest the multidimensional orientation of the transformative mixed methods approach is appropriate for acting upon urgent crises. However, it challenges core methodological values and research constellations including (i) tackling unequal engagement opportunities, (ii) navigating social and epistemic control, and (iii) paying attention to situatedness and positionality. The article concludes that, in anticipation of future crises, favorable conditions for multi-targeted, collaborative research need to be fostered, both on the institutional and on personal levels. These conditions should enable fast and adequate team formation, as well as reflection and negotiation of positionalities, and divergent interests and objectives, of both scientists and citizens.

Chronic non-bacterial osteomyelitis (CNO) is caused by aseptic inflammation of bones, primarily driven by the innate immune system. CNO may display different clinical presentations (acute vs chronic, uni- vs multifocal) and is accompanied by other inflammatory disorders in up to a third of patients. Once considered a rare disorder, it has become clear that many patients were underdiagnosed. With increasing awareness and the development of total-body MRI protocols, CNO recognition and diagnosis have greatly improved. Our knowledge of the clinical manifestations and outcomes of CNO has been refined in recent years, especially thanks to the recruitment of large international series. Similarly, new insights into the pathogenesis have been gained by the development of mice models and identification of rare monogenic diseases that resemble CNO. Unfortunately, these advances have not been paralleled in the therapeutic management. In the absence of prospective controlled trials, therapeutic strategies still rely on low-level evidence studies. About half of the patients respond to first-line therapies, but a more refractory and/or chronic disease course requires additional treatments. This narrative review aims to provide the practicing physician with an update on CNO pathogenesis, clinical presentation, associated inflammatory conditions, and diagnostic investigations, and includes a concise summary of current therapeutic recommendations. Conclusion: While major progresses have been made in the recognition and management of CNO, significant challenges remain, in particular regarding the treatment of refractory patients, and those with associated inflammatory disorders. What is Known: • Many physicians caring for children will encounter patients suffering of (suspected) CNO. CNO diagnosis requires exclusion of numerous conditions included in the differential diagnosis, which may be challenging. What is New: • We provide an updated review of recent findings in the field CNO, including imaging and diagnostic strategies, associated inflammatory diseases and long-term outcomes data. • We focus particularly on the challenges encountered by clinicians in the diagnosis and treatment of these patients. • We highlight knowledge gaps in the understanding and treatment of CNO, that should stimulate future research. Replication Protein A (RPA) plays a pivotal role in DNA replication by coating and protecting exposed single-stranded DNA, and acting as a molecular hub that recruits additional replication factors. We demonstrate that archaeal RPA hosts a winged-helix domain (WH) that interacts with two key actors of the replisome: the DNA primase (PriSL) and the replicative DNA polymerase (PolD). Using an integrative structural biology approach, combining nuclear magnetic resonance, X-ray crystallography and cryo-electron microscopy, we unveil how RPA interacts with PriSL and PolD through two distinct surfaces of the WH domain: an evolutionarily conserved interface and a novel binding site. Finally, RPA is shown to stimulate the activity of PriSL in a WH-dependent manner. This study provides a molecular understanding of the WH-mediated regulatory activity in central replication factors such as RPA, which regulate genome maintenance in Archaea and Eukaryotes. The single stranded DNA binding protein RPA plays a pivotal role in DNA replication. The archaeal RPA hosts a WH domain that interacts with the DNA primase and the replicative polymerase PolD. In the context of the oral cavity, an organic layer known as the mucosal pellicle (MP) adheres to the surface of the oral epithelium, playing a pivotal role in lubricating and safeguarding oral tissues. The formation of the MP is driven by interactions between a transmembrane mucin known as MUC1, located on the oral epithelium, and salivary secreted mucin, namely MUC5B and MUC7. This study aimed to investigate the function of MUC1 and the influence of its structure on MP lubrication properties. We proposed a novel methodology to study oral lubrication based on four different models of oral epithelium on which we conducted in vitro tribological studies. These models expressed varying forms of MUC1, each possessing on of the distinct domain constituting the mucin. Mechanical parameters were used as indicators of lubrication efficiency and, consequently, of the role played by MUC1 in oral lubrication. The results from the tribological tests revealed that the presence of full MUC1 resulted in enhanced lubrication. Furthermore, the structure of MUC1 protein drive the lubrication. In conclusion, the mechanical tests conducted on our epithelium models demonstrated that MUC1 actively participates in epithelium lubrication by facilitating the formation of the MP. Time series data are pivotal in diverse fields such as finance, meteorology, and health data analysis. Accurate analysis of these data is crucial for identifying temporal trends and making informed decisions. However, frequent occurrences of missing values, often due to device failures or data collection errors, pose a significant challenge. In this work, we introduce TS-Pothole, a method for imputing missing values in univariate time series. This method leverages cyclic pattern analysis and a recursive strategy to handle univariate datasets in which missing values are distributed both continuously and randomly. We evaluate TS-Pothole on four real-world datasets representing different configurations of missing values, and assess its performance in terms of accuracy and execution speed. In particular, we compare our approach with state-of-the-art methods, such as GANs and autoencoders. Our experiments show that TS-Pothole outperforms such methods by providing more accurate (up to 1.5 times) and faster (up to 2 times) imputations, even as the proportion of missing data increases, representing the best alternative in handling univariate time series with missing values. Anastomotic leak occurrence is a severe complication after colorectal surgery. Considering the difficulty of treating these leaks and their impact on patient care, there is a strong need for an efficient prevention strategy. We evaluated a combination of extracellular vesicles (EVs) from rat adipose-derived stromal cells with a thermoresponsive gel, Pluronic® F127 (PF-127) to prevent anastomotic leaks. The pro-regenerative and immunomodulatory potencies of EVs are assessed in vitro. In vivo efficacy are assessed in rat with a colonic anastomosis model after irradiation. Endoscopic, anatomical and histological data show a consistent effect of EVs + gel on the healing of colonic anastomosis. These results are illustrated by a smaller anastomotic ulcer size, less fibrosis and less inflammatory infiltrations in the EVs + gel group. This multi-modal investigation is the first to point-out the translational potential of EVs combined with PF-127 for the healing of high-risk colorectal anastomosis. Extracellular vesicles from adipose-derived rat stromal cells embedded in a thermoresponsive gel display pro-regenerative and immunomodulatory effects, resulting in the healing of colorectal anastomosis in rats. The Controls Over Mesopelagic Interior Carbon Storage (COMICS) cruise DY086 took place aboard the RRS Discovery in the South Atlantic during November and December, 2017. Physical, chemical, biogeochemical and biological data were collected during three visits to ocean observatory station P3, off the coast of South Georgia, during an austral spring bloom. A diverse range of equipment including CTD-rosette, Acoustic Doppler Current Profiler (ADCP), net deployments, marine snow catchers (MSCs), Stand Alone Pump System (SAPS) and PELAGRA Sediment Traps were used to produce a comprehensive, high-quality dataset. The data can provide excellent insight into regional biological carbon pump (BCP) processes; it is recommended for use by observational scientists and modellers to enhance understanding of ecosystem interactions relating to mesopelagic carbon storage. During their final transformation, insects emerge from the pupal case and deploy their wings within minutes. The wings deploy from a compact origami structure, to form a planar and rigid blade that allows the insect to fly. Deployment is powered by a rapid increase in internal pressure, and by the subsequent flow of hemolymph into the deployable wing structure. Using a combination of imaging techniques, we characterize the internal and external structure of the wing in Drosophila melanogaster, the unfolding kinematics at the organ scale, and the hemolymph flow during deployment. We find that, beyond the mere unfolding of the macroscopic folds, wing deployment also involves wing expansion, with the stretching of epithelial cells and the unwrinkling of the cuticle enveloping the wing. A quantitative computational model, incorporating mechanical measurements of the viscoelastic properties and microstructure of the wing, predicts the existence of an operating point for deployment and captures the dynamics of the process. This model shows that insects exploit material and geometric nonlinearities to achieve rapid and efficient reconfiguration of soft deployable structures. Upon emerging from their pupal case, insects swiftly deploy their wings from a compact origami-like structure to a fully extended blade. Augmenting the portfolio of therapeutics for type 2-driven diseases is crucial to address unmet clinical needs and to design personalized treatment schemes. An attractive therapy for such diseases would consist in targeting the recruitment of T helper 2 (Th2) lymphocytes to inflammatory sites. Herein, we show the degradation of filamins (FLN) a and b by the ASB2α E3 ubiquitin ligase as a mechanism sustaining Th2 lymphocyte functions. Low levels of FLNa and FLNb confer an elongated shape to Th2 lymphocytes associated with efficient αVβ3 integrin-dependent cell migration. Genes encoding the αVβ3 integrin and ASB2α belong to the core of Th2-specific genes. Using genetically modified mice, we find that increasing the levels of FLNa and FLNb in Th2 lymphocytes reduces airway inflammation through diminished Th2 lymphocyte recruitment in inflamed lungs. Collectively, our results highlight ASB2α and its substrates FLNa and FLNb to alter Th2 lymphocyte-mediated responses. Filamin proteins are degraded by ASB2α E3 ubiquitin ligase and may affect T cell function. Here the authors show that increased levels of Filamin A and Filamin B in Th2 cells reduces allergic inflammation in mouse models through reduced Th2 cell recruitment into inflamed lungs. Séraphin (1747–1800) is considered the founder of shadow puppetry in France. This "facetious hunchback", well known to Parisians in the late 18th century, ran his own theater until his death in 1800 at the age of 53. His deformity seems to have left its mark on popular memory, and was an integral part of the "Théâtre Séraphin". Part of his skeleton was preserved in the Dupuytren Museum in Paris. In 1897, it was described as a "particular kind of ankylosis, probably different from ordinary kyphosis". Then, in 1899, Dr. André Léri, a pupil of Pierre Marie, hypothesized from skeletal analysis that Séraphin must have suffered from rhizomelic spondylosis (the first french name for ankylosing spondylitis). In this article, we present the recent analysis of his skeleton, restored at the Dupuytren Museum, as well as old documents, a graphic representation and texts that allow us to make the diagnosis of ankylosing spondylitis. Karst aquifers are a significant source of drinking water and highly vulnerable to pollution and microbial contamination. Microbiological regulations for the quality of drinking water mostly focus on bacterial levels and lack guidance concerning fungal contamination. Moreover, there is no standardised microbial analysis methodology for identifying fungi in water. Our main objective was to establish the most effective culture and identification methodology to examine yeast diversity in karst waters. We assessed the comparative efficacy of four culture media (CHROMagar Candida, dichloran glycerol 18% [DG18], dichloran rose Bengal chloramphenicol [DRBC], and SYMPHONY agar) for yeast isolation from karst water samples. Furthermore, we investigated the comprehensiveness of databases used in MALDI-TOF mass spectrometry (MALDI-TOF MS) for identifying environmental yeast species. In total, we analysed 162 water samples, allowing the identification of 2479 yeast isolates. We demonstrate that a combination of four culture media, each with distinct specifications, more efficiently covers a wide range of yeast species in karst water than a combination of only two or three. Supplementation of a MALDI-TOF MS database is also critical for analysing environmental microbial samples and improved the identification of yeast biodiversity. This study is an initial step towards standardising the analysis of fungal biodiversity in karst waters, enabling a better understanding of the significance of this environmental reservoir in relation to public health. Hereditary C1q deficiency (C1QDef) is a rare monogenic disorder leading to defective complement pathway activation and systemic lupus erythematosus (SLE)-like manifestations. The link between impairment of the complement cascade and autoimmunity remains incompletely understood. Here, we assessed type 1 interferon pathway activation in patients with C1QDef. Twelve patients with genetically confirmed C1QDef were recruited through an international collaboration. Clinical, biological and radiological data were collected retrospectively. The expression of a standardized panel of interferon stimulated genes (ISGs) in peripheral blood was measured, and the level of interferon alpha (IFNα) protein in cerebrospinal fluid (CSF) determined using SIMOA technology. Central nervous system (encompassing basal ganglia calcification, encephalitis, vasculitis, chronic pachymeningitis), mucocutaneous and renal involvement were present, respectively, in 10, 11 and 2 of 12 patients, and severe infections recorded in 2/12 patients. Elevated ISG expression was observed in all patients tested (n = 10/10), and serum and CSF IFNα elevated in 2/2 patients. Three patients were treated with Janus-kinase inhibitors (JAKi), with variable outcome; one displaying an apparently favourable response in respect of cutaneous and neurological features, and two others experiencing persistent disease despite JAKi therapy. To our knowledge, we report the largest original series of genetically confirmed C1QDef yet described. Additionally, we present a review of all previously described genetically confirmed cases of C1QDef. Overall, individuals with C1QDef demonstrate many characteristics of recognized monogenic interferonopathies: particularly, cutaneous involvement (malar rash, acral vasculitic/papular rash, chilblains), SLE-like disease, basal ganglia calcification, increased expression of ISGs in peripheral blood, and elevated levels of CSF IFNα. Atmospheric simulation chambers can become valuable tools for studying the impact of different air pollutants and sources on preclinical models. We developed a novel experimental approach for the generation of realistic polluted atmospheres, by coupling, for the first time, two advanced chambers, the FORTH (Foundation for Research & Technology—Hellas) mobile simulation chamber and the CESAM (Chamber for Experimental Multiphase Atmospheric Simulation) chamber, to continuously generate air pollution levels that represent a variety of urban atmospheric conditions heavily influenced by biomass burning (BB). The combination of the two chambers enables the production of stable and representative aerosols, allowing for long-term exposure studies on preclinical models (i.e., healthy, and wild type mice) under controlled conditions. This work describes the coupling methodology, the operational conditions of the FORTH and CESAM chambers and the integration of the animal exposure devices to this dual chamber set-up. The protocols applied for the continuous production (here up to 72 h) of both fresh and aged BB emissions and several other pollutants are described. The range of aerosol concentrations, compositions, and properties achieved in these experiments are summarized. The aged BB aerosol had up to twice the oxidative potential (OP) of the fresh BB emissions. The study revealed significant changes in aerosol composition during the photochemical processing of the BB emissions, with the oxygen to carbon (O:C) ratio of aged BB increasing by 33% compared to the fresh. Notable volatile organic compounds (VOC) emissions, such as formaldehyde and acetonitrile, and their levels were also highlighted. The main objective of this work is to provide useful insights for the future development of robust protocols for effective long-term exposure (several days or weeks) of preclinical models under controlled and stable conditions. Optimization problems constrained by partial differential equations (PDEs) naturally arise in scientific computing, as those constraints often model physical systems or the simulation thereof. In an implicitly constrained approach, the constraints are incorporated into the objective through a reduced formulation. To this end, a numerical procedure is typically applied to solve the constraint system, and efficient numerical routines with quantifiable cost have long been developed for that purpose. Meanwhile, the field of complexity in optimization, that estimates the cost of an optimization algorithm, has received significant attention in the literature, with most of the focus being on unconstrained or explicitly constrained problems. In this paper, we analyze an algorithmic framework based on quadratic regularization for implicitly constrained nonlinear least squares. By leveraging adjoint formulations, we can quantify the worst-case cost of our method to reach an approximate stationary point of the optimization problem. Our definition of such points exploits the least-squares structure of the objective, and provides new complexity insights even in the unconstrained setting. Emamectin benzoate based insecticide and glyphosate based herbicides are widely used in developing countries for agricultural purposes. The problem lies in the total lack of knowledge of data on their uses as well as the absence of scientific data on the potential effects on local biodiversity. This study evaluates the acute and chronic effects of two commercial formulations EMACOT® and SUNPHOSATE-G® on juveniles of a prawn species (Macrobrachium macrobrachion) native of large watersheds in Benin. Juveniles with an average weight of 1.49 ± 0.2 g were subjected to six and seven lethal doses of EMACOT® and SUNPHOSAT-G® respectively. Thus, swimming behavior, loss of mobility and mortality were monitored every hour for 72 h. Then, sub-lethal effects on molting effectiveness, weight and size gain as well as survival were evaluated on juveniles individually exposed to nominal concentrations of 0.06 and 0.11 mg.L-1 of EMACOT, and 13.99 and 27.98 mg.L-1 of SUNPHOSATHE-G. The results showed that the LC50-72 h of the herbicide formulation on juveniles is 279.76 mg.L-1 which corresponds to 211.78 mg.L-1 of glyphosate. That of the insecticide is 1.14 mg.L-1 corresponding to 0.06 mg.L-1of emamectin benzoate. As for the chronic effects, both concentrations of emamectin benzoate prevented the prawn from molting (1 molt at the start of the experiment against 3 molts for the control subjects in 28 days). As a result, weight gain in these two treatments was very low (0.06 and 0.19 g, respectively) compared to 0.66 g in the controls. For glyphosate, findings with 13.99 mg.L-1 are similar to those of both doses of emamecin benzoate. On the other hand, no significant effect was observed on the effectiveness of molts within 27.98 mg.L-1 treatment (3 molts in 28 days). The weight gain was 0.11 g and 0.46 g respectively for both glyphosate sub-lethal doses. The results of this study indicate a significant sensitivity of the prawn juveniles to emamectin benzoate (EMACOT) and a relatively low sensitivity to glyphosate (SUNPHOSATE-G). But the risk is significant when these two pesticides are used in the flood plains of large rivers. Background: International consensus on classifications of appendiceal mucinous neoplasms (AMNs) and associated pseudomyxoma peritonei (PMP) have been carefully made but clinicopathological associations supporting decision making remain scarce. Objective: This study aimed to assess interdependence between AMNs and PMP and provide directions for clinical management. Methods: This two-center retrospective cohort study reviewed patients with PMP treated with cytoreductive surgery and hyperthermic intraperitoneal chemotherapy between 2005 and 2021. The primary objective was to reassess histopathologic grade of AMNs and PMP according to the Peritoneal Surface Oncology Group International classification and to establish its interdependence. Secondary outcomes were recurrence rate, PMP grade progression, ovarian involvement, and overall survival (OS). Results: Of 105 patients included, 78 (74.3%) had low-grade AMNs as the primary tumor, 8 (7.6%) had high-grade AMNs, 7 (6.7%) had mucinous adenocarcinoma (MAC), 1 (0.9%) had MAC with signet ring cells (SRC), and 11 (10.5%) had unidentified tumors. Overall, 11 patients (10.5%) had no PMP, 21 (20.0%) had acellular mucin, 56 (53.3%) had low-grade PMP, 12 (11.4%) had high-grade PMP, and 5 (4.8%) had PMP-SRC. In 11 cases (13.3%), AMNs and matching PMP grade differed. Over a 16-year follow-up, recurrence occurred in 31.8%, with three cases showing histopathologically changed PMP. Ovarian involvement was observed in 43/65 females (66.2%). Median OS was 13.8 years, and 5-year OS rates were 100%, 74.4%, 44.4%, and 20% for acellular mucin, low-grade PMP, high-grade PMP and PMP-SRC, respectively (p < 0.001). Conclusions: AMN histology does not always reflects its associated PMP grade, while PMP grade strongly influences survival. Background: Targeted radionuclide therapy with 177Lu-labelled small conjugates is expanding rapidly, and its success is linked to appropriate patient selection. Companion diagnostic conjugates are usually labelled with 68Ga, offering good imaging up to ≈2 h post-injection. However, the optimal tumor-to-background ratio is often reached later. This study examined promising positron-emitting radiometals with half-lives between 3 h and 24 h and β+ intensity (Iβ+) ≥ 15% and compared them to 68Ga. The radiometals included: 43Sc, 44Sc, 45Ti, 55Co, 61Cu, 64Cu, 66Ga, 85mY, 86Y, 90Nb, 132La, 150Tb and 152Tb. 133La (7.2% Iβ+) was also examined because it was recently discussed, in combination with 132La, as a possible diagnostic match for 225Ac. Methods: Total electron and photon doses per decay and per positron; possibly interfering γ-ray emissions; typical activities to be injected for same-day imaging; positron range; and available production routes were examined. Results: For each annihilation process useful for PET imaging, the total energy released (MeV) is: 45Ti (1.5), 43Sc (1.6), 61Cu and 64Cu (1.8), 68Ga (1.9), 44Sc and 133La (2.9), 55Co (3.2), 85mY (3.3), 132La (4.8), 152Tb (6.5), 150Tb (7.1), 90Nb (8.6), and 86Y (13.6). Significant amounts (≥ 10%) of ≈0.5 MeV photons that may fall into the acceptance window of PET scanners are emitted by 55Co, 66Ga, 85mY, 86Y, 132La, and 152Tb. Compton background from more energetic photons would be expected for 44Sc, 55Co, 66Ga, 86Y, 90Nb, 132La,150Tb, and 152Tb. The mean positron ranges (mm) of 64Cu (0.6), 85mY (1.0), 45Ti (1.5), 133La (1.6), 43Sc and 61Cu (1.7), 55Co (2.1), 44Sc and 86Y (2.5), and 90Nb (2.6) were lower than that of 68Ga (3.6). DOTA chelation is applicable for most of the radiometals, though not ideal for 61Cu/64Cu. Recent data showed that chelation of 45Ti with DOTA is feasible. 90Nb requires different complexing agents (e.g., DFO). Finally, they could be economically produced by proton-induced reactions at medical cyclotrons. Conclusion: In particular, 43Sc, 45Ti, and 61Cu have overall excellent β+ decay-characteristics for theranostic applications complementing 177Lu-labelled small conjugates, and they could be sustainably produced. The black soldier fly, Hermetia illucens (L. 1758), is an omnivorous saprophagous insect with a high potential for valorizing organic by-products rich in carbohydrates. Among carbohydrates, H. illucens relies on soluble sugars for the growth and storage lipid synthesis. This study aimed to assess the impact of common soluble sugars on the development, survival, and fatty acid composition of H. illucens. Monosaccharides and disaccharides were individually incorporated into a chicken feed diet. Cellulose was used as a control. Larvae fed glucose, fructose, sucrose, and maltose grew faster than control larvae. In contrast, lactose exhibited anti-nutritional effects on larvae, slowing down growth and reducing final individual weight. However, all soluble sugars produced larvae fatter than the control diet. Notably, the tested sugars shaped the fatty acid profile. Maltose and sucrose increased saturated fatty acid content compared to cellulose. Conversely, lactose increased the bioaccumulation of dietary unsaturated fatty acids. This study is the first to demonstrate the effect of soluble sugars on the fatty acid profile of H. illucens larvae. The vertebrate heart tube extends by progressive addition of epithelial second heart field (SHF) progenitor cells from the dorsal pericardial wall. The interplay between epithelial mechanics and genetic mechanisms during SHF deployment is unknown. Here, we present a quantitative single-cell morphometric analysis of SHF cells during heart tube extension, including force inference analysis of epithelial stress. Joint spatial Principal Component Analysis reveals that cell orientation and stress direction are the main parameters defining apical cell morphology and distinguishes cells adjacent to the arterial and venous poles. Cell shape and mechanical forces display a dynamic relationship during heart tube formation. Moreover, while the T-box transcription factor Tbx1 is necessary for cell orientation towards the arterial pole, activation of Tbx5 in the posterior SHF correlates with the establishment of epithelial stress and SHF deletion of Tbx5 relaxes the progenitor epithelium. Integrating findings from cell-scale feature patterning and mechanical stress provides new insights into cardiac morphogenesis. The embryonic heart tube undergoes elongation via the addition of progenitors from the second heart field, though how epithelial mechanics and genetics interact during this process remains unknown. This study investigates the alteration processes affecting the smalt pigment in historical artworks through a combination of analytical techniques including X-ray Absorption Near Edge Structure (XANES) spectroscopy at the potassium (K) and cobalt (Co) K-edges, coupled with scanning electron microscopy with energy-dispersive X-ray spectroscopy (SEM-EDX) and micro X-ray fluorescence elemental mapping (µ-XRF). Laboratory mock-up samples prepared with different oil binding media were artificially aged, revealing varying rates of smalt degradation. µ-XANES analyses at the K K-edge provided grain-scale insights into smalt alteration, highlighting the influence of binding media composition on degradation kinetics. Crossed clustering data analysis over K µ-XANES acquired on both model samples and historical cross-sections corroborated findings and identified distinct spectral signatures associated with different stages of smalt degradation. Although the use of verditer, a synthetically produced copper carbonate pigment chemically analogous to azurite (Cu3(CO3)2(OH)2), in early modern wall paintings is regularly reported in grey literature, its detection in wall paintings is typically unavailable in a public forum. This article focusses on two particular samples (M9 and T1) initially suspected to contain copper blue pigments from decorative wall painting schemes in domestic houses across a range of status groups, in eastern and south-eastern England, dated to 1550-1650. Interest in verditer synthesis procedures is found in textual sources including both historic manuals and current scientific literature. The unreliability of the synthesis, particularly for blue verditer, however, is widely documented. Here, we report a multimodal study employing SEM-EDX, SR- μ PXRD mapping, and SR- μ XANES spectroscopy to characterise the phases present in two wall painting samples, one c. 1600/early 1600s (M9) and the other c. 1600 (T1). In addition to assessment of the presence of verditer, this study presents the first identification of rouaite, a synthetic copper hydroxynitrate mineral (Cu2(NO3)(OH)3), in wall painting samples. Rouaite, shown in a previous publication to be a common byproduct of replications of the historical verditer synthetic recipes, appears in combination with verditer and alone. Its infrequent identification in paintings and murals is considered in light of analytical challenges. The presence of copper hydroxychlorides is also discussed in the context of rouaite stability and degradation. These results represent a significant development in our knowledge of the early modern palette in England and historical verditer synthesis and advances our understanding of how these painting schemes may be best preserved in the future. Purpose: Chemotherapy-induced neutropenia poses a significant risk to cancer patients, with pegfilgrastim being commonly used for its prevention. While pegfilgrastim can be administered via prefilled syringe or pen device, patient preferences and experiences with these delivery methods remain unclear. Methods: We conducted a prospective, open-label, randomized, observational trial (NCT05910164) at the Rafael Institute, France, comparing patient preferences for pegfilgrastim administration using a prefilled syringe versus a prefilled pen device. Patients undergoing chemotherapy and requiring pegfilgrastim were enrolled and randomized 1:1 to receive either syringe or pen first, with crossover administration. Questionnaires assessed patient preferences, learning experiences, autonomy, pain levels, emotional responses, satisfaction with nursing care, and empowerment. Results: Among 150 randomized patients (mean age 58 years; 69% female), both groups showed a preference for the pen device, with significantly higher mean scores favoring pen administration (4.94 ± 1.70 vs. 4.27 ± 1.84; p = 0.00106). Patients reported significantly lower perceived pain with pen administration and stronger positive emotions compared to syringe use. Satisfaction with nursing care was higher with syringe use. Empowerment levels were similar across groups but significantly stronger when using the pen in complete autonomy. Conclusion: A preference for pegfilgrastim administration via the pen device was observed, though this may have been influenced by the administration sequence and the absence of syringe self-administration. The insights gained can help inform clinical decision-making and improve patient-centered care in managing chemotherapy-induced neutropenia. Trial registration: NCT05910164 on June 15, 2023. The placement of peripheral venous catheters (PVC) is a frequent procedure in the emergency department (ED), which exposes patients to complications (hematoma, fluid leakage, phlebitis, edema, infection), increases hemolysis of blood samples, is time-consuming and costly. The main aim of this study is to analyze the rate of PVC nonuse in the ED and to identify predictive factors of their nonuse. This prospective single-center observational study was conducted in the ED of the Saint-Antoine Hospital in Paris, France between February and March 2022. Adult patients receiving a PVC were included. In addition to demographic and medical data, the reason for PVC prescription and the prescribing physician's expectation of PVC use were collected. A total of 304 patients were included, with a median age of 61.5 years (IQR: 43–79 years), of whom 152 (50%) were men. PVC were primarily prescribed for intravenous medication administration. Seventy-two (23.7%) PVC were not used. In multivariable analysis, the predictive factors of nonuse were the prescribing physician's expectation of nonuse [OR 6.35, CI 95% (2.64–15.29), for "no" and "not sure" vs. "yes" responses] and the reason for prescribing "just in case" [OR 3.54, CI 95% (1.37–9.17)]. PVC were not used in 23.7% of cases. Predictors of nonuse were the prescribing physician's expectation of nonuse and the reason for prescribing "just in case". Extreme precipitation events have substantial implications for water resources, ecosystems, and human populations in Central Africa (CA). Consequently, understanding the spatial variability of these events is crucial for effective climate change adaptation and water management strategies. In this study, we assess the performance of the state-of-the-art global climate models from Phase 6 of the Coupled Model Intercomparison Project (CMIP6) in simulating extreme precipitation events over CA. By considering three observational datasets, we evaluated the ability of sixteen CMIP6 models as well their multi-model ensemble (MME), to capture the patterns of extreme precipitation events. We then focus on key metrics such as duration and intensity, based on a total of ten indices of extreme precipitation events, defined by the Expert Team on Climate Change Detection and Indices (ETCCDI). The results showed that the individual models as well as the MME exhibited acceptable performance in reproducing the patterns of extreme precipitation events, especially when dealing with wet-day amount, frequency, dry spell duration and persistence of extreme precipitation over the two northern CA subregions (i.e., Sudano-Sahelian and Northern Equatorial). Moreover, the analyses revealed that CMIP6 models generally lack the ability of in simulating not only the wet spell duration, but also heavy precipitation indices, particularly over the central and southeastern CA subregions (i.e., Equatorial East and Southern Equatorial, respectively). These results highlight the remaining challenges in reproducing local-scale processes governing precipitation variability and extreme precipitation events over the region. The results of this study provide an understanding of both the strengths and limitations of the CMIP6 models over CA, which would help to improve regional climate projections and strengthen the ability of decision makers to assess the future risks associated with extreme precipitation events in the region. This paper presents an approach for extracting points from conic intersections by using the concept of pencils. This method is based on QC2GA—the two-dimensional version of QCGA (Quadric Conformal Geometric Algebra)—that is demonstrated to be equivalent to GAC (Geometric Algebra for Conics). A new interpretation of QC2GA and its objects based on pencils of conics and point space elements is presented, enabling the creation, constraining, and exploitation of pencils of conics. A Geometric Algebra method for computing the discriminants and center point of a conic will also be presented, enabling the proposition of an algorithm for extracting points from a conic intersection object. Transposable elements are ubiquitous mobile DNA sequences generating insertion polymorphisms, contributing to genomic diversity. We present GraffiTE, a flexible pipeline to analyze polymorphic mobile elements insertions. By integrating state-of-the-art structural variant detection algorithms and graph genomes, GraffiTE identifies polymorphic mobile elements from genomic assemblies or long-read sequencing data, and genotypes these variants using short or long read sets. Benchmarking on simulated and real datasets reports high precision and recall rates. GraffiTE is designed to allow non-expert users to perform comprehensive analyses, including in models with limited transposable element knowledge and is compatible with various sequencing technologies. Here, we demonstrate the versatility of GraffiTE by analyzing human, Drosophila melanogaster, maize, and Cannabis sativa pangenome data. These analyses reveal the landscapes of polymorphic mobile elements and their frequency variations across individuals, strains, and cultivars. Tebentafusp, a bispecific immune therapy, is the only drug that demonstrated an overall survival benefit in patients with metastatic uveal melanoma (MUM). Circulating tumor DNA (ctDNA) has emerged as a potential prognostic and predictive marker in the phase 3 IMCgp100-202 trial using multiplex PCR-based next-generation sequencing (NGS). In this study (NCT02866149), ctDNA dynamics were assessed using droplet digital PCR (ddPCR) in 69 MUM patients undergoing tebentafusp treatment. Notably, 61% of patients exhibited detectable ctDNA before treatment initiation, which was associated with shorter overall survival (median 12.9 months versus 40.5 months for patients with undetectable ctDNA; p < 0.001). Patients manifesting a 90% or greater reduction in ctDNA levels at 12 weeks demonstrated markedly prolonged overall survival (median 21.2 months versus 12.9 months; p = 0.02). Our findings highlight the potential of ddPCR-based ctDNA monitoring as an economical, pragmatic and informative approach in MUM management, offering valuable insights into treatment response and prognosis. Background: Bleeding events are common complications in critically ill patients with haematological malignancies. The objective of this study was to assess the incidence and identify determinants of ICU-acquired severe bleeding events in critically ill patients with haematological malignancies. We conducted a single-center retrospective study including all adult patients with a history of haematological malignancy requiring unplanned ICU admission over a 12-year period (2007–2018). The primary endpoint was the occurrence of ICU-acquired (i.e. after the first 24 h in the ICU) severe bleeding events, as defined as grades 3 or 4 of the World Health Organization classification. Results: A total of 1012 patients were analysed, mainly with a diagnosis of lymphoma (n = 434, 42.9%) and leukaemia or myelodysplastic syndrome (n = 266, 26.3%). Most patients were recently diagnosed (n = 340, 33.6%) and under active cancer treatment within the last 3 months (n = 604, 59.7%). The main cause for admission was infection (n = 479, 47.3%), but a significant proportion of patients were admitted for a primary haemorrhage (n = 99, 10%). ICU-acquired severe bleeding events occurred in 109 (10.8%) patients after 3.0 days [1.0–7.0] in the ICU. The main source of bleeding was the gastrointestinal tract (n = 44, 40.3%). Patients experiencing an ICU-acquired severe bleeding event displayed prolonged in-ICU length of stay (9.0 days [1.0–6.0] vs. 3.0 [3.5–15.0] in non-bleeding patients, p < 0.001) and worsened outcomes with increased in-ICU and in-hospital mortality rates (55% vs. 18.3% and 65.7% vs. 33.1%, respectively, p < 0.001). In multivariate analysis, independent predictors of ICU-acquired severe bleeding events were chronic kidney disease (cause-specific hazard 2.00 [1.19–3.31], p = 0.008), a primary bleeding event present at the time of ICU admission (CSH 4.17 [2.71–6.43], p < 0.001), non-platelet SOFA score (CSH per point increase 1.06 [1.01–1.11], p = 0.02) and prolonged prothrombin time (CSH per 5-percent increase 0.90 [0.85–0.96], p = 0.001) on the day prior to the event of interest. Conclusions: Major bleeding events are common complications in critically ill patients with haematological malignancies and are associated with a worsened prognosis. Objective: To retrospectively evaluate the long-term outcomes after percutaneous cryoablation of abdominal wall endometriosis (AWE). Method: The Institutional Review Board approved this retrospective observational review of 40 consecutive patients, of a median age of 37 years (interquartile range [IQR] 32–40 years), presenting with a total of 52 symptomatic AWE nodules. All patients underwent cryoablation between January 2013 and May 2022 with a minimum follow-up period of 12 months. Outcomes were assessed using a visual analog scale (VAS) that measured pain, as well as by magnetic resonance imaging (MRI). The pain-free survival rates were derived using the Kaplan–Meier estimator. Adverse events were analyzed and graded using the classification system of the Cardiovascular and Interventional Radiological Society of Europe. Results: The median follow-up time was 40.5 months (IQR 26.5–47.2 months). The median VAS score before cryoablation was 8 (IQR 7–9). Complete relief of symptoms was documented in 80% (32/40) of patients at 3 months after initial cryoablation and correlated with the absence of residual endometriosis nodules on MRI. The median pain-free survival rates were 89.2% [95% CI, 70.1–96.4%] at 36 months and 76.8% [95% CI, 55.3–83.8%] after 60 months. No patient or lesion characteristics were found to be prognostic of failure. No major adverse events or side effects were reported in long term. Conclusion: Cryoablation safely and effectively afforded long-term pain relief for patients with AWE nodules. Clinical relevance statement: AWE cryoablation was found to be safe and effective in the long-term. Reidl et al. (Eur J Comb 75:152–168, 2019) characterized graph classes of bounded expansion as follows: A class C closed under subgraphs has bounded expansion if and only if there exists a function f : N → N such that for every graph G ∈ C , every nonempty subset A of vertices in G and every nonnegative integer r, the number of distinct intersections between A and a ball of radius r in G is at most f(r) |A|. When C has bounded expansion, the function f(r) coming from existing proofs is typically exponential. In the special case of planar graphs, it was conjectured by Sokołowski (Electron J Comb 30(2):P2.3, 2023) that f(r) could be taken to be a polynomial. In this paper, we prove this conjecture: For every nonempty subset A of vertices in a planar graph G and every nonnegative integer r, the number of distinct intersections between A and a ball of radius r in G is O (r 4 | A |) . Purpose: Social eating (SE) is a corner stone of daily living activities, quality of life (QoL), and aging well. In addition to feeding functional disorders, patients with head and neck cancer (HNC) face individual and social psychological distress. In this aging population, we intended to better assess the influence of age on these challenges, and the role of self-stigmatization limiting SE in patients with and beyond HNC. Methods: This was an exploratory multicenter cross-sectional mixed method study. Eligibility criteria were adults diagnosed with various non-metastatic HNC, before, during, or until 5 years after treatment. SE disorders were explored with the Performance Status Scale Public Eating rate (PSS-HN PE). In the quantitative part of the study, SE habits, Functional Assessment of Cancer Therapy Body Image Scale (FACT-MBIS) and specific to HNC (FACT-HN35) were also filled in by the patients. In the qualitative study, the semi-structured interview guide was drawn out to explore stigma, especially different dimensions of self-stigmatization. Results: A total of 112 patients were included, mean age 64.7 years, 23.2% of female. One-third (n = 35) of patients had an abnormal PSS-HN PE rate < 100. Younger patients had more often an impaired Normalcy of Diet mean (70.4 vs 82.7, p =.0498) and PE rates (76 vs 86.9, p =.0622), but there was no difference between age subgroups in MBIS nor FACT-HN scores. Seventy patients (72.2%) found SE and drinking « important» to « extremely important» in their daily life. The qualitative study reported self-stigmatization in two older patients and strategies they have developed to cope with in their behaviors of SE. Conclusion: This study confirms that SE remains of high concern in patients with and beyond HNC. Even in older patients experiencing less often functional feeding disorders, body image changes and SE issues are as impaired as in younger patients and need to be addressed. [ABSTRACT FROM AUTHOR]

Objectives: Hysterosalpingography (HSG) is widely used for evaluating the fallopian tubes; however, controversies regarding the use of water- or oil-based iodine-based contrast media (CM) remain. The aim of this work was (1) to discuss reported pregnancy rates related to the CM type used, (2) to validate the used CM in published literature, (3) to discuss possible complications and side effects of CM in HSG, and (4) to develop guidelines on the use of oil-based CM in HSG. Methods: A systematic literature search was conducted for original RCT studies or review/meta-analyses on using water-based and oil-based CM in HSG with fertility outcomes and complications. Nine randomized controlled trials (RCTs) and 10 reviews/meta-analyses were analyzed. Grading of the literature was performed based on the Oxford Centre for Evidence-Based Medicine (OCEBM) 2011 classification. Results: An approximately 10% higher pregnancy rate is reported for oil-based CM. Side effects are rare, but oil-based CM have potentially more side effects on the maternal thyroid function and the peritoneum. Conclusions: 1. HSG with oil-based CM gives approximately 10% higher pregnancy rates. 2. External validity is limited, as in five of nine RCTs, the CM used is no longer on the market. 3. Oil-based CM have potentially more side effects on the maternal thyroid function and on the peritoneum. 4. Guideline: Maternal thyroid function should be tested before HSG with oil-based CM and monitored for 6 months after. Clinical relevance statement: Oil-based CM is associated with an approximately 10% higher chance of pregnancy compared to water-based CM after HSG. Although side effects are rare, higher iodine concentration and slower clearance of oil-based CM may induce maternal thyroid function disturbance and peritoneal inflammation and granuloma formation. The electron beam-induced current technique (EBIC) developed at CEA-Leti has been a valuable tool for studying the transport properties of minority carriers in HdCdTe. Indeed, previous work has demonstrated the use of this technique for estimating diffusion length from the exponential decay of EBIC as a function of junction distance, as well as direct measurement of the modulation transfer function (MTF) of small pixel pitch diodes. In this work, a modulated electron beam and a lock-in amplifier are used to measure the variation in current and phase shift with the scan distance to the junction. Increasing interest in reducing the size, weight, and power consumption (SWaP) of devices motivates the development of high-operating-temperature (HOT) infrared imaging systems. HgCdTe photodiodes are in a privileged position to produce HOT detectors with background-limited infrared performance (BLIP). However, the proportion of unstable pixel responses tends to rise with increased operating temperature and degrades the detection performance. Multiple studies suggest that this issue is related to the surface states generated and affected by the manufacturing steps. In this work, native oxide and a residual halogen impurity susceptible to being introduced at the interface during passivation processes were investigated. For this purpose, surface contaminations were intentionally amplified to study their impact on the interface behavior of simplified electrical test structures. The chemical environments of the sample surface before insulator deposition were analyzed using x-ray photoelectron spectroscopy (XPS). After passivation deposition, time-of-flight secondary ion mass spectrometry (TOF-SIMS) enabled the evaluation of the contaminants' presence levels at the interface before and after annealing. The influence of the contaminants on the interface behavior was studied using capacitance–voltage measurements on metal–insulator–semiconductor structures (C(V) MIS). Finally, photoluminescence decay (PLD) enabled the extraction of the minority charge carrier effective lifetime to determine the contaminants' global impact on the structures. The coupling of physicochemical (XPS, TOF-SIMS) and electro-optical characterization techniques (PLD, C(V) MIS) allowed us to evaluate and discuss the effects of interfacial oxide and halogen impurities on passivation quality and subsequent device electrical behavior. Atrial arrhythmias are an important cause of morbidity and mortality in adults with congenital heart disease (ACHD). In acquired heart disease, the left atrial (LA) strain has been shown to predict supraventricular tachyarrhythmias (SVT). This study aimed to investigate whether reduced LA strain is associated with SVT in ACHD patients. This retrospective, single-center cohort study collected baseline clinical and echocardiographic data of 206 ACHD patients (157 left heart defect, 49 right heart defect). Patients with sinus rhythm at baseline and a 5-year follow-up (median age 29, IQR 22–41 years) were included. Diagnosis of sustained SVT was determined from clinical reports during the follow-up period. New or recurrent sustained SVT occurred in 16 patients (7.8%, median follow-up of 6.2 years). Patients who developed SVT were older, more likely to have diastolic dysfunction, and had larger LA dimensions, left ventricular mass, and a lower peak LA longitudinal strain (PALS). Lower PALS was associated with higher risk of SVT in patients with left and right heart defects. Patients in the lowest quartile for PALS had a 15.9-fold higher hazard ratio of SVT (95% confidence interval, 4.5 to 56.0, p < 0.001) in comparison with the top three quartiles. PALS provides information about the occurrence of SVT in the ACHD population. Lithium-ion batteries are expected to operate within a narrow temperature window around room temperature for optimal performance and lifetime. Therefore, in cold environments, electric vehicle battery packs must be extensively preheated prior to charge or discharge. However, conventional preheating is accomplished externally, which is slow and thus significantly increases charging times. Recently, internal heating has been demonstrated as a potential solution to quickly and uniformly preheat a lithium-ion pouch cell. However, internal heating has not been evaluated in other battery formats such as cylindrical batteries. In this work, we present a numerical model of a 4680 battery with internal heaters for fast preheating in cold environments. The effects that the number of heater layers, heating duration, resting duration, environmental temperature, and boundary heat transfer coefficient have on the temperature heterogeneity of the battery were investigated. The results show that internal heating alone reduces the temperature variation within the battery by a factor of 5 compared to external heating, and by a factor of 20 when combining internal and external heating. This study further proves that internal preheating of lithium-ion batteries is a promising thermal management strategy, and provides guidance on potential design considerations and heating protocols to implement internal heating. Several models of working memory (WM), the cognitive system devoted to the temporary maintenance of a small amount of information in view of its treatment, assume that these two functions of storage and processing share a common and limited resource. However, the predictions issued from these models concerning this resource-sharing remain usually qualitative, and at which precise extent these functions are affected by their concurrent implementation remains undecided. The aim of the present study was to quantify this resource sharing by expressing storage and processing performance during a complex span task in terms of the proportion of the highest level of performance each participant was able to reach (i.e., their span) in each component when performed in isolation. Two experiments demonstrated that, despite substantial dual-task decrements, participants managed to preserve half or more of their best performance in both components, testifying for a remarkable robustness of the human cognitive system. This paper describes the Data Domotopia a 2300 + respondent self-administered web-based survey. It includes 100 + multi-purpose items about home-making and stillness in a moving world. We suppose that home-making can reveal coping strategies and resilience practices to make everyday life work – as home is a central location in people's activity-travel patterns. To describe this phenomenon, the concept of Domotopia is introduced, defining how people arrange, use, and experience their homes to cope with the pathologies of accelerated and liquid modernity (Bauman 2005). While the Data Domotopia is based on a mixed-method combining qualitative and quantitative material, this paper focuses mainly on the description of the questionnaire – which is organized into three interrelated layers: the dwelling, the dwellers, and the neighborhood. Each of these layers unfolds in functional, social, emotional and sensory components. The survey covers most of the contemporary issues related to home-making. This includes the domestic space and gender issues; the socio-spatial resources (mobility, action space, core, and wider social network); lifestyles, ideals, and residential aspiration; time pressures, time use, organization and stress; equipment, rules and arrangements; interpersonal relations, cohabitation and negotiation, dominance and power. Intakes on the Data Domotopia is given by two concrete cases about the time-space coverage of the habitual action space, and about inter-personal task allocation. These examples show the potential of the data to study domocentric stillness and resilience to urban pathologies. The deleterious impact of antibiotics (ATB) on the microbiome negatively influences immune checkpoint inhibitors (ICI) response in patients with cancer. We conducted a randomized phase I study (EudraCT:2019-A00240-57) with 148 healthy volunteers (HV) to test two doses of DAV132, a colon-targeted adsorbent, alongside intravenous ceftazidime-avibactam (CZA), piperacillin-tazobactam (PTZ) or ceftriaxone (CRO) and a group without ATB. The primary objective of the study was to assess the effect of DAV132 on ATB plasma concentrations and both doses of DAV132 did not alter ATB levels. Secondary objectives included safety, darkening of the feces, and fecal ATB concentrations. DAV132 was well tolerated, with no severe toxicity and similar darkening at both DAV132 doses. DAV132 led to significant decrease in CZA or PTZ feces concentration. When co-administered with CZA or PTZ, DAV132 preserved microbiome diversity, accelerated recovery to baseline composition and protected key commensals. Fecal microbiota transplantation (FMT) in preclinical cancer models in female mice from HV treated with CZA or PTZ alone inhibited anti–PD-1 response, while transplanted samples from HV treated with ATB + DAV132 circumvented resistance to anti–PD-1. This effect was linked to activated CD8+ T cell populations in the tumor microenvironment. DAV132 represents a promising strategy for overcoming ATB-related dysbiosis and further studies are warranted to evaluate its efficacy in cancer patients. The oral antibiotic adsorbent DAV132 can reduce fecal concentrations of antibiotics while preserving their pharmacokinetic properties. Here, in a randomized trial in healthy volunteers treated with antibiotics, the authors show that DAV132 does not affect plasma concentrations of the antibiotics but preserves microbiome diversity and composition, with implications for cancer immunotherapy. Purpose: To report the oncological outcomes and the tolerance between 6 instillations and more than 6 cycles of hyperthermic intravesical chemotherapy(HIVEC) in patients with non-muscle invasive bladder cancer(NMIBC). Methods: This is a multicenter retrospective study from a national database including 9 expert centers. All patients treated with HIVEC between 2016 and 2023 for NMIBC were included. Patients were classified into two groups according to the total number of HIVEC instillations, including induction plus maintenance. Kaplan-Meier curves were computed to present survival outcomes. Results: 261 patients with a median follow-up of 25.5 months were included. 199(76.2%) and 62(23.8%) were treated by 6 and more than 6 cycles of HIVEC, respectively. The 2-years RFS(40.2% vs. 34.4%,p = 0.3) and the 2-years PFS(86% vs. 87%,p = 0.85) were similar between group treated with 6 and more than 6 instillations. 2-years CSS and OS were also similar between both groups. Univariate Cox regression showed no association between the number of bladder instillation and RFS (HR = 1.2 95%CI[0.8–1.84], p = 0.3) or PFS (HR = 0.8 95%CI[0.29–2.02], p = 0.2). In the group treated with more than 6 cycles, 2-years RFS and 2-years PFS were similar between patients who received induction plus maintenance compared to those treated with induction only. Finally, hematuria and urinary burning were significantly higher in the group treated by more than 6 cycles (21% vs. 8.5%(p < 0.01),and 29% vs. 17% (p = 0.03), respectively). Serious side effects(grade ≥ 3) are rare(3.1%) and similar in both groups. Conclusions: Results show no significant difference in two years RFS, PFS, CSS and OS according to number of instillations received, while toxicity profile seems better in the group receiving six instillations only.
